(54) Analogifremgangs måde til fremstil« ling af 1 -fenyl-2-amino-1,5-pro= pandiol-N-alkyl-derivater eller fysiologisk acceptable salte deraf.
Den foreliggende opfindelse angår en analogifremgangsmåde til fremstilling af hidtil ukendte l-fenyl-2-amino-l,3-propan-diol-N-alkyl-derivater og fysiologisk acceptable salte deraf. De hidtil ukendte forbindelser er virksomme mod angina.
De ved fremgangsmåden ifølge opfindelsen fremstillede forbindelser har den almene formel I B _ N \—u Oh nh oh $ Ah ? i\2

2 2 U4299 hvor R·*· er et hydrogena tom, en alkylgruppe med 1-8 kulstofatomer eller en fenoxymetylgruppe og 2 R er et hydrogenatom, en alkylgruppe med 1-8 kulstofatomer eller en 2,2-difenylætylgruppe med det forbehold, at hvis R1 er en fe- 2 2 noxymetylgruppe skal R være en metylgruppe, og hvis R er en 2,2-difenylætylgruppe skal R^ være et hydrogenatom.
Til de hidtil ukendte propandiol-N-alkyl-derivater med den almene formel I hører naturligvis også samtlige mulige stereoisomerer af disse forbindelser. Som bekendt foreligger 1- fenyl-2-amino-l,3-propandiol i form af treo- og erytroisomerer, som begge danner optiske isomerer. Dette gælder naturligvis også for N-alkylderivater deraf. Når alkylgruppen på nitrogenatomet indeholder et yderligere asymmetrisk kulstofatom, foreligger der en forbindelse med tre asymmetriske centre, og antallet af mulige diastereoisomerer er dobbelt så højt. I den foreliggende beskrivelse vil der i overensstemmelse med den normale nomenklatur for de fire racemater blive brugt betegnelserne treo, allotreo, erytro og allo- erytro.
Fremgangsmåden ifølge opfindelsen er ejendommelig ved det i krav l's kendetegnende del angivne.
Blandt de kendte alkyleringsmetoder anvendes sådanne, ved hvilke den indtrædende disubstitution er så ringe, at det ikke forstyrrer adskillelsen af hovedproduktet.
Fortrinsvis kondenseres l-fenyl-2-amino-l,3-propandiol med formel II reduktivt med et aldehyd eller en keton med den almene formel III

XR2 144299 3 1 2 hvor R og R har de ovenfor angivne betydninger og Hal er et halo-genatom, og fra det vundne N-benzyl-alkyl-derivat fjernes benzyl-gruppen ved katalytisk hydrogenering. Udgangsforbindelsen med formel II fremstilles på kendt måde (treo- og erytroracemat f.eks. ifølge J.A.C.S. 1949, 2463-2468; adskillelse af de optiske antipoder HU-PS nr. 154.951). N-benzylderivatet af forbindelsen med formel II kan vindes ved at man på palladium katalytisk reducerer den på kendt måde fremstillede benzal-forbindelse, treo-2-benzylidenamino-l-fenyl-l,3-propandiol (J.Org.Chem. 1954, 596).
De vundne forbindelser med den almene formel I kan om ønsket omsættes med uorganiske eller organiske syrer til deres fysiologisk acceptable salte.
Til den reduktive kondensation anvendes fortrinsvis en af de kendte hydrogenaktive katalysatorer (platin, palladium, raneynikkel eller jernholdigt raneynikkel). I laboratoriemålestok er anvendelsen af platinoxyd hensigtsmæssig, mens der til den tekniske målestok snarere kommer raneynikkel under tryk på tale. Reaktionen udføres i alkohol, vandig alkohol eller i et andet, med vand blandbart organisk opløsningsmiddel, hvis pH-værdi fortrinsvis indstilles svagt surt med en organisk syre.
Basen med den almene formel I er i almindelighed tungt opløselig. For at forbedre den biologiske anvendelighed (for at fremme resorptionen) er det fordelagtigt at fremstille forbindelsernes let opløselige salte (f.eks. hydroklorid, fumarat, glukonat).
