Description
- Diese Erfindung bezieht sich auf Druckguss-Membranen, die insbesondere zur peroralen Verabreichung eines therapeutischen Mittels in weiterentwickelten Verabreichungsformen, die als osmotische Verabreichungsformen bezeichnet werden können, hergestellt werden. Die Erfindung betrifft außerdem Zusammensetzungen, die wenigstens ein Polymer enthalten, das für das Druckgießen eines Fertigungsprodukts verwendet wird.
- Einzigartige Verabreichungsformen, die als osmotische Verabreichungsformen hergestellt werden, tauchten im Bereich der Medizin und der Pharmazie mit der Erfindung von osmotischen Verabreichungssystemen durch die Erfinder Theeuwes und Higuchi, die in den US-Patenten Nrn. 3.845.770 und 3.916.899 offenbart sind, auf. Die in diesen Patenten offenbarten osmotischen Systeme umfassen eine halbdurchlässige Membran, die eine Zelle umgibt, die das therapeutische Mittel enthält. Die Membran ist für den Durchtritt eines äußeren Fluids durchlässig und für den Durchtritt des Medikaments undurchlässig. Es ist wenigstens ein Ausgang durch die Membran vorhanden, um das therapeutische Mittel aus der osmotischen Verabreichungsform abzugeben.
- Eine Pionierarbeit in der Verabreichungstechnik im Hinblick auf osmotische Systeme wurde im US-Patent Nr. 4.327.727 von den Patentinhabern Cortese and Theeuwes geleistet. Diese Erfindung stellt ein osmotisches Verabreichungssystem zur Verabreichung eines therapeutischen Mittels, das wegen seiner Löslichkeit in wässerigen und biologischen Fluiden nur schwer in bedeutenden Mengen und bei zeitlich gesteuerter Dosis zu verabreichen ist, bereit. Das osmotische Verabreichungssystem dieses Patents umfasst eine halbdurchlässige Wand, die eine Zelle umgibt, die ein therapeutisches Mittel, das in wässerigen und biologischen Fluiden unlöslich bis stark löslich ist, und ein schäumbares Hydrogel enthält. Bei Gebrauch schäumt das Hydrogel in Gegenwart eines äußeren Fluids, das vom Verabreichungssystem aufgenommen wird, auf und schiebt das therapeutische Mittel durch einen Austrittskorridor aus dem Verabreichungssystem.
- Ein weiterer Beitrag zur Verabreichungstechnik in einem neu erfundenen Ver abreichungssystem wurde im US-Patent Nr. 5.023.088, deren Patentinhaber Wong, Theeuwes, Eckenhoff, Larson und Huynh sind, offenbart. In diesem Patent offenbarten die Patentinhaber ein Verabreichungssystem, das ein halbdurchlässiges Gerüstelement mit einer Zelle, die mehrere verschiebbare therapeutische Einheiten enthält, umfasst. Die Patentinhaber offenbarten ferner ein Treibmittel, das die therapeutischen Einheiten durch einen in den Gerüstelementen ausgebildeten offenen Ausgang schiebt, wenn das Verabreichungssystem in einer entsprechenden Umgebung in Gebrauch ist.
- Fachleuten auf dem Gebiet der Verabreichungstechnik ist klar, dass die oben dargelegten Verabreichungsformen einen positiven erfinderischen Wert haben und einen praktischen und sinnvollen Fortschritt in der Verabreichungstechnik darstellen. Obwohl die oben beschriebenen Verabreichungsformen in vielen Gebrauchsumgebungen eine ideale Kinetik zur Verabreichung zahlreicher und unterschiedlicher Medikamente bei gesteuerter und kontinuierlicher Dosis besitzen, gibt es auch einen Fall, in dem der Hersteller solcher Verabreichungsformen beim Erzielen noch besserer Ergebnisse vorangebracht werden kann. Beispielsweise werden die Verabreichungsformen des Standes der Technik nach einem Membranauftragverfahren hergestellt, bei dem ein organisches Lösungsmittel wie etwa Aceton oder Methylenchlorid verwendet wird, um die Membran auf die Verabreichungsform aufzutragen. Obwohl diese Lösungsmittel ausgezeichnete Membranen bilden, gibt es schwerwiegende Mängel, die mit ihrem Gebrauch verbunden sind. Diese organischen Lösungsmittel sind nämlich teuer und explosibel, wobei Spuren davon möglicherweise giftig sein können und wobei während der Herstellung Lösungsmitteldämpfe entweichen können, die für die Umwelt bedenklich sind. Die Bedingungen, die zur Fertigungsoptimierung bei der Herstellung einer dünnen oder einer dicken Membran erforderlich sind, sind bei der Massenproduktion nicht einhaltbar, da die Veränderlichkeit des Membrangewichts und der Dickengleichmäßigkeit zu unterschiedlichen Freigabeprofilen der Verabreichungsformen führen kann. Ferner ist die Optimierung der Auftragsbedingungen für jede folgende Schichtstaffelung kostspielig, wobei die Herstellung einer dicken Membran bei der Massenproduktion infolge der extrem langen Auftragszeit häufig undurchführbar wird. Zudem können manche spezielle Membranformen wegen ihrer komplizierten Geometrie nicht durch einen Auftragsprozess gefertigt werden.
