Die Erfindung bezieht sich auf Komplexsalze mit pharmakologischer
Aktivität, insbesondere Komplexsalze, deren Kation von einem Cyclohexyl-piperazinoacetamid oder -propionamid und deren
Organometall-Anion von einem Dihydroxyaluminium-^-hydroxycarboxylat
gebildet wird. Diese Komplexsalze bieten gleichzeitig geschwür-, sekrethemmende sowie puffernde Aktivität.

Zwei der Verbindungen der Formel (II) haben sich als unter pharmakologischem
Gesichtspunkt besonders brauchbar erwiesen, da sie eine beträchtliche antisekretorische und Antigeschwüraktivität
bieten. Es sind N-Cyclohexyl-1-piperazinylacetamid und N-4-Methylcyclohexy1-1-piperazinylacetamid.
Die Verbindungen der oben erwähnten US-PS können jedoch nicht in allen pharmazeutischen Formen angewandt werden, und zwar aufgrund
ihrer hohen Basizität.
Auch die anorganischen und organischen Säure-Salze, die gewöhnlich
in der pharmazeutischen Industrie eingesetzt werden, können nicht in allen pharmazeutischen Formen angewandt werden, mit Ausnahme
des Monohydrochlorids, das jedoch bemerkenswerte Herstellungsprobleme
bietet, da u.a. eine von disubstiuiertem Produkt freie N-Cyclohexylpiperazinylacetamid-Base
verwendet werden muß.
Organometall-Anionen, wie sie sich durch Kombinieren von Aluminiumhydroxid
mit ^-Hydroxycarbonsäuren ableiten,"sind aus der technischen
Literatur bereits bekannt. Im allgemeinen werden sie durch Salzbildung mit Tris-(hydroxymethyl)-aminomethan (THAM, auch als
TRIS-Puffer bezeichnet), kombiniert. TRIS-Puffer ist eine organische
Base mit Puffer-Vermögen, die jedoch keinerlei pharmakologische Eigenschaft systemischen Charakters besitzt.
Es wurde nun gefunden, daß durch Salzbildung der N-Cyclohexylpiperazinoacetamid-Basen
mit einem Dihydroxy-aluminium-oC-hydroxycarboxylat,
vorzugsweise von Milch-, Wein- oder Glucoleptonsäure und noch bevorzugter von Gluconsäure stammend, Korn-
plexsalze erhalten werden, die zugleich eine Antigeschwür- und
antisekretorische Aktivität sowie eine ausgezeichnete Pufferwirkung ergeben.
Diese neue Komplexsalze sind voll wasserlöslich«
Erfindungsgemäß können zusätzlich zu den N-Cyclohexylpiperazinoacetamid-Basen
andere Kationen der Formel (II) verwendet werden, worunter besonders N-4'-Methyl-cyclohexyl-i-piperazinoacetamid
zu erwähnen ist.
Dihydroxyaluminat-gluconat von N-Cyclohexyl-1-piperazinoacetamid,
das derzeit die am meisten bevorzugte erfindungsgemäße Verbindung darstellt, kann durch die folgende Strukturformel wiedergegeben

Das erfindungsgemäße Verfahren wird im folgenden Beispiel im Einzelnen
angegeben. Es läuft kontinuierlich ohne Isolierung der Zwischenstufen in praktisch quantitativer Ausbeute ab. Dieses
Beispiel schränkt die Erfindung nicht ein.
In einen mit einem Heizmantel, Thermometer, Rückflußkühler mit
Sammelkopf für das Destillat und Rührer ausgestatteten 5 1-Reaktor werden 329,5 g einer 50 (Gew.-/Gew.-)prozentigen wässrigen
Lösung von G.P. Gluconsäure (MG 196,16; 0,840 Mol) gebracht, und dann werden nach Verdünnen mit 480 ml Wasser 175,07 g 98%iges
Alumxniumisopropylat (entsprechend 171,57 g 100 %ig; MG 204,25; 0,840 Mol) eingeführt und das Gemisch 4 h bei 75°C unter Rühren
unter einem Stickstoffstrom erwärmt, um die Bestandteile vollständig
zu lösen. Beim Erwärmen erfolgt Sammeln verdünnten Isopropanols. Ist die Umsetzung beendet, wird das Gemisch wieder mit
Wasser auf das Ausgangsvolumen gebracht, kann sich auf Raumtemperatur
abkühlen, Diatomeenerde (Celite) wird zugesetzt und das anfallende Gemisch filtriert, um kleine suspendierte Teilchen zu
entfernen. Eine komplexometrische Titration erfolgt zur Kontrolle des Titers sowie der Ausbeute, die sich als praktisch quantitativ
erweist (188,9 g 100%iges Aluminiumgluconat; MG 225,131; 0,839 Mol).
Zur so erhaltenen Lösung wird allmählich unter Kontrolle mit Hilfe
eines pH-Meßgeräts eine 10(Gew.-/VoI.-)prozentige wässrige Lösung
sehr reinen N-Cyclohexy1-1-piperazinoacetamids in einer
0,839 Mol äquivalenten Menge gegeben, d.h. 189,05 g 100%ig (MG =■ 225,3), wobei der pH auf 8,35 - 8,36 eingestellt wird. Eine vollständige Lösung wird erhalten, die mit einem Sprühtrockner getrocknet
wird.
Das trockene Produkt hat einen Titer von 98,5 - 99 % bei einer Ausbeute von 97-98 %.

