Abstract
【解決手段】I-SceI酵素をコ−ドした、単離DNAを提供する。該DNA配列は、クロ−ニングベクタ及び発現ベクタ、形質転換細胞及びトランスジェニック動物へ導入することが可能である。該ベクタは遺伝子マッピングと、遺伝子の部位特異的導入に有益である。また、染色体DNA中に少なくとも一つの、部位特異的な切れ目を誘導し、ポリペプチドをコードするDNAを挿入する方法により毛質転換する組換え細胞の提供は(a)I-SceI制限部位を具備した二本鎖DNAを有する細胞を用意する;(b)I-SceIをコ−ドするDNAを具備した、少なくとも一つのプラスミドで前記細胞を形質転換する;および、(c)少なくとも一つの、二本鎖の切れ目が誘導された細胞を選択する。

Description
遺伝子を哺乳類の生殖系列に導入することは、生物学で非常に興味のもたれていることである。外来的に加えたDNAを取り込み、またそのDNA中に含まれている遺伝子を発現するという哺乳類細胞の性向が、長い間知られている。遺伝子操作の結果は、これらの動物の子孫へも遺伝する。これら子孫の細胞全ては、導入遺伝子をゲノムの遺伝的構成の一部として遺伝している。このような動物は、トランスジェニックと呼ばれている。
したがって本発明は、当該技術分野でのこれらの必要性を実現するのを助ける。特には、本発明はI-Sce I酵素をコ−ドする単離されたDNAに関連する。該DNAは以下の配列を有している:
真性のミトコンドリア遺伝子(参考文献8)は、ミトコンドリアの遺伝コ−ドの特性のために、大腸菌(E.coli)、酵母、またはその他の生物体で発現することはできない。「ユニバ−サル・コ−ド・エクゥイバレント」が、試験管内部位特異的変異導入により構築された。その配列は図1に示してある。非ユニバ−サルなコドン(二つのCTNは除く)が、大腸菌(E.coli)では非常にまれな、幾つかのコドンとともに置換されていることに注目されたし。
本発明は、酵素I-Sce Iをコ−ドする、単離したDNA配列に関する。酵素I-Sce Iは、エンドヌクレア−ゼである。この酵素の性質は以下の通りである(参考文献14)。
I-Sce Iは認識配列でDNAを開裂させる二本鎖エンドヌクレア−ゼである。I-Sce Iは、3'OHが張り出した4bpのよろめいた(staggered)開裂部を生成する。
基質:二本鎖DNAに作用する。基質DNAは弛緩していても、ネガティブなス−パ−コイルであってもよい。
カチオン:酵素活性には、Mg++(8 mMが最適)が必要である。Mn++は、Mg++の代わりになりえるが、この場合には、認識の厳密度が低下する。
最適な活性条件:高いpH(9から10)、20−40℃の温度、一価のカチオンが存在しないこと。
酵素の安定性:I-Sce Iは室温で不安定である。酵素−基質複合体は酵素のみよりもより安定である(認識部位の存在により、酵素が安定化する)。

−1−部位の変異体、
−2−酵母の全ゲノム(サッカロマイセス・セレビシアエは、ゲノムの複雑さは1.4 X 107bpである)、
で調べる。デ−タは未発表である。
−1−部位の変異:図3に示してあるように、厳密度の一般的なシフトが起きていて、すなわちMg++で重度に影響を被る変異体は、部分的にMn++で影響を受けるようになり、Mg++で部分的に影響を受ける変異体が、Mn++で影響を受けなくなる。
−2−酵母:マグネシウムの条件では、正常な酵母において開裂は観察されなかった。同じ条件では、トランスジェニックな酵母由来のDNAは、人工的に挿入されたI-Sce I部位で完全に開裂して、その他の開裂部位は全く検出されなかった。マグネシウムをマンガンで置換すると、酵母の全ゲノム中で5つの開裂部位が更に発見されたが、この何れもが完全には開裂しなかった。よって、マンガン存在下ではこの酵素は、ca3百万塩基対(5/1.4 X 107bp)当たり平均して1部位をあらわす。
−1−それぞれの部位での可能な変異の全てが決定されてはいない:よって、重度に影響された変異体に相当する塩基は、別の変異体で同じ部位を試験した場合に重要であるかもしれない。
−2−非常に重要な塩基(変異体全てにおいて重度に影響されている)と、中程度に重要な塩基(変異体の幾つかが重度に影響されている)との間のはっきりとした境目はない。酵素に対しての、上等の基質と、悪い基質との間は連続している。

酵素I-Sce Iをコ−ドする、本発明のDNA配列はまた、組織や体液のような生物学的試料中でのヌクレオチド配列の検出用のプロ−ブにもしようすることができる。このプロ−ブは、原子または無機のラジカル、もっとも普通には放射性核種で標識することが可能であるが、また重金属でもおそらく可能であろう。放射性標識には32P、3H、14C などが含まれる。適切なシグナルと十分な半減期をゆうする、如何なる放射性標識を用いることが可能である。他の標識には、標識済み抗体、蛍光体(fluorescers)、化学蛍光体、酵素、標識済みリガンドに対して、対で特異的に結合する抗体などに特異的に結合できるリガンドがある。標識の選択はプロ−ブの、DNAまたはRNAへのハイブリダイゼーション速度への、標識の影響により決まる。標識は、ハイブリダイゼーションが可能なDNA、またはRNAの量を検出するのに十分な感度を提供することが必要であろう。
