Es wird ein Verfahren zur Herstellung von 3-Brommethylbenzoesäuren der allgemeinen Formel (II) durch Bromierung der entsprechenden 3-Methylbenzoesäuren beschrieben. Weiterhin werden Verbindungen der allgemeinen Formel II beschrieben, worin R<1> für Fluor, Chlor oder Brom, und R<2> für (C1-C4)-Alkyl stehen.

Beschreibung
Verfahren zur Herstellung von 3-Brommethylbenzoesäuren
Die Erfindung betrifft ein Verfahren zur Herstellung von 3-Brommethylbenzoesäuren durch Bromierung der entsprechenden 3-Methylbenzoesäuren. Weiterhin betrifft sie bestimmte 3-Brommethylbenzoesäuren.
Derivate der 3-Brommethylbenzoesäure stellen wertvolle Edukte in der Synthese von bestimmten Herbiziden dar. Brommethylaromaten können grundsätzlich durch Seitenkettenbromierung aus den entsprechenden Methylaromaten hergestellt werden, vergl. dazu Houben-Weyl, Bd. 5, Seite 331 ff. (1960). Es ist jedoch bekannt, daß eiektronegative Substituenten, wie Carboxyl, Alkylcarbonyl, Cyano und Nitro die Reaktion deutlich behindern, sodaß nur schlechte Ausbeuten erzielbar sind. Aus WO 99/06339 ist ein Verfahren zur Herstellung von substituierten Benzylbromiden durch Bromierung der entsprechenden Methylaromaten in Anwesenheit von Azocarbonsäureestem oder -nitrilen und in Anwesenheit eines Oxidationsmittels bekannt. Dabei ist ein Substituent elektronegativ und stammt aus der Gruppe Fluor, Chlor, Brom, Alkoxycarbonyl, Cyano und Nitro. Nachteile dieses Verfahrens sind die Verwendung eines zusätzlichen Oxidationsmittels und die nicht immer ausreichenden Ausbeuten. In EP-A 0 292 944 ist die Herstellung von 3-Brommethyl- 2-chlor-4-methylsulfonyl-benzoesäuremethlester durch Radikalstarter induzierte Bromierung von 2-Chlor-3-methyl-4-methylsulfonyl-benzoesäure-methylester in Tetrachlorkohlenstoff beschrieben. Aus dieser letztgenannten Verbindung wird dann durch Verseifung die freie Säure 3~Brommethyl-2-ch!or-4-methylsu!fonyl- benzoesäure hergestellt.
Wünschenswert sind, gerade in Hinblick auf die Herstellung von bestimmten Herbiziden, Verfahren zur direkten Herstellung von 3-Brommethylbenzoesäuren.
Es wurde nun gefunden, daß 2-Halogen-3-methyl-4-alkyIsulfonyl-benzoesäuren in sehr guten Ausbeuten und in sehr hohen Reinheiten durch Bromierung zu den
entsprechenden 3-Brommethyl-2-halogen-4-alkylsulfonyl-benzoesäuren umgesetzt werden können.
Ein Gegenstand der vorliegenden Erfindung ist somit ein Verfahren zur Herstellung von 3-Brommethylbenzoesäuren der allgemeinen Formel II
durch Bromierung von 3-Methylbenzoesäuren der allgemeinen Formel
mit
A) N-Bromsuccimid in Gegenwart eines Radikalstarters, oder
B) elementarem Brom und Bestrahlen mit einer Photolampe,
worin in Formeln I und II
R1 für Fluor, Chlor oder Brom, und
R2 für (C C4)-Alkyl stehen.
Als Radikalstarter für die Bromierung nach Variante A) sind handelsübliche und dem Fachmann bekannte Radikalstarter wie Diaroylperoxyde, Azocarbonsäureester und Azocarbonsäurenitrile geeignet. Beispielsweise seien Azoisobutyronitril und
Dibenzoylperoxyd genannt. Die Bestrahlung mit Licht nach Variante B) erfolgt mit handelsüblichen Fotolampen und ist dem Fachmann grundsätzlich bekannt.
Neben den sehr hohen Ausbeuten und sehr hohen Reinheiten sind als weitere Vorteile des erfindungsgemäßen Verfahrens die Verwendung von weniger toxischen Lösungsmittel und die besonders einfache Aufarbeitung der Endprodukte zu sehen.
