Abstract
Description
本発明は、生化学、分子生物学、構造生物学および医学の分野に関する。更に詳細には、本発明は、交差−β構造、それらの結合タンパク質、およびそれらの生物学的役割に関する。
増加しつつある一連の証拠は、球状タンパク質の変性により毒性を生じる可能性があることを示唆している。l変性タンパク質は、部分構造化コンホメーションを採用することによってタンパク質凝集とフィブリル化を開始することができる。このようなフィブリル状凝集体は、様々な種類の組織に(ゆっくりと)蓄積し、様々な退行性疾患に関連している。「アミロイド」という用語は、これらのフィブリル状沈着物を記載するのに用いられる。アミロイドを特徴とする疾患はアミロイドーシスと呼ばれ、アルツハイマー病(AD)、軽鎖アミロイドーシス、糖尿病II型、および海綿状脳障害が挙げられる。最近、毒性がミスフォールドタンパク質の固有の特性であることが見出された。本発明によれば、これらのコンホメーション病(conformational disease)についてのこの共通の機構1に関連する。
1) 交差−β構造の存在を検出、
2) アミロイドフィブリルの形成を抑制、
3) 交差−β構造によって誘発される毒性を調節、
4) 循環から交差−β構造含有分子を除去
するための化合物および方法を提供する。
試薬
ウシ血清アルブミン(BSA)画分V pH7.0およびD−グルコース−6−リン酸二ナトリウム(g6p)、D,L−グリセルアルデヒド、およびニワトリ卵白リゾチームはICN製であった(Aurora, Ohio, 米国)。ウサギ抗−組換え組織型プラスミノーゲンアクチベーター(tPA) 385Rおよびマウス抗−組換えtPA 374BはAmerican Diagnostica (Veenendaal, オランダ)から購入した。抗−ラミニン(L9393)はSigma製であった。ブタ抗−ウサギ免疫グロブリン/HRP(SWARPO)およびウサギ抗−マウス免疫グロブリン/HRP(RAMPO)はDAKO Diagnostics B.V.(オランダ)製であった。Alteplase(組換え組織型プラスミノーゲンアクチベーター,tPA)はBoehringer-Ingelheim (ドイツ)から入手した。クリングル2と触媒ドメインだけを含む組換え突然変異体tPA(K2P−tPA)であるReteplase(Rapilysin)は、Roche, Hertfordshire,英国から入手し、ブタ膵臓カルボキシペプチダーゼB(CpB)はRoche, Mannheim, ドイツ製であった。カルボキシペプチダーゼインヒビター(CPI)はCalbiochem(La Jolla, CA, 米国)製であった。Tween20は、Merck-Schuchardt(Hohenbrunn, ドイツ)から購入した。コンゴレッドはAldrich(Milwaukee, WI, 米国)製であった。Thioflavin Tおよび凍結乾燥したヒトヘモグロビン(Hb)はSigma (St. Louis, MO, 米国)製であった。凍結乾燥したヒトフィブリノーゲンはKordia (Leiden, オランダ)製であった。発色性プラスミン基質S−2251はChromogenix (Milan, イタリア)から購入した。オリゴヌクレオチドは、Sigma-Genosys (英国)から購入した。ホウ素ガラス毛細管(0.5 mm φ)は、Mueller(Berlin, ドイツ)製であった。
上記のヒトアルツハイマーペプチドに存在するアミノ酸を含むペプチドAβ(1−40)、(DAEFRHDSGYEVHHQKLVFFAEDVGSNKGAIIGLMVGGVV)、フィブリンペプチド85(またはFP13)(KRLEVDIDIKIRS)、86(またはFP12)(KRLEVDIDIKIR)および87(またはFP10)(KRLEVDIDIK)、ヒトフィブリン(フィブリノーゲン)に由来、およびランゲルハンス島アミロイドポリペプチド(IAPP)ペプチドまたは誘導体(fl−hIAPP: KCNTATCATQRLANFLVHSSNNFGAILSSTNVGSNTY,ΔhIAPP(SNNFGAILSS)、ΔmIAPP(SNNLGPVLPP)は、Pepscan, Inc.(オランダ)またはthe Netherlands Cancer Institute (NCI, Amsterdam, オランダ)のペプチド合成施設から入手した。ペプチドをリン酸緩衝食塩水(PBS)に溶解して1mg/mlの最終濃度とし、室温(RT)で3週間保管し、フィブリルを形成させた。この期間中に、懸濁液を毎週2回攪拌した。3週間後、懸濁液を4℃で保管した。NCI製の凍結乾燥したAβ(1−40)を、同様の方法で交差−β構造を形成させた。交差−β構造の形成を適時にコンゴレッド結合および偏光での緑色複屈折の検査によって追跡した。
Thioflavin T蛍光測定のため、1mg/mlのフィブリノーゲンを2U/mlのIIa因子と共に150mM NaCl、20mMトリス−HCl pH7.5、10mM CaCl2、50μMチオフラビンT中で37℃でインキュベーションした。血餅のバックグラウンド蛍光をThioflavin Tの非存在下で記録し、Thioflavin Tのバックグラウンド蛍光をIIa因子の非存在下で測定した。蛍光は、Sarstedt REF67.754キュベットを用いてHitachi F-4500蛍光分光光度計(Ltd.,Tokyo,日本)で測定した。装置の設定値: 励起435nm(スリット幅10nm)、発光485nm(スリット幅10nm)、PMT電圧950V、測定時間10”、遅延0”。コンゴレッド結合の検出のため、フィブリン血餅を上記のようにして室温で形成した(Thioflavin Tは、緩衝液では省いた)。血餅をコンゴレッド溶液と共にインキュベーションして、製造業者の勧める方法に従って洗浄した(Sigma Diagnostics,MO,米国)。血餅を、偏光下で分析した。
高次グリケーション最終生成物で修飾したウシ血清アルブミン(アルブミン−g6p)の調製のため、アルブミン100mg/mlを1M g6pおよび0.05%m/v NaN3を含むPBSと共に37℃にて暗所でインキュベーションした。1つのアルブミン溶液を2週間グリケーションを行い、別のバッチのアルブミンは4週間グリケーションを行った。グリケーションは、残りのバッチの一部では23週間まで延長した。ヒトHb 5mg/mlを、1M g6pおよび0.05%m/v NaN3を含むPBSと共に37℃にて10週間インキュベーションした。更に、50mg/mlのHb溶液を、同じ緩衝液で8週間インキュベーションした。グリセルアルデヒドで修飾したアルブミン(アルブミン−グリセルアルデヒド)、およびニワトリ卵白リゾチーム(リゾチーム−グリセルアルデヒド)の濾過殺菌したタンパク質溶液15mg/mlを、10mMグリセルアルデヒドを含むPBSと共に2週間インキュベーションした。コントロールでは、g6pまたはグリセルアルデヒドを、溶液で省いた。インキュベーションの後、アルブミンおよびリゾチーム溶液を蒸留水に対して十分に透析した後、−20℃で保管した。タンパク質濃度を、高次タンパク質分析試薬ADV01(Cytoskeleton, Denver,CO,米国)を用いて決定した。グリケーションは、高次グリケーション最終生成物からの固有蛍光シグナルを励起波長380nm、発光波長435nmで測定することによって確かめた。