本発明は更に、本発明の酵素I-Sce Iをコ−ドするDNA配列に関しているが、該ヌクレオチド配列は他の核酸に連結されている。この核酸は、例えばプラスミド、DNA若しくはRNA、原核生物や真核生物が含まれる如何なる起源の天然のDNA若しくはRNA、のような如何なるものよりから得られる。DNAまたはRNAは、文献に記載されるような種々の技術(Maniatis et al., Molecular Cloning: A Laboratory Manual, Cold Spring Harbor Laboratory, New York (1982))により、生物学的液体や組織のような、生物学的物質より抽出することができる。慨して核酸は、細菌、酵母、ウイルス、または植物や動物のような高等生物より得られるであろう。該核酸は、ヒトのDNA全体に含まれる遺伝子の一部、または特異的な微生物の核酸配列の一部のような、より複雑な混合物の画分であってもよい。該核酸は、より大きい分子の画分であるか、または遺伝子全体、若しくは遺伝子の集合を構成するものであってもよい。DNAは、一本鎖か、または二本鎖の形態であてもよい。上記の断片が一本鎖の形態である場合には、通常の技術により、DNAポリメラ−ゼを使用して、二本鎖に変換されてもよい。
本発明は更に、酵素I-Sce Iをコ−ドする本発明のDNA配列を有するクロ−ニング、及び発現用のベクタに関する。
pSCM525:tacプロモ−タと、I-Sce Iの合成遺伝子とを挿入したpUC12に由来する、大腸菌(E.coli)の発現ベクタである。発現はIPTGで誘導される。
PEX7:ガラクト−スプロモ−タの制御下に合成遺伝子を挿入したpRP51-Bam O(pLG-SD5のLEU2d誘導体)から得られる、酵母発現ベクタ。発現は、ガラクト−スで誘導される。
pRSV I-Sce I:合成遺伝子(pSCM525のBamHI−PstI断片)が、ラウス肉腫ウイルスのLTRプロモ−タ制御下にある、pRSV誘導体。この発現ベクタは、図8に記載してある。
本発明のベクタは、通常の技術により宿主生物内へ挿入することが可能である。例えば、ベクタは形質転換、形質移入、電気穿孔、マイクロインジェクション、またはリポソ−ムによる方法(リポフェクション)により挿入することができる。
1.種々のゲノム内での天然部位の発生度
精製したI-Sce I酵素を使用して、天然、または縮重した部位の発生度を、複数の種の完全なゲノムで調べた。サッカロマイセス・セレビシアエ(Saccharomyces cerevisiae)、バシラス・アントラシス(Bacillus anthracis)、ボレリア・ブルグドルフェリ(Borrelia burgdorferi)、レプトスピラ・ビフレキサ(Leptospira biflexa)、及びL.インテロガンス(L. interrogans)では、天然の部位は全く見られなかった。T7ファ−ジDNAで、一つの縮重した部位が見られた。
天然のI-Sce I部位がないのであれば、人工部位を形質転換か形質移入により導入することができる。二つの場合を区別する必要がある:相同性組み換えによる部位特異的組み込みと、非相同的組み換え、トランスポゾン移動、またはレトロウイルス感染によるランダムな組み込みである。最初のものは酵母と、2、3の細菌種では容易であるが、高等な真核細胞ではより困難である。第二のものは全ての系で可能である。
二つのタイプを区別することができる:
−1−I-Sce I部位を選択標識とともに導入する、特異的カセット;
酵母に対して:標識遺伝子を含む以下のもの、全ては、pAF100の誘導体である(Thierry et al., (1990) YEAST 6:521-534):
pAF101:URA3(Hind III部位に挿入);
pAF103:NeoR(BglII部位に挿入);
pAF104:HIS3(BglII部位に挿入);
pAF105:KanR(BglII部位に挿入);

ほ乳類細胞に対して:
pMLV LTR SAPLZ:MLVとPhleo-LacZのLTR内にI-Sce I部位を有する(図13)。このベクタはまずΨ2細胞(3T3の誘導体;R. Muliiganより入手)で増殖させる。ゲノム中の未同定の位置にI-Sce I部位を有する、二つのトランスジェニック細胞株が入手可能である。:1009(多能性神経細胞)、及びD3(トランスジェニック動物を産生できるES細胞)。
