で表される化合物である。上記の通り、硫酸基は、それらが多数アセンブリされて活性化及び増殖細胞への取り込み効果を生じると、直接的エフェクター機能を呈する。Mは、好ましくはナトリウムである。
スクロース,マンノース又はトレハロースは、本発明の硫酸化ポリグリセリン及びその複合体の量の好ましくは1−100倍,さらに好ましくは5−20倍の量で使用される。最も好ましくは、トレハロースの5−20倍量での使用(例えば、10mgの薬物に対して100mgのトレハロース)である。
Bioconjugate Chemistry 15, 2004, 162-167; Advanced Materials 12, 2000, 235-239; Macromolecules 32, 1999, 4240に従って、樹状ポリグリセリンを得、特性決定する。
異なる平均分子量の様々な高分子材料を、異なる出発分子(1,1,1−トリス(ヒドロキシメチル)プロパン(TMP)、1,1,1−トリス(ヒドロキシメチル)エタン(TME)及びペンタエリスリトール(PE))から得、硫酸化反応に供する。
硫酸化ポリグリセリンを、Bioconjugate Chemistry 15, 2004, 162-167の実験に関する記載に基づいて合成する。高度の硫酸化を達成するために、文献記載の方法を改良する。SO3/ピリジン複合体を、90℃下、1.2倍モル過剰で加え、90℃下での攪拌を18時間継続する。透析及び限外ろ過(MWカットオフ2000)によって精製を行う。硫酸化度を元素分析によって測定する。全ての化合物はナトリウム塩として得られる(表3)。
所望の硫酸化度は、反応混合物中の硫酸化試薬を規定モル比で使用することによって調節することはできない。なぜなら、その反応性は、試薬の性質、純度及び製造元(origin)、並びに使用される出発物質に依存するからである。しかしながら、過剰の硫酸化試薬を使用することにより、90%を上回る硫酸化度が保証される一方、それよりも少ない硫酸化試薬、好ましくは0.6〜1モル当量を使用することにより、50〜90%の硫酸化度の産物が得られる。
表3は、実施例1bで得られた硫酸化ポリグリセリンを示す。
反応は、攪拌装置を備えたリアクターで実施する。装置全体を減圧下で乾燥し、乾燥アルゴンをフラッシュする。乾燥工程は4回繰り返す。Acrosからのグリシドール(純度>96%)をCaH2とともに一晩攪拌し、45℃及び1mbarで蒸留する。フラクション1(グリシドール使用量は5%)は、ダイマー/トリマーからなる。フラクション2は、所望の純グリシドールである。蒸留されたグリシドールは、冷蔵庫に保管し、乾燥条件下でのみ開く。TMP(2.68g,20mmol)をリアクターに加え、減圧下、60℃で融解する。アルゴン雰囲気下、KOtBu(1MのTHF溶液,6mL)を加え、沈殿物をNMPの添加により溶解する。混合物を120℃に加熱し、2時間攪拌して、t−ブチルアルコールを除去する。グリシドール(100g,1.35mol)を、ドーシングポンプ(dosing pump)で18時間かけて加えた225mLの無水THF(比1:2.5)に溶解する。混合物を90℃に冷却し、200mLの無水DMFで希釈する。ピリジン−SO3複合体(215g,1.35mol)を固体として加え、さらに50mLの無水DMFを加える。24時間の攪拌後、350mLの水を、別のフラスコ中の反応混合物に加え、混合物を2M NaOHで中和し、pHを約9−10とする。蒸発乾固の後、結晶性固体をジエチルエーテル中で攪拌し、NMPを除去する。限外ろ過(水,標準的セルロース膜;MWCO1000)によって精製を行う。高減圧下での蒸発乾固の後、生成物が淡黄色無定形固体として得られる。
実施例2a:追加の機能性リンカーユニットが重合反応の過程で結合した樹状ポリグリセリンコアの合成のための一般的方法:
改良した重合合成を、文献記載の合成(Bioconjugate Chemistry 15, 2004, 162-167)に基づいて実施する。モノマー反応パートナーであるグリシドールの添加が終了した後、脱プロトン化ヒドロキシル基に結合する反応性リンカー種を添加して、重合反応を60〜100℃の温度で継続する。リンカーは、グリシドールに対して1〜50mol%加え、反応は2〜24時間継続する。メタノール/アセトンからの沈殿を繰り返し、高減圧下、50〜80℃で24時間、乾燥することにより、粗製ポリマーを得る。この物質が硫酸化反応で使用される。
実施例1aで得られた樹状ポリグリセリンを、透析(水,MWCO3000)により精製し、減圧濃縮し、高減圧下(0.05mbar)、60℃で24時間、乾燥する。1gのポリグリセリンを、60℃下、無水DMF(20mL)に溶解し、水素化ナトリウム(リンカーで誘導体化される1個のOHあたり2.5モル当量)で処理した後、80℃で18時間、攪拌する。次いで、ブロマイド又はトシレート脱離基あるいはエポキシド反応基(表2)を含む等モル量のリンカーのDMF溶液を80℃で加え、溶液をさらに18時間攪拌する。メタノールの添加により生成物を急冷し、アセトンで沈殿させ、減圧下で乾燥し、メタノールで2日間透析(MWCO1000)した後、高減圧下、60℃で乾燥する。
