Während Herstellungsverfahren fur heterocyclische

Beide Verfahren gehen von verhältnismäßig schwer zugänglichen Ausgangsprodukten aus, deren Synthesen
mehrere Reaktionsschritte erfordern. Darüber hinaus sind beide Synthesen für die Herstellung von
Aza-azulen mit funktionalen Gruppen im 7gliedrigen
stickstoffhaltigen Ring ungeeignet.
Das erfindungsgemäße Verfahren ist dadurch gekennzeichnet, daß man ein 1,2-DiacyIcyclopentadien-Derivat
der allgemeinen Formel II
wurde an der Ratte als LD50 nach Miller und
Tainter, Proc, Soc, Exper. Bio), Med. 57,261,1944,
bestimmt. Zum Vergleich ist das gut wirksame, im Handel befindliche Antiphlogistikum Phenylbutazon
angegeben. Das Ergebnis zeigt die folgende Tabelle.

Die Herstellung von Verbindungen der Formel II
ist in den Patentschriften 1 104 955 und 1 105 411 beschrieben.
Aza-azulene mit einem Stickstoffatom im 7g!iedrigen Ring des Bicyclus sowie deren funktionell substituierten
Derivaten kommt im Hinblick auf zahlreiche in neuerer Zeit als hochwirksame Psychopharmaka bekanntgewordene
Azepin- und Diazepinderivate ein besonderes Interesse zu.
Die erfindungsgemäße Verbindung zeichnet sich darüber hinaus durch eine hohe antiphlogistische
Wirksamkeit aus.
Die erfindungsgemäße Verbindung 6-Dimeihylamino-5-aza-azulen wurde am Carrageenin-Ödem
der Rattenpfote (Domenjozu. Mitarb.. Arch.exp.
Pharm. Path. 230,325,1957. und W i η t e r u. Mitarb..
J. Pharmacol, exp. Therap. 141. 369. 1963) auf anti phlogistische Wirkung geprüft. Die akute Toxizitä«

Die Erfindung wird in den nachstehe;^ auitefuhrten
Beispielen weiter erläutert:
In einem lOO-ml-Kolben. der durch ein kurzes, gewinkeltes
Übergangsstück mit einer auf —60 C gekühlten Falle verbunden ist. wird bei 1 Torr das gut
verriebene Gemisch aus 2,1 g (KJmMoI) 6-Dimethvlamino-fulven-2-(N.N-dimelhyla!dimrnonium)-chlorid
und 1.2 g (lOmMol) N.N- Dimethylacetamidin - hydrochlorid
mit freier Flamme erhitzt. Dabei geht ein orangegelbes Destillat über, aus dem nach chromatographischer
Reinigung an Aluminiumoxyd (basisch. Akt.-St. II) mit Äther 210 mg (12.2% der Theorie)
bei 99 C schmelzendes 6-Dimethylamino-5-aza-azulen
erhalten werden. Die Konstitution des Produktes wird durch Elementaranaiyse. Molekulargewichtsbestimmung
sowie durch UV- und NMR-^pektren bewiesen.

Pikrat des 6-Dimethylamino-5-aza-azulens
Man versetzt eine Lösung von 50.0 mg (0.29 m.Mol) 6-Dimethyl-amino-5-aza-azulen in 30 ml Methanol
mit einer Lösung von 65.0 mg (0.29 mMol) Pikrinsäure in 30 ml Methanol. Das bei —50 C ausfallende
P'krat wird aus Methanol umkristallisiert. Man erhält 95.5 mg (85% der Theorie, bezogen auf eingesetzte
Pikrinsäure) gelber Kristalle, die sich ab 180 C
allmählich zersetzen.

In einem durch ein Calciumchlorid-Rohr verschlossenen 100-ml-Kolben werden 1.5 g HOmMoI) 6-Dimethylamino-fulven-2-aldehyd und 1,2 g (10 mMol) N.N-Dimethyl-acetamidin-hydrochlorid im Ölbad langsam erwärmt. Bei 90 C erhält man eine klare
Schmelze, die sich allmählich dunkel färbt. Man er hitzt die Schmelze 2 Stunden auf HXJ bis 120 C. löst
s erkaltete Reaktiansprodukt in Methanol, gießt
β Lösung in 250 ml Wasser und schüttelt fünfmal it Äther aus. Die vereinigten Ätherlösungen werden
it Wasser gewaschen und über Magnesiumsulfat !trocknet. Nach Entfernen des Lösungsmittels im
»kuum erhält man einen braunen Rückstand, aus
weisen.