真核ゲノムを遺伝的にマッピングするための、入れ子状(nested)染色体断片化方法では、18塩基対の長さの認識のような、制限エンドヌクレア−ゼI-Sce Iのユニ−クな性質をうまく利用している。ほとんどの真核ゲノムには、天然にはI-Sce I認識部位がないこともまた、このマッピング方法ではうまく利用されている。
この方法を、酵母サッカロマイセス・セレビシアエ(Saccharamyces serevisiae)の染色体XIのマッピングに適用した。I-Sce I部位を二倍体株FY1679の染色体XI上の7つの異なる部位に挿入し、その結果該染色体中に8つの物理的間隔を定義した。相同性組み換えにより、URA3-1I-Sce Iカセットより部位を挿入した。二つの部位を、遺伝的に定義されている遺伝子、TIF1とFAS1内に挿入し、その他のものは発明者が有する、オ−バ−ラップしていない5つのコスミドをランダムに選択して、染色体中の未知の位置に挿入した。7つのトランスジェニック株のそれぞれがアガロ−スに包埋したものを次いでI-Sce Iで消化して、パルスフィ−ルドゲル電気泳動で分析した(図14A)。それぞれのトランスジェニック株の染色体XI上でのI-Sce I部位の位置はまず、断片の左右の向きを考慮に入れずに断片サイズより推定される。向きは、以下のようにして決定する。この株のセットのなかで、最もテロメアに近いI-Sce I部位は、トランスジェニックE40の中にあるが、それは全ての断片の中で最も短いものが50kbであるからである(図15A)。よってここで、トランスジェニックE40にI-Sce Iを導入するのに使用したコスミドクロ−ンpUKGO40を、全ての染色体断片に対するプロ−ブとして使用する。予想したとおり、pUKG040はE40株より二つの断片を明るみにだす(それぞれ50kb及び630kb)。大きい断片はほぼ染色体XI全体であり、長さが38kbであるpUKG040は、その大きい染色体断片内に4kb未満のものを含むという事実により、弱いハイブリダイゼーションシグナルを示す。該トランスジェニック株は、二つの相同物のうち一つの中のみにI-Sce I部位が挿入されている二倍体であるために、染色体XI全体は、I-Sce I消化後も視覚的に認識できることに注目されたし。現在では、pUKG040プロ−ブはその他の全てのトランスジェニック株のうちのたった一つの断片にハイブリダイズして、I-Sce I部位の左右の向きを明瞭にする(図15B)。コスミドベクタと、I-Sce I部位を有する染色体サブ断片との間での顕著な交差ハイブリダイゼーションは見られない。トランスジェニック株は現在では、I-Sce I部位が該染色体のハイブリダイズする末端からの距離が短い順に配置されていて(図15C)、I-Sce Iマップが推定できる(図15D)。マッピングの精度はPFGEの分解能と、最適な補正に依存する。遺伝子地図に関しての、染色体の正確な左右の向きは、この段階では分からない。本方法を可視化し、I-Sce I間のI-Sce I部位の間隔サイズのより正確な測定をするために、新たに並べた、同じトランスジェニック株を新規のパルスフィ−ルドゲル電気泳動にかけた(図16)。トランスファ−後、断片をすぐにコスミドpUKG040とpUKG066にハイブリダイズさせたが、これらはそれぞれが該染色体の反対側の末端全ての断片を明るみに出す(クロ−ンpUKG066は、遺伝子地図に定義される染色体の右末端を定義するが、これはそれにはSIR1遺伝子が含まれているからである)。通常の段階的な染色体断片サイズの列が見られる。プロ−ブUKG066と染色体IIIとの間に幾つかの交差ハイブリダイゼーションがあることに注目されたいが、これはおそらく幾つかのDNA繰り返し配列のためである。
この方法をYACマッピングに適用するのには、二つの可能性が考えられる。
−1−酵母内での相同性組み換えを利用して、興味ある遺伝子内にI-Sce I部位を挿入する。これにより、試験管内でのI-Sce I消化によるYAC挿入物内の遺伝子のマッピングが可能になる。これは実際に行なわれ成功した。
−2−高い反復性の配列(例:マウスのB2や、ヒトのAlu)を使用した、酵母内での相同性組み換えにより、YAC挿入物内にI-Sce I部位をランダムに組み込む。トランスジェニック株を次いで、参考文献P1に記載されているようにして、ライブラリ−またはマップ遺伝子を並べる。
入れ子状の、染色体断片は、分離用PFGより精製することができ、染色体XI特異的サブライブラリ−からのクロ−ンに対するプローブとして使用することができる。このサブライブラリ−は138のコスミドクロ−ンよりなっているが(8回カバ−するのに相当する)、これはPFGで精製した染色体XIでコロニ−ハイブリダイゼーションして、出願人の有する完全な酵母ゲノムライブラリ−から、以前に分類しておいたものである。次いで整列させていないクロ−ンのコレクションを、該染色体の左末端からの短い順の染色体断片とハイブリダイズさせた。I-Sce Iマップ上のそれぞれのコスミドの位置はそれぞれのハイブリダイゼーションより明確に決定される。更にこの結果を確認するためと、より正確なマップを提供するため、順番に並べられているコスミドクロ−ン全てのサブセットを、EcoRIで消化して電気泳動にかけ、該染色体の左末端からの短い順番の、入れ子状シリ−ズの染色体断片とハイブリダイズさせた。結果は図17に示してある。
