Abstract
Description
(1)菌株の分離及び選別
抗菌活性のある菌株を選別するために、在来式味噌玉麹から分離したバチルス菌株を用いた。菌株は大豆発酵液体培地に37で36時間前培養した。病原性カビ及び食中毒誘発細菌がローン細胞(lawn cell)となっている寒天固体培地に直径8mmのペーパーディスクを載せ、前培養した菌株を接種した。12時間培養し、その後培地に接種した菌株の周囲に真菌類及び細菌に対して同時に透明な円を形成する菌株を選別した。
当業界で公知の通常の方法により、前記選別した菌株の16S rDNAとgyrAの塩基配列を分析した。塩基配列は、NCBIのBLASTを用いて、GenBankデータベースと、選別した菌株の16S rDNAの塩基配列(配列番号1)及びgyrAの塩基配列(配列番号2)とを比較した。その結果、本菌株の16S rDNA塩基配列はバチルス・ベレゼンシスと99%の相同性を示し、gyrA塩基配列はバチルス・ベレゼンシスと100%の相同性を示した。

新たに分離したバチルス・ベレゼンシスCJBVが産生する抗菌物質を分離するための実験を行った。その過程を図1に示す。
新たに分離したバチルス・ベレゼンシスCJBVが真菌類及び食中毒誘発細菌に対して抑制する効果があるか否かを確認するために、次の抗菌活性実験を行った。
まず、カビであるコレトトリカム・オルビクラレ、フザリウム・オキシスポラムf.sp.ライコペルシシJCM12575(Fusarium oxysporum f.sp. lycopersici JCM12575)、カンジダ・アルビカンス、病原性酵母であるジゴサッカロマイセス・バイリに対して新たに分離したバチルス・ベレゼンシスCJBVが抗菌活性を示すか実験した。
また、新たに分離したバチルス・ベレゼンシスCJBVが発酵食品において食中毒誘発細菌にのみ特異的に抗菌活性を有するか調べるために、次の実験を行った。

Claims (16)
- 真菌類及び食中毒誘発細菌に特異的抗菌活性を有する、受託番号KCCM11528Pのバチルス・ベレゼンシスCJBV(Bacillus velezensis CJBV)菌株。
- 前記菌株は、ジャン(醤)類発酵細菌であるバチルス・サブティリス(Bacillus subtilis)、バチルス・アミロリケファシエンス(Bacillus amyloliquefaciens)に対して抗菌活性を有さない、請求項1に記載のバチルス・ベレゼンシスCJBV菌株。
- 前記真菌類は、コレトトリカム・オルビクラレ(Colletotrichum orbiculare)、フザリウム・オキシスポラム(Fusarium oxysporum)、カンジダ・アルビカンス(Candida albicans)、及びジゴサッカロマイセス・バイリ(Zygosaccharomyces bailii)からなる群から選択されるものである、請求項1に記載のバチルス・ベレゼンシスCJBV菌株。
- 前記食中毒誘発細菌は、バチルス・セレウス(Bacillus cereus)又はリステリア・モノサイトゲネス(Listeria monocytogenes)である、請求項1に記載のバチルス・ベレゼンシスCJBV菌株。
- 請求項1〜4のいずれかによるバチルス・ベレゼンシスCJBV菌株、前記菌株の胞子、前記菌株の培養物、その濃縮物、その抽出物、及びその乾燥物からなる群から選択される少なくとも1つを有効成分として含む、抗菌組成物。
- 請求項5による抗菌組成物を含む、食品組成物。
- 請求項5による抗菌組成物を含む、飼料組成物。
- 請求項5による抗菌組成物を含む、植物作物用抗菌剤。
- 請求項5による抗菌組成物を含む、微生物感染疾患の予防又は治療用薬学組成物。
- 前記微生物は、真菌類又は食中毒誘発細菌である、請求項9に記載の薬学組成物。
- 前記微生物感染疾患は、食中毒、皮膚真菌症、膣炎、下痢、嘔吐、腸炎、胃腸管炎、便泌、腹痛、腹部膨満、コレラ、リステリア症、蜂窩織炎(cellulitis)、尿路感染症、髄膜炎、腹膜炎、膀胱炎、リンパ管炎、ひょう疽(felon)、中耳炎、呼吸器疾患、肺炎、化膿性炎症、及び敗血症からなる群から選択されるものである、請求項9に記載の薬学組成物。
- 請求項5による抗菌組成物を含む、微生物感染疾患の予防又は改善用健康機能食品。
- 請求項5による抗菌組成物を含む、医薬部外品組成物。
- 請求項14によるバシロマイシンD類似体を含む、抗菌組成物。
- 前記抗菌組成物は、コレトトリカム・オルビクラレ、フザリウム・オキシスポラム、カンジダ・アルビカンス、ジゴサッカロマイセス・バイリ、バチルス・セレウス、及びリステリア・モノサイトゲネスの生育を抑制する、請求項15に記載の抗菌組成物。
