Description
4,5-ungesättigte 16-Hydroxyprostansäurederivate
Die Erfindung betrifft neue 4,5'-ungesättigte 16-Hydroxyprostansäurederivate
der folgenden allgemeinen Formel

worin R -CH-OH oder -COOR", worin R" Wasserstoff oder Alkyl
mit 1 bis 6 C-Atomen ist, R- Wasserstoff oder Alkyl mit 1
bis 6 C-Atomen, η eine ganze Zahl von 2 bis 4, Y eine eis-Vinylen-
oder trans-Vinylengruppe und (+) die gezeigte
Verbindung, deren Spiegelbild und Gemische von Racematen bedeuten.
Die Niederalkylreste in vorstehender Formel sind typischerweise Methy, Ethyl, Propyl, Butyl, Pentyl, Hexyl und deren
verzweigtkettige Reste.
Ebenso von der Erfindung umfaßt sind die einzelnen Stereoisomeren, wobei Isomerengemische durch die Wellenlinien
in obiger Formel gekennzeichnet sind.

Erf indungsgexnäße Verbindungen, worin Y eine cis-Vinylengruppe
bedeutet, lassen sich durch folgende Formel darstellen

Verbindungen dieser Gruppe sind solche, worin R -COOR1 bedeutet, worin R1 Wasserstoff oder Alkyl mit 1 bis 6
C-Atomen ist. Besonders bevorzugte Verbindungen dieser Gruppe sind diejenigen Verbindungen, worin η = 3 bedeutet.

Die erfindungsgemäßen. Verbindungen besitzen wertvolle
pharmakologische Eigenschaften, wie beispielsweise ihre
Fähigkeit, die gastrische Sekretion zu inhibieren, die durch sekretionsanregende Mittel wie Histamin und Pentagastrin
stimuliert würde, während sie weiterhin den überraschenden Vorteil besitzen, daß sie nicht die potenten
unerwünschten Nebenwirkungen aufweisen, die verwandte Substanzen zeigen.
Der spezifische Versuch zur Bestimmung der gastrischen antisekretorischen Wirksamkeit wird nachstehend beschrieben.
13 bis 20 kg schwere erwachsene Beagle-Hündinnen werden mit entnervten Heidenhain-Fundustaschen präpariert.
Nach einer Genesungszeit von mindestens 4 Wochen nach
diesem Eingriff läßt man die Tiere etwa 20 Stunden fasten, wonach sie in Pavlov-Ständer getan werden und ihnen intravenös
eine Kochsalzlösung infundiert wird. Die Taschensekrektionen werden alle 15 Minuten gesammelt und auf
Volumen und Gesamtsäure durch Titration mit 0,1N Natrium—; hydroxid auf einen pH-Wert von 7,0 gemessen.
Nach 30 minütiger Basalsekretion erhalten die Hunde eine
Salzlösung von Histamindihydrochlorid in einer Dosis von 1,0 mg/Std. infundiert. Das Volumen der Diffusion wird
bei etwa 13 ml/Std. gehalten. Ein gleichbleibender Spiegel
der gastrischen Sekretion wird nach etwa einer Stunde nach Beginn der Histamininfusion erzielt, wonach die Testverbindung,
die in einer ethanolischen iso-osmotischen Phosphatpufferlösung
gelöst ist, durch eine einzige intravenöse Injektion verabreicht wird. Die Dauer der antisekretorischen
Wirkungen wird bestimmt und die Nebenwirkungen, falls vorhanden, aufgezeichnet. Die Verbindung wird als

wirksam bewertet, wenn statistisch signifikante Inhibierung der sekretorischen Parameter nach Behandlung mit
der Verbindung erfolgt.
Geeignete Ausgangsmaterialien zur Herstellung der erfindungsgemäßen
Verbindungen sind die Cyclopent-1-enalkansäuren
und -ester der folgenden Formel

Schutzgruppe bedeutet, wie Tri(niederalkyl)silyl, Tetrahydrofuranyl oder Tetrahydropyranyl. Die Herstellung

