Description
- Die vorliegende Erfindung betrifft eine neue Klasse von Verbindungen, deren Salze, pharmazeutische Zusammensetzungen, die diese enthalten, Verfahren für deren Herstellung und deren Verwendung bei der Herstellung eines Medikaments zur Behandlung des menschlichen Körpers. Insbesondere betrifft die Erfindung neue Sulfone, welche die Verarbeitung von APP durch γ-Sekretase modulieren und somit zur Herstellung eines Medikaments zur Behandlung oder Prävention von Alzheimer-Krankheit geeignet sind.
- Alzheimer-Krankheit (AD) ist die häufigste Form von Demenz. Zwar ist sie hauptsächlich eine Erkrankung, die ältere Menschen betrifft und an der bis zu 10% der Bevölkerung mit einem Alter von über 65 leiden, an AD leidet jedoch auch eine bedeutende Zahl von jüngeren Patienten mit einer genetischen Veranlagung. Sie ist eine neurodegenerative Störung, die klinisch durch einen fortschreitenden Gedächtnisverlust und Verlust der Wahrnehmungsfunktion gekennzeichnet ist und pathologisch durch die Ablagerung extrazellulärer proteinhaltiger Plaques in den kortikalen und assoziativen Gehirnregionen des Erkrankten gekennzeichnet ist. Diese Plaques enthalten hauptsächlich fibrilläre Aggregate aus Amyloidpeptid (Aβ). Die Rolle von Sekretasen, einschließlich putativer γ-Sekretase, bei der Verarbeitung von Amyloidvorläuferprotein (APP), um Aβ zu erzeugen, ist in der Literatur gut dokumentiert und z. B. in der
WO 01/70677 - In der Literatur gibt es relativ wenige Berichte über Verbindungen mit inhibitorischer Wirkung gegenüber γ-Sekretase, gemessen in auf Zellen basierenden Assays. Diese sind in der
WO 01/70677 - Die vorliegende Erfindung stellt eine neue Klasse von nichtpeptidischen Verbindungen zur Verfügung, welche sich zur Behandlung oder Prävention von AD durch Modulierung der Verarbeitung von APP durch die putative γ-Sekretase, so dass die Produktion von Aβ aufgehalten wird, eignen.
- Die vorliegende Erfindung stellt eine Verbindung der Formel I oder ein pharmazeutisch annehmbares Salz davon zur Verfügung: wobei:

Z CN, OR2a, CO2R2a oder CON(R2a)2 bedeutet,
R1b H, C1-4-Alkyl oder OH bedeutet,
R1c H oder C1-4-Alkyl bedeutet,
Ar1 Phenyl oder Pyridyl bedeutet, wobei jedes davon 0–3 Substituenten trägt, unabhängig ausgewählt aus Halogen, CN, NO2, CF3, OH, OCF3, C1-4-Alkoxy oder C1-4-Alkyl, welches gegebenenfalls einen Substituenten trägt, ausgewählt aus Halogen, CN, NO2, CF3, OH und C1-4-Alkoxy,
Ar2 Phenyl bedeutet, das in den 2- und 5-Positionen mit Halogen substituiert ist,
R2a H, C1-6-Alkyl, C3-6-Cycloalkyl, C3-6-Cycloalkyl-C1-6-alkyl, C2-6-Alkenyl bedeutet, wobei jedes davon gegebenenfalls einen Substituenten trägt, ausgewählt aus Halogen, CN, NO2, CF3, OR2b, CO2R2b, N(R2b)2, CON(R2b)2, Ar und COAr, oder R2a Ar bedeutet, oder zwei R2a-Gruppen zusammen mit einem Stickstoffatom, an das sie gemeinsam gebunden sind, eine N-Heterocyclylgruppe vervollständigen können, die 0–4 Substituenten trägt, unabhängig ausgewählt aus =O, =S, Halogen, C1-4-Alkyl, CN, NO2, CF3, OH, C1-4-Alkoxy, C1-4-Alkoxycarbonyl, CO2H, Amino, C1-4-Alkylamino, Di(C1-4-alkyl)amino, Carbamoyl, Ar und COAr,
R2b H, C1-6-Alkyl, C3-6-Cycloalkyl, C3-6-Cycloalkyl-C1-6-alkyl, C2-6-Alkenyl bedeutet, wobei jedes davon gegebenenfalls einen Substituenten trägt, ausgewählt aus Halogen, CN, NO2, CF3, OH, C1-4-Alkoxy, C1-4-Alkoxycarbonyl, CO2H, Amino, C1-4-Alkylamino, Di(C1-4-alkyl)amino, Carbamoyl, Ar und COAr, oder R2b Ar bedeutet, oder zwei R2b-Gruppen zusammen mit einem Stickstoffatom, an das sie gemeinsam gebunden sind, eine N-Heterocyclylgruppe vervollständigen können, die 0–4 Substituenten trägt, unabhängig ausgewählt aus =O, =S, Halogen, C1-4-Alkyl, CN, NO2, CF3, OH, C1-4-Alkoxy, C1-4-Alkoxycarbonyl, CO2H, Amino, C1-4-Alkylamino, Di(C1-4-alkyl)amino, Carbamoyl, Ar und COAr, - Ar Phenyl oder Heteroaryl bedeutet, das 0–3 Substituenten trägt, ausgewählt aus Halogen, C1-4-Alkyl, CN, NO2, CF3, OH, C1-4-Alkoxy, C1-4-Alkoxycarbonyl, Amino, C1-4-Alkylamino, Di(C1-4-alkyl)amino, Carbamoyl, C1-4-Alkylcarbamoyl und Di(C1-4-alkyl)carbamoyl.
- Wenn eine Variable mehr als einmal in Formel I oder in einem Substituenten davon vorkommt, sind die einzelnen Vorkommen dieser Variablen unabhängig voneinander, sofern nichts anderes angegeben ist.
- So wie hier verwendet, bedeutet die Bezeichnung "C1-x-Alkyl", wobei x eine Zahl größer als 1 ist, geradkettige und verzweigte Alkylgruppen, wobei die Anzahl der einzelnen Kohlenstoffatome im Bereich von 1 bis x liegt. Spezielle Alkylgruppen sind u. a. Methyl, Ethyl, n-Propyl, Isopropyl und t-Butyl. Abgeleitete Bezeichnungen, wie z. B. "C2-6-Alkenyl", "Hydroxy-C1-6-alkyl", "Heteroaryl"-C1-6-alkyl, "C2-6-Alkinyl" und "C1-6-Alkoxy" sollen auf analoge Weise ausgelegt werden.
- Die Bezeichnung "C3-6-Cycloalkyl", so wie hier verwendet, bedeutet nichtaromatische monocyclische oder kondensierte bicyclische Kohlenwasserstoff-Ringsysteme, die 3 bis 6 Ringatome enthalten. Beispiele sind u. a. Cyclopropyl, Cyclobutyl, Cyclopentyl, Cyclohexyl und Cyclohexenyl.
- Die Bezeichnung "C3-6-Cycloalkyl(C1-6)alkyl", so wie hierin verwendet, umfasst Cyclopropylmethyl, Cyclobutylmethyl, Cyclopentylmethyl und Cyclohexylmethyl.
- Die Bezeichnung "N-Heterocyclyl", so wie hier verwendet, bedeutet ein cyclisches oder polycyclisches System mit bis zu 10 Ringatomen, ausgewählt aus C, N, O und S, wobei keines der beteiligten Ringe aromatisch ist und wobei wenigstens ein Ringatom Stickstoff ist und die Bindung durch den Ring-Stickstoff erfolgt. Bevorzugte N-Heterocyclylgruppen sind monocyclische Systeme aus 4–6 Gliedern, wie z. B. Azetidinyl, Pyrrolidinyl, Piperidinyl, Piperazinyl, Morpholinyl, Thiomorpholinyl, Imidazolidinyl, Oxazolidinyl und Thiazolidinyl.
- Die Bezeichnung "Heteroaryl", so wie hier verwendet, bedeutet ein cyclisches oder polycyclisches System mit bis zu 10 Ringatomen, ausgewählt aus C, N, O und S, wobei wenigstens einer der beteiligten Ringe aromatisch ist und wenigstens ein Ringatom anders als Kohlenstoff enthält. Bevorzugte Heteroarylgruppen sind monocyclische Systeme aus 5 oder 6 Gliedern, wie z. B. Pyridinyl-, Pyridazinyl-, Pyrimidinyl-, Pyrazinyl-, Pyrrolyl-, Furyl-, Thienyl-, Pyrazolyl-, Oxazolyl-, Isoxazolyl-, Thiazolyl-, Isothiazolyl-, Imidazolyl-, Oxadiazolyl-, Triazolyl- und Thiadiazolylgruppen. Weitere Beispiele für Heteroarylgruppen sind u. a. Tetrazol, 1,2,4-Triazin und 1,3,5-Triazin.
