Description
Die Erfindung betrifft neue ß-Rezeptoren-Blocker. Im besonderen
betrifft die Erfindung ß-adrenergische Blocker, welche sich zur Therapie oder Prophylaxe von Angina pectoris,
Herzarrhythmie und Bluthochdruck eignen.
Die ß-adrenergischen Blocker stellen eine Klasse von Wirkstoffen
dar, welche sich für die Therapie von Herzarrhythmien und Angina pectoris eignen. Das Kennzeichen von
Angina pectoris ist eine ungenügende Myokardsauerstoffzufuhr
trotz höherem Sauerstoffbedarf. Der erhöhte Säuerstoff-

bedarf bei körperlichen Anstrengungen wird durch die adrenergischen
ß-Rezeptoren-Stimulantien, Epinephrin und Norepinephrin,
ausgelöst. Dieselben adrenergischen Amine führen auch zu zahlreichen
Arrhythmien während oder nach einem Myokardialinfarkt. Die Blockierung der ß-adrenergischen Stimulantien
durch ß-adrenergische Blocker mildert viele Arten von Arrhythmien und verringert den Sauerstoffbedarf des Herzens
ι wodurch eine ausgeprägte Schmerzlinderung und erhöhte Toleranz gegenüber körperlichen Anstrengungen bei Angina
pectoris erzielt wird.
Die erfindungsgemäßen ß-Rezeptoren-Blocker sind die Verbindungen

Die Verbindungen der Erfindung enthalten asymmetrische Kohlenstoffatome
und können daher als optisch aktive Isomere oder als racemische Gemische auftreten.
Spezielle Beispiele für als ß-Rezeptoren-Blocker wirksame
Verbindungen sind:

Die Verbindung 5-[2-Hydroxy-3-(3»4-dimethoxyphenäthylamino)-]-propoxy-3»4-dihydröcarbostyril
hat sich auch als kardioselektiv erwiesen.
Die erfindungsgemäßen Verbindungen können generell durch Umsetzung
des entsprechenden Amins mit 5-(2,3-Epoxy)-propoxy-3,4-dihydrocarbostyril
hergestellt werden. 5-[2-Hydroxy-3-(3»4-dimethoxyphenäthylamino)-]-propoxy-3

Das Epoxid wird durch Umsetzung von 5-Hydroxy-3»4-dihydroearbostyril
[Y. Tamura et al., Chem. Ind., (1970), 1435] mit Epichlorhydrin in Dimethylformamid in Gegenwart von
Matriumhydrid hergestellt. Diese Methode liefert das
Epoxid in "besserer Qualität als das von K. Nakagawa et al.
in Journal of Medicinal Chemistry* .17. >
(1974)» 529, beschriebene Verfahren. 3»4-Dimethoxyphenäthylamin ist im
Handel erhältlich.
4-(2-Aminoäthoxy)-benzamid kann nach einem der nachstehenden
Verfahren aus handelsüblichen Ausgangsverbindungen unter Anwendung herkömmlicher Synthesetechniken hergestellt

Die nachstehenden Beispiele erläutern die Synthese und
pharmakologische Wirksamkeit der neuen Verbindungen. Sie
gehen jedoch nicht die einzigen Synthesemöglichkeiten wieder und sollen die Erfindung in keiner Weise beschränken.
Herstellung von 5-[2-Hydroxy~3-(3»4-dimethoxyphenäthylamino)-]-propoxy-3»4-dihydrocarbostyril
Ein Gemisch von 1,27 g (0,007 Mol) frisch destilliertem 3»4-Dimethoxyphenäthylamin und 1,5 g (0,0068 Mol) 5-(2,3-Epoxy)-propoxy-3»4-dihydrocarbostyril
[K. Nakagawa et al., J. Med. Chem., JT7, (1947), 529] in 35 ml Methanol wird
3 Std. unter Rückfluß gekocht. Anschließend dampft man das Lösungsmittel ab, reibt den öligen Rückstand mit absolutem
Äthanol/1ther an und filtriert einige Verunreinigungen ab.
Man säuert das Eiltrat mit ätherischer Salzsäure an, trennt das gummiartige Hydrochlorid ab, löst es in Wasser und
extrahiert mit Ithylacetat. Die wäßrige Schicht wird un-' ter vermindertem Druck zur Trockene eingedampft. Durch UnH-kristallisation
des Rückstands aus absolutem Äthanol erhält man die Analysenprobe vom Ep. 177 bis 1800Q; Rf 0,58
(Methanol/CHCl-, 1:9).

5-[2-Hydroxy-3-(4-carbamoylphenoxyäthylamino) - ]-propoxy-3»4-dihydrοcarbostyril
Ein Gemisch von 0,5 g (0,0028 Mol) 4-(2-Aminoäthoxy)-benzamid und 0,52 g (0,0024 Mol) 5-(2,3-Epoxy)-propoxy-3»4-dihydrocarbostyril
in 25 ml absolutem Äthanol wird 72 Std. unter Rückfluß gekocht und anschließend heiß filtriert.
Der Rückstand (0,8 g; Fp. 180 bis 1840C), welcher ein
Dimeres enthält, wird durch präparative Dünnschichtchromatographie gereinigt. Man erhält 406 mg der reinen
Verbindung vom Pp. 207 bis 2090C; Rf 0,57 (Methanol/Chloroform;
3:7).

Die Struktur wird durch das Kernresonanz- und Massenspektrum
bestätigt.
Das als Ausgangsverbindung benötigte 4-(2-Aminoäthoxy)-benzamid wird durch 8- bis 10-stündiges Erhitzen von 45 g
4-(2-Bromäthoxy)-benzoesäureäthylester mit" 250 ml flüssigem Ammoniak in einem Autoklaven auf 100 bis 1200C hergestellt.
Diese Umsetzung liefert

folgt hergestellt:
20 g (O»122 Mol) wasserfreies 5-Hydroxy-3»4-dihydrocarbostyril
[Y. Tamura et al», Chem. Ind., (1970), 1435] werden in einer StickstoffatmoSphäre unter Rühren in eine Suspension
einer 50 ^igen NaH-Dispersion (6,14 g; 0,128 Mol) in
110 ml wasserfreiem Dimethylformamid eingetragen. Anschliessend fügt man bei 300C 33,8 g (0,366 Mol) Epichlorhydrin in
Form von Teilmengen hinzu. ¥enn die anfängliche Reaktion nachläßt, erhitzt man das Gemisch 2 Std. auf 650C, rührt
es anschließend über Nacht bei Raumtemperatur, filtriert und dampft das FiItrat im Vakuum ein« Wenn sich ein Feststoff
auszuscheiden beginnt, verdünnt man das Gemisch mit absolutem Äthanol und filtriert. Dabei erhält man 10,5 g
des Produkts vom Fp. 164 bis 1680C, dessen durch Dünnschichtchromatographie
bestimmte Reinheit 96 $ beträgt. Das äthanolische FiItrat wird eingedampft. Der gummiartige
Rückstand wird mehrmals mit Wasser gewaschen und mit Äthanol überschichtet. Durch Filtration erhält man 0,6 g
derselben Substanz; Gesamtausbeute 11,1 g (42 $). Die Verbindung
ist identisch mit jener, die bei dem von K. Uakagawa
et al., J. Med. Chem., V7, (1974), 529, beschriebenen Verfahren anfällt.

5-[2-Hydroxy-3-(3»4-methylendioxyphenäthylamino)-]-propoxy-3,4-dihydrocarbo3tyril
Ein Gemisch von äquivalenten Mengen 5-Hydroxy-3»4-dihydrocarbostyril
und 3,4-Methylendioxyphenäthylamin-hydrochlorid
[j. Org. Chem., 2J5, (1958), 1982] in 30 ml absolutem Äthanol,
das 2 ml Triäthylamin enthält, wird 5 Std. unter Rückfluß gekocht.
Die dabei erhaltene klare Lösung wird unter vermindertem Druck zur Trockene eingedampft. Man reibt die entstehende
pastöse Masse mit absolutem Äthanol an, filtriert und sammelt den Rückstand (710 mg; Cl positiv). Das Piltrat
wird mit ätherischer Salzsäure angesäuert und zur Trockene eingedampft. Man reibt den erhaltenen Rückstand mit absolutem
Äthanol an und filtriert. Der 3?ilterrückstand (230 mg) wird mit dem zuvor erhaltenen Filterrückstand (710 mg) vereinigt.
Man löst die Gesamtheit in warmem Wasser, extrahiert mit Äthylacetat und verwirft den organischen Extrakt. Die
wäßrige Schicht wird zur Trockene eingedampft, wobei das Produkt zurückbleibt. Durch Umkristallisation aus Wasser
erhält man die Verbindung vom Pp. 242 bis 2440C (Zersetzung).

Man verabreicht die gemäß Beispiel 1 bis 3 hergestellten Verbindungen intravenös (i.v.) anästhesierten Hunden, um
ihre Wirksamkeit als ß-Rezeptoren-Blocker nachzuweisen.
Man verabfolgt die Verbindungen den anästhesierten Hunden in unterschiedlichen Dosen, um die prozentuale Hemmung der
Wirkung von Isoproterenol auf den Blutdruck und die Herzfrequenz der Hunde zu bestimmen. Die Testverbindungen
sind:

Die in der Tabelle zusammengestellten Werte zeigenf daß
die Verbindungen I, II und III wirksame ß-Rezeptoren-Blocker darstellen. Aus den Werten geht ferner hervor,
daß die Verbindung I, d.h. 5-[2-Hydroxy-3-(3»4-diinethoxyphenäthylamino)-]-propoxy-3»4-dihydrocarbostyril
kardioselektiv ist. Dies bedeutet, daß die Verbindung bevorzugt auf den Herzmuskel einwirkt und bei empfindlichen Personen
keine Asthmaanfälle verursacht. Die Kardioselektivität der Verbindungl wird dadurch angezeigt» daß die Hemmung
der Wirkung von Isoproternol auf den Blutdruck im Verlauf der Zeit rasch den Wert null erreicht, während die Inhibierung
der Wirkung auf die Herzfrequenz wesentlich langer andauert.
