Description

感染性組換え非分節型マイナス鎖ウイルスを作出する第一の方法は:
a.RNAポリメラーゼ、非分節型マイナス鎖RNAウイルスの核タンパク質(N)及び非分節型マイナス鎖RNAウイルスのポリメラーゼ補助因子リン酸化タンパク質(P)を安定して産生する細胞又は細胞培養物を、非分節型マイナス鎖RNAウイルスのcDNAクローンにより、及び非分節型マイナス鎖RNAウイルスのRNAポリメラーゼラージタンパク質(L)をコードする核酸を含むベクターにより組換える工程、
b.非分節型マイナス鎖RNAウイルスの複製及び作出を維持するのにコンピテントな細胞に、該組換細胞又は組換細胞培養物を移送する工程、並びに
c.工程bの共培養物から、感染性組換え非分節型マイナス鎖RNAウイルスを回収する工程:を含んでなる。
本明細書において使用される「移送」は、組換細胞の、異なる細胞型上の、特に異なる細胞型の単層上の配置をいう。これらのうち後者の細胞は、感染性組換え非分節型マイナス鎖RNAウイルスの複製及び作出の両方、すなわち細胞内の感染性ウイルスの形成及び可能性のあるこれらの感染性ウイルスの細胞外への放出の各々を維持するためにコンピテントである。この移送は、本発明の組換細胞の、前文で定義されたようなコンピテント細胞との共培養を生じる。前記移送は、組換細胞が効率的にウイルス産生する培養でない、すなわちその感染性ウイルスはこれらの組換細胞から効率的に回収することができない場合の、追加の、すなわち任意の工程であってよい。この工程は、本発明の組換細胞の、非分節型マイナス鎖RNAウイルスのcDNAクローン、及び任意に非分節型マイナス鎖RNAウイルスのRNAポリメラーゼラージタンパク質(L)をコードする核酸を含むベクターによる更なる組換え後に導入される。