アルブミン-AGEの調製のため、100mg/mlウシ血清アルブミン(画分V,カタログ番号;A−7906,熱ショックによる初期分画化,純度:98%(電気泳動),残りのほとんどはグロブリン, Sigma-Aldrich, St. Louis, MO, 米国)を、リン酸緩衝食塩水(PBS、140mM塩化ナトリウム、2.7mM塩化カリウム、10mMリン酸水素二ナトリウム、1.8mMリン酸二水素カリウム、pH7.3)、1M D−グルコース−6−リン酸二ナトリウム塩水和物(乾燥)(ICN, Aurora, Ohio,米国)、および0.05%(m/v)NaN3と共に37℃で暗所にてインキュベーションした。ウシアルブミンは、83個の潜在的なグリケーション部位を有する(59個のリシン、および23アルギニン残基、N末端).アルブミンを、2週間(アルブミン−AGE:2)、4週間(アルブミン−AGE:4)、または23週間(アルブミン−AGE:23)グリケーションした。コントロールでは、g6pを省いた。インキュベーションの後、溶液を蒸留水に対して十分に透析した後、4℃で保管した。タンパク質濃度は、高次タンパク質分析試薬ADV01(Cytoskeleton,CO,米国)で決定した。あるいは、アルブミンを、1M g6p、250mM DL−グリセルアルデヒド(ICN, Aurora,Ohio,米国)/100mM NaCNBE3、1Mβ−D−(−)−フルクトース(ICN,Aurora,Ohio,米国)、1M D(+)−グルコース(BDH, Poole,英国)、500mMグリオキシル酸一水和物(ICN, Aurora,Ohio,米国)/100mM NaCNBH3、および相当するPBSおよびPBS/NaCNBH3緩衝液コントロールと共に86週間インキュベーションした。グリケーションは、(i)濃い褐色染色の観察、(ii)SDS−ポリアクリルアミドゲル上のマルチマーの存在、(iii)AGE特異抗体moab抗−アルブミン−g6p 4B546とpoab抗フィブロネクチン−g6p(Ph. De Groot/I. Bobbink, UMC Utrecht;未公表データ)の結合の分析、および(iv)AGE由来の固有蛍光シグナルの測定(励起波長380nm、発光波長445nm)によって確かめた。アルブミンコントロールの自己蛍光シグナルはアルブミン−AGEについて測定したシグナルの4%未満であり、バックグラウンド補正に用いた。
ヒトHbは、3名の健常人と16名の糖尿病患者のEDTAで血液凝固防止した血液の赤血球から単離した。全血100μlを生理食塩水(154mM NaCl)5mlで希釈し、細胞を緩やかに分離し、生理塩5mlに再懸濁した。室温で16時間インキュベーションした後、細胞を再度分離した。ペレット状細胞に0.1Mホウ酸,pH6.5,2mlを加えてリーシスした後、細胞細片を分離した。上清を集めて、−20℃で保管した。
健常人ドナーまたは糖尿病患者のEDTA−血液中のグリコヘモグロビンまたはHbAlcとも呼ばれるグリコHbの濃度を、溶血した全血を用いる比濁分析的阻害イムノアッセイを用いて、製造業者の推奨に従って(Roche Diagnostics, Mannheim, ドイツ)決定した。標準偏差は、測定したHbAlc濃度の2.3%である。
コンゴレッドの、炭水化物であるグルコース、フルクトースおよびグルコース−6−リン酸、または炭水化物誘導体であるグリセルアルデヒドおよびグリオキシル酸を用いて86週間グリコアルブミン−AGEへの結合を、風乾試料を用いて試験した。このため、5μgアルブミンをガラスカバースリップに塗布して、風乾した。試料をコンゴレッドとインキュベーションした後、製造業者の推奨に従って洗浄した(Sigma Diagnostics, St Louis, MO,米国)。それぞれ596nmおよび620nmの励起および発光波長を用い、Leica DMIRBE蛍光顕微鏡(Rijswijk, オランダ)で撮影を行った。
エンドスタチンは、本質的に文献記載の方法でEscherichia coliから精製した。47簡単に説明すれば、エンドスタチンを発現するB121.DE3細菌を8M尿素、10mMトリス(pH8.0)、10mMイミダゾールおよび10mMβ−メルカプト−エタノールを含む緩衝液でリーシスした。Ni2+−アガロース上で精製した後、タンパク質試料をH2Oに対して十分に透析した。透析中に、エンドスタチンは微細な白色固形物として沈澱する。この材料の一部を後で使用するために−80℃で保管するか、または凍結乾燥の後に保管した。酵母株Pichia pastorisで産生した可溶性エンドスタチンは、Dr. Kim Lee Sim(EntreMed, Inc., Rockville, MA)の厚意により提供された。エンドスタチン凝集体は、下記のようにして可溶性エンドスタチンから調製した。可溶性の酵母エンドスタチンを8M尿素中で一晩透析した後、H2Oに対して3回透析を行った。細菌性エンドスタチンと同様に、酵母エンドスタチンは、微細な白色固形物として沈澱する。
凍結乾燥した細菌性エンドスタチンを0.1%ギ酸(FA)またはジメチルスルホキシドに再懸濁し、ガラス毛細管に集めた。溶媒を蒸発させ、生成するエンドスタチン材料を製造業者のプロトコールに従ってコンゴレッド(Sigma Diagnostics, St. Louis, MO, 米国)で染色した。
ペプチドおよびタンパク質溶液(100μg/ml、H2O中)のUV円偏光二色性(CD)スペクトルを、JASCO J−810 CD分光偏光計(Tokyo,日本)で測定した。フィブリンペプチド85、86、87、またはアルブミン、グリコアルブミンおよびヒトAp(16−22)について、それぞれ190−240nmまたは190−250nmの5または10回の単回測定の平均吸収スペクトルを測定する。Aβ(16−22)のCDスペクトルを、ポジティブコントロールとして測定した。Aβ(16−22)をH2O45中でインキュベーションすると、容易に交差−β構造を有するアミロイドフィブリルコンホメーションを採用する。存在する二次構造要素のアルブミンおよびAp(16−22)の相対比率は、k2dソフトウェア48を用いて推計した。