I mange tilfælde som f.eks. ved 3,3-difenylpropyl-derivatet kan det forekomme, at opløsningen bliver uklar på grund af hurtig hydrolyse af glukonatet som følge af den ringe basicitet og basens dårlige vandopløselighed. Dette fænomen, som frem for alt ville virke forstyrrende ved injektionsopløsninger, kan dog overvindes ved at man anvender et overskud af glukonsyre og derved tilbagetrænger hydrolysen.

Forbindelserne med den almene formel I viser sig ved forskellige prøver at være biologisk aktive. Blandt disse biologiske virkninger skal navnlig virkningen mod angina fremhæves, idet denne virkning i mange tilfælde flerfoldigt overgår virkningen af klinisk velgennemprøvede midler, f.eks. virkningen af prenilamin (3,3-difenyl-propyl-l-methyl-fenætylamin). Desuden udviser de fleste forbindelser en lokalanæstetisk virkning og en krampeløsende virkning på det glatte muskulatur, hvilke virkninger i enkelte tilfælde er kraftigere end for kendte og velanvendte midler, som f.eks. virkningen af lidokain. Forbindelserne er allerede virksomme i overordentlig små doser, og deres toxicitet er meget lav (LD5Q-værdier på over 2000 mg/kg p.o. er ingen sjælden- 5 144299 hed), således at man praktisk talt kan se bort fra toxiciteten.
Toxiciteten ved oral indgift blev fastlagt på mus vejende 20-25 g af stammen CFLP af begge køn, idet der for hver dosis blev anvendt 10 forsøgsdyr. Iagttagelsesperioden varede 96 timer. De på denne måde bestemte LD^g-værdier er sammenstillet i tabel 2.

Ifølge in vitro gennemførte forsøg udviste de heller ikke nogen antikolinerg-, antihistamin- eller antiserotonin-virkning. Blodtryk og hjertefrekvens påvirkes ikke vedvarende af dem og åndingen beskadiges ikke.
Virkningen mod angina blev bestemt på rotter fra stammen CFY ifølge Nieschulz's metode (E.Nieschulz, K.Popendiker og I.Hoffmann: Arzneimittelforschung 1955, 5, 680) . Hanrotter med en vægt på 180-220 g blev narkotiseret. Elektrokardiogrammet blev registreret med et EKG-apparat af typen NEK 2 i standard-shuntning II. Den eksperimentelle coronarutilstrækkelighed blev fremkaldt med glanduitrin (4 i.e./kg i.v.). Som referenceforbindelse anvendtes prenilamin.
Den akutte intravenøse toxicitet blev målt på rotter 6 144299 af begge køn. For hver dosis blev der anvendt 5 forsøgsdyr.
Den relative effekt mod angina blev beregnet efter følgende ligning: LD50(mg/kg i.v.) -:—r-:- x hæmning (%) (prenilamin = 1)

Den lokalanæstetiske virkning blev ifølge Truant og d'Ama tos metode (A.P.Truant, S.Wiedling: Acta Chirurg.Scand. 116, 351(1958)) undersøgt på rottens nervus ischiadicus. Antallet af de dyr, som udviste den karakteristiske motoriske paralyse, blev fastslået. Som referenceforbindelse blev der anvendt lidocain. EC^-værdien og den i forhold til lidocain beregnede relative virkning er sammenstillet i tabel 4. Ved denne undersøgelse kan der kun anvendes vandopløselige forbindelser.
Den krampeløsende virkning på den glatte muskulatur blev undersøgt efter Brocks et al metode (N.Brock, J.Geks, D.Lorenz: Arch. Exper. Pathol, u. Pharmacol. 215, 492 (1952)) på rottens isolerede ileum. Som referenceforbindelse anvendtes papaverin. EC5Q-værdien og den i forhold til papaverin relative virkning er an- 7 144299 givet i tabel 4. Den relative virkning er beregnet ud fra ECgg- værdien for referenceforbindelsen divideret med EC„--værdien 50 for den undersøgte forbindelse.