Die Einführung der sauerstoffhaltigen trans-Vinylseitenkette
in der 2-Stellung des Cyclopentanrings wird
durch Umsetzung mit einem geeigneten örganometallischen
Reagens bewirkt. Die sauerstoffhaltigen trans-Vinylseitenkettengruppen
werden aus den entsprechenden Acetylenen nach einem Verfahren hergestellt, das durch Pappo et al in
Chemistry, Biochemistry and Pharmacological Acitivity of Prostanoids, 17-26 (1979) beschrieben wird. Beispiel 13
beschreibt die Herstellung eines trans-Vinylstannidausgangsmaterials
aus dem entsprechenden Acetylen. Nachdem die Seitenkette eingeführt worden ist, werden die Sauerstoff schutzgruppen
zweckmäßigerweise mit einer schwachen Säurelösung wie Essigsäure entfernt.
Die Verbindungen, worin R eine Hydroxymethylgruppe bedeutet, werden aus dem Enolether des entsprechenden Esters wie in
den Beispielen 17 und 18 erläutert ist hergestellt. Der geschützte Prostansäureester, der durch die Reaktion der
Ausgangsmaterialien hergestellt wurde, wird mit Triethyloxoniumtetrafluorborat
behandelt, wobei der entsprechende Enolether entsteht. Der Enolether wird mit Lithiumaluminiumhydrid
reduziert und mit einer schwachen Säure angesäuert, wobei man das entsprechende 9-Oxo-11,16-dihydroxyprost-4,13-dien-1-ol
erhält.
Wenn anstelle der racemi sehen Seitenkette eine aufgespaltene
Seitenkette verwendet wird, bildet sich ein Gemisch aus Diastereoisomeren (Beispiele 20 und 21). Dieses Gemisch aus
Diastereoisomeren kann dann chromatographiert werden, wobei
man die einzelnen stereoisomeren Produkte (Beispiel 22) erhält.

Nachstehende Beispiele dienen der weiteren Erläuterung der Erfindung. In diesen Beispielen sind die Temperaturen
in 0C und die Mengen der Materialien in Gew.-Teilen angegeben,
außer wenn etwas anderes vermerkt wird.
4,0 Teile 5-Chlor-2-pentanonethylenketal wurden mit 9,0 Teilen Lithiumbromid und 2,0 Teilen Diisopropylethylamin
in 30 Volumenteilen Tetrahydrofuran, das von Lithiumaluininiumhydrid
abdestilliert worden war, vermischt. Das Gemisch wurde unter Stickstoff 48 Stunden lang unter Rückfluß
gehalten, gekühlt und in ein Gemisch aus Ether und Wasser zur Extraktion gegossen. Die Etherschicht wurde
2 χ mit Wasser, dann mit 1N Salzsäure und dann 2 χ wieder
mit Wasser gewaschen. Die Etherschicht wurde dann über Natriumsulfat getrocknet und unter vermindertem Druck eingeengt,
-wobei man 5-Brom-2-pentanonethylenketal der Formel erhielt

0,1 Teil p-Toluolsulfonsäure wurde unter Rühren einem Gemisch
aus 4,2 Teilen 4-Pentyn-1-ol und 5,0 Teilen Dihydropyran zugesetzt. Nach etwa 30 Minuten wurde das Gemisch mit

0,5 Teilen Triethylamin behandelt und vakuumdestilliert, wobei man 2-Tetrahydropyranyl-4-pentynylether der Formel
erhielt

Eine Lösung, die 18,5 Teile 2-Tetrahydropyranyl-4-pentynylether
von Beispiel 2 enthielt, in 125 Volumenteilen Tetrahydrofuran» das von Lithiumaluminiumhydrid frisch destilliert
worden war, wurde auf etwa -300C gekühlt und mit 46 Volumenteilen
einer 2,4 molaren n-Butyllithiumlösung in Hexan
behandelt. Man ließ die Lösung sich über Nacht auf Raumtemperatur erwärmen. Nach etwa 30 Minuten bei Raumtemperatur
wurden 21 Teile 5-Brom-2-pentanonethylenketa1 des Beispiels
und anschließend 30 Volumenteile Hexamethylphosphortriamid unter Rühren zugesetzt. Nach 1 Stunde wurde das Reaktionsgemisch
in ein Gemisch aus Ether und 1N Salzsäure gegossen. Die Etherschicht wurde mit Wasser gewaschen, über Natriumsulfat
getrocknet und das Lösungsmittel im Vakuum abgestreift, wobei man das Produkt als eine farblose viskose
Flüssigkeit der folgenden Formel erhielt

