(i)第1のHBVウイルスに由来するポリメラーゼタンパク質の少なくとも450個のアミノ酸残基を含んでなるポリメラーゼ部分;
(ii)第2のHBVウイルスに由来するコアタンパク質の少なくとも100個のアミノ酸残基を含んでなるコア部分;および
(iii)第3のHBVウイルスに由来するHBsAgタンパク質の連続した15〜100個のアミノ酸残基の1以上の免疫原性ドメインを含んでなるenv分;または
該ポリメラーゼ部分、該コア部分、該env部分、該ポリメラーゼ部分をコードする該核酸分子、該コア部分をコードする該核酸分子および/または該env部分をコードする該核酸分子の任意の組合せ
からなる群から選択される、免疫原性組成物を提供する。
本出願全体を通じて本明細書において用いられるように、「1つの(a)」および「1つの(an)」とは、特に断りがない限り、それらの用語が「少なくとも1つ」、「少なくとも最初」、「1つ以上」または「複数」の参照される化合物または工程を意味するという意味で用いられる。例えば、「1つの細胞(a cell)」という用語は、複数の細胞を含み、その混合物を含む。
一実施形態によれば、本発明の組成物に含まれるまたは本発明の組成物によってコードされるポリメラーゼ部分は、対応する天然HBVポリメラーゼと比べて改変されている。

あるいはまた、上記の組成物と組み合わせて、本発明はまた、第2のHBVに由来するコア部分を含んでなる組成物を提供する。HBVウイルスに関連して本明細書において記載されるように、本発明において使用されるコア部分は、ポリメラーゼ部分の起源であるHBVウイルスと同じまたは異なるHBVを起源とする。好ましくは、コア部分およびポリメラーゼ部分は両方とも遺伝子型D HBVを起源とし、Y07587 HBV分離株あるいは、中国で多いHBV遺伝子型(例えば、遺伝子型BまたはC)のものが特に好ましい。

(iii)E1領域の代わり挿入された、CMVプロモーターの制御下に置かれた核酸分子であり、配列番号7、8もしくは9に、または494番のAsp残基のHis残基への置換および672番のGlu残基のHis残基への置換を含む配列番号7に示されるアミノ酸配列を含んでなるポリメラーゼ部分をコードする核酸分子を含んでなるE1欠損型Adベクター。
(iv)E1領域の代わり挿入された、CMVプロモーターの制御下に置かれた、配列番号18、19もしくは20に、または残基1からはじまり残基251で終わる配列番号20の部分(core−env1−env2)に、または残基1で始まり残基221で終わる配列番号20の部分(core−env1)に示されるアミノ酸配列を含んでなるコア部分およびenv部分をコードする核酸分子を含んでなるE1欠損型Adベクター。
(a)本発明のウイルスベクターを好適な細胞系統に導入する工程、
(b)該感染性ウイルス粒子の産生を可能にする好適な条件下で該細胞系統を培養する工程、
(c)産生された感染性ウイルス粒子を該細胞系統の培養物から回収する工程、および
(d)所望により、該感染性ウイルス粒子回収物を精製する工程。
・1Mサッカロース、150mM NaCl、1mM MgCl2、54mg/l Tweet 80、10mM Tris pH8.5(特に活性剤がアデノウイルスベクターである場合)、および
・10mg/mlマンニトール、1mg/ml HSA、20mM Tris、pH7.2および150mM NaCl、
・生理食塩水
を含む。
下記の構築を、Maniatis et al. (1989, Laboratory Manual, Cold Spring Harbor Laboratory Press, Cold Spring Harbor NY)に詳説されている一般的遺伝子操作および分子クローニング技術に従い、または市販のキットを用いる場合には製造者の推奨に従って行う。PCR増幅技術は当業者に公知である(例えば、Innis, Gelfand, Sninsky and White, Academic Pressにより出版されているPCR protocols -A guide to methods and applications, 1990参照)。アンピシリン耐性遺伝子を担持する組換えプラスミドを、100μg/mlの抗生物質を添加した寒天培地または液体培地の大腸菌C600(Stratagene)で複製する。組換えワクシニアウイルスの構築は、上記に引用されている文献およびMackett et al. (1982, Proc. Natl. Acad. Sci. USA 79, 7415-7419)およびMackett et al. (1984, J. Virol. 49, 857-864)中の当技術分野の従来技術に従って行う。組換えワクシニアウイルスの選択を容易にするために、大腸菌の選択遺伝子gpt(キサンチングアニンホスホリボシルトランスフェラーゼ)(Falkner and Moss, 1988, J. Virol. 62, 1849-1854)を用いる。
1.1.1.選択された抗原およびHBV配列株
以下に具体的に示すベクターは、ポリメラーゼ、コアポリペプチドおよびエンベロープタンパク質の免疫原性ドメインを発現するように操作されたものであった。このベクターはHBV株Y07587に由来し、その配列は国際データベース(Genbank Y07587)および刊行物に記載されている。このベクターは血清型aywの遺伝子型Dのウイルスである。コアポリペプチドは野生型(aa1〜183)またはアミノ酸77〜8を欠くコアポリペプチド(すなわち、core*で示されるアミノ酸1〜76および85〜183を含むコア)またはC末端切断型ポリペプチド(1〜148)またはさらにアミノ酸77〜84を欠くC末端切断型コア(1〜148)(すなわち、core*tで示されるアミノ酸1〜76および85〜148を含むコア)のいずれかである。