- Fachleuten auf dem Gebiet der Verabreichungstechnik ist klar, dass dann, wenn eine Verabreichungsform geschaffen werden kann, die ein hohes Niveau an zuverlässiger Abgabeaktivität aufweist und nach einem Verfahren hergestellt wird, das die Mängel und Nachteile, die mit dem Stand der Technik verbunden sind, beseitigt, eine solche Verabreichungsform und das damit einhergehende Herstellungsverfahren einen positiven Wert hätten und außerdem einen Fortschritt in der Verabreichungstechnik darstellen würden. Fachleuten auf dem Gebiet der Verabreichungstechnik ist auch sofort klar, dass eine Verabreichungsform, die als osmotische Verabreichungsform gestaltet ist und als solche im Wesentlichen frei von organischen Lösungsmitteln ist, im Bereich der Pharmazie und der Medizin praktische Anwendung finden wird. Fachleuten auf dem Gebiet der Verabreichungstechnik ist gleichermaßen klar, dass eine zum Druckgießen vorgesehene neuartige Zusammensetzung, die die bisher bekannten Probleme beseitigt, einen positiven Fortschritt in der Technik darstellen wird.
- Eine weitere Aufgabe der Erfindung ist es, Membranen zu schaffen, die nach einem Druckgussverfahren hergestellt werden, wobei die Membran eine Durchlässigkeit für Wasser besitzt, jedoch für ein therapeutisches Mittel im Wesentlichen undurchlässig ist, und mechanische Eigenschaften besitzt, die bei der Herstellung einer osmotischen Verabreichungsform nützlich sind.
- Eine weitere Aufgabe der Erfindung ist es, eine Zusammensetzung vorzusehen, die für Druckgussartikel im Bereich der Medizin und des Gesundheitswesens nützlich ist.
- Eine weitere Aufgabe der Erfindung ist es, eine Zusammensetzung mit thermoplastischen Eigenschaften vorzusehen.
- Eine weitere Aufgabe der Erfindung ist es, eine Zusammensetzung vorzusehen, die wenigstens ein Polymer enthält, wobei die Zusammensetzung zu jeder gewünschten Form, vorzugsweise als Gerüstelement für eine Verabreichungsform, druckgegossen werden kann.
- Eine weitere Aufgabe der Erfindung ist es, eine Zusammensetzung vorzusehen, die zwei oder mehr Polymere enthält, die für das Thermoplastgießen nützlich sind.
- Weitere Aufgaben, Merkmale und Aspekte dieser Endung werden Fachleuten auf dem Gebiet der Verabreichungstechnik aus der folgenden genauen Beschreibung deutlicher, wenn diese in Verbindung mit den begleitenden Ansprüchen genommen wird.