実施例1で得られた樹状ポリグリセリンを、透析(水,MWCO3000)により精製し、減圧濃縮し、高減圧下(0.05mbar)、60℃で24時間、乾燥する。1gのポリグリセリンを無水DMF(20mL)に溶解する。この溶液に、カルボニルジイミダゾール(CD1;リンカーで誘導体化される1個のOHのあたり5モル当量)を加え、溶液を24時間攪拌する。次いで、アセトンを加えて活性化ポリグリセリンを沈殿させる。残渣を15mLのDMFに溶解し、LP12(1個のOHあたり1.5当量;全体では12当量;表2参照)のDMF溶液を加え、混合物を室温でさらに18時間攪拌する。生成物をアセトンで沈殿させ、減圧下で乾燥し、メタノールで2日間透析(MWCO2000)した後、高減圧下、60℃で乾燥する。
Bioconjugate Chemistry 15, 2004, 162-167及び実施例1bに従って、反応を実施する。透析及び限外ろ過により精製を行う。硫酸化度を元素分析により決定する。全ての化合物はナトリウム塩として得られる(表4)。
表4は、実施例2a−d(保護基の切除前)で得られた、リンカーを有するポリグリセリン硫酸エステルP14〜P27及び実施例2eで得られたP28を示す。
カルボベンジルオキシ(Cbz)−保護化アミノ基,ベンジル及びジベンジル−保護化アミノ基,ベンジルエステル:100mgのポリマーを10mLのメタノール/水(9:1)に溶解し、この溶液に10mgの10%Pd/C触媒を加える。溶液を、水素下、3mbarで24時間、攪拌する。混合物をセライトでろ過し、溶媒を減圧留去し、残渣を繰り返しジクロロメタンで処理し、エバポレーションする。次いで、5mLの水を加える。1M NaOH溶液の添加により、溶液のpHを8に調節する。蒸留水を用いた限外ろ過により、最終産物の精製を行う。
反応を、攪拌装置を備えたリアクターで実施する。装置全体を減圧乾燥し、乾燥アルゴンをフラッシュする。乾燥工程を4回繰り返す。Acrosからのグリシドール(純度>96%)をCaH2とともに一晩攪拌し、45℃及び1mbarで蒸留する。フラクション1(グリシドールの使用量は5%)は、ダイマー/トリマーからなる。フラクション2は、所望の純グリシドールである。蒸留したグリシドールを冷蔵庫に保管し、乾燥条件下でのみ開く。TMP(2.68g,20mmol)をリアクターに加え、減圧下、60℃で融解する。アルゴン雰囲気下、KOtBu(1MのTHF溶液,6mL)を加え、NMPを加えて沈殿物を溶解する。混合物を120℃に加熱し、2時間攪拌して、t−ブチルアルコールを除去する。グリシドール(100g,1.35mol)を、ドーシングポンプで18時間かけて加えた225mLの無水THF(比1:2.5)に溶解する。N−(2,3−エポキシプロピル)フタルイミド(27.4g,135mmol)を、加熱により、シリンジを用いて70分かけて加えた55mLの無水THFに溶解し、一晩攪拌する。混合物を90℃に冷却し、200mLの無水DMFで希釈する。ピリジン−SO3複合体(215g,1.35mol)を固体として加え、さらに50mLの無水DMFを加える。24時間の攪拌後、350mLの水を、別のフラスコ中の反応混合物に加え、混合物を2M NaOHで中和し、pHを約9〜10にする。蒸発乾固の後、結晶性固体をジエチルエーテル中で攪拌し、NMPを除去する。限外ろ過(水,標準的セルロース膜;MWCO1000)により精製を行う。高減圧下での蒸発乾固の後、生成物が淡黄色無定形固体として得られる。
200mgのP14を実施例2dに従ってヒドラジンで脱保護し、これにより透析後に160mgの物質が得られる。この160mgのポリマーP14を1mLの酢酸ナトリウム緩衝液(100mM)に溶解し、イソチオシアネートシアニン色素E3(3当量)を用いて40℃で24時間処理する。水を用いた限外ろ過(標準的セルロース,MWCO3000)により未反応色素を分離して生成物を精製した後、凍結乾燥する。
生成物:140mgの緑色無定形固体(図5参照)。
ポリマーP26(30mg)及び色素E2(5.5mg)を0.6mLのリン酸緩衝生理食塩水(PBS;pH7.4)及び0.2mLのメタノールの混合物に溶解する。この混合物に、CuSO4五水和物(PBS中に12mg/mL)及び0.1mLのアスコルビン酸ナトリウム(PBS中に3.8mg/mL)を含む0.1mLの溶液を加える。この溶液を、激しく振盪しながら、遮光下、40℃で5日間、インキュベーションする。蒸留水を用いた限外ろ過(標準的セルロース,MWCO4000−6000)の後、サイズ排除クロマトグラフィー(Sephadex G−50)を実施することにより、精製を実施する。凍結乾燥により、緑色固体(19mg)が得られる(図5参照)。
ポリマーPI7(30mg)及び色素E5(5mg)を、0.4mLのリン酸緩衝生理食塩水(PBS;pH7.4)及び0.4mLのエタノールの混合物に溶解する。この混合物に、0.1mLのCuSO4五水和物溶液(PBS中に12mg/mL)及び0.1mLのアスコルビン酸ナトリウム(PBS中に3.8mg/mL)を加える。この溶液を、激しく振盪しながら、遮光下、40℃で5日間、インキュベーションする。メタノールを用いた限外ろ過(標準的セルロース,MWCO4000−6000)及び分取HPLC(RP18,水)を組み合わせて精製を実施することにより、生成物は、直ちに溶離したピークとして得られる。凍結乾燥により、18mgのインドカルボシアニン(ICC)複合体(紫がかった赤の凍結乾燥物)が得られた(図5参照)。実施例3b及び3cの反応工程を、ポリマー/色素の組み合わせ(例えば、P17/E1,P17/E2,P26/E1,P26/E5,P27/E1,P27/E2,P27/E5)に基づいて、複合体の調製にさらに適用することができる。
