Description
- Die Erfindung betrifft ein neues Verfahren für die Synthese von 1-(Aminomethyl)cyclohexyl-essigsäure der Formel (I) über das neue intermediäre 1-(Nitromethyl)cyclohexyl-essigsäure-Derivat der allgemeinen Formel (II), wobei R Wasserstoff, eine Benzyl-Gruppe, eine Diphenylmethyl-Gruppe oder am aromatischen Ring mit C1-C4-Alkyl oder Alkoxy substituierte Derivate davon darstellt.
- Die 1-(Aminomethyl)cyclohexyl-essigsäure der Formel (I), sonst als Gabapentin bekannt, ist der Wirkstoff des GABA-Antagonist-Medikaments. Verschiedene Verfahren zur Synthese dieser Verbindung sind aus der Literatur bekannt.
- In den meisten der bekannten Verfahren wird ein Zwischenprodukt mit Säure hydrolysiert, und Gabapentin wird aus dem so gebildeten Gabapentinhydrochloridsalz durch Verwenden eines Ionenaustauschharzes erhalten. Dieses Verfahren ist in dem deutschen Patent Nr.
DE 2 460 891 beschrieben, in dem das 1,1-Cyclohexyldiessigsäureanhydrid in Hydroxamsäure umgewandelt und das letztere über Lossen-Abbau in das Hydrochloridsalz des Produkts transformiert wird. Das US-Patent Nr.US 4 024 175 beschreibt ein Verfahren, in dem das gleiche 1,1-Cyclohexyldiessigsäureanhydrid als Ausgangsmaterial verwendet wird. Das Anhydrid wird zuerst in ein Monomethylestermonosalz transformiert, und dann wird ein Monosäuremonoazid aus ihm erhalten. Das Gabapentinhydrochlorid wird aus letzterem über Curtius-Abbau hergestellt. - Ähnlich wird Gabapentinhydrochlorid durch die in dem europäischen Patent Nr.
EP 414 274 - Die Nachteile der oben erwähnten Vorgehensweise sind wie folgt. Gabapentin wird als sein Hydrochloridsalz erhalten, und Gabapentin selbst kann nur unter Verwenden von arbeitsaufwendigen und teuren Ionenaustauschverfahren isoliert werden. Das Vermeiden der ungewünschten Lactambildungs-Nebenreaktion verlangt auch eine arbeitsaufwendige und teure Technik. Weitere Nachteile dieser Vorgehensweise sind die Verwendung von gefährlichen Reagenzien, z. B. Kaliumcyanid, Natriumazid, und das teure druckbeständige Gerät.
- Die im europäischen Patent Nr.
EP 414 275 EP 414 274 - Das Ziel der Erfindung ist es, ein ökonomisches, industriell anwendbares Verfahren für die Synthese von Gabapentin zu entwickeln, das die Nachteile der oben erwähnten Vorgehensweisen vermeidet und die einfache Synthese von sehr reinem Endprodukt der Formel (I) in weniger Schritten und in guter Ausbeute möglich macht.
- Die erfindungsgemäße Synthese von Gabapentin ist wie folgt:
- a) der Alkylester von Cyclohexyliden-essigsäure der allgemeinen Formel (VI) – worin R2 eine C1-C4-Alkyl-Gruppe darstellt – wird mit Nitromethan in Gegenwart einer Base in den Alkylester von 1-(Nitromethyl)-cyclohexyl-essigsäure der allgemeinen Formel (V) – worin die Bedeutung von R2 wie oben definiert ist – umgewandelt, letzterer wird mit wäßriger methanolischer Lösung von Kaliumhydroxid hydrolysiert, und die erhaltene 1-(Nitromethyl)cyclohexyl-essigsäure der Formel (IIa) wird in einem Lösungsmittel in Gegenwart eines Katalysators hydriert, um das gewünschte Produkt der Formel (I) zu ergeben, und gegebenenfalls Umwandlung der erhaltenen Verbindung in ein pharmazeutisch annehmbares Salz oder b) der Alkylester von Cyclohexyliden-essigsäure der allgemeinen Formel (VI) – wobei die Bedeutung von R2 wie oben definiert ist – wird mit wäßriger methanolischer Lösung von Kaliumhydroxid hydrolysiert, und die erhaltene Cyclohexyliden-essigsäure der Formel (IV) wird mit einem Reagens der Formel R1-X – worin R1 eine Benzyl-Gruppe, Diphenylmethyl-Gruppe oder gegebenenfalls am aromatischen Ring mit C1-C4-Alkyl oder Alkoxy substituierte Derivate davon darstellt und X Halogenatome darstellt – umgesetzt, um das entsprechende Cyclohexylidensäure-Derivat der allgemeinen Formel (III) – worin die Bedeutung von R1 wie oben definiert ist – zu ergeben, und dieses Zwischenprodukt wird mit Nitromethan in das 1-(Nitromethyl)cyclohexylessigsäure-Derivat der allgemeinen Formel (IIb) – wobei die Bedeutung von R1 wie oben definiert ist – umgewandelt, und letzteres wird in einem Lösungsmittel in Gegenwart eines Katalysators hydriert, und gegebenenfalls Umwandlung der erhaltenen Verbindung in ein pharmazeutisch annehmbares Salz.
