En un conjunto de realizaciones, un polímero (por ejemplo, copolímero, por ejemplo, copolímero de bloque) de la presente invención incluye un polímero biocompatible, es decir, el polímero que no induce típicamente una respuesta adversa cuando se inserta o inyecta en un sujeto vivo, por ejemplo, sin una inflamación significativa y/o rechazo agudo del polímero por el sistema inmune, por ejemplo, a través de una respuesta de linfocitos T. En determinadas realizaciones, el polímero biocompatible es biodegradable, es decir, el polímero es capaz de degradarse química y/o biológicamente dentro de un entorno fisiológico, tal como dentro del cuerpo. Por ejemplo, el polímero puede ser uno que se hidrolice espontáneamente tras su exposición a agua (por ejemplo, dentro de un sujeto), el polímero puede degradarse tras su exposición a calor (por ejemplo, a temperaturas de aproximadamente 37 ºC). La degradación de un polímero puede producirse a velocidades variables, dependiendo del polímero o copolímero usado. Por ejemplo, la semivida del polímero (el tiempo en que se degrada el 50 % del polímero en monómeros y/u otros restos no poliméricos) puede ser del orden de días, semanas, meses o años dependiendo del polímero. Los polímeros pueden degradarse biológicamente, por ejemplo, por actividad enzimática o por la maquinaria celular, en algunos casos, por ejemplo, a través de la exposición a una lisozima (por ejemplo, que tenga un pH relativamente reducido). En algunos casos, los polímeros pueden degradarse en monómeros y/u otros restos no poliméricos que las células pueden volver a usar o desechar sin efectos tóxicos significativos sobre las células (por ejemplo, la polilactida puede hidrolizarse para formar ácido láctico, la poliglicolida puede hidrolizarse para formar ácido glicólico etc.). etc.).

En algunas realizaciones, El polímero biocompatible es un polímero hidrófobo. Los ejemplos no limitativos de polímeros biocompatibles incluyen polilactida, poliglicolida y/o poli (lactida-co-glicolida).
En una realización, se describe en el presente documento una nanopartícula que comprende 1) una matriz polimérica; 2) un compuesto o capa anfífila que rodea o está dispersa dentro de la matriz polimérica formando una envoltura continua o discontinua para la partícula; 3) un polímero recubierto, y 4) un ligando de PSMA de bajo peso molecular unido covalentemente. Una capa anfífila puede reducir la penetración de agua en la nanopartícula, aumentando de este modo la eficacia de encapsulación del fármaco y ralentizando la liberación del fármaco. Además, estas nanopartículas protegidas por capas anfífilas pueden proporcionar ventajas terapéuticas liberando el fármaco encapsulado y el polímero en los momentos apropiados.
Tal como se usa en el presente documento, el término "anfífilo" se refiere a una propiedad en la que una molécula tiene tanto una porción polar como una porción no polar. A menudo, un compuesto anfífilo tiene una cabeza polar unida a una larga cola hidrófoba. Los ejemplos específicos de compuestos anfífilos incluyen, pero no se limitan a, fosfolípidos, tales como 1,2diestearoil-sn-glicero-3-fosfoetanolamina (DSPE), dipalmitoilfosfatidilcolina (DPPC), diestearoilfosfatidilcolina (DSPC), diaraxidoilfosfatidilcolina (DAPC), dibehenoilfosfatidilcolina (DBPC), ditricosanoilfosfatidilcolina (DTPC), y dilignoceroilfatidilcolina (DLPC), incorporado en una proporción de entre 0,01-60 (peso de lípido/peso de polímero), más preferentemente entre 0,1-30 (peso de lípido/peso de polímero). Los fosfolípidos que se pueden usar incluyen, pero sin limitación, ácidos fosfatídicos, fosfatidil colinas con lípidos saturados e insaturados, fosfatidil etanolaminas, fosfatidilgliceroles, fosfatidilserinas, fosfatidilinositoles, derivados lisofosfatidilos, cardiolipina y fosfolípidos β-acil-yalquilo. Los ejemplos de fosfolípidos incluyen, pero no se limitan a, fosfatidilcolinas tales como dioleoilfosfatidilcolina, dimiristoilfosfatidilcolina, dipentadecanoilfosfatidilcolina, dilauroilfosfatidilcolina, dipalmitoilfosfatidilcolina (DPPC), diestearoilfosfatidilcolina (DSPC), diaraquidoilfosfatidilcolina (DAPC), dibehenoilfosfatidilcolina (DBPC), ditricosanoilfosfatidilcolina (DTPC), dilignoceroilfatidilcolina (DLPC); y fosfatidiletanolaminas tales como dioleoilfosfatidiletanolamina o 1-hexadecil-2-palmitoilglicerofosfetoetanolamina. También pueden usarse fosfolípidos sintéticos con cadenas acilo asimétricas (por ejemplo, con una cadena acilo de 6 carbonos y otra cadena acilo de 12 carbonos).
