Description
- Die vorliegende Erfindung betrifft eine neue Verbindung, die gegenüber Prolylendopeptidase (EC 3.4.21.26) enzyminhibierende Aktivität aufweist. Die Erfindung betrifft auch ein Verfahren zur chemischen Synthese einer solchen neuen Verbindung, ebenso wie ihre Verwendung als Inhibitor der Prolylendopeptidase-Aktivität, und ein Medikament, insbesondere ein antiamnesisches Mittel, das sie als aktiven Inhaltsstoff enthält.
- Prolylendopeptidase inaktiviert bekannterweise Neurotransmitter wie die Substanz P, das Thyrotropin freisetzende Hormon (TRH) und Neurotensin oder Vasopressin, das spekulativ mit dem Gedächtnis in Verbindung gebracht wird. Tsuru und Yoshimoto vom Department of Pharmaceutical Sciences, Nagasaki University, fanden heraus, daß Verbindungen, die zur Inhibierung der Prolylendopeptidase- Aktivität in der Lage waren, wirksam waren zur Verhinderung von experimenteller Amnesie, die in Ratten durch Scopolamin verursacht worden war. Auf der Basis dieser Entdeckung schlugen sie die potentielle Verwendung von Prolylendopeptidase-Aktivitätsinhibitoren als antiamnesische Mittel vor (T. Yoshimoto und D. Tsuru, Agr. Biol. Chem. 42, 2417, 1978).
- EP-A-0172458 offenbart ein antiamnesisches Mittel, das als aktiven Bestandteil ein N-Acylpyrrolidinderivat der Formel
- enthält, worin n eine Zahl von 1 bis 4 darstellt; und R eine niedere Alkylestergruppe, -CH&sub2;OH oder eine Aldehydgruppe ist.
- Motiviert durch den Bericht von Tsuru und Yoshimoto machten die gegenwärtigen Erfinder verschiedene Anstrengungen, neue Verbindungen zu finden, die starke antiamnesische Aktivität aufweisen und dennoch einen hinreichend niedrigen Toxizitätsgrad haben. Als Ergebnis fanden die Erfinder, daß N-Acylpyrrolidinderivate mit Anti-Prolylendopeptidase- Aktivität der Formel (I), wie unten gezeigt, ausgezeichnete Wirkung gegen Amnesie aufwiesen. Die vorliegende Erfindung wurde auf der Grundlage dieser Befunde erreicht.
- Die N-Acylpyrrolidinderivate der vorliegenden Erfindung werden durch die allgemeine Formel (I) dargestellt:
- worin R¹ ein Wasserstoffatom, eine niedere Alkylgruppe oder eine Phenylgruppe ist,
- R³ eine Formylgruppe ist,
- wenn R¹ ein Wasserstoffatom ist, R² eine substituierte oder unsubstituierte Phenyloxygruppe oder Phenylcarbonylgruppe oder eine substituierte Phenylgruppe ist, und wenn R¹ eine niedere Alkylgruppe oder Phenylgruppe ist, R² eine substituierte oder unsubstituierte Phenylgruppe, eine niedere Alkylgruppe, eine Aralkylgruppe von 7 bis 10 Kohlenstoffatomen oder eine Hydroxygruppe ist; oder R¹ und R² zusammen eine Benzylidengruppe bilden, welche unsubstituiert oder substituiert an ihrer Aromateneinheit ist, und
- n eine Ganzzahl von 0 bis 5 ist.
- R¹ und R² können als niedere Alkylgruppen z. B. geradkettige oder verzweigte Alkylgruppe von 1 bis 4 Kohlenstoffatomen wie etwa Methyl und Isopropyl usw. sein.
- Wenn R² eine substituierte Phenyloxygruppe oder eine substituierte Phenylgruppe ist, kann der Substituent z. B. ein Halogenatom oder eine niedere Alkoxygruppe von 1 bis 4 Kohlenstoffatomen sein.
- Wenn R² besagte Aralkylgruppe ist oder R² eine Benzylidengruppe zusammen mit R¹ bildet, können ihre aromatischen Ringe durch ein Halogenatom oder eine niedere Alkoxygruppe von 1 bis 4 Kohlenstoffatomen substituiert sein.