エンドスタチン凝集体を0.1%FA中で可溶化し、凍結乾燥したフィブリンペプチドをH2Oに溶解し、グリコアルブミンを水に対して十分に透析した。試料をガラス毛細血管に集めた。次に、溶媒を数日間かけて蒸発させた。乾燥試料を含む毛細管を、Nonius・カッパー・CCD回折計(Bruker-Nonius, Delft, オランダ)に入れた。散乱を、CCD領域検出器上で黒鉛モノクロメーターで密封チューブMoKα線を用いて16時間測定した。空気およびガラス毛細管壁からの散乱を、社内ソフトウェアを用いて差し引いた(VIEW/EVAL, Dept. of Crystal- and Structural Chemistry, Utrecht University, オランダ)。
エンドスタチン、ヘモグロビンおよびアルブミン試料を、炭素コーティングしたコロジオンフィルムで被覆した400メッシュ標本グリッドに塗布した。5分後、液滴を濾紙で除去し、調製物を1%メチルセルロースおよび1%酢酸ウラシルで染色した。H2Oで洗浄した後、試料を一連の等級付けしたのEtOHとヘキサメチルジシラザンで脱水した。透過型電子顕微鏡法(TEM)画像を、JEM−1200EX電子顕微鏡(JEOL,日本)上で60kVで記録した。
tPAのアルブミン−g6p(4週間および23週間インキュベーション)、アルブミン−グリセルアルデヒド、コントロールアルブミン、ヒトHb−g6p(10週間インキュベーション)、Hbコントロール、またはAβ(1−40)への結合を、固相酵素免疫検定法(ELISA)の装備を用いて試験した。アルブミン−g6pおよびコントロールアルブミン(2.5μg/ml、コート緩衝液中、50mM Na2CO3/NaHCO3 pH9.6、0.02%m/v NaN3、50μl/ウェル)を、96穴タンパク質イモビライザー(Immobilizer)プレート(Exiqon, Vedbaek, デンマーク)に室温で1時間固定した。Aβ(l−40)(10μg/ml、コート緩衝液中)を、96穴タンパク質イモビライザープレート(Exiqon, Vedbaek, デンマーク)に室温で75分間固定した。コントロールウェルを、コート緩衝液のみと共にインキュベーションした。PBS/0.1%v/v Tween20 200μlを用いる線上段階の後、プレートを振盪しながらPBS/1%v/v Tween20 300μlで室温にて2時間ブロックした。その後の総てのインキュベーションは、振盪を行いながら、PBS/0.1%v/v Tween20中で、ウェル当たり50μlの容量を用いて室温にて1時間行った。それぞれのインキュベーションの後、ウェルをPBS/0.1%v/v Tween20 200μlで5回洗浄した。fl.tPAまたはK2−P tPAの増加量を、コーティングを施したウェルおよびコントロールウェルに3回加えた。抗体385Rおよび続いてSWARPO、または抗体374Bおよび続いてRAMPOを1μg/mlの濃度でウェルに加えた。結合したペルオキシダーゼを標識した抗体を、オルトフェニレンジアミン8mgおよび0.0175%v/v H2O2を50mMクエン酸/100mM Na2HPO4 pH5.0 20ml中に含む溶液100μlを用いて可視化した。染色は、2M H2SO4溶液50μlを加えて停止した。吸光度を、Vmax動力学的マイクロプレートリーダー(Molecular Devices, Sunnyvale, CA, 米国)上で490nmで読み取った。競合実験は、tPA 20または40nMを用いて、それぞれアルブミン−g6pまたはAβ(1−40)およびコンゴレッドをPBS/0.08%v/v Tween20/2%v/v EtOHに溶解したものの増加量を用いて行った。
交差−β構造マーカーtPAのアルブミン−AGEへの結合を、ELISA装置を用いて試験した。本発明者らは、tPAがプロトタイプアミロイドペプチドヒトAβ(l−40)およびヒトIAPP49(本明細書)へ結合することを示した。従って、本発明者らは、これら2種類のペプチドへ結合するtPAをポジティブコントロールとして用いた。86週間のグリケーション試料およびコントロールを、Greinerマイクロロン・プレート(microlon plates)(カタログ番号655092, Greiner, Frickenhausen, ドイツ)にコーティングした。ウェルは、Superblock(Pierce, Rockford, IL, 米国)でブロックした。それに続く総てのインキュベーションは、PBS/0.1%(v/v) Tween20中で、50μl/ウェルの容量で、振盪を行いながら室温で1時間行った。インキュベーションの後、ウェルを300μlのPBS/0.1%(v/v) Tween20で5回洗浄した。増加濃度のtPAを、コーティングしたウェル並びにコントロールウェルに3回加えた。86週間インキュベーションした試料のtPAインキュベーション中に、少なくとも123,000倍モル過剰量のε−アミノカプロン酸(εACA,10mM)を溶液に加えた。εACAはリシン類似物であり、tPAのそのクリングル2ドメイン50を介するアルブミンへの潜在的結合を回避する目的で用いられる。モノクローナル抗体374b(American Diagnostica, Instrumentation laboratory, Breda, オランダ)、および続いてRAMPO(Dako diagnostics, Glostrup, デンマーク)を、0.3μg/mlの濃度でウェルに加えた。結合したペルオキシダーゼ標識抗体を、50mMクエン酸/0.0175%(v/v)H2O2を含む100mM Na2HPO4 pH5.0 20mlに8mgのオルトフェニレンジアミンを含む溶液100μlを用いて可視化した。染色は、2M H2SO4溶液50μlを加えて停止した。吸光度をVmax動力学的マイクロプレートリーダー(Molecular Devices, Sunnyvale, CA, 米国)上で490nmで読み取った。コーティングを行っていないコントロールウェルからのバックグラウンドシグナルを、相当するコーティングを施したウェルから差し引いた。
他の実験の蛍光測定では、アルブミン−g6p:2、アルブミン−g6p:4、アルブミン−g6p:23およびコントロールを50mMグリシン−NaOH,pH9に溶解したものを、アミロイド交差−β構造のまま51である増加量のThT(Sigma-Aldrich Chemie, Steinheim, ドイツ)とインキュベーションした。アルブミン− AGE:4濃度は175nMであり、他のタンパク質濃度は500nMであった。86週間のグリケーション試料の蛍光測定のため、140nMのタンパク質を固定濃度の20μM ThTと共にインキュベーションした。蛍光は、室温で1時間インキュベーションした後、Hitachi F-4500蛍光分光光度計(Ltd., Tokyo,日本)上で3回測定した。励起および発光治療は、それぞれ435nm(スリット幅10nm)および485nm(スリット幅10nm)であった。緩衝液およびThTなしの溶液からのバックグラウンドシグナルを、ThTとインキュベーションしたタンパク質溶液での相当する測定値から差し引いた。
