Arornamittel und ihre Herstellung und
Vertuen dung
Die vorliegende Erfindung bezieht sich auf Aromarnittel, nämlich
auf die Verwendung bestimmter Acylpyrimidinderivate, insbesondere
der Derivate der Formel (i):

Die Verbindungen der Formel (i) besitzen interessante organoleptische
Eigenschaften und können daher mit Erfolg in der Aromaindustrie
vertuendet werden.
Im Lauf des letzten Jahrzehnts haben verschiedene Forschungsgruppan
ihre Aufmerksamkeit auf die Untersuchung der Aromaeigenschaften
heterocyclischer Verbindungen, nämlich stickstoffhaltiger
heterocyclischer Verbindungen, gerichtet. Verschiedene Pyrazine

zeigen laut Angaben in der Literatur Eignung als Aromamittel und
sind als wesentliche Bestandteile zur Rekonstituierung von unterschiedlichen
Aromapräparaten angesehen morden (vgl. z.B. Fenaroli's
Handbook of Flavor Ingredients, CRC Press Inc., Cleveland (1975), Bd. II, Seite 692 ff.; und die GB PSS 1 156 472, 1 156 475 und
1 156 484).
Bestimmte stickstoffhaltige, heterocyclische Derivate der Klasse
der Pyridine, Pyrrole sowie Thiazole haben ebenfalls, laut Angaben,
wertvolle gustative Eigenschaften (vgl. die GB PSS 1 156 483,
1 156 482 und 1 156 485). Bisher ist jedoch noch nirgends vorgeschlagen morden, Pyrimidinverbindungen als Aromabestandteile zu
verwenden.
Überraschenderweise wurde nun festgestellt, daß das Aroma und der Geschmack von Lebensmitteln im allgemeinen, Getränken, Tiernahrung,
pharmazeutischen Präparaten und Tabakprodukten durch Zugabe einer
wij^samen Menge mindestens einer Verbindung der Formel (l) verändert
werden können.
Eine besonders bevorzugte Klasse dieser Verbindungen umfaßt

Die meisten der obigen Verbindungen sind, tuie angegeben, neu.
Die obigen neuen Verbindungen umfassen besonders interessante Ver
bindungen der folgenden Formel (i1):

in welcher R für ein Wasserstoffatom steht und R einen Isobutyl-

rest bedeutet oder R für einen Methylrest steht und R " einen
Methyl-, Äthyl- oder Isobutylrest bedeutet.
Ziel der vorliegenden Erfindung ist ein Verfahren zum Verstärken,
Verbessern oder Modifizieren der Aromaeigenschaften von Lebensmitteln,
Getränken, Tiernahrung, pharmazeutischen Präparaten und Tabakprodukten, das dadurch gekennzeichnet ist, daß man diesen
Materialien eine geringe, aber wirksame Menge mindestens einer Verbindung
der Formel (i) zufügt. Weiter betrifft die vorliegende Erfindung Aromapräparate, denen mindestens eine Verbindung der
Formel (i) zugegeben ist.
Die vorliegende Erfindung schafft insbesondere ein Verfahren zum Modifzieren der organoleptischen Eigenschaften von Fleisch, Fleisch
imitierenden oder fleischhaltigen eßbaren Materialien, das dadurch
gekennzeichnet ist, daß man diesen Materialien mindestens eine Verbindung der Formel (i) zugibt.
In ihrem reinen Zustand entwickeln die Verbindungen der Formel
(I) gustative Noten verschiedener Art, wie z.B. tierische, gegrilltB, geröstete und fettige Aromanoten. Ihr süßlicher und
karamellartiger Aromacharakter erinnert an den von Fleisch entwickelten Geschmack. Die Verbindungen dar Formel (i) eignen sich
in typischer Weise sehr gut bei der Herstellung von Aromapräparaten, die zur Aromatisierung von Fleisch, Flefech imitierenden
oder fleischhaltigen eßbaren Materialien bestimmt sind. Aber selbstverständlich haben die Pyrimidine der Formel (i) aufgrund
ihrer gustativen Eigenschaften ein breites Verwendungsspektrum und können in vielen verschiedenen Materialien, wie Kaffee,
Schokolade, Milchprodukten, Yoghurts, Konditor- und Bäckereiwaren, verwendet u/erden.
Weiterhin eignen sich die Verbindungen (i) zur Aromatisierung
von Tabak-und Tabakersatzprodukten.
Die Anteile, bei welchen die Verbindungen eine interessante gustative
Wirkung erzielen können, variieren innerhalb weiter Grenzen. Diese Anteile liegen vorzugsweise zwischen etwa 0,02-20 ppm (Gew.),
bezogen auf das Gesamtgewicht des Materials, in welches sie einverleibt werden. Diese Werte sollten jedoch nicht restriktiv interpretiert
werden, wobei der Fachmann selbstverständlich höhere oder niedrigere Konzentrationen zur Erzielung besonderer Wirkungen verwendet
kann. Weiterhin ist es bekannt, daß die Konzentration eines gegebenen Aromamittels von der Natur des besonderen, zu aromatisierenden
Materials und von der Natur der anderen Bestandteile in einem gegebenen Präparat abhängt. Die Pyrimidine (i) können per se
durch direkte Zugabe in reinem Zustand oder - häufiger - durch
(zulässigen) Zugabe in Form einer Lösung in einem derzeit eßbaren/Lösungsmittel,