- Die Verbindungen der Formel (I) weichen stark von den bekannten 2-Oxopyrrolidin enthaltenden antiamnesischen Mitteln wie Piracetam oder Aniracetam ab, und zwar dadurch, daß sie einen Prolinrest oder ein Derivat davon in ihrer Struktur enthalten. Wegen dieses Merkmales zeigen die Verbindungen der Formel (I) extrem niedrigere Toxizitätsgrade in Organismen.
- Die Verbindung der Formel (I) der vorliegenden Erfindung können durch die folgenden Verfahrensschritte synthesisiert werden:
- (i) Zwischenverbindungen der Formel (Ia)
- worin R¹, R² und n die oben angegebenen Bedeutungen haben und R&sup4; ein Wasserstoffatom oder eine niedere Alkylgruppe ist, werden durch eine Kondensationsreaktion einer Carbonsäure der Formel (II):
- worin R¹, R² und n die selben Bedeutungen wie oben angegeben haben, oder eines reaktiven Derivats davon mit Prolin, Prolinalkylester oder eines Salzes davon der Formel (III)
- worin R&sup4; die oben angegebenen Bedeutungen hat, erhalten.
- Wenn das Ausgangsmaterial eine Carbonsäure der allgemeinen Formel (II) ist, ist bei der Kondensationsreaktion die Verwendung eines Kondensationsmittels, wie es gewöhnlich bei der Peptidsynthese verwendet wird, wie etwa eines N- Ethyl-N'N'-dimethylaminopropylcarbodiimids vorteilhaft. Beispiele der reaktiven Derivate der Caronbsäure der Formel (II) sind Säurechloride, Säureanhydride und aktive Ester der Carbonsäure. Wenn das Ausgangsmaterial aus diesen reaktiven Derivaten ausgewählt wird, kann die Kondensation unter Verwendung einer Base wie Trialkylamin als Kondensierungsmittel ausgeführt werden. Einige reaktive Derivate der Carbonsäuren können ohne Zugabe eines Kondensierungsmittels reagieren.
- (ii) Die Verbindungen der Formel (Ia) können mit einem Reduktionsmittel behandelt werden, woraus die Zwischenverbindungen der allgemeinen Formel (Ib) entstehen:
- worin R¹, R² und n die oben angegebenen Bedeutungen haben unter der Voraussetzung, daß R² nicht Phenylcarbonyl ist.
- Jedes beliebige Reduktionsmittel kann eingesetzt werden, wenn es nicht Amide reduziert. Das vorgezogene Reduktionsmittel ist Natriumborhydrid. Die Reduktion wird in einem etherischen oder alkoholischen Lösungsmittel in einem Temperaturbereich von Raumtemperatur bis unter 100 ºC und vorzugsweise zwischen 40 und 70 ºC durchgeführt.
- In einem besonders vorgezogenen Verfahren wird eine Verbindung der allgemeinen Formel (Ia) zusammen mit einem Überschuß von Natriumborhydrid in t-Butanol oder Tetrahydrofuran gelöst, gefolgt von Zutropfen von Methanol unter Erwärmung. Das Verhältnis von t-Butanol oder Tetrahydrofuran zu Methanol ist vorzugsweise ungefähr 5:1 Volumenteile.
- (iii) Die so erhaltenen Verbindungen der Formel (Ib) werden dann oxidiert, woraus Verbindungen der vorliegenden Erfindung (I) entstehen:
- worin R¹, R² und n die oben angegebenen Bedeutungen haben, unter der Voraussetzung, daß R² keine Hydroxygruppe ist. (iv) Alternativ dazu werden die Verbindungen der vorliegenden Erfindung, ausgedrückt durch die allgemeine Formel (I), auch erhalten, indem man Prolinalkylester- Derivate der obigen Formel (III) durch ein 2-Formylpyrrolidinderivat, dessen Formylgruppe geschützt ist, und das durch die allgemeine Formel (IV) dargestellt ist, ersetzt:
- worin Y eine Schutzgruppe des Aldehyds ist, und dieses Derivat (IV) mit der Carbonsäure der allgemeinen Formel (II) kondensiert.
- Die in der Reaktionsmischung gebildete erfindungsgemäße Verbindung kann durch eine geeignete konventionelle Reinigungsmethode wie Säurenchromatographie, preparative Dünnschichtchromatographie oder Destillation isoliert werden.