- Die Bezeichnung "Halogen", so wie hier verwendet, umfasst Fluor, Chlor, Brom und Iod, wobei Fluor und Chlor bevorzugt sind.
- Zur medizinischen Verwendung können die Verbindungen der Formel I vorteilhafterweise in Form von pharmazeutisch annehmbaren Salzen vorliegen, es können jedoch auch andere Salze zur Herstellung der Verbindungen oder ihrer pharmazeutisch annehmbaren Salze geeignet sein. Geeignete pharmazeutisch annehmbare Salze der Verbindungen dieser Erfindung sind u. a. Säureadditionssalze, wie z. B. diejenigen, die mit Salzsäure, Schwefelsäure, Methansulfonsäure, Fumarsäure, Maleinsäure, Succinsäure, Essigsäure, Benzoesäure, Oxasäure, Citronensäure, Weinsäure, Kohlensäure oder Phosphorsäure gebildet werden, und, wenn die Verbindungen der Erfindung einen sauren Rest tragen, Natrium-, Kalium, Calcium- oder Magnesiumsalze und Salze, die mit geeigneten organischen Liganden gebildet werden, z. B. quaternäre Ammoniumsalze und Pyridiniumsalze.
- Wenn die erfindungsgemäßen Verbindungen wenigstens ein Asymmetriezentrum besitzen, können sie entsprechend als Enantiomere vorliegen. Wenn die Verbindungen gemäß der Erfindung zwei oder mehrere Asymmetriezentren besitzen, können sie zusätzlich als Diastereoisomere vorliegen. Es ist zu verstehen, dass alle solchen Isomere und Mischungen davon in beliebigen Proportionen vom Umfang der vorliegenden Erfindung umfasst sind.
- Ungeachtet des Vorliegen oder der Abwesenheit von Asymmetriezentren können bestimmte erfindungsgemäße Verbindungen aufgrund der Asymmetrie des Moleküls als Ganzes als Enantiomere vorliegen. Es ist zu verstehen, dass in solchen Fällen beide Enantiomere und Mischungen davon in einem beliebigen Anteil vom Umfang der vorliegenden Erfindung umfasst sind, und dass die Strukturformeln, die Moleküle dieses Typs zeigen, für beide möglichen Enantiomere stellvertretend sein sollen, sofern nichts anderes angegeben ist.
- Bei den Verbindungen der Formel I bedeutet Ar1 gegebenenfalls substituiertes Phenyl oder Pyridyl, speziell gegebenenfalls substituiertes Phenyl oder 3-Pyridyl. Ar1 ist vorzugsweise ausgewählt aus Phenylgruppen, die in der 4-Position mit Halogen, Methyl oder Trifluormethyl substituiert sind, und Phenylgruppen, die in den 3- und 4-Positionen durch Halogen substituiert sind.
- Ar2 ist vorzugsweise 2,5-Difluorphenyl.
- Bei speziellen Ausführungsformen ist Ar1 4-Chlorphenyl oder 4-Trifluormethylphenyl, und Ar2 ist 2,5-Difluorphenyl.
- R1b bedeutet typischerweise H, Methyl oder OH, vorzugsweise H.
- R1c bedeutet typischerweise H oder Methyl, vorzugsweise H.
- Wenn m 1 ist, bedeuten R1b und R1c vorzugsweise nicht beide C1-4-Akyl.
- Spezielle Werte für R2a sind u. a. H, Phenyl, Pyridyl, C3-6-Cycloalkyl (wie z. B. Cyclopropyl, Cyclobutyl und Cyclopentyl), C3-6-Cycloalkyl-C1-6-alkyl (wie z. B. Cyclopropylmethyl), C2-6-Alkenyl (wie z. B. Allyl) und lineares oder verzweigtes C1-6-Alkyl, das gegebenenfalls substituiert ist mit CF3, Ar, OR2b, N(R2b)2, CO2R2b oder CON(R2b)2.
- Beispiele für N-Heterocyclylgruppen, die durch N(R2a)2 dargestellt sind, sind u. a. Piperidin-1-yl (gegebenenfalls substituiert mit OH, CO2H, CO2C1-4-Alkyl, Me oder Ph), Piperizin-1-yl (gegebenenfalls substituiert mit Me oder Ph), Morpholin-4-yl, Thiomorpholin-4-yl, 1,1-Dioxothiomorpholin-4-yl, 2-Oxoimidazolidin-1-yl, 5,5-Dimethyl-2,2-dioxooxazolidin-3-yl, 2,5-Dioxoimidazolidin-1-yl, 2-Oxooxazolidin-3-yl, 2-Oxopyridin-1-yl und 2-Oxopyrrolidin-1-yl.
- R2b bedeutet typischerweise H oder C1-4-Alkyl.
- Wenn Z OR2a bedeutet, bedeutet R2a geeigneterweise H, Ar (insbesondere Pyridyl), Alkyl (wie z. B. Methyl, Ethyl, Propyl oder Butyl) oder substituiertes Alkyl (insbesondere CH2Ar, wie z. B. Benzyl oder Pyridylmethyl).
- Wenn Z CO2R2a bedeutet, bedeutet R2a geeigneterweise H oder Alkyl (wie z. B. Methyl, Ethyl, Propyl oder Butyl).
- Wenn Z CON(R2a)2 bedeutet, bedeuten die R2a-Gruppen unabhängig H oder gegebenenfalls substituiertes Alkyl, Cycloalkyl, Cycloalkylalkyl oder Alkenyl oder vervollständigen zusammen eine N-Heterocyclylgruppe. Besonders bevorzugt bedeutet einer der Reste R2a H, und der andere bedeutet H, Alkyl (wie z. B. Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, sek.-Butyl, tert.-Butyl oder 1-Ethylpropyl), Alkenyl (wie z. B. Allyl), Cycloalkyl (wie z. B. Cyclopropyl, Cyclobutyl oder Cyclopentyl), Cycloalkylalkyl (wie z. B. Cyclopropylmethyl) oder substituiertes Alkyl (wie z. B. Alkyl, substituiert mit Ar, insbesondere 2-Pyridylethyl, 3-(imidazol-1-yl)propyl oder 2-Phenylethyl; oder Alkyl, substituiert mit CF3, CO2R2b oder CON(R2b)2, insbesondere 2,2,2-Trifluorethyl, Methoxycarbonylmethyl oder Carbamoylmethyl). Alternativ vervollständigen die zwei R2a-Gruppen eine N-Heterocyclylgruppe, wie z. B. Morpholin, Thiomorpholin, Thiomorpholin-1,1-dioxid, 4-Methylpiperazin, 4-Phenylpiperazin, Piperidin, 4-Hydroxypiperidin oder Piperidin, welche in der 3- oder 4-Position mit CO2R2b und/oder C1-4-Alkyl substituiert ist, insbesondere 3- oder 4-Carboxypiperidin, 3- oder 4-Ethoxycarbonylpiperidin, 3-Carboxy-3-methylpiperidin und 3-Ethoxycarbonyl-3-methylpiperidin.
- Beispiele für einzelne Verbindungen gemäß Formel I sind in dem sich anschließenden Beispielteil angegeben.
- Die Verbindungen der Formel I besitzen eine Wirkung als Modulatoren der Verarbeitung von APP durch γ-Sekretase.