- Gemäß der Art und Weise dieser Erfindung sind neuartige Zusammensetzungen vorgesehen für das Druckgießen von Membranen, wie sie in Anspruch 1 beansprucht sind, die zu Verabreichungsformen, wie sie in Anspruch 6 beansprucht sind, geformt werden können. Die Zusammensetzungen enthalten ein thermoplastisches Polymer oder ein Gemisch aus thermoplastischen Polymeren und optional Druckgusszutaten. Das thermoplastische Polymer, das für den gegebenen Zweck verwendet werden kann, umfasst Polymere, die einen niedrigen Erweichungspunkt, beispielsweise unterhalb von 200°C, vorzugsweise im Bereich von 40°C bis 180°C besitzen. Die Polymere sind vorzugsweise Kunstharze, beispielsweise Linearpolykondensationsharze, kondensationspolymerisierte Harze, additionspolarisierte Harze wie etwa Polyamide, Harze, die aus Diepoxiden und primären Alkanolaminen erhalten werden, Harze des Glyzerins und der Phthalanhydride, Polymethan-, Polyvinylharze, Polymerharze mit freien Endstellen oder mit veresterten Carboxyl- oder Carboxyamidgruppen, beispielsweise Acrylsäure, Acrylamid oder Acrylsäureestern, Polycaprolacton und seine Copolymere mit Dilactid, Diglycolid, Valerolacton und Decalacton, eine Harzzusammensetzung, die Polycaprolacton und Polyalkenoxid umfasst, und eine Harzzusammensetzung, die Polycaprolacton, ein Polyalkenoxid wie etwa Polyethylenoxid, Poly(Zellulose) wie etwa Poly(Hydroxypropyl-methylzellulose), Poly(Hydroxyethyl-methylzellulose) und Poly(Hydroxypropylzellulose) umfasst. Die membranbildende Zusammensetzung kann optional membranbildende Zutaten wie etwa Polyethylenglykol, Talcum, Polyvinylalkohol, Lactose oder Polyvinylpyrrolidon umfassen. Die Zusammensetzungen zum Bilden einer Druckguss-Polymerzusammensetzung können 100% thermoplastisches Polymer enthalten. Die Zusammensetzung enthält in einer weiteren Ausführungsform 10% bis 99% eines thermoplastischen Polymers und 1% bis 70% eines anderen Polymers, die insgesamt 100% ergeben. Die Erfindung sieht außerdem eine Thermoplast-Polymerzusammensetzung vor, die 1% bis 98% eines ersten thermoplastischen Polymers, 1% bis 90% eines anderen, zweiten Polymers und 1% bis 90% eines weiteren, dritten Polymers enthält, wobei die Polymere zusammen 100% ergeben. Die repräsentative Zusammensetzung enthält 20% bis 90% thermoplastisches Polycaprolacton und 10% bis 80% Poly(Alkenoxid); während eine Zusammensetzung 20% bis 90% Polycaprolacton und 10% bis 60% Poly(Ethylenoxid), die mit den Zutaten 100% ergeben, enthält; eine Zusammensetzung 10% bis 97% Polycaprolacton, 10% bis 97% Poly(Alkenoxid) und 1% bis 97% Poly(Ethylenglykol), die mit allen Zutaten 100% ergeben, enthält; eine Zusammensetzung 20% bis 90% Polycaprolacton und 10% bis 80% Poly(Hydroxypropylzellulose), die mit allen Zutaten 100% ergeben, enthält; und eine Zusammensetzung 1% bis 90% Polycaprolacton, 1% bis 90% Poly(Ethylenoxid), 1% bis 90% Poly(Hydroxypropylzellulose) und 1% bis 90% Poly(Ethylenglykol), die mit allen Zutaten 100% ergeben, enthält. Die Prozent,%, werden in Gewichtsprozent, Gew.-%, ausgedrückt.
- In einer weiteren Ausführungsform der Erfindung wird eine Zusammensetzung für das Druckgießen zur Herstellung einer Membran vorbereitet, indem eine Zusammensetzung, die 63 Gew.-% Polycaprolacton, 27 Gew.-% Polyethylenoxid und 10 Gew.-% Polyethylenglykol enthält, in einer herkömmlichen Mischmaschine wie etwa einem Moriyama®-Mischer bei 65°C bis 95°C vermischt werden, wobei die Zutaten in der folgenden Zugabereihenfolge: Polycaprolacton, Polyethylenoxid und Polyethylenglykol in den Mischer gegeben werden. Alle Zutaten werden für 135 Minuten bei einer Rotordrehzahl von 10 bis 20 min–1 vermischt. Als Nächstes wird das Gemisch einem Baker Perkins Kneader®-Extruder bei 80°C bis 90°C, einer Pumpendrehzahl von 10 min–1 und einer Schraubendrehzahl von 22 min–1 zugeführt und danach auf 10°C bis 12°C abgekühlt, bis eine gleichmäßige Temperatur erreicht wird. Danach wird die gekühlte gespritzte Zusammensetzung einer Albe-Körnungsmaschine zugeführt und bei 25 ° in Körner mit einer Länge von 5 mm umgewandelt. Die Körner werden als Nächstes in eine Druckgussmaschine, eine Arburg Allrounder®, mit 200°F bis 350°F (93°C bis 177°C) geleitet und erwärmt, bis sie eine schmelzflüssige polymere Zusammensetzung ergeben, die dann mit unter hohem Druck und mit hoher Geschwindigkeit in eine Gussform gedrückt wird, bis die Form gefüllt ist und sich die die Polymere enthaltende Zusammensetzung zu einer vorgegebenen Gestalt verfestigt hat. Die Parameter für das Druckgießen umfassen eine Bandtemperatur durch die Zonen 1 bis 5 der Trommel mit 195°F (91°C) bis 375°F (191°C), einen Druckgussdruck von 1818 bar, eine Geschwindigkeit von 55 cm3/s und eine Formtemperatur von 75°C.