200mgのP28を、DMF/水が9:1である混合物(2mL)に溶解する。この混合物に86mgのNHS−エステル色素E7(4モル当量)を加えた後、室温で48時間攪拌する。蒸発乾固の後、固体残渣の精製を実施例3cに記載したように実施した。凍結乾燥により、185mgのインドジカルボシアニン複合体(青色凍結乾燥物)が得られた。
実施例3dの反応工程を、P28と、色素E8及び診断及び治療エフェクター分子のその他のNHSエステルとの複合体の調製にさらに適用することができる(実施例4bも参照)。
プロパギルシアニン色素(E1)を、フェニルボロン酸前駆体を用いた鈴木反応によるIR−820の誘導体化に関するLee等の方法(J. Org. Chem. 73 : 723 (2008))を改良して合成する。本発明では、IR−820を4−カルボキシエチルフェニルボロン酸と反応させ、対応する鈴木−カップリング生成物を得た。アミド形成の公知の工程により、プロパギルアミン及びHBTUを用いて、E1への変換を実施した。この工程において、芳香環がスルホン酸基で高度に置換された、IR−820の新規アナログが得られ、その後、鈴木カップリング及びプロパギルアミンを用いたアミド化が実施される(表1中のE2−E4参照)。
合成により、16個の硫酸基を含む所定のデンドロンとの色素複合体が生じる。実施例3bに記載したように、Elを[G3.0]−アジド(Wyszogrodzka等, Chemistry 14: 9292 (2008)中の化合物14)と反応させた。Wyszogrodzka等が記載するように、アセタールの脱保護を実施し、その後、実施例2dに従って硫酸化を実施する。HPLC分析により、複合体が収率25%で得られる。
ポリマーP17(30mg)及びプロパギル−DOTA E13(3mg)を0.4mLのリン酸緩衝生理食塩水(PBS;pH7.4)及び0.4mLのエタノールの混合物に溶解する。この混合物に、CuSO4五水和物(PBS中に12mg/mL)及び0.1mLのアスコルビン酸ナトリウム(PBS中に3.8mg/mL)を含む0.1mLの溶液を加える。この溶液を、激しく振盪しながら、遮光下、40℃で5日間、インキュベーションする。水を用いた限外ろ過(標準的セルロース,MWCO4000−6000)により精製を実施し、これにより20mgの複合体が白色固体として得られる(図6)。
実施例4aに記載したように、反応を実施する。
さらに、例えば、P28及びE12を用いて実施例3dの手順に従い、アミノで修飾された硫酸化ポリグリセリンをDOTA−NHSエステルと反応させることにより、DOTAキレート剤複合体を得ることができる。
多数のアジド基(15%)が、高いエフェクター/ポリマー比を得るために必要となる。ポリマーP27(50mg)及び複合体E16(36mg)を0.8mLのリン酸緩衝生理食塩水(PBS;pH8.5)及び0.4mLのメタノールの混合物に溶解する。この混合物に、CuSO4五水和物(PBS中に12mg/mL)及び0.2mLのアスコルビン酸ナトリウム(PBS中に3.8mg/mL)を含む0.2mLの溶液を加える。この溶液を、25℃で3日間、激しい振盪下に維持する。水を用いた限外ろ過(標準的セルロース,MWCO4000−6000)により精製を実施し、これにより52mgの複合体が白色固体として得られる。ICP−MSによる金属分析により、ポリマー1モルあたりガドリニウム複合体5モルというガドリニウム含量が生じる(図6)
ポリマーP16を用いて、硫酸化 ポリグリセリンのチオール−修飾を実施する。50mgのポリマーP16を、1mLの水/トリフルオロ酢酸中で2時間攪拌した後、エタノールで沈殿させ、次いで、高減圧下で乾燥する。これを0.5mLの10mMリン酸緩衝液(pH7.0)に溶解し、10モル当量の2−イミノチオランを用いて室温で1時間処理する。この混合物に、0.67mL(10モル当量)のマレイミド−ピロフェオホルビド(E16)のDMF溶液(濃縮,20mg/mL)を加える。反応混合物を、45℃で24時間、激しく振盪する。混合物をエバポレーションし、残渣を数回、ジクロロメタンで洗浄/遠心することにより、生成物を分離する。分取HPLC(RP18,水/メタノール)により最終的な精製を実施し、これにより20mgの複合体が得られる(図7)。
200mgのP14を、実施例2eに従って、水素化ホウ素ナトリウム/酢酸で脱保護し、これにより透析後に160mgの物質が得られる。この160mgのポリマーP14を、DMF及び50mMリン酸緩衝液(pH8.0)の1mLの混合物(9:1)に溶解した。この混合物に、E25(15当量)を加えた後、40℃で24時間、激しく攪拌する。混合物をエバポレーションし、残渣を数回、ジクロロメタンで洗浄/遠心することにより、生成物を分離する。1H−NMRの芳香族シグナルにより、ポリマー1個あたり平均で約1.8のクロランブシル分子の複合化が判明する(図7)。
同様に、パクリタキセルスクシネートNHSエステル(E28)との反応により、ポリマー1個あたり平均で1.2個のパクリタキセル分子が得られる。
