実施例1:タバコ抽出物の限外濾過
熱水(140〜155℃)およびタバコを8:1の水:タバコの比で混合することにより、タバコ抽出物を調製する。混合物を1時間撹拌し、次いで遠心分離する。こうして得られた上清を約25%固形分まで蒸発させる。抽出物を、50mL円錐管で再度遠心分離(4000rpmで10分間)して残っている任意の大きな粒子を除去する。真空濾過システムおよび7μm濾紙を使用して、上清を濾過する。
イソマルト、NaClおよびバニリンをポットで混合し、混合物の温度を143℃にする。イソマルトが溶融するまで混合物を143℃に維持し、次いで、温度を166℃に上昇させる。別の容器で、実施例1からの処理タバコ抽出物、マルチトールシロップ、H2O、スクラロースおよび任意選択によりL−リジンを混合して溶液を形成する。任意選択により、第2の別の容器で、水および水酸化ナトリウムを混合して溶液を形成する。
特定の無煙タバコ製品は、異なる添加剤で処理してアクリルアミドの量を減少させたタバコ抽出物を使用して調製する。実施例1により調製した限外濾過タバコ抽出物を添加剤と合わせて水中アクリルアミドを減少させ、溶液が形成するまで撹拌することにより、熱処理タバコ抽出物を調製する。得られた混合物を88℃に加熱し、60分間この温度で保持する。混合物を冷却し、追加の水を添加して混合物を200gの出発重量に戻す。
イソマルトおよびマルチトールシロップを、イソマルトが溶融するまでポットで混合する。別の容器で、実施例1により調製した処理タバコ抽出物、H2O、NaCl、スクラロースおよび香味剤を混合して溶液を形成する。
a)咽喉刺激緩和剤としてのショウガ
官能試験員に、3重量%のタバコ抽出物および1.5重量%のショウガ粉末を含む口内錠、ならびに3重量%のタバコ抽出物を含む(ショウガを含まない)対照口内錠を与える。対照口内錠を最初にサンプリングし、引き続いてショウガ含有口内錠をサンプリングする。1〜15の評価スケールを使用して、使用者が対照口内錠に関連するある程度の咽喉刺激を記述する。使用者は、ピーク口感覚強度とピーク咽喉感覚強度の両方の減少を記述し、ショウガ含有口内錠の全体的嗜好を記述する。
官能試験員に、3重量%のタバコ抽出物と2重量%のクエン酸ナトリウムおよび0.35重量%のオレオレジンショウガを含む口内錠、ならびに3重量%のタバコ抽出物を含む(クエン酸ナトリウムおよびオレオレジンショウガを含まない)対照口内錠を与える。オレオレジンショウガおよびクエン酸ナトリウム含有口内錠を最初にサンプリングし、引き続いて対照口内錠をサンプリングする。1〜15の評価スケールを使用して、使用者は、オレオレジンショウガおよびクエン酸ナトリウム含有口内錠についてのピーク口感覚強度とピーク咽喉感覚強度の両方の増加を記述する。オレオレジンショウガおよびクエン酸ナトリウム含有口内錠を最初にサンプリングしたので、この結果は官能試験員が試料間の差を識別することができないことによるものであり得る。強いぴりっとする感覚ならびに強力なショウガ風味を本質的に有するオレオレジンショウガは、口内錠の他の成分により引き起こされる任意の咽喉刺激をマスクするが、それ自体が特定の包含レベルで咽喉刺激に寄与し得ることも可能である。
タバコ抽出物を、凍結乾燥の代わりに、液体限外濾過抽出物をBiotage LLC、Charlotte、NCから入手可能な非インプリントポリマー(NIP)カートリッジに通過させてタバコ特異的ニトロソアミン(TSNA)を除去することを除いて、実施例1に記載のように調製する。驚くべきことに、NIPカートリッジが限外濾過抽出物中に残っている茶色タバコ色素の複数さえ除去して、抽出物を通る光透過の有意な増加をもたらすことが分かる。得られた処理抽出物を約20%固形分に濃縮し、噴霧乾燥する。
実施例1のプロセスを、0.1〜約0.5重量%のアスパラギナーゼ(DSM Food Specialties、Heerlen、NLからPreventASe(商標)としてまたはNovozymes、A/S、Bagsvaerd、DKからAcrylaway(登録商標)として入手可能)を実施例1の8:1の水:タバコスラリーに添加し、スラリーの温度を約60℃以下に維持するまたは実施例1に示すようにスラリーを加熱し、その後スラリーを約60℃以下に冷却した後、アスパラギナーゼを添加することにより変更する。酵素処理後、任意選択によりスラリーの温度を少なくとも約70℃に30分間以上上昇させることにより酵素を不活性化する。
Claims (33)
- a.半透明タバコ抽出物を含むタバコ材料と;
b.無煙タバコ製品の少なくとも約80重量%の量の砂糖代替物と;
c.糖アルコールシロップと
を含む、口の中で溶融可能および実質的に半透明な無煙タバコ製品。 - 前記砂糖代替物がガラス状マトリックスを形成することができる非吸湿性糖アルコールである、請求項1に記載の無煙タバコ製品。
- 前記砂糖代替物がイソマルトである、請求項1に記載の無煙タバコ製品。
- 前記砂糖代替物が無煙タバコ製品の少なくとも約85重量%の量で存在する、請求項1に記載の無煙タバコ製品。
- 前記砂糖代替物が無煙タバコ製品の少なくとも約90重量%の量で存在する、請求項1に記載の無煙タバコ製品。
- 前記糖アルコールシロップがマルチトールシロップである、請求項1に記載の無煙タバコ製品。