- Die Begriffe "therapeutisches Mittel" und "Arzneimittel" werden hier austausch bar verwendet, wobei sie sich auf eine Arzneimittelmischung, eine Stoffzusammensetzung oder ein Gemisch davon beziehen, die eine therapeutische und häufig nützliche Wirkung erzielen. Diese umfassen Pestizide, Herbizide, Germizide, Biozide, Algizide, Rodentizide, Fungizide, Insektizide, Oxidationsinhibitoren, Pflanzenwuchsförderer, Pflanzenwuchshemmer, Konservierungsmittel, Antikonservierungsmittel, Desinfektionsmittel, Sterilisationsmittel, Katalysatoren, chemische Reaktanden, Gärungsmittel, Nahrungsmittel, Nahrungsergänzungsmittel, Nährstoffe, Kosmetika, Medikamente, Vitamine, Mittel zur sexuellen Sterilisation, Fruchtbarkeitshemmer, Fruchtbarkeitsförderer, Mittel zur Schwächung von Mikroorganismen und andere Mittel, die in einer Gebrauchsumgebung von Nutzen sind. Die Begriffe, wie sie hier verwendet werden, umfassen ferner jede physiologisch oder pharmakologisch aktive Substanz, die eine lokale oder systemische Wirkung oder Wirkungen bei Tieren einschließlich warmblütiger Säugetiere, Menschen und Primaten, Vögeln, Haustieren oder auf Bauernhöfen gehaltenen Tiere wie etwa Katzen, Hunde, Schafe, Ziegen, Vieh, Pferden und Schweinen, Labortieren wie etwa Mäusen, Ratten und Versuchkaninchen, Fischen, Reptilien, Zoo- und Wildtieren und dergleichen hervorruft. Das aktive Medikament, das verabreicht werden kann, umfasst anorganische und organische Verbindungen einschließlich, jedoch nicht ausschließlich, Medikamenten, die auf die peripheren Nerven, adrenerge Rezeptoren, cholinerge Rezeptoren, die Skelettmuskeln, das kardiovaskuläre System, glatte Muskeln, das Blutkreislaufsystem, synoptische Stellen, Neuroeffektorübergangsstellen, das endokrine System und Hormonsysteme, das Immunsystem, das reproduktive System, das Skelettsystem, autocoide Systeme, die alimentären und exkretorischen Systeme, das Histaminsystem und das zentrale Nervensystem wirken. Geeignete Mittel können beispielsweise unter Proteinen, Enzymen, Hormonen, Polynukleotiden, Nukleoproteinen, Polysacchariden, Glucoproteiden, M.lipoproteiden, Polypeptiden, Steroiden, Hypnotika und Sedativa, Aufputschmitteln, Beruhigungsmitteln, krampflösenden Mitteln, Muskelentspannungsmitteln, Mitteln gegen Parkinson-Krankheit, Schmerzmitteln, Entzündungshemmern, Mitteln zur lokalen Betäubung, Muskel kontrahierenden Mitteln, Antibiotika, Mitteln gegen Malaria, Hormonmitteln einschließlich Kontrazeptiva, Symphatikomimetika, Polypeptiden und Proteinen, die physiologische Wirkungen hervorrufen können, Diuretika, flüssigkeitsregulierenden Mitteln, antiandrogenen Mitteln, Mitteln gegen Parasiten, Mitteln für die Neubildung von Gewebe, Mitteln gegen die Neubildung von Gewebe, Hypoglykämika, Nahrungsmitteln und Nahrungsergänzungsmitteln, Zusatzstoffen für das Wachstum, Fetten, Mitteln gegen Augenentzündung, Mitteln gegen Darmentzündung, Elektrolyten und diagnostischen Mitteln gewählt werden.

Weitere Beispiele sind die Proteine und Proteine, die einschließlich, jedoch nicht ausschließlich, Insulin, Colchicin, Glucagon, die Schilddrüse stimulierende Hormone, Nebenschilddrüsen- und Hirnanhangdrüsenhormone, Calcitonin, Renin, Prolaktin, Corticotropin, thyreotrope Hormone, follikelstimulierende Hormone, Chorion-Gonadotropin, vom Schwein gewonnenes Somatotropin, Oxytocin, Vasopressin, Prolaktin, Somatostatin, Lypressin, Pancreozymin, luteinisierende Hormone, LHRH, Interferone, Interleukine, Wachstumshormone wie etwa menschliche Wachstumshormone, Wachstumshormone von Rindern und von Schweinen, Fruchtbarkeitshemmer wie etwa die Prostaglandine, Fruchtbarkeitsförderer, Wachstumsfaktoren und den Releasingfaktor des Hormons der menschlichen Bauchspeicheldrüse enthalten.