Opfindelsen belyses nærmere i det følgende ved hjælp af nogle eksempler.
Eksempel 1 L(+)-treo-l-fenyl-2-isopropylamino-l,3-propandiol-hydroklorid
En opløsning af 8,36 g (0,05 mol) L-(+)-treo-l-fenyl- 2-amino-l,3-propandiol, 5,8 g (0,1 mol) acetone og 3,1 ml eddikesyre i absolut ætanol hydrogeneres i nærværelse af 0,5 g for-hydrogeneret platinoxyd til optagelse af den teoretiske mængde hydrogen (2400 ml hydrogen, varighed ca. 16 timer). Under hydrogeneringen sættes yderligere tre gange 0,2 g platinoxyd til reaktionsblandingen.
Katalysatoren frafiltreres og filtratet inddampes under vakuum. Der vindes 14,7 g af en viskos olie. Denne inddampes på ny under vakuum sammen med 20 ml ætanol og 10 ml 20% saltsur alkohol (pH 3-4). Det vundne rosafarvede hydroklorid opløses varmt i 40 ml af en blanding af ætanol og ætylacetat i forholdet 2:3, opløsningen filtreres , der tilsættes yderligere 20 ml ætylacetat efterfulgt af henstand i køleskab til krystallisering. Efter fra-filtrering af krystallerne vindes 5,5 g L-(+)-treo-l-fenyl-2-isopropylamino-1,3-propandiol-hydroklorid, som smelter ved 162- 8 144299 165°C} α^° = +59,11° (c = 2, H20) .
Efter inddampning af moderluden og behandling af remanensen med ætano1/ætylacetat i forholdet 2:3 kan der yderligere vindes 2,85 g af hydrokloridet. Det samlede udbytte udgør dermed 8,35 g (68%).
Eksempel 2 (Treo + allotreo)-l-fenyl-2-fenoxy-isopropylamino-l,3-propandiol- fumarat._ 17,3 g (0,103 mol) treo-l-fenyl-2-amino-l,3-propandiol og 31,0 g (0,206 mol) frisk destilleret fenoxyacetone opløses i 150 ml ætanol. Opløsningen syrnes med 6,2 ml eddikesyre og i nærværelse af 0,91 g i forvejen i 15 ml ætanol forhydrogeneret platinoxyd hydrogeneres til optagelse af den teoretiske hydrogenmængde. (Ved 20°C 4960 ml, varighed af hydrogenering: ca. 18-20 timer).
Katalysatoren frafiltreres og filtratet inddampes under vakuum. Fra remanensen fjernes opløsningsmiddelsporene og eddikesyren ved azeotrop destillation med benzen. Derpå opløses remanensen i 100 ml absolut alkohol og til opløsningen sættes en opløsning af 12 g fumarsyre i 300 ml alkohol. Blandingen henstår i køleskab natten over og derpå frafiltreres det udskilte produkt. Moderluden inddampes til en trediedel af dens omfang, og på denne måde vindes en yderligere fraktion. De forenede produktfraktioner (-30,2 g) omkrystalliseres fra 185 ml ætanol. Der vindes 24,75 g (57,5%) (treo + allotreo)-l-fenyl-2-fenoxy-isopropyl-amino-l,3-propandiol-fumarat som smelter ved 145-147°C.
Eksempel 3 (+)-(treo + allotreo)-1-fenyl-2-fenoxy-isopropylamino-1,3-propan-diol-fumarat_ ._
En opløsning af 13,0 g L-(+)-treo-l-fenyl-2-amino-l,3- propandiol og 23,2 g frisk destilleret fenoxyacetone i 124 ml ætanol syrnes med 4,66 g eddikesyre og hydrogeneres i nærværelse af 1 g platinoxyd, som i forvejen er blevet forhydrogeneret i 30 ml ætanol, indtil den teoretiske mængde hydrogen er optaget (3700 ml, hydrogeneringens varighed: 17 timer).
