Die Erfindung betrifft Mittel für die Behandlung von Krebs und malignen Neoplasmen sowie die Herstellung der
darin enthaltenen Wirksubstanz. Sie zeigt auch die Anwendung solcher· Mittel.
Gemäß der Erfindung enthält ein Mittel zur Behandlung von Krebs oder malignen Tumoren einen Koordinationskomplex von
Platin(II) mit einem Phenylendiamin, der die folgende Struktur
besitzt:

worin der sechsgliedrige Ring ein aromatischer Kern und R^.,
Rp, R5, und R vorzugsweise Wasserstoff oder niederes Alkyl
sind, jedoch allgemein Wasserstoff, Alkyl-, Aryl-, Aralkyl-, Alkaryl-, Halogen-, Pseudohalogen-, Nitro-, Amido-, Amino-,
Hydroxy-, Formyl-, Carbonsäure-, Carbonsäuresalζ-, SuIfonsäure-
oder Sulfonsäure sal zgruppen oder mit den soeben genannten Gruppen substituiertes Alkyl, Aryl, Aralkyl oder Alkaryl
bedeuten können, und worin X und Y halogenoide Gruppen sind, die gleich oder verschieden sein können, jedoch vorzugsweise
beide Chlorid oder ein anderes Halogenid oder ein Pseudohalogenid, insbesondere Cyanid, Cyanat, Thiocyanat,
Azid oder Ozid bedeuten.
Unter dem Ausdruck "Halogenoid" sind Halogen und Pseudohalogenverbindungen
zu verstehen.
_cos-Dichloro-diammin-platin(Il) ist eine bekannte antineoplastische
Verbindung mit einem therapeutischen Index von 8,1 Dichloro-(o-phenylendiammin)-platin(II) und Dichloro-(4,5-dimethyl-o-phenylendiammin)-platin(II)
haben entsprechende therapeutische Indices von 20,4· bzw. mehr als 56,7. Die
biologischen Daten für diese Verbindungen und chemisch ähnliche, Jedoch strukturell verschiedene Verbindungen sind zu
Vergleichszwecken nachfolgend angegeben.

Diese Ergebnisse zeigen die Aktivität für die Platinverbindungen beim ADJ/PCö-Plasmazelltumor. '

Die Versuche wurden an weißen Mäusen BALB/C unter Verwendung
von 3 Testmäusen für jede Dosierung und einem Minimum von 6 unbehandelten Kontrollmäusen durchgeführt.
Ein ADJ/PCö-Plasmazelltumor, der von einer getöteten Maus
entnommen ist, wird frei von überflüssigem Gewebe seziert und unter sterilen Bedingungen in Stücke von etwa 10 mg
Größe geschnitten. Die Gewebestücke werden dann nach der Trocar-Methode subkutan in die linke Achselregion einer
anderen Maus implantiert. Die Mäuse waren durchschnittlich etwa 7 Wochen alt und wogen 18 bis 20 g. Wenn man den Tag 0
als den Tag der Behandlung nimmt (24 Tage nach der Tumorimplantation), wurden die Tiere am Tag 10 getötet. Die Tumo
ren wurden herausgeschnitten und gewogen und das Verhältnis der Tumorgewichte in den behandelten Tieren zu dem des
Kontrollsatzes der Tiere wurde bestimmt.
Bei den Testtieren wurden die Verbindungen am Tag 0 in öl
als Träger intraperitoneal injiziert. Das Injektionsvolumen
betrug 1 ml pro 100 g Körpergewicht.
Die EDg0 ist die Minimaldosis, die eine 90 %ige Regression
in der Tumorgröße hervorruft. LD ist die Dosierung, welche
bei 50 % der Tiere den Todesfall hervorruft. Der therapeutische
Index ist LDcq/EDqq und ist ein Maß der Selektivität
der fraglichen Verbindung bei der Behandlung des Tumors. ID/3 bedeutet einen Tod bei 3 Tieren, 3D/3 bedeutet 3 Todesfälle
bei 3 Tieren etc.
Diese Ergebnisse zeigen, daß die Verbindungen gemäß der Erfindung gute Ergebnisse hinsichtlich der Antitumoraktivitat
zeigen und sehr günstig im Vergleich mit chemisch ähnlichen, jedoch strukturell verschiedenen Verbindungen abschneiden.

Es sei bemerkt, daß das 4,5-Dimethyl-o-phenylendiamminderivat
einen beträchtlich besseren therapeutischen Index und geringere Toxizität zeigt, als der unsubstituierte
o—Phenyl endi amminkomp 1 ex .
Wesentliche Anwendung der Erfindung ist also die Behandlung von Krebs oder malignen Tumoren mittels der erfindungsgemäßen
Verbindungen der Struktur:
worin X, Y, E^, E?, E^ und E^ die oben angegebenen Bedeutungen
besitzen bzw. mit Mitteln, welche solche Verbindungen als Wirksubstanz in pharmazeutisch üblichen Trägern und
gegebenenfalls Hilfsstoffen und Exzipientien enthalten.
Dichloro-(o-phenylendiammin)-Pt(II) wurde auch beim Walker-Tumor
untersucht. Es hat einen therapeutischen Index für diesen Tumor von 3 verglichen mit einem Index von 2 für cis-Dichlorodiammin-Pt(II).
Die zur Prüfung gegen den Walker-Tumor angewandte Arbeitsweise ist die gleiche,wie sie für
den PCe-Plasmazelltumor oben beschrieben ist, mit der Ausnahme,
daß Chester-Beatty-Wistar-Eatten verwendet wurden,
und die Behandlung einen Tag nach der Tumortransplantation begann.

