Abstract
Description
ヒト新産児および新産動物を、外来抗原ならびにそれぞれヒトおよび動物の疾患に関係する抗原に対して、それぞれ予防接種する手段を提供することが、本発明の目的である。具体的に述べると、新産動物およびヒト新産児の免疫系の成熟を加速させうる手段を提供することが、本発明の目的である。また、ヒトを含む新生動物をポックスウイルス感染に対して、特に天然痘に対して、予防接種することを可能にする手段を提供することも、本発明の目的である。
本発明によれば、意外なことに、ヒトを含む新生動物または出生前動物の細胞に感染する能力を持つが上記細胞内で感染性子孫ウイルスに複製される能力は持たないウイルスを用いて、ヒトを含む新生動物または出生前動物を安全にしかも効率よく予防接種及び(又は)処置できることが見いだされた。特に、本発明で使用されるウイルス、例えばMVA、とりわけMVA−BNおよびその誘導体(下記参照)は、新産仔に投与しても有害な作用を何も示さないことが明らかになった。以下に詳述するように、上記のウイルスによる動物の予防接種は、予防接種に使用したウイルスに対する特異免疫応答をもたらし、かつ/または外来抗原および腫瘍抗原に対する一般予防接種になる。さらに、本発明で使用されるウイルスは、樹状細胞数の増加およびインターフェロンなどの因子の増加を伴う、免疫系の成熟の誘導及び(又は)強化をもたらす。本発明で使用されるウイルスによる予防接種は、動物に投与される製剤がアジュバントを含んでいなくても可能である。
(i)ニワトリ胚線維芽細胞(CEF)および細胞株BHKでは増殖的に複製する能力を持つが、ヒト細胞株HaCatでは増殖的に複製する能力を持たない、
(ii)生体内では複製不能、
(iii)致死チャレンジモデルで既知の株MVA575(ECACC V00120707)よりも高い免疫原性を誘導及び(又は)
(iv)DNA初回免疫/ワクシニアウイルス追加免疫方式と比較して、ワクシニアウイルス初回免疫/ワクシニアウイルス追加免疫方式で少なくとも実質的に同レベルの免疫を誘導。
AGR129マウスの代わりに、成熟したB細胞およびT細胞を産生することができず、したがって重度の免疫無防備状態であり、複製型ウイルスに著しく感染しやすいマウス株であれば、他のどのマウス株でも使用することができる。
さらに、免疫系の成熟の加速は、好ましくは、新産動物へのMVA−BNの投与の7日後に、MVA−BNを投与されていない対照動物と比較して、血中及び(又は)脾臓中のCD11c陽性細胞数の少なくとも1.5倍の増加、好ましくは少なくとも2.0倍の増加と関連づけられる。さらに、免疫系の成熟の強化は、好ましくは、本発明のウイルスによる新生仔の予防接種の2日後に、年齢対応対照群と比較して、Flt3−L濃度の少なくとも1.5倍の増加、好ましくは少なくとも2.0倍の増加と関連づけられる。
ヒトを含む新生動物または出生前動物を予防接種または処置するための医薬の製造にウイルスを使用する方法において、当該ウイルスは、ヒトを含む上記新生動物または出生前動物の細胞に感染する能力を持つが、ヒトを含む上記新生動物または出生前動物では感染性子孫ウイルスに複製される能力を持たないことを特徴とする、上記使用する方法。
図1A:誕生後24〜48時間以内の新産マウスに、106p.f.u.のMVAまたはDISC HSV−1を1回注射するか、または対照として生理食塩水(NaCl)による処置を行った。7日齢時に、汎DCマーカーであるCD11cを使って、末梢血中のこれらの細胞を、フローサイトメトリーで決定した。3〜5回の実験の平均および標準偏差を示す。
(i)MVA−BNおよびDISC−HSVは、新産動物において、CD11c+およびCD8+表現型のDCを誘導する。
b組換え活性遺伝子にRAGマウス欠失(すなわち機能的なT細胞およびB細胞なし)

(iii)MVA−BNで処置した新生マウスでは血清Flt3−Lのレベルが上昇している。
(i)MVA−BN処置新生マウスは100〜500 LD50のHSV−1によるチャレンジに耐えて生き残る。
(ii)MVA−BNは成体期まで持続する特異免疫を新生仔に誘導する。