430nMアルブミン−g6pおよび19μMチオフラビンTを、増加濃度のtPAと共に室温で1時間インキュベーションした。ブランクの読みには、アルブミン−g6pを省いた。試料は、50mMグリシン−NaOH pH9中で4回調製した。発光測定は、上記の通りに行った。
アルブミン−g6p(500nM)およびチオフラビンT(10μM)を、増加量のtPAと共に50mMグリシン−NaOH pH9中で室温にて1時間インキュベーションした。吸光度測定は、420nmのアルブミン−g6pチオフラビンT吸光度最大値で行った。試料は4回調製した。ブランクの読みには、アルブミン−g6pを溶液から省いた。吸光度は、Pharmacia Biotech Ultrospec 3000 UV/可視分光光度計(Cambridge,英国)上で石英セル中で読み取った。
プラスミノーゲン(200μg/ml)を、補助因子(5μMのエンドスタチン、Aβ(l−40)、またはフィブリン由来のペプチド85、86および87の一つ)の存在下または非存在下でtPA(200pM)と共にインキュベーションした。所定の時間間隔で試料を採取し、反応を5mM EDTAおよび150mMεACAを含む緩衝液で停止した。試料を集めた後、発色性プラスミン基質S−2251を加え、プラスミン活性を分光光度計で37℃にて動的に決定した。
NIE−115マウス神経芽腫細胞は、抗生物質を補足した5%FCSを含むDMEM中で常法により培養した52。細胞は、有糸分裂後ニューロンに分化した。細胞を20μg/mlのプラスミノーゲンの存在下または非存在下にて50μg/mlのCpBの存在下または非存在下でAβ(50μg/ml)に24時間暴露した。細胞を写真撮影し、計数して、培地に4x試料緩衝液(250mMトリス pH6.8,8%SDS,10%グリセロール,100mMDTT,0.01%w/vブロモフェノールブルー)を加えてリーシスした。接着および浮遊(乾燥および/または死んでいると思われる)細胞並びに培地を含む溶解生成物を、プラスミノーゲンおよびプラスミン、並びにラミニンの存在についてプラスミノーゲン(MoAb 3642, American Diagnostics)、ラミニン(PoAb L9393, Sigma)に対する特異抗体を用いるウェスタンブロット分析によって分析を行った。
本発明者らは、ヒトFXII(Calbiochem, La Jolla, CA, 米国,カタログ番号233490)のアミロイド(ポリ)ペプチドへの結合を試験した。プロトタイプアミロイドペプチドヒトアミロイド−β(1−40)(hAβ(1−40))およびヒトフィブリン断片α147−159FP13、およびグルコース−6−リン酸グリケーションしたウシアルブミン(アルブミン−高次グリケーション最終生成物(AGE))およびグルコース−6−リン酸グリケーションしたヒトヘモグロビン(HbAGE)であって、総て交差−β構造を有するもの、並びにネガティブコントロールであるマウスΔランゲルハンス島アミロイドポリペプチド(ΔmIAPP)、アルブミンコントロールおよびHbコントロールであって、総てアミロイド特異構造を書いているものをELISAプレートにコーティングし、一連の濃度のヒトXII因子を積層した。FXIIの結合は、ウサギポリクローナル抗−FXII抗体(Calbiochem, La Jolla, CA, 米国,カタログ番号233504)およびペルオキシダーゼ標識ブタ抗−ウサギIgGを用いて検出した。ウェルは、3回コーティングした。FXII結合緩衝液は、10mM HEPES pH7.3、137mM NaCl、11mM D−グルコース、4mM KC1、1mg/molアルブミン、50μM ZnCl2、0.02%(m/v)NaN3、および10mMε−アミノカプロン酸(εACA)からなっていた。リシン類似物εACAを加えて、FXIIクリングルドメインを介する交差−β構造へのFXIIの推定的結合を回避した。更に、FXIIのhAβ(1−40)およびプロトタイプアミロイドヒトアミリン断片hΔIAPPをドットブロット分析を用いて試験した。交差−β構造を含むペプチド10μg、並びにネガティブコントロールペプチドmΔIAPPおよびリン酸緩衝食塩水(PBS)を、メタノール活性化ニトロセルロースに2回スポットした。次いで、スポットを2nM FXII/FXII緩衝液またはFXII緩衝液のみ、抗FXII抗体、およびSWARPOと共にインキュベーションした。FXIIの結合は、強化ルミノール試薬(PerkinElmer Life Sciences, Boston, MA, 米国)とインキュベーションしたときの化学発光によって可視化した。交差−β構造フォールド49を有するポリペプチドへ結合する能力について知られているFXIIおよびtPAがアミロイド(ポリ)ペプチド上の積層結合部位に結合するかどうかを試験するため、競合的ELISAを行った。コーティングしたhAβ(1−40)またはアミロイドアルブミン−AGEを、一連の濃度のヒト組換え組織型プラスミノーゲンアクチベーター(Actilyse(商標),完全長tPA)またはReteplase(商標)(K2P−tPA)の存在下にて2.5nMまたは15nM FXII/結合緩衝液を用いてインキュベーションした。Reteplaseは、切頭型のtPAであって、第二のクリングルドメインとプロテアーゼドメインからなっている。f.l.tPA−およびK2P−tPA濃度は、最大でhAβ(1−40)へ結合するtPAのkDの135倍(50nM)またはアルブミン−AGEへ結合するtPAのkDの150倍(1nM)であった。
プロペプチド(残基Met35-Arg1)およびBg1II制限部位が先行する残基Ser1-Ser50であるヒトtPAのアミノ末端フィンガードメイン(F)のクローニングは、PCRおよび標準的組換えDNA手法を用いて行った。簡単に説明すれば、プロペプチド−フィンガー領域を、tPAを鋳型としてコードするcDNAを含むプラスミンZpl753 1ngを用いるPCRによって増幅した。用いたオリゴヌクレオチドは、それぞれSalI−またはNotI制限部位(下線部)を含んでなる下記の通りのものであった。
最初に、293T細胞を、5%v/vウシ胎児血清、ペニシリン、ストレプトマイシンおよびグアニジンを補足したRPMI1640培地(Invitrogen, Scotland, 英国)で15%のコンフルエンシーまで増殖した。細胞を、製造業者の推奨に従ってFugene-6を用いて一過的にトランスフェクションした(Roche, IN, 米国)。tPA断片を含むpMT2−tPA−F−GST、または受容体様タンパク質チロシンホスファターゼμ(RPTPμ)54の細胞外ドメインを含むコントロールプラスミドpMT2−RPTPμ−GSTをトランスフェクションし、培地を48時間のトランスフェクションの後に回収した。293T培地でのtPA−F−GSTおよびRPTPμ−GSTの発現を、イムノブロッティングによって証明した。集めた試料を、2x試料緩衝液を加えた後、SDS−PAAゲル上で展開した。膜を1%ミルク(Nutricia)でブロックし、最初にモノクローナル抗−GST抗体2F354および二番目にHRP接合ウサギ抗−マウスIgG(RAMPO)とインキュベーションした。ブロットを、Western Lightning Chemiluminescence Reagent Plus (PerkinElmer Life Sciences, MA, 米国)を用いて展開した。