- Die Verbindungen der vorliegenden Erfindung üben eine inhibitorische Wirkung gegen die Prolylendopeptidase aus. Zum Beispiel wurde die inhibitorische Wirkung der Verbindungen gegen dieses Enzym überprüft, indem man Z- Glycyl-prolyl-β-naphthylamid als Substrat verwendete. Wie später im Experimentalbeispiel gezeigt, zeigte die Verbindung (I) inhibitorische Aktivität gegenüber Prolylendopeptidase, jedoch keine gegenüber Papain, Bromelain, Trypsin, Chymotrypsin und Thermolysin.
- Diese inhibitorische Wirkung ermöglicht die Anwendung der vorliegenden Erfindung zum Zwecke der Verbesserung von Gehirndysfunktionen unter Einschluß der Anwendung bei Amnesie.
- Die vorliegende Erfindung wird im folgenden im größeren Detail durch Beispiele beschrieben. BEISPIEL 1 Synthese von N-(3-phenoxy)propionyl-prolinal (Verbindung Nr. 21):
- Eine Mischung von N-(3-phenoxy)propionyl-prolinol (1,4 g) und Triethylamin (1,7g) wurde in wasserfreiem Dimethylsulfoxid (16 ml) gelöst, und zu der gerührten Lösung wurde eine Lösung (16 ml) von Schwefeltrioxidpyridinkomplex (2,6 g) in Dimethylsulfoxid zugegeben. Hach zehn minütigen Rührung der Mischung bei Raumtemperatur wurde die Reaktionslösung in Eiswasser (150 ml) eingegossen und mit Ethylacetat extrahiert. Der Extrakt wurde sukzessive mit 10 %-iger wäßriger Zitronensäure, Wasser, gesättigtem wäßrigem Natriumhydrogencarbonat und Salzlösung gewaschen und über wasserfreiem Magnesiumsulfat getrocknet. Hach Abdestillieren des Lösungsmittels unter Vakuum wurde das resultierende Rohprodukt durch Mitteldruck Flüssigsäulenchromatographie auf Silicagel (Lösungsmittel Chloroform) gereinigt, woraus das Endprodukt als Öl (1,2 g) erhalten wurde.
- Die analytischen Daten für die im Beispiel erhaltenen Verbindungen sind in Tabelle 1 aufgelistet. Die anderen in Tabelle 1 aufgelisteten Verbindungen wurden ebenfalls durch Verfahren, die ähnlich zu obigem Beispiel waren, aus den entsprechenden Startverbindungen erhalten. - Das Verfahren von Yoshimoto und Tsuru (T. Yoshimoto und D. Tsuru, Agr. Biol. Chem. 42, 2417, 1978) wurde verwendet, um die Anti-Prolylendopeptidase-Aktivitäten der Verbindungen der vorliegenden Erfindung zu messen. Eine Mischung von 0,0025 M Z-glycyl-prolin-β-naphthylamid (0,25 ml), 0,1M Phosphatpuffer (pH 7,0; 0,99 ml) und eine Lösung einer speziellen Anti-Prolylendopeptidase-Verbindung (0,01 ml) wurde in einem Testrohr bei 37 ºC drei Minuten inkubiert. Danach wurden 0,1 ml einer Lösung von Prolylendopeptidase (0,2 U/ml) zugegeben und die Mischung auf 35ºC während 10 Minuten erwärmt. Hach der Reaktion wurden 2,0 ml Triton X- 100 in 1M Acetatpuffer (pH 4,0) zur Reaktionsmischung zugegeben, bis die Endkonzentration des Tensides 10 % betrug. Die Mischung wurde bei Raumtemperatur 15 Minuten stehen gelassen und die Absorption (a) bei 410 nm wurde gemessen.
- Eine Blindprobe wurde hergestellt unter Verwendung des Puffers anstelle der Anti-Prolylendopeptidase-Verbindung und ihre Absorption (b) wurde ebenfalls gemessen. Der Prozentsatz der Inhibierung der Prolylendopeptidase wurde durch die Formel ((b-a)/b) · 100 berechnet, und die Menge einer spezifischen Verbindung, um 50 %-ige Inhibierung (IC&sub5;&sub0;) zu erreichen, wurde bestimmt. - Messung des Präventiveffekts gegen experimentelle Amnesie, die in Ratten durch Scopolamin (intraperitoneale Verabreichung) verursacht wurde.