wie Äthanol, Dipropylenglykal oder Triacetin, oder in Mischung mit
anderen Aromabestandteilen in Form eines Aromapräparates verwendet
werden.
Die Pyrimidine (i) können nach üblichen Verfahren hergestellt
u/erden, die im folgenden mit den üblichen Abkürzungen genauer dargestellt
werden.
4-Acetylpyrimidin, 4-Propionylpyrimidin und 4-Isoualerylpyrimidin
wurden nach einem Verfahren gemäß dem folgenden Reaktionsschema I hergestellt:

a. 2,4-Dihydraxy-6-methylpyrimidin wurde nach dam Verfahren in
Org.Synth.Coll,, Bd. 2, 422 (1969), ausgehend von Äthylacetoacetat
und Harnstoff, hergestellt.
b. 2, 4-Dichlor-6-tnethylpyrimidin
45 g (0,36 Mol) 2,4-Dihydroxy-6-methylpyrimidin wurden mit Phosphoroxychlorid
in Aniuesenheit von Diäthylanilin behandelt. Nach
3-stündigem Erhitzen zum Rückfluß wurde die Reaktionsmischung abgekühlt
und das überschüssige Phosphoroxychlorid über Eis zersetzt; dann wurde die Mischung mit MaOH neutralisiert, mit einer
NaCl Lösung gesättigt und mit Äther extrahiert. Durch Abtrennen und Eindampfen der ätherischen Extrakte erhielt man einen Rückstand,
der zu 57 g der obigen Verbindung mit einem Kp.99 C./11 Torr destilliert vuurde.
c. 4-Methylpyrimidin
Das in Absatz b oben erhaltene Produkt wurde in 5Ü0 ecm Diäthyläther,
die eine äquivalente Menge Triethylamin enthielt, und 5 g Palladium-über-Tierkohle (5 %) gelöst und die Lösung 5 1/2
Stunden in einer VJasserstoffatmosphäre gerührt. Nach Filtrieren, Eindampfen und Destillieren erhielt man die obige Verbindung in
einer Ausbeute von etiua 80 % mit einem Kp. 35°C./12 Torr.
d. 4-Formylpyrimidinoxim
0,1 Mol 4-Methylpyrimidin wurde zu 100 ecm Äthanol zugefügt ,
die 3,5 g (0,1 Mol) HCl enthielten. Innerhalb von 60 Minuten uiurde eine Lösung aus 9 g (0,12 Mol) Äthylnitrit in 60 ecm Äthylalkohol
zugefügt, wobei die Reaktionsmischung auf 5°C. gehalten
wurde; dann iuurde 1 Stunde bei Zimmertemperatur gerührt. Das so gebildete 4-Formylpyrimidinoxim-hydrochlorid wurde abfiltriert,
in Wasser gelöst und die wässrige Lösung durch Zugabe von Na2CO,