Katalysatoren frafiltreres, filtratet inddampes og mod 9 144299 destillationens slutning uddrives opløsningsmiddelresten ' med benzen. Der vindes 37,1 g af et olieagtigt produkt. Dette opløses i 100ml ætanol og der sættes 8,6 g fumarsyre i 200 ml ætanol til opløsningen. Efter inddampning under vakuum vindes en gullig olie som krystalliserer ved rivning med ætylacetat. Blandingen henstår natten over i køleskab hvorpå den filtreres.
Der vindes 17,9 g (55%) (+)-(treo- + allotreo)-l-fenyl-2-fenoxy- isopropylamino-propandiol-l,3-fumarat, der smelter ved 61-67°C; cip0 = +37,93° (c = 2, H20) .
Eksempel 4 (Erytro + alloerytro)-l-fenyl-2-fenoxy-isopropylamino-l,3-propan- diol-fumarat._
En opløsning af 17,3 g (0,103 mol) erytro-l-fenyl-2-amino-1,3-propandiol og 31,0 g (0,206 mol) fenoxyacetone i 350 ml absolut ætanol syrnes med 6,2 ml eddikesyre og hydrogeneres i nærværelse af l,0g platinoxyd, som er forhydrogene-ret i 30 ml ætanol, indtil optagelsen af den teoretiske mængde hydrogen (4960 ml). Derpå frafiltreres katalysatoren og filtratet inddampes under vakuum. Der bliver 51,0 g viskost produkt tilbage. Dette opløses i 100 ml ætanol og til opløsningen sættes en til 60°C opvarmet opløsning af 5,81 g fumarsyre i 120 ml ætanol. Blandingen henstår i køleskab natten over, hvorpå produktet frafiltreres og tørres. Der vindes 31,2 g af et produkt, som smelter ved 170-179°C. Dette omkrystalliseres fra 1100 ml ætanol, hvorved der vindes 18,7 g (50,5%) (erytro + alloerytro)-1-fenyl-2-fenoxy-isopropylamino-1,3-propandiol-fumarat, som smelter ved 189-190°C.
Eksempel 5
Treo-l-fenyl-2- [(31,31-difenylpropyl)-aminoJ-l,3-propandiol-hy- droklorid (racemat) ,_
En opløsning af 5,01 g (0,03 mol) racemisk treo-1-fenyl-2-amino-l,3-propandiol, 6,3 g (0,03 mol) 3,3-difenylpropion-aldehyd og 2,7 g (0,045 mol) eddikesyre i 100 ml ætanol hydrogeneres i nærværelse af 0,5 g platinoxyd, som i forvejen er blevet forhydrogeneret i 30 ml ætanol, ved stuetemperatur og atmosfærisk tryk til optagelse af den teoretiske mængde hydrogen (720 ml), hvilket varer ca. 16 timer. Derpå frafiltreres katalysatoren 144299 10 og filtratets pH-værdi indstilles på 3 med ca. 20% saltsur ætanol. Opløsningen inddampes under vakuum til tørhed. Den krystallinske remanens opløses varmt i 350 ml absolut ætanol, opløsningen filtreres og inddampes under vakuum til 60 ml. Efter henstand i køleskab natten over frafiltreres produktet. Ved inddampning af filtratet til 15 ml vindes en yderligere fraktion. De forenede produktfraktioner (ca. 8,2 g) opløses i 350 ml absolut ætanol, efter filtrering inddampes opløsningen til 60 ml og det udskilte produkt frafiltreres. Der vindes 5,55 g (46,5%) treo-l-fenyl-2-β3',3'-difenylpropyl)-aminoj-1,3-propandiol-hydroklorid i form af racematet. Smeltepunkt: 217-221°C.
Dannelse af den frie base;
De vundne 5,5 g af hydrokloridet opløses i 400 ml 80%, vandig metanol. Opløsningen omrøres i to timer med 66 g anionbytterharpiks Dovex ® 1 (basisk form) hvorefter der filtreres og ionbytterharpiksen vaskes først med 250 ml 70% metanol to gange, og derpå en gang med 250 ml ren metanol. Filtratet forenes med vaskevæskerne og inddampes under vakuum, hvorved der vindes 4,97 g rå base. Denne rives med 30 ml n-hexan, henstilles i fem timer i køleskab og frafiltreres derpå. Der vindes 4,28 g (85%) base, som smelter ved 79-83°C.
