Abstract
Description
本出願は、2011年1月12日出願の米国仮特許出願第61/431,944号および2011年11月11日出願の第61/558,527号に関連する。
図1は、神経幹細胞および神経前駆細胞の脳における3つの主な細胞型−ニューロン、アストロサイトおよびオリゴデンドロサイトへの発達を示す。実矢線は1つの細胞型のもう1つへの発達を示した。HFSC細胞からニューロン限定前駆細胞(NRP)までの破線は、HFSC細胞がニューロンとなる傾向がより低いことを示す。HFSC細胞からO2A細胞までの太線は、HFSC細胞がオリゴデンドロサイト系列細胞を生成する傾向がより大きいことを示す。HFSC細胞のニューロン、アストロサイトおよびオリゴデンドロサイトに分化する多能性は実施例7(図14参照)において示された。HFSC細胞のオリゴデンドロサイト系列細胞に分化する傾向は実施例2(図7参照)および実施例8(図15参照)において示された。
細胞生物学、タンパク質化学、および抗体技術における一般的な方法は、Current Protocols in Protein Science(J.E.Colligan et al.,eds.,Wiley&Sons);Current Protocols in Cell Biology(J.S.Bonifacino et al.,Wiley&Sons)およびCurrent Protocols in Immunology(J.E.Colligan et al.eds.,Wiley&Sons)において見つけることができる。細胞培養方法は一般的にはCulture of Animal Cellsの現行版:A Manual of Basic Technique(R.I.Freshney et al,Wiley&Sons);General Techniques of Cell Culture(M.A.Harrison&I.F.Rae,Cambridge Univ.Press)に記載される。組織培養供給用品および試薬は、Chemicon(登録商標)、Millipore(登録商標)、R&D Systems(登録商標)、Invitrogen(商標)、Nalgene−Nunc(商標)International、Sigma−Aldrich(商標)、およびScienCellのような商業供給業者から市販される。
いくつかの培地およびサプリメントは、ドナーのインフォームドコンセントとともにAdvanced Bioscience Resources,Inc.(アラマダ、カリフォルニア州、米国)から入手された、12週のヒト胎児脊髄に由来するHFSC細胞を用いる予備実験において試験された。細胞はまず、2mMのグルタミン(Invitrogen(商標))、1mMのピルベート(Invitrogen(商標))および2%のB27サプリメント(Invitrogen(商標))が添加されたDMEM:F−12(1:1)(DMEM/F12)(Invitrogen(商標)、カールスバド、カリフォルニア州、米国)において培養された。各種増殖因子は、それらがHFSC細胞の増殖を刺激するか試験された。もっとも効果的な増殖因子はPDGF−AAおよびbFGFの組み合わせだった。細胞はPDGF−AAおよびbFGFの存在下で増殖することができたが、液胞を示し、不健康に見えた。いくつかのサプリメントが試験され、2mMのグルタミン、1mMのピルベートおよび2%のB27サプリメントに加えて1%のNEAAが添加されたDMEM/F12は液胞を減少させ、細胞数をわずかに増加させることができたことが観察された(データ示さず)。1mMのN−アセチル−システイン(Sigma−Aldrich(商標)、セントルイス、ミズーリ州、米国)および55μMのβ−メルカプトエタノール(Invitrogen(商標))を含む他のサプリメントは細胞の状態を向上させると考えられたが、向上はNEAAほど顕著ではなかった(データ示さず)。よって、2mMのグルタミン、1mMのピルベート、2%のB27サプリメント、1%のNEAA、1mMのN−アセチル−システインおよび55μMのβ−メルカプトエタノールが添加されたDMEM/F12は最適なベース培地と同定され、その後HFSC細胞を培養するのに用いられた。