- Die Erfindung stellt auch pharmazeutische Zusammensetzungen zur Verfügung, die eine oder mehrere Verbindungen der Formel I oder die pharmazeutisch annehmbaren Salze davon und einen pharmazeutisch annehmbaren Träger enthalten. Vorzugsweise liegen diese Zusammensetzungen in Einheitsdosisformen, wie z. B. Tabletten, Pillen, Kapseln, Pulvern, Granulaten, sterilen parenteralen Lösungen oder Suspensionen, Aerosol- oder Flüssigsprays mit Dosiervorrichtung, Tropfen, Ampullen, Transdermalpflaster, Autoinjektionsvorrichtungen oder Zäpfchen, zur oralen, parenteralen, intranasalen, sublingualen oder rektalen Verabreichung oder zur Verabreichung durch Inhalation oder Insufflation vor. Zur Herstellung fester Zusammensetzungen, wie z. B. Tabletten, wird der Hauptwirkstoff mit einem pharmazeutischen Träger, wie z. B. herkömmlichen Tablettierungsbestandteilen, die den Fachleuten bekannt sind, z. B. wie in der
WO 01/70677 WO 01/70677 - Die flüssigen Formen, in welche die neuen Zusammensetzungen der vorliegenden Erfindung zur oralen Verabreichung oder Verabreichung durch Injektion eingebracht werden können, sind u. a. wässrige Lösungen, geeignet aromatisierte Sirupe, wässrige oder Ölsuspensionen und aromatisierte Emulsionen mit Speiseölen, wie es in der
WO 01/70677 - Die vorliegende Erfindung stellt auch eine Verbindung der Formel I oder ein pharmazeutisch annehmbares Salz davon zur Verwendung bei einem Verfahren zur Behandlung des menschlichen Körpers zur Verfügung. Vorzugsweise erfolgt die Behandlung für einen Zustand, der mit der Ablagerung von β-Amyloid verbunden ist. Vorzugsweise ist der Zustand eine neurologische Erkrankung mit verbundener β-Amyloid-Ablagerung, wie z. B. Alzheimer-Krankheit.
- Die vorliegende Erfindung stellt ferner die Verwendung einer Verbindung der Formel I oder ein pharmazeutisch annehmbares Salz davon bei der Herstellung eines Medikaments zur Behandlung oder Prävention von Alzheimer-Krankheit zur Verfügung.
- Ebenfalls offenbart ist ein Verfahren zur Behandlung eines Subjekts, das an Alzheimer-Krankheit leidet oder zu Alzheimer-Krankheit neigt, umfassend die Verabreichung einer wirksamen Menge einer Verbindung gemäß Formel I oder eines pharmazeutisch annehmbaren Salzes davon an dieses Subjekt.
- Zur Behandlung oder Prävention von Alzheimer-Krankheit beträgt eine geeignete Dosismenge etwa 0,01 bis 250 mg/kg pro Tag, vorzugsweise etwa 0,05 bis 100 mg/kg pro Tag und speziell etwa 0,1 bis 50 mg/kg Körpergewicht pro Tag. Die Verbindungen können in einem Regime 1 bis 4 Mal pro Tag verabreicht werden. In einigen Fällen kann jedoch eine Dosierung außerhalb dieser Grenzen verwendet werden.
- Verbindungen der Formel I, bei denen m 0 ist und Z CO2R2a oder CON(R2a)2 ist, können durch Kupplung einer Carbonsäure (1) mit R2aOH bzw. HN(R2a)2 hergestellt werden, wobei Ar1, Ar2, R1c und R2a die gleichen Bedeutungen wie zuvor besitzen. Sämtliche Standard-Kupplungsverfahren können verwendet werden, einschließlich der Verwendung von Kupplungsmitteln wie Dimethylaminopyridin, Hydroxybenzotriazol, Dicyclohexylcarbodiimid, Carbonyldiimidazol und dergleichen. Bei einem bevorzugten Verfahren wird die Säure in das entsprechende Säurechlorid umgewandelt (z. B. durch Behandlung mit Oxalylchlorid in DMF-Lösung) und direkt mit dem erwünschten Nukleophil umgesetzt. Bei einem weiteren bevorzugten Verfahren wird die Säure in ein aktives Esterderivat, wie z. B. den Pentafluorphenolester, umgewandelt (z. B. durch Kupplung mit dem Phenol in Gegenwart von Dicyclohexylcarbodiimid), und dieses Zwischenprodukt wird mit dem erwünschten Nukleophil umgesetzt.

- Die Ester (2) sind durch Reduktion der Alkylidenderivate (3), gegebenenfalls gefolgt von Alkylierung mit (C1-4-Alkyl)-L, wobei L eine Abgangsgruppe (insbesondere Bromid oder Iodid) ist, wenn R1c anders als H ist, erhältlich: wobei Ar1, Ar2 und R2 die gleichen Bedeutungen wie zuvor besitzen. Die Reduktion kann unter Verwendung von Natriumborhydrid und Nickel(II)chlorid in Ethanol durchgeführt werden, während die optionale Alkylierung durch Behandlung des Esters (2, R1c = H)) mit starker Base (z. B. Natriumbis(trimethylsilyl)amid) in einem aprotischen Lösungsmittel bei niedriger Temperatur, gefolgt von der Behandlung mit (C1-4-Alkyl)-L- und Erwärmen auf Raumtemperatur, bewirkt werden kann.
- Falls erwünscht, können die ungesättigten Ester (3) zu den entsprechenden Säuren hydrolysiert und durch Umsetzung mit HN(R2a)2 in Amide umgewandelt werden, bevor die olefinische Bindung reduziert wird.
- Die ungesättigten Ester (3) sind durch Kondensation eines Ketons (4) mit Ph3P=CHCO2R2 erhältlich: wobei Ar1, Ar2 und R2 die gleichen Bedeutungen wie zuvor besitzen, während die Ketone (4) durch Decarboxylierung des Enols (5) erhältlich sind, die wiederum durch Reaktion eines Sulfons (6) mit wenigstens zwei Äquivalenten eines Acrylats (7) gebildet werden können: wobei Ar1, Ar2 und R2 die gleichen Bedeutungen wie zuvor besitzen. Die Decarboxylierung kann durch Erhitzen auf 150°C in DMSO in Gegenwart von Natriumchlorid und Wasser erreicht werden, während die Reaktion von (6) mit (7) bei Umgebungstemperatur in einem inerten Lösungsmittel, wie z. B. THF, in Gegenwart einer starken Base, wie z. B. Kalium-t-butoxid, durchgeführt werden kann.
- Die Sulfone (6) werden durch Oxidation der Thioether Ar2-CH2SAr1 (8) hergestellt, die wiederum durch Umsetzung von Thiolen Ar1SH (9) mit Benzylderivaten Ar2CH2-L (10) gebildet werden, wobei L eine Abgangsgruppe ist, wie z. B. Chlorid oder Bromid, und Ar1 und Ar2 die gleichen Bedeutungen wie zuvor besitzen. Die Reaktion zwischen (9) und (10) findet in einem inerten Lösungsmittel, wie z. B. Dichlormethan, in Gegenwart einer Base, wie z. B. Triethylamin, statt, während die Oxidation von (8) zu (6) zweckmäßigerweise durch m-Chlorperoxybenzoesäure ebenfalls in einem inerten Lösungsmittel, wie z. B. Dichlormethan, bewirkt wird.
- Verbindungen der Formel I, bei denen m 0 ist und Z CN oder OR2a ist, können durch Umsetzung eines Sulfonatesters (11) mit Cyanid-Ion bzw. R2aOH erhalten werden: wobei L1 eine Sulfonat-Abgangsgruppe (wie z. B. Mesylat, Tosylat oder Triflat) bedeutet und Ar1, Ar2, R1c und R2a die gleichen Bedeutungen wie zuvor besitzen. Die Verdrängungsreaktion kann in DMF bei erhöhter Temperatur, z. B. bei etwa 80°C, durchgeführt werden. Wenn das Nukleophil R2aOH ist, ist es von Vorteil, das entsprechende Anion durch Behandlung mit Natriumhydrid vor der Reaktion mit (11) zu erzeugen.
- Die Sulfonate (11) werden durch Reaktion der Alkohole (12) mit dem entsprechenden Sulfonylchlorid (z. B. unter wasserfreien Bedingungen bei niedriger Temperatur in Gegenwart eines tertiären Amins) hergestellt.
- Die Alkohole (12) sind durch die Hydroborierung von Alkenen (13) erhältlich: wobei Ar1, Ar2 und R1c die gleichen Bedeutungen wie zuvor besitzen. Das Verfahren umfasst typischerweise die Reaktion mit Boran in THF bei Raumtemperatur, gefolgt von der Behandlung mit alkalischem Wasserstoffperoxid und Abtrennung des erwünschten cis-Isomers durch Chromatographie. Alkene (13) sind durch Kondensation mit Ph3P=CHR1c aus Ketonen (4) erhältlich, wobei R1c die gleiche Bedeutung besitzt wie zuvor.
