Abstract
Предложен способ и устройство, позволяющие одновременно определять реакционные свойства более чем 10000 из стимулированных антигеном лимфоцитов, расположенных на чипе, а также раздельно определять состояние индивидуальных клеток. Микролуночный чип включает множество лунок и слой покрытия на одной из основных поверхностей основного элемента, при этом лунки имеют такой размер, который позволяет проходить в каждую лунку только одной клетке. Слой покрытия вещества, способного к связыванию с веществом, вырабатываемым клетками, содержащимися в лунках, присутствует на основной поверхности вокруг лунок. Способ скрининга целевой клетки включает внесение образцов клеток и клеточной культуральной среды в лунки вышеуказанного микролуночного чипа; погружение слоя покрытия и лунок в культуральную среду и культивирование клеток в состоянии, допускающем диффузию веществ в культуральной среде из лунок в слой покрытия; добавление меченого вещества, специфично связывающегося с веществом, вырабатываемым целевой клеткой, присутствующей среди образцов клеток, на слое покрытия; и детектирование вещества, выработанного целевой клеткой, которое связалось с веществом в слое покрытия, посредством меченого вещества, с определением целевой клетки.
