Abstract
Procedimiento para preparar (1R,2R)-3-(3-dimetilamino-1-etil-2-metilpropil)- fenol de fórmula (IV)**Fórmula** o una sal de adición de ácido del mismo, comprendiendo el proceso las siguientes etapas: Etapa a): preparar un compuesto de fórmula (II)**Fórmula** opcionalmente en forma de una sal de adición de ácido, haciendo reaccionar un compuesto de fórmula general (I)**Fórmula** con bromuro de etil-magnesio o cloruro de etil-magnesio en un éter orgánico seleccionado de entre el grupo consistente en dietil éter, tetrahidrofurano, 2- metiltetrahidrofurano, tert-butil metil éter o cualquier mezcla de los mismos, bajo condiciones de Grignard; Etapa b): preparar un compuesto de fórmula general (III)**Fórmula** opcionalmente en forma de una de adición de ácido, donde el paso b) se desarrolla: b') sometiendo el compuesto de fórmula general (II) a una deshidratación y b'') hidrogenando el compuesto así obtenido de fórmula general (VII)**Fórmula** empleando un catalizador en un medio de reacción inerte en presencia de hidrógeno: Etapa c): desproteger el compuesto de fórmula (III) para obtener (1R,2R)-3- (3-dimetilamino-1-etil-2-metilpropil)fenol de fórmula (V) y opcionalmente una etapa d), esto es convertir el compuesto de fórmula (IV) en una sal de adición, donde, en los compuestos arriba citados de fórmulas generales (I), (II) y (III), R representa metilo, bencilo o tetrahidropiranilo.

10 Son adecuadas para su uso en la deshidratación, en particular, aquellas resinas de intercambio iónico que tienen grupos ácido sulfónico.
Tienen preferencia las resinas de intercambio iónico basadas en copolímeros de tetrafluoroetileno/perfluorovinil éter, opcionalmente en forma de sus nanocompuestos de sílice, tal como se describen, por ejemplo, en las publicaciones
15 de Olah et al., Synthesis, 1996, 513-531 y Harmer et al., Green Chemistry, 2000, 714, cuyas descripciones correspondientes se incorporan aquí por referencia y forman parte de la descripción. Los productos correspondientes están disponibles comercialmente, por ejemplo bajo el nombre Nafion®, y también se pueden utilizar en aquella forma en los procedimientos de acuerdo con la presente invención.
20 Además, son preferentes resinas de intercambio iónico basadas en copolímeros de estireno/divinilbenceno, las cuales se pueden preparar por procedimientos convencionales conocidos por los expertos en la técnica.
De forma particularmente preferente, para la deshidratación se tienen en cuenta resinas de intercambio iónico que tienen un grupo ácido sulfónico basadas en 25 copolímeros de estireno/divinilbenceno, tales como las comercializadas, por ejemplo, bajo el nombre Amberlyst®, Rohm & Haas, y que también se pueden utilizar como tales en los procedimientos de acuerdo con la presente invención. Estas resinas de intercambio iónico se distinguen en particular por su estabilidad frente al agua y alcoholes incluso a temperaturas elevadas, por ejemplo de 130 a
El grado de reticulación y la estructura de estas resinas de intercambio iónico puede variar. Por ejemplo, se pueden mencionar resinas de intercambio iónico macroporosas con una distribución de diámetro de poro heterogénea, resinas de intercambio iónico isoporosas con una distribución de diámetro de poro virtualmente 35 uniforme o resinas de intercambio iónico tipo gel que no tienen o que virtualmente
no tienen poros. Las resinas macroporosas, en particular, se pueden utilizar ventajosamente para catalizadores heterogéneos en la fase líquida.
Resinas macroporosas particularmente adecuadas con un diámetro de poro medio de 20 a 30 nm y una concentración mínima de grupos activos de 4,70 a 5,45 equivalentes por kg de resina están disponibles comercialmente bajo los nombres Amberlyst® 15, Amberlyst® 35 y Amberlyst® 36, y, en consecuencia, también se pueden utilizar en los procedimientos de acuerdo con la presente invención.
De igual manera, preferentemente la deshidratación se realiza en presencia de un catalizador ácido basado en óxidos de metal, por ejemplo SiO2, Al2O3, TiO2, Nb2O5, B2O3 o basados en óxidos de metal mixtos, por ejemplo Al2O3/SiO2 o Al2O3/B2O3.