- Ein alternativer Weg zu den Alkoholen (12), bei denen R1c H ist, umfasst die Reduktion eines Ketons (4) (z. B. durch Verwendung von Borhydrid) zum entsprechenden sekundären Alkohol (14), die Umwandlung des Alkohols (14) in das entsprechende Mesylat (oder eine äquivalente Abgangsgruppe), die Durchführung einer nukleophilen Verdrängung mit Cyanid-Ion, die Hydrolyse des resultierenden Nitrils zur entsprechenden Carbonsäure, gefolgt von der Reduktion zum primären Alkohol. Die Hydrolyse wird typischerweise unter sauren Bedingungen (z. B. in einer Mischung aus Essigsäure und konz. HCl bei 110°C) durchgeführt, und die Reduktion wird zweckmäßigerweise durch sequentielle Behandlung mit Isobutylchlorformiat und Borhydrid in THF durchgeführt.
- Verbindungen der Formel I, bei denen m 1 ist und R1b H oder C1-4-Alkyl ist, können durch Oxidation eines Alkohols (12) zum entsprechenden Aldehyd oder Keton und Umwandlung der Carbonylgruppe darin in einer zuvor in Verbindung mit der Umwandlung von Ketonen (4) in Verbindungen der Formel I, bei denen m 0 ist, beschriebenen Weise erhalten werden. Zum Beispiel ergibt die Oxidation der Alkohole (12), bei denen R1c H ist, die entsprechenden Cyclohexancarboxaldehyde, die mit Ph3P=CO2Et kondensiert werden können, um Ethylester von 2-Cyclohexylpropensäuren zu ergeben. Diese wiederum können zu entsprechenden 2-Cyclohexylpropanoaten hydriert werden, die gegebenenfalls der Alkylierung und/oder der Hydrolyse zu den entsprechenden Säuren unterworfen werden können und/oder in eine Reihe von Amid- oder alternativ Esterderivaten umgewandelt werden können.
- Wenn sie selbst nicht im Handel erhältlich sind, können die in den oben beschriebenen Syntheseschemata eingesetzten Ausgangsmaterialien und Reagenzien durch Verwendung von Standardverfahren der organischen Synthese für im Handel erhältliche Materialien erhalten werden.
- Man wird erkennen, dass viele der oben beschriebenen Syntheseschemata Mischungen aus Stereoisomeren ergeben werden. Insbesondere können bestimmte Produkte als Mischungen aus cis- und trans-Isomeren gebildet werden, wobei ein spezieller Ringsubstituent sich auf derselben oder auf der gegenüberliegenden Seite des Rings wie die Arylsulfonylgruppe befindet. Solche Mischungen können durch herkömmliche Mittel, wie z. B. fraktionierte Kristallisation und präparative Chromatographie, getrennt werden.
- Bestimmte Verbindungen gemäß der Erfindung können aufgrund der Anwesenheit von einem oder mehreren Chiralitätszentren oder aufgrund der Gesamt-Asymmetrie des Moleküls als optische Isomere existieren. Solche Verbindungen können in racemischer Form hergestellt werden, oder einzelne Enantiomere können entweder durch enantiospezifische Synthese oder durch Auftrennen hergestellt werden. Die neuen Verbindungen können zum Beispiel durch Standardverfahren, wie z. B. präparativer HPLC oder Bildung diastereomerer Paare durch Salzbildung mit einer optisch aktiven Säure, wie z. B. (–)-Di-p-toluoyl-d-weinsäure und/oder (+)-Di-p-toluoyl-1-weinsaure, gefolgt von fraktionierter Kristallisation und Gewinnung der freien Base, in ihre Enantiomerenkomponenten aufgetrennt werden. Die neuen Verbindungen können auch durch Bildung diastereomerer Ester oder Amide, gefolgt von der chromatographischen Trennung und der Entfernung des chiralen Hilfsstoffs, aufgetrennt werden.
- Während irgendwelcher der obigen Synthesesequenzen kann es notwendig und/oder wünschenswert sein, empfindliche oder reaktive Gruppen an irgendwelchen der betroffenen Moleküle zu schützen. Dies kann durch herkömmliche Schutzgruppen erreicht werden, wie z. B. durch diejenigen, die in Protective Groups in Organic Chemistry, Hrsg. J. F. W. McOmie, Plenum Press, 1973, und von T. W. Green & P. G. M. Wuts, Protective Groups in Organic Synthesis, John Wiley & Sons, 1991, beschrieben sind. Die Schutzgruppen können an einem zweckmäßigen nachfolgenden Punkt unter Verwendung von im Fachgebiet bekannten Verfahren entfernt werden.
- Geeignete Verfahren zur Untersuchung des Wirkungsgrades von Verbindungen der vorliegenden Erfindung in Bezug auf γ-Sekretase sind in der
WO 01/70677 - Die Beispiele der vorliegenden Erfindung hatten alle einen ED50-Wert von weniger als 10 μM, besonders bevorzugt von weniger als 1 μM und ganz besonders bevorzugt von weniger als 100 nM in wenigstens einem der oben genannten Tests.
- Die folgenden Beispiele veranschaulichen die vorliegende Erfindung.
- BEISPIELE
- 4-Chlorthiophenol (3,6 g, 0,025 mol) in Dichlormethan (100 ml) wurde mit 2,5-Difluorbenzylbromid (5,17 g, 0,025 mol) und Triethylamin (3,9 ml, 0,028 mol) behandelt, die Reaktion wurde 2 Stunden gerührt, dann mit Dichlormethan (250 ml) verdünnt und mit Wasser (100 ml) und Salzlösung (100 ml) gewaschen. Die abgetrennt organische Schicht wurde getrocknet (MgSO4) und zur Trockene eingedampft. Das Produkt wurde durch Leiten durch einen Kieselgelpfropfen mit Hexan-Ethylacetat-Mischungen als Elutionsmittel gereinigt. 5,12 g. 1H-NMR CDCl3 7,23 (4H, s), 6,69-6,86 (3H, m) und 4,04 (2H, s).
- Dieser Thioether (5,12 mol) wurde in Dichlormethan (100 ml) gelöst und mit m-Chlorperoxybenzoesäure (14,3 g, 0,042 mol (50% Gew./Gew.)) behandelt und 2 Stunden gerührt. Die Reaktion wurde anschließend mit Na2S2O5 (5%ige Lösung, 100 ml) und Salzlösung (50 ml) gewaschen, getrocknet (MgSO4) und zur Trockene eingedampft. Das Sulfonprodukt wurde auf Kieselgel mit Hexan-Ethylacetat-Mischungen als Elutionsmittel gereinigt, 3,6 g. 1H-NMR CDCl3 7,61 (2H, d, J = 8,6 Hz), 7,45 (2H, d, J = 8,6 Hz), 7,13-7,08 (1H, m), 7,05-7,01 (1H, m), 7,05-7,00 (1H, m), 6,99-6,87 (1H, m), und 4,36 (2H, s).
- Hergestellt wie bei Zwischenprodukt 1 unter Verwendung von 4-Trifluormethylthiophenol und erhalten als ein Feststoff. 1H-NMR (360 MHz, CDCl3) δ 7,85-7,83 (2H, m), 7,76-7,74 (2H, m), 7,15-7,10 (1H, m), 7,06-7,0 (1H, m), 6,92-6,86 (1H, m) und 4,46 (2H, s).
- Zwischenprodukt 1 (1 g, 3,31 mmol) und Methylacrylat (0,84 ml, 9,27 mmol) in Tetrahydrofuran (30 ml) wurde tropfenweise mit Kalium-tbutoxid (3,64 ml, 1 M Lösung in Tetrahydrofuran, 3,64 mmol) behandelt. Die Reaktion wurde 2 Stunden gerührt, mit Ethylacetat (100 ml) verdünnt und mit Wasser (50 ml) und Salzlösung (50 ml) gewaschen. Die organische Phase wurde abgetrennt, getrocknet (MgSO4) und zur Trockene eingedampft, und das Produkt wurde auf Kieselgel mit Hexan-Ethylacetat-Mischungen als Elutionsmittel gereinigt. (1,0 g). 1H-NMR CDCl3 12,0 (1H, s), 7,41 (4H, s), 7,06-7,0 (2H, m), 6,87-6,81 (1H, s), 3,81 (3H, s), 3,38 (1H, dd, J = 3,2, 15,8 Hz), 3,02-2,92 (2H, m), 2,52 (1H, dd, J = 5,7, 18,5 Hz), 2,3-2,2 (1H, m) und 2,2-2,1 (1H, m).