30 Teile des Decyny!ketals des Beispiels 3 wurden in einem
Gemisch aus 150 Volumenteilen 1N Salzsäure, 200 Volumenteilen Tetrahydrofuran und 50 Volumenteilen Methanol gelöst- Die
Lösung wurde 48 Stunden lang' auf Raumtemperatur gehalten und dann 5 bis 6 Stunden unter Rückfluß behandelt- Die Lösung
wurde dann auf Raumtemperatur gekühlt,und festes Kaliumcarbonat wurde zugesetzt, bis zur Erreichung eines pH-Wertes von 7.
Die Lösung wurde dann bis zur Hälfte ihres Volumens abgestreift, mit Wasser verdünnt und 2 χ mit Ether extrahiert- Die Etherextrakte
wurden vereinigt, mit Wasser gewaschen, über Natriumsulfat getrocknet, wonach das Lösungsmittel abgestreift wurde,
wobei man 9-Oxodec-4-yn-1~ol erhielt, das ohne Reinigung in Beispiel 5 verwendet wurde.
20 Teile 9-Oxodec-4-yn-1-ol des Beispiels 4 wurden in 200
Volumenteilen Aceton gelöst und auf 00C gekühlt. Die kalte
Lösung wurde gerührt und tropfenweise mit 90 Volumenteilen eines 2,67 molaren Jones-Reagens (Chromsäure in Schwefelsäure
und Wasser) behandelt. Die Acetonlösung wurde von den festen Chromsalzen abdekantiert, die dann mit frischem Aceton gespült
wurden. Die Acetonlösungen wurden vereinigt und in ein Gemisch aus Ether und Wasser gegossen. Die Etherschicht wurde vom
Wasser abgetrennt, Ί χ mit Wasser gewaschen und dann 3 χ mit einer 5%igen Kaliumcarbonatlösung extrahiert. Die alkalischen
Extrakte wurden vereinigt, mit konzentrierter Salzsäure angesäuert und 2 χ mit Ether und 1 χ mit Ethylacetat extrahiert.

Die Extrakte wurden vereinigt, über Natriumsulfat getrocknet und anschließend das Lösungsmittel abgestreift, wobei man
die reine 9-Oxodec-4-yn· säure erhielt.
10 Teile der 9-Oxodec-4-yn ^äure des Beispiels 5 wurden bei
Raumtemperatur in Toluol, das etwa 0,5 % Chinolin mit 5 % Palladium auf Bariumsulfat als Katalysator enthielt,
hydriert. Die Tolüollösung wurde mit 1N Salzsäure und dann mit Wasser gewaschen. Die Lösung wurde über Nacht über
Natriumsulfat getrocknet und das Lösungsmittel abgestreift, wobei man das Produkt cis-9-Oxodec-4-ensäure als gelbes
öl erhielt.
50 Volumenteile t-Butylalkohol wurden mit 3,2 Teilen
metallischem Kalium versetzt und unter Argon rückflußbehandeli Nachdem sich das Kalium gelöst hatte, wurde eine Lösung aus
2,5 Teilen cis-9-Oxodec-4-ensäure und 4,85 Teilen Dimethyloxalat, das aus Hexan in 25 Volumenteilen t-Butylalkohol
umkristallisiert worden war, tropfenweise der rückflußbehandelten Lösung binnen 1 Stunde zugesetzt. Das Reaktionsgemisch wurde zwei weitere Stunden rückflußbehandelt, auf
Raumtemperatur gekühlt und unter Argon filtriert. Der orangefarbige Filterkuchen wurde einem Gemisch aus Chloroform
und 1N Salzsäure zugesetzt. Die Chloroformschicht wurde
mit einer gesättigten Natriumchloridlösung gewaschen, über Natriumsulfat getrocknet und das Lösungsmittel abgestreift,
wobei man das Produkt 7-(2,3,5-Trioxo-4-methoxalylcyclopentan)hept-4-cis-ensäure
der folgenden Formel erhielt