2−イミノチオランを用いたポリグリセリン硫酸エステルのチオール修飾を、実施例6aに記載したように実施する。チオール修飾された硫酸化ポリグリセリン(50mg/mL)の溶液に、10モル当量のマレイミドパクリタキセル(E20;10mg/mLのDMF溶液)を加える。反応混合物を、25℃で24時間、激しく振盪する。混合物をエバポレーションし、残渣を数回、ジクロロメタンで洗浄/遠心することにより、生成物を分離する。1H−NMRの芳香族シグナルにより、ポリマー1個あたり平均で約2個のパクリタキセル分子の複合化が判明する(図7)。
さらに、P28及びE28を用いて実施例3dの手順に従い、アミノ−修飾された硫酸化ポリグリセリンをパクリタキセル−NHSエステルと反応させることにより、パクリタキセル複合体が得られ、これにより、ポリマー1個あたり平均で1個のパクリタキセル分子の複合化が生じる。
実施例6aに記載したように、2−イミノチオランを用いたポリグリセリン硫酸エステルのチオール−修飾を実施する。Warnecke等の方法(Bioconjugate Chem. 2004, 15(6), 1348-1359)に従って、カルボプラチンマレイミド(E31)を合成する。チオール−修飾された硫酸化ポリグリセリン(50mg/mL)の溶液に、15モル当量のE31(20mg/mLのエタノール溶液)を加え、25℃で24時間、反応混合物を激しく振盪する。混合物をエバポレーションし、残渣を数回、ジクロロメタン及びn−ブタノールで洗浄/遠心することにより、生成物を分離する。ICP−MSにより白金含量を測定する(金属:ポリマー比は2:1)。
VEGF siRNAを、J. Contr. Release 2006, 1 16, 123-129に従って得た。

上皮ヒト肺癌細胞株を、次の通り、常法に従って増殖させた:DEMEM培養液に、10%ウシ胎児血清,2%グルタミン及びペニシリン/ストレプトマイシン(全てPAN Biotech社製)を加えた。細胞を、1×105細胞/mLで培養液に播種し、5%CO2下、37℃で培養し、1週間に2回、1:5に分割した。in vitro蛍光イメージングのために、細胞を、2×105細胞/mLで24ウェル培養プレート(Sigma社製)のカバーガラスに播種し、37℃で24時間、培養した。その後、細胞を、インドカルボシアニン(ICC)プロパギルエステル(化合物E5)と複合化した様々な分子量を有する10-6M ポリグリセリンを含有する培養液、又は対照として10-6M ICC−トリグリセロール複合体(Angew Chem Int Ed Engl. 48: 7540, 2009)を含有する培養液中、37℃で1時間、培養した。次いで、細胞を、冷アセトンで固定した。4,6−ジアミジノ−2−フェニルインドール(DAPI,Abeam)を用いて核を対比染色した。イメージ獲得を、Leica DMRB顕微鏡(Leica)を用いて行った。イメージは、デジタルカメラ(Spot32,Diagnostic Instruments)で得た。
低分子量(最大20kDaまで)のポリグリセリン又はICC−トリグリセロール複合体の細胞取り込みは観察されなかった。高分子量(120又は208kDa)のポリグリセリンの細胞内局在が判明した。結果を図8に示す。
ヒト上皮肺癌細胞株を、常法に従って増殖し、実施例7に記載したように、24ウェル培養プレート中で処理した。細胞を、10-6M ヘパリン−ICC複合体若しくは硫酸化ポリグリセリン−ICC複合体(実施例3c)を含有する培養液、又は対照として10-6M ICC−トリグリセロール複合体(Angew Chem Int Ed Engl. 48: 7540, 2009)を含有する培養液を用いて、37℃で4時間、培養した。その後、細胞を冷アセトンで固定した。4,6−ジアミジノ−2−フェニルインドール(DAPI,Abeam)を用いて核を対比染色した。イメージ獲得を、Leica DMRB顕微鏡(Leica)を用いて実施した。イメージは、デジタルカメラ(Spot32,Diagnostic Instruments)で得た。ヘパリン又はグリセロール−ICC複合体の細胞取り込みは観察されなかったが、硫酸化ポリグリセリン−ICCはA549細胞の細胞質に存在することが判明した。結果を図9に示す。
Ficoll−Hypaqueでの分画遠心分離により、ヒト血液の単核球フラクション由来の細胞を分離し、次いで、10%ウシ胎児血清を含有するRPMI(PAN Biotech)で洗浄した。2×105細胞/mLの細胞を、24ウェルプレート中、RPMI培養液、又は10-6M 硫酸化ポリグリセリン−ICC複合体(実施例3c)を含有する培養液、又は対照として10-6M ICC−トリグリセロール複合体(Angew Chem Int Ed Engl. 48: 7540, 2009)を含有する培養液で、4時間、培養した。その後、細胞をPBSで洗浄し、200μL/ウェルのアキュターゼ(PAA)で剥がし、PBSで2回洗浄した。細胞を、4℃で10分間、500μLの3%パラホルムアルデヒドで固定し、2mLのPBSで停止させ、4℃で10分間、遠心(250g)した。上清を除去し、細胞を200μLのPBS(0.5%ウシ血清アルブミン(Roth)含有)に懸濁した。固定した細胞を、FACS Calibur分析装置(Becton−Dickinson)で分析するまで、4℃で保存した。細胞のサイズ及び粒度(前方散乱/側方散乱)の分析により、単球又はリンパ球を同定するとともに、硫酸化ポリグリセリン−ICC複合体の取り込みを定量した。単球において、リンパ球よりも約100倍(hundertfold)大きな量の硫酸化ポリグリセリンが検出された。結果を図10に示す。