- Der Ester aus Herstellung 1 (1,0 g, 2,25 mmol) in Dimethylsulfoxid (10 ml) wurde mit NaCl (0,3 g, 4,96 mmol) und Wasser (0,9 ml, 4,96 mmol) behandelt und 2 Stunden auf 150°C erhitzt. Die abgekühlte Reaktionsmischung wurde mit Ethylacetat (100 ml) verdünnt, mit gesättigtem NH4Cl (100 ml) gewaschen und die organische Phase abgetrennt, getrocknet (MgSO4) und zur Trockene eingedampft. Das Produkt wurde auf Kieselgel mit Hexan-Ethylacetat-Mischungen als Elutionsmittel gereinigt, 0,5 g. 1H-NMR CDCl3 7,43-7,37 (4H, m), 7,22-7,1 (2H, m), 6,97-6,9 (1H, m), 3,05-2,98 (2H, m) und 2,61-2,53 (2H, m).
- Hergestellt durch die Verfahren der Herstellungen 1 und 2 unter Verwendung von Zwischenprodukt 2, um das Produkt als einen Feststoff zu ergeben. (0,3 g) 1H-NMR (360 MHz, CDCl3) δ 7,71-7,69 (2H, d, J = 7,5 Hz), 6,62-6,60 (2H, d, J = 7,4 Hz), 7,22-7,11 (2H, m), 6,95-6,88 (1H, m), 3,02-2,99 (2H, m), 2,63-2,54 (4H, m) und 2,25-2,16 (2H, m).
- Ethyl(diethoxyphosphinyl)acetat (5,16 ml, 26 mmol) wurde tropfenweise zu einer Aufschlämmung von Natriumhydrid (60%ige Dispersion in Mineralöl, 988 mg, 24,7 mmol) in Tetrahydrofuran (60 ml) gegeben und die Mischung 1 Stunde bei Raumtemperatur gerührt. Das Keton aus Herstellung 2 (5 g, 13 mmol) in Tetrahydrofuran (50 ml) wurde tropfenweise innerhalb von 20 Minuten zugegeben und die Mischung 18 Stunden bei Raumtemperatur gerührt. Wasser wurde zugegeben und die Mischung mit Ethylacetat extrahiert. Die vereinten organischen Fraktionen wurden mit Wasser gewaschen, getrocknet (MgSO4) und das Lösungsmittel unter vermindertem Druck abgedampft. Der Rückstand wurde durch Flash-Säulenchromatographie auf Kieselgel mit Isohexan:EtOAc (85:15) als Elutionsmittel gereinigt, um das Produkt als einen weißen Feststoff zu ergeben (5,2 g, 88%). 1H-MMR (400 MHz, CDCl3) δ 7,41-7,36 (4H, m), 7,18-7,13 (1H, m), 7,11-7,05 (1H, m), 6,93-6,86 (1H, m), 5,64 (1H, s), 4,14-4,10 (2H, m), 3,99-3,96 (1H, m), 2,91-2,80 (2H, m), 2,42-2,38 (1H, m), 2,31-2,04 (3H, m), 1,89-1,78 (1H, m), 1,28-1,24 (3H, m).
- Das Keton aus Herstellung 2 (0,1 g, 0,26 mmol) in Methanol (2 ml) wurde mit NaBH4 (0,098 g, 0,26 mmol) behandelt und 1 Stunde gerührt. Die Reaktion wurde mit HCl (1 N, 10 ml) gequencht, mit Ethylacetat (20 ml) verdünnt, dann die organische Phase abgetrennt, getrocknet (MgSO4) und zur Trockene eingedampft. Die cis- und trans-Produkte wurden auf Kieselgel mit Hexan-Ethylacetat-Mischungen als Elutionsmittel gereinigt.

- Das trans-Cyclohexanol aus Herstellung 5 (2,7 g, 6,9 mmol) und Triethylamin (1,45 ml, 10,3 mmol) in Dichlormethan (50 ml) wurden mit Methansulfonylchlorid (0,645 ml, 8,9 mmol) bei –30°C behandelt. Nach 30 Minuten wurde die Mischung mit Wasser (20 ml), 10%iger wässriger Citronensäure (20 ml) und gesättigtem wässrigem Natriumhydrogencarbonat (50 ml) gewaschen, getrocknet (MgSO4) und zur Trockene eingedampft. Der Feststoff wurde mit Ether verrieben, um das Mesylat zu ergeben (2,6 g). 1H-NMR (CDCl3) 7,40-7,37 (4H, m), 7,12-7,07 (2H, m), 6,92-6,83 (1H, m), 4,78-4,65 (1H, m), 2,96 (3H, s), 2,88-2,52 (2H, m), 2,29-2,21 (4H, m) und 1,59-1,47 (2H, m).
- Das trans-Mesylat aus Herstellung 6 (103 mg, 0,22 mmol) wurde in Toluol (20 ml) gelöst und zu einer zuvor azeotrop behandelten Probe von Tetrabutylammoniumcyanid (354 mg, 1,32 mmol) zugegeben und die Mischung innerhalb von 18 Stunden auf 70°C erwärmt und anschließend auf RT abgekühlt. Die Lösung wurde mit Wasser (10 ml) verdünnt und mit Ethylacetat (2 × 50 ml) gewaschen. Die organische Phase wurde mit Salzlösung (10 ml) gewaschen, getrocknet (MgSO4) und eingedampft. Das erhaltene klare Öl wurde durch Säulenchromatographie auf Kieselgel mit 10–20% Ethylacetat in Hexanen als Elutionsmittel gereinigt, um das Cyanid zu ergeben. 1H-NMR (CDCl3) 7,42-7,36 (4H, s), 7,10-7,05 (2H, m), 6.89-6,84 (1H, m), 2,88-2,86 (1H, m), 2,76-2,72 (2H, m), 2,52-2,45 (1H, m), 2,12-2,07 (1H, m) und 1,56-1,49 (1H, m).
- Das Cyanid aus Herstellung 7 (143 mg, 0,36 mmol) wurde in einer Mischung aus Eisessig (10 ml) und konz. HCl (6 ml) gelöst/suspendiert und 15 Stunden auf 110°C erhitzt. Die Mischung wurde abgekühlt, mit Ethylacetat verdünnt und mit Wasser (3 Mal) gewaschen, getrocknet (MgSO4) und zur Trockene eingedampft. Dieser rohe Rückstand (153 mg) wurde durch präparative DC (5% Methanol in Dichlormethan/1% Essigsäure) gereinigt. 1H-NMR (CDCl3) 7,38-7,35 (4H, s), 7,08-7,06 (2H, m), 6,90-6,84 (1H, m), 2,65-2,58 (2H, m), 2,38-2,33 (3H, m) und 1,75-1,49 (4H, m).
- Das Cyanid aus Herstellung 8 (50 mg, 0,12 mmol) wurde in einer Mischung aus Tetrahydrofuran (4,5 ml) und Wasser (0,5 ml) gelöst und bei 20°C gerührt. Die Mischung wurde mit Wasserstoffperoxid (20 ml, 0,6 mmol) und anschließend mit Lithiumhydroxid (6 mg, 0,25 mmol) 2 Stunden behandelt. Wasserstoffperoxid (20 ml, 0,6 mmol) und anschließend Lithiumhydroxid (6 mg, 0,25 mmol) wurden zugegeben und die Mischung bei RT 72 Stunden gerührt. Die Mischung wurde abgekühlt, mit Ethylacetat verdünnt und mit Wasser (2 Mal) und gesättigtem Natriumhydrogensulfit gewaschen, getrocknet (MgSO4) und zur Trockene eingedampft. Dieser rohe Rückstand (51 mg) wurde durch präparative DC (20% Ethylacetat in Hexanen) gereinigt. 1H-NMR (CDCl3) 7,37 (4H, s), 7,10-7,02 (2H, m), 6,90-6,84 (1H, m), 5,57 (2H, br. s), 2,54-2,48 (3H, m), 2,43-2,39 (1H, m), 2,19-2,15 (2H, m) und 1,62-1,50 (3H, m).