4,0 Teile der 7-(2,3/5-Trioxo-4-methoxalylcyclopentan)-hept-4-cis-ensäure
des Beispiels 7 wurden 100 Volumenteilen einer 1N Salzsäure zugesetzt und 3 Stunden lang unter Argon
rückflußbehandelt. Die Lösung wurde auf Raumtemperatur gekühlt, abfiltriert und 2 χ mit Ethylacetat extrahiert. Die
Extrakte wurden vereinigt und 2 χ mit gesättigter Natrium-Chloridlösung
gewaschen, getrocknet und das Lösungsmittel
abgestreift, unter Erzielung eines roten Öles. Das rote
öl wurde an Silicagel chromatographiert (60 % Ethylacetat, 39 % Hexan und 1 % Essigsäure als Eluierungsmittel), wobei man 7-(2,3,5-Trioxocyclopentan)hept-4-cis-ensäure als
einen gelben Feststoff mit einem Schmelzpunkt von 78 bis 800C der allgemeinen Formel erhielt
abgestreift, unter Erzielung eines roten Öles. Das rote
öl wurde an Silicagel chromatographiert (60 % Ethylacetat, 39 % Hexan und 1 % Essigsäure als Eluierungsmittel), wobei man 7-(2,3,5-Trioxocyclopentan)hept-4-cis-ensäure als
einen gelben Feststoff mit einem Schmelzpunkt von 78 bis 800C der allgemeinen Formel erhielt

1,15 Teile 7-(2,3,5-Trioxocyclopentan)hept-4-cis-ensäure
wurden in 35 Volumenteilen Ethanol und 30 Volumenteilen Wasser gelöst und auf 00C gekühlt. 0,55 Teile Natriumborhydrid
wurden in 5,0 Volumenteilen Wasser gelöst und tropfenweise der Ethanollösung zugesetzt. Nach Beendigung der Zugabe
wurde die Lösung 30 Minuten lang bei 00C gerührt. Die Lösung
wurde in Ethylacetat und 1N Salzsäure gegossen. Die wäßrige Schicht wurde 3 χ mit zusätzlichem Ethylacetat extrahiert.
Die Ethylacetatextrakte wurden vereinigt, 1 χ mit gesättigtem Natriumchlorid gewaschen, über Natriumsulfat getrocknet
und das Lösungsmittel abgestreift, wobei man als ein viskoses gelbes öl (-i) -7- (2,5-Dioxö-3-hydroxycyclopentan)hept-4-cis-ensäure
der allgemeinen Formel erhielt

und deren verschiedene tautomere Enolformen.
Beispiel 10
Eine Lösung aus 2,0 Teilen (+)-7-(2,5-Dioxo-3-hydroxycyclopentan)hept-4-cis-ensäure
in 30 Volumenteilen trockenem Methanol wurden mit 10 Volumenteilen 2,2-Dimethoxypropan
und 4 Volumenteilen 1%iger methanolischerSalzsäure versetzt. Man ließ das Gemisch 48 Stunden lang bei Raumtemperatur steher
und streifte es bei Raumtemperatur unter vermindertem Druck zur Trockne ab. Man setzte etwa 4 Volumenteile Ether zu und
ließ das Gemisch bei Raumtemperatur weitere 48 Stunden lang stehen. Das erstarrte Gemisch wurde in Toluol aufgenommen,

das 1 % Triethylamin enthielt,und die Lösung nacheinander
mit verdünntem Kaliumcarbonat und Wasser gewaschen, über Natriumsulfat getrocknet und das Lösungsmittel abgestreift.
Der Rückstand wurde aus Ether umkristallisiert und ergab das Produkt, (+_) -Methyl-7- ('i-hydroxy-^-meth.oxy-S-oxocyclopent-1-en)hept-4-cis-enoat
als einen weißen Feststoff mit einem Schmelzpunkt von 82 bis 84?C der allgemeinen Formel