neutralisiert. Durch Filtrieren und Trocknen erhielt man die obige
Verbindung mit einem F. von 154-1550C. in 71-^iger Ausbeute.
e. 4-Cyanpyrimidin
Eine Lösung aus 0,01 Mol des in Absatz d. oben hergestellten
Oxims, 0,012 Mol Triphenylphosphin, 0,01 Mol Tetrachlorkohlenstoff
und 0,01 Mol Triethylamin in 50 ecm 1,2-Dichloräthan wurde 2 Stunden
auf 6O0C. erhitzt. Die flüchtigen Komponenten wurden entfernt
und der Rückstand 4 Mal mit je 100 ecm Petroläther (Kp. 30-500C.)
extrahiert. Nach Eindampfen und Destillieren der organischen
Extrakte erhielt man 4-Cyanpyrimidin in 61-/oiger Ausbeute.
f. 4-Acetylpyrimidin
Eine Lösung aus 0,05 Mol Methylbromid in 15 ecm Äther wurde zu
0,04 g-Atom Magnesiumspänen in 15 ecm trockenem Äther zugefügt und die erhaltene Lösung bis zum vollständigen Lösen des anwesenden
Magnesiums zum .Rückfluß erhitzt (30 Minuten). Dann wurde die Lösung auf -15°C. abgekühlt und eine Lösung aus 0,025 Mol 4-Cyanpyrimidin
20 ecm Äther zugefügt. Weiter wurden der Reaktionsmischung 50 ecm Äther zugefügt, und es wurde 30 Minuten bei -100C.
und dann 30 Minuten bei Zimmertemperatur gerührt.
Dann wurde die Mischung auf 100 g Eis gegossen, mit konz. H2SO.
angesäuert und 15 Minuten bei Zimmertemperatur gerührt. Nach Neutralisation
mit.festem Natriumcarbonat, ständiger Extraktion mit Äther und Eindampfen der ätherischen Extrakta erhielt man einen *
Rückstand, der durch präparative Gas-chromatographie unter Verwendung
einer CARBOWAX Kolonne 2OM gereinigt wurde; so erhielt man 4-Acetylpyrimidin in 53-^iger Ausbeute.

Eine Lösung aus 405 g (2,5 Mol) Äthytorttoacetat und 295 g (2,5 Mol)
Athylacetoacetat in 500 ecm Methylalkohol wurde 15 Minuten zum
Rückfluß erhitzt, wobei ein ständiger Strom aus gasförmigem Ammoniak durch die Lösung geleitet ujurde. Nach Abdampfen des Lösungsmittels
uiurde das gebildete Pyrimidin durch Zugabe von 500 ecm
Äther ausgefällt, wodurch man 190 g der obigen Verbindung in 61,3-Ausbeute
erhielt.
Die weiteren Stufen zur Umwandlung des erhaltenen 4-Hydroxy-2,6-dimethylpyrimidins
in das gewünschte Acylpyrimidin erfolgten nach den oben in Stufen b. bis f. beschriebenen l/erfahren·

Haselnuß', geröstet, getreideartig, weniger "Körper" als
3. 2-Acetylpyrazin 1,0 tierisch, weniger fleischig
und geröstet als 1j fettig
4. 5-Methylchinoxalin 2,5 weniger fleischig und tierisch
als 1; Haselnuß, geröstet, leicht grün
Der obige Vergleichstest zeigt den grundsätzlichen Unterschied bezüglich des Aromacharakters zwischen 2-Methyl-4-acetylpyrimidin
und den Verbindungen 2 bis 4, die bekanntlich fleischartige Aromanoten
begünstigen sollen.
Durch Mischen der folgenden Bestandteile wurde eine rekonstituierte
Rindfleischbrühe hergestellt:
handelsübliches Fleischextrakt Mononatriumglutamat
50:50 Mischung aus Natriuminositati Natriurnguanilat
Natriumchlorid
Milchsäure
Wasser

Die so hergestellte Brühe wurde in 3 Teile von gleichem Volumen
geteilt. Zwei Anteile wurden mit 0,15 bzw. 0,30 ppm 2-Methyl-4-
acetylpyrimidin aromatisiert und von einem Cxpertengremium wie
folgt bewertet:
a. nicht aromatisierte Brühe milder Geschmack, fleischiger
Charakter, Protein, Gemüsegeschmack

Durch Mischen der folgenden Bestandteile u/urde eine künstliche
,Rindfleischbrühe hergestellt:

Mononatriumglutamat 9,00
hydrolysiertes Pflanzenprotein 11,00
Karamellfarbe 1,20
gemahlener u/eißer Pfeffer 0,15
Zuiiebelpulv/er 0,15
Zucker 4,75
Weizenmehl ' 19,50
Maisstärke '; 13,50
insgesamt 80,00
130 g der so erhaltenen Mischung wurde unter Rühren zu 1 1 heißem Wasser zugefügt und 2 Minuten gekocht. Die erhaltene Soßenmischung
wurde in 3 Teile gleichen Volumens geteilt· Zu/ei derselben wurden
mit 0,30 bzui. 0,50 ppm 2-Methyl-4-acetylpyrimidin aromatisiert
und durch Vergleich mit der nicht aromatisieren Brühe probiert. Das Expertengremium beu/ertete die Aromaqualität der geprüften
Nahrungsmittel wie folgt:
a. nicht aromatisierte Brühe eüß, fruchtig, typischer Protein-
hydro Iysatgeschmack
b. aromatisierte Brühe deutlicherer fleischiger Charak-(0,30 ppm) terj geröstst>

Durch Mischen dar folgenden Bestandteile wurde eine Praline-Füllung
für Schokolade hergestellt:

insgesamt 201,1
Das so hergestellte Nahrungsmittel wurde in 3 Teile gleichen Volumens
geteilt, von denen zwei mit 0,30 bzw. 0,50 ppm 2-Methyl-4-acetylpyrimidin
aromatisiert und u/ie folgt bewertet wurden:
a. nicht-aromatisierte Schokolade süß, mild gerösteter Hasalnuß-
charakter
b. aromatisierte Schokolade typischer NuBcharakter (0,30 ppm)
c. aromatisierte Schokolade stärker geröstet als a; (0'50 ppm) Nußcharakter
Kaffeearoma tisierung
3,2 g handelsübliches Sofort-Kaffeepulver wurden in etwa 240 ecm
siedendem Wasser gelöst und das so erhaltene Getränk in zwei Anteile
gleichen Volumens geteilt, von denen einer mit 2-Methyl-4-acetylpyrimidin
in einer Konzentration von 0,075 ppm aromatisiert wurde; das so erhaltene, aromatisierta Getränk wurde durch l/er- gleich
mit nicht-aromatisiertem Kaffee organoleptisch ausgewertet.
Geschmack und Aroma des aromatisieren Getränkes hatten, wia festgestellt
wurde, einen deutlicher gerösteten Charakter sowie ein merklicheres Nußaroma.

Tabakaromatisierung
0,3 g einer 1°/oo Lösung aus 2-Methyl-4-acetylpyrimidin in 95 %
Äthanol wurde auf 100 g einer Tabakmischung vom Typ "American
Blend" gesprüht und der so aromatisierte Tabak zur Herstellung
von Testzigeretten verwendeten, deren Rauch durch Vergleich mit
nicht-aromatisierten Kontrollzigaretten organoleptisch ausgewertet
wurde. Der zur Herstellung der Kontrollzigaretten verwendete Tabak war vorher mit 95 % Äthanol behandelt worden. Das
Expertengremium erklärte, daß der Geschmack der Testzigeretten mehr "body" und einen deutlicheren, an den Geschmack von Haselnüssen
erinnernden Aromacharakter hatte. Beispiel 7 bis 10
Die Auswertung der Aromaeigenschaften verschiedener Acylpyrimidine
erfolgte in\ klarem Quellu/asser, wobei die folgenden Ergebnisse
erzielt wurden«
süß, leicht fruchtig, karamell fruchtig, ätherisch, leicht blumig leicht fruchtig
a. 4-Acetylpyrimidin
b. 4-Propionylpyrimidin
c. 2-Methyl-4~propionyl-
pyrimidin
d. i-Methyl-A-isovalerylpyrimidin

1,- Verbindungen der Formel:[T:-R21 2in welcher R für ein Wasserstoffatom steht und R einen Isobutyl-1 2rest bedeutet oder R für einen Methylrest steht und R einen Methyl-, Äthyl- oder Isobutylrest bedeutet. - 2.- 2-Methyl-4-acetylpyΓimidin.
- 3.- 2-^/tethyl-4-pΓopionylpyrimidin.
- 4.- 4-Isovalerylpyrimidin.
- 5.- 2-Methyl-4-iso\/alerylpyriinidin.6,- Verfahren zum Verstärken, Verbessern oder Modifizieren der Aromaeigenschaften, von Lebensmitteln, Getränken, Tiernahrung, pharmazeutischen Präparaten und Tabakprodukten, dadurch gekennzeichnet, daß man diesen Materialien eine geringe, jedoch wirksame Menge mindestens einer Verbindunq der Formel:R1C-R2
