Abstract
THE PRESENT INVENTION PROVIDES AN ACCURATE AND PROMPT DETECTION OF METHICILLIN RESISTANT STAPHYLOCOCCUS AUREUS (MRSA) WHICH IS IMPORTANT FOR INFECTION CONTROL BY USING OLIGONUCLEOTIDES AS PRIMERS AND GENE-SPECIFIC LABELED PROBES IN PCR FOR THE SIMULTANEOUS AMPLIFICATION AND DETECTION OF GENES CARRIED BY METHICILLIN RESISTANT STAPHYLOCOCCUS AUREUS (MRSA). IN ORDER TO DETECT MRSA BY THE PCR METHOD IN DISTINCTION FROM OTHER STAPHYLOCOCCI, TWO KINDS OF PRIMERS ARE PROVIDED FOR AMPLIFYING SA-442 GENE EXPRESSING SPECIFICITY TO S. AUREUS SPECIES, TWO KINDS OF PRIMERS ARE PROVIDED FOR AMPLIFYING MECA GENE EXPRESSING THE METHICILLIN RESISTANCE AND TWO KINDS OF PRIMERS ARE PROVIDED FOR AMPLIFYING COA GENE EXPRESSED BY THE COAGULASE ENZYME PRODUCING STAPHYLOCOCCI FOR DISCRIMINATING S. AUREUS FROM THE OTHER STAPHYLOCOCCI, ARE COMBINED WITH THREE GENE-SPECIFIC PROBES TO THE TARGETED AMPLIFIED NUCLEIC ACID SEQUENCES FOR SPECIFIC DETECTION OF THE PRESENCE AND/OR ABSENCE OF THE AMPLIFIED GENE SEQUENCES, WHEREIN THE PRESENCE AND DETECTION OF ALL THREE AMPLIFICATION PRODUCTS ARE INDICATIVE OF METHICILLIN RESISTANT STAPHYLOCOCCUS AUREUS (MRSA) AND THE ABSENCE OF SA-442 AND/OR COA AMPLIFIED PRODUCT(S) IS INDICATIVE OF OTHER STAPHYLOCOCCI.
