Abstract

Description
SNS−595は、新規なナフチリジン細胞傷害剤であり、すでにAG−7352として
知られている(例えば、Tsuzukiらの論文(Tetrahedron-Asymmetry 12: 1793-1799 (2001

様において、該赤血球生成剤はダルベポエチンアルファである。別の実施態様において、
該赤血球生成剤はダルベポエチンアルファであり、かつ治療的有効量は約1.5μg/k
gから約4.5μg/kgである。別の実施態様において、該赤血球生成剤はダルベポエ
チンアルファであり、かつ治療的有効量は1週間につき約1.5μg/kgから約4.5
μg/kgの投与量である。

(注射、又は静脈内注射に適した医薬組成物)
酸性組成物(<pH4)は、SNS−595の増加した溶解度の適切なバランスと、好
ましい薬学的特性(例えば、配達位置においてより少ない刺激によって患者の快適さが増
加する)をもたらす。適切な組成の実例には、メタンスルホン酸を用いてpHを2.5に
調整した4.5%ソルビトールの水溶液の毎mLに対して10mgのSNS−595を含
む。そのような溶液を作るための一つの手順は、100mg/10mLの製造において以
下を含む:100mgのSNS−595、及び450mgのD−ソルビトールを蒸留水に
加える;該容積を10mLにする;及び、結果として生じた溶液のpHを、メタンスルホ
ン酸を用いて2.5に調整する。また、その結果の組成物は凍結乾燥にも適している。凍
結乾燥態は、それから蒸留水を用いて、使用前に適切な濃度にもどされる。
(ガン患者におけるSNS−595の薬物動態)
SNS−595を、最長6周期まで被験患者へ投与した。1周期を3週間として定義し
、各周期の初日(0日)においてSNS−595を投与し、追って少なくとも21日観察
した。SNS−595を少なくとも3人から成る患者群に投与し、順次的に群ごとに投与
量を増加させた。SNS−595の量は、濃度曲線下面積が無限大である直線であり、そ
の薬物動態特性は、同じ群の患者間で非常に一致していた。図1は、様々な患者群間での
経時的なSNS−595の血漿濃度を示しており、表1はそれから得られた薬物動態パラ
メータを示している。
Claims (26)
- SNS−595を10mg/m2〜150mg/m2の投与量で患者に投与することを
含む、ガンの治療方法。 - 前記投与量が10mg/m2〜100mg/m2である、請求項1記載の方法。
- 前記投与量が30mg/m2〜75mg/m2である、請求項1記載の方法。
- 前記投与量が40mg/m2〜80mg/m2である、請求項1記載の方法。
- 前記投与量が50mg/m2〜90mg/m2である、請求項1記載の方法。
- 前記治療されるガンが、膀胱ガン、乳ガン、子宮頸ガン、結腸ガン、食道ガン、頭頸部
ガン、白血病、肝臓ガン、肺ガン、リンパ腫、黒色腫、骨髄腫、神経芽細胞腫、卵巣ガン
、膵臓ガン、前立腺ガン、腎臓ガン、肉腫、皮膚ガン、胃ガン、精巣ガン、甲状腺ガン、
及び子宮ガンからなる群から選択される、請求項1記載の方法。 - 前記投与量が10mg/m2〜100mg/m2であり、治療されるガンが固形ガンで
ある、請求項1記載の方法。 - 前記投与量が60mg/m2〜150mg/m2であり、治療されるガンが血液ガンで
ある、請求項1記載の方法。 - 患者に治療的に有効な投与量の対症療法剤を投与することをさらに含む、請求項1記載
の方法。 - 前記対症療法剤が制吐薬である、請求項9記載の方法。
- 前記対症療法剤が造血薬である、請求項9記載の方法。
- i)患者に、10mg/m2〜150mg/m2の投与量のSNS−595を投与する
こと;ii)該対象にSNS−595が投与されない場合、少なくとも一日間待機すること
;iii)患者に、10mg/m2〜150mg/m2の投与量の別のSNS−595を投
与すること;及び、iv)ii)〜iii)のステップを複数回繰り返すことを含む方法。 - 該待機期間が、少なくとも2日である、請求項12記載の方法。
- 該待機期間が、少なくとも3日である、請求項12記載の方法。
- 該待機期間が、少なくとも6日である、請求項12記載の方法。
- 該待機期間が、少なくとも13日である、請求項12記載の方法。
- 該待機期間が、少なくとも20日である、請求項10記載の方法。
- 該待機期間が、少なくとも27日である、請求項10記載の方法。
- 患者に、毎週SNS−595を投与することを含む方法。
- 患者に、3週間ごとにSNS−595を投与することを含む方法。
- 3週間の間、毎週、対象にSNS−595を投与すること、続いて少なくとも2週間は
前記対象にSNS−595を投与しないことを含むサイクルを含む方法であって、該サイ
クルが複数回繰り返される、前記方法。 - 10mg/m2〜150mg/m2のSNS−595を患者に投与すること、及び治療