Für das erfindungsgemäße Verfahren haben sich solche Lösungsmittel als geeignet erwiesen, die gegenüber den Bedingungen der Bromierung als inert anzusehen sind. Dazu zählen beispielsweise halogenierte Kohlenwasserstoffe, wie Methylenchlorid, Chloroform, Tetrachlorkohlenstoff , 1 ,2-Dichlorethan und Chlorbenzol sowie Verbindungen wie Acetonitril. Als vorteilhaft haben sich für Verfahrensvariante A) die Lösungsmittel Chlorbenzol und Acetonitril und für Verfahrensvariante B) die Lösungsmittel Chlorbenzol, Methylenchlorid und 1 ,2-Dichlorethan herausgestellt. Selbstverständlich können auch Gemische dieser Lösungsmittel verwendet werden.
In beiden Verfahrensvarianten wird die Reaktion in der Regel bei einer Temperatur von 40 bis 100°C, vorzugsweise 70 bis 100°C durchgeführt. Im Fall der niedrig siedenden Lösungsmitteln Acetonitril, 1 ,2-Dichlorethan und Methylenchlorid arbeitet man vorzugsweise unter Rückfluß.
In Verfahrensvariante A) wird zweckmäßigerweise eine Verbindung der Formel I mit N-Bromsuccinimid (NBS) und Radikalstarter in Lösungsmittel vorgelegt und dann langsam erwärmt. Gegebenenfalls kann durch Zugabe von wenig Brom die Reaktion beschleunigt werden. NBS setzt man vorzugsweise im Überschuß ein.
In Verfahrensvariante B) wird zweckmäßigerweise eine Verbindung der Formel I in Lösungsmittel vorgelegt und dann nach Erwärmen unter Bestrahlen mit einer Fotolampe Brom zugetropft. Brom setzt man vorzugsweise im Überschuß ein.
Je nach Löslichkeiten der Verbindungen der Formeln I und II im verwendeten Lösungsmittel liegen diese Verbindungen vollständig oder teilweise gelöst vor. In der
Regel liegen sie teilweise gelöst vor, so daß ein Teil in suspendiert ist. Üblicherweise ist in beiden Verfahrensvarianten nach etwa 2 bis 6 Stunden die Reaktion vollständig abgelaufen. Der genaue Zeitpunkt des vollständigen Reaktionsablauf kann beispielsweise dünnschichtchromatographisch oder per HPLC verfolgt werden.
Zur Aufarbeitung läßt man das Reaktionsgemisch abkühlen. Erfolgte die Bromierung nach Verfahrensvariante A), so fügt man man zweckmäßigerweise zunächst Natriumbisulfit-Lösung und gegebenenfalls Lösungsmittel hinzu. Die weitere Aufarbeitung richtet sich dann nach der Löslichkeit und Reinheit des Produkts. Im Falle der Verfahrensvariante B) kann das Reaktionsprodukt in der Regel nach Abkühlen direkt abgesaugt werden. In beiden Fällen wird das Reaktionsprodukt gewaschen und falls erforderlich durch Umkristallisieren beispielsweise aus Ethylacetat oder Butylacetat weiter gereinigt.
Verbindungen der Formel II sind mit Ausnahme der aus EP-A 0 292 944 bekannten Verbindung 3-Brommethyl-2-chlor-4-methylsulfonyl-benzoesäure neu. Ein weiterer Gegenstand der vorliegenden Erfindung sind somit Verbindungen der Formel II,
worin
R1 für Fluor, Chlor oder Brom, und
R2 für (Cι-C4)-Alkyl stehen, und wobei die Verbindung 3-Brommethyl-2-chlor-4-methylsulfonyl-benzoesäure ausgenommen sein soll.
Die nachfolgenden Ausführungsbeispiele erläutern die Erfindung:
Beispiel 1
Herstellung von 3-Brommethyl-2-chlor-4-methylsulfonyl-benzoesäure
Verfahrensvariante A):
In einer 1 I Rührapparatur wurden 100 g 2-Chlor-3-methyl-4-sulfonylmethylbenzoe- säure in 400 ml Chlorbenzol suspendiert. Bei Raumtemperatur wurden 85,9 g
N-Bromsuccinimid und 6,6 g Azoisobutyronitril zugegeben und anschließend auf
60°C erwärmt. Danach addierte man 2,6 g Brom und erhitzte das Reaktionsgemisch weiter langsam auf 90°C. Es wurde noch 2 h bei 90°C nachgerührt, nach dieser Zeit zeigte eine HPLC-Analyse einen Umsatz von mehr als 98% an. Nach Abkühlen des
Reäktionsgemisches wurden nacheinander 100 ml einer 2%igen Natriumbisulfit-
Lösung und 400 ml Wasser zugegeben und gut verrührt. Die Suspension wurde filtriert, der Feststoff gewaschen und bei 60°C im Vakuum getrocknet.