この態様においては、人工的に挿入したI-Sce I部位でのDNAの完全消化の後で、選択した細菌性制限エンドヌクレア−ゼによるパ−シャル消化を行う。制限断片を次いで電気泳動により分離してブロットした。間接的末端標識を、左または右のI-Sce Iの半分の部位を使用して行う。この技術は酵母染色体では成功し、またYACに対しても困難なく適用できるはずである。
1.酵母内でのI-Sce Iの発現。
合成のI-Sce I遺伝子を、マルチコピ−プラスミドであるpPEX7及びpPEX408上の、ガラクト−ス誘導性プロモ−タの制御下に置いた。発現は正常におき、以下に示すような、部位への影響を誘導する。I-Sce Iの合成遺伝子が染色体内の誘導性プロモ−タの制御下に挿入されたトランスジェニック酵母を構築することが可能である。
プラスミドの有するI-Sce I部位への影響:分子内効果は、参考文献18に詳細が記載されている。分子間(プラスミドから染色体)組み換えを予想することができる。
一倍体の細胞では、人工的I-Sce I部位での染色体内単一切断は、細胞増殖の停止とそれに引き続く細胞死を起こす(ほんの数%しか生き延びない)。切断部位に相同性のある無傷の配列が存在すると、修復がおきて、100%細胞が生存する。二倍体細胞では、染色体内の人工的I-Sce I部位の単一の切断は染色体相同部を使用した修飾がおきて、100%細胞が生存する。この両方の場合、誘導された二本鎖切断の修飾により、切断部の脇の非相同性配列の欠失と、ドナ−DNA分子に由来する非相同性配列の挿入を伴った、異型接合性が失われる。
クロ−ニング部位の次にI-Sce I認識部位を有するYACベクタの構築により、挿入部が一部オ−バ−ラップしている場合には、別のYACとの相同性組み換えを誘導することができる。これは、保有物(contigs)の構築にも有益である。
−1−マウス細胞
−a−マウス細胞(Ψ2)を標準的なリン酸カルシウム形質転換法を利用して、I-Sce I部位を有するベクタpMLV LTR SAPLZで形質転換した。
−b−形質転換済み細胞を、フレオマイシン(phleomysin)と5%ウシ胎児血清を含んだDMEM培地で選択し、12%CO2、100%湿度、37℃でコロニ−が形成されるまで増殖させた。
−c−フレオマイシン抵抗性コロニ−を同じ培地で一回サブクロ−ニングした。
−d−1ml当たり105の力価のウイルス粒子を産生するクロ−ンMLOP014を選択した。 このクロ−ンをCNCMに1992年5月5日に培養物コレクションアクセス番号I-1207の下に寄託した。
−e−10%ウシ胎児血清と5mg/mlの「ポリブレイン(polybrain)」とを有するDMEM培地内で105細胞当たり、このクロ−ンの上清を使用した105のウイルス粒子をまいて、他のマウス細胞(1009)を感染させた。培地は新鮮な同じ培地で、感染6時間後に置き換えた。
−f−感染後24時間で、フレオマイシン抵抗性細胞を、上記と同じ培地内で選択した。
−g−フレオマイシン抵抗性のコロニ−を同じ培地でサブクロ−ニングした。
−h−一つのクロ−ンを取り上げて、分析した。DNAを標準的な方法で精製して、最適な条件下でI-Sce Iで消化した。
I-Sce I部位を有するミニTn5トランスポゾンを、標準的なDNA組み換え技術により構築した。ミニTn5トランスポゾンを共役プラスミド(conjugative plasmid)上にのせる。大腸菌(E.coli)とエルシニア(Yersinia)との間の細菌共役を使用して、エルシニア内にミニTn5トランスポゾンを組み込む。カナマイシン、ストレプトマイシン、またはテトラサイクリンに対して抵抗性のあるエルシニア細胞を選択する(それぞれ、ベクタpTKM-ω、pTSM-ω、及びpoTTc-ω)。
−1−I-Sce I認識部位が染色体のユニ−クな位置に挿入されている、トランスジェニックな細胞を構築する。トランスジェニック細胞を、発現ベクタと、興味ある遺伝子、及びI-Sce Iが内部に挿入されている配列に対して相同性な部分を有しているプラスミドとで共形質転換する。
−2−プラスミド上の興味ある遺伝子内、またはその隣への、I-Sce I認識部位の挿入。正常な細胞を、合成I-Sce I遺伝子を有する発現ベクタと、I-Sce I認識部位を有するプラスミドとで共形質転換する。
−3−I-Sce I遺伝子が、ゲノム中の、誘導性、または構成性の細胞性プロモ−タの制御下に組み込まれた、安定なトランスジェニック細胞株の構築。興味ある遺伝子内、またはその隣にI-Sce I部位を有しているプラスミドによる、細胞株の形質転換。