- Das Verfahren der Erfindung wird in Schema 1 dargestellt.
- Die Erfindung basiert auf der Beobachtung, daß die Reduktion der neuen Verbindungen der allgemeinen Formel (II) bei atmosphärischem Druck direkt das reine gewünschte Endprodukt ergibt.
- Überraschenderweise wurde gefunden, daß beim Verwenden der Verbindungen der allgemeinen Formel (II) als Ausgangsmaterialien in dem Reduktionsschritt die Lactam-Verbindung nicht gebildet wird, sondern das sehr reine Gabapentin direkt erhalten wird. Dies war durch die Kenntnis der früheren Vorgehensweisen nicht vorhersehbar, daß die Fähigkeit dieser Art von Verbindungen zur Lactam-Bildung aus der Literatur bekannt ist (z. B.
EP 414 274 - Der als Ausgangsmaterial verwendete Alkylester von Cyclohexylidenessigsäure der allgemeinen Formel (VI) kann gemäß der Literatur über die Reaktion von Cyclohexanon und dem geeigneten Ester der Diethylphosphonessigsäure hergestellt werden.
- In dem letzten Hydrierungsschritt können alle Katalysatoren verwendet werden, die im allgemeinen in Hydrierungsreaktionen anwendbar sind, Seltenerdmetall-Katalysatoren z. B. Rhodium oder Palladium, Raney-Nickel oder Cobalt-Katalysatoren, gegebenenfalls auf einem Träger z. B. auf Kohlenstoff, bevorzugt Palladium-auf-Aktivkohle, bevorzugter 10% der zu reduzierenden Verbindung.
- Die Hydrierung wird in einem inerten organischen Lösungsmittel, bevorzugt in einem C1-C4-Alkohol, bevorzugter in Methanol, bei 10 bis 50°C, unter 1 bis 20 kPa Druck, bevorzugt bei Raumtemperatur und unter atmosphärischen Druck, durchgeführt.
- Die Michael-Addition der Ester der Cyclohexyliden-essigsäure mit Nitromethan wird in Gegenwart einer Base, bevorzugt Kaliumhydroxid, durchgeführt.
- Die Hydrolyse der Alkylester-Gruppe wird mit Base, bevorzugt wäßriger methanolischer Lösung von Kaliumhydroxid, bei Raumtemperatur durchgeführt, dann wird die Säure mit 10%iger wäßrige Kaliumdihydrogenphosphat-Lösung freigesetzt.
- Nach Filtration des Katalysators wird das Produkt durch Konzentration des Filtrats isoliert. Das bei der Konzentration erhaltene Produkt ist 98 bis 99% rein, die Ausbeute ist 50 bis 70%.
- Die Vorteile dieser Vorgehensweise sind wie folgt:
- – das erhaltene Produkt ist sehr rein,
- – die Anzahl an Reaktionsschritten ist geringer als bei den bekannten Vorgehensweisen,
- – die Lactam-Verbindung, die sehr schwierig zu entfernen ist, wird nicht gebildet,
- – es wird weder spezielles druckresistentes Gerät noch ein teuerer Katalysator benötigt,
- – das Endprodukt kann ohne Anwenden der schwierigen und komplizierten Ionenaustauschtechnik erhalten werden,
- – es werden keine giftigen oder gefährlichen Materialien be nötigt.
- Beispiele
- Beispiel 1
- a) Synthese von 1-(Nitromethyl)cyclohexyl-essigsäure
- Eine Lösung von 4,3 g (0,02 mol) Methyl-1-(nitromethyl)cyclohexylacetat in einer Mischung von 50 ml Methanol und 20 ml l0%igem wäßrigem Kaliumhydroxid wird bei Raumtemperatur 24 h gerührt, dann wird das Methanol im Vakuum abdestilliert. Der pH der resultierenden wäßrigen Lösung wird mit l0%igem wäßriger Kaliumdihydrogenphosphat-Lösung auf 7 eingestellt. Die Lösung wird dreimal mit 30 ml Ethylacetat extrahiert, die vereinigten organischen Schichten werden über Natriumsulfat getrocknet und konzentriert, um 3,2 g (80%) der Titelverbindung als Öl zu ergeben.