実施例1に記載されたように、培地にPDGF−AAおよびbFGFを含むことは、最初はHFSC細胞の適切な増殖を支持したが、増殖速度は2回の継代後に遅くなった。NT−3(R&D Systems)およびIGF−1(Sigma−Aldrich(商標))はHFSC細胞の増殖を向上することができたかを測定するため試験された。20ng/mlのPDGF−AAおよび10ng/mlのbFGFの存在は、細胞の増殖率を刺激するには不十分だった(増殖率は<1だった)(図4参照)。その後の5ng/mlのNT−3および/または10ng/mlのIGF−1の添加は、増殖率の向上をもたらした(増殖率は>1となった)。NT−3およびIGF−1の組み合わせは継代3でもっとも効果的だった。各条件から入手された細胞はそれらの効果を確認するためさらに継代された。細胞は、スフィアを形成し始めたので、早く回収された。継代4で、NT−3およびIGF−1の組み合わせによる増殖率の回復は低下し、IGF−1の単一添加が継代4でもっとも効果的となった。NT−3およびIGF−1の組み合わせは細胞の分化を引き起こし、またはより多くのスフィアを形成し得、培地交換中に失われた。よって、IGF−1の添加はもっとも効果的な生存因子と同定され、その後HFSC細胞を培養するのに用いられた。しかしながら、HFSC細胞の増殖速度は継代4の終わりで再度遅くなり始め、細胞数は播種細胞数と比較して低下した。従って、別のサプリメント、1−チオグリセロールが継代5でHFSC細胞の増殖を向上させることができたか試験された。
各種培養条件を試験しながら、HFSC細胞が培地からbFGFを除去して培養された場合、細胞の多くは(オリゴデンドロサイトを示す)二極または多極細胞に分化し、すぐに死滅した。これらの細胞死を回避するため、自発的分化技術が開発された。
図1に示されるように、従来の神経幹細胞はHFSC細胞の前駆細胞であると考えられる。この関係を確認するため、HFSC細胞は以前報告された従来の神経幹細胞から誘導することができたか試験された。ヒト胎児脊髄(胎齢11週)の第2組織サンプルからの細胞はまず、従来の神経幹細胞を増殖するため、2.5mMのグルタミン、15mMのHEPES、2%のB27サプリメント、1%のNEAA、1.5mMのピルベート、55μMのβ−メルカプトエタノール、および1mMのN−アセチル−システインを含有するDMEM/F12において、10ng/mlのEGFおよび10ng/mlのbFGFの存在下で15日間培養された。多くの異なる細胞型が最初は存在したが、いくつかの増殖可能な細胞集団は一次培養の終わりで入手された。これらの結果は継代0の15日目の図8、スライドAにおいて示される。第1継代後、細胞はクローン#2bと同じ条件(すなわち10ng/mlのbFGF、10ng/mlのIGF−1、100ng/mlのPDGF−AAおよび50μMの1−チオグリセロールを有するHFSCM1培地)において15回の継代によって培養された。