- Selbstverständlich kann in der Verabreichungsform, die Membranen gemäß der Erfindung aufweist, mehr als ein therapeutisches Mittel enthalten sein, wobei die Verwendung der Ausdrücke "therapeutisches Mittel" und "Arzneimittel" keineswegs die Verwendung von zwei oder mehrerer solcher therapeutischen Mittel oder Arzneimittel ausschließt. Das therapeutische Mittel kann in einer breiten Vielfalt chemischer und physikalischer Formen wie etwa als ungeladene Moleküle, Komponenten von molekularen Komplexen, nicht reizende pharmazeutisch annehmbare Salze, therapeutische Derivate des therapeutischen Mittels wie etwa Ether, Ester, Amide usw. vorkommen, wobei in der Erfindung auch therapeutische Derivate des therapeutischen Mittels, die vom Körper ohne weiteres pH-hydrolisiert werden können, und Enzyme eingeschlossen sind. Die Menge des therapeutischen Mittels in der Verabreichungsform entspricht der zum Hervorrufen der gewünschten therapeutischen Reaktion erforderlichen Menge. In der Praxis wird diese weitgehend vom speziellen therapeutischen Mittel, von der Verabreichungsstelle, vom Gesundheitszustand und von der gewünschten therapeutischen Wirkung abhängen. Somit entspricht es häufig nicht der Praxis, einen speziellen therapeutischen Bereich für die therapeutisch wirksame Dosis des in der Verabreichungsform enthaltenen therapeutisch aktiven Mittels zu definieren, jedoch enthält die Verabreichungsform im Allgemeinen 10 ng bis 2,5 g des therapeutischen Mittels. Die therapeutisch aktiven Arzneimittel sind in Pharmaco-therapy, Bd. 8, S. 147–157 (1988); Drugs, Bd. 30, S. 333–354 (1985); Remington's Pharmacological Basis of Therapeutics von Goodman und Gilman, 4. Ausgabe, 1970, herausgegeben von The Man Million Company, London offenbart.
- Der Begriff "Osmagent", wie er hier verwendet wird, umfasst auch osmotisch wirksame Lösungsstoffe, osmotisch wirksame Verbindungen und osmotisch Mittel. Osmotisch wirksame Mischungen, die zum Zweck dieser Erfindung verwendet werden können, umfassen anorganische und organische Verbindun gen, die einen osmotischen Druckgradienten entlang einer für ein äußeres Fluid halbdurchlässige Membran aufweisen. Osmotisch wirksame Verbindungen, die für den gegebenen Zweck nützlich sind, umfassen Magnesiumsulfat, Magnesiumchlorid, Natriumchlorid, Lithiumchlorid, Kaliumsulfat, Natriumkarbonat, Natriumsulfat, Lithiumsulfat, Kaliumchlorid, Natriumsulfat, Kalziumbikarbonat, Kalziumsulfat, Kaliumsäurephosphat, Kalziumlactat, Mannit, Urea, Inositol, Magnesiumsuccinat, Weinsäure, Kohlenhydrate, Raffinose, Saccharose, Glucose, Lactosemonohydrat und Mischungen davon. Der osmotisch wirksame Lösungsstoff kann in irgendeiner physikalischen Form, etwa als Partikel, Kristall, Pille, Tablette, Streifen, Film, in Körnchenform, gemahlen oder pulverisiert vorkommen. Osmotisch wirksame Lösungsstoffe und Verfahren zum Messen osmotischer Drücke sind im US-Patent Nr. 5.232.705 offenbart.