Man erhielt 121 ,3 g (88,4% der Theorie) 3-Brommethyl-2-chlor-4-methylsulfonyl- benzoesäure.
Reinheit: 96% (HPLC) Festpunkt: 207-208 °C
Verfahrensvariante B):
In einem 250 ml Glaskolben wurden 5 g 2-Chlor-3-methyl-4-sulfonylmethyl- benzoesäure in 50 ml Methylenchlorid und 50 ml Wasser vorgelegt. Unter Bestrahlung mit einer 300W Lampe wurden während 3 h langsam 11 g Brom zudosiert, wobei das Gemisch unter Rückfluss siedete. Nach weiteren 2h Bestrahlen war das Reaktionsgemisch weitgehend entfärbt. Nach HPLC-Analyse enthielt das Reaktionsgemisch 87% Produkt und 10% Edukt.
Beispiel 2
Verfahrensvariante A):
Herstellung von 3-Brommethyl-2-chlor-4-ethylsulfonyI-benzoesäure
In einer 0,5 I Rührapparatur wurden 20 g 2-Chlor-3-methyl-4-sulfonylethylbenzoe- säure in 150 ml Acetonitril suspendiert. Bei Raumtemperatur wurden 17,6 g N-
Bromsuccinimid und 0,7 g Dibenzoylproxid zugegeben und anschließend auf
Rückfluß erhitzt. Nach jeweils 1 h wurden in 3 Portionen je 0,7 g Dibenzoylproxid weiter zugegeben und nach der letzten Zugabe noch 2 h nachgerührt. Nach dieser Zeit zeigte eine HPLC-Analyse vollständigen Umsatz an. Nach Abkühlen des Reaktionsgemisches wurden nacheinander 100 ml einer 2%igen Natriumbisulfit- Lösung und 300 ml Ethylacetat zugegeben. Die organische Phase wurde abgetrennt, gewaschen und im Vakuum eingeengt. Der feste Rückstand wurde in 50 ml Diethylether und 20 ml Heptan suspendiert, abfiltriert und getrocknet. Man erhielt 28,2 g (95,6% der Theorie) 3-Brommethyl-2-chlor-4-ethylsulfonyl- benzoesäure. Reinheit: 88% (HPLC) Festpunkt: 103°C
Claims
1. Verfahren zur Herstellung von 3-Brommethylbenzoesäuren der allgemeinen Formel II
durch Bromierung von 3-MethyIbenzoesäuren der allgemeinen Formel
mit
A) N-Bromsuccimid in Gegenwart eines Radikalstarters, oder
B) elementarem Brom und Bestrahlen mit einer Photolampe,
worin in Formeln I und II
R1 für Fluor, Chlor oder Brom, und
R2 für (d-C )-Alkyl stehen.
2. Verfahren nach Anspruch 1 , worin als Radikalstarter für die Bromierung nach Variante A) Azoisobutyronitril oder Dibenzoylperoxyd verwendet wird.
3. Verfahren nach Anspruch 1 oder 2, worin als Lösungsmittel für die Verfahrensvariante A) Chlorbenzol oder Acetonitril und für Verfahrensvariante B) Chlorbenzol oder Methylenchlorid eingesetzt wird.
4. Verfahren nach einem der Ansprüche 1 bis 3, worin die Reaktion bei einer Temperatur von 70 bis 100°C und im Falle von Acetonitril und Methylenchlorid als Lösungsmittel bei deren Rückflußtemperatur durchgeführt wird.
5. Verbindungen der Formel II ,
worin
R1 für Fluor, Chlor oder Brom, und
R2 für (C1-C )-Alkyl stehen, und wobei die Verbindung 3-Brommethyl-2-chlor-4-methylsulfonyl-benzoesäure ausgenommen sein soll.