100 Volumenteile trockenes Toluol wurden in einen Dreihalskolben gegeben und in einem Isopropylalkohol/Trockeneisbad
auf -700C gekühlt. 15,5 Volumenteile 1,83molares Natriumdihydrobis-(2-methoxyethoxy)aluminat, verdünnt mit
100 Volumenteilen Toluol und eine Lösung aus 6,92 Teilen {+)-Methyl-7-(4-hydroxy-2-methoxy-5-oxocyclopent-1-en)hept-4-cis-enoat
in 200 Volumenteilen Toluol wurden in getrennte Tropftrichter gegeben. Die beiden Lösungen wurden
tropfenweise und gleichzeitig dem Kolben zugesetzt. Während der Zugabe durfte die Temperatur des Kolbens nicht -600C
übersteigen. Das Gemisch wurde bei -700C 3,5 Stunden und
dann bei 00C 15 Minuten gerührt, mit einer Lösung von 5,0
Volumenteilen Methanol in 10 Volumenteilen Toluol abgeschreckt
und mit 150 Volumenteilen 1N Salzsäure angesäuert. Die organische Schicht wurde abgetrennt, mit Wasser gewaschen,
über Natriumsulfat getrocknet und das Lösungsmittel abgestreift. Der Rückstand wurde an Silicagel chromatographiert (70 %

Ethylacetat, 30 % Hexan als EIuierungsmittel), wobei man
(_+) -Methyl-7- (3-hydroxy-5~oxocyclopent-1 -en) hept-4-cisenoat
als ein viskoses öl der folgenden Formel erhielt.

2,6 Teile (+_) -Methyl-7- (S-hydroxy-S-oxocyclopent-i -en) hept-4-cis-enoat
wurden in 20 Volumenteilen Dimethylformamid gelöst und nacheinander mit 1,0 Teilen Imidazol und 1,9 Teilen
Triethylsilylchlorid behandelt. Die Lösung wurde 1 Stunde lang bei Raumtemperatur gerührt, mit Ether verdünnt, 3 bis
4 χ mit Wasser gewaschen, über Natriumsulfat getrocknet und das Lösungsmittel abgestreift. Das Produkt, das direkt in
Beispiel 14 verwendet wurde, ist (_+) -Methyl-7- (3-triethylsilyloxy-5-oxocyclopent-1-en)hept-4-cis-enoat
der folgenden Formel

Beispiel 13
2,12 Teile 4(RS)-Trimethylsiloxy~4-methyl-1-octyn und
3,0 Teile Tri-n-butylzinnhydrid wurden vermischt und
unter Argon mit einer Sonnenlampe 2 Stunden lang bei 00C
und dann 2 Stunden lang bei 550C bestrahlt. Das dabei
entstehende Produkt wurde direkt in Beispiel 14 verwendet.
1,0 Teile des trans-Vinylstannidproduktes von Beispiel 13
wurden in 3,0 Volumenteilen trockenem Tetrahydrofuran gelöst,
auf -600C gekühlt und mit 0,87 Volumenteilen einer 2,3molaren
Lösung von n-Butylüthium in Hexan behandelt. Die Lösung wurde
1 Stunde lang bei -600C gerührt und dann mit einer Etherlösung
behandelt, die 0,26 Teile Kupfer-1-pentynylid und 0,64 Teile
Hexamethylphosphortriamid enthielt. Die Lösung wurde dann weitere 10 Minuten bei -600C gerührt und eine Etherlösung,
die 0,35 Teile des (+)-Methyl-7-n-triethylsilyloxy-S-oxocyclopent-1-en)hept-4-cis-enoats
des Beispiels 12 enthielt, wurde zugesetzt. Die Lösung wurde 1 Stunde lang gerührt und
in ein Gemisch aus Ether und 1N Salzsäure gegossen. Die Etherschicht wurde abgetrennt, 2 χ mit Wasser gewaschen, filtriert,
über Natriumsulfat getrocknet und das Lösungsmittel abgestreift. Der Rückstand wurde an Silicagel Chromatographiert (10 %
Ethylacetat, 90 % Hexan als Eluierungsmittel) und ergab als
Produkt racemisches Methyl-7-/3~σc-triethylsil·yl·oxy-2ß-(4
(RS) ^-trimethylsilyloxy^-methyl-i-trans-octenyl) -5-oxocyclopentan/-1(X-hept-4-cis-enoat
der folgenden Formel