Preferiblemente, la temperatura para la deshidratación (b') cuando se utiliza un catalizador ácido, como se describe anteriormente, en cada caso es de 20 a 250ºC, de manera particularmente preferible de 50 a 180ºC y de manera muy particularmente preferible de 100 a 160ºC.
La proporción entre el catalizador ácido y el compuesto de fórmula general (II) preferiblemente está en el intervalo de 1:200 a 1:1, en particular de 1:4 a 1:2.
Después de la deshidratación, el catalizador se puede separar de la mezcla de reacción de forma sencilla, preferiblemente por filtración. El compuesto particular de fórmula general (VII) obtenido se aísla y/o purifica por métodos convencionales conocidos por el experto en la técnica.
Alternativamente, la etapa de deshidratación (b') también se puede llevar a cabo sometiendo un compuesto de fórmula general (II) a un exceso de cloruro de tionilo, opcionalmente en un medio de reacción, preferiblemente en un medio de reacción seleccionado del grupo consistente en dietil éter, tetrahidrofurano, tolueno, 2metiltetrahidrofurano, dioxano, tert-butil metil éter y mezclas de los mismos, y calentamiento subsecuente de la mezcla de reacción así obtenida de 40ºC a 120ºC, preferiblemente de 80ºC a 120ºC.
La hidrogenación de la etapa (b") también se puede llevar a cabo vía catálisis heterogénea con hidrógeno. El hidrógeno preferiblemente está en forma gaseosa, aunque también es posible que al menos parte del mismo esté disuelto en una fase liquida.
La catálisis heterogénea en el contexto de la presente invención significa que los catalizadores utilizados en la etapa (b") en cada caso están presentes en el estado sólido de agregación.
Preferiblemente, el catalizador heterogéneo utilizado para hidrogenación en la etapa (b’’) según la presente invención contiene uno o más metales de transición; estos metales preferiblemente se pueden seleccionar del grupo consistente en Cu, Ag, Au, Zn, Cd, Hg, V, Nb, Ta, Cr, Mo, W, Fe, Ru, Os, Co, Rh, Ir, Ni, Pd, Pt, de
5 manera particularmente preferible del grupo consistente en Ru, Rh, Pd, Pt y Ni.
Los catalizadores correspondientes preferiblemente pueden contener uno o más de los metales de transición mencionados anteriormente en estados de oxidación iguales o diferentes. También puede ser preferible que el catalizador correspondiente contenga uno o más de los metales de transición mencionados en
10 dos o más estados de oxidación diferentes.
La preparación de catalizadores dopados con metales de transición se puede llevar a cabo por procedimientos convencionales conocidos por el experto en la técnica.
Preferiblemente, el catalizador utilizado para hidrogenación en la etapa (b’’) se selecciona del grupo consistente en níquel de Raney, paladio, paladio sobre 15 carbono (1 -10% en peso, preferiblemente 5% en peso), platino, platino sobre carbono (1 -10% en peso, preferiblemente 5% en peso), rutenio sobre carbono (1 -10% en peso, preferiblemente 5% en peso) y rodio sobre carbono (1 -10% en peso, preferiblemente 5% en peso), de manera más preferible paladio sobre carbono (1 -10% en peso, preferiblemente 5% en peso) para catalizador de
20 hidrogenación en la etapa (b’’).
Los compuestos de fórmula general VII o III utilizados en la etapa (b'’) según la presente invención preferiblemente están en fase líquida y, para ello, preferiblemente se mezclan o se disuelven en un medio de reacción líquido bajo las condiciones de reacción particular.
25 Ejemplos de medios de reacción adecuados son metanol, etanol, isopropanol, tolueno, heptano, hexano, pentano, ácido acético, acetato de etilo, ácido fórmico, ácido clorhídrico, ácido bromhídrico, ácido sulfúrico y mezclas de los mismos. De manera más preferible se utiliza etanol corno medio de reacción en la etapa (b"). Por supuesto, también es posible utilizar mezclas o sistemas multifásicos que estén
30 constituidos por dos o más de los líquidos mencionados anteriormente en los procedimientos según la presente invención. También es posible una reacción en CO2 supercrítico como disolvente.