- Die schiebende Zusammensetzung enthält ein schäumbares Mittel, das auch als Osmopolymer oder Hydrogel bekannt ist, wobei das schäumbare Element in der Verabreichungsform für den Zweck dieser Erfindung eine schiebende Zusammensetzung enthält, die mit Wasser oder wässerigen biologischen Fluiden zusammenwirkt und aufquillt oder schäumt, bis ein Gleichgewichtszustand erreicht ist. Die Osmopolymere besitzen die Eigenschaft, in Wasser aufzuquellen und einen großen Teil des aufgenommenen Wassers in der Polymerstruktur zu halten. Die Osmopolymere quellen oder schäumen in einem starken Maße auf und weisen eine Volumenzunahme um das 2fache bis 50fache auf. Die Osmopolymere können vernetzt oder nicht vernetzt sein. Die aufquellenden, wasseraufsaugenden Polymere sind in einer gegenwärtig bevorzugten Ausführungsform insofern vernetzt, dass durch Kovalenz- oder Ionenbindungen Vernetzungen gebildet sind. Die Osmopolymere können pflanzlichen, tierischen oder synthetischen Ursprungs sein. Hydrophile Polymere, die für den gegebenen Zweck geeignet sind, umfassen Poly(Hydroxyalkyl-methacrylsäureester) mit einem Molekulargewicht von 30.000 bis 5.000.000, Poly(Vinylpyrrolidon) mit einem Molekulargewicht von 10.000 bis 360.000, anionische und kationische Hydrogele, Polyelektrolytkomplexe, Poly(Vinylalkohol), der einen geringen Acetat-Rest aufweist, mit Formaldehyd oder Glutaraldehyd vernetzt ist und einen Polymerisationsgrad von 200 bis 30.000 aufweist, ein Gemisch aus Methylzellulose, vernetztem Agar-Agar und vernetzter Carboxymethylzellulose, einem wasserunlöslichen, in Wasser aufquellenden Copolymer, das durch Bilden einer Dispersion von fein verteiltem Copolymer aus Maleinsäure-anhydrid mit Styrol hergestellt ist, Ethylen, Propylen und Butylen oder Isobutylen, das mit 0,0001 bis etwa 0,5 mol poly-ungesättigtem Vernetzungsmittel pro Mol Maleinsäureanhydrid vernetzt ist, und in Wasser aufquellende Polymere von N-vinyllactamen und dergleichen.
- Weitere Osmopolymere umfassen Polymere, die Hydrogele wie etwa acidische Carbopol®-Carboxy-Polymere bilden, die im Allgemeinen ein Molekulargewicht von 450.000 bis 4.000.000 besitzen, das Natriumsalz und andere Metallsalze von acidischen Carbopol®-Carboxy-Polymeren, Cyanamer®-Polyacrylamide, vernetzte, in Wasser aufquellende Inden-Maleinsäure-anhydrid-Polymere; die Goodrite®-Polymeracrylsäure, die beispielsweise, jedoch nicht darauf eingeschränkt, ein Molekulargewicht von 80.000 bis 200.000 besitzt, deren Natriumsalz und deren andere Metallsalze, Polyox®-Polyethylenoxid-Polymere mit einem Molekulargewicht von 100.000 bis 7.800.000, Stärke-Graphtcopolymere; Auqua-Keeps®-Acrylat-Polymere, mit Diester vernetztes Polyglucan und dergleichen. Beispiele von Hydrogele bildenden Polymeren sind dem Stand der Technik aus dem US-Patent Nr. 3.865108, erteilt an Hartop, aus dem US-Patent Nr. 4,207.893, erteilt an Michaels, und aus dem Handbook of Common Polymers von Scott und Roff, veröffentlicht von Chemical Rubber, CRC Press, Cleveland, OH bekannt.
- Weitere Osmopolymere, die in der ersten Schicht vorhanden sein können, umfassen Agarose, Alginate, Amylopectin, Arabinoglactan, Carrageen, Eucheuma, Fuciodan, Furcellaran, Gelatine, Guar-Gummi, Gummi agar, Gummiarabikum, Gummi ghatti, Gummi karaya, Gummi tragacanth, Hypnea, Laminarin, Heuschreckenbohnengummi, Pectin, Polyvinylalkohol, Polyvinylpyrrolidon, Propylenglycolalginate, N-vinyllactam-polysaccharide, Xanthan-Gummi und dergleichen. Die Osmopolymere sind aus Controlled Release System, Fabrication Technology, Bd. II, S. 46 (1988), veröffentlicht von CRC Press, Inc. bekannt.
