Claims (12)
1. Transgena biljka kukuruza sadrži kromosom koji obuhvaća heterologni polinukleotidni umetak koji eksprimira cspB gen reguliran od strane skraćenog riža-aktin promotora, pri čemu navedeni kromosom sadrži SEQ ID NO: 24 pod-sekvencu, pri čemu je selektivni marker gen neomicin fosfotransferaze tipa II lociran između lox mjesta isječen, i pri čemu navedeni heterologni polinukleotidni umetak prenosi poboljšanu toleranciju navedene biljke kukuruza na manjak vode.
2. Sjeme transgene biljke kukuruza prema zahtjevu 1, pri čemu navedeno sjeme sadrži kromosom koji obuhvaća heterologni polinukleotidni umetak koji eksprimira cspB gen reguliran od strane strane skraćenog riža-aktin promotora, pri čemu navedeni kromosom sadrži SEQ ID NO: 24 pod-sekvencu, pri čemu je selektivni marker gen neomicin fosfotransferaze tipa II lociran između lox mjesta isječen, i pri čemu navedeni heterologni polinukleotidni umetak prenosi poboljšanu toleranciju na manjak vode u biljki kukuruza uzgojenog spomenutim sjemenom.
3. Sjeme prema zahtjevu 2, pri čemu je sjeme, sjeme hibridnog kukuruza koje je heterozigotno za heterologni polinukleotidni umetak koji eksprimira cspB gen.
4. Postupak određivanja zigotnosti heterolognog polinukleotidnog umetka koji prenosi poboljšanu toleranciju na manjak vode kod transgenih biljaka kukuruza, pri čemu spomenuti postupak uključuje kontakt DNK uzorka iz transgenih biljka kukuruza ili sjemena koje sadrži kromosom obuhvaćajući heterologni polinukleotidni umetak iz SEQ ID NO: 24, ili SEQ ID NO: 24 pod-sekvence, pri čemu je selektivni marker gen neomicin fosfotransferaze tipa II lociran između lox mjesta isječen setom začetnica koji obuhvaća prvi set začetnica za proizvodnju amplikona DNK transgenog kukuruza koji ima SEQ ID NO: 1 ili 2, koji je dijagnostički za kromosom koji sadrži heterologni polinukleotidni umetak, drugi set začetnica za proizvodnju amplikona DNK ne-transgenog kukuruza na mjestu insercije, podvrgavanje kontaktnog uzorka polinukleotidnoj amplifikaciji i koristeći sonde za detekciju amplikona DNK transgenog kukuruza i amplikona DNK ne-transgenog kukuruza, pri čemu prisutnost samo amplikona DNK transgenog kukuruzna iz prvog seta začetnica indicira da je DNK homozigotan za heterologni polinukleotidni umetak, a prisutnost amplikona proizvedenih iz prvog i drugog seta začetnica indicira da je DNK heterozigotan za heterologni polinukleotidni umetak, na taj način određujući zigotnost heterolognog polinukleotidnog umetka.
5. Postupak prema zahtjevu 4, pri čemu navedeni prvi set začetnica obuhvaća SEQ ID NO: 14 i SEQ ID NO: 15, a navedeni drugi set začetnica obuhvaća SEQ ID NO: 17 i SEQ ID NO: 18.
6. Postupak prema zahtjevu 5, pri čemu sonda za otkrivanje amplikona DNK transgenog kukuruza sadrži SEQ ID NO: 16 a sonda za otkrivanje amplikona DNK ne-transgenog kukuruza sadrži SEQ ID NO: 19.
7. Postupak prema zahtjevu 4, pri čemu navedeni prvi set začetnica obuhvaća začetnicu od najmanje 16 u nizu poredanih nukleotida iz SEQ ID NO: 5 ili SEQ ID NO: 6, i začetnicu od najmanje 16 u nizu nukleotida, koji se nalaze u SEQ ID NO: 7.
8. Prerađena hrana ili stočna hrana dobivena od sjemena kukuruza ima kromosom koji sadrži transgeni/genomski križani polinukleotid iz SEQ ID NO: 1 i heterologni transgeni umetak koji sadrži skraćeni riža-aktin promotor koji je operativno vezan na cspB gen, pri čemu se 5' terminalni kraj navedenog umetka preklapa sa 3' terminalnim krajem iz SEQ ID NO: 1, pri čemu spomenuta prerađena hrana ili stočna sadrži mjerljivu količinu polinukleotida koji obuhvaća nukleotidnu sekvencu iz SEQ ID NO: 1, SEQ ID NO: 2, ili njihov komplement.
9. Prerađena hrana ili stočna hrana prema zahtjevu 8, pri čemu spomenuta hrana ili stočna hrana obuhvaća kukuruznu krupicu, kukuruzno brašno ili kukuruzni gluten.
10. Prerađena hrana ili stočna hrana prema jednom od zahtjeva 8 ili 9, pri čemu navedeni polinukleotid nadalje sadrži nukleotidnu sekvencu iz SEQ ID NO: 3, SEQ ID NO: 4, ili njihov komplement.
11. Prerađena hrana ili stočna hrana prema jednom od zahtjeva 8 do 10, pri čemu navedeni polinukleotid nadalje sadrži nukleotidnu sekvencu sadržanu u SEQ ID NO: 5, SEQ ID NO: 6, SEQ ID NO: 7 ili SEQ ID NO: 24.
12. Sastav obuhvaća polinukleotidnu sekvencu koja je dijagnostička za prisutnost MON87460 DNK, pri čemu polinukleotid ima sekvencu navedenu u SEQ ID NO: 2, SEQ ID NO: 3, SEQ ID NO: 4, ili SEQ ID NO: 25.
