REVENDICATION 4-Acétyl-2-méthylpyrimidine.
Au cours de la dernière décennie, nombreuses ont été les publications relatives à l'utilisation de certains dérivés hétérocycli-ques, notamment azotés, en tant qu'agents aromatisants. Parmi ces composés, les pyrazines ont retenu une attention particulière de la part des aromatiseurs, leur emploi étant déterminant pour la reconstitution de compositions de nature variée (voir à cet effet: «Fenaroli's Handbook of Flavor Ingrédients», CRC Press, Inc., Cleveland (1975), vol. II, p. 692 et ss.; brevets britanniques Nos 1156472,1156475 et 1156484). Certains dérivés hétérocycliques appartenant à la classe des pyridines, des pyrroles, ainsi qu'à celle des thiazoles ont également été décrits comme possédant des propriétés gustatives intéressantes (voir par exemple: brevets britanniques Nos 1156483,1156482 et 1156485).
A ce jour cependant, aucune mention n'a été faite quant à la possibilité d'utiliser des composés pyrimidiques comme ingrédients aromatisants.
Nous avons maintenant découvert de façon surprenante que les composés de formule:

dans laquelle R représente un reste alkyle contenant de 1 à 6 atomes de carbone, possédaient des propriétés organoleptiques fort intéressantes et que, de ce fait, ils pouvaient être employés très avantageusement dans l'industrie des arômes.
A l'état pur, les composés de formule I servent à développer des notes gustatives variées, telles des notes de type animal, grillé, rôti ou gras. Leur caractère douceâtre et caramel rappelle le goût particulier de la viande. C'est en effet dans la reconstitution d'arômes synthétiques destinés à l'aromatisation de produits à base de viande, ou imitant la viande, que les composés de l'invention trouvent une application préférentielle. Cependant, en mélange avec d'autres ingrédients aromatisants, les composés de formule I peuvent également servir à l'aromatisation d'aliments variés ainsi que de boissons en général, de préparations pharmaceutiques et du tabac. A titre d'exemple, on peut citer l'aromatisation du chocolat, du café, de boissons à base de lait, des yaourts, des desserts, des produits de pâtisserie et boulangerie.
Ces propriétés aromatisantes sont particulièrement marquées pour la 4-acétyl-2-méthylpyrimidine.
La présente invention a donc pour objet la 4-acétyl-2-méthyl-pyrimidine à titre de composé nouveau.
Les proportions dans lesquelles le composé de l'invention peut développer des effets aromatisants intéressants peuvent varier dans une gamme de valeurs très étendue. De façon préférentielle, on utilisera des concentrations comprises entre environ 0,02 et 20 ppm (parties par million) en poids par rapport au poids total de l'aliment aromatisé. De telles concentrations, cependant, ne peuvent être considérées restrictivement et des valeurs situées au-delà de la limite supérieure indiquée ci-dessus peuvent être employées, notamment lorsque des effets aromatisants spéciaux sont recherchés.
La 4-acétyl-2-méthylpyrimidine peut être synthétisée conformément à la méthode illustrée ci-dessous:

La méthode de synthèse suivie est décrite en détail dans l'exemple donné ci-après (températures en degrés centigrades).
Exemple
1. Diéthylacétal d'x-formylacétate d'èthyle
On active 105 g ( 1,6 atome-gramme) de copeaux de Zn (par traitement à l'HCl 1 % et lavages t uccessifs à l'eau, méthanol, puis acétone et chauffage à 100°) que l'on couvre de 25 ml de toluène absolu et additionne d'un cristal d'iode. Sous bonne agitation et chauffage à reflux, on y ajoute en 1 h une solution de 134 g (0,8 M) d'a-bromacétate d'éthyle et de 144 g (0,97 M) d'orthoformiate d'éthyle dans 300 ml de toluène absolu. Une surveillance de l'agitation est nécessaire vu la prise en masse du mélange réactionnel (gomme noire). On poursuit le chauffage à reflux durant 6 h et sous bonne agitation. Après refroidissement, on verse sur 500 g de glace, on dissout le précipité par addition de 60 ml d'acide acétique glacial, filtre le Zn excédentaire et extrait la phase aqueuse en continu par
CH^Wgl l'éther. Les phases organiques sont lavées avec 100 ml d'une solution aqueuse saturée d'hydrogénocarbonate de sodium, puis séchées, filtrées et évaporées. Par distillation, on élimine les produits de
55 départ n'ayant pas réagi (Eb10 <70°) et collecte 39 g de fraction principale (Eb10:83-85°), liquide incolore contenant 2 produits (Chromatographie en phase gazeuse, colonne Apiezon L., 125°).
"2. $-Ethoxyacrylate d'éthyle
6o On chauffe progressivement jusqu'à 175° un mélange de 39 g du mélange obtenu ci-dessus et 0,6 g d'acide p-toluènesulfonique. L'alcool formé distille lentement (1 h), puis le produit est distillé sous pression réduite: Eb10:76-78° - Obtenu 27 g
65 3. 4-Hydroxy-2-méthylpyrimidine
A une solution de 0,5 g (21,7 milli-atomes-grammes) de sodium dans 10 ml d'éthanol absolu, on ajoute 1,9 g (20,1 mM) de chlorhydrate d'acétamidine et agite durant 15 min. Après refroidissement

à 0", on ajoute 2,9 g (20,1 mM) de ß-ethoxyacrylate d'éthyle et agite durant 2 h à cette température. On filtre sur Supercell, lave le support à l'êther et évapore les filtrats à sec. On ajoute 10 ml de soude aqueuse 10% et agite à reflux durant 1 h. Après refroidissement, on ajoute à pH 6 2 ml d'acide acétique glacial, sature avec NaCl, extrait avec 3 x 10 ml de chloroforme, sèche, filtre et évapore le solvant. On obtient ainsi 2 g de masse pâteuse. Par recristallisation et dissolution dans 5 ml d'éthanol, puis précipitation par 40 ml d'éther et filtration, on obtient une masse solide qui, par lavage à l'éther, donne un produit ayant :211-214°.
4. 4-Chloro-2-méthylpyrimidine
On chauffe à reflux durant 45 min 7,5 g (69 mM) de 4-hydroxy-2-méthylpyrimidine et 46 ml d'oxychlorure de phosphore. On refroidit, décompose sur la glace, neutralise par addition d'hydrogé-nocarbonate de sodium (230 g) et extrait à l'éther. On sèche, purifie sur norit et évapore le solvant. Le produit est ensuite purifié par sublimation à 45° sous une pression de 5 x 10~3 Torr, pour fournir une poudre cristalline blanche, 4,7 g (rdt:53%), F:51-56 '.
5. 4-Cyano-2-méthylpyrimidine a) Chlorure de 2-mèthyl-4-pyrimidinyltriméthylammonium
A une solution de 1,3 g (10 mM) de 4-chloro-2-méthylpyrimidine dans 7 ml de toluène, refroidie dans la neige carbonique, on ajoute lentement 2 ml (22 mM) de triméthylamine anhydre et laisse agiter durant 24 h à la température ambiante en récipient fermé. La solution se trouble progressivement. Après addition de 50 ml d'éther, on filtre le sel, le lave à l'éther et le sèche sous vide. (Obtenu 1,53 g de poudre rose, Fdéc : 183-208°.)
b) 4-Cyano-2-mèthylpyrimidine
On chauffe à 90° un mélange de 3,5 g (53,8 mM) de cyanure de potassium et 6 g (0,1 M) d'acétamide. A ce mélange fondu, on ajoute en 15 min, par petites portions, 1,3 g (10 mM) du chlorure obtenu 5 suivant a ci-dessus. On agite durant 30 min à 90°, refroidit, dissout dans 20 ml d'eau, extrait en continu à l'éther. Une purification au norit, puis un séchage et une évaporation fournissent 0,4 g d'un liquide brun. Après distillation, on obtient 0,33 g de liquide incolore (rdt: 27%), F:22-24°.

6. 4-Acètyl-2-méthylpyrimidine
A 0,18 g (7,4 atomes-grammes) de Mg dans 3 ml d'éther absolu, on ajoute 1,4 g (9,86 mM) d'iodure de méthyle et 3 ml d'éther et chauffe à reflux jusqu'à dissolution complète du métal (15 min). A ce 15 réactif, refroidi à —15°, on ajoute en l'espace de 30 min une solution de 0,59 g de 4-cyano-2-méthylpyrimidine dans 10 ml d'éther. Après 30 min d'agitation à cette température, puis 30 min à la température ambiante, on verse le mélange sur 25 ml d'eau glacée, acidifie par addition de 1 g de H2S04 concentré, puis agite durant 15 min à la 20 température ambiante. Après neutralisation par addition de carbonate de sodium solide, on extrait en continu à l'éther, puis sèche et évapore le solvant. Par distillation, on obtient 0,49 g (73%) de liquide jaunâtre (Eb j 0:85-90°).