- 前記半透明タバコ抽出物が、前記タバコ抽出物をリジン、システイン、アスパラギナーゼまたは過酸化水素を含む水溶液中で加熱することにより、無煙タバコ製品への包含前に処理された熱処理タバコ抽出物である、請求項1に記載の無煙タバコ製品。
- 前記水溶液がNaOHをさらに含む、請求項7に記載の無煙タバコ製品。
- 前記半透明タバコ抽出物が約50000Da未満の分子量を有する化合物からなる、請求項1に記載の無煙タバコ製品。
- 前記半透明タバコ抽出物が約5000Da未満の分子量を有する化合物からなる、請求項1に記載の無煙タバコ製品。
- 前記半透明タバコ抽出物が、約600nmより大きい可視光波長で少なくとも約30%の光透過率%により特徴づけられる、請求項1に記載の無煙タバコ製品。
- 前記半透明タバコ抽出物が、約600nmより大きい可視光波長で少なくとも約40%の光透過率%により特徴づけられる、請求項1に記載の無煙タバコ製品。
- 前記半透明タバコ抽出物が、約600nmより大きい可視光波長で少なくとも約50%の光透過率%により特徴づけられる、請求項1に記載の無煙タバコ製品。
- 前記タバコ材料が粒子状タバコ材料をさらに含む、請求項1に記載の無煙タバコ製品。
- 前記半透明タバコ抽出物と前記粒子状タバコの重量比が約5:1〜約1:10である、請求項14に記載の無煙タバコ製品。
- 香味剤、甘味剤、塩、およびこれらの組み合わせからなる群から選択される1種または複数の成分をさらに含む、請求項1に記載の無煙タバコ製品。
- 以下:
無煙タバコ製品の約0.1〜約15重量%の量の香味剤;
無煙タバコ製品の約0.01〜約2重量%の量のスクラロース;および
無煙タバコ製品の約0.5〜約10重量%の量の塩化ナトリウム
の1種または複数を含む、請求項16に記載の無煙タバコ製品。 - 咽喉刺激緩和剤をさらに含む、請求項1に記載の無煙タバコ製品。
- 前記咽喉刺激緩和剤がクエン酸ナトリウム、ハチミツ、ショウガ、ペクチン、カプサイシン、カンフル、デキストロメトルファン、エキナセア、グルコン酸亜鉛、ペパーミント油、スペアミント油、ユーカリ油、グリセリン、有機酸、およびこれらの組み合わせもしくは抽出物からなる群から選択される、請求項18に記載の無煙タバコ製品。
- a.少なくとも約1.0重量%の約50000Da以下の分子量を有する成分からなる半透明タバコ抽出物と;
b.少なくとも約85重量%の量のガラス状マトリックスを形成することができる非吸湿性糖アルコールと;
c.前記非吸湿性糖アルコールの再結晶を遅くするのに十分な量の糖アルコールシロップと
を含む、請求項1に記載の無煙タバコ製品。 - a.少なくとも約0.5重量%の約50000Da以下の分子量を有する成分からなる半透明タバコ抽出物と;
b.少なくとも1.0重量%の粒子状タバコ材料と;
c.少なくとも約85重量%の量のガラス状マトリックスを形成することができる非吸湿性糖アルコールと;
d.前記非吸湿性糖アルコールの再結晶を遅くするのに十分な量の糖アルコールシロップと
を含む、請求項1に記載の無煙タバコ製品。 - 前記タバコ材料が限外濾過タバコ抽出物を含む、請求項1から21のいずれかに記載の無煙タバコ製品。
- (i)半透明タバコ抽出物、溶融状態でガラス状マトリックスを形成することができる非吸湿性糖アルコール、および糖アルコールシロップを混合して混合物を形成するステップと;
(ii)前記混合物を室温に冷却して半透明性を示す固形無煙タバコ製品を形成するステップと
を含む、半透明無煙タバコ製品を調製する方法。 - タバコ抽出物をサイズ排除クロマトグラフィー、精密濾過、限外濾過、ナノ濾過、逆浸透、またはこれらの組み合わせに供するステップにより前記半透明タバコ抽出物を形成するステップをさらに含む、請求項23に記載の方法。
- 前記供するステップが前記タバコ抽出物から約50000Daより上の分子量を有する成分を除去する、請求項24に記載の方法。
- 前記半透明タバコ抽出物を、タバコ特異的ニトロソアミンを除去するのに適した非インプリントポリマーによる処理に供するステップをさらに含む、請求項23に記載の方法。
- 前記タバコ抽出物をリジン、システイン、アスパラギナーゼまたは過酸化水素を含む水溶液中で加熱することにより、前記半透明タバコ抽出物を熱処理するステップをさらに含む、請求項23に記載の方法。
- 前記水溶液がNaOHをさらに含む、請求項27に記載の方法。
- 前記半透明無煙タバコ製品が約500ppb未満のアクリルアミド含量を有する、請求項23に記載の方法。
- 前記混合するステップが、前記半透明タバコ抽出物の非存在下で、前記非吸湿性糖アルコールをハードクラック段階より上の温度に加熱するステップと、次いでハードクラック段階未満の温度で、前記半透明タバコ抽出物を前記非吸湿性糖アルコールに混合するステップとを含む、請求項23に記載の方法。
- 前記ハードクラック段階が約145℃〜約155℃であり、前記非吸湿性糖アルコールが前記ハードクラック段階と約171℃との間の温度で加熱される、請求項30に記載の方法。
- 前記冷却ステップの前に、前記混合物を複数の型に導入して個々の製品単位を製造するステップをさらに含む、請求項23に記載の方法。
- 前記混合するステップが、粒子状タバコを前記混合物に添加するステップをさらに含む、請求項23から32のいずれか一項に記載の方法。