CASKI細胞を、次の通り、培養した:10%ウシ胎児血清(FCS),2%グルタミン及びペニシリン/ストレプトマイシン(全てPAN Biotech社製)を加えたDEMEM培養液を使用した。細胞を、1×105細胞/mLで培養液に播種し、5%CO2下、37℃で培養し、1週間に2回、1:5に分割した。様々な硫酸化度(30%,70%,86%又は92%の硫酸化)又は様々な分子量のポリグリセリンコア(2600又は7500Da)を有する硫酸化ポリグリセリンをCASKI細胞に10-8Mの濃度で48時間適用した。培養液のみでの培養を対照として用いた。培養上清を採取し、遠心し、−20℃で保管した。市販のELISAキット(eBioscience Inc)を用いて、培養上清中のTGFβ1含量を検出した。結果を図11に示す。
BIACORE X装置(Biacore AB,Uppsala,Sweden)を用いて、25℃で実験を行った。リガンド固定には、10mM HEPES(pH7.4),150mM NaCl,3mM EDTA及び0.005%界面活性剤P20からなるHBS−EP緩衝液(Biacore AB)を使用した。アッセイには、HBS−EP緩衝液及びランニングバッファーを使用した。κB DNA配列モチーフ(下線)のビオチン化一本鎖(5’−ビオチン−AGTTGAGGGGACTTTCCCAGGC−3’(フォワード)及び5’−ビオチン−GCCTGGGAAAGTCCCCTCAACT−3’(リバース))を、metabion international AG(Martinsried,Germany)から購入した。30μLの両一本鎖溶液(各濃度は100μM)を混合し、80℃で5分間加熱し、完全なハイブリダイゼーションが可能となるように、ゆっくりと室温まで冷却した。次いで、60μLのHBS−EP緩衝液をサンプルに加え、ビオチン化プローブを、ストレプトアビジン官能化センサーチップSA(Biacore AB)に固定した。参照用として、同じチップの2番目のレーンを空のままにした。性能を高めるために、1M NaClの50mM NaOH溶液を固定開始前に3回連続して1分間インジェクションすることにより、センサーチップを初期調整した。チップにκB DNA配列モチーフをローディングしたところ、ベースラインがシフトして1300レゾナンスユニット(resonance unit)(RU)を示した。固定工程の後、ランニングバッファーで数回洗浄し、チップ表面を平衡化した。組換えNF−κB p50タンパク質は、Active Motif(Carlsbad,CA,USA)から購入したものである。NF−κBタンパク質の最終的な作用濃度(working concentration)は、約28.6nMであった(モル濃度は、NF−κBモノマーの分子量(50,000Da)に基づく)。ローディングの前に、各サンプルを、室温で18分間、最終的な阻害濃度(inhibitor concentration)を0nM,0.1nM,1nM,10nM,100nM,1μM,10μM又は100μMをして、それぞれの硫酸化ポリグリセリン化合物とインキュベーションした(表5参照)。サンプルを、参照及びリガンド(κB DNAオリゴヌクレオチド)レーンに、流速20μL/分でインジェクションした。各測定サイクルは、105秒間のサンプルインジェクション(会合段階),180秒間維持される解離段階及び1M NaClによるフローシステムの洗浄から構成した。データ評価のために、リガンド(κB DNA配列)レーンのデータから、参照レーンのデータを差し引いた。サンプルインジェクションの応答を、インジェクションの開始時(0秒)及びat the end of the 解離段階の終了時(285秒)にセットされたリポートポイント間で抽出した。最終応答値を、曲線作成に使用した。各データ点は、2つの測定値の平均値(±SEM)を表す。結合曲線を図12に示す。IC50値を表5に示す。結合アフィニティーは、IC50値の減少によって示される硫酸化度及び分子量の増加に伴って増加する。リンカー及びエフェクター分子は、結合アフィニティーを阻害しない。デンドロンにアセンブリされた硫酸エステルの数が16個だけである場合、十分な結合アフィニティーを示さない(G3.0−デンドロン/E1)。
表5は、硫酸化ポリグリセリン、リンカーを有する硫酸化ポリグリセリン、及びエフェクター分子との複合体(実施例1,2,3)のIC50値を示す。
肺腫瘍細胞の細胞増殖に対する硫酸化ポリグリセリンの潜在的な効果を試験するために、ヒトA549細胞株を使用した。A549細胞株を、常法に従って、次の通り増殖させた。10%ウシ胎児血清(FCS),2%グルタミン及びペニシリン/ストレプトマイシン(すべてPAN Biotech社製)を添加したDEMEM培養液を使用した。細胞を、1×105細胞/mLで培養液に播種し、5%CO2下、37℃で培養し、1週間に2回、1:5に分割した。
培養2日後、10μLのMTT(5mg/mLのPBS溶液,Sigma社製)を各ウェルに加え、プレートを4時間インキュベーションした。生じたホルマザン生成物を酸イソプロパノールで溶解し、波長570nmでの吸光度(Ex570)をマイクロプレート分光光度計(Anthos htll,Microsystems)で測定した。