- Mehrere der Anti-Prolylendopeptidase-Verbindungen der vorliegenden Erfindung wurden auf ihre Fähigkeit, die Inhibierung des Langzeitgedächtnisses durch Scopolamin zu verhindern, überprüft. Physiologische Kochsalzlösungen, die ausgewählte Verbindungen der vorliegenden Erfindung in einer Menge von 1 mg/kg enthielten, wurden intraperitoneal an Wister männliche Ratten (100-120 g) verabreicht. Eine Stunde nach der Verabreichung wurden auf die Ratten Elektroschocks ausgeübt, so daß sie passives Vermeiden erlernen konnten. Unmittelbar danach wurde Scopolamin intraperitoneal jeder Ratte in einer Menge von 3 mg pro kg Körpergewicht verabreicht.
- Das Ergebnis des Tests wurde 24 Stunden und 48 Stunden nach der Verabreichung von Scopolamin ermittelt. Die Anzahl amnesischer Ratten und gesunder Ratten wurde sowohl für die Kontrollgruppe (Ratten, welchen die Testverbindungen nicht gegeben wurden, sondern welchen nur Scopolamin und physiologische Kochsalzlösung intraperitoneal verabreicht wurde) und die behandelte Gruppe (Ratten, welchen sowohl die Testverbindung wie auch Scopolamin verabreicht wurden) gezählt. Die Ergebnisse sind in Tabelle 3 gezeigt. Unter den getesteten Verbindungen zeigten SUAM 1051, SUAM 1055 und SUAM 1065 besonders bemerkenswerte Wirkungen. - Die vorliegende Erfindung betrifft auch ein antiamnesisches Mittel, das zur Behandlung von Krankheiten, die von organischen Dysfunktionen im Gehirn herrühren, nützlich ist. Das anitamnesische Mittel umfaßt mindestens eine Verbindung der Formel (I) zusammen mit einem pharmazeutisch zulässigen Träger.
- Die Formulierung des Mittels der Erfindung schließt entweder feste Formulierungen wie Kapseln, Tabletten und Pulver oder flüssige Formulierungen wie Elixiere, Sirups oder Suspensionen zur oralen Verabreichung ein. Alternativ dazu können die aktiven Verbindungen (I) als Injektionen oder Suppositorien formuliert werden.
- Der im Mittel der Erfindung eingeschlossene Träger kann aus pulverförmigen festen Trägern wie Lactose, Saccharose, Dextrose, Mannitol, Sorbitol, Cellulose und Glycin usw. ausgewählt werden.
- Das Mittel der Erfindung kann ferner ein Schmiermittel, einen Binder oder einen Disintegrator enthalten. Beispiele für Schmiermittel sind Siliciumdioxid, Talk, Magnesiumstearat und Polyethylenglycol. Beispiele für Binder sind Stärke, Gelatine, Tragacanth, Methylcellulose und Polyvinylpyrrolidon. Beispiele für Disintegratoren sind Stärke, Agar usw.
- Der aktive Bestandteil (I) des erfindungsgemäßen Mittels wird oral einem erwachsenen Patienten in einer Dosis von 10 bis 4000 mg, vorzugsweise 100 bis 1000 mg/Tag verabreicht, oder parentaral in einer Dosis von 1 bis 2000 mg vorzugsweise 50 bis 500 mg/Tag verabreicht. Die Dosis kann abhängig von der Krankheit, Alter, Gewicht oder Zustand des Patienten oder der Formulierung des Arzneimittels variieren.

Claims (4)
1. N-Acylpyrrolidinderivate der allgemeinen Formel (I):
worin R¹ ein Wasserstoffatom, eine niedere Alkylgruppe
oder eine Phenylgruppe ist,
R³ eine Formylgruppe ist,
wenn R¹ ein Wasserstoffatom ist, R² eine substituierte
oder unsubstituierte Phenyloxygruppe oder
Phenylcarbonylgruppe oder eine substituierte
Phenylgruppe ist, und wenn R¹ eine niedere Alkylgruppe
oder Phenylgruppe ist, R² eine substituierte oder
unsubstituierte Phenylgruppe, eine niedere Alkylgruppe,
eine Aralkylgruppe von 7 bis 10 Kohlenstoffatomen oder
eine Hydroxygruppe ist; oder R¹ und R² zusammen eine
Benzylidengruppe bilden, welche unsubstituiert oder
substituiert an ihrer aromatischen Einheit sein kann,
und n eine Ganzzahl von 0 bis 5 ist.