Eksempel 6 D(-)-treo-l-fenyl-2-f~(3',3'-difenylpropyl)-amino] -1,3-propandiol-hydroklorid._
En blanding af 10,03 g (0,06 mol) D-(-)-treo-l-fenyl- 2-amino-l,3-propandiol og 12,6 g (0,06 mol) 3,3-difenylpropional-dehyd holdes i 30 minutter ved 100°C. Det vundne produkt opløses i 200 ml absolut ætanol og hydrogeneres derpå i nærværelse af 0,5 g platinoxyd, der i forvejen er blevet forhydrogeneret i 30 ml ætanol, efter tilsætning af 5,4 g eddikesyre indtil optagelse af den teoretiske mængde hydrogen (1440 ml).Katalysatoren frafiltreres og filtratets pH-værdi indstilles på 3 med 20% saltsur ætanol. Derefter inddampes opløsningen under vakuum. Det tilbageblivende hydroklorid opløses varmt i 1400 ml absolut ætanol, opløsningen filtreres og inddampes til 300 ml. Opløsningen henstår natten over i køleskab, hvorpå den filtreres. Det primære udbytte af hydrokloridet udgør 13,1 g, smeltepunkt: 233-234°C, sønderdeling: 144299 11 238°C. Ved inddampning af moderluden vindes yderligere 2,7 g produkt. De forenede 15,8 g af hydrokloridet opløses i 1500 ml absolut metanol, opløsningen filtreres, filtratet inddampes til 250 ml og den ved køling af opløsningen udfældede substans fra-filtreres. Der vindes 9,8 g (41%) D-(-)-treo-l-fenyl-2-jT(3',3'-difenylpropyl)-amino] -1,3-propandiol-hydroklorid. Produktet smelter ved 235-237,5°C, og dets sønderdelingspunkt ligger ved 240°C.
Ved inddampning af moderluden til 60 ml vindes yderligere 3,34 g hydroklorid. Smeltepunkt: 237,5-239°C : -36,68° (ætanol, c = 1,48). Det samlede udbytte udgør dermed 55%.
Dannelse af den frie base: 13,14 g af det vundne hydroklorid behandles i 800 ml (S) 80% vandig metanol med 90 g anionbytterharpiks Dovex ^ 1. Efter filtreringen vaskes harpiksen to gange med 500 ml 70% metanol og en gang med 500 ml absolut metanol. Filtratet forenes med vaskevæskerne og inddampes under vakuum, idet der mod destillationens slutning tilsættes og afdampes benzen. Remanensen krystalliseres ved tilsætning af n-hexan i kulden. Der vindes 9,1 g (76,25%) D-(-)-treo-l-fenyl-2- [(3',3'-difenylpropyl)-aminoj-1,3-propandiol, som smelter ved 79-82,5°C.
