Claims (14)
1. Fremgangsmåde til detektion af tilstedeværelsen af methicillin-resistente Staphylococcus aureus (MRSA) stammer af MREJ typen i, ii, iii og viii, hvilken fremgangsmåde omfatter, at: a) en prøve, der skal analyseres for tilstedeværelsen af MRSA stammer af MREJ typen i, ii, iii og viii, hvor hver af MRSA stammerne indeholder et Staphylococ kassette-kromosom mec {SCCmec) element, som indeholder et mecA gen indsat i et kromosomalt DNA, hvorved der genereres en polymorf højre-ekstremitet-forbindelsessekvens (MREJ) af typen i, ii, iii eller viii, som omfatter sekvenser fra både den højre ekstremitet af SCCmec elementet og kromosomalt DNA, der støder op til den højre ekstremitet af SCCmec elementet, bringes i kontakt med en første primer og en anden primer for hver af MREJ typen i, ii, iii og viii, hvor den første primer hybridiserer med den højre ekstremitet af SCCmec elementet af en MREJ type i, ii, iii eller viii sekvens udvalgt fra gruppen bestående af: SEQ ID NO: 1, 20 til 25, 41 og 199 og komplementer deraf, for MREJ typen i, SEQ ID NO: 2, 17 til 19, 26, 40, 173 til 183, 185, 186 og 197 og komplementer deraf, for MREJ typen ii, SEQ ID NO: 4 til 16, 104, 184 og 198 og komplementer deraf, for MREJ typen iii, og SEQ ID NO:167 og komplementet deraf, for MREJ typen viii; og hvor den anden primer hybridiserer med en kromosomal sekvens af S. aureus for specifikt at generere et amplikon (amplikoner), hvis en sådan MRSA stamme af MREJ typen i, ii, iii eller viii er til stede i prøven; og b) detektion af tilstedeværelsen af amplikonet (amplikonerne).
2. Fremgangsmåde ifølge krav 1, hvorved den kromosale sekvens af S. aureus er orfX.
3. Fremgangsmåde ifølge krav 1 eller 2, hvorved fremgangsmåden omfatter anvendelsen af mindst en primer og/eller sonde udvalgt fra følgende SEQ ID NO: 115, 116, 187, 188, 207, 208, 64, 71, 72, 73, 74, 75, 76, 70, 103, 130, 132, 158, 159, 59, 62, 126, 127, 128, 129, 131,200, 201,60, 61,63, 32, 83, 84, 160, 161, 162,163 og 164 til detektion af MREJ typen viii.
4. Fremgangsmåde ifølge krav 3, hvilken fremgangsmåde omfatter anvendelsen af et primerpar bestående af SEQ ID NO:64 og 115 eller 64 og 116.
5. Fremgangsmåde ifølge krav 4, hvilken fremgangsmåde desuden omfatter anvendelsen af mindst en sonde, som har en sekvens udvalgt fra gruppen bestående af SEQ ID NO:32, 83, 84, 160, 161,162, 163 og 164.
6. Fremgangsmåde ifølge krav 5, hvorved primerne og sonderne har følgende nukleotidsekvenser: SEQ ID NOs:64, 115, 116, 84, 163, 164 til detektion af MREJ typen viii.
7. Fremgangsmåde ifølge et af kravene 1 til 6, hvorved der anvendes flere primere og/eller sonder sammen i det samme fysiske indelukke.
8. Kit til detektion af tilstedeværelsen af MRSA stammer af MREJ typen i, ii, iii og viii i en prøve omfattende: a) et første sæt af oligonukleotider, som hybridiserer med den højre ekstremitet af SCCmec elementet af sekvenser af MREJ typen i, ii, iii og viii udvalgt fra gruppen bestående af SEQ ID NO: 1,20 til 25, 41 og 199 og komplementer deraf, for MREJ typen i, SEQ ID NO: 2, 17 til 19, 26, 40, 173 til 183, 185, 186 og 197 og komplementer deraf, for MREJ typen ii, SEQ ID NO: 4 til 16, 104, 184 og 198 og komplementer deraf, for MREJ typen iii, og SEQ ID NO:167 og komplementet deraf for MREJ typen viii; og b) et andet oligonukleotid, som hybridiserer med en kromosal sekvens af S. aureus; hvor oligonukleotiderne af a) og b) muliggør den selektive generering af et amplikon (amplikoner), som omfatter sekvenser fra både den højre ekstremitet af SCCmec elementet og kromosalt DNA, der støder op til den højre ekstremitet af MRSA stammer af MREJ typen i, ii, iii og viii.
9. Kit ifølge krav 8, hvor den specifikke sekvens af det S. aureus kromosale DNA er orfX.
10. Kit ifølge krav 8 eller 9, hvor det andet oligonukleotid i b) omfatter en sekvens udvalgt fra gruppen bestående af SEQ ID NO: 32, 59, 60 til 64, 70 til 76, 83, 84, 103, 126 til 132, 160 til 164, 200 og 201.
11. Kit ifølge krav 8 eller 9, omfattende et par af oligonukleotider bestående af SEQ ID NO: 64 og 115 eller 64 og 116.
12. Kit ifølge et af kravene 8 til 11, desuden omfattende mindst en sonde udvalgt fra gruppen bestående af SEQ ID NO: 32, 83, 84, 160, 161, 162, 163 og 164.
13. Fremgangsmåde ifølge et af kravene 1 til 7 eller kit ifølge et af kravene 8 til 12, hvorved den anden primer har en sekvens som angivet i SEQ ID NO: 64.
14. Kit ifølge et af kravene 8 til 13, omfattende primere og sonder, som har sekvenserne som angivet i SEQ ID NQ:64, 115, 116, 84, 163 og 164.