- Eine Zusammensetzung, die ein therapeutisches Mittel zur Verwendung in Verabreichungsformen mit Membranen gemäß der Erfindung enthält, wird durch Standard-Herstellungstechniken hergestellt. Beispielsweise wird bei einer Herstellung ein therapeutisches Mittel mit Zutaten, die die Zusammensetzung bilden, vermischt und dann in eine feste Form gepresst, die den Innenabmessungen des Raums innerhalb der Verabreichungsform entspricht. In einer anderen Ausführungsform wird das therapeutische Mittel mit anderen Zutaten, die die Zusammensetzung bilden, durch herkömmliche Verfahren wie etwa die Verarbeitung in einer Kugelmühle, die Kalandrierung, das Vormischen oder die Verarbeitung in einem Mischwalzwerk zu einer festen oder halbfesten Masse vermischt und dann zu einer vorgegebenen, eine Schicht bildenden Form gepresst. Eine schiebende, aufschäumende Zusammensetzung, die ein Osmopolymer enthält, wird in ähnlicher Weise präpariert und zu einer Form gepresst, die der inneren Struktur der Verabreichungsform entspricht. Verfahren zum Präparieren einer therapeutische Mittel enthaltenden Zusammensetzung und einer Osmopolymere enthaltenden Zusammensetzung sind im US-Patent Nr. 5.024.834 offenbart.
- Die folgenden Beispiele veranschaulichen die Erfindung.
- Eine Verabreichungsform, die eine aus einer Zusammensetzung aus Polycaprolacton, Polyethylenodxid und Polyethylenglykol bestehende Druckgussmembran enthält, die einen Innenraum mit einer offenen Ausmündung und einem dosierten Boden umgibt, wird an ihrem Boden mit einer schiebenden Zusammensetzung und danach mit einer Arzneimittelzusammensetzung an der offenen Ausmündung gefüllt. Die schiebende Zusammensetzung enthält 58,75 Gew.-% Natriumcarboxymethylzellulose, 30,00 Gew.-% Natriumchlorid, 5,00 Gew.-% Hydroxypropyl-methylzellulose, 5,00 Gew.-% Hydroxypropylzellulose, 1,00 Gew.-% rotes Eisenoxid und 0,25 Gew.-% Magnesiumstearat. Die Arzneimittelzusammensetzung enthält 66,70 Gew.-% Gemfibrozil, 14,30 Gew.-% Säure-Di-Sol, ein Natrium-Croscarmellos, 9,50 Gew.-% Polyethylenoxid mit einem Molekulargewicht von 200.000, 5,00 Gew.-% Myrj 52-S-Tensid, auch als Poly(Ethylenglycolat 40)Stearat bekannt, 3,00 Gew.-% Hydroxypropyl-methylzellulose mit einem Molekulargewicht von 9.200, 1,00 Gew.-% Cab-O-sil, ein kolloidales Siliciumdioxid, und 0,50 Gew.-% Magnesiumstearat. Die offene Ausmündung der Verabreichungsform ist zu einer Öffnung von 15 mil (0,381 mm) gekröpft, um eine osmotische Verabreichungsform zu schaffen. Eine durch eine Lösungsmitteltechnik gebildete Verabreichungsform des Standes der Technik ist im US-Patent Nr. 4.327.725, erteilt an Cortese und Theeuwes und übertragen an die Firma ALZA, offenbart.
- Eine weitere durch die Endung geschaffene Verabreichungsform umfasst ein Gerüst aus einem ersten Membranabschnitt und einem zweiten Membranabschnitt. Der erste Abschnitt und der zweite Abschnitt sind so beschaffen, dass sie in teleskopischer Anordnung aneinander anschließen. Der membranbildende erste Abschnitt ist aus einer Polycaprolacton-Hydroxypropylzellulose-Zusammensetzung druckgegossen, während der zweite Abschnitt Polycaprolacton, Polyethylenoxid und Ethylenglykol in verschiedenen Verhältnissen enthält. Der erste Abschnitt enthält eine therapeutische Zusammensetzung für die Verabreichung an ein Tier oder einen Menschen. Der zweite Abschnitt ist ein Mittel, das den ersten Abschnitt während der Lagerung als Kappe verschließt. Der zweite Abschnitt ist ein Mittel, das die Verabreichungsform öffnet, wenn sie in einer Fluidumgebung in Gebrauch ist, um eine therapeutische Zusammensetzung aus dem ersten Abschnitt an die Umgebung abzugeben. Der zweite Abschnitt umfasst eine osmotische Schicht, die 58,75 Gew.-% Natriumcarboxymethylzellulose, 30,00 Gew.-% Natriumchlorid, 5,00 Gew.-% Hydroxypropyl-methylzellulose mit einem Molekulargewicht von 9.200, 5,00 Gew.-% Hydroxypropylzellulose, 1,00 Gew.-% rotes Eisenoxid und 0,25 Gew.-% Magnesiumstearat enthält. Die osmotische Schicht wird am Boden des zweiten Abschnitts angeordnet. Als nächstens wird eine Grenzschicht, die 95,00 Gew.-% Stearinsäure und 5,00 Gew.-% Hydroxypropyl-methylzellulose mit einem Molekulargewicht von 9.200 enthält, in Zweischichtanordnung mit der osmotischen Schicht am Abschnitt angeordnet. Die Verabreichungsform wird so zusammengesetzt, dass das schmälere offene Ende innerhalb des offenen Endes der Schicht angebracht ist, und zusammengedrückt, bis der erste Abschnitt und der zweite Abschnitt eng miteinander verbunden sind. Eine durch Lösungsmitteltechniken hergestellte Verabreichungsform ist im US-Patent Nr. 5.312.388, erteilt an Wong, Theeuwes und Larson und übertragen an die Firma ALZA offenbart. Zum Zweck dieser Erfindung können herkömmliche Druckgussmaschinen, wie sie in Encyclopedia of Polymer Science and Engineering, Bd. 8, Injection-Molding, S. 102–138, 1987 offenbart sind, verwendet werden.