改変型HBVポリメラーゼポリペプチドをコードするヌクレオチド配列は、合成オリゴヌクレオチドおよびPCR産物を用い、Geneart社によって合成された。改変型HBVポリメラーゼは、HBV Y07587のポリメラーゼタンパク質(配列番号1)が、天然HBVポリメラーゼによって示されるRNアーゼ HおよびRTアーゼ活性を排除するために540番(DがH)および718番(EがH)において変異されたものに相当する(結果として配列番号8に示されるアミノ酸配列および配列番号22に示されるヌクレオチド配列が得られる)。次に、再構築されたPol配列をプラスミドベクターにクローニングして、PGA15−pol(配列番号27)を得る。天然HBVポリメラーゼのN末端に存在する最初の47のアミノ酸が欠失された末端切断型を、以下のプライマーOTG19037
を用いてpGA15−PolプラスミドからPCRによって増幅した。得られたフラグメントを、MVAトランスファープラスミドの、p7.5Kプロモーター(Cochran et al 1985, J, Virol, 54:30)の下流のNheIおよびEcoRV制限部位に挿入し、pTG17842を得た。この変異および末端切断型ポリメラーゼを以下pol*と呼ぶ。
次に、HBV Pol*配列を、そのN末端においてペプチドシグナル(SS)と、また、そのC末端において狂犬病ウイルス(ERA分離株;Genbank No M38452に記載)の糖タンパク質に由来する膜係留配列(TMR)と融合させることによって改変した。このSS配列およびTMR配列を、それぞれプライマー対OTG19045(配列番号30)
Pol*−TMR発現の分析は、ウエスタンブロットによって行った。増殖培地にプロテアソーム阻害剤MG−132(10μM)を添加して、または添加せずに、A549細胞にMVA TG17843をMOI 1で感染させた。24時間後、細胞を回収した。ウエスタンブロット分析は、市販のモノクローナル抗Pol抗体Hep B Pol(8D5, Santa Cruz, # sc-81591)を用いて行った。
Core*は、アミノ酸77〜84が欠失したHBV Y07587(配列番号2)のコア配列に相当する。
Core*コード配列は、pGA4−コアプラスミドから2重PCRによって再構成した。このプラスミドはGeneart社によって作製された。このプラスミドは、合成オリゴヌクレオチドおよび/またはPCR産物から組み立てられた改変型HBVコア遺伝子の全長コード配列を含む。コード配列の最後の2つのコドンCAA TGTは、Polとの配列相同性を避けるためにCAG TGCに改変した(配列番号36)。
Core*発現の分析は、ウエスタンブロットによって行った。ニワトリ胚繊維芽細胞にMVA TG17971をMOI 0.2で感染させた。24時間後、細胞を回収した。ウエスタンブロット分析は、市販のモノクローナル抗体Hep B cAg(13A9)(Santa Cruz, # sc-23946)を用いて行った。
Core*tは、アミノ酸148の後で末端切断され、かつ、アミノ酸77〜84が欠失されたHBV Y07587(配列番号2)のコア配列に相当する。
Core*tコード配列は、コード配列の最後の2つのコドンCAA TGTを、Pol(配列番号36)との配列相同性を避けるためにCAG TGCに改変したこと以外は、合成オリゴヌクレオチドおよびPCR産物から組み立てられた全長HBVコア遺伝子をコードする配列を含むpGA4−コアプラスミドからダブルPCRによって再構成した。
Core*t発現の分析は、ウエスタンブロットによって行った。ニワトリ胚繊維芽細胞にMVA TG17972をMOI 0.2で感染させた。24時間後、細胞を回収した。ウエスタンブロット分析は、市販のモノクローナル抗コア抗体Hep B cAg(13A9)(Santa Cruz, # sc-23946)を用いて行った。
Core−t*部分を、HBsタンパク質のアミノ酸14〜51にわたるEnv1ドメインと融合させた。
このCore*t−Env1配列は、ダブルPCRによって再構成した。Core*t配列は、以下のプライマーOTG19317(配列番号41)
Core*t−env1発現の分析は、ウエスタンブロットによって行った。ニワトリ胚繊維芽細胞にMVA TG17993をMOI 0.2で感染させた。24時間後、細胞を回収した。ウエスタンブロット分析は、市販のモノクローナル抗コア抗体Hep B cAg(13A9)(Santa Cruz, # sc-23946)を用いて行った。
1.1.5に記載されているCore*tポリペプチドを、次に、HBsタンパク質のアミノ酸14〜51(Env1)およびアミノ酸165〜194(Env2)にわたる2つの免疫原性ドメインと融合させた。
Core*t−Env1−Env2をコードするヌクレオチド配列をトリプルPCRによって再構築した。Core*t配列は、以下のプライマーOTG19317(配列番号41)およびOTG19319(配列番号42)を用い、pTG17972からPCRによって増幅した。Env1は、以下のプライマーOTG19318(配列番号44)およびOTG19322(配列番号46)
Core*t−env1−env2発現の分析は、ウエスタンブロットによって行った。ニワトリ胚繊維芽細胞にMVA TG17994をMOI 0.2で感染させた。24時間後、細胞を回収した。ウエスタンブロット分析は、市販のモノクローナル抗コア抗体Hep B cAg(13A9, Santa Cruz, # sc-23946)を用いて行った。
CORE−Env1−Env2−Env4融合物をコードする合成遺伝子(831ヌクレオチド)は、ダブルPCRによって再構成した。COREは、以下のプライマーOTG19152(配列番号49)