- Natriumborhydrid (313 mg, 8,23 mmol) wurde zu einer Mischung aus dem ungesättigten Ester von Herstellung 4 (3,74 g, 8,23 mmol) und Nickel(II)chlorid (2,67 g, 20,6 mmol) in Ethanol (100 ml) gegeben. Die Mischung wurde 20 Minuten bei Raumtemperatur gerührt, dann mit Wasser (100 ml) versetzt. Die Mischung wurde durch HyfloTM filtriert, mit Ethanol und Ethylacetat gewaschen. Das Lösungsmittel wurde unter vermindertem Druck abgedampft und der Rückstand zwischen Ethylacetat und Wasser aufgetrennt. Die organische Schicht wurde gesammelt, getrocknet (MgSO4) und das Lösungsmittel unter vermindertem Druck abgedampft. Der Rückstand wurde durch Flash-Säulenchromatographie auf Kieselgel mit Isohexan:EtOAc (85:15) als Elutionsmittel gereinigt, um das schneller laufende cis-Isomer als ein Öl zu ergeben (1,36 g, 36%), 1H-NMR (400 MHz, CDCl3) δ 7,37-7,30 (4H, m), 7,09-7,00 (2H, m), 6,86-6,79 (1H, m), 4,14 (2H, q, J = 7,1 Hz), 2,47 (2H, d, J = 7,6 Hz), 2,46-2,38 (2H, m), 2,19-2,14 (1H, m), 1,76-1,71 (2H, m), 1,57-1,48 (4H, m), 1,27 (3H, t, J = 7,1 Hz);
und auch das langsamer laufende trans-Isomer als ein Öl (200 mg, 5,3%). - Lithiumhydroxid (350 mg, 14,57 mmol) wurde zu einer Lösung des cis-Esters aus Beispiel 1 (1,33 g, 2,91 mmol) in Ethanol (40 ml) zugegeben. Die Mischung wurde entgast und bei Raumtemperatur unter Stickstoffgas 5 Stunden gerührt. Die Mischung wurde in wässrige Salzsäure (1 M) gegossen und mit Ethylacetat extrahiert. Der organische Extrakt wurde getrocknet (MgSO4) und das Lösungsmittel unter vermindertem Druck abgedampft, um einen weißen Feststoff zu ergeben, der anschließend aus IPA kristallisiert wurde, um das Produkt als einen weißen Feststoff zu ergeben (950 mg, 76%). 1H-NMR (400 MHz, CD3OD) δ 7,51-7,49 (2H, m), 7,40-7,37 (2H, m), 7,19-7,10 (2H, m), 7,00-6,94 (1H, m), 2,51-2,35 (6H, m), 2,13-2,10 (1H, m), 1,78-1,74 (2H, m), 1,57-1,50 (2H, m).
- Die Säure von Beispiel 2 (50 mg, 0,117 mmol), Morpholin (30 μl, 0,351 mmol), 1-Hydroxybenzotriazol (24 mg, 0,176 mmol) und Triethylamin (65 μl, 0,468 mmol) wurden in Tetrahydrofuran bei Raumtemperatur unter Stickstoffgas 10 Minuten gerührt. 1-(3-Dimethylaminopropyl)-3-ethylcarbodiimid-Hydrochlorid (45 mg, 0,234 mmol) wurde zu der Mischung zugegeben und 24 Stunden gerührt. Die Mischung wurde in wässriges Natriumhydroxid (1 M) gegossen und mit Ethylacetat extrahiert. Der organische Extrakt wurde getrocknet (MgSO4) und das Lösungsmittel unter vermindertem Druck abgedampft. Der Rückstand wurde durch Flash-Säulenchromatographie auf Kieselgel mit 5 bis 10% Methanol in Dichlormethan als Elutionsmittel gereinigt, um das Produkt als einen weißen Schaum (50 mg, 86%) zu ergeben. 1H-NMR (400 MHz, CD3OD) δ 7,50 (2H, d, J 8,6 Hz), 7,37 (2H, d, J 8,6 Hz), 7,19-7,09 (2H, m), 7,00-6,93 (1H, m), 3,69-3,63 (4H, m), 3,59-3,56 (4H, m), 2,55 (2H, d, J 7,4 Hz), 2,47-2,39 (4H, m), 2,16-2,07 (1H, m), 1,78-1,74 (2H, m), 1,58-1,51 (2H, m). m/z (ES+) (M+1) 498 + 500.
- Beispiele 4–15
- Die folgenden Verbindungen wurden gemäß dem Verfahren von Beispiel 3 hergestellt, wobei das passende Amin anstelle von Morpholin verwendet wurde.
- Diese Beispiele wurden durch das folgende Verfahren unter Verwendung des passenden Amins in Form der freien Base oder des passenden Aminsalzes mit vorhergehender Neutralisation hergestellt.
- Zu einer gerührten Suspension von cis-4-(4-Chlorbenzolsulfonyl)-4-(2,5-difluorphenyl)cyclohexanessigsäure (Beispiel 2, 0,15 g, 0,35 mmol) in Dichlormethan (5 ml) wurden Oxalylchlorid (0,05 ml, 0,57 mmol) und Dimethylformamid (1 Tropfen) zugegeben. Nach 30 Minuten wurde die Lösung zu einem kleinen Volumen eingedampft und zu einer Lösung des Rückstandes in Dichlormethan (5 ml) das erwünschte Amin (1,75 mmol) zugegeben. Nach 20-minütigem Rühren der Lösung wurde das Lösungsmittel im Vakuum entfernt und der Rückstand durch Chromatographie auf Kieselgel mit steigenden Konzentrationen von Ethylacetat in Isohexan (25%, 50%) als Elutionsmittel gereinigt. Die produkthaltigen Fraktionen wurden eingedampft, um das Produktamid zu ergeben. Die chromatographische Reinigung erfolgte auf Kieselgel unter Verwendung geeigneter Konzentrationen an Ethylacetat in Isohexan, Ethylacetat oder Methanol in Ethylacetat, wo passend.
- Zu einer Lösung des cis-Amids aus Herstellung 9 (46 mg) und von Pyridin (0,053 ml) in Tetrahydrofuran (1 ml) wurde Trifluoressigsäureanhydrid (0,056 ml) zugegeben. Die Lösung wurde bei Raumtemperatur 2 Stunden gerührt, wonach 0,5 M HCl (wässrig) und Ethylacetat zugegeben wurden. Die organische Phase wurde getrocknet (MgSO4), zu einem kleinen Volumen eingedampft und durch Chromatographie auf Kieselgel mit Isohexan:Ethylacetat (5:1) als Elutionsmittel gereinigt, um das erwünschte Produkt als einen farblosen Feststoff zu ergeben. 1H-NMR (360 MHz, CDCl3) δ 1,61-1,70 (2H, m), 1,86-1,94 (2H, m), 2,03-2,10 (1H, m), 2,42-2,45 (4H, m), 2,51 (2H, d, J 8,0 Hz), 6,8 (1H, m), 7,02-7,09 (2H, m), 7,30 (2H, d, J 8,6 Hz), 7,36 (2H, d, J 8,7 Hz).
- Die Säure aus Herstellung 8 (153 mg) wurde in trockenem THF (10 ml) gelöst und unter Stickstoff auf 0°C abgekühlt. Triethylamin (61 μl, 0,43 mmol) und Isobutylchlorformiat (57 μl, 0,43 mmol) wurden zugegeben und die Mischung eine Stunde bei 0°C gerührt. Der Niederschlag, der sich gebildet hatte, wurde durch Filtration entfernt und mit weiteren 5 ml trockenem THF gewaschen. Die vereinten THF-Schichten wurden wieder auf 0°C abgekühlt und mit Natriumborhydrid (70 mg, 1,84 mmol) als eine Lösung in Wasser (2 ml) versetzt, wobei die Lösung aufschäumte. Nach 30-minütigem Rühren bei 0°C wurde die Reaktion mit Ethylacetat verdünnt, mit Ammoniumchloridlösung, Natriumhydrogencarbonatlösung und Salzlösung gewaschen, dann getrocknet (MgSO4) und zur Trockene eingedampft. Der Rückstand wurde durch Säulenchromatographie mit Ethylacetat:Hexan (1:3) als Elutionsmittel gereinigt, um den erwünschten Alkohol (75 mg) zu ergeben. 1H-NMR (CDCl3) 7,39-7,31 (4H, m), 7,10-7,01 (2H, m), 6,88-6,81 (1H, m), 3,71 (2H, d, J = 7,5 Hz), 2,46-2,32 (4H, m), 1,90-1,85 (2H, m), 1,78-1,74 (1H, m) und 1,54-1,44 (2H, m). m/z = 423 [MNa]+.