2. Verfahren zur Herstellung von N-Acylpyrrolidinderivaten
der allgemeinen Formel (I):
worin R¹ ein Wasserstoffatom, eine niedere Alkylgruppe
oder eine Phenylgruppe ist,
R³ eine Formylgruppe ist,
wenn R¹ ein Wasserstoffatom ist, R² eine substituierte
oder unsubstituierte Phenyloxygruppe oder eine
Phenylcarbonylgruppe oder eine substituierte
Phenylgruppe ist, und wenn R¹ eine niedere Alkylgruppe
oder Phenylgruppe ist, R² eine substituierte oder
unsubstituierte Phenylgruppe, eine niedere Alkylgruppe,
eine Aralkylgruppe von 7 bis 10 Kohlenstoffatomen oder
eine Hydroxygruppe ist; oder R¹ und R² zusammen eine
Benzylidengruppe bilden, welche unsubstituiert oder
substituiert an ihrer aromatischen Einheit sein kann,
und n eine ganze Zahl von 0 bis 5 ist,
welches umfaßt:
Kondensierung einer Carbonsäure oder eines Chlorides
oder Anhydrides davon, die folgende Formeln haben:
worin R¹, R² und n dieselben Bedeutungen wie oben
haben, mit Prolin, Prolinalkylester oder einem Salz von
diesen der Formel:
worin R&sup4; ein Wasserstoffatom oder eine niedere
Alkylgruppe ist, wodurch eine Verbindung der Formel
(I') entsteht:
in welcher R³ -COOR&sup4; darstellt (worin R&sup4; ein
Wasserstoffatom oder eine niedere Alkylgruppe ist), die
gebildete Verbindung wird dann reduziert, woraus eine
Verbindung der Formel (I') entsteht, in welcher R³ -
CH&sub2;OH darstellt, und die so erhaltene Verbindung wird
danach oxidiert, woraus eine Verbindung der Formel (I)
entsteht.
3. Ein Anti-Prolylendopeptidase-Mittel umfassend als
aktiven Bestandteil mindestens ein N-
Acylpyrrolidinderivat der allgemeinen Formel (I):
worin R¹ ein Wasserstoffatom, eine niedere Alkylgruppe
oder eine Phenylgruppe ist,
R³ eine Formylgruppe ist,
wenn R¹ ein Wasserstoffatom ist, R² eine substituierte
oder unsubstituierte Phenyloxygruppe oder
Phenylcarbonylgruppe oder eine substituierte
Phenylgruppe ist, und wenn R¹ eine niedere Alkylgruppe
oder eine Phenylgruppe ist, R² eine substituierte oder
unsubstituierte Phenylgruppe, eine niedere Alkylgruppe,
eine Aralkylgruppe von 7 bis 10 Kohlenstoffatomen oder
eine Hydroxygruppe ist; oder R¹ und R² zusammen eine
Benzylidengruppe bilden, welche unsubstituiert oder
substituiert an ihrer aromatischen Einheit ist, und
n eine Ganzzahl von 0 bis 5 ist,
zusammen mit einem zulässigen Träger.
4. Ein antiamnesisches Mittel, umfassend als aktiven
Bestandteil mindestens ein N-Acylpyrrolidinderivat der
Formel (I) :
worin R¹ ein Wasserstoffatom, eine niedere Alkylgruppe
oder Phenylgruppe ist,
R³ eine Formylgruppe ist,
wenn R¹ ein Wasserstoffatom ist, R² eine substituierte
oder unsubstituierte Phenyloxygruppe oder
Phenylcarbonylgruppe oder eine substituierte
Phenylgruppe ist, und wenn R¹ eine niedere Alkylgruppe
oder Phenylgruppe ist, R² eine unsubstituierte oder
substituierte Phenylgruppe, eine niedere Alkylgruppe,
eine Aralkylgruppe von 7 bis 10 Kohlenstoffatomen oder
eine Hydroxygruppe ist; oder R¹ und R² zusammen eine
Benzylidengruppe bilden, welche unsubstituiert oder
substituiert an ihrer aromatischen Einheit ist, und
n eine Ganzzahl von 0 bis 5 ist,
zusammen mit einem pharmazeutisch zulässigen Träger.
