haben, bei 100 bis 3000C umsetzt.
Es ist seit langem bekannt, daß polycyclische Ringverbindungen in Naturprodukten vorkommen. Viele
dieser polycyclischen Verbindungen, insbesondere diejenigen mit verschmolzenen Ringen, haben physiologische
Anwendungen. Die Synthese derartiger Ringstrukturen ist im allgemeinen schwierig. So gelang bisher
die Herstellung von in 4-Stellung substituierten Derivaten der Bicyclo[2,2,2]-okt-2-en-l-carbonsäure nur
auf Umwegen.
Demgegenüber stellt die Erfindung einen großen technischen Fortschritt dar, da durch sie ein einstufiges
Verfahren zur direkten Herstellung von Bicyclooktenen aus leicht erhältlichen Ausgangsmaterialien verfügbar
wird. Zwar wurde die Umsetzung von äthylenisch ungesättigten Verbindungen mit einem verhältnismäßig
W) stabilen, festen, am Ring perhalogenierten Pyron bereits
beschrieben (US-PS 30 92 641), jedoch verleiht die Halogenierung dem Ring Eigenschaften, die wesentlich
anders sind als die Eigenschaften eines gemäß der Erfindung benutzten nichthalogenierten Pyronrings. Zur
Herstellung von Verbindungen ähnlich den erfindungsgemäß hergestellten mit einem halogenfreien Pyronring
nach bekannten Verfahren war aber bisher eine mehrstufige Synthese erforderlich.
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Bicyclo[2,2,2]-okt-2-en-l-carbonsäuren und
deren Estern der allgemeinen Formel

in welcher einer der Reste R1 oder R2 einen Carboxy-
oder niederen Alkoxycarbonylrest und der andere dieser Reste einen gesättigten aliphatischen Ci —Ce-Kohlenwasserstoffrest
oder einen aromatischen oder heteroaromatischen Rest mit 5 bis 10 C-Atomen und R3 und R4
Wasserstoff bedeuten, wobei wahlweise in dem Fall, daß der Rest R2 ein Ci—Cg-gesättigter aliphatischer Kohlenwasserstoffrest
ist, einer der Reste R3 oder R4 auch ein von aliphatischen Kohlenstoff-Kohlenstoff-Mehrfachbindungen
freier Ci-Cs-Kohlenwasserstoff rest sein
kann. Das Verfahren ist dadurch gekennzeichnet, daß man Äthylen mit einem «-Pyron der allgemeinen Formel

bei 100 bis 300° C umsetzt
Von den nach dem Verfahren gemäß der Erfindung erhaltenen Verbindungen werden solche bevorzugt, in
denen sowohl R3 als auch R4 Wasserstoff sind, und worin R2 einen niederen Alkyl (Ci-C4)-, Phenyl- oder Pyridylsubstituenten
bedeutet, der über eines seiner Ringkohlenstoffatome gebunden ist
Da gesättigte aliphatische Kohlenwasserstoffreste mit 6 oder weniger C-Atomen, vorzugsweise mit 1—4 C-Atomen,
billiger und leicht verfügbar sind, werden sie als Reste R2 bevorzugt, wenn diese gesättigte Ci-Cs-Kohlenwasserstoffreste
sein sollen.
Als »aromatisch« bzw. »heteroaromatisch« gelten im Rahmen der Beschreibung carbocyclische und heterocyclische
Verbindungen von aromatischem Charakter.
Wenn R2 ein aromatischer oder heteroaromatischer Rest sein soll, wird es aus Gründen der Verfügbarkeit des
Pyrans und unter Berücksichtigung der Anwendungsmöglichkeiten der Produkte bevorzugt, daß der cyclische
Anteil 6 Ringatome enthält, und daß nicht mehr als ein > Heteroatom im Ring vorhanden ist. Der aromatische
oder heteroaromatische Rest R2 ist über eines seiner Ringkohlenstoffatome an das Brückenkopfkohlenstoffatom
gebunden, und die Gesamtzahl der Kohlenstoffatome in dem(n) Ring(en) ist mindestens 5 und nicht ι ο
höher als 10. Der aromatische Rest ist vorzugsweise a) Phenyl, und zwar entweder unsubstituiert oder mit
einem oder zwei Substituenten aus der folgenden Gruppe substituiert: Alkyl, Halogen, Alkoxy, Hydroxy,
halogeniertes Alkyl, Cyan, Nitro, Carboxy und Alkoxy- i j
carbonyl; oder (heteroaromatisch) b) Pyridyl, und zwar entweder unsubstituiert oder mit einem der folgenden
Reste substituiert: Alkyl, Halogen, halogeniertes Alkyl und Alkoxy.