発明者らはHFSC細胞(クローン#2b)が構築された後各種培養条件を試験したが、本発明者らは各増殖因子の用量反応が変化したことに気づいた。HFSC細胞(クローン#2b)を単離するため、50μMの1−チオグリセロールに加えてより高濃度のPDGF−AA(100ng/ml)が必要だった。このクローンが絶えず増殖するようになった後、このクローンを増殖するのにより高濃度のPDGF−AA(100ng/ml)はこれ以上必要なかった。増殖率は約10〜20ng/mlのPDGF−AAで飽和され、より高濃度のPDGF−AA(100mg/ml)はそれらの増殖に対してさらなる効果を有さなかった。これは長期培養または1−チオグリセロールの継続使用のためであり得る。HFSC細胞のクローン#2bおよびクローン#3を構築するため、何回かの継代後1−チオグリセロールが用いられた。より高濃度のPDGF−AAの効果を試験するため、新たなクローンは初期培養から1−チオグリセロールの存在下で構築された。加えて、bFGFおよびIGF−1に対する反応はそれぞれ20ng/mlおよび40ng/mlで飽和されると考えられた。より高濃度のIGF−1(50〜500ng/ml)が用いられた場合、より多くの分化細胞を見ることができた(約1%)。従って、HFSC細胞を増殖するには20ng/mlのIGF−1が好ましいと考えられた。20ng/mlのbFGFおよび20ng/mlのIGF−1の使用は、10ng/mlのbFGFおよび10ng/mlのIGF−1の使用と比較して増殖率を5〜10%向上させた。従って、この実験には20ng/mlのbFGFおよび20ng/mlのIGF−1が用いられた。
クローン#2bの細胞は継代10、11および12で8%のDMSOの存在下で凍結保存された。継代11で凍結されたHFSC細胞(クローン#2b)はATCC(受入番号PTA−12291)に寄託された。クローン#3の細胞も継代9および10で8%のDMSOの存在下で凍結保存された。クローン#4Aおよび#4Bの細胞はクローン#2bまたは#3より速く増殖し、従ってそれらは同じ条件において継代4および5(クローン#4A)または継代3、4および5(クローン#4B)で8%のDMSOの存在下で凍結保存された。HFSC細胞を凍結するため、HFSC細胞を培養するのと同じ培地(HFSCM1培地および50μMの1−チオグリセロール)が用いられた。細胞はその後解凍され、10ng/mlのbFGF、10ng/mlのIGF−1、100ng/mlのPDGF−AAまたは20ng/mlのbFGF、20ng/mlのIGF−1、20ng/mlのPDGF−AAを有する上記無血清HFSCM1培地および50μMの1−チオグリセロールにおいて培養された。これらの細胞は凍結前と同様の速度で増殖したことが観察された(図6、図9および図11参照)。凍結細胞は図12〜15に示されるその後の実験に用いられた。
実施例2のHFSC細胞が100ng/mlのPDGF−AA、10ng/mlのbFGF、10ng/mlのIGF−1および50μMの1−チオグリセロールの存在下で培養された場合、それらは図3、スライドD〜Fに示されるようにクラスターおよび/またはスフィアに増殖した。クラスターから分離された拡散細胞は、図3、スライドDおよびEに示されるようにオリゴデンドロサイトに分化する傾向があった。細胞が単一細胞状態で継代された場合でも、それらは最終的には再度集まり、クラスターを形成し始めた。この増殖状態でのそれらの形状はオリゴデンドロサイトプレ前駆細胞に似ていたが(Ben−Hur et al,J.Neurosci.,18:5777,1998;Gago et al,Mol.Cell.Neurosci.,22:162,2003参照)、いずれのクラスターも形成することなく二極形態で増殖するO−2A前駆細胞とは違った。ラットオリゴデンドロサイトプレ前駆細胞はずっと以前に報告されたものの、ヒト対応物は報告されていない。しかしながら、それらはいずれの場合も(以下に示されるような)オリゴデンドロサイトに分化するそれらの能力を保持したので、増殖細胞培養におけるオリゴデンドロサイト前駆細胞の正確な段階を同定することは不要だった。