0,30 Teile Methyl-7-/3££-triethylsilyloxy-2ß- (4 (RS) -4-trimethylsilyloxy-4-methyl-1-trans-octenyl)-5-oxocyclopentan7-1o6-hept-4-cis-enoat
wurden in 5,0 Volumenteilen
eines 3:1 :1-Gernischs aus Essigsäure:Tetrahydrofuran;Wasser
gelöst und 30 Minuten lang bei Raumtemperatur stehengelassen. Die Lösung wurde mit Ether verdünnt, 5 χ mit Wasser gewaschen,
über Natriumsulfat getrocknet und das Lösungsmittel abgestreift.. Der Rückstand wurde an Silicagel chromatographier
(100 % Ethylacetat als Eluierungsmittel) und ergab als
Produkt als ein viskoses farbloses öl racemisches Methyl-7-/3oC-hydroxy-2ß-(4 (RS) -^-hydroxy-^-methyl-i-trans-octenyl) -5-oxocyclopentan7-1o;-hept-4-cis-enoat der Formel

1,0 Teile racemisches Methyl-7-/3cC-hydroxy-2ß-(4 (RS) -4-hydroxy-4-methyl-1
-trans-octenyl) -5-oxocyclopentan7-1<^>hept-4-cis-enoat
wurden in 50 Volumenteilen 95%igem Ethanol gelöst und 30 Volumenteilen eines Tris(2-amino-2-hydroxymethyl

1,3-propandiol)-puffers mit einem pH-Wert von 7,8 zugesetzt.
Dieses Gemisch wurde mit 0,15 Teilen einer Schweineleberesterase
(Sigma Chemical Co. No. E-3128) behandelt und 3 bis 4 Stunden bei Raumtemperatur gerührt. Das Gemisch wurde mit
Ether verdünnt, mit 1N Salzsäure und dann mit Wasser gewaschen, über Natriumsulfat getrocknet und das Lösungsmittel abgestreift.
Der Rückstand wurde an Silicagel chromabographiert (100 %
Ethylacetat als Eluierungsmittel) und ergab als Produkt racemisches 7-/3CC- Hydroxy-2ß—(4 (RS) -4-hydroxy-4-methyl-1-trans-octenyl)
-5-oxocyclopentan7-10C-hept-4-cis-ensäure der Formel

1,0 Teile des trans-Vinylstannidproduktes von Beispiel
wurdai in 3,0 Volumenteilen trockenem Tetrahydrofuran gelöst, auf -600C gekühlt und mit 0,87 Volumenteilen einer 2,3molaren
Lösung von n-Butyllithium in Hexan behandelt. Das Reaktionsgemisch
wurde 1 Stunde lang bei -6O0C gerührt und dann mit einer
Etherlösung behandelt, die 0,26 Teile Kupfer-1-pentynilid
und 0,64 Teile Hexamethylphosphortriamid enthielt. Die Lösung wurde dann weitere 10 Minuten bei -6O0C gerührt und eine
Etherlösung zugesetzt, die 0,35 Teile des (_+)-Methyl-7-(3-

triethylsilyloxy-5-oxocyclopentan)-hept-4-cis-enoats
von Beispiel 12 enthielt. Die Lösung wurde 1 Stunde lang
gerührt und dann mit 3,0 Volumenteilen Triethyloxoniumtetrafluorfoorat
behandelt (1 molar in Methylenchlorid), Man rührte das Gemisch 30 Minuten lang bei -50°C und ließ
es dann langsam sich auf 00C erwärmen. Das Gemisch wurde
dann in Ether und eine verdünnte Kaliumbicarbonatlösung gegossen. Die Schichten wurden getrennt und die Etherschicht
mit zusätzlicher verdünnter Kaliumbicarbonatlösung gewaschen, über Natriumsulfat getrocknet und das Lösungsmittel abgestreift.
Der Rückstand wurde an Silicagel chromatographiert (10 % Ethylacetat, 90 % Hexan mit 0,2 % Triethylamin als
Eluierungsmittel) und ergab racemisches Methyl-7-/^3-triethylsilyloxy-2ß~(4(RS)~4-trimethylsilyloxy-4-methyl-1-transoctenyl)-5-ethyloxycyclopent-i(5)-eny-ICtder