Met開始部(48〜832)を除く最初の47のアミノ酸が末端切断され、かつ、540番(DがH)および718番(EがH)において変異された(野生型ポリメラーゼに対して)ポリメラーゼタンパク質であるPol*をコードする遺伝子をアデノウイルスベクターに挿入した。Pol遺伝子(2364ヌクレオチド)は、プライマーOTG19155(配列番号53)
抗原の免疫原性は、HLAトランスジェニックマウスの免疫誘導の後に、Elispot IFNγおよび細胞内サイトカイン染色(ICS)アッセイによってin vivoで評価した。
試験に用いたHLA−A2.1トランスジェニックマウスは、Pascolo et al. (1997, J, Exp. Med. 185:2043)によって記載されている。これらのマウスはH−2Dbおよびネズミβ2−ミクログロブリン遺伝子ノックアウトを有し、ヒトβ2mのC末端がキメラ重鎖に共有結合されているトランスジェニック単鎖組織適合性クラスI分子(HHD分子)(HLA−A*0201 α1−α2,H−2Db α3トランスメンブランおよび細胞室内ドメイン)を発現する。7〜10週齢のマウス(雄および雌)に免疫誘導を行った。マウスの平均体重は25〜30g前後であった。
マウスを、群1 AdTG17909(env1、env2およびenv4免疫原性ドメインと融合したHBV Coreをコードする)による免疫誘導、群2 AdTG17910(末端切断および変異型Pol*をコードする)による免疫誘導、群3 両ベクターによる免疫誘導および群4 陰性対照としてのエンプティーアデノウイルス(AdTG15149)による免疫誘導、の4群に分けた。総ての動物を、尾の基部に皮下注射を行うことによって免疫誘導し、群1および2の動物には、108IUの各アデノウイルス(TG17909またはTG17910)の皮下注射を1回施し、群3には、108IUの各アデノウイルスを含む混合物(計2.108IU:108IUのAdTG17909+108IUのAdTG17910)の皮下注射を1回施し、陰性対照には、2.108IUのAdTG15149の皮下注射を1回施した。細胞性免疫応答は、免疫誘導2週間後にIFNg Elispotおよび細胞内サイトカイン染色(ICS)アッセイによって評価した。
in vitro細胞刺激に用いたペプチドは、HLA−A2制限エピトープであると記載されているまたは推定される9〜10アミノ酸の短鎖ペプチドか、または総ての対象抗原をカバーするペプチドライブラリーに含まれる15アミノ酸の長鎖ペプチドのいずれかであった。
ポリメラーゼタンパク質の記載されているまたは推定されるHLA−A2制限エピトープ、コアタンパク質またはEnvドメインに相当する短鎖ペプチドは、Eurogentec (Belgium)によって合成され、100%DMSO(sigma, D2650)に10mM濃度で溶かした。
HBVコアタンパク質は、コアライブラリーに由来する2プールの21〜22ペプチド(プール1:残基1〜100をカバーする22ペプチド;プール2:残基89〜183をカバーする21ペプチド)によってカバーされ;
HBV Envタンパク質は、Envライブラリーに由来する3プールの6〜10ペプチド(プール1:HBs残基9〜59をカバーする10ペプチド;プール2:HBs残基157〜194をカバーする9ペプチド;プール4:HBs残基193〜226をカバーする6ペプチド)によってカバーされる。
免疫マウスからの脾細胞を回収し、赤血球を溶解させた(Sigma, R7757)。2.105細胞ウェルを、抗マウスIFNγモノクローナル抗体(BD Biosciences;10μg/ml,551216)でコーティングしたMultiscreenプレート(Millipore, MSHA S4510)にて、10%FCS(Sigma, F7524またはJRH, 12003-100M)、80U/mLペニシリン/80μg/mLストレプトマイシン(PAN, P06-Q7-100)、2mM L−グルタミン(Gibco, 25030)、1×非必須アミノ酸(Gibco, 11140)、10mM Hepes(Gibco, 15630)、1mMピルビン酸ナトリウム(Gibco, 31350)および50μM β−メルカプトエタノール(Gibco, 31350)を添加したMEM培養培地(Gibco, 22571)中、陰性対照としての10単位/mlの組換えネズミIL2(Peprotech, 212-12)単独の存在下、または
・AdベクターによってコードされるHBV抗原中に存在する1つのHLA−A2制限ペプチド(PolではSLY、CoreではFLP、ILC、EnvではVLQ、FLGおよびGLS)または無関連のもの10μM、
・各ペプチド終濃度5μg/mlのペプチドプール、
・陽性対照としてのコンカナバリンA(Sigma, C5275)5μg/ml
とともに、3反復で40時間培養した。
各群の各動物からの脾細胞に対してICSを行った。溶解バッファー(Sigma, R7757)で赤血球を溶解させた後、ウェル当たり2×106個の細胞を平底96ウェルプレートにて、完全α(MEM培養培地(Gibco BRL, 22571)中、陰性対照として10単位/mlのネズミ組換えIL−2(Peprotech, 212-12)単独の存在下、または10μMの特異的HBVペプチドまたは各ペプチド終濃度5μg/mlのペプチドプールまたは10μMの無関連のペプチドとともにインキュベートした。GolgiPlug(BD Biosciences, 555029)を終濃度1μl/mlですぐに5時間加えた。次に、細胞をV底96ウェルプレートに回収し、1%FCS−PBSで洗浄した。