染色体と外来性DNAとの間の相同性組み換え(HR)は、ゲノムへの遺伝的変化を導入する方法の基礎である(5B、20B)。組み換えイベントのパラメ−タは、細胞内へ導入されているプラスミド配列の研究と(1B、4B、10B、12B)、in vitro系での研究により決定された。HRは、哺乳類細胞では非効率的であるが、DNA中の二本鎖切断により促進される。
プラスミドの構築
pG-MPLは4つの工程により得た:
(I)モロニ−マウス白血病ウイルス(MoMuLV)の3’LTRのU3配列中の、NheI部位−XbaI部位間のSAを有するMoMuLVのenv遺伝子の、0.3kbからなるBgl II−SmaI断片(クレノウ酵素で処置済み)の挿入;
(II)リンカ−アダプタ−を有するこの修飾済みLTR中の、SAの隣のXbaI部位における、PhleoLacZ融合遺伝子(15B)(Cayla laboratoryより入手したpUT65由来)を含む、3.5kbのNcoI−XhoI断片の挿入;
(III)p5'LTRプラスミド(XhoIとEcoRIとの間に、MoMuLVの、ヌクレオチド番号563までの5'LTRを有するプラスミド)の、SalIとEcoRIとの二重消化により回収される、この3’LTR(SAとPhleoLacZを有する)の挿入;
(IV)この3’LTR内のNcoI部位(SAとPhleoLacZとの間)への、合成I-Sce I認識部位の挿入。
(I)BamHIとSalIとで開裂させたphCMV1(F. Meyer、個人的に入手)プラスミドへの、I-Sce Iを含有する断片(pSCM525, A. Thierry, 個人的に入手)である0.73kbのBamHI−SalIの挿入;
(II) SV40のポリアデニル化シグナルを有する1.6kbの断片の、phCMV1のPstI部位への挿入。
3T3、PCC7 S、Ψ2は、参考文献(7B)、及び(13B)で参照されている。細胞選択培地:ガンシクロビル(14B、23B)を、組織培養液に2μMの濃度で加えた。ガンシクロビル選択は、6日間、細胞上で行い続けた。G418を適切な培養液に、PCC7-Sに対しては1mg/mlの濃度で、3T3に対しては、400μg/mlの濃度で加えた。選択は、細胞培養中を通して行った。フレオマイシンは10μg/mlの濃度で使用した。
−Ψ細胞株は、I-Sce I認識部位を有するプロウイルス組み換えベクタを含んでいるプラスミドで形質転換した:pG-MPL、pG-MtkPL、pG-MtkΔPAPL。
G-MPL 複数のクロ−ン(30以上)が回収された。1から14のプロウイルス組み込み、及び複数の異なる組み込み点が、分子的解析により確認された。
−胚性幹細胞株D3をG-MPLで感染させる:4クロ−ンを回収(3つは正常なプロウイルスの組み込みが起きていて、一つは4つのプロウイルス組み込みが起きていた)。
レトロウイルスの組み込み(プロウイルス組み込み)により、I-Sce I含有LTRの複製がおきる。細胞は該部位に関してヘテロ接合である。
これらの方法は、2B、及び3Bに記載のようにして行った。
I-Sce IのHRを検出するために、図20に示した実験系を設計した。欠損した組み換えレトロウイルス(24B)は、I-Sce I認識部位とPhloeLacZ(15B)誘導遺伝子を、3'LTR内に挿入して構築した(図20a)。レトロウイルスの組み込みにより、細胞ゲノム内に、互いに5.8kb、または7.2kb離れた二つのI-Sce I部位の組み込みがおきた(図20b)。これらの部位でのI-Sce I誘導性二本鎖切断(DSB)(図20c)により、DSBの脇の領域との相同性配列を有するドナ−プラスミド(pSVneo, 図20d)とのHRが開始されることと、また非相同性配列でドナ−プラスミドが有するものは、この組み換え中に複製される(図20e)可能性とを推定した。
より特には、二つのプロウイルス配列を本研究で使用した。G-MtkPLプロウイルス配列(G-MtkPLウイルス由来)には、形質移入済み細胞の(フレオマイシン含有培地内での)陽性選択用のPheleoLacZ誘導遺伝子と、(ガンシクロビル含有培地内での)陰性選択用のtk遺伝子が含まれている。G-MPLプロウイルス(G-MPLウイルス由来)はPhleoLacZ配列のみが含まれている。G-MtkPL及びG-MPLはエンハンサ非含有モロニ−マウス白血病プロウイルスより構築された欠損型組み換えウイルス(16B)である。該ウイルスベクタは、プロモ−タトラップとして機能し、故に細胞のフランキング配列により活性化される。