硫酸化ポリグリセリン(化合物P3)を、10-5M〜10-9Mの濃度で、A549細胞に48時間適用した。培養液単独での培養を対照として使用した。
結果を図13に示す。
硫酸化ポリグリセリンが、in vivoにおける腫瘍増殖の抑制に有効であるか否かを試験するために、よく知られている肺癌ヌードマウスモデルを使用した。ヒト非小細胞肺癌細胞A549(0.7×107細胞/マウス)を、無胸腺雄NMPJ nu/nu マウス(Taconic Europe)の脇腹に皮下注射した。1日あたり100μLのPBS又は30mg/kg体重の硫酸化ポリグリセリン(化合物P3)を含む100μLs.c.中の投与を、細胞移植後12−16,19−23及び26−30日目に実施した。腫瘍容積を細胞移植後12〜44日の間の10の時点で測定した。腫瘍サイズをキャリパーで測定し、容積を次式に従って計算した:容積(cm3)_1/2(Z_W2)、L及びWは移植腫瘍の長さ(cm)及び幅(cm)である。結果を図14に示す。さらに、薬物処理による毒性及び有害事象の兆候は、マウス処理の間を通じて、観察されなかった。
硫酸化ポリグリセリンが関節リウマチの抑制に有効であるか否かを試験するために、実験用コラーゲン誘導性関節リウマチ(CIA)をラットに誘導した。CIAは、ウシII型コラーゲン及び不完全フロイントアジュバント(DIFCO Laboratories)のエマルションで雌Lewisラットを免疫することにより誘導した。肢の炎症を、2週間後に、肢の容積の増加により評価した。ラットを、疾患の臨床兆候について毎日モニタリングし、0〜3の疾患スコアを決定した。in vivo処置について、硫酸化ポリグリセリン(化合物P3)30mg/kg体重のPBS溶液200μLを、免疫後14日目から16日目まで毎日、対照及びCIAラットに皮下注射により投与した。骨及び軟骨の病変及び炎症性浸潤を、膝関節のパラフィン切片の組織学的変化に基づいて評価した。肥満細胞をトルイジンブルー(Sigma)の0.5N HCL(Roth)溶液で一晩染色し、滑膜における数を計測した。結果を図15に示す。さらに、毒性及び有害事象の兆候は、マウス処理の間を通じて、観察されなかった。
硫酸化ポリグリセリンが実験用多発性硬化症の抑制に有効であるか否かを試験するために、実験用自己免疫性脳炎(EAE)をマウスに誘導した。EAEは、PLP100μgのPBS溶液100μL及びCFA100μLをマウスに皮下注射して免疫することにより、雌SJLマウス(8〜12週齢)に誘導した。CFAは、不完全フロイントアジュバント(DIFCO Laboratories)を8mg/mLの結核菌H37RA(乾燥物;DIFCO Laboratories)と混合することにより調製した。免疫の時点及びその2日後に、マウスに、百日咳毒素(List Biological Laboratories)200ngのPBS溶液100μLを静脈内注射した。疾患の臨床兆候についてマウスを毎日モニタリングし、次のようなEAE重症度に基づいて0〜5の疾患スコアを決定した:0は疾患なし;1は尾を引きずる;2は後肢が弱化;3は後肢が麻痺;4は後肢及び前肢が麻痺;5は病的状態及び死亡。in vivo処置において、硫酸化ポリグリセリン(化合物P3)を、2日目から20日目まで毎日、皮下注射によりPLP免疫化マウスに投与した。疾患の臨床兆候について、マウスを毎日モニタリングした。表6は、1日投与量30mg/kgで皮下注射された硫酸化ポリグリセリンの治療効果を示す。対照動物は、免疫後10日目及び20日目に、それぞれ3.1及び2.3という臨床スコアを示したのに対して、1日投与量30mg/kg体重の硫酸化ポリグリセリンでは、臨床スコアがそれぞれ1.7及び1.1まで減少した。さらに、薬物処置による毒性及び有害事象の兆候は、マウス処置の間を通じて観察されなかった。
表6は、EAEマウスモデルの臨床スコアに対する、硫酸化ポリグリセリン(P3)の効果を示す。
硫酸化ポリグリセリンを、雌NMRIマウスの亜急性敗血症モデルで試験した。敗血症は、単回投与量0.2mg/kgでのLPSの単回腹腔内注射により誘導した。陰性対照群には、生理食塩水を単回注射した。LPS(Sigma Aldrich)投与の30分前に、硫酸化ポリグリセリン(化合物P3)を単回投与量15mg/kgでの皮下注射により投与した。非致死性敗血症の指標として、補体タンパク質C5a(Alpco Diagnostics)の血清レベルを測定した。測定は、LPS投与の2時間後又は6時間後に実施した。LPSは、血清C5aの亢進を誘導し、陰性対照群と比較して245%となった。単回投与量15mg/kgの硫酸化ポリグリセリンは、LPS媒介性亢進の強力な阻害を誘導し、陰性対照群のレベルの105%となった。この阻害は、LPS投与の6時間後も顕著であった。
表7は、マウスに誘導されたLPS誘導敗血症に対する、硫酸化ポリグリセリンの効果を示す。