染色は、50μlの1%FCS−PBS中、CD3(ハムスターMAb抗CD3e−PE、1/200希釈)、CD8(ラットMAb抗CD8a−APC、1/600希釈)およびCD4(ラットMAb抗CD4−PerCP、1/600希釈)(総てBD Biosciencesから、それぞれ553063、553035および553052)に対するモノクローナル抗体を用いて室温で15分間行った。洗浄後、細胞を固定し、Cytofix/Cytopermで透過処理を施し、Perm/Wash溶液(BD Biosciences, 554714)で洗浄した。その後、抗マウスIFNg−PE抗体(BD Biosciences, 554412557724)および抗マウスTNFa−Alexa488抗体(BD Biosciences, 557719)または抗マウスIFNg−PE抗体(BD Biosciences, 554412557724)および抗マウス1L2−Alexa488抗体(BD Biosciences, 557719)を室温で15分間加え、Perm/Washで洗浄した後、細胞を1%FCS−PBSに再懸濁させ、FacsCalibur(Becton Dickinson)を用いてフローサイトメトリーにより分析した。CD3e+、CD8a+細胞またはCD3e+、CD4+細胞にゲートをかけ、IFNg+CD8+またはIFNg+CD4+TまたはTNFa+CD8+またはTNFa+CD4+TまたはIL2+CD8+またはIL2 CD4+TまたはIFNg+TNFa+CD8+またはIFNg+TNFa+CD4+またはIFNg+IL2+CD8+またはIFNg+IL2+CD4+T細胞集団の割合%を求めた。媒体単独で得られた割合%をバックグラウンドとみなした。
in vivo CTLアッセイは、記載されているように行った(Fournillier et al., 2007)。脾細胞懸濁液を同系マウスの脾臓から得、赤血球を溶解させた後に20×106細胞/mlに調整した。3分の1の細胞をHBV特異的ペプチドの1つともにインキュベートし、別の3分の1の細胞を別のHBVペプチドとともにインキュベートし(総て終濃度10μMにて37℃で1時間)、最後の3分の1はパルスを行わずにおいた。5(6)−カルボキシフルオレセインジアセテートスクシンイミジルエステル(CFSE)(Molecular probes, C1157)を、unパルスを行わなかった細胞には16μM(CFSE−高)で、ILCまたはVLQペプチドでパルスした細胞には4μM(CFSE−中)で、また、SLYまたはFLPペプチドでパルスした細胞には1μM(CFSE−低)で10分間加えた。PBSで洗浄した後、これらの3つの集団(非パルス、ILCおよびSLYペプチドでパルスした細胞、または非パルス、FLPおよびILCペプチドでパルスした細胞)を混合し、計30.106個の細胞を、麻酔したマウスに後眼窩静脈から注射した(ケタミン−キシラジン−PBS混合物(Ketamine Virbac, Centravet KET204,終濃度25mg/ml;塩酸キシラジン(Rompun Bayer, Centravet,終濃度5mg/mlを使用)。従って、CFSE−低および中集団は、細胞傷害性T細胞によって溶解されると思われる特異的標的に相当し、CFSE−高集団アッセイのノーマライゼーションを可能とする内部参照であった。24時間後、レシピエントマウスからの脾細胞をフローサイトメトリーにより分析して、CFSE標識細胞を検出した。リンパ球(SSC/FSC)にゲートをかけた後、次のゲートを事象数/CFSE蛍光(FL1)に基づいて設定したところ3つのピークが明らかになり、すなわち、1つ目はCFSE−低細胞に相当し、2つ目はCFSE−中細胞に相当し、3つ目はCFSE−高細胞に相当する。動物ごとに、CFSE+ペプチドでパルスされた標的とCFSE+でパルスされていない標的との比を計算した(R=CFSE−低細胞の数/CFSE−高細胞の数)。2匹のナイーブマウスを用いてR参照値を求めた。各動物の特異的溶解の割合%を下式:溶解率%=(1−Rマウス/R参照)×100によって求めた。応答は、特異的溶解の割合%が10%(カットオフ)より大きければ陽性と見なした。
2.1 ウイルスベクターによる抗原の発現
2.1.1 アデノウイルス構築物AdTG17909およびAdTG17910からの抗原の発現
core−env1−env2−env4融合タンパク質の発現は、ウエスタンブロットによって評価した。A549細胞(106細胞)に、AdTG17909または陰性対照としてのエンプティーアデノウイルスを48時間、MOI 10または50で感染させた。細胞ペレットを回収し、抗コアマウスモノクローナル抗体(C1−5,sc−23945,Santa Cruz)でプローブした。図1Aに示されるように、AdTG17909感染細胞から回収されたサンプルにおいて、予測分子量(31.6kDa)を有する主要バンドが現れた。
Pol*、Pol*TMR、Core*t、Core*tEnv1、Core*t−Env1−Env2の発現の分析は、ウエスタンブロットによって行った。MVA TG17842およびMVA TG17843では増殖培地にプロテアソーム阻害剤MG−132(10μM)を加えて、または加えずに、A549細胞またはCEFに、それぞれMVA TG17842、MVA TG17843、MVA TG17971、MVA TG17972、MVA TG17993およびMVA TG17994をそれぞれMOI 1または0.2で感染させた。24時間後、細胞を回収した。
MVA TG17993およびMVA TG17994では、ウエスタンブロット分析は、市販のモノクローナル抗コア抗体Hep B cAg(13A9,Santa Cruz,#sc-23946)を用いて行った。図1Bに示されるように、見掛けの分子量がそれぞれ19.9および23.4kDaのタンパク質の発現が検出された。このバンドはCore*t−Env1タンパク質およびCore*t−Env1−Fnv2の予測分子量を有する。