En una realización particular, un componente anfífilo que puede usarse para formar una capa anfífila es la lecitina y, en particular, fosfatidilcolina. La lecitina es un lípido anfífilo y, como tal, forma una bicapa fosfolipídica que tiene las cabezas hidrófilas (polares) orientadas a su entorno, que son a menudo acuosas, y las colas hidrófobas enfrentadas entre sí. La lecitina tiene una ventaja de ser un lípido natural que está disponible de, por ejemplo, soja, y ya tiene aprobación de la FDA para su uso en otros dispositivos de administración. Además, una mezcla de lípidos tales como la leticina, es más ventajosa que un único lípido puro.

Agentes terapéuticos
Cualquier agente ("carga útil"), incluyendo, por ejemplo, agentes terapéuticos (por ejemplo, agentes anticancerosos), agentes de diagnóstico (por ejemplo, agentes de contraste; radionucleidos; y restos fluorescentes, luminiscentes y magnéticos), agentes profilácticos (por ejemplo, vacunas), y/o agentes nutracéuticos (por ejemplo, vitaminas, minerales, etc.) pueden administrarse por las nanopartículas descritas en el presente documento. Los agentes ejemplares a administrar incluyen, pero sin limitación, moléculas pequeñas (por ejemplo, agentes citotóxicos), ácidos nucleicos (por ejemplo, agentes de ARNsi, ARNi y microARN), proteínas (por ejemplo, anticuerpos, péptidos, lípidos, carbohidratos, hormonas, metales, elementos y compuestos radiactivos, fármacos, vacunas, agentes inmunológicos, etc. y/o combinaciones de los mismos. En algunas realizaciones, el agente a administrar es un agente útil en el tratamiento del cáncer (por ejemplo, cáncer de próstata).
Por ejemplo, el resto de direccionamiento puede dirigirse a o provocar que la partícula se localice en partes específicas dentro de un sujeto, y la carga útil puede administrarse a esas partes. Por ejemplo, una porción de direccionamiento puede provocar que las partículas se localicen en un tumor, un sitio patológico, un tejido, un órgano, un tipo de célula, etc. dentro del cuerpo de un sujeto, dependiendo del resto de direccionamiento usado. emparejamientos erróneos en el centro del ARNsi son más críticos y esencialmente suprimen la escisión del ARN diana. Por el contrario, los nucleótidos 3' del ARNsi no contribuyen significativamente a la especificidad del reconocimiento de la diana. Generalmente, los restos en el extremo 3' de la secuencia de ARNsi que es complementaria al ARN diana (por ejemplo, la secuencia de guía) no son críticos para la escisión del ARN diana.
La identidad de secuencia puede determinarse fácilmente por comparación de secuencias y algoritmos de alineamiento conocidos en la técnica. Para determinar el porcentaje de identidad de dos secuencias de ácido nucleico (o de dos secuencias de aminoácidos), las secuencias se alinean con el fin de una comparación óptima (por ejemplo, pueden introducirse huecos en la primera secuencia o en la segunda secuencia para un alineamiento óptimo). Los nucleótidos (o restos de aminoácidos) en posiciones de nucleótidos (o aminoácidos) correspondientes se comparan entonces. Cuando una posición en la primera secuencia está ocupada por el mismo resto que la posición correspondiente en la segunda secuencia, entonces las moléculas son idénticas en esa posición. El porcentaje de identidad entre las dos secuencias está en función del número de posiciones idénticas compartidas por las secuencias (es decir, % de homología = n.º de posiciones idénticas/n.º total de posiciones x 100), penalizando opcionalmente la puntuación por el número de huecos introducidos y/o la longitud de los huecos introducidos.
La comparación de secuencias y la determinación del porcentaje de identidad entre dos secuencias pueden efectuarse usando un algoritmo matemático. En una realización, el alineamiento se generaba a lo largo de una porción determinada de la secuencia alineada que tenía una identidad suficiente pero no a lo largo de porciones que tenían un bajo grado de identidad (es decir, un alineamiento local). Un ejemplo preferido no limitante de un algoritmo de alineamiento local utilizado para la comparación de secuencias es el algoritmo de Karlin y Altschul (1990) Proc. NatL Acad Sci. USA 87:2264-68, modificado como en Karlin y Altschul (1993) Proc. NatL Acad. Sci. USA 90:5873. Dicho algoritmo se incorpora en los programas BLAST (versión 2.0) de Altschul, y col. (1990) J Mol Biol. 215:403-10.