G-MtkPLウイルスによりNIH3T3に与えられた表現型は、PhleoR β-gal+ glsSであり、G-MLPによりPCC7-Sに与えられた表現型は、PhleoR、βgal+である(図20b)。I-Sce Iにより誘導される組み換えイベントの直接的選択を可能にするため、pVRneoドナ−プラスミドを構築した。pVRneoにおいては、neo遺伝子は、左側の染色体切断に対して5'である配列に相同性である300bpと、右側の切断に対して3’である配列に対して相同性である2.5kbとで挟まれている(図20d)。ポリアデニレ−ションシグナルはneo遺伝子に対して3’に配置して、組み換え後にPhleoLacZ伝令を阻害するようにした。プロウイルスとプラスミドとの間の誘導性組み換えが起こると、得られる表現型はneoRであり、ドナ−プラスミド内のポリアデニレ−ションシグナルの存在のため、PhleoLacZ遺伝子は発現しないはずであり、phleoSβ-gal―表現型になる。
neoR組換え体の分子的構造をサザンブロット分析で調べた(図23、及び表1)。I-Sce I部位でのHRからは、組み換えDNAは、4.2kbの親断片の代わりに6.4kbのLacZ断片が産生されることが予想される。NIH3T3細胞からのneoR glsR β-gal―の15の組換え体全ては6.4kbのKpnI断片のみを示した。よって、二重の選択方法により、予想した遺伝子置換(二重部位相同性組み換え:DsHR)による組み換え体のみ産生された。
本願で示された結果は、二本鎖切断が、サッカロマイセス・セレビシアエ(Saccharomyces cerebisiae)のI-Sce I系により、ほ乳類細胞中で誘導されることと、標的染色体配列中の切断は、使用したドナ−プラスミドDNAとの部位特異的組み換えを誘導することを証明している。哺乳類細胞内で操作するために、この系においては、内在性のI-Sce I様活性が哺乳類細胞内ではないことと、I-Sce Iタンパク質が哺乳類細胞内で中性であることが必要である。I-Sce I認識部位の導入は、インプットしたDNA配列内での再構成や変異を起こさないようであるので、内在性のI-Sce I様活性は、哺乳類細胞内で作用しないであろう。例えば、I-Sce I認識部位を有するレトロウイルスで感染させたNIH3T3及びPCC7-Sクロ−ンの全ては、安定にウイルスを増殖させた。I-Sce I遺伝子産物の毒性を試験するために、I-Sce I発現プラスミドをNIH3T3細胞株へ導入した(デ−タ非掲載)。機能性のI-Sce I遺伝子のコ・トランスファ−が、高い割合で起きることが判明し、これは該遺伝子に関しての選択が全く起きていないことを示唆している。I-Sce I遺伝子の機能性は、遺伝子産物と生物学的機能の転写分析、免疫蛍光検出で証明した(Choulika et al., 投稿準備中)。
この例は、I-Sce Iメガヌクレア−ゼ(サッカロマイセス・セレビシアエ(Saccharomyces cerevisiae)のミトコンドリアのイントロンホ−ミングに関わる)(6B、28B)を使用して、DSBを誘導して哺乳類細胞での組み換えを仲介することを記載している。I-Sce Iは、18bpの長さの認識部位を有する(29B、22B)、非常にまれに切断する制限エンドヌクレア−ゼである。in vivoでは、I-Sce Iエンドヌクレア−ゼは、特異的DSBを開始して細胞によるギャップ修復を導くことにより、修飾した酵母の核での組み換えを誘導することができる(30B、17B、21B)。よってこの方法は、生細胞中での染色体の操作を目指して、特異的DSBを染色体標的DNAに導入する方法として使用することが潜在的には可能である。I-Sce I仲介性組み換えは、染色体工学に関してリコンビナ−ゼ系[11]よりも優れているが、それは後者では宿主とドナ−のDNA分子の両方に標的部位が存在する必要があり、可逆的な反応に至るからである。
プラスミドの構築
pG-MtkPL は、5つの工程により得た:(I)NheIとXbaI部位(クレノウ酵素で処理済み)との間にスプライシングの受容部(SA)を有している、モロニ−マウス白血病ウイルス(MoMuLV)のenv遺伝子の、0.3kbのBgl II-SmaI断片(クレノウ酵素で処理済み)を、中間体プラスミド中にある、MoMuLVの3’LTRのU3配列中に挿入する;(II)この修飾済みLTR内に、PhleoLacZ融合遺伝子([13], pUT65由来;Cayla Loratory, Zone Commercial du Gros , Toulouse, France)を含む3.5kbのNcoI-XhoI断片を、SAの隣のXbaI部位で挿入する;(III)Sal I-EcoRIの二重消化で回収したこの3’LTR(SAとPhleoLacZとを含む)を、p5'LTRプラスミド(MoMuLVのヌクレオチド番号563までの5'LTRを含むプラスミド)のXhoI部位とEcoRI部位との間に挿入する;(IV)合成I-Sce I認識部位を、3'LTR内のNcoI部位へ挿入する;(V)pG-MPLのPstI部位に、リンカ−アダプタ−を有している1.6kbのtk遺伝子を挿入する(レトロウイルスゲノムとはアンチセンスで)。