アデノウイルスベクターによって発現されるHBVポリペプチドの免疫原性は、AdTG1 7909またはAdTG17910単独か、またはこの2つのアデノウイルスの混合物のいずれかで免疫誘導したHLA−A2トランスジェニックマウスにおいて評価した。1回の皮下注射の後に誘導された特異的T細胞応答を、ポリメラーゼ、コアもしくはエンベロープドメイン、または/および着目するHBV抗原をカバーするオーバーラップペプチドのプールに存在する既知の(患者における特異的T細胞応答の標的であると記載されている)HLA−A2エピトープを用いた、Elispot IFNg、ICSおよびin vivo細胞溶解アッセイによって評価した。
Elispot IFNgアッセイでは、HLA−A2制限エピトープ(SLYADSPSV)(HBVポリメラーゼ内の816〜824番に位置する配列番号55)に特異的なIFNgを産生する細胞を誘導し得ることが示された(図2A)。AdTG17909による免疫誘導もまた、2つのコアHLA−A2制限エピトープ(18〜27番のFLPSDFFPSV(配列番号56)および99〜108番のILCWGELMTL(配列番号57)、ならびに3つのエンベロープHLA−A2制限エピトープ(双方ともEnv1に存在する14〜22番のVLQAGFFLL(配列番号58)および41〜49番のFLGGTTVCL(配列番号59)、ならびにEnv2に存在する185〜194番のGLSPTVWLSV(配列番号60))に特異的なIFNgを産生する細胞の高頻度での誘導をもたらした(図2BおよびC)。AdTG17909とAdTG17910の混合物による免疫誘導もまた、3つの抗原の同じエピトープ、すなわち、Polに存在するSLYエピトープ、コアタンパク質に存在するFLPおよびILCエピトープ、そしてエンベロープドメインの3つのエピトープ(VLQ、FLGおよびGLS)を標的とする、匹敵するレベルの特異的IFNg産生細胞を誘導した(図2A、BおよびC)。単一のAdまたは2つの混合物による免疫誘導後に検出されたT細胞応答の頻度は匹敵するものであったが、これは記載したベクターから発現した3つの抗原の間に主要な免疫優性が無いことを示す。
ポリメラーゼドメイン(SLY)、コアドメイン(FLPおよびILC)およびエンベロープドメイン(VLQ、FLGおよびGLS)に存在するHLA−A2制限エピトープを標的とする、IFNg単独またはIFNg+TNFaのいずれかを誘導することができるCD8 T細胞の数を、ICSアッセイによって評価した。これら総てのエピトープが二重および単一分泌細胞の標的であった。結果を図3に示す。AdTG17909単独またはAdTG17910との組合せで免疫誘導した動物では、およそ同等のCore特異的CD8 T細胞応答およびEnv特異的CD8 T細胞応答(図3Bおよび3Cに示されるように、FLP、ILC、VLQ、FLGおよびGLSペプチドで再刺激した後にIFNgまたはIFNg+TNFaを産生する特異的CD8 T細胞の割合%が同じ)が誘導された。他方、ポリメラーゼ特異的CD8 T細胞応答(SLYエピトープ)に関しては、Pol*を発現するAdTG1710で処置されたマウス(図3A)ならびに程度は低いが、AdTG17910とAdTG17909の混合物で免疫誘導されたマウス(図3C)において、極めて高い割合%のIFNgまたはIFNg+TNFa産生CD8+細胞が検出された。
ポリメラーゼドメイン(SLY)、コアドメイン(FLPおよびILC)およびエンベロープドメイン(VLQ、FLGおよびGLS)またはコアタンパク質とEnvドメインをカバーするオーバーラップペプチドのプールに存在するHLA−A2制限エピトープを標的とする、IFNg単独またはIFNg+TNFaまたはIFNg+IL2のいずれかを誘導することができるCD8およびCD4 T細胞の割合%をICSアッセイによって評価した。供試した総てのHLA−A2制限エピトープが単一および二重分泌(IFNgおよびIFNg+TNFa)細胞の標的であり、およびオーバーラップペプチドの一部のプールも、単一および二重産生細胞(IFNgおよびIFNg+TNFおよびIFNg+1L2)の標的であった。結果を図4に示す。5匹のHLA−A2トランスジェニックマウスをAdTG17909とAdTG17910の混合物で免疫誘導し、3匹のHLA−A2トランスジェニックマウスをAdTG15149(陰性対照)で免疫誘導した。AdTG15149で免疫誘導した動物は、HBV特異的T細胞応答を示さなかった(データは示されていない)。AdTG17909とAdTG17910を組み合わせて免疫誘導した動物は、HBV標的抗原に特異的な強いCD8 T細胞応答を示し(図4A)、ポリメラーゼドメイン、コアドメインおよびEnvドメインに存在するHLA−A2エピトープに特異的であり、かつ、ペプチドの「コア1」プールならびにEnv1およびEnv2ドメインをカバーするペプチドのプールに特異的な単一(IFNg)および二重(IFNg+TNFa)産生細胞の割合%は高かった。図4Bおよび4Cに示されるように、これらのワクチン接種マウスはまた、HBV抗原に特異的なCD4 T細胞応答、特に、ペプチドの「コア2」プールおよびEnv2をカバーするペプチドのプールに特異的な単一(IFNg)および二重(IFNg+TNFaおよびIFNg+IL2)産生細胞を示した。