En otra realización, el alineamiento se optimiza por introducción de huecos apropiados y el porcentaje de identidad se determina a lo largo de la longitud de las secuencias alineadas (es decir, un alineamiento con huecos). Para obtener alineamientos con huecos con fines comparativos, puede utilizarse el Gapped BLAST como se describe en Altschul y col., (1997) Nucleic Acids Res. 25(17):3389. En otra realización, el alineamiento se optimiza introduciendo huecos apropiados y el porcentaje de identidad se determina a lo largo de la longitud completa de las secuencias alineadas (es decir, un alineamiento global). Un ejemplo preferido no limitante de un algoritmo matemático utilizado para la comparación global de secuencias es el algoritmo de Myers y Miller, CABIOS (1989). Dicho algoritmo se incorpora en el programa ALIGN (versión 2.0) que es parte del paquete de programas informáticos de alineamiento de secuencias GCG. Cuando se utiliza el programa ALIGN para comparar secuencias de aminoácidos, puede usarse una tabla de residuos de peso PAM120, una penalización por longitud de huecos de 12, y una penalización por huecos de 4.
Se prefiere una identidad de secuencia superior al 90 %, por ejemplo, 91 %, 92 %, 93 %, 94 %, 95 %, 96 %, 97 %, 98 %, 99 % o incluso el 100 % de identidad de secuencia, entre el ARNsi y la porción del ARNm diana. Como alternativa, el ARNsi puede definirse funcionalmente como una secuencia de nucleótidos (o secuencia oligonucleotídica) que es capaz de hibridar una porción del transcrito de ARNm diana (por ejemplo, NaCI 400 mM, PIPES 40 mM, pH 6,4, EDTA 1 mM, hibridación a 50 ºC o 70 ºC durante 12-16 horas; seguida de lavado). En una realización, las moléculas de ARNsi se modifican para mejorar la estabilidad en suero o en medio de cultivo para cultivos celulares. Para aumentar la estabilidad, los residuos 3' pueden estabilizarse frente a la degradación, por ejemplo, pueden seleccionarse de modo que consistan en nucleótidos de purina, particularmente nucleótidos de adenosina o guanosina. Como alternativa, la sustitución de nucleótidos de pirimidina por análogos modificados, por ejemplo, la sustitución de uridina por 2'-desoxitimidina se tolera y no afecta a la eficacia de la interferencia del ARN. Por ejemplo, la ausencia de un 2'-hidroxilo puede aumentar significativamente la resistencia a nucleasa de los ARNsi en medio de cultivo de tejidos.
En otra realización, la molécula de ARNsi puede contener al menos un análogo de nucleótido modificado. Los análogos de nucleótidos pueden localizarse en posiciones en las que la actividad específica de diana, por ejemplo, la actividad de mediación de la ARNi no se realiza sustancialmente, por ejemplo, en una región en el extremo 5' y/o el extremo 3' de la molécula de ARN. Los análogos de nucleótidos incluyen ribonucleótidos modificados con azúcares y/o en su cadena principal (es decir, incluyen modificaciones en la cadena principal de fosfato-azúcar). En ribonucleótidos modificados con azúcares preferidos, el grupo 2'-OH se sustituye por un grupo seleccionado de H, OR, R, halo, SH, SR, NH2, NHR, NR2 o NO2, en los que R es alquilo C1-C6, alquenilo o alquinilo y halo es F, Cl, Br o I.
Los análogos de nucleótidos también incluyen ribonucleótidos de nucleobases modificadas, es decir, ribonucleótidos que contienen al menos una nucleobase de origen no natural en lugar de una nucleobase de origen natural. Las bases pueden modificarse para bloquear la actividad de la adenosina desaminasa. El ARN puede producirse enzimáticamente o mediante síntesis orgánica parcial/total, cualquier ribonucleótido modificado puede introducirse mediante síntesis enzimática u orgánica in vitro. En una realización, un ARNsi se prepara químicamente. Se conocen en la técnica procedimientos de síntesis de moléculas de ARN, en particular, los procedimientos de síntesis química que se describen en Verina y Eckstein (1998), Annul Rev. Biochem. 67:99. En otra realización, un ARNsi se prepara enzimáticamente. Por ejemplo, un ARNsi puede prepararse por procesamiento enzimático de un ARN bicatenario largo que tenga una complementariedad suficiente con el ARNm diana deseado. El procesamiento del ARN largo puede efectuarse in vitro, por ejemplo, usando lisados celulares apropiados, y los ARNsi pueden purificarse posteriormente mediante electroforesis en gel o filtración en gel. Después, el ARNsi puede desnaturalizarse de acuerdo con metodologías reconocidas en la técnica. En una realización ejemplar, el ARNsi puede purificarse a partir de una mezcla por extracción con un disolvente o resina, precipitación, electroforesis, cromatografía o una combinación de los mismos. Como alternativa, el ARNsi puede usarse con ninguna o con un mínimo de purificación para evitar pérdidas debido al procesamiento de la muestra.