100 Volumenteilen trockenem Tetrahydrofuran gelöst, auf 00C gekühlt und mit 0,3 Teilen Lithiumaluminiumhydrid be-

handelt. Nach 15 Minuten bei O0C wurde das Reaktionsgemisch
in Ether und Wasser gegossen. Die Etherschicht wurde abgetrennt, wieder mit Wasser gewaschen, über Natriumsulfat
getrocknet und das Lösungsmittel abgestreift. Der Rückstand wurde in 50 Volumenteilen eines 3:1:1-Gemische aus Essigsäure:
Wassertetrahydrofuran aufgenommen und 30 Minuten lang bei
Raumtemperatur stehengelassen. Die Lösung wurde mit Ether verdünnt, 4 bis 5 χ mit Wasser gewaschen, über Natriumsulfat
getrocknet und das Lösungsmittel abgestreift. Der Rückstand wurde an Silicagel chromatographiert (100 % Ethylacetat
als Eluierungsmittel) und ergab als farbloses viskoses Öl als Produkt racemisches 7-/3aCrHydroxy-2ß- (4 (RS) -4-hydroxy-4~inethyl~1-trans-octenyl)
-S-hept-4-cis-en-1-öl der Formel

Die erfindungsgemäßen A ' -trans-Prostansäurederivate werden
nach folgenden Verfahren hergestellt:
0,10 Teile metallisches Lithium wurden 15 Volumenteilen
wasserfreiem Ammoniak in einem Dreihalskolben zugesetzt.

der mit einem Trockeneiskühler ausgestattet und in ein Trockeneis/Isopropanolbad eingetaucht war. 1,5 Teile
des Produktes von Beispiel 3 und 0,5 Völumenteile t-Butanol wurden vermischt und tropfenweise der Ammoniaklösung
zugesetzt. Nach beendeter Zugabe wurde das Reaktionsgemisch 2 Stunden lang gerührt- Der Kolben wurde aus dem Eisbad
entfernt und das Reaktionsgemisch mit festem Ammoniumchlorid abgeschreckt. Danach wurde Ether tropfenweise zugesetzt,
und man ließ den Ammoniak verdampfen. Die Lösung wurde in Ether und verdünnte Salzsäure gegossen. Die Etherschicht
wurde abgetrennt, 2 χ mit verdünnterSalzsäure und dann mit Wasser gewaschen, über Natriumsulfat getrocknet und
das Lösungsmittel abgestreift, wobei man das Produkt der folgenden Formel erhielt
Wenn die obige trans-Verbindung anstelle der Acetylenverbindung in Beispiel 4 eingesetzt wird und die Beispiele
5, 7, 8, 9, 10, 11, 12, 14 und 15 durchgeführt werden, erhält man als Produkt racemisches Methyl-7-/3Kt -hydroxy-2ß-(4(RS)-4-hydroxy-4-methyl-1-trans-octenyl)-5-OXO-cyclopentan7-1o£-hept-4-trans-enoat

in Beispiel 4 eingesetzt und die Beispiele 5, 7, 8, 9, 10, 11, 12, 17 und 18 durchgeführt werden,
erhält man als Produkt racemisches 7-/3OC-Hydroxy-2ß-(4(RS)-4-hydroxy-4-methyl-1-trans-octenyl)-S-oxocyclopentan/-1o6-hept-4-trans-en-1-ol