アデノウイルスベクターAdTG17909およびAdTG179I0の、HBV HLA−A2エピトープを提示するagainsT細胞に対してin vivo細胞溶解を誘導する能力を、in vivoCTLアッセイによって評価した。4つのHLA−A2エピトープ、すなわち、SLY(Pol)、FLPおよびILC(Core)およびVLQ(Env1ドメイン)をそれぞれ試験した。6匹の動物をAdTG17909+AdTG17910で免疫誘導し、2匹の動物をAdTG15149(陰性対照)で免疫誘導した。各群の半数(AdTG17909+AdTG17910免疫誘導マウスは3匹、AdTG15149免疫誘導マウスは1匹)の、SLYペプチドでパルスされた細胞およびILCペプチドでパルスされた細胞をin vivoで溶解する能力を試験した。残りの半数で、ワクチン接種動物の、FLPペプチドでパルスされた細胞およびVLQペプチドでパルスされた細胞をin vivoで溶解する能力を試験した。結果を図5に示す。予測されたように、AdTG15149で免疫誘導したマウスでは、HBV特異的in vivo細胞溶解は検出できなかった(データは示されていない)。AdTG17909+AdTG17910で免疫誘導したマウスでは、2つのコアエピトープFLPおよびILCに対するin vivo細胞溶解は弱かった。しかしながら、対照的に、AdTG17909とAdTG17910の混合物で免疫誘導した動物は、ポリメラーゼエピトープSLY(図5A)およびEnv1 エピトープVLQ(図5B)に対して強いin vivo細胞溶解を示し、両場合とも50%を超える特異的溶解に達した。
実施例1.1.2〜1.1.7に記載されているMVAベクターの1つ(MVA TG17842、MVA TG17843、MVA TG17971またはMVA TG17972単独)で、または2つのMVAの混合物(MVA TG17843+MVA TG17972、MVA TG17843+MVA TG17993またはMVA TG17843+MVA TG17994)で免疫誘導したHLA−A2トランスジェニックマウスにおいて、MVAに基づく組成物の免疫原活性を評価した。マウスに1週間おきに3回の皮下注射を施して免疫誘導し、特異的T細胞応答を、ポリメラーゼドメイン、コアドメインもしくはエンベロープドメインまたは/および着目するHBV抗原をカバーするオーバーラップペプチドのプールに存在する上記のHLA−A2エピトープを用いたElispot IFNgおよびICSによって評価した。
3匹のマウスを、末端切断および変異型ポリメラーゼ抗原を発現するMVA TG17842または同じ末端切断および変異型ポリメラーゼの膜標的型を発現するMVA TG17843またはMVA N33.1(陰性対照)のいずれかで免疫誘導した。ポリメラーゼ特異的T細胞応答を、SLY HLA−A2制限エピトープおよび該ポリメラーゼをカバーするペプチドのプールを用いたIFNg Elispotアッセイによって評価した。MVA N33.1およびMVA TG17842で免疫誘導されたマウスでは、HBV特異的T細胞応答は検出されなかった(データは示されていない)。しかしながら、図6Aに示されるように、MVA TG17843で免疫誘導した後では、HLA−A2制限エピトープSLY、およびポリメラーゼのC末端部分をカバーするペプチドプール8に特異的なIFNg産生細胞が誘導された(調べた試験条件下では、他のペプチドプール1〜7に対する特異的応答は検出できなかった)。これらのデータは、MVAに基づく組成物において膜係留抗原としてポリメラーゼを発現することの利点を強調する。
8匹のマウスを、残基77〜84を欠失したコア部分を発現するMVA TG17971、またはその末端切断型(残基149からのC末端切断)を発現するMVA TG17972、またはMVA N33.1(陰性対照)のいずれかで免疫誘導した。コア特異的T細胞応答を、HLA−A2制限エピトープ(FLPおよびILCペプチド)およびペプチドの上記コア1およびコア2プールを用いたIFNg Elispotアッセイによって測定した。図6Bに示されるように、MVA TG17971による免疫誘導により、HLA−A2制限FLPおよびILCペプチド、ならびにコア抗原をカバーする2つのペプチドプールに対して特異的な散在性T細胞応答を誘導することができる(図6B)。調べて試験条件下で供試ペプチドを用いた場合、MVA TG17972で免疫誘導したマウスにおいてコア特異的T細胞応答は検出できなかった(データは示されていない)。
3匹のマウスをMVA TG17843と、MVA TG17972、MVA TG17993およびMVA TG17994との混合物で免疫誘導し、HBV特異的T細胞応答を、上記のペプチドを用いたElispot IFNgアッセイによって評価した。
・ポリメラーゼ特異的T細胞応答は、MVA TG17843+MVA TG17972(図7Aに示されるように2/3の動物で陽性応答)、MVA TG17843+MVA TG17993(図7Bに示されるように3/3の動物で陽性応答)およびMVA TG17843+MVA TG17994の組合せ(図7Cに示されるように2/3の動物で陽性応答)を接種した動物の大多数に検出された。IFNg産生細胞の頻度は、MVA TG17843を単独で注射した場合(図6A)に見られるものに匹敵すると思われ、ベクターの組合せは誘導される免疫応答に悪影響を及ぼさないことを示す。
・接種を行った動物において、調べた試験条件下では、HLA−A2またはペプチドプールを用いた場合、コア特異的応答は検出できなかった(データは示されていない)。
・MVA TG17993を含む組合せで接種を行った動物において、調べた試験条件下では、Env1 HLA−A2またはペプチドプールを用いた場合、env特異的応答は検出できなかった(データは示されていない)。
・図7Cに示されるように、MVA TG17994を含む組合せで免疫誘導した3匹のうち2匹の動物において、HLA−A2制限GLSエピトープおよびEnv2をカバーするペプチドプールの双方に対するEnv2−特異的T細胞応答が検出された(これらの試験条件下ではEnv1ドメインに対するT細胞応答は検出できなかった(データは示されていない))。