Das Verfahren gemäß der Erfindung kann unter Druck _>
<> durchgeführt werden. Der angewendete Druck ist abhängig von den eingesetzten Verbindungen und kann
bis zu 5000 bar und mehr betragen. Im allgemeinen liegt er im Bereich von 50 bis 3000, vorzugsweise im Bereich
von 75—1000 bar. Die Reaktionstemperatur beträgt 2>
etwa 100-300°C, vorzugsweise 125-275°C. Die Reaktionsdauer ist verschieden je nach den angewendeten
Bedingungen. Vorzugsweise werden höhere Temperaturen bei niedrigeren Drücken angewendet, um die
Reaktionszeit zu verkürzen. Vorzugsweise werden x> Zeiten von 0,5—24 Stunden angewendet, wobei
10—20 Stunden besonders bevorzugt werden.
Da die eingesetzten Reaktionsteilnehmer Kohlenstoff-Kohlenstoff-Doppelbindungen
enthalten, können Hydrochinon, Phenthiazin oder ähnliche freie Radikal r> bildende Inhibitoren in geringen Mengen (Spurenmengen)
vorhanden sein, um Nebenreaktionen, insbesondere Polymerisationen, zu verhindern.
Die durch die Reaktion gebildeten Bicyclooktenalkylcarboxylate lassen sich leicht in üblicher Weise
isolieren. Unter normalen Raumbedingungen sind sie niedrigschmelzende Feststoffe. Sie haben niedrige
Flüchtigkeit Die so hergestellten Carboxylate können dann durch übliche Esterhydrolysereaktionen in die gesättigten
freien Carbonsäuren umgewandelt werden. Zur 4 > Herstellung der in 2-Stellung ungesättigten freien Carbonsäuren
wird man am besten unmittelbar von dem entsprechenden carboxy-substituierten a-Pyron ausgehen,
wie erfindungsgemäß vorgesehen.
Die gemäß der Erfindung hergestellten Verbindungen, ~>o
in denen der Rest R1 eine niedere Alkylestergruppe und der Rest R2 ein gesättigter aliphatischer Ci-Cs-Hydrocarbylrest
ist, haben einen beständigen, angenehmen, fruchtartigen Geruch. Diese neuen Verbindungen sind
wertvoll für die Formulierung von Riechstoffen oder als ■-,■-> Aromastoffe, wo ein fruchtiger z. B. ein traubenartiger
Geruch und Geschmack gewünscht wird. Diese wertvolle Eigenschaft und die sich daraus ergebende
Verwendung stehen im deutlichen Gegensatz zu bekannten analogen Verbindungen, z. B. solchen, bei denen die t>o
Reste R2, R3 und R4 sämtlich Wasserstoff darstellen.
Diese Verbindungen haben einen starken, unangenehmen Geruch und sind demgemäß völlig ungeeignet für
die Anwendungszwecke, für die die gemäß der Erfindung
hergestellten Verbindungen wertvoll sind. Wenn Carb- t»
oxygruppen in beiden Stellungen R1 und R2 vorhanden sind, haben die Verbindungen praktisch keinen Geruch.
Die gemäß der Erfindung hergestellten Verbindungen sind ferner geeignet für die Herstellung von 1-Aminobicyclooktanen
durch übliche Reaktionen.
Verschiedene Ausführungsformen der Erfindung sind ausführlicher in den folgenden Beispielen beschrieben.
Beispiel 1
4-Methyl-bicyclo[2,2,2]-okt-2-en-1 -äthylcarboxylat
4-Methyl-bicyclo[2,2,2]-okt-2-en-1 -äthylcarboxylat
In einem Druckgefäß wurden 53 g 6-Methyl-<x-pyron-3-äthylcarboxylat,
1 g Hydrochinon und 80 cm3 Benzol mit Äthylen etwa 10 Stunden auf eine maximale
Temperatur von 180° C unter einem Druck von 3000 bar
erhitzt. Das Druckgefäß wurde gekühlt, der gasförmige Inhalt vorsichtig abgeblasen, das Benzol entfernt und das
flüssige Produkt destilliert, wobei 4-Methylbicyclo[2,2,2]-okt-2-en-l-äthylcarboxylat
erhalten wurde.