Herstellung der Platinkomplexe
Dichloro-(o-phenylendiammin)-platinClI)
0,456 g o-Phenylendiammin-dihydrochlorid wurden zu einer
filtrierten Lösung von 1 g KpPtCl2, in 50 ml 1n HCl, die
sich in einem zum Lichtausschluß geschwärzten Kolben befanden,
gegeben. Die Lösung wurde 1 Tag bei 50 0C gehalten.
Die gebildeten braungelben Nadeln wurden abfiltriert, mit Wasser, Aceton und Äther gewaschen und im Vakuum getrocknet,
was eine Ausbeute von 80 % ergab.

Der Dichloro-(4,5-dimethyl-o-phenylendiammin)-platin(lI)-komplex
kann in analoger Weise hergestellt werden.
Der Ausdruck Alkyl, der hier verwendet wird, bedeutet vorzugsweise niedermolekulares Alkyl mit 1 bis 6 Kohlenstoffatomen,
insbesondere 1 bis 4 Kohlenstoffatomen, wobei Methyl und Äthyl besonders bevorzugt sind. Der Ausdruck
Aryl bedeutet vorzugsweise Phenyl. Diese Definition gilt auch für Substituenten der Gruppen R. bis E^ sowie für den
Alkyl-oder Arylteil von Aralkyl und Alkaryl. Unter dom
Ausdruck Carbonsäure bzw. Carbon säur es al ζ oder Sulfonsäurc
bzw. SuIfonsäuresalz ist vorzugsweise nur die Carboxyl-,
Carboxylat-, Sulfonsäure- bzw. SuIfonatgruppe zu verstehen.

- Pat entansprüche/ Tj Koordinationskomplexe von Platin(II) der Strukturworin H^ bis IL, die gl-eich oder verschieden sein können, Wasserstoff, Alkyl-, Aryl-, Aralkyl-, Alkaryl-, Halogen-, Pseudolialogen-, Nitro-, Amido-, Amino-, Hydroxy-, IOrmyl-, Carbonsäure-, Carbonsäuresalζ-, Sulfonsäure- oder SuIfonsäuresalzgruppen oder mit solchen Gruppen substituiertes Alkyl, Aryl, Aralkyl oder Alkaryl bedeuten, und worin X und T halogenoide Gruppen sind, die gleich oder verschieden sein können.
- 2. Platinkomplexe nach Anspruch 1, dadurch gekennzeichnet , daß X und Y identisch sind.
- 3. Platinkomplexe nach Anspruch 1 oder 2, dadurch gekennzeichnet , daß X und Y beide Halogen sind.

- 5· Platinkomplexe nacli Anspruch 3, dadurch gekennzeichnet , daß X und Y Pluorid, Bromid oder Jodid bedeuten.
- 6. Platinkomplexe nach Anspruch 1, dadurch gekennzeichnet , daß X oder Y eine Pseudohalogengruppe bedeuten·
- 7. Platinkomplexe nach Anspruch 1, dadurch gekennzeichnet, daß X und Y beide Pseudohalogengruppen bedeuten.
- 8. Platinkomplexe nach Anspruch 6 oder 7» dadurch gekennzeichnet , daß als Pseudohalogengruppe Cyanid, Cyanat, Thiocyanat oder Azid vorliegt.
- 9· Platinkomplexe nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet , daß alle vier Gruppen R Wasserstoff bedeuten.
- 10. Platinkomplexe nach Anspruch 1 bis 8, dadurch gekennzeichnet , daß wenigstens zwei Gruppen E Methyl oder Äthyl sind.
- 11. Platinkomplexe nach Anspruch 10, dadurch gekennzeichnet , daß H2 und R3, Methyl gruppen sind.
- 12. Pharmazeutisches Mittel zur Behandlung von Krebs bzw. malignen Tumoren oder Neoplasmen bei Mensch und Tier, gekennzeichnet durch einen Gehalt an mindestens einer Verbindung gemäß Anspruch 1 bis 11 als Wirksubstanz zusammen mit einem üblichen pharmazeutischen Träger.309845/1U113- Mittel nach Anspruch 12, gekennzeichnet durch einen für die orale, parenterale oder intraperitoneale Verabreichung geeigneten Träger sowie gegebenenfalls üblichen Hilfsstoffen und/oder Exzipientien.14·. Verfahren zur Herstellung von Dichloro-(o-phenylendiamminplatin(II) oder Dichloro-(4,5~dimethyl-o-phenylendiamminplatin(II), dadurch gekennzeichnet , daß man o-Phenylendiammin-dihydrochlorid oder 4-,5-Dimethyl-ophenylendiammin-dihydrochlorid zu einer filtrierten Lösung eines Alkalichlorplatinats in Salzsäurelösung in einem zum LichtausSchluß abgedeckten Gefäß zugibt, und das erhaltene Gemisch wenigstens 12 Stunden bei erhöhter Temperatur, insbesondere im Bereich von 50 0C, hält.309845/ 1141