興味深いことに、pol部分、コア部分およびenv部分を発現するMVAベクターの組合せは、同時注射したpolおよびenv2抗原を標的とする特異的T細胞応答の誘導を可能とする。
これらのデータを考え合わせると、記載されている組成物の免疫原活性およびそれらの、主要なHBV抗原に対するT細胞応答誘導能が証明される。
Claims (59)
- 少なくとも1つのポリペプチドまたは該少なくとも1つのポリペプチドをコードする核酸分子を含んでなる免疫原性組成物であって、前記少なくとも1つのポリペプチドが、
(i)第1のHBVウイルスに由来するポリメラーゼタンパク質の少なくとも450個のアミノ酸残基を含んでなるポリメラーゼ部分;
(ii)第2のHBVウイルスに由来するコアタンパク質の少なくとも100個のアミノ酸残基を含んでなるコア部分;および
(iii)第3のHBVウイルスに由来するHBsAgタンパク質の連続した15〜100個のアミノ酸残基の1以上の免疫原性ドメインを含んでなるenv部分;または
該ポリメラーゼ部分、該コア部分、該env部分、該ポリメラーゼ部分をコードする該核酸分子、該コア部分をコードする該核酸分子および/または該env部分をコードする該核酸分子の任意の組合せ

- 前記コア部分とenv部分の融合ポリペプチドが、
・配列番号18に示されるアミノ酸配列と少なくとも80%の同一性を示すアミノ酸配列を含んでなるポリペプチド;
・配列番号19に示されるアミノ酸配列と少なくとも80%の同一性を示すアミノ酸配列を含んでなるポリペプチド;および
・配列番号20に示されるアミノ酸配列と、もしくは残基1で始まり、残基251で終わる配列番号20に示されるアミノ酸配列の一部と、あるいはまた残基1で始まり、残基221で終わる配列番号20に示されるアミノ酸配列の一部と少なくとも80%の同一性を示すアミノ酸配列を含んでなるポリペプチド;または配列番号20において残基77〜84を欠くその欠失型
からなる群から選択される、請求項28に記載の免疫原性組成物。 - 請求項1および8〜18のいずれか一項に定義されるポリメラーゼ部分、請求項1および19〜23のいずれか一項に定義されるコア部分、ならびに請求項1および24〜29のいずれか一項に定義されるenv部分またはそれらの任意の組合せをコードする核酸分子。
- ・配列番号21に示されるヌクレオチド配列と少なくとも80%の同一性を示すヌクレオチド配列を含んでなる核酸分子;
・配列番号22に示されるヌクレオチド配列と少なくとも80%の同一性を示すヌクレオチド配列を含んでなる核酸分子;
・1480番のGヌクレオチドのCへの置換、2014番のGヌクレオチドのCへの置換、および2016番のAヌクレオチドのTへの置換を含む、配列番号21に示されるヌクレオチド配列と少なくとも80%の同一性を示すヌクレオチド配列を含んでなる核酸分子;
・配列番号23に示されるヌクレオチド配列と少なくとも80%の同一性を示すヌクレオチド配列を含んでなる核酸分子;
・配列番号24に示されるヌクレオチド配列と少なくとも80%の同一性を示すヌクレオチド配列を含んでなる核酸分子;
・配列番号25に示されるヌクレオチド配列と少なくとも80%の同一性を示すヌクレオチド配列を含んでなる核酸分子;および
・配列番号26に示されるヌクレオチド配列と、またはヌクレオチド1で始まりヌクレオチド753で終わる配列番号26に示されるヌクレオチド配列の部分と、またはヌクレオチド1で始まりヌクレオチド663で終わる配列番号26に示されるヌクレオチド配列の部分と、少なくとも80%の同一性を示すヌクレオチド配列を含んでなる核酸分子;または配列番号26の229番のGから252番のAまでに及ぶ部分を欠失しているそれらの欠失型
からなる群から選択される、請求項30に記載の核酸分子。 - 請求項30または31に定義される1種以上の核酸分子を含んでなるベクター。
- プラスミドまたはウイルスベクターである、請求項32に記載のベクター。
- 前記ウイルスベクターが、複製欠損型アデノウイルスベクターである、請求項33に記載のベクター。
- 前記核酸分子が、E1領域の代わりに挿入されている、請求項34に記載のベクター。
- 前記ウイルスベクターが、ポックスウイルスベクター、より詳しくはカナリア痘ウイルス、鶏痘ウイルスまたはワクシニアウイルスから得られる、請求項33に記載のベクター。
- 前記ワクシニアウイルスが、改変アンカラ(MVA)系統である、請求項36に記載のベクター。
- 前記ポリメラーゼ部分、前記コア部分、および前記env部分をコードする核酸分子を担持する、請求項32〜37のいずれか一項に記載のベクター。
- 前記ポリメラーゼ部分、前記コア部分、および前記env部分をコードする核酸分子の1つだけまたは2つを担持する、請求項32〜37のいずれか一項に記載のベクター。
- (i)7.5Kプロモーターなどのワクシニアプロモーターの制御下に置かれた、配列番号7、8もしくは9に、または494番のAsp残基のHis残基への置換および672番のGlu残基のHis残基への置換を含む配列番号7に示されるアミノ酸配列を含んでなるポリメラーゼ部分をコードする、核酸分子を含んでなるMVAベクター;
(ii)pH5rプロモーターなどのワクシニアプロモーターの制御下に置かれた、配列番号18または19に示されるアミノ酸配列を含んでなるコア部分およびenv部分をコードする核酸分子を含んでなるMVAベクター;
(iii)E1領域の代わり挿入された、CMVプロモーターの制御下に置かれた核酸分子であり、配列番号7、8もしくは9に、または494番のAsp残基のHis残基への置換および672番のGlu残基のHis残基への置換を含む配列番号7に示されるアミノ酸配列を含んでなるポリメラーゼ部分をコードする核酸分子を含んでなるE1欠損型Adベクター;および