増殖細胞の分化能を試験するため、HFSC細胞(クローン#3)は分離/単一細胞期に継代され、分化を刺激する血清含有培地[オリゴデンドロサイト前駆細胞分化培地(OPCDM)](ScienCell(商標)Research Laboratories)において培養された。細胞は次にニューロン(抗βIIIチューブリン抗体、抗ニューロフィラメント−L抗体および抗MAP2抗体)、オリゴデンドロサイト前駆細胞およびオリゴデンドロサイト[O4抗体、O1抗体、抗GalC抗体および抗ミエリン塩基性タンパク質(MBP)抗体]、ならびにアストロサイト(抗GFAP抗体)を識別する抗体、その後蛍光染色標識二次抗体(DyLight488またはDyLight594、Jackson ImmunoResearch)で染色された。DAPIは細胞核を対比染色するために用いられた。
HFSC細胞は、図5において示されるように、PDGF−AA濃度を低減し、bFGFを補充しないことにより、オリゴデンドロサイトへの良好な分化能を示した。しかしながら、分化細胞は総細胞の半分未満だった。HFSC細胞が、bFGFなしで、10ng/mlのPDGF−AAおよび100ng/mlのIGF−1とともに、非常に低密度(<0.5×104細胞/cm2)で播種された場合、ほとんどの細胞は分化したと考えられたが、それらは1日以内に失われた。それらのオリゴデンドロサイトへの分化能をさらに試験するため、分化の誘導および長期細胞生存は非常に重要であると考えられた。環状AMP(cAMP)は多くの細胞型の分化を誘導することが知られる。cAMPの細胞透過性アナログであるpCPT−cAMPはHFSC細胞において分化を誘導することができたか試験された。100μMのpCPT−cAMPは高密度(3×104細胞/cm2)でのHFSC細胞の分化を誘導することができたが、細胞は100ng/mlのIGF−1の存在下でも十分に生存することができなかった。BDNFはオリゴデンドロサイト前駆細胞の分化を向上し、分化細胞の細胞生存を支持することが報告されている。HFSC細胞がグルタミンおよびHEPESを含有し、B27サプリメント、N2サプリメントおよび50μMの1−チオグリセロールが添加されたDMEM/F12において10ng/mlのPDGF−AA、100ng/mlのIGF−1、100μMのpCPT−cAMPおよび10ng/mlのBDNFの存在下で分化された場合、HFSC細胞の少なくとも半分より多くはプロセスベアリング細胞に分化された。HFSC細胞はO4またはNG2のようないくつかのオリゴデンドロサイトマーカーを発現するので、HFSC細胞およびオリゴデンドロサイト系列細胞を識別する新規方法が必要だった。いくつかのトライアルに基づき、本発明者らは。未分化細胞がオリゴデンドロサイト前駆細胞より強くGD3を発現し、O4について逆だったことに気づいた(図11、スライドA、スライドBおよびスライドCの矢頭)。細胞がGD3およびO4で染色された場合、オリゴデンドロサイトはそれらの染色パターンおよびそれらの形態を用いて識別することができた。加えて、ニューロンまたはアストロサイトのような他の細胞型も、それらはGD3またはO4抗原を発現しないため、識別することができた。図15、スライドDは各細胞型の割合を示す。未分化細胞は総細胞の23.5%±2.0%だった。オリゴデンドロサイト前駆細胞は総細胞の75.8%±2.1%だった。他の細胞型は0.9%±0.6%のみだった。上記のように、オリゴデンドロサイト前駆細胞の分化細胞(オリゴデンドロサイト前駆細胞+他の細胞型)に対する割合は分化細胞の99.1%±0.56%だったが、他の細胞型は分化細胞の0.9%±0.56%だった。このデータは、HFSC細胞がオリゴデンドロサイト前駆細胞への高い分化能を有することを示す。