- Schritt (1)
- Eine Lösung der Säure aus Beispiel 2 (1 g) in DCM (50 ml) und Ethylacetat (30 ml) wurde mit Pentafluorphenol (1,5 Äquiv.) und DCC (1,5 Äquiv.) versetzt und 1 Stunde bei Raumtemperatur gerührt. Die Reaktionsmischung wurde im Vakuum eingedampft, in Ethylacetat aufgenommen und filtriert. Das Filtrat wurde im Vakuum eingedampft, um den Pentafluorphenolester in ausreichender Reinheit zu ergeben, um ihn bei den nachfolgenden Reaktionen zu verwenden.
- Schritt (2)
- Zu dem in Schritt (1) hergestellten Pentafluorphenolester (155 mg, 0,25 mmol), gelöst in DMF (3 ml) und unter Stickstoff, wurden Glycinmethylester-Hydrochlorid (125 mg, 1,0 mmol) und Triethylamin (0,15 ml) zugegeben. Nach 2 Stunden wurde die Reaktion mit Wasser verdünnt, mit Ethylacetat (3 Mal) extrahiert, mit Wasser und Salzlösung gewaschen, getrocknet (MgSO4), filtriert und eingedampft. Die Reinigung durch Flash-Säulenchromatographie (1:1 iHexan/Ethylacetat bis 9:1 Ethylacetat/Methanol) ergab einen weißen Feststoff (55 mg). 1H-NMR (CDCl3) 1,08-1,16 (1H, m), 1,30-1,37 (1H, m), 1,67-1,71 (1H, m), 1,75-1,79 (2H, m), 1,91-1,95 (1H, m), 2,20-2,26 (1H, m), 2,41 (4H, d, J = 7,8 Hz), 3,77 (3H, s), 4,05 (2H, d, J = 5,1 Hz), 6,19 (1H, br.), 6,79-6,85 (1H, m), 7,00-7,07 (2H, m), 7,30-7,37 (4H, m).
- Der in Beispiel 35 hergestellte Glycinester (50 mg, 0,1 mmol) in einem verschlossenen Rohr und gelöst in einer 2 M Ammoniak-in-Methanol-Lösung (3 ml) wurde 3 Stunden auf 50°C erwärmt. Nach dem Abkühlen auf Raumtemperatur wurde die Reaktionsmischung eingeengt und durch Verreiben mit Ether gereinigt, um einen weißen Feststoff (28 mg) zu ergeben. MS (EI+): 485 (MH+).
- Der Alkohol aus Beispiel 34 (4 g, 10 mmol) wurde in Dichlormethan (280 ml) gelöst und mit Dess-Martin-Periodinan (4,66 g, 11 mmol) versetzt und die Mischung 45 Minuten gerührt, bevor gesättigtes wässriges Natriumhydrogensulfit (100 ml) zugegeben wurde, und nach 5 Minuten wurde die Mischung aufgetrennt und die organische Phase mit gesättigtem wässrigem Natriumhydrogencarbonat (100 ml) gewaschen, getrocknet (MgSO4) und zur Trockene eingedampft. Der rohe Rückstand (4 g) wurde in trockenem Dichlormethan (100 ml) gelöst und mit Methyltriphenylphosphinoacetat (4,7 g, 14 mmol) behandelt, wobei 16 Stunden bei RT gerührt wurde. Das Lösungsmittel wurde abgedampft und der Rückstand durch Säulenchromatographie auf Kieselgel mit 10-20% Ethylacetat in Hexanen gereinigt, um das Produkt zu ergeben. 1H-NMR (CDCl3) 7,37-7,36 (4H, m), 7,10-7,02 (3H, m), 6,87-6,83 (1H, m), 5,91 (1H, d, J = 16 Hz), 3,77 (3H, s), 2,55-2,45 (3H, m), 2,40-2,38 (2H, m), 1,95-1,90 (2H, m) und 1,65-1,52 (2H, m).
- Das Alken aus Beispiel 37 (3,6 g, 9 mmol) wurde in Ethylacetat (350 ml) gelöst. Der Kolben wurde entgast und anschließend mit 10% Palladium auf Kohle (400 mg) versetzt und die Mischung unter einer Wasserstoffatmosphäre 45 Minuten gerührt. Die Lösung wurde durch CeliteTM filtriert und eingedampft. Das erhaltene klare Öl wurde durch präparative DC mit 5% Ethylacetat in Hexanen als Elutionsmittel gereinigt. Das erhaltene Öl wurde anschließend weiter durch Säulenchromatographie auf Kieselgel mit 5-10% Ethylacetat in Hexan als Elutionsmittel gereinigt, um das Produkt zu ergeben. 1H-NMR (CDCl3) 7,37-7,34 (4H, m), 7,08-7,00 (2H, m), 6,85-6,81 (1H, m), 3,67 (3H, s), 2,45-2,39 (4H, m), 2,33 (2H, t, J = 8,4 Hz), 1,81 (2H, q, J = 8,4 Hz), 1,72-1,68 (2H, m) und 1,60-1,43 (3H, m).
- Der Ester aus Beispiel 38 (104 mg, 0,23 mmol) wurde in einer Mischung aus Ethanol (10 ml) und Wasser (3 ml) gelöst und bei 20°C gerührt. Der Kolben wurde entgast und anschließend mit Lithiumhydroxid (27 mg, 1,15 mmol) versetzt. Die Mischung wurde 3 Stunden bei Raumtemperatur gerührt. Anschließend wurde 1 N Salzsäure zugegeben und die Mischung mit Ethylacetat (2 × 50 ml) gewaschen. Die organische Phase wurde mit Salzlösung (50 ml) gewaschen, getrocknet (MgSO4) und eingedampft. Das erhaltene Öl wurde anschließend weiter durch präparative DC mit Ethylacetat als Elutionsmittel gereinigt, um die Säure zu ergeben. 1H-NMR (CDCl3) 7,37-7,30 (4H, m), 7,09-6,99 (2H, m), 6,85-6,79 (1H, m), 2,42-2,36 (6H, m), 1,85-1,79 (2H, m), 1,73-1,69 (2H, m), 1,63-1,58 (1H, m) und 1,53-1,45 (2H, m).
- Die Säure aus Beispiel 39 (52 mg, 0,118 mmol) in Dichlormethan (2 ml) wurde mit Oxalylchlorid (88 μl, 2 M Lösung in Dichlormethan, 0,176 mmol) behandelt. Ein Tropfen N,N-Dimethylformamid wurde zugegeben, und man ließ die Lösung 2 Stunden rühren. Nach dieser Zeit wurde das Lösungsmittel im Vakuum entfernt und der Rückstand wieder in Dichlormethan (1 ml) gelöst. Diese Lösung wurde in methanolischen Ammoniak (2 M, 2 ml) getropft. Die Reaktion wurde im Vakuum eingedampft und der Rückstand auf Kieselgel chromatographiert, wobei mit 80% Ethylacetat in Hexanen eluiert wurde. Das resultierende Material wurde durch präparative DC mit 100% Ethylacetat als Elutionsmittel weiter gereinigt, gefolgt von der Umkristallisation aus heißem Hexan, um das Produkt zu ergeben (7,4 mg, 14%). 1H-NMR (360 MHz, CDCl3), 1,45-1,53 (2H, m), 1,57-1,65 (1H, br.), 1,70-1,75 (2H, m), 1,78-1,84 (2H, m), 2,32 (2H, t, J = 15,3 Hz), 2,38-2,44 (4H, br.), 2,95 (3H, s), 3,02 (3H, s), 6,79-6,86 (1H, m), 7,00-7,09 (2H, m), 7,31-7,37 (4H, m); MS. (ES+), 470 (M++1), 294 (M+175).
- Der cis-Ester aus Beispiel 1 (669 mg, 1,467 mmol) in Tetrahydrofuran (14 ml) wurde auf –78°C abgekühlt, mit Natriumbis(trimethylsilyl)amid (2,20 ml, 1 M Lösung in Tetrahydrofuran, 2,20 mmol) behandelt und unter Erwärmen auf Raumtemperatur 2 Stunden gerührt. Anschließend wurde Methyliodid (457 μl, 7,36 mmol) zu der Mischung bei –20°C zugegeben und das Rühren fortgesetzt, wobei erneut innerhalb von 2 Stunden auf Raumtemperatur erwärmt wurde. Die Reaktion wurde mit Eisessig (132 μl, 2,20 mmol) gequencht, mit Ammoniumchlorid (50%ig, wässr., 80 ml) verdünnt und mit Ethylacetat (3 × 100 ml) extrahiert. Die vereinten organischen Phasen wurden anschließend mit Salzlösung (gesätt., 200 ml) gewaschen, getrocknet (MgSO4) und im Vakuum eingedampft, um Rohprodukt (670 mg) zu ergeben. Dieses Material wurde auf Kieselgel mit 8% Ethylacetat in Hexanen als Elutionsmittel chromatographiert, um das Produkt zu ergeben (272 mg, 40%). 1H-NMR (400 MHz, CDCl3), 1,16 (3H, d, J = 6,9 Hz), 1,28 (3H, t, J = 7,1 Hz), 1,45-1,51 (2H, m), 1,71-1,77 (2H, m), 1,89-1,94 (1H, m), 2,28-2,48 (3H, br), 2,54-2,60 (1H, br.), 2,70-2,74 (1H, m), 4,09-4,18 (2H, m), 6,77-6,84 (1H, m), 6,99-7,08 (2H, m), 7,26-7,36 (4H, m).