エフェクター複合体P26/E2(実施例3b)を0.9%NaCl(濃度:1mg/mL)に溶解し、コラーゲン誘導性関節リウマチのラット(実施例14に記載した動物モデル;投与量:2mg/kg)の尾静脈に注射した。レーザーユニット(励起波長760nm)及びロングパスフィルター(観察>780nm)を有するCCDカメラを備える平面蛍光イメージングセットアップを、J. Biomed. Optics 10, 41205 (2005)に従って使用した。麻酔ラットの蛍光イメージングを、インジェクション後10分,30分,1時間,6時間及び24時間に実施した。結果を図16に示す。図16に示すように、関節炎の関節における迅速かつ高い取り込み及び蛍光コントラスト(10分)が最大24時間持続する(データは示さない)一方、健常な関節は、蛍光の亢進を示さない。矢印は、疾患進行度(疾患スコア1,2及び3)が増加する関節炎の関節であり、蛍光コントラストはスコアとともに増加し、このことは、疾患活性をモニタリングする複合体の能力を示す。
450μgのTNCBを腹部皮膚へ局所適用することにより、C57B1/6マウス(雌,8週齢)をTNCBに感作した。5日後、45μgのTNCBを右耳へ局所適用することにより、マウスをチャレンジした。文献記載の方法に従って、エフェクター複合体P17/E13(実施例4a)をCu−64で標識化した。(CuCl2溶液の形態でのCu−64の生成: Appl. Radiat Isot. 2007, 65, 1 1 15)。標識化は、0.1mgの複合体を用いて、酢酸緩衝液(pH5)中、37℃で1時間実施した(100MBq)。放射化学的純度をHPLC及びラジオTLCにより測定した。放射化学的収率は90%であった。SEC精製の後、Cu−64標識化HSPG DOTA複合体をリン酸緩衝液(pH7.4)の溶液として得た。100μCiをマウスの尾静脈に注射した。臨床PETスキャナーを用いて、高解像10分PETイメージングを繰り返し得た。麻酔マウスのイメージは、Cu−64標識HSPG DOTA複合体のインジェクション後30分に得た。結果を図17に示す。図17は、炎症領域(マウス耳における炎症組織)における迅速で高い取り込みを矢印で示す。
細胞増殖抑制薬の細胞毒性に対する硫酸化ポリグリセリン複合体の潜在的効果を試験するために、ヒトA549細胞株を使用した。A549細胞を、常法に従って、次のように増殖した。10%ウシ胎児血清(FCS),2%グルタミン及びペニシリン/ストレプトマイシン(全てPAN Biotech社製)を添加したDEMEM培養液を使用した。細胞を、1×105細胞/mLで培養液に播種し、5%CO2下、37℃で培養し、1週間に2回、1:5に分割した。
細胞増殖の分析を、24ウェルプレートで培養した細胞を用いて実施した。2×105細胞/mLを、漸増する濃度の試験物質を含有する1mLの培養液中でインキュベーションした。 培養の24時間後、試験物質を含有する培養液を除去し、標準的な培養液で置換した。培養のさらに24時間後、アポトーシスのプロセスのパラメーターである細胞数、生存率及び細胞直径を細胞計数器及び分析システム(CASY(R),Scharfe Systems)で分析した。さらに、細胞代謝活性試験であるMTTアッセイ(3−(4,5−ジメチルチアゾール−2−イル)−2,5−ジフェニルテトラゾリウムブロミドの細胞還元)により、薬物の影響をin vitroで評価した。すなわち、1ウェルあたり1×104個の細胞を、漸増する濃度の試験物質(実施例6bのパクリタキセル複合体)を含有する100μLの培養液中、96ウェルプレートにプレーティングする。培養2日後、10μLのMTT(5mg/mLのPBS溶液,Sigma社製)を各ウェルに加え、プレートを4時間インキュベーションした。生じたホルマザン生成物を酸イソプロパノールで溶解し、波長570nmでの吸光度(Ex570)をマイクロプレート分光光度計(Anthos htll,Microsystems)で測定した。パクリタキセル(タキソール)を、10-7Mの濃度でA549細胞に適用し、試験物質を濃度10-7〜10-10Mで適用した。培養液単独での培養を対照として使用した。
結果を図18に示す。図18は、10-7Mに相当する濃度での優れた効果、及び1/100よりも低い濃度での細胞毒性を示す。
肺癌細胞株A549を、実施例11に記載したように増殖させた。VEGF検出用に上清を回収するために、細胞を、1×106細胞/mLで24ウェル培養プレートに播種し、24時間培養した。その後、硫酸化ポリグリセリン(複合体P16/E33,実施例6e)に結合したVEGF−siRNA又はVEGF−siRNAを用いて、その他のトランスフェクション試薬を添加することなく、細胞をインキュベーションした。トランスフェクションの4時間後、培養液を新しい培養液(10%FCS含有)で置換した。さらに48時間後、培養上清をELISA用に回収した。上清中のVEGF含量を、市販のELISAキット(Quantikine,R&D Systems)を用いて検出した。結果を図19に示し、図19は、試験物質とインキュベーションした後のA549肺癌細胞株のVEGF産生を示す。VEGFタンパク質を、48時間調整した細胞培養液中、ELISAにより測定した。各バーは、3回の独立した試験で得られた3つの測定値の平均値±SEMである。
シアニン色素と複合化した硫酸化ポリグリセリン(化合物P17/E1)の0.9%NaCl(生理食塩水)溶液を濃度0.1μMで調製した。暗所中、室温で様々な時間(0,1,2.5,3,4,24時間)保管した溶液の蛍光強度を測定した(励起760nm,検出>780nm)。結果を図21に示し、図21は、溶液中の凝集による蛍光の定常減少を示す。蒸留水又はメタノールでは、そのような減少は観察されなかった。蒸留水を用いて透析し、トレハロース(硫酸化ポリグリセリン1mgあたり10mg)を添加し、凍結乾燥して、固形物質を得た。蒸留水中に再懸濁し、初期レベルの蛍光シグナルを得た。