本発明は、CD133陽性、CD140a陽性、CD9陽性、CD44陰性、PSA−NCAM陽性、A2B5陽性、O4陽性、およびNG2陽性であるHFSC細胞の表現型について開示した。この情報は培養なしにHFSC細胞を選択または富化することを可能にする。CD133は神経幹細胞のマーカーであり、前駆細胞において発現しない。CD9も神経幹細胞のマーカーとして用いられるが、いくつかのオリゴデンドロサイトはCD9を発現することが知られる。PSA−NCAMおよびA2B5はニューロン限定前駆細胞またはグリア限定前駆細胞を検出するのに用いられる。ほとんどの神経前駆細胞はPSA−NCAMおよびA2B5またはPSA−NCAMもしくはA2B5を発現すると考えられ、それらの使用は、とくに第1および第2三半期においてHFSC細胞をそれほど十分には富化することができない。CD140a、NG2、A2B5およびO4はオリゴデンドロサイト前駆細胞、プロ−オリゴデンドログリアおよびオリゴデンドロサイトのマーカーとして用いられる。CD140aおよびNG2の発現レベルはHFSC細胞においてオリゴデンドロサイト前駆細胞、プロ−オリゴデンドログリアまたはオリゴデンドロサイトより高かったが、A2B5およびO4の発現レベルはHFSC細胞においてオリゴデンドロサイト前駆細胞、プロ−オリゴデンドログリアまたはオリゴデンドロサイトより低かった。CD140aおよびNG2の使用はHFSC細胞を富化するのにより適していると考えられる。上記情報および本発明において記載されるデータに基づき、HFSC細胞を富化する各マーカーの効果は、CD140a>NG2>CD9>CD133>A2B5>O4、PSA−NCAMとなるが、この順はそれらの胎齢によって変わるだろう。
Claims (44)
- 前駆細胞または幹細胞であり、
ニューロン、アストロサイト、およびオリゴデンドロサイトに分化するその能力を維持し、
その後の継代を通して効率的にオリゴデンドロサイト系列細胞に分化するその能力を維持し、
少なくとも細胞表面抗原CD133およびCD140αを発現する、
単離された増殖可能なヒト神経細胞。 - ATCC受入番号PTA−12291として寄託された、請求項1に記載の単離された増殖可能なヒト神経細胞。
- 脊髄、大脳皮質、海馬、線条、前脳基底核、腹側中脳、青斑核、視床下部、小脳、脳梁および視神経からなる群から選択されたヒト胎児神経組織に由来する、請求項1に記載の増殖可能なヒト神経細胞。
- 前記神経組織が胎齢8〜24週のヒト脊髄から単離される、請求項3に記載の増殖可能なヒト神経細胞。
- 効果的な量の1つの増殖サプリメント、2つの増殖因子、および1つの生存因子を含む培地を含む、これを富化するのに効果的な条件下で培養された、請求項1に記載の増殖可能なヒト神経細胞。

- その後の継代を通してニューロン、アストロサイト、およびオリゴデンドロサイトに分化するその能力、ならびに少なくとも細胞表面抗原CD133およびCD140αを発現するその能力を失うことなく、凍結および解凍することができる、請求項1に記載の増殖可能なヒト神経細胞。
- 哺乳類中枢神経系から単離された前駆細胞または幹細胞であり、ニューロン、アストロサイト、およびオリゴデンドロサイトに分化するその能力ならびに効率的にオリゴデンドロサイト系列細胞に分化するその能力を維持する、増殖可能な神経細胞のインビトロ培養方法であって、
a)ヒト胎児神経組織から少なくとも1つの細胞を単離および分離するステップ、
b)該細胞を37℃の温度で、1〜20%のO2、および5%のCO2を含む雰囲気において、ならびに
−少なくとも5ng/mlのPDGF−AA
−少なくとも0.5ng/mlのbFGF、および
−少なくとも10μMの1−チオグリセロール
を含む既知組成の無血清培地において培養するステップ、
c)b)からの該細胞を継代し、該増殖可能なヒト神経細胞を入手するステップ
を含む、方法。 - 前記培地が少なくとも1.0ng/mlのIGF−1をさらに含む、請求項17に記載の方法。
- 前記神経組織が脊髄、大脳皮質、海馬、線条、前脳基底核、腹側中脳、青斑核、視床下部、小脳、脳梁および視神経からなる群から選択される、請求項17に記載の方法。