Analyse (Fraktion 3) für C12H18O2:

Diese Verbindung wurde unter Verwendung von Palladiumkohle als Katalysator in Methanol bei einem
Wasserstoffdruck von etwa 2,1 kg/cm2 hydriert, wobei 4-Methyl-bicyclo[2,2,2]-oktan-1 -äthylcarboxylat vom
Schmelzpunkt 22,5—23,50C erhalten wurde. Diese Verbindungen
ergab durch Erhitzen am Rückflußkühler mit methanolischem Natriumhydroxyd und anschließendes
Ansäuern 4-Methy!bicyclo[2,2,2]-oktan-1 -carbonsäure vom Schmelzpunkt 194—195°C.
Anlagerung von Äthylen an
ö-Äthyl-a-pyron-S-äthylcarboxylat
ö-Äthyl-a-pyron-S-äthylcarboxylat
Das in Beispiel 1 beschriebene allgemeine Verfahren wurde wiederholt mit der Ausnahme, daß 6-Äthyl-
«-pyron-3-äthylcarboxylat bei einer Reaktionszeit von
14 Stunden verwendet wurde, wobei 4-Äthylbicyclo-[2,2,2]-okt-2-en-l-äthylcarboxylat
vom Schmelzpunkt 120—130°C/13 mm Hg erhalten wurde. Die Hydrierung
dieser Verbindung ergab 4-Äthylbicyclo[2,2,2]-oktan-äthylcarboxylat,

Anlagerung von Äthylen an
6-n-Propyl-a-pyron-3-äthylcarboxylat
6-n-Propyl-a-pyron-3-äthylcarboxylat
Das in Beispiel 2 beschriebene allgemeine Verfahren wurde wiederholt mit der Ausnahme, daß 6-n-Propyla-pyron-3-äthylcarboxylat
ohne Zusatz eines Lösungsmittels verwendet wurde. Erhalten wurde 4-n-Propylbicyclo[2,2,2]-okt-2-en-l-äthylcarboxylat
vom Siedepunkt 112°C/2,2 mm Hg. Durch Hydrierung dieser Verbindung
wurde 4-n-Propylbicyclo[2,2,2]-oktan-l-äthyl-
carboxylat erhalten, das durch Verseifung und Ansäuerung 4-n-PropyIbicycIo[2,2,2]-oktan-l-carbonsäure vom
Schmelzpunkt 208—209° C ergab.
Analyse für Q2H20O2:
Berechnet: C 73,5, H 9,9;
gefunden: C 73,45, H 9,66.
gefunden: C 73,45, H 9,66.
ö-t-Butyl-a-pyron-S-äthylcarboxylat wurde wie folgt
hergestellt:
Zu einem Gemisch von 12 g (0,5MoI) Natriumhydrid
in 350 cm3 Äthylenglykoldimethyläther wurde unter Rühren ein Gemisch von 50 g (0,5 Mol) Pinakolon
und 108 g (0,5MoI) Diäthyläthoxymethylenmalonat
tropfenweise innerhalb von 30 Minuten gegeben. Das
Anlagerung von Äthylen an ö-Isopropyl-a-pyron-S-äthylcarboxylat
Das in Beispiel 2 beschriebene allgemeine Verfahren wurde wiederholt mit der Ausnahme, daß 6-Isopropyl-«-
pyron-3-äthylcarboxylat verwendet wurde. Erhalten wurde 4- Isopropylbicyclof 2,2,2]-okt-2-en-1 -äthylcarboxylat
vom Siedepunkt 120—122°C/6 mm Hg. Durch Hydrierung dieser Verbindung wurde Isopropylbicyclo-[2,2,2]-oktan-l-äthyIcarboxylat
erhalten, das nach Verseifung und Ansäuerung 4-IsopropylbicycIo[2,2,2]-oktan-1-carbonsäure
vom Schmelzpunkt 210—212°C ergab.
Anlagerung von Äthylen an ö-n-Butyl-a-pyron-S-äthylcarboxylat
Das in Beispiel 2 beschriebene allgemeine Verfahren wurde wiederholt mit der Ausnahme, daß 6-n-Butyla-pyron-3-äthyIcarboxylat
verwendet wurde. Erhalten wurde 4-n-Butylbicyclo[2,2,2]-okt-2-en-1 -äthylcarboxylat.
Durch Hydrierung dieser Verbindung wurde 4-n- Butylbicyclo[2,2,2]-oktan-1 -äthylcarboxylat erhalten,
das nach Verseifung und Ansäuerung 4-n-ButyI-bicyclo[2,2,2]-oktan-l-carbonsäure
vom Schmelzpunkt 158 -158,5° C ergab.
Analyse für C13H22O2:
Berechnet: C 74,3, H 10,5;
gefunden: C 74,11, H 10,22.