T3及びΨ2は、参考文献(7B)と(13B)のものである。細胞選択培地:ガンシクロビル(14B、23B)を2μMの濃度で組織培養培地へ加えた。ガンシクロビル選択を6日間続けた。フレオマイシンを10μg/mlの濃度で使用した。同じ条件で二重の選択を行った。
これらのプロトコ−ルは、記載されているものである(2B、3B)。
ウイルス産生細胞株は、pG-MtkPLをΨ2パッケ−ジング細胞株へ形質導入して得られる。ウイルスは、形質転換済みΨ2細胞株の培養液の濾過により調製され、クロ−ンはフレオマイシン含有培地で単離した。
哺乳類細胞でのI-Sce Iエンドヌクレア−ゼ活性をアッセイするために、G-MtkPLプロウイルスを含むNIH3T3細胞を使用した。G-MtkPLプロウイルス(図25a)は、ガンシクロビル含有培地での陰性選択用のtk遺伝子を含み、また二つのLTR内にI-Sce I認識部位とPhleoLacZ融合遺伝子とを含む。このPhleoLacZ遺伝子はフレオマイシン含有培地での、形質導入済み細胞の陽性選択に使用することが可能である。これらの細胞でのI-Sce Iエンドヌクレア−ゼの発現は、以下の機構(図25に例示されている)のうちの一つにより修復されるI-Sce I認識部位での二本鎖切断(DSB)を誘導すると仮定した:
(a)仮にI-Sce Iエンドヌクレア−ゼが二つのLTR(図1−bの1と2)のうちの一つのみで切断を誘導するならば、二つのLTRの間で相同性である配列は対を形成し、組み換えを起こして染色体内相同性組み換え(すなわち一本鎖アニ−リング(SSA )(12B、10B)または交差)を起こす;
(b)仮にI-Sce Iエンドヌクレア−ゼが、二つのLTRのそれぞれにおいて切断を誘導するならば、二つのフリ−な末端は再連結することが可能であり(末端結合機構(31B))染色体内組み換え(図25−b 3)がおきる;または、
(c)二つのDSBにより作られるギャップは、相同性染色体かその他の染色体断片上の配列を使用したギャップ修復機構により修復されて、プロウイルス配列の喪失がおきる(32B)(図25−c)。
ここに示した結果は、酵母のI-Sce Iエンドヌクレア−ゼが、哺乳類細胞中で染色体組み換えを誘導することを証明している。これは、I-Sce Iがin vivoにおいて、予め決めておいた標的で染色体を切断することが可能であることを強く示唆している。
ドナ−ベクタの多様性:
ドナ−プラスミド中のI-Sce I部位のフランキング領域のサイズと配列(左側300bpと、右側2.5kbpに関して):I-Sce I部位の左右のフランキング領域が最高で、合計11kbまでの種々のサイズである、異なる構築物が存在する。配列は、構築したものに依存している(LTR、遺伝子)。300bpから11kbの間の、如何なる配列も使用可能である。
I-Sce I部位を有する組み換えのレトロウイルス(すなわち、pG-MPL, pG-MtkPL, pG-MtkΔPAPL)は、種々のパッケ−ジング細胞株(両種向性、または異種栄養性)で産生させてもよい。
I-Sce I部位の挿入は、所望の遺伝子座、及び適切な位置での、古典的な遺伝子置換により行う。次いで、細胞中(人工的に挿入したI-Sce I部位へのドナ−遺伝子の挿入)、またはトランスジェニック動物中の同じ位置での異なる遺伝子の発現を、スクリ−ニングすることが可能である。複数の薬剤、リガンド、医用タンパク質等の効果は、組織特異的な方法で試験することが可能である。遺伝子は一貫して、染色体内の同じ位置に挿入されるであろう。
・I-Sce I部位を有するベクタ(種々のドナ−プラスミド):該遺伝子内(またはその脇)に一つの部位、あるいはドナ−遺伝子の脇に二つの部位。
一過性発現:同じプラスミド上、または別のもの(コ・トランスフェクション)の上のORF。
高力価である、種々の感染性レトロウイルス粒子を産生することが可能な細胞株;
I-Sce I部位、レポ−タ−セレクタ−遺伝子、活性LTR、及び他のレトロウイルスの必須配列を有する、欠失レトロウイルスを有するプラスミド;上記の改変したレトロウイルス中のI-Sce I部位のフランキング領域に対して相同性な配列を含み、またマルチプルなクロ−ニング部位を含む、プラスミド;並びに、I-Sce Iエンドヌクレア−ゼの発現を可能にし、特異的適用に順応させたベクタ。
1.G-MPLのプロウイルス組み込みを、1から14個有している、マウス線維芽細胞株、NIH3T3。複数のクロ−ン(30よりも多い)を回収した。個となるゲノム組み込み(未解析)の存在と多重度を分子的分析により確認した。
1.部位特異的遺伝子挿入