仔ハムスター腎細胞を、5%v/vウシ胎児血清(FCS)、ペニシリン、ストレプトマイシンおよびグアニジンを補足したDMEM/NUT混合F−12(HAM)培地(Invitrogen,英国)で1%のコンフルエンシーまで播種した。細胞を、Ca3(PO4)2−DNA沈澱法を用いて、安定にトランスフェクションした。24時間後、培地に1mg/mlゲネティシン(geneticin)G−418硫酸塩(Gibco,英国)を補足した。G−418を含む培地を10日間に数回新しくして、死細胞を除去した。その後、安定な単一コロニーを新たな培養フラスコに移して、細胞をコンフルエンシーまで増殖させた。次に、構築物の発現をイムノブロッティングによって立証した。集めた試料を、2x試料緩衝液を加えた後にSDS−PAAゲル上で展開した。ゲルは、ニトロセルロース膜上でブロッティングした。膜を、1.5%m/vBSAを含む5%ミルク(Nutricia)中でブロックし、最初にモノクローナル抗−GST抗体(Santa Cruz Biotechnology, Santa Cruz, CA, 米国,カタログ番号Z-5)および二番目にHRP接合ウサギ抗−マウスIgG(RAMPO)とインキュベーションした。ブロットを、Western Lightning Chemiluminescence Reagent Plus(PerkinElmer Life Sciences, MA, 米国)を用いて展開した。安定なクローンを、以後250μg/mlのG−418の存在下で増殖させた。プルダウン実験のために、目的とする構築物を産生する安定なクローンの5%FCSを有するコンディショニングした培地を用いた。精製のため、tPA F−EGF−GSTの安定なクローンの細胞を三層の培養フラスコに移して、0.5%v/v Ultroser G(ITK Diagnostics, Uithoorn, オランダ)を含む培地で増殖させた。培地は、3または4日毎に新しくした。TPA F−EGF−GSTは、Glutathione Sepharose 4B (Amersham Biosciences, Uppsala, スウェーデン)カラム上で培地から単離して、100mM還元グルタチオン(Roche Diagnostics, Mannheim, ドイツ)で溶出した。
構築物の純度を、SDS−PAGEの後、クーマシー染色またはウェスタンブロット法によってチェックした。これらの分析から、幾らかのGSTが調製物中に存在することは明らかである。精製したtPA F−EGF−GSTをPBSに対して透析し、−20℃で保管した。
培地をNanosep 10KΩ遠心分離装置(Pall Gelman Laboratory, MI, 米国)を用いて20倍に濃縮し、製造業者の推奨に従ってSepharose 4Bにカップリングしたグルタチオンと共にインキュベーションした(Pharmacia Biotech, Uppsala, スウェーデン)と共にインキュベーションした。結合した構築物をPBSで洗浄し、50mMトリス−HCl pH8.0中10mMグルタチオンで溶出した。構築物を、使用前に−20℃で保管した。
GSTで標識したtPA F、F−EGF、EGF、K1、F−EGF−K1、FXII F、HGFa F、Fn F4、Fn F5、Fn F4−5およびGSTを発現するBHK細胞のコンディショニングした培地を用いて、アミロイド結合分析を行った。最初に、構築物を、ウェスタンブロット法を用いてほぼ等濃度に調整した。組換え断片の定性的結合は、「プルダウン」分析法を用いて評価した。この目的のため、組換えによって作製した断片をAβまたはIAPPフィブリルと共にインキュベーションした。これらのペプチドは不溶性繊維を形成するので、未結合タンパク質は遠心分離によって繊維から容易に除去することができる。結合断片を含むペレットは、続いて水で数回洗浄した。結合断片をSDS−試料緩衝液中で可溶化し、PAGEによって分析し、上清画分および出発材料中の未結合タンパク質も同様に処理した。ゲルを、抗−GST抗体Z−5を用いるイムノブロッティングを用いて分析する。
精製したtPA F−EGF−GST組換えタンパク質の親和性を画定するため、固定化アミロイド(ポリ)ペプチドおよび非アミロイドコントロールΔmIAPPを用いてELISAを行った。Aβ、FP13、IAPPまたはΔmIAPP 25μg/mlを、Exiqonペプチド固定化プレート上で固定した。過剰のεACAの存在下にて一連の濃度のtPA F−EGF−GSTをウェルに加えて、抗−GST抗体Z−5を用いて結合を分析した。コントロールとして、GST(Sigma-Aldrich, St. Louis,MO, 米国,カタログ番号G-5663)を同じ濃度で用いた。
ADに冒されたヒトのパラフィン脳切片は、Slootweg教授(Dept. of Pathology, UMC Utrecht)の厚意により送られた。切片を、一連のキシレン−エタノールで脱パラフィン化した。内在性ペルオキシダーゼを、メタノール/1.5%H2O2で15分間ブロックした。H2Oで洗浄した後、切片を未希釈ギ酸と共に10分間インキュベーションした後、PBS中で5分間インキュベーションした。切片を、10% HPS/PBS中で15分間ブロックした。切片を、PBS/0.3% BSA中で7nM tPA F−EGF−GSTまたはGSTに2時間暴露した。PBSで3回洗浄した後、切片に200ng/mlの抗−GST抗体Z−5を1時間積層した。洗浄後、使用準備のできたヤギ抗−ウサギPowervision(Immunologic, Duiven, オランダ,カタログ番号DPVR-55AP)を適用して、1時間インキュベーションした。洗浄後、切片を3,3’−ジアミノベンジジン(Sigma-Aldrich, St Louis, MO, 米国,カタログ番号D-5905)で10分間染色した後、ヘマトキシリン染色を10秒間行った。H2Oで洗浄した後、切片を製造業者の推奨に従ってコンゴレッドと共にインキュベーションした(Sigma Diagnostics, St Louis, MO, 米国)。切片をキシレンで洗浄し、D.P.X. Mounting Medium(Nustain, Nottingham, 英国)で固定した。切片の分析は、Leica DMIRBE蛍光顕微鏡(Rijswijk, オランダ)上で行った。コンゴレッドの蛍光は、励起波長596nmおよび発光波長620nmを用いて分析した。