Anlagerung von Äthylen an 6-tert.-Butyl-a-pyron-3-äthylcarboxylat
Das in Beispiel 2 beschriebene allgemeine Verfahren wurde wiederholt mit der Ausnahme, daß 6-t-Butyl-
«-pyron-3-äthylcarboxylat verwendet wurde. Erhalten
wurde 4-t-Butylbicyclo[2,2,2]-okt-2-en-l-carboxylat. Durch Hydrierung dieser Verbindung wurde 4-t-Butylbicyclo[2,2,2]-oktan-l
-äthylcarboxylat erhalten, das nach Verseifung und Ansäuerung 4-t-ButylbicycIo[2,2,2]-oktan-1-carbonsäure
vom Schmelzpunkt 281—282,5° C ergab.
Analyse für C13H22O2:
Berechnet: C 74,3, H 10,5;
gefunden: C 74,46, H 10,16.
gefunden: C 74,46, H 10,16.
Gemisch wurde 3 Stunden unter Rühren am Rückflußkühler erhitzt und dann auf Raumtemperatur gekühlt.
Das überschüssige Natriumhydrid wurde mit Äthanol zerstört, und das Reaktionsgemisch wurde unter Rühren
in eine Lösung von 116 cm3 (120 g, 2MoI) Eisessig in
500 cm3 Eiswasser gegossen. Das Gemisch wurde mit Äther extrahiert Die Ätherextrakte wurden mit
gesättigter wäßriger Natriumchloridlösung gewaschen und mit wasserfreiem Magnesiumsulfat getrocknet
Nach Entfernung des Äthers bei vermindertem Druck wurden 132,28 g eines Öls erhalten, aus dem durch
Destillation 59,13 g (52%) 6-tert-Butyl-«-pyron-3-äthylcarboxylat als gelbes Öl vom Siedepunkt 125° C bei
0,25 mm, 130°C bei 0,5 mm, 145°C bei 0,8 mm und 148°C bei 1,0 mm Hg erhalten wurden.
Anlagerung von Äthylen an
6-Methyl-a-pyron-3-äthylcarboxylat
6-Methyl-a-pyron-3-äthylcarboxylat
Eine Reihe von Reaktionen zwischen 6-MethyI-a-pyron-3-äthylcarboxylat
und Äthylen wurde bei verschiedenen Temperaturen und Drücken durchgeführt Jede Reaktion wurde 16 Stunden in einem Schüttelrohr
ausgeführt. Das Reaktionsprodukt wurde mit Benzol aus dem Rohr gespült und die erhaltene Lösung unter vermindertem
Druck zu einem öl eingeengt. Das öl wurde in Äthanol gelöst und zur Entfernung von Polyäthylen
filtriert. Das Äthanol wurde dann durch Abdampfen unter vermindertem Druck entfernt. Bei jedem Versuch
ergab die Entfernung des Äthanols ein öl, das durch Vergleich seines Infrarotspektrums und seines Verhaltens
in der Gaschromatographie mit einer gemäß Beispiel 5 hergestellten authentischen Probe als 4-Methylbicyclo[2,2,2]-okt-2-en-l-äthylcarboxylat
identifiziert wurde. Die Ergebnisse des Versuchs sind in der folgenden Tabelle zusammengestellt.

Anlagerung von Äthylen an
e-Phenyl-a-pyron-S-äthylcarboxylat
e-Phenyl-a-pyron-S-äthylcarboxylat
Ein Gemisch von 60 g ö-Phenyl-a-pyron-S-carbonsäureäthylester
und 25 cm3 Benzol wurde mit Äthylen in einem Schüttelrohr auf 1000 bar gebracht, worauf die
Temperatur auf 180° C erhöht wurde. Der Druck im Rohr
betrug dann 3000 bar. Das Rohr wurde bei dieser Temperatur 7 Stunden geschüttelt. Während dieser Zeit fiel
der Druck auf 2920 bar. Das Rohr wurde mit Äthylen wieder auf 3000 bar aufgedrückt und die Reaktion 6 Stunden
fortgesetzt. Das Schüttelrohr wurde gekühlt und der Inhalt entnommen, mit 500 cm3 Äthanol verdünnt
und filtriert. Der Alkohol und das Benzol wurden abfiltriert, wobei 57 g 4-Phenylbicyclo[2,2,2]-okt-2-en-
1-äthylcarboxylat als weißer Feststoff vom Schmelzpunkt
44—45,5° C zurückblieben.