Eksempel 7
Erytro-l-fenyl-2-[(3',31-difenylpropyl)-amino]-1,3-propandioi
En blanding af 10,03 g (0,06 mol) erytro-l-fenyl-2-amino-l,3-propandiol, 12,6 g (0,06 mol) 3,3-difenylpropionalde-hyd og 5,4 g eddikesyre i 250 ml absolut ætanol hydrogeneres i nærværelse af 0,3 g platinoxyd, som i forvejen er blevet hydrogeneret i 30 ml ætanol, indtil optagelse af den teoretiske mængde hydrogen (1440 ml). Katalysatoren frafiltreres, filtratet inddampes og remanensen behandles i 200 ml metanol med 200 g anion-bytterharpiks Dovex ^ 1. Efter frafiltrering af harpiksen inddampes opløsningen. Det tilbageblivende 21,5 g viskose produkt opløses i 30 ml ætanol og der sættes n-hexan til opløsningen indtil den bliver uklar (ca. 400 ml). Blandingen stilles i køleskab og filtreres senere. Det vundne 14,3 g krystallinske råprodukt omkrystalliseres fra 260 ml af en blanding af ætanol og destilleret vand i forholdet 1:1. Der vindes 10,85 g (50,02%) erytro-l-fenyl-2-[(3',3'-difenylpropyl)-amino]-1,3-propandiol 144299 12 som smelter ved 108-109°C-Eksempel 8
Treo-1-feny1-2-methylamino-1,3-propandiol-hydroklorid
En blanding af 10,4 g (40,4 mmol) l-fenyl-2-benzylamino- 1,3-propandiol, 7,16 g (0,05 mol) metyljodid og 13,6 g pulveriseret natriumkarbonat i 250 ml absolut acetone koges under tilbagesvaling i 70 timer. Derpå filtreres reaktionsblandingen og filtratet inddampes. Til fjernelse af natriumkloridet koges remanensen med 100 ml destilleret vand i 5 minutter og derpå ekstraheres den organiske base med 5 x 50 ml æter. De forenede ekstrakter tørres over natriumsulfat og æteren afdestilleres hvorved der vindes 6,45 g af et olieagtigt produkt som krystalliserer ved behandling med petroleumsæter og 48 timers henstand. De ved filtrering vundne 5,35 g produkt optages i vand og ekstraheres fire gange med 80 ml æter. Den æteriske opløsning klares med aktivt kulstof og filtreres derpå og æteren afdestilleres. Remanensen krystalliseres ved rivning med n-hexan til hvide krystaller. Der vindes 3,4 g (31 %) treo-l-fenyi-2-benzylmetylamino-1.,3-propandiol som smelter ved 69-72°C.
Dette produkt hydrogeneres i 135 ml ætanol i nærværelse af 0,5 g palladiumkulstof indtil optagelse af den teoretiske mængde hydrogen. Katalysatoren frafiltreres og filtratet inddampes. pH-Værdien af det tilbageblivende viskose produkt indstilles på 3 med 20 % saltsur ætanol hvorpå produktet inddampes til tørhed. Hydrokloridet opløses varmt i 7 ml ætanol og der sættes ætylacetat til opløsningen indtil denne bliver uklar (ca. 30 ml). Opløsningen henstår og senere frafiltreres det krystallinske hydroklorid. Der vindes 2,72 g (80 %) treo-l-fenyl-2-metylamino-1,3-propandiol-hydroklorid som smelter ved 125-127°c.

Claims (2)
1. Analogifremgangsmåde til fremstilling af l-fenyl-2-amino-l,3-propandiol-N-alkyl-derivater med den almene formel C CH - CH - CH0 \ // I I I 2 OH NH OH CH X /\2 RX R hvor R1 er et hydrogenatom, en alkylgruppe med 1-8 kulstofatomer eller en fenoxymetylgruppe og 2 R er et hydrogenatom, en alkylgruppe med 1-8 kulstofatomer eller en 2,2-difenylætylgruppe, med det forbehold at hvis R"^ er en fenoxy- 2 2 metylgruppe skal R være enmetylgruppe og hvis R er en 2,2-difenylætylgruppe skal R^ være et hydrogenatom,eller fysiologisk acceptable salte deraf, kendetegnet ved at den racemiske eller optisk aktive treo- eller erytro-l-fenyl-2-araino-l,3-propan-diol med formlen Or-r-r. OH NH2 OH eller N-benzylderivatet deraf underkastes skånsom alkylering, hvorpå man i tilfælde af N-benzylderivatet fjerner benzyigruppen, hvorpå den vundne forbindelse om ønsket omsættes med en uorganisk eller organisk syre til dannelse af et fysiologisk acceptabelt salt.
2. Fremgangsmåde ifølge krav 1, kendetegnet ved at alkyleringen af 1-fenyl-2-amino-l,3-propandiol med formel II foretages som reduktiv kondensation med et aldehyd eller en keton med den almene formel ^ Ri