- Eine Verabreichungsform, die eine flüssige Mischung nach Formel enthalten soll, wird durch Druckgießen eines Gerüstelements, das die flüssige Mischung nach Formel enthält, hergestellt. Das Gerüstelement umfasst eine halbdurchlässige Membran, die eine innere Zelle umgibt. Das Gerüst besitzt eine weit geöffnete Ausmündung und einen geschlossenen Boden. Die druckgegossene Membran umfasst ein Gemisch aus Polycaprolacton und einem Zellulosederivat oder Polycaprolacton und Polyethylenoxid-Polyethylenglykol, um eine halbdurchlässige Membran zu bilden. Das Zellulosederivat ist aus der Gruppe aus gewählt, die aus Zelluloseester, Zelluloseether, Hydroxyalkylzellulose und Hydroxypropyl-alkylzellulose besteht. Die Verabreichungsform wird hergestellt, indem zuerst in dem Boden des Gerüsts eine aufschäumbar-schiebende, gepresste Schicht, die 79,00 Gew.-% Kelton HV, Natriumalginat, 15,00 Gew.-% Kalziumsulfat, 5,00 Gew.-% Poly(Vinylpyrrolidon), 0,50 Gew.-% Eisenoxid und 0,50 Gew.-% Magnesiumstearat enthält, angeordnet wird; als Nächstes wird eine Grenzschicht, die verhindert, dass sich eine flüssige Mischung nach Formel mit der aufschäumbar-schiebenden Schicht vermischt, in einer kontaktherstellenden Zweischichtanordnung in das offene Gerüstelement eingeführt. Die Grenzschicht enthält 95,00 Gew.-% Stearinsäure und 5,00 Gew.-% Hydroxypropyl-methylzellulose. Als Nächstes wird die Membran, die die offene Ausmündung umgibt, gegen den Luftdruck gekröpft, was eine Öffnung von 30 mil hinterlässt. In das gekröpfte Gerüstelement wird danach durch die Öffnung hindurch eine flüssige Mischung nach Formel eingeführt. Die flüssige Mischung nach Formel enthält 41,75 Gew.-% Cremophor, ein hydriertes Polyoxyl-40-Rizinusöl, 35,05 Gew.-% Maisöl, 21,97 Gew.-% Propylenglykol, 1,10 Gew.-% Tocopherol und 0,14 Gew.-% blaue Farbe.
Claims (6)
- Membran, die 20% bis 90% Polycaprolacton und 10% bis 80% Poly(alkenoxid) umfasst, wobei die Prozentsätze insgesamt 100 Gew.-% ergeben.
- Membran, die 10% bis 97% Polycaprolacton, 10% bis 97% Poly(Alkenoxid) und 1% bis 97% Poly(Ethylenglykol) umfasst, wobei die Prozentsätze insgesamt 100 Gew.-% ergeben.
- Membran, die 1% bis 90% Polycaprolacton, 1% bis 90% Poly(alkenoxid), 1% bis 90% Poly(Hydroxypropylzellulose) und 1% bis 90% Poly(Ethylenglykol) umfasst, wobei die Prozentsätze insgesamt 100 Gew.-% ergeben.
- Membran, die 20% bis 90% Polycaprolacton und 10% bis 80% Poly(Hydroxypropylzellulose) umfasst, wobei die Prozentsätze insgesamt 100 Gew.-% ergeben.
- Membran nach einer der vorhergehenden Ansprüche, wobei die Membran durch Druckguss gebildet ist.
- Verabreichungsform, die eine Membran nach einem der vorhergehenden Ansprüche umfasst.