tPA−F−GSTおよびRPTPμ−GSTの繊維状アミロイドヒトAβ(1−40)およびアルブミン−g6pへの結合を、ELISAで分析した。簡単に説明すれば、ヒトAβ(1−40)、アルブミン−g6pまたは緩衝液のみを、それぞれペプチドI イモビライザーまたはタンパク質I イモビライザーにコーティングした。ウェルを精製したGSTで標識した構築物またはコントロール培地と共にインキュベーションし、結合を一次抗−GSTモノクローナル抗体2F3およびRAMPOを用いて検出した。ウェルはまた、10mM eACAの存在下にて500nM tPAと共にインキュベーションした。次に、tPAの結合は、クリングル2ドメインに位置したリシル結合部位から独立している。tPAの結合を、一次抗体374BおよびRAMPOを用いて測定した。実験は、3回行う、コーティングを行っていないウェルのブランク読みを差し引いた。
グルコース−6−リン酸でグリケーションしたウシ血清アルブミンに対する抗体を、標準的免疫機構を用いてウサギで誘導した。2週間グリケーションしたアルブミン−AGE(Prof. Dr. Ph. G. de Groot/Dr. I. Bobbink; 未公表データ)で免役した後、抗−AGE1を得た。抗体を、Protein Gカラムを用いて血清から精製した。抗−AGE2を、Davids Biotechnologie (Regensburg, ドイツ)によって展開した。アルブミン−AGE:23で免役した後、抗体をEMD−Epoxy活性化ビーズ(Merck, Darmstadt, ドイツ)に接合したヒトAβ(1−40)上でアフィニティー精製した。ポリクローナルマウス抗−AGE抗体は、アルブミン−AGE:23およびヒトAβ(1−40)で9:1のモル比で免役した後に得た。ポリクローナル血清は、標準的免疫手順を用いて得て、これはAcademic Biomedical Cluster Hybridoma Facility (Utrecht University, オランダ)によって行った。次に、モノクローナルは、標準的手順を用いて生成した。
ELISAのため、アミロイド化合物を、上記と同様にして、Exiqonペプチドまたはタンパク質イモビライザー(Vedbaek, デンマーク)に固定した。抗−AGE抗体および市販の抗−Aβ(l−42) H-43(Santa Cruz Biotechnology, Santa Cruz, CA, 米国)を、0.1% v/v Tween20を含むPBSで希釈した。ウサギ抗−ヒトビトロネクチンK9234はDr. H. de Boer (UMC Utrecht)の厚意により提供を受け、ネガティブコントロールとして用いた。マウスポリクローナル抗−アルブミン−AGE/Aβを用いるELISAのため、関連のないタンパク質に対して誘導された抗体を含むコントロール血清を用いた。マウスポリクローナル抗−アルブミン−AGE/Aβの結合は、PBS/0.1% Tween20中の血清の希釈系列を用いて行った。IAPPとの競合結合分析のために、抗−AGE1を様々な濃度のIAPPとインキュベーションした。IAPPフィブリルを分離し、上清をAβをコーティングしたELISAプレートのウェルに3回塗布した。マルチリガンド交差−β構造結合セリンプロテアーゼtPAを用いる競合結合分析は、若干異なる方法で行った。コーティングしたAβおよびIAPPにkDに関連した抗−AGE1または抗−Aβ(l−42)H−43濃度を一連の濃度のtPAと共に積層した。107−104倍モル過剰量のリシン類似物εACA(10mM)が結合緩衝液に存在し、tPAのAβおよびIAPPのリシン残基への結合を回避し、これはアミロイド構造の存在とは独立していた。
抗−AGE1を、Aβ(16−22)、Aβ(1−40)およびIAPPのアミロイド凝集体と共にインキュベーションした。遠心分離の後、ペレットを非還元性試料緩衝液(1.5%(m/v)ドデシル硫酸ナトリウム、5%(v/v)グリセロール、0.01%(m/v)ブロモフェノールブルー、30mMトリス−HCl pH6.8)に溶解したPBS/0.1% Tween20で3回洗浄した。アミロイドフィブリルをペレット化の後、上清を2x試料緩衝液で1:1に希釈した。試料をポリアクリルアミドゲルに適用して、ウェスタンブロット法の後、抗−AGE1をSWARPOで検出した。
Aβカラム上でアフィニティー精製したウサギ抗−AGE2を用いて、ADに罹っているヒトの脳切片におけるアミロイドプラークに対する結合特性を分析した。手順は、本質的に上記の通りに行った。11μg/mlの抗−AGE2のタンパク質−AGE付加物または脳切片におけるヒトアルブミンへの終局的な結合を回避するため、300nMのg6p−グリケーションジペプチドGly−Lysを0.3%m/v BSAと共に結合緩衝液に加えた。免疫組織化学的染色の後、切片をコンゴレッドで染色した。
溶液中でのアミロイド交差−β構造を検出するため、サンドイッチELISA法を用いた。Actilyse tPAを、96穴タンパク質イモビライザープレート(Exiqon, Vedbaek, デンマーク)のウェルに10μg/mlの濃度で固定した。一連の濃度のアルブミン−AGE:23およびアルブミン−コントロール:23をtPAをコーティングしたウェル並びにコーティングしていないコントロールウェルに加えた。アミロイド構造の結合は、1μg/mlの抗−Aβ(1−42)H−43(Santa Cruz Biotechnology, Santa Cruz, CA,米国)、続いて0.3μg/mlのSWARPOと共にインキュベーションした後に、オルト−フェニレン−ジアミン/H2O2/H2SO4染色によって検出した。
1. 交差−β構造は、フィブリンおよびフィブリン由来の合成ペプチドに存在する。
本発明者らは、フィブリン血餅がコンゴレッドで染色し(図示せず)チオフラビンT蛍光を示し(図2A)、フィブリン血餅におけるアミロイド構造の存在を示していることを明らかにした。コンゴレッド染色(図示せず)、円偏光二色性測定およびX線回折分析を用いて、本発明者らは、フィブリンの配列から誘導される合成ペプチドが交差−β構造を採用していることを示す(図2B,C)。これらのペプチドは、以前にtPA−結合とtPA活性化特性18を有することが見出された。これらのペプチドにおける交差−β構造の存在により、tPAによって伝達されるプラスミノーゲン活性化を刺激する能力と相関することを見出した(図2D)。