in ß zum Carbonyl); 70,8 und 69,6 (2 CHOH in γ und cf zum Carbonyl);
62,8 (CH2OH); 60,3 (COCH2N); 31,9, 25,0 und 24,5 (CH3 des Cyclohexan-Rings).
Pufferwirkung: Zu 4,6 g der Probe in einem 1000 ml-Becherglas
werden 400 ml 0,015 η HCl gegeben, wobei die Temperatur bei 37°C
(Thermostat) gehalten wird. Der pH-Wert wird nach 30 min gemessen, wobei kontrolliert wird, daß er zwischen 4,8 und 5,0 bleibt.
Dann wird mit einer Bürette 0,1 η HCl mit einer Geschwindigkeit von 1 ml/min eingebracht, um einen pH von 3,0 einzustellen. Als
Mindestmenge müssen 165 ml 0,1 η HCl verbraucht werden.
Die akute Toxizität des Komplexsalzes des Dihydroxyaluminiumgluconats
von N-Cyclohexyl-piperazinoacetamid wird nach dem üblichen
pharmakotoxikologischen Test an der Ratte und der Maus oral geprüft, die Ergebnisse finden sich in der folgenden Tabel
le.

Die Toxizität ist sehr gering, selbst in Relation zu der hohen
therapeutischen Dosis, die in diesem Falle erforderlich ist.
Was die pharmakologische Aktivität betrifft, zeigt das erfindungsgemäße
Komplexsalz eine neutralisierende Wirkung bei Magenübersäuerung, begleitet von einer antisekretorischen und Antigeschwürwirkung,
was durch die ED_ -Werte an der Ratte in Erscheinung
tritt, wie nachfolgend angegeben:

Die Antigeschwüraktivität wurde an der Ratte für verschiedene Arten von experimentellem Ulcer getestet. Die erhaltenen Werte
finden sich in Tabelle 2.

Die erfindungsgemäßen Komplexsalze können vorteilhaft als orale
pharmazeutische Formen, gekennzeichnet durch ein starkes.Dispersionsvermögen,
besonders Kautabletten mit hohem Zerfalls-
• ·
grad, und Suspensionen von Granulaten oder nikroverkapselten
Pulvern in inerten, pharmazeutisch annehmbaren Trägern verwendet werden.
Im Falle der vorgenannten speziellen Formen, in denen der disperse
Zustand den Kontakt mit der Schleimhaut fördern soll, in der der pH-Anstieg erfolgen soll, kann das Komplexsalz mit den
üblichen, pharmazeutisch annehmbaren Hilfsmitteln sowie-Aromatisierungs-
und Süßungsmitteln zusammengemischt werden.
Der erfindungsgemäß erhaltene Wirkstoff ist brauchbar zur Therapie
aller Formen von Magenübersäuerung, wie bei Magen-Darm-Geschwüren, sowie zur Behandlung von Pyrose, Gastralgie und
Gastritis.
Eine empfohlene Medikation ist 400 mg, 2-3 mal täglich, bis insgesamt
800 - 1200 mg/Tag.
Claims (6)
- PatentansprücheΠ.ι Komplexsalze von N-Cyclohexylpiperazino-acetamid oder -propionamid mit Dihydroxyaluminium-K-hydroxycarboxylaten.
- 2. Komplexsalz nach Anspruch 1, bestehend im wesentlichen aus Dihydroxyaluminiumgluconat von N-Cyclohexylpiperazino-acetamid, mit folgender FormelCH2-OH CH -OHCH -OH ICH -OH I
CH -0-.C -0" 8OH OHN-CH0-CO-NH 2 - 3. Verfahren zur Herstellung des Komplexsalzes, bestehend im wesentlichen aus Dihydroxyaluminiumgluconat von N-Cyclohexylpiperazino-acetamid, dadurch gekennzeichnet, daß es in nur einer Stufe, ausgehend von einer 50%igen Gluconsäurelösung, unter Umsetzen mit einer stöchiometrischen Menge Aluminiumisopropylat, Erwärmen unter einem Stickstoffstrom bis zum Ende der Reaktion zur Bildung des· Dihydroxyaluminiumgluconat-Anions und, nach Entfernen des Isopropylalkohols, Verdünnen der filtrierten Masse mit Wasser, wobei die tatsächliche stöchiometrische Menge durch Titration bestimmt wird, Salzbildung mit einer A'quivalent-Lösung von N-Cyclohexylpiperazino-acetamid und Trocknen des so erhaltenen Salzes durch Verdampfen durchgeführt wird.
- 4. Verfahren nach Anspruch 2, dadurch gekennzeichnet, daß die N-Cyclohexylpiperazino-acetamid-Lösung von disubstituiertem Produkt frei ist.
- 5. Pharmazeutische Zusammenstellung mit Antigeschwür-, antisekretorischer und puffernder Wirkung, enthaltend ein Komplexsalz gemäß Anspruch 1 zusammen mit einem oder mehreren Trägerin) und/oder pharmakologisch inerten und nicht-toxischen Excipientien.
- 6. Pharmazeutische Zusammenstel^ng mit Antigeschwür-, antisekretorischer und puffernder Wirkung in einer Einheitsdosis mit 400 mg eines Komplexsalzes gemäß Anspruch 2.
