Abstract

Изобретение описывает способ получения конъюгата "антитело-майтансиноид". Способ включает стадии: (а) приведение антитела и майтансиноида, включающего тиоловую группу, в контакт с образованием первой смеси, включающей антитело и майтансиноид, затем приведение первой смеси в контакт с бифункциональным реагентом, образующим перекрестные связи и включающим линкер, в растворе, имеющем pH от приблизительно 4 до приблизительно 9, с получением второй смеси, включающей (i) конъюгат "антитело-майтансиноид", в котором антитело химически соединено посредством линкера с майтансиноидом, (ii) несвязанный майтансиноид и (iii) побочные продукты реакции; и (б) очистка второй смеси, включающей конъюгат "антитело-майтансиноид", с получением очищенного конъюгата "антитело-майтансиноид". Также предложен способ получения конъюгата "антитело-майтансиноид", включающий стадии: (а) приведение антитела и майтансиноида, включающего тиоловую группу, в контакт с образованием первой смеси, включающей антитело и майтансиноид, затем приведение первой смеси в контакт с бифункциональным реагентом, образующим перекрестные связи и включающим линкер, в растворе, имеющем pH от 4 до 9, с получением второй смеси, включающей (i) конъюгат "антитело-майтансиноид", в котором антитело химически соединено посредством линкера с майтансиноидом, (ii) несвязанный майтансиноид и (iii) побочные продукты реакции; (б) приведение второй смеси в контакт с дополнительным майтансиноидом с образованием третьей смеси и затем приведение третьей смеси в контакт с дополнительным бифункциональным реагентом, образующим перекрестные связи, имеющим pH от приблизительно 4 до приблизительно 9, с образованием четвертой смеси; и (в) очистка четвертой смеси, включающей конъюгат "антитело-майтансиноид", с получением очищенного конъюгата "антитело-майтансиноид".