Los parámetros de reacción para la hidrogenación heterogénea en la etapa (b’’), tales como presión, temperatura o tiempo de reacción, pueden variar en un amplio
35 intervalo.
Preferiblemente, la temperatura durante la hidrogenación heterogénea en la etapa (b’’) en cada caso es de 0 a 250ºC, de manera particularmente preferible de 15 a 180ºC y de manera muy particularmente preferible de 15 a 30ºC.
La hidrogenación heterogénea en la etapa (b’’) preferiblemente se puede llevar a
5 cabo a presión reducida, a presión normal o a presión elevada, preferiblemente en el intervalo de 1 a 300 bar. Con particular preferencia, las reacciones se llevan a cabo bajo una presión en un intervalo de 2 a 10 bar, en particular de 4 a 10 bar.
El tiempo de reacción puede variar en base a parámetros diversos, tales como la temperatura, la presión, la naturaleza del compuesto a reaccionar o las propiedades
10 del catalizador y puede ser determinado para el procedimiento en cuestión por el experto en la técnica utilizando ensayos preliminares.
También es posible la separación continua de muestras con el fin de controlar la reacción, por ejemplo mediante métodos de cromatografía de gas, opcionalmente en combinación con la regulación de los parámetros de procedimiento
15 correspondientes.
La cantidad total de catalizador(es) utilizada depende de uno o más de los diversos factores, tales como la proporción del componente catalíticamente activo respecto a cualquier material inerte presente o la naturaleza de la superficie de los uno o más catalizadores. La cantidad óptima del uno o más catalizadores para una
20 reacción particular puede ser determinada por el experto en la técnica utilizando ensayos preliminares.
El compuesto particular de fórmula general (III) obtenido se puede aislar y/o purificar por métodos convencionales conocidos del experto en la técnica.
En otra realización de la invención, la etapa b (Esquema 1) es una reacción de
25 sustitución directa del grupo OH por H, que preferiblemente se lleva a cabo en una reacción de un recipiente. En especial, se sustituye un OH-por H-.
Las etapas de acuerdo con la presente invención se pueden llevar a cabo en cada caso de forma discontinua (por lotes) o continua, siendo preferente el proceso discontinuo.
30 Como reactor para el proceso discontinuo se toma en consideración, por ejemplo, un reactor de suspensión y, para el procedimiento continuo, un reactor de lecho fijo
o un reactor de ciclo.
A continuación se describe un procedimiento para la preparación de clorhidrato de (1R,2R)-3-(3-dimetilamino-1-etil-2-metilpropil)fenol.

Etapa (a"): Preparación de (S)-3-(dimetilamino)-1-(3-metoxifenil)-2-metilpropan-1ona (la)
1. a. Preparación de (2R,3R)-O,O'-dibenzoiltartrato de (S)-3-(dimetilamino)-1-(3metoxifenil)-2-metilpropan-1-ona en acetona
5 Se disuelve ácido (2R,3R)-O,O'-dibenzoiltartárico monohidratado (189,1 g, 0,5 mol) en acetona (550 ml) en una planta de reacción de 2 l equipada con agitador mecánico, equipo de medida de temperatura y baño de aceite y se agrega la 3(dimetilamino)-1-(3-metoxifenil)-2-metilpropan-1-ona (110,6 g, 0,5 mol). La mezcla de reacción se calienta de 35ºC a 40ºC durante 27 horas y se deja enfriar a 25ºC.
10 La suspensión se extrae por efecto sifón y se obtiene el (2R,3R)-O,O'dibenzoiltartrato de (S)-3-(dimetilamino)-1-(3-metoxifenil)-2-metilpropan-1-ona como un sólido incoloro (233,2 g, 80,5%, ee 96,9%, ee = exceso enantiomérico).
1. b. Preparación de (2R,3R)-O,O'-dibenzoiltartrato de (S)-3-(dimetilamino)-1-(3metoxifenil)-2-metilpropan-1-ona en acetona/metanol

La suspensión se separa por efecto sifón y se obtiene (2R,3R)-O,O’-dibenzoiltartrato de (S)-3-(dimetilarnino)-1-(3-metoxifenil)-2metilpropan-1-ona como un sólido incoloro (2,38 kg, 74%, ee 98,4%).