(iv)E1領域の代わり挿入された、CMVプロモーターの制御下に置かれた、配列番号18、19もしくは20に、または残基1で始まり残基251で終わる配列番号20の部分に、または残基1で始まり残基221で終わる配列番号20の部分に示されるアミノ酸配列を含んでなるコア部分およびenv部分をコードする核酸分子を含んでなるE1欠損型Adベクター
からなる群から選択される、請求項39に記載のベクター。 - 請求項30もしくは31に記載の核酸分子または請求項32〜40のいずれか一項に記載のベクターを含んでなる、感染性ウイルス粒子。
- 以下の工程:
(e)本発明のウイルスベクターを好適な細胞系統に導入する工程、
(f)該感染性ウイルス粒子の産生を可能にする好適な条件下で該細胞系統を培養する工程、
(g)産生された感染性ウイルス粒子を該細胞系統の培養物から回収する工程、および
(h)所望により、該感染性ウイルス粒子回収物を精製する工程
を含んでなる方法によって産生される、請求項41に記載の感染性ウイルス粒子。 - 請求項30もしくは31に記載の核酸分子、請求項32〜40のいずれか一項に記載のベクター、または請求項42もしくは43に記載の感染性ウイルス粒子を含んでなる、宿主細胞。
- 請求項30もしくは31に記載の核酸分子、請求項32〜40のいずれか一項に記載のベクター、または請求項42もしくは43に記載の感染性ウイルス粒子、または請求項43に記載の宿主細胞と、薬学上許容されるビヒクルとを含んでなる、組成物。
- ヒトにおける全身適用または粘膜適用に好適な1種以上のアジュバントをさらに含んでなる、請求項44に記載の組成物。
- 筋肉内投与または皮下投与用に処方される、請求項44または45に記載の組成物。
- 約105〜約1013感染単位のウイルスベクターまたは感染性ウイルス粒子を含んでなる、請求項44〜46のいずれか一項に記載の組成物。
- HBV感染またはHBV関連疾患もしくは病的状態の治療または予防を意図した薬物の製造のための、請求項1および8〜29のいずれか一項に定義されるHBV部分、請求項1〜29のいずれか一項に記載の免疫原性組成物、請求項30もしくは31に記載の核酸分子、請求項32〜40のいずれか一項に記載のベクター、請求項41もしくは42に記載の感染性ウイルス粒子、請求項43に記載の宿主細胞、または請求項44〜47のいずれか一項に記載の組成物の少なくとも1つの使用。
- HBV感染またはHBV関連疾患および病的状態を治療または予防する方法であって、それを必要とするヒトまたは動物生物体に、治療上有効な量の、請求項1および7〜29のいずれか一項に定義されるHBV部分、請求項1〜29のいずれか一項に記載の免疫原性組成物、請求項30もしくは31に記載の核酸分子、請求項32〜40のいずれか一項に記載のベクター、請求項41もしくは42に記載の感染性ウイルス粒子、請求項43に記載の宿主細胞、または請求項44〜47のいずれか一項に記載の組成物の少なくとも1つを投与することを含んでなる、方法。
- 慢性B型肝炎ウイルス感染を有する患者を治療するための、請求項48に記載の使用または請求項49に記載の方法。
- 前記方法または使用が、標準治療と併用される、請求項48〜50のいずれか一項に記載の方法または使用。
- 前記標準治療が、ラミブジン、エンテカビル、テルビブジン、アデホビル、ジピボキシル、またはテノホビルからなる群から選択されるサイトカインおよび/またはヌクレオチドまたはヌクレオシド類似体を含んでなる、請求項51に記載の方法または使用。
- 宿主生物においてHBVに対する免疫応答を誘導または刺激する方法であって、該免疫応答を誘導または刺激するために、該生物に、請求項1および7〜29のいずれか一項に定義されるHBV部分、請求項1〜29のいずれか一項に記載の免疫原性組成物、請求項30もしくは31に記載の核酸分子、請求項32〜40のいずれか一項に記載のベクター、請求項41もしくは42に記載の感染性ウイルス粒子、請求項43に記載の宿主細胞、または請求項44〜47のいずれか一項に記載の組成物の少なくとも1つをを投与することを含んでなる、方法。
- 前記免疫応答が、細胞性であり、CD4+および/またはCD8+に媒介される、請求項53に記載の方法。
- HBV感染の治療に使用するためのパーツキットであって、請求項1および7〜29のいずれか一項に定義されるHBV部分、請求項1〜29のいずれか一項に記載の免疫原性組成物、請求項30もしくは31に記載の核酸分子、請求項32〜40のいずれか一項に記載のベクター、請求項41もしくは42に記載の感染性ウイルス粒子、請求項43に記載の宿主細胞、または請求項44〜47のいずれか一項に記載の組成物からなる群から選択される複数の活性物質を含んでなる、パーツキット。
- 請求項1および7〜18のいずれか一項に定義されるポリメラーゼ部分をコードする核酸分子を含んでなる第1のベクターと、請求項1および19〜29のいずれか一項に定義されるコア部分および/またはenv部分をコードする核酸分子を含んでなる第2のベクターとを含んでなる、請求項55に記載のパーツキット。
- 前記第1のベクターが、請求項40(i)に定義されるMVAベクターであり、前記第2のベクターが、請求項40(ii)に定義されるMVAベクターである、請求項56に記載のパーツキット。
- 前記第1のベクターが、請求項40(iii)に定義されるAdベクターであり、前記第2のベクターが、請求項40(iv)に定義されるAdベクターである、請求項56に記載のパーツキット。
- 免疫調節剤を発現する第3のベクターをさらに含んでなる、請求項56〜58のいずれか一項に記載のパーツキット。