Anlagerung von Äthylen an
3-Phenyl-a-pyron-6-äthylcarboxylat
3-Phenyl-a-pyron-6-äthylcarboxylat
Ein Gemisch von 60 g S-Phenyl-a-pyron-e-äthylcarboxylat
(hergestellt durch Behandeln von 3-Phenyla-pyron-6-carbonsäure mit Diazoäthan (M. J. D.
van Dam, Rec. Trav. Chim. 83, S. 31 [1964]) und
25 ml Benzol wurden in einen 200 ml Schüttelautoklav gegeben und mit Äthylen auf 1000 bar gebracht. Die
Temperatur wurde stufenweise auf 1800C gebracht. Zusätzliches
Äthylen wurde periodisch aufgedrückt, um den Druck auf 2900 bis 3000 bar für eine Gesamtzeit von
13 Stunden zu halten. Der Autoklav wurde gekühlt und sein Inhalt mit 500 ml Äthanol verdünnt und filtriert. Die
Lösungsmittel wurden durch Destillation entfernt, wobei weißes, festes 4-Phenylbicyclo[2,2,2]-okt-2-en-l-äthylcarboxylat
zurückblieb, dessen Infrarotspektrum mit dem des Produkts von Beispiel 8 identisch war.
Ausbeute und Schmelzpunkt wie in Beispiel 8.
4-(4'-Pyridyl)-bicyclo[2,2,2]-okt-2-en-1 -äthylcarboxylat
Eine Lösung von 12 g 6-(4'-Pyridyl)-a-pyron-3-äthylcarboxylat in 50 cm3 Benzol wurde unter einem Äthylen druck
von 3000 bar 15 Stunden in einem Schüttelrohr bei 170° C gehalten. Das Rohr wurde gekühlt und durch eine
Falle abgeblasen, die mit festem Kohlendioxyd gekühlt war. Das Reaktionsgemisch und der Inhalt des Abscheiders
wurden in 11 absolutem Äthanol gelöst und zur Entfernung von polymerem Material filtriert. Das
Äthanol wurde durch Abdampfen unter vermindertem Druck entfernt, wobei 10,2 g eines Öls erhalten wurden,
das stehen gelassen wurde und hierbei zu einem niedrigschmelzenden Feststoff kristallisierte. Das Produkt bestand
aus der Verbindung 4-(4'-Pyridyl)-bicyclo[2,2,2]-okt-2-en-l-carbonsäureäthylester,
Fp. 59,5 bis 62,00C.
Das 6-(4'-Pyridyl)-a-pyron-3-äthylcarboxylat wurde wie folgt erhalten: Eine Suspension von 14 g Natriumhydrid
in 300 cm3 wasserfreiem Dimethylsulfoxyd wurde unter Rühren auf 60—700C erwärmt. Die Wasserstoffentwicklung
wurde mit einer Naßuhr gemessen. Nach Beendigung der Wasserstoffentwicklung wurde die
Lösung auf 25° C gekühlt. Unter Rühren wurden 61,5 g 4-Acetylpyridin und anschließend 108 g Diäthyläthoxymethylenmalonat
zugesetzt. Während dieser Zusätze wurde die Temperatur durch Kühlen mit einem Eisbad
κι
bei 20—25°Cgehalten. DasGemisch wurde 10— 15Stunden stehen gelassen und dann auf 500 g Eis gegossen,
das 35 cm3 Essigsäure enthielt. Durch Zusatz von Wasser wurde das Volumen auf 1500 cm3 gebracht. Ein dunkelroter
Feststoff kristallisierte aus. Dieser Feststoff wurde abfiltriert, mit Wasser gewaschen und getrocknet. Der
Feststoff wurde in Xylol suspendiert und unter einer Destillationskolonne mit Rückflußkühler bis zum Rückfluß
erhitzt. Das Äthanol wurde entfernt, solange es gebildet wurde. Das Xylol wurde dann bei vermindertem Druck
entfernt. Dann wurde der Kolbeninhalt gekühlt und der Kristallisation überlassen. Dieses rohe Material ergab
bei Umkristallisation aus Toluol reines 6-(4'-Pyridyl)-a-pyron-3-äthylcarboxylat
als gelben Feststoff vom Schmelzpunkt 156-1570C.
Die folgenden weiteren speziellen aromatisch substituierten Bicyclo[2,2,2]-okt-2-en-l-carbonsäuren und
Ester werden erhalten, wenn Äthylen und das entsprechende a-Pyron mit dem erforderlichen aromatischen
Substituenten nach dem beschriebenen Verfahren umgesetzt werden. Wenn das Produkt als Carbonsäure angegeben
ist, wurde es nach einem üblichen Esterhydrolysenverfahren oder sonst unmittelbar aus dem entsprechenden
carboxy-substituierten a-Pyron erhalten.