2. Preparación de (S)-3-(dimetilamino}-1-(3-metoxifenil)-2-metilpropan-1-ona (la)
25 Se suspende (2R,3R)-O,O’-dibenzoiltartrato de (S)-3-(dimetilamino)-1-(3metoxifenil)-2-metilpropan-1-ona (968 g, 1,67 mmol, ee 98%) en 6 l de tert-butil metil éter en un recipiente de camisa doble de 10 l equipado con agitador eléctrico, línea de transición de gas, equipo de medición de temperatura Pt100 enfriamiento/calentamiento basado en un sistema de aceite y se agrega dietilamina
30 (384 g, 5,25 mol). La mezcla de reacción se agita entre 20ºC y 25ºC durante 90 minutos y el sólido se separa por efecto sifón. El filtrado se concentra a una temperatura de 40ºC a vacío hasta que se alcanza una presión de 4 mbar. Se obtiene la (S)-3-(dimetilamino)-1-(3-metoxifenil)-2-metilpropan-1-ona (356,7 g, 96,5%, ee 98%) como un aceite incoloro.
35 Etapa (a): Preparación de (2S,3R)-1-(dimetilamino)-3-{3-metoxifenil)-2metilpentan-3-ol (lIa)
2. El clorhidrato de (2S,3R)-1-(dimetilamino)-3-(3-metoxifenil)-2-metilpentan-3-ol obtenido como se describe anteriormente se dispone en un matraz de tres bocas de 250 mI equipado con termómetro, agitador de aire comprimido mecánico, condensador de reflujo y baño de aceite y se agrega una disolución acuosa de 5 cloruro de hidrógeno (150 ml, 36% en peso). La mezcla de reacción se calienta a 55ºC durante 5 horas y se deja enfriar a 20ºC. Se agrega una disolución acuosa de hidróxido de sodio (33% en peso) mientras se enfría hasta alcanzar un pH 11. Se agregan 150 ml de acetato de etilo, la mezcla de reacción se agita durante 10 minutos, se separan las fases y se extrae el acetato de etilo al vacío a 60ºC hasta
10 alcanzar una presión de 10 mbar. Se obtiene (R)-3-(3-metoxifenil)-N,N,2trimetilpent-3-eno-1-amina (21 g, 90%) como un residuo oleoso (proporción Z/E 4,5:1).
Etapa (b"): Preparación de clorhidrato de (2R,3R)-3-(3-metoxifenil}-N,N,2trimetilpentan-1-amina (IIIa)
15 1. Se disuelve (R)-3-(3-metoxifenil)-N,N,2-trimetilpent-3-eno-1-amina (5 kg, 21,43 mmol) en 13 l de etanol seco a una temperatura de 25ºC y con una frecuencia de agitación rotacional de 850 ± 150 por minuto en un aparato de hidrogenación de camisa doble equipado con una tapa montada estacionaria, con suministro de hidrógeno y nitrógeno, agitador de gasificación eléctrico, equipo de medición de
20 temperatura Pt100, controlador e inspección de vidrio y gas "Büchi bpc". El aparato de hidrogenación se purga con nitrógeno. Se suspende paladio sobre carbono vegetal (375 g, 5% en peso) en cloruro de hidrógeno acuoso (675 g, 32% en peso) y se agrega a la mezcla de reacción. El aparato de hidrogenación se purga nuevamente con nitrógeno y la reacción se lleva a cabo una presión nominal de
25 hidrógeno de 5 bar y una presión de hidrógeno interna de 1 bar hasta que finaliza la reacción. El aparato de hidrogenación se purga con nitrógeno y el catalizador se separa por filtración sobre un filtro estratificado con tierra de filtración. El filtrado se concentra al vacío. El residuo se recoge en acetato de etilo y se agrega hidróxido de sodio acuoso (10% en peso, 3,7 l) a 20ºC hasta alcanzar un pH de 10 a 12. La
30 fase orgánica se concentra al vacío entre 45ºC y 50°C hasta alcanzar una presión de 5 mbar. El residuo oleoso es una mezcla de (2R,3R)-3-(3-metoxifenil)-N,N,2trimetilpentan-1-amina y (2R,3S)-3-(3-metoxifenil)-N,N,2-trimetilpentan-1-amina (4,5 kg, 95%, proporción 5,5(R,R):1(R,S)).