- ステップa)では、前記神経組織が胎齢8〜24週のヒト胎児脊髄からのものである、請求項17に記載の方法。
- ステップa)では、前記単離ステップが蛍光活性化細胞選別(FACS)または蛍光パニングの少なくとも1つを用いる単離方法により行われる、請求項17に記載の方法。
- 前記培地が
−40〜200ng/mlのPDGF−AA、
−5〜100ng/mlのbFGF、
−10〜100μMの1−チオグリセロール、および
−5〜100ng/mlのIGF−1
を含む、請求項18に記載の方法。 - 前記培地が
−約100ng/mlのPDGF−AA、
−約20ng/mlのbFGF、
−約50μMの1−チオグリセロール、および
−約20ng/mlのIGF−1
を含む、請求項18に記載の方法。 - 前記培地が
−5〜100ng/mlのPDGF−AA、
−1〜50ng/mlのbFGF、
−10〜100μMの1−チオグリセロール、および
−5〜100ng/mlのIGF−1
を含む、請求項18に記載の方法。 - 前記培地が
−約20ng/mlのPDGF−AA、
−約20ng/mlのbFGF、
−約50μMの1−チオグリセロール、および
−約20ng/mlのIGF−1
を含む、請求項18に記載の方法。 - 前記培養ステップがポリオルニチンまたはポリリシンでコーティングされた培養容器において行われる、請求項18に記載の方法。
- 前記既知組成の無血清培地が非必須アミノ酸、グルタミン、ピルベート、B27、N−アセチル−システインおよびβ−メルカプトエタノールが添加されたDMEM/F12を含む、請求項18に記載の方法。
- 哺乳類中枢神経系から入手された少なくとも1つの単離神経細胞を含むインビトロ培養であって、該分離細胞を
−少なくとも5ng/mlのPDGF−AA、
−少なくとも5ng/mlのbFGF、および
−少なくとも10μMの1−チオグリセロール
を含む既知組成の無血清培地に浸漬する、培養。 - 少なくとも1.0ng/mlのIGF−1をさらに含む、請求項28に記載のインビトロ培養。
- 前記細胞がヒト胎児脊髄から入手される、請求項29に記載のインビトロ培養。
- ポリオルニチンまたはポリリシンでコーティングされた培養容器における、請求項30に記載のインビトロ培養。

対象に治療効果的な量の請求項1に記載の単離された増殖可能なヒト神経細胞を含む組成物を投与するステップ
を含む、方法。 - 前記症状が脱髄疾患または神経変性疾患である、請求項34に記載の方法。
- 前記脱髄疾患が脊髄傷害(SCI)、多発性硬化症(MS)、遺伝性白質ジストロフィ、横断性脊髄障害/脊髄炎、進行性多発性局所白質脳症および他の先天性脱髄疾患からなる群から選択される、請求項35に記載の方法。
- 前記神経変性疾患がアルツハイマー病、アルツハイマー型の老年痴呆(SDAT)、パーキンソン病、ハンチントン病、筋委縮性側索硬化症(ALS)、虚血症、視覚消失症および有髄ニューロンへの傷害により引き起こされる神経変性疾患からなる群から選択される、請求項35に記載の方法。
- 前記投与ステップが前記脱髄疾患または前記神経変性疾患に影響される神経組織または側脳室への前記増殖可能なヒト神経細胞の注射を含む、請求項35に記載の方法。
- 前記神経組織が脊髄、脳室下帯、脳梁、小脳、基底核、および黒質からなる群から選択される、請求項38に記載の方法。
- 前記投与ステップが前記増殖可能なヒト神経細胞の前記対象への、トランス子宮胎児脳室内注射、脳室内注射、実質内注射、硝子体注射および血管内投与からなる方法による注射を含む、請求項35に記載の方法。
- 前記投与ステップの前に前記増殖可能なヒト神経細胞を分化するステップをさらに含む、請求項35に記載の方法。
- 請求項1のような単離された増殖可能なヒト神経細胞を含む医薬神経幹細胞組成物。
- 請求項17に記載の方法によりインビトロ培養された増殖可能な神経細胞を含む医薬神経幹細胞組成物。