tPA、プラスミノーゲンおよびプラスミンがAβと結合するかどうかを試験するため、固相結合分析法を行った。Aβをプラスチック製の96穴プレートにコーティングし、ペプチドのプラスミン(プラスミノーゲン)またはtPAへの結合を、コーティングしたペプチドに増加濃度のtPA、プラスミノーゲンまたはプラスミンを積層することによって評価した。結合は、ELISAを行うことによってプラスミン(プラスミノーゲン)またはtPAに対する特異抗体を用いて評価した。図3Aは、tPAがAβにフィブリンに結合するtPAのKdと同様の約7nMのKdで結合することを示す。対照的に、プラスミノーゲンのAβへの検出可能な結合は見られなかった(図3B)。しかしながら、活性化プラスミノーゲン(プラスミン)はAβに結合し、47nMのKdで結合する。(活性な)プラスミンはAβに結合するが、(不活性)プラスミノーゲンはAβに結合しないという事実は、プラスミン活性、従って遊離リシンの生成はプラスミンのAβへの結合に重要であることを示唆している。これを試験するため、リシン類似物であるε−アミノカプロン酸(εACA)を用いてプラスミンおよびtPAのその存在におけるAβへの結合を試験した。本発明者らは、プラスミンのAβへの結合はεACAの存在下で完全に廃止されることを示す(図3D)。対照的に、εACAはtPA−Aβ相互作用に対して効果がない(図3C)。従って、プラスミンは、恐らくは(複数の)リシン結合クリングルドメインを介してインキュベーション期間中に生成した遊離リシンに結合すると結論される。これと協力して、フィブリン中の遊離リシンへのプラスミノーゲンクリングルドメインのKdは、Aβに対するプラスミンの結合のためのKdに類似している。
コンゴレッド染色(図示せず)、X線回折分析およびTEMを用いて、Escherichia coli並びにPichia pastors由来のエンドスタチン凝集体における交差−β構造の存在およびエンドスタチンがアミロイドフィブリルを形成する能力を示した(図6A−B)。細菌性エンドスタチンは4.7オングストローム(水素結合距離)並びに10−11オングストローム(シート間距離)で反射線を生じることを見出した。反射線は、反対回折核で最大強度を示す。水素結合繰り返し距離が4.7オングストロームの繊維軸は、軸方向の毛細管軸に沿って配向されている。これは、10−11オングストロームのシート間距離がこれらの水素結合に直交していることを意味する。これは、タンパク質が交差−β構造を有する交差−βシートコンホメーションであることと一致する。球状タンパク質の分子内βシートは、この方法で配列されている回折パターンを生じることができない。バックグラウンド散乱の量から、タンパク質のごく一部だけが交差−β構造形成に参加していることになる。本発明者らは、エンドスタチンにおける交差−β構造の存在がtPAによって伝達されるプラスミノーゲン活性化を刺激する能力と相関し(図6C)、且つニューロン細胞死と相関する(図6D)ことを見出した。
IAPPのアミロイド沈着物は、糖尿病II型患者の膵臓で形成される59。IAPPはイン・ビトロで細胞死を引き起こすことができ、従ってイン・ビボで見られるβ細胞の破壊に寄与し、これによりインスリン産生が不十分となると考えられる。IAPPは、交差−β構造を含んでなるフィブリルを形成する60。
幾つかのタンパク質のグリケーションによって、これらのタンパク質がtPAと結合する能力を誘発しまたは増加することができ、且つtPAによって伝達されるプラスミン形成を刺激することができる22;61。本発明者らは、g6pを用いるアルブミンのグリケーションがアルブミンに対する高親和性tPA結合を与えるだけでなく(図8A)、チオフラビンTに結合することもできるようになることを示す(図8C)。tPAの結合は、コンゴレッドと競合することができる(図8B)。更に、チオフラビンTのグリコアルブミンへの結合をtPAと競合させることができる(図8D,E)。コンゴレッドおよび/またはチオフラビンTとtPAが競合するという事実は、それらが同一のまたは重複している結合部位を有することを示している。
ヒトヘモグロビンのg6pとのインキュベーションにより、高親和性tPA結合が生じた(図9A)。フィブリルを含む非晶質の凝集体Hb−g6p付加物がTEMで観察されたが(図9B)、コントロールHbは凝集しなかった(図示せず)。糖尿病患者のヒトHbは、12.4%を上回るHbがグリケーションされると、フィブリル状凝集体を形成する傾向を有する(表II)。
上記の観察から、アルブミンの−NH2基のg6pによる修飾によって、アミロイド交差−β構造の形成が誘発されることは明らかである。本発明者らが検討した次の疑問は、球状アルブミンのアミロイドフォールドへのリフォールディングの誘発がg6pの制限特性であるかどうか、またはアミロイド形成がAGE形成に用いた元の炭水化物または炭水化物誘導体とは無関係に起こるかどうかであった。アルブミン溶液を、1M g6p、250mM DL−グリセルアルデヒド/100mM NaCNBH3、1Mβ−D−(−)−フルクトース、1M D(+)−グルコース、500mMグリオキシル酸/100mM NaCNBH3、および相当するPBSおよびPBS/NaCNBH3緩衝液コントロールと共に37℃で86週間インキュベーションした。グリセルアルデヒドおよびグリオキシル酸は、メイラード反応におけるAGEの前駆体である炭水化物誘導体である63;64。86週間後に、アルブミン− グリセルアルデヒドおよびアルブミン−フルクトースは淡黄/褐色懸濁液となった。コントロールは無色の透明溶液であった。アルブミン−グルコースおよびアルブミン−グリオキシル酸は透明な単黄−淡褐色溶液であった。アルブミン−g6p:86は、透明な暗褐色溶液であった。AGE形成は、AGEに特異的な励起/発光波長(図示せず)、moab抗−AGE4B5の結合(図示せず)およびpoab抗−AGEの結合(図示せず)を用いる自己蛍光測定によって確かめた。予想されたように、アルブミン−グリオキシル酸は(主として)非蛍光性カルボキシメチル−リシン(CML)付加物が形成されるという事実によって自己蛍光シグナルを示さなかった63。
AβはtPAおよびコンゴレッドに結合することができることは明らかにされている。本発明者らは、tPAのAβへの結合をコンゴレッドと競合させることができることを示す(図11)。これらの結果は、Aβ、フィブリンおよびグリコアルブミンの構造は類似しており、tPAへの結合を伝達することができるという本発明者らの知見を支持している。
図12のグラフは、FXIIが、試験した総てのアミロイド化合物に特異的に結合することを示している。hAβ(1−40)、FP13、アルブミン−AGEおよびHb−AGEについてのkDは、それぞれ約2、11、8および0.5nMである。競合的FXII−tPA ELISAを用いて得たデータは、tPAがFXIIのアミロイド(ポリ)ペプチドへの結合を効率的に阻害することを示している(図12)。これらのデータから、FXIIおよびf.l.tPAはhAβ(1−40)上の結合部位の重複について競合していると結論した。K2P−tPAは、FXII結合を阻害しない。FXIIのアルブミン−AGEへの結合も、hAβ(1−40)について観察されたのと同様に、tPAによって効果的に廃止されるが、K2P−tPAによっては廃止されない。これは、FXIIが同様にしてhAβ(1−40)およびアルブミン−AGEに結合することができることを示している。更に、これらのデータは、tPAの最初の3個のドメイン(フィンガー、EGF様、クリングル1)が、FXIIとアミロイドhAβ(1−40)またはアルブミン−AGEとの相互作用に対するf.l.tPAの阻害効果に関与していると思われることを示している。ドットブロット分析法を用いて、FXIIのスポットしたアミロイドhΔIAPPおよびhAβ(1−40)への結合を試験した。ネガティブコントロールPBSおよびmΔIAPPについては、FXIIの結合は見られなかった(図12)しかしながら、FXIIは、初期の報告65と一致してhAβ(1−40)並びにhΔIAPPと特異的に結合した(図12)。これらのデータは、図12A−Fに示されたELISAデータと共に、FXIIがアミノ酸配列相同性を共有しないポリペプチドに結合することができ、これによって交差−β構造フォールドを共有することを示唆している。これは、アミロイド特異的フォールドを含むtPAとポリペプチドの相互作用についての本発明者らの最新のデータと一致している。