2,12 Teile (4S) ^-Trimethylsilyloxy^-methyl-i-octyn, das
nach der in "Recent Developments in the Synthesis of Antisecretory Prostaglandins", R. Pappo et al in Chemistry,
Biochemistry and Pharmacological Activity of Prostanoids, 1979 beschriebenen Methode erhalten worden war, und 3,0
Teile Tri-n-butylzinnhydrid wurden gemischt und unter Argon mit einer Sonnenlampe 2 Stunden lang bei 00C und dann 2 Stunden
lang bei 550C bestrahlt. Das dabei entstehende Produkt
wurde direkt in Beispiel 21 verwendet.
1,0 Teile des trans-Vinylstannidproduktes von Beispiel 20
wurden in 3,0 Volumenteilen trockenem Tetrahydrofuran...gelöst, auf -600C gekühlt und mit 0,87 Volumenteilen einer 2,3 molaren
Lösung von n-Butyllithium in Hexan behandelt. Die Lösung wurde 1 Stunde bei -600C gerührt und dann mit einer Etherlösung
behandelt, die 0,26 Teile Kupfer-T-pentynylid und 0,64 Teile Hexamethylphosphortriamid enthielt. Die Lösung
wurde dann weitere 10 Minuten bei ^600C gerührt und eine
Etherlösung, die 0,35 Teile des racemischen Methyl-7-(3-triethylsilyloxy-5-oxocyclopent-1-en)hept-4-cis-enoats
des Beispiels 12 enthielt, zugesetzt. Die Lösung wurde
gerührt und in ein Gemisch aus Ether und 1N Salzsäure gegossen.
Die Etherschicht wurde abgetrennt, 2 χ mit Wasser gewaschen, filtriert, über Natriumsulfat getrocknet und
das Lösungsmittel abgestreift. Der Rückstand wurde an SiIicagel
chromatographiert (10 % Ethylacetat, 90 % Hexan

und 0,35 Teilen Ν,Ν-Diisopropylethylamin behandelt.
Die Lösung wurde über Nacht gerührt. Die Lösung wurde mit Ether verdünnt, mit kalter verdünnter Salzsäure und ....
dann 2 bis 3 χ mit Wasser gewaschen, getrocknet und an Silicagel chromatographiert (100 % Ethylacetat als Eluierungsmittel)
und ergab als Produkt racemisches Isopropyl-7-/3
(Qi) -hydroxy-2ß- (4 (RS) -4-hydroxy-4-methyl-1 -trans-octenyl)-5-oxocyclopentan7-1Ai-hept-4-cis-enoat

Claims (10)
- Patentansprüche: ( 1.\ Verbindung der Formelworin R -CH2OH oder -C-OR1, worin R1 Wasserstoff oder Alkyl mit 1 bis 6 C-Atomen ist, R- Wasserstoff oder Alkyl mit 1 bis 6 C-Atomen, η eine ganze Zahl von 2 bis 4, Y eine cis-Vinylen- oder trans-Vinylengruppe und (+_) die gezeigte Verbindung, deren Spiegelbild oder das Gemisch von Racematen bedeuten.
- 2. Verbindung nach Anspruch 1 der FormelO.Y'(CH2Jn-CH3worin R1 Wasserstoff oder Alkyl mit 1 bis 6 C-Atomen, R- Wasserstoff oder Alkyl mit 1 bis 6 C-Atomen, η eine ganze Zahl von 2 bis 4 und Y eine cis-Vinylen- oder trans-Vinylengruppe bedeuten.130035/0479

- 6. Verbindung nach Anspruch 2, nämlich das racemische Methyl-7-/3ct-hydroxy-2ß-(4 (RS) -hydroxy-4-methyl-1 -trans-octenyl) 5-oxocyclopentan7-1<il-hept-4-trans-enoat.
- 7. Verbindung nach Anspruch 1 der FormelCH2OHHO R,HO130035/0479worin R1 Wasserstoff oder Alkyl mit 1 bis 6 C-Atomen/ η eine ganze Zahl von 2 bis 4 und Y eine cis-Vinylen- oder trans-Vinylengruppe bedeuten.
- 8. Verbindung nach Anspruch 7, nämlich racemisches 7-/3^ Hydroxy-2ß-(4-(RS)-hydroxy-4-methyl-1-octenyl)-5-oxocyclopentan7-'üb>i-hept-4-cis-en-1-öl.
- 9. Verbindung nach Anspruch 1, nämlich Methy1-7-/3(S) hydroxy-2ß-(4(S)-hydroxy-4-methyl-i-trans-octenyl)-5-oxocyclopentan7-10i-hept-4-ciS'-enoat.