I-Sce I部位を前もって組み込んでおくことにより、予め決めた位置に、種々の遺伝子若しくはある遺伝子の変異体が挿入されている、限定されない数の細胞株を産生することが、この方法により可能になる。よってこのような細胞株は、表現型、リガンド、薬剤のスクリ−ニングに対して有益であり、また、該細胞株がレトロウイルス産生に関してのトランス相補細胞株であるならば、組み換えレトロウイルスベクタの発現を、非常に高レベルで行うのに有益である。
同様の細胞株と、導入遺伝子、種々の、プロモ−タ、制御因子、及び/または構造遺伝子とを使用して、タンパク質、代謝物、またはその他の、生物学的もしくは生物工学的に興味ある化合物を産生することが可能である。遺伝子は常に、染色体内の同じ位置に挿入される。トランスジェニック動物においては、複数の薬剤、リガンド、または医用タンパク質の効果を組織特異的方法により試験することを可能にする。
A.遺伝子治療では、患者由来の細胞をI-Sce I含有レトロウイルスで感染させ、欠失レトロウイルスの組み込みでスクリ−ニングし、次いでI-Sce I産生ベクタとドナ−配列とによる共形質転換が可能である。
I-Sce I部位を遺伝子座に導入し(外来配列とともに、または外来配列なしで)、細胞内で異型接合型を作りだす。修復DNAがないと、誘導される二本鎖切断部は、非特異的エクソヌクレア−ゼにより伸長されて、娘染色分体の無傷の配列によりギャップ修復され、よって、該細胞はこの遺伝子座に関して同型接合型になる。
染色体の特異的転座、または欠失が、I-Sce I開裂により誘導される。遺伝子座挿入は、「古典的な遺伝子置換」により、染色体中の特異的な位置に一つ組み込むことにより得られる。認識配列の、I-Sce Iエンドヌクレア−ゼによる開裂は、非致命的転座、または欠失とその後の末端連結により修復可能である。染色体断片の欠失はまた、二つ以上のI-Sce-I部位を遺伝子座のフランキング領域内に挿入することにより得られる(図32を参照)。開裂は、組み換えにより修復することが可能であり、上記の二つの部位の間の完全な領域が欠失する(図32を参照)。

Claims (9)
- 動物または植物の細胞中の染色体DNAに部位特異的に、二本鎖の切れ目を少なくとも一つ誘導する方法によって得られる組み換え真核細胞であって、前記方法は:
(a)少なくとも一つのI-SceI制限部位を具備した二本鎖染色体DNAを有する細胞を用意すること;
(b)メガヌクレア−ゼ(meganuclease)I-SceIをコードするDNAを具備した、少なくとも一つのプラスミドで前記細胞を形質転換すること;および、
(c)少なくとも一つの、二本鎖の切れ目が誘導された細胞を選択すること、
を含む組み換え真核細胞。 - 少なくとも一つの、部位特異的な切れ目を動物または植物の細胞の染色体DNAへ誘導し、ポリペプチドをコードするDNAを挿入する方法によって得られる組み換え真核細胞であって、前記方法は;
(a)二本鎖DNAを有する動物または植物の細胞を用意して、I-SceI制限部位を具備したDNAで該細胞を形質転換すること;
(b)I-Sce-I酵素を加えるか、またはI-Sce-I酵素をコードするDNAで前記細胞を形質転換すること;
(c)前記ポリペプチドをコードするDNA、または前記DNAを含むベクタで、前記細胞を形質転換すること;および、
(d)前記ポリペプチドをコードする前記DNAまたは前記ベクタが染色体DNAに挿入されている細胞であって、前記ポリペプチドを発現する細胞を選択すること、

- ポリペプチドを発現する組換え幹細胞であって、前記幹細胞は、以下の工程によって得られる組換え幹細胞:
a.I-SceI制限部位を具備したDNAによって形質転換された細胞である幹細胞を用意することと、
b.前記細胞へI-SceI酵素を加えるか、または前記細胞を、I-SceI酵素をコードする遺伝子を有するベクタで形質転換することと、
c.前記細胞を前記ポリペプチドをコードする前記DNAで形質転換することと、
d.前記ポリペプチドをコードする前記DNAが、前記細胞の前記染色体DNAに挿入された細胞であって、前記ポリペプチドを発現する細胞を選択すること。 - 少なくとも一つのタンパク質産物を発現する動物または植物の細胞の染色体DNA中に少なくとも一つの、部位特異的な切れ目を誘導し、ポリペプチドをコードするDNAを挿入する方法によって形質転換された組換え細胞であって、前記方法は;
(a)二本鎖DNAを有する動物または植物の細胞を用意して、I-SceI制限部位を具備したDNAで形質転換すること;
(b)前記細胞へI-Sce-I酵素を加えるか、またはI-Sce-I酵素をコードしたDNAで該細胞を形質転換すること;
(c)前記ポリペプチドをコードした前記DNA、または該DNAを有するベクタで前記細胞を形質転換すること;および、
(d)前記ポリペプチドをコードする前記DNAまたは前記ベクタが前記細胞の前記染色体DNAに挿入されている細胞であって、前記細胞は前記ポリペプチドを発現し、かつ前記タンパク質産物を発現しない細胞を選択すること、
を含む組み換え細胞。