Como alternativa, los ARNsi también pueden prepararse por transcripción enzimática a partir de plantillas de ADN sintético o a partir de plásmidos de ADN aislados a partir de bacterias recombinantes. Típicamente, se usan ARN polimerasas de fago tales como ARN polimerasa de T7, T3 o SP6 (Milligan y Uhlenbeck (1989) Methods EnzynioL 180:51-62). El ARN puede secarse para su almacenamiento o disolverse en una solución acuosa. La solución puede contener tampones o sales para inhibir la hibridación y/o promover la estabilización de las dobles cadenas.
Las herramientas de diseño y kits disponibles en el mercado, tales como las disponibles en Ambion, Inc. (Austin, TX) y el Whitehead Institute of Biomedical Research en el MIT (Cambridge, MA), permiten el diseño y la producción de ARNsi. A modo de ejemplo, una secuencia de ARNm deseada puede introducirse en un programa de secuencias que generará secuencias de cadena diana con sentido y antisentido. Estas secuencias pueden introducirse después en un programa que determine las plantillas oligonucleotídicas de ARNsi de sentido y antisentido. Los programas también pueden usarse para añadir, por ejemplo, insertos de tipo horquilla o secuencias de cebadores de promotor T1. También pueden emplearse después kits para construir casetes de expresión de ARNsi.
En diversas realizaciones, los ARNsi se sintetizan in vivo, in situ, e in vitro. La ARN polimerasa endógena de la célula puede mediar la transcripción in vivo o in situ, o puede usarse ARN polimerasa clonada para la transcripción in vivo o in vitro. Para la transcripción de un transgen in vivo o una construcción de expresión, una región reguladora (por ejemplo, promotor, potenciador, silenciador, donador y aceptor de corte y empalme, poliadenilación) para transcribir los ARNsi. La inhibición puede dirigirse por transcripción específica en un órgano, tejido o tipo celular; la estimulación de una condición ambiental (por ejemplo, infección, estrés, temperatura, inductores químicos); y/o transcripción por ingeniería genética en una fase o edad del desarrollo. Un organismo transgénico que expresa ARNsi a partir de una construcción recombinante puede producirse por introducción de la construcción en un cigoto, una célula madre embrionaria, u otra célula multipotente derivada del organismo apropiado.
En una realización, el ARNm diana especifica la secuencia de aminoácidos de al menos una proteína tal como una proteína celular (por ejemplo, una proteína nuclear, citoplasmática, transmembrana o asociada a membrana). En otra realización, el ARNm diana de la invención especifica la secuencia de aminoácidos de una proteína extracelular (por ejemplo, una proteína de la matriz extracelular o proteína secretada). Tal como se usa en el presente documento, la expresión "especifica la secuencia de aminoácidos" de una proteína significa que la secuencia de ARNm se traduce en la secuencia de aminoácidos de acuerdo con las normas del código genético. CBL, CSFIR, ERBA, ERBB, EBRB2, ERBB2, ERBB3, ETSI, ETSI, ETV6, FGR, FOS, FYN, HCR, HRAS, JUN, KRAS, LCK, LYN, MDM2, MLL, MYB, MYC, MYCLI, MYCN, NRAS, PIM 1, PML, RET, SRC, TALI, TCL3 y YES);

Ejemplo 4: Unión/captación de nanopartículas mediada por resto de direccionamiento de molécula pequeña en células LNcap
La unión y captación de nanopartículas (NP-GL1, NP-GL2) con ligandos unidos a superficie GL1 (basado en ácido glutámico/4-amino-fenilalanina) y GL2 (basado en ácido glutámico/lisina) por células LNCap que expresan altos 5 niveles de PSMA se ensayó por comparación con nanopartículas (NP) de PLGA-PEG desnudas como control negativo y NP que llevaban aptámero (Apt) de antígeno de membrana específico de próstata (PSMA) A10 terminado en amina (NP-Apt) como control positivo. La captación de NP-GL1, NP-GL2, NP y NP-Apt por células LNCap y células PC3 que expresan bajos niveles de PSMA se comparó con la unión/captación mediada por PSMA específica de NP-GL1, NP-GL2.