- b) Synthese von 1-(Aminomethyl)cyclohexyl-essigsäure
- Eine Lösung von 2,01 g (0,01 mol) 1-(Nitromethyl)cyclohexyl-essigsäure in 50 ml Methanol wird in Gegenwart von 0,2 g Palladium-auf-Aktivkohle bei atmosphärischem Druck hydriert. Der Katalysator wird abgefiltert, und das Filtrat wird auf 10 ml konzentriert. 20 ml Tetrahydrofuran werden zu dem Rückstand zugegeben, und die ausgefällten Kristalle wurden abfiltriert und getrocknet, um 1,3 g (80%) der Titelverbindung zu ergeben.
Smp.: 164–169°C. - Beispiel 2
- Synthese von 1-(Aminomethyl)cyclohexyl-essigsäure
- Eine Lösung von 5 g (0,017 mol) Benzyl-1-(nitromethyl)cyclohexylacetat in 50 ml Methanol wird zu einer Mischung von 0,5 g verhydriertem Palladium, 10 g auf Aktivkohle in 50 ml Methanol zugegeben. Diese Mischung wird bei Raumtemperatur unter atmosphärischem Druck hydriert, bis der berechnete Wasserstoff verbraucht ist, dann wird der Katalysator abgefiltert, das Filtrat wird auf etwa 15 ml konzentriert, und 30 ml Tetrahydrofuran werden zugegeben, um die Titelverbindung auszufällen. Ausbeute: 1,5 g (51%). Smp.: 168°C.
Claims (9)
- Verfahren zur Synthese von 1-(Aminomethyl)cyclohexyl-essigsäure und pharmazeutisch annehmbaren Salzen davon, gekennzeichnet durch a) Umwandlung des Alkylesters von Cyclohexyliden-essigsäure der Formel (VI) – wobei R2 eine C1-C4-Alkyl-Gruppe darstellt – mit Nitromethan in Gegenwart einer Base in den Alkylester von 1-(Nitromethyl)cyclohexylessigsäure der Formel (V) – wobei die Bedeutung von R2 wie oben definiert ist – Hydrolyse mit wäßriger methanolischer Lösung von Kaliumhydroxid und Hydrierung der erhaltenen 1-(Nitromethyl)cyclohexyl-essigsäure der Formel (IIa) in einem Lösungsmittel in Gegenwart eines Katalysators und gegebenenfalls Umwandlung der erhaltenen Verbindung in ein pharmazeutisch annehmbares Salz oder b) Hydrolyse des Alkylesters der Cyclohexyliden-essigsäure der Formel (VI) – worin R2 eine C1-C4-Alkyl-Gruppe darstellt – mit wäßriger methanolischer Lösung von Kaliumhydroxid in die Cyclohexyliden-essigsäure der Formel (IV), Umsetzung der erhaltenen Säure der Formel (IV) mit einem Reagens der Formel R1-X – worin R1 eine Benzyl-Gruppe, Diphenylmethyl-Gruppe oder gegebenenfalls am aromatischen Ring mit C1-C4-Alkyl oder Alkoxy substituierte Derivate davon darstellt und X ein Halogenatom darstellt – um das intermediäre Cyclohexylidensäure-Derivat der (III) zu ergeben – worin die Bedeutung von R1 wie oben definiert ist – Umwandlung dieses Zwischenprodukts mit Nitromethan in das 1-(Nitromethyl)cyclohexylessigsäure-Derivat der Formel (IIb) – wobei die Bedeutung von R1 wie oben definiert ist – und Hydrierung des letzteren in einem Lösungsmittel in Gegenwart eines Katalysators, und gegebenenfalls Umwandlung der erhaltenen Verbindung in ein pharmazeutisch annehmbares Salz.
- Verfahren b) gemäß Anspruch 1, dadurch gekennzeichnet, daß Benzylhalogenid als Reagens der Formel R1-X verwendet wird.
- Verfahren b) gemäß Anspruch 1, dadurch gekennzeichnet, daß Diphenylmethylhalogenid als Reagens der Formel R1-X verwendet wird.
- Verfahren gemäß Anspruch 1 bis 3, dadurch gekennzeichnet, daß die Hydrierung in einem inerten organischen Lösungsmittel durchgeführt wird.
- Verfahren gemäß Anspruch 1 bis 3, dadurch gekennzeichnet, daß Palladium-auf-Aktivkole als Katalysator verwendet wird.
- Neue Verbindungen der Formel (II), worin R Wasserstoff, eine Benzyl-, eine Diphenylmethyl-Gruppe oder gegebenenfalls am aromatischen Ring mit C1-C4-Alkyl oder Alkoxy substituierte Derivate davon darstellt.
- 1-(Nitromethyl)cyclohexyl-essigsäure.
- Benzyl-1-(nitromethyl)cyclohexyl-acetat.
- Diphenylmethyl-1-(nitromethyl)cyclohexyl-acetat.
