dessen Salze und ein Verfahren zur Herstellung dieser Verbindungen, das dadurch gekennzeichnet ist, daß
man ein Phosphoran der allgemeinen Formel II
(R)3P=CH-CH2CH2-N N-R1
bis etwa 10O0C, mit dem Keton um und isoliert dann
das Produkt, indem man das Lösungsmittel z. B.
durch Destillation in einem Vakuum entfernt. Das Produkt kann durch Umkristallisation gereinigt und
gegebenenfalls in pharmazeutische Dosierungsformen umgewandelt werden.

in der R eine Alkylgruppe mit 1 bis 6 Kohlenstoffatomen, eine Phenyl-, Aminophenyl- oder Benzylgruppe
darstellt und R1 ein Wasserstoffatom oder eine 2-Hydroxyäthylgruppe bedeutet, mit einem Keton
der Formel III
(R)3P=CH-CH2-CH2-N NR1
in der R und R1 die vorstehende Bedeutung haben,
werden erhalten, indem man eine Suspension einer (III) 30 trisubstituierten Piperazinopropylphosphoniumverbindung
der Formel IV
umsetzt und gegebenenfalls das nicht substituierte Stickstoffatom des Piperazinringes durch Umsetzung
mit Äthylenoxid substituiert.
Nach einer besonderen Ausführungsform der Erfindung verwendet man ein Phosphoran der Formel II,
in der R eine Phenylgruppe bedeutet. Besonders erwähnt sei (3-Piperazinopropyliden- l)-triphenylphosphoran.
Die erfindungsgemäße Propylidenverbindung ist wertvoll als Monomeres, insbesondere — jedoch
nicht ausschließlich — wegen der C — C-Doppelbindung.
Im übrigen findet diese Propylidenverbindung Verwendung als therapeutisches Mittel.
Bekanntlich sind viele Aminopropylidenverbindungen wertvoll als psychotherapeutische Mittel. Sie
werden in der Chemotherapie von Geisteskrankheiten und insbesondere für die Behandlung von Depressionsoder Erregungszuständen verwendet.
Die erfindungsgemäße Verbindung ist besonders wertvoll als psychotherapeutisches Mittel. Sie erwies
sich gegenüber dem bekannten N-[3-(4-/S-Hydroxyäthyl-piperazino)-propyliden]-phenothiazin(Perphenazin)
in klinischen Versuchen als überlegen, über diese Versuche liegt ein ausführlicher Versuchsbericht vor.
Bei der Herstellung der erfindungsgemäßen Verbindung entstehen die eis- und trans-Isomeren und
die Säureanlagerungssalze dieser Verbindung. Das cis-Isomere ist besonders wertvoll.
Wie anschließend noch ausführlich durch das Beispiel erläutert wird, setzt man das trisubstituierte
Piperazinopropylidenphosphoran ausreichend lange bei einer geeigneten Temperatur, z. B. von etwa —10
(R)3Pe-CH2-CH2-CH2-N NR1
mit einer starken Base behandelt. Man kann die Reaktion in einem mit den Reaktionsteilnehmern
oder dem Produkt nicht reagierenden Medium, z. B. trockenem Tetrahydrofuran, durchführen. Geeignete
starke Basen sind z. B. Alkoholate oder Natriumhydrid oder metallorganische Verbindungen, z. B.
Butyllithium.
Bisher wurde vorgeschlagen, Aminopropylidenverbindungen mit psychotherapeutischen Eigenschaften
durch Behandlung eines Ketons mit einem Aminoalkylmagnesiumhalogenid als Grignardverbindung,
anschließende Hydrolyse und Dehydratisierung des dadurch entstandenen Carbinols herzustellen. Das
Grignard-Verfahren ist als Verfahren zur Herstellung von Aminopropylidenverbindungen bekannt. In
diesem Zusammenhang sei erwähnt, daß die USA.-Patentschriften 2 951082, 2 996 503, 3 046 283,
3 047 580 und 3 055 888 die Verwendung einer Grignardverbindung zeigen. Zu den Vorteilen des
erfindungsgemäßen Verfahrens zählen die Tatsachen, daß sich die Piperazinopropylidenverbindung erfindungsgemäß
einfacher herstellen läßt, als es mit der Grignardverbindung möglich ist. Das erfindungsgemäße
Piperazinopropylidendibenzoxepin kommt in zwei geometrisch isomeren Formen vor und eine
der beiden (eis und trans) Konfigurationen der Piperazinopropyliden-Seitenkette
ist pharmakologisch viel wirksamer. Nach dem erfindungsgemäßen Verfahren wird anscheinend die wirksamere Form erzeugt. Auf
der anderen Seite führt die Grignard-Reaktion mit
dem gleichen Keton zur Herstellung eines Gemisches, in welchem das Isomere mit geringerer Wirksamkeit
überwiegt. Das aus dem erfindungsgemäßen Verfahