indol-hydrochlorid als Wirkstoff.
3. Pharmazeutische Zusammensetzungen nach Anspruch 1, gekennzeichnet durch 5-Methoxy-3-(4'-piperidyl)-indol-hydrochiorid
als Wirkstoff.
Die Erfindung betrifft pharmazeutische Zusammensetzungen, die mindestens ein Piperidylindol-Derivat
der Formel 1 J >

R Wasserstoff oder Methoxy und
Ri und Rj zugleich oder unabhängig voneinander Wasserstoff
oder Methyl bedeuten,
und/oder mindestens ein Additionssalz der Piperidylindol'Derivate
mit pharmazeutisch geeigneten Säuren als Wirkstoff enthalten.
Beispiele für die Additionssalze mit pharmazeutisch geeigneten Säuren sind etwa Salze mit Bromwasserstoffsäure,
Jodwasserstoffsäure, Schwefelsäure, Phosphorsäure, Essigsäure, Benzoesäure, Maleinsäure, Fumarsäure,
Bernsteinsäure, Weinsäure, Glyoxylsäure, Asparaginsäure, Alkansulfonsäuren, Arylsulfonsäuren
sowie insbesondere mit Chlorwasserstoffsäure.
Zu den erfindungsgemäß verwendeten Piperidylindol-Derivaten
der Formel I gehören insbesondere:
3-(4'-Piperidyl)-indnl,
l-Methyl-3-(4'-piperidyl)-indol,
6-Methoxy-3-(4'-piperidyl)-indol,
5-Methoxy-3-(4 -piperidyl)-indi)l und
6-Methoxy-2-methyl-3-(4'-piperidyl)-indi
l-Methyl-3-(4'-piperidyl)-indol,
6-Methoxy-3-(4'-piperidyl)-indol,
5-Methoxy-3-(4 -piperidyl)-indi)l und
6-Methoxy-2-methyl-3-(4'-piperidyl)-indi
sowie die entsprechenden Additionssalze mit pharmazeutisch geeigneten Säuren, insbesondere
die entsprechenden Hydrochloride.
Die Piperidylindol-Derivate der Formel I sowie deren Salze weisen außerordentlich interessante pharmakologischen
Eigenschaften auf; sie besitzen insbesondere bemerkenswerte antidepressive und antiemetische
Eigenschaften sowie Anti-Parkinson-Wirkung. .
Aufgrund dieser Eigenschaften können die erfindungsgemäßen Zusammensetzungen als Medikamente
beispielsweise bei der Behandlung psychischer Störungen, Verhaltensstörungen, Charakterstörungen od. dgl.
sowie bei der Behandlung akineüscher Zustände verwendet werden.
Die übliche Dosis hängt vom verwendeten Produkt, der behandelten Person sowie der vorliegenden
Erkrankung ab und beträgt beispielsweise 5 bis 500 mg pro Tag bei oraler Verabreichung der Humanmedizin.
Die Derivate der Forme! ί sowie ihre Additionssalze
mit pharmazeutisch geeigneten Säuren können dabei in pharmazeutischen Zusammensetzungen vorliegen, die
zur parenteralen Verabreichung oder zur Verabreichung über den Verdauungstrakt bestimmt sind.
Derartige pharmazeutische Zusammensetzungen können beispielsweise fest oder flüssig sein und in
üblicherweise in der Humanmedizin verwendeten pharmazeutischen Formen vorliegen, beispielsweise als
einfache oder dragierte Tabletten bzw. Dragees, Gelatinekapseln, Granulate, Suppositorien oder etwa
injizierbare Präparationen, die nach üblichen Verfahren herstellbar sind. Der oder die Wirkstoffe können dabei
in üblicherweise bei pharmazeutischen Zusammensetzungen verwendete Excipientien eingebracht werden,
beispielsweise in Talk, Gummiarabicum, Lactose, Stärke, Magnesiumstearat, Kakaobutter, wäßrige oder
nichtwäßrige Träger, Fettmaterialien tierischen oder pflanzlichen Ursprungs, Paraffinderivate, Glycole, verschiedene
Netzmittel, Dispergiermittel oder Emulgatoren sowie etwa Konservierungsmittel.
Einige der Piperidylindol-Derivate der Formel I können nach der BE-PS 8 02 912 hergestellt werden.
Weitere Derivate sind in der Fachliteratur beschrieben.
Die Herstellung der erfindungsgernäß verwendeten Wirkstoffe wird im folgenden anhand von Beispielen
näher erläutert.
Beispiel 1
3-(4'-Piperidyl)-indol-hydrochIorid
3-(4'-Piperidyl)-indol-hydrochIorid
12 g 3-(4'-Piperidyl)-indol werden in 70 ml Methanol suspendiert. Das Gemisch wird anschließend in einem
Eisbad abgekühlt, worauf langsam eine gesättigte methanolische Lösung von Chlorwasserstoffsäure bis zu
pH 1 (etwa 20 ml) zugegeben wird. Man läßt darauf 30 min bei 0°C kristallisieren, filtriert, spült mit
Methanol und trocknet im Vakuum. Es werden 123 g des rohen Hydrochloride erhalten, das in 160 mg
Methanol am Rückfluß gelöst wird.
Anschließend wird auf etwa 100 ml aufkonzentriert und 3h bei Raumtemperatur sowie lh bei 0"C
kristallisieren gelassen. Anschließend wird filtriert, mit Methanol gespült und im Vakuum getrocknet. Es
werden 8,4 g 3(4'-Piperidyl)-indol-hydrochlorid in Form eines cremefarbenen Produkts erhalten; F. 2280C.

1 -Methyl-S^'-piperidylJ-indol-hydrochlorid

26 g l-Methyl-3-(4'-piperidyl)-indol werden in 210 ml
Äthylacetat gelöst Man kühlt mit Eis ab, fügt unter Rühren 30 ml mit Chlorwasserstoffsäure gesättigtes
Äthylacetat zu und läßt im Eisbad kristallisieren.
Anschließend wird filtriert und mit Äthylacetat gewaschen. Es werden 19,2 g Hydrochlorid erhalten, das
kalt und warm in Äthanol am Rückfluß gereinigt wird. Es werden 12 g i-MethyI-3-(4'-piperidyI)-indol-hydrochlorid
in Form farbloser Kristalle erhalten; F. 264°C.

%N Beispiel 5 6-Methoxy-3-(4'-piperidyl)-indol-hydrochlorid
9,7 g 6-Methoxy-3-(4'-piperidyl)-indol werden in ml heißem Isopropanol gelöst Anschließend werden
ml einer mit Chlorwasserstoffsäure gesättigten Isopropanollösung zugesetzt.
Darauf wird in der Wärme wieder gelöst filtriert und aufkonzentriert Nach dem Abkühlen wird mit Eis
gekühlt, abgesaugt, mit Isopropanol gewaschen und im Vakuum getrocknet. Es werden 9,5 g 6-Methoxy-3-(4'-piperidyl)-indol-hydrochlorid
in Form farbloser Kristalle erhalten; F. 213° C.

10 g 5-Methoxy-3-(4'-pipe'idyl)-indol werden in
100 ml Methanol gelöst Anschließr-.d wird mit Eis
abgekühlt wonach 30 ml mit Chlorwasserstoffsäure gesättigtes Äthylacetat sowie anschließend langsam
100 ml Äthylacetat zugesetzt werden, worauf im Eisbad kristallisieren gelassen wird. Anschließend wird filtriert
und mit Methylacetat gewaschen. Es werden 11 g Hydrochlorid erhalten, das kalt und warm in Äthanol am
Rückfluß gereinigt wird. Es werden 7,5 g 5-Methoxy-3-(4'-piperidyl)-indol-hydrochlorid
in Form farbloser Kristalle erhalten; F. 217°C.
Analyse: CMH,9C1N2O
Beispiele für pharmazeutische Zusammensetzungen
1. Es wurden Tabletten folgender Formulierung hergell

12,3 g 6-Methoxy-2-methyl-3-(4'-piperidyl)-indol werden in 200 ml Äthanol gelöst Man kühlt mit Eis ab, setzt
unter Rühren und Aufrechterhalten einer Temperatur wi
unterhalb 1O0C eine äthanolische Chlorwasserstoffsäurelösung
bis pH 1 zu.
Anschließend wird filtriert, mit Äthanol gewaschen und im Vakuum getrocknet. Es werden 12 g Hydrochlorid
erhalten, das kalt und heiß in Äthanol am Rückfluß "">
gereinigt wird. Es werden 7,2 g 6-Methoxy-2-methyl-3-(4'-piperidyl)-indol-hydrochlorid
in Form beigefarbener Kristalle erhalten: F. 252°C.
Es wurden Tabletten folgender Formulierung hergestellt:
- I -Methyl-3-(4'-piperidyl)-i7idolhydrochlorid 25 mg
= Excipiens auf 1 Tablette ad 200 mg
(Excipiens: Lactose, Stärke,Talk, Magnesiumstearat).
Es wurde eine injizierbare Lösung folgender Formulierung hergestellt:
- 5-Methoxy-3-(4'-piperidyl)-indolhydrochlorid 25 mg
- Excipiens, wäßrig, steril ad 2 ml
6. Es wurden Tabletten folgender Formulierung hergestellt:
- 6-Methoxy-2-methyI-3-(4'-piperidyl)-indol-hydrochlorid 25 mg
- Excipiens auf 1 Tablette ad 200 mg
(Excipiens: Lactose, Stärke, Talk,
Magnesiumstearat).

Pharmakologische Untersuchungen
Die pharmakologische Wirkung folgender Substanzen wurden im einzelnen untersucht:
3-(4'-Piperidyl)-indo5-hydrochlorid (Verbindung A), 6-Methoxy-2-methyl-3-(4'-piperidyl)-indol (Verbindung
B),
1 -Methyl-3-(4'-piperidyl)-indoI-hydrochlorid (Verbindung
C),
5-Methoxy-3-(4'-piperidyl)-indol-hydrochlorid
(Verbindung D) und
(Verbindung D) und
6-Methoxy-2-methyl-3-(4'-piperidyl)-indoI-hydrochlorid (Verbindung E).
1. Test zur Potentialisierung der
Amphetamin-Stereotypien
Amphetamin-Stereotypien
Die Versuche wurden an Gruppen zu 5 münnlichsn
Ratten von 150 bis 180 g vorgenommen. Jedes Tier wurde allein in einen Gitterkäfig (29 χ 25 χ 17 cm)
gesetzt, der einige Holzspäne enthielt
Zwischen der Verabreichung der untersuchten Verbindung und der Injektion von 5 mg/kg Dexamphetamin-sulfat
wurde 1 h gewartet.
Das Verhalten der Tiere wurde anschließend 5 h lang jede '/2 h nach einem von Halliwell et al (Brit. J.
Pharmacol. 23 [1964] 330-350) angegebenen Bewertungsverfahren festgehalten.
Eingeschlafenes Tier (0); erwacht, jedoch unbeweglich
(1); bewegt sich im Käfig (2); beschnüffelt den Deckel (3); leckt die Wände ab (4); berührt die Späne
oder Gitterstäbe des Käfigs mit den Zähnen (5); beißt die Späne oder die Gitterstäbe des Käfigs (6).
Die Intensität der Stereotypien wurde in Form einer zwischen 0 und 30 liegenden Bewertung ausgedrückt,
die der Summe der bii den fünf Ratten einer Gruppe zu
jeder Zeit erhaltenen Werte entspricht. Die Summe der in 5 h erhaltenen Bewertungen wurde berechnet.
Die Verbindungen wurden intraperitoneai verabreicht; die Verbindungen A, C, D und E lagen dabei in
wäßriger Lösung, die Verbindung B in wäßriger Suspension vor.
Die Dosis der Verbindungen, die die Summe der Bewertungen in 5 h um etwa 100% erhöht, beträgt
20 mg/kg für Verbindung A, 5 mg/kg für Verbindung B, über 20 mg/kg für Verbindung C, etwa 1 mg/kg für
Verbindung D sowie etwa 5 rng/kg für Verbindung E.
2. Test des Antagonismus gegenüber der
Prochlorpemazin-Katalepsie
Prochlorpemazin-Katalepsie
Die Versuche wurden an Gruppen zu fünf männlichen Ratten von etwa 100 g durchgeführt
Die untersuchten Verbindungen wurden intraperitoneai oder oral zugleich mit einer Dosis von 15 mg/kg
Prochlorpemazin, das intraperitoneai gegeben wurde, verabreicht.
ίο Die Katalepsie wurde 7 h jede Stunde nach dem
Kreuztest der Pfoten der gleichen Seite nach Boissier und Simon (vgl. Boissier und Simon, Therapie 18 [1963]
1257—1277) unter folgender Auswertung ermittelt:
Das Tier weigert sich gegen ein Oberkreuzen der Vorderpfoten mit den Hinterpfoten der jeweils gleichen
Seite (0); es akzeptiert ein derartiges Überkreuzen lediglich an einer Seite (0,5); es akzeptiert das
Überkreuzen auf beiden Seiten (1).
Die Verbindungen A, C und D lagen dabei in wäßriger Lösung vor.
Die Verbindung A wirkt der -aiit Prochlorpemazin
induzierten Katalepsie ab einer Dosis *on 03 mg/kg bei
intraperitonealer und ab einer Dosis von 2 mg/kg bei oraler Darreichung entgegen.
Die Verbindung C wirkt der mit Prochlorpemazin indizierten Katalepsie ab einer Dosis von 10 mg/kg bei
intraperitonealer Verabreichung entgegen, die Verbindung D ab einer Dosis von 2 mg/kg bei intraperitonealer
Verabreichung.
3. Antiemetische Wirkung
Der Antagonismus gegenüber durch Apomorphin hervorgerufenem Erbrechen wurde bei Hunden untersucht
(vgl. Chen und Ensor, J. Pharmac. Exp. Therap. 93
[1959] 245-250).
Die Anzahl der durch eine subkutane Injektion von 0,1 mg/kg Apomorphin-hydrochlorid hervorgerufenen
Erbrechvorgänge wurde bei jedem Tier 8 Tage vor dem Versuch ermittelt
Die untersuchte Verbindung wurde in wäßriger Lösung subkutan in variablen Dosen 1^h vor dem
Aponiorphin-hydrochlorid verabreicht.
Die Verbindung A antagonisiert das durch Apomorphin hervorgerufene Erbrechen ab einer Dosis von
0,5 mg/kg bei subkutaner Verabreichung.
4. Ermittlung der akuten Toxität
Die akute Toxität wurde an Gruppen von 10 Mäusen von etwa 20 g Körpergewicht bestimmt, denen steigende
Dosen der untersuchten Verbindung intraperitoneai und oral verabreicht wurden.
Die Mortalität wurde 48 h nach der Verabreichung der Verbindung ermittelt
Die 50-%-LetaIdosen (DL 50) der Verbindung A betrugen bei intraperitonealer Verabreichung 95 mg/kg
bzw. bei oraler Verabreichung 200 mg/kg.
Die 50-%-Letaldosen (DL 50) der Verbindungen C, D und E betrugen bei intraperitonealer Verabreichung
60 mg/kg, 100 mg/kg bzw. 95 mg/kg.