- Hergestellt aus der Säure von Beispiel 42 durch das Verfahren von Beispiel 35 unter Verwendung von Ammoniak im zweiten Schritt. MS MH+ 462(463).
- Hergestellt aus dem Keton von Herstellung 3 durch Nacharbeiten der Verfahren der Herstellungen 5–8 und der Beispiele 34, 37, 38 und 39.
1H-NMR (360 MHz, CDCl3) δ 10,1 (1H, m), 7,64 (2H, d, J = 8,3 Hz), 7,53 (2H, d, J = 8,3 Hz), 7,09-7,00 (2H, m), 6,83-6,76 (1H, m), 2,50-2,37 (6H, m), 1,85-1,81 (2H, q, J = 7,4 Hz), 1,75-1,70 (2H, m), 1,63-1,59) (1H, m), 1,55-1,45 (2H, m).
MS(EI+) 477 (MH+).
Claims (14)
- Eine Verbindung der Formel I: wobei: m 0 oder 1 ist, Z CN, OR2a, CO2R2a oder CON(R2a)2 bedeutet, R1b H, C1-4-Alkyl oder OH bedeutet, R1c H oder C1-4-Alkyl bedeutet, Ar1 Phenyl oder Pyridyl bedeutet, wobei jedes davon 0–3 Substituenten trägt, unabhängig ausgewählt aus Halogen, CN, NO2, CF3, OH, OCF3, C1-4-Alkoxy oder C1-4-Alkyl, welches gegebenenfalls einen Substituenten trägt, ausgewählt aus Halogen, CN, NO2, CF3, OH und C1-4-Alkoxy, Ar2 Phenyl bedeutet, das in den 2- und 5-Positionen mit Halogen substituiert ist, R2a H, C1-6-Alkyl, C3-6-Cycloalkyl, C3-6-Cycloalkyl-C1-6-alkyl, C2-6-Akenyl bedeutet, wobei jedes davon gegebenenfalls einen Substituenten trägt, ausgewählt aus Halogen, CN, NO2, CF3, OR2b, CO2R2b, N(R2b)2, CON(R2b)2, Ar und COAr, oder R2a Ar bedeutet, oder zwei R2a-Gruppen zusammen mit einem Stickstoffatom, an das sie gemeinsam gebunden sind, eine N-Heterocyclylgruppe vervollständigen können, die 0–4 Substituenten trägt, unabhängig ausgewählt aus =O, =S, Halogen, C1-4-Alkyl, CN, NO2, OF3, OH, C1-4-Alkoxy, C1-4-Alkoxycarbonyl, CO2H, Amino, C1-4-Alkylamino, Di(C1-4-alkyl)amino, Carbamoyl, Ar und COAr, R2b H, C1-6-Alkyl, C3-6-Cycloalkyl, C3-6-Cycloalkyl-C1-6-alkyl, C2-6-Alkenyl bedeutet, wobei jedes davon gegebenenfalls einen Substituenten trägt, ausgewählt aus Halogen, CN, NO2, CF3, OH, C1-4-Alkoxy, C1-4-Alkoxycarbonyl, CO2H, Amino, C1-4-Alkylamino, Di(C1-4-alkyl)amino, Carbamoyl, Ar und COAr, oder R2b Ar bedeutet, oder zwei R2b-Gruppen zusammen mit einem Stickstoffatom, an das sie gemeinsam gebunden sind, eine N-Heterocyclylgruppe vervollständigen können, die 0–4 Substituenten trägt, unabhängig ausgewählt aus =O, =S, Halogen, C1-4-Alkyl, CN, NO2, OF3, OH, C1-4-Alkoxy, C1-4-Alkoxycarbonyl, CO2H, Amino, C1-4-Alkylamino, Di(C1-4-alkyl)amino, Carbamoyl, Ar und COAr, Ar Phenyl oder Heteroaryl bedeutet, das 0–3 Substituenten trägt, ausgewählt aus Halogen, C1-4-Alkyl, CN, NO2, CF3, OH, C1-4-Alkoxy, C1-4-Alkoxycarbonyl, Amino, C1-4-Alkylamino, Di(C1-4-alkyl)amino, Carbamoyl, C1-4-Alkylcarbamoyl und Di(C1-4-alkyl)carbamoyl, oder ein pharmazeutisch annehmbares Salz davon.
- Eine Verbindung gemäß Anspruch 1, wobei Ar1 ausgewählt ist aus Phenylgruppen, substituiert in der 4-Position mit Halogen, Methyl oder Trifluormethyl, und Phenylgruppen, substituiert in den 3- und 4-Positionen durch Halogen.
- Eine Verbindung gemäß Anspruch 1 oder Anspruch 2, wobei Ar1 4-Chlorphenyl oder 4-Trifluormethylphenyl ist und Ar2 2,5-Difluorphenyl ist.
- Eine Verbindung gemäß irgendeinem der Ansprüche 1–3, wobei Z CO2R2a bedeutet und R2a H oder C1-6-Alkyl bedeutet.
- Die Verbindung gemäß Anspruch 1, wobei m 1 ist, Ar1 4-Chlorphenyl bedeutet, Ar2 2,5-Difluorphenyl bedeutet, R1b und R1c beide H bedeuten und Z CO2H bedeutet, oder ein pharmazeutisch annehmbares Salz davon.
- Die Verbindung nach Anspruch 5 in Form ihres Natriumsalzes.
- Die Verbindung gemäß Anspruch 1, wobei m 1 ist, Ar1 4-Trifluormethylphenyl bedeutet, Ar2 2,5-Difluorphenyl bedeutet, R1b und R1c beide H bedeuten und Z CO2H bedeutet, oder ein pharmazeutisch annehmbares Salz davon.
- Die Verbindung gemäß Anspruch 1, wobei m 0 ist, Ar1 4-Chlorphenyl bedeutet, Ar2 2,5-Difluorphenyl bedeutet, R1c H bedeutet und Z CONH2 bedeutet, oder ein pharmazeutisch annehmbares Salz davon.
- Die Verbindung gemäß Anspruch 1, wobei m 0 ist, Ar1 4-Trifluormethylphenyl bedeutet, Ar2 2,5-Difluorphenyl bedeutet, R1c H bedeutet und Z CONH2 bedeutet, oder ein pharmazeutisch annehmbares Salz davon.
- Die Verbindung gemäß Anspruch 1, wobei m 0 ist, Ar1 4-Chlorphenyl bedeutet, Ar2 2,5-Difluorphenyl bedeutet, R1c H bedeutet und Z CONHCH2CH3 bedeutet, oder ein pharmazeutisch annehmbares Salz davon.
- Die Verbindung gemäß Anspruch 1, wobei m 0 ist, Ar1 4-Chlorphenyl bedeutet, Ar2 2,5-Difluorphenyl bedeutet, R1c H bedeutet und Z CN bedeutet, oder ein pharmazeutisch annehmbares Salz davon.
- Eine pharmazeutische Zusammensetzung, die eine Verbindung gemäß irgendeinem der Ansprüche 1–11 oder ein pharmazeutisch annehmbares Salz davon und einen pharmazeutisch annehmbaren Träger enthält.
- Eine Verbindung gemäß irgendeinem der Ansprüche 1–11 oder ein pharmazeutisch annehmbares Salz davon zur Verwendung bei einem Verfahren zur Behandlung des menschlichen Körpers.
- Die Verwendung einer Verbindung gemäß irgendeinem der Ansprüche 1–11 oder eines pharmazeutisch annehmbaren Salzes davon bei der Herstellung eines Medikaments zur Behandlung oder Prävention von Alzheimer-Krankheit.