tPAの幾つかのドメインは、フィブリンまたはフィブリン断片への結合を伝達することが示されている12;53;66;67。しかしながら、tPAのどのドメインがAβまたは他の交差−β構造を含む分子への結合にとって必要であるかは知られていない。本発明者らは、レテプラーゼと呼ばれ、N末端フィンガー、EGFおよびクリングル1ドメイン(K2−tPA)を欠いている突然変異tPAが交差−β構造を含んでなる分子と結合することができないことを示す(図13A,B)。これらの結果は、N末端領域がtPAのフィブリルを含んでなる交差−β構造への結合に必要であることを示唆している。
GSTで標識した構築物tPA−F−GSTおよびRPTPμ−GST(コントロール)を、Sepharose 4Bビーズにカップリングしたグルタチオンを用いて293T培地から精製したところ、それぞれ約35kDaおよび150kDaの単一バンドを生じた(図示せず)。培地で用いたFCSに由来する微量のBSAも、存在した。
ELISAにおいて、コントロールtPAは、過剰のεACAの存在下でヒトAβ(1−40)およびアルブミン−g6pへ結合した(図13C)これは、用いた分析法では、tPAは繊維状アミロイドにクリングル2から独立して結合することができることを示している。tPA−FドメインはヒトAβ(1−40)およびアルブミン−g6pに結合したが、RPTμ−GSTでは結合は見られなかった。従って、tPA−F−GSTで見られた結合は特異的であり、GSTタグに由来しない。この結果は、tPAフィンガードメインを交差−β構造dアミロイドフィブリルへの結合についての性質によってデザインされた特異的ドメインとして示している。
tPAのフィンガードメインは、フィブリンへの高親和性結合に重要であることが示されているl2; 66。レテプラーゼ(K2−P tPA)およびF−tPA、F−EGF−tPAおよびF−EGF−K1−tPAを用いる本発明の結果は、フィブリン以外の刺激因子への結合におけるtPAのN末端フィンガードメインの重要な役割を示している。個々までは、これら総ての因子はコンゴレッドと結合し、交差−β構造を含む。更に、フィブリンについてのフィブロネクチンの結合部位は、フィンガードメインタンデムF4−F5に対してマッピングされている68。フィブリン断片FCB2の存在下における完全長tPAによるプラスミノーゲン活性化はフィブロネクチンによって阻害することができることが示されている69。まとめて考えると、これらの観察は、tPAとフィブロネクチンがフィブリン上の同一または重複結合部位についてフィンガードメインによって競合することを示唆している。本発明者らのデータは、FnのF4−5ドメインがアミロイドAβに結合することを示している。
最近、O'Nuallain and Wetzel70は、アミロイドの特徴を有するペプチドに対して誘発された抗体は、アミノ酸配列には関係なく同様なアミロイド特性を有する任意の他のペプチドに結合することができることを示した。これらのデータ、および組織型プラスミノーゲンアクチベーターおよびXII因子は、アミロイド特異的交差−β構造フォールドを共有する配列に関係のないポリペプチドのファミリーに結合することができるという本発明者らの観察に基づいて、一層広汎なクラスのタンパク質が特異的アミノ酸残基によって構成された線形またはコンホメーションエピトープに対するよりはこの構造単位に対する親和性を示すことができると仮定した。この仮定により、アミロイド交差−β構造フォールドを含むアルブミン−AGEに対して誘発された抗体が、この交差−β構造フォールドを有する任意の(ポリ)ペプチドに対して広範囲の特異性をも示すかどうかという疑問を刺激した。
溶液状のアミロイドアルブミン−AGE:23を積層したコーティングtPAでのサンドイッチELISA法を用いた後、広範囲の抗−Aβ(1−42)H−43を検出し(図17)、溶液状のタンパク質を含む交差−β構造を検出することができた。

- 循環中に含まれる細胞外タンパク質の交差−β構造形成を調節することを含んでなる、細胞外タンパク質分解および/またはタンパク質クリアランスを調節する方法。
- 循環中に含まれる細胞外タンパク質の交差−β構造形成を増加させることを含んでなる、細胞外タンパク質分解および/またはタンパク質クリアランスを増加させる方法。
- 循環中に含まれる細胞外タンパク質の交差−β構造形成を増加させることができる化合物を供給することを含んでなる、細胞外タンパク質分解および/またはタンパク質クリアランスを増加させる方法。
- 前記化合物がグルコースである、請求項3に記載の細胞外タンパク質分解および/またはタンパク質クリアランスを増加させる方法。
- 循環中に含まれる細胞外タンパク質の交差−β構造を安定化することを含んでなる、細胞外タンパク質分解および/またはタンパク質クリアランスを増加させる方法。
- 循環中に含まれる細胞外タンパク質の交差−β構造形成を減少させることを含んでなる、細胞外タンパク質分解および/またはタンパク質クリアランスを減少させる方法。
- 循環中に含まれる細胞外タンパク質の交差−β構造形成を減少させることができる化合物を供給することを含んでなる、細胞外タンパク質分解および/またはタンパク質クリアランスを減少させる方法。
- 組織プラスミノーゲンアクチベーター(tPA)様活性を調節することを含んでなる、細胞外タンパク質分解および/またはタンパク質クリアランスを調節する方法。
- tPA様活性を増加させることができる化合物を供給することを含んでなる、細胞外タンパク質分解および/またはタンパク質クリアランスを増加させる方法。
- 前記化合物が交差−β構造を含んでなる、請求項9に記載の細胞外タンパク質分解および/またはタンパク質クリアランスを増加させる方法。

- 前記化合物が、カルボキシペプチダーゼインヒビター(CPI)またはCPI様活性を含んでなる、請求項9または11に記載の細胞外タンパク質分解および/またはタンパク質クリアランスを増加させる方法。
- tPA様活性を減少させることができる化合物を供給することを含んでなる、細胞外タンパク質分解および/またはタンパク質クリアランスを減少させる方法。