- ヒトを含む新生動物または出生前動物を予防接種または処置するための医薬の製造にウイルスを使用する方法において、当該ウイルスは、ヒトを含む上記新生動物または出生前動物の細胞に感染する能力を持つが、ヒトを含む上記新生動物または出生前動物では感染性子孫ウイルスに複製される能力を持たないことを特徴とする、上記使用する方法。
- ヒトを含む新生動物または出生前動物を予防接種または処置するためのウイルスにおいて、当該ウイルスが、ヒトを含む上記新生動物または出生前動物の細胞に感染する能力を持つが、ヒトを含む上記新生動物または出生前動物では感染性子孫ウイルスに複製される能力を持たないウイルス。
- 当該ウイルスがDNAウイルスである、請求項1又は2記載の使用する方法又はウイルス。
- 当該ウイルスがDISC−ヘルペスウイルスおよび変異ワクシニアウイルスアンカラ(MVA)から選ばれる、請求項1〜3のいずれか1つに記載の使用する方法又はウイルス。
- 上記MVA株が、動物細胞培養物の欧州収集所( European Collection of Animal Cell Cultures(ECACC))に受託番号V00083008として寄託されているMVA−BNおよびその誘導体である、請求項4に記載の使用する方法又はウイルス。
- MVAが経口適用、鼻腔適用、筋肉内適用、静脈内適用、腹腔内適用、皮内適用、子宮内適用及び(又は)皮下適用によって投与される、請求項4又は5記載の使用する方法又はウイルス。
- MVAが、1回目の接種(「初回接種」)および2回目の接種(「追加接種」)において、治療上有効な量で投与される、請求項4〜6のいずれか1つに記載の使用する方法又はウイルス。
- MVAが、ヒトを含む上記動物に、少なくとも101 TCID50(組織培養感染価)の量で投与される、請求項4〜7のいずれか1つに記載の使用する方法又はウイルス。
- 上記予防接種がポックスウイルス感染に対して行われる、請求項4〜8のいずれか1つに記載の使用する方法又はウイルス。
- 上記ポックスウイルス感染が天然痘感染である、請求項9に記載の使用する方法又はウイルス。
- ウイルスゲノムが少なくとも1種の異種核酸配列を含む、請求項1〜10のいずれか1つに記載の使用する方法又はウイルス。
- 上記異種核酸配列が、少なくとも1種の抗原、抗原エピトープ及び(又は)治療化合物をコードする配列から選ばれる、請求項11記載の使用する方法又はウイルス。
- 上記予防接種が、上記異種配列の由来となった病原体に対するものであるか、又は上記少なくとも1種の抗原もしくは抗原エピトープを含む病原体に対するものである、請求項11又は12記載の使用する方法又はウイルス又はウイルス又はウイルス。
- 上記予防接種が、腫瘍抗原及び外来抗原から選ばれる抗原に対して、ヒトを含む上記動物を保護するためのものであって、その際上記腫瘍抗原及び(又は)上記外来抗原は当該ウイルスに関連する抗原と異なる、請求項1〜8又は11〜12のいずれか1つに記載の使用する方法又はウイルス。
- 上記外来抗原が感染性病原体および毒素から選ばれる、請求項14記載の使用する方法又はウイルス。
- 上記感染性病原体がウイルス、細菌、プリオンおよび真菌から選ばれる、請求項15記載の使用する方法又はウイルス。
- 上記予防接種または処置が、免疫系の成熟及び(又は)活性化を誘導又は強化することにある、請求項1〜8のいずれか1つに記載の使用する方法又はウイルス。
- 上記の処置が、(i)樹状細胞またはその前駆細胞を活性化及び(又は)動員(mobilize)する因子のレベルを増加させることにあるか、(ii)樹状細胞またはその前駆細胞の数を増加させることにあるか、あるいは(iii)インターフェロン(IFN)又はIL−12の産生量及び(又は)細胞含有量を増加させることにある、請求項1〜8のいずれか1つに記載の使用する方法又はウイルス。
- 樹状細胞を活性化する因子がFlt3−L及び(又は)GM−CSFである、請求項18記載の使用する方法又はウイルス。
- 上記インターフェロンがIFNα及び(又は)IFNβである、請求項18又は19記載の使用する方法又はウイルス。