Claims (24)
- 硫酸化ポリグリセリンと、前記硫酸化ポリグリセリンに共有結合している治療又は診断エフェクター分子とを含む、医薬組成物。
- 式:P(OSO3 -M+)n(L−G−E)m
[式中、
Pは、n個のヒドロキシル基が硫酸基OSO3 -M+で置換されたポリオール高分子であり、
Mは、アニオン性硫酸基に対するカチオン性無機又は有機対イオンであり、
Eは、治療又は診断エフェクター分子であり、
Lは、P及びE間のリンカー又はスペーサーであり、
Gは、L及びE間の共有結合のための反応基であり、
mは、1−100の数である。]
で表される、請求項1に記載の医薬組成物。 - 前記硫酸化ポリグリセリン及び治療又は診断エフェクター分子の活性化細胞又は増殖細胞への細胞内取り込みにより、並びに、前記細胞におけるNF−κB及び/又はAP−1の阻害及び/又はTGF−β合成の阻害により、疾患を治療するための、請求項1又は2に記載の医薬組成物。
- 硫酸化ポリグリセリンと、前記硫酸化ポリグリセリンに共有結合している治療又は診断エフェクター分子とを含む、複合体。
- 式:P(OSO3 -M+)n(L−G−E)m

- 前記エフェクター分子の割合が前記複合体の50重量%未満であり、前記複合体の水への溶解度が100mg/mLよりも大きい、請求項4〜6のいずれか1項に記載の複合体。
- 活性化細胞又は増殖細胞への細胞内取り込みにより、並びに、前記細胞におけるNF−κB及び/又はAP−1の阻害及び/又はTGF−β合成の阻害により、疾患を治療するための、硫酸化ポリグリセリンと、前記硫酸化ポリグリセリンに共有結合している治療又は診断エフェクター分子とを含む、請求項4〜7のいずれか1項に記載の複合体。
- 癌、炎症、自己免疫疾患及び線維症を含む群から選択される疾患を治療するための、硫酸化ポリグリセリンと、前記硫酸化ポリグリセリンに共有結合している治療又は診断エフェクター分子とを含む、請求項4〜8のいずれか1項に記載の複合体。
- 1投与あたりの投与量が1mg/kg〜1000mg/kgである複数回処置が実施される、請求項3又は4に記載の、疾患を治療するための、硫酸化ポリグリセリンと、前記硫酸化ポリグリセリンに共有結合している治療又は診断エフェクター分子とを含む、請求項4〜9のいずれか1項に記載の複合体。
- a)1〜1000個のOH基を有するポリヒドロキシ化合物である多機能性出発分子上に、式:(RO−CH2)2CH−ORで表されるグリセロールの繰り返し単位を有する高分子ポリグリセリンPであって、RがH又はさらに別のグリセロール単位であり、コアの分岐度が0〜67%であり、平均分子量が500〜1,000,000g/molである前記高分子ポリグリセリンP、
b)前記グリセロール単位の複数のOH基の、−OSO3H又は−OSO3 -M+による置換であって、−OSO3H又は−OSO3 -M+基の好ましい数が10よりも大きく、得られる硫酸化度Xが30〜100%であり、M+がカチオン性無機又は有機対イオンである前記置換、
c)1,000〜5,000,000g/molという、得られる硫酸化ポリグリセリンの平均分子量、
d)少なくとも1個から最大100−X%までのOH基に結合した官能基Gを有するリンカー単位Lであって、前記官能基が、さらに別の治療又は診断エフェクター分子と複合化することができ、Xが硫酸化度である前記リンカー単位L、
e)1から可能な最大数までの前記官能基に共有結合した診断及び/又は治療エフェクター分子であって、前記診断エフェクター分子が、蛍光色素及び放射性又は常磁性金属のキレート剤からなる群より選択され、前記治療エフェクター分子が、細胞増殖抑制剤、抗血管新生薬、光増感剤及びsiRNAからなる群より選択される前記診断及び/又は治療エフェクター分子、
を含む、請求項4〜10のいずれか1項に記載の複合体。 - Lが、1又は2以上の非連続メチレン基がO、S、NH、C(O)NH、C(O)、SO2、SO、アリール、エテン又はエチンから選択される基で置換されていてもよい分岐鎖状又は直鎖状のC1-20アルキル基であり、Gが、−OH、−OSO3H、−OSO3 -、−NH2、−N3、−COOH、−SH、−SO3 -及び−C≡Cを含む群より選択される、式:P(OSO3 -M+)n(L−G−E)mで表される請求項5〜11のいずれか1項に記載の複合体。
- 活性化細胞又は増殖細胞への細胞内取り込みにより、並びに、前記細胞におけるNF−κB及び/又はAP−1の阻害、及び/又はTGF−β合成の阻害により、疾患を治療するための、式:P(OSO3 -M+)n(L−G)m

- 癌、炎症、自己免疫疾患及び線維症を含む群より選択される疾患を治療するための、請求項19又は20に記載の無水製剤。
- 1投与あたりの投与量が1mg/kg〜1000mg/kgである複数回処置が実施される、請求項19又は20に記載の無水製剤。
- 緩衝塩、並びに/又は、スクロース、マンノース及びトレハロースからなる群より選択される少なくとも1つの抗凍結剤を含有する凍結乾燥物を含む、請求項19又は20に記載の無水製剤。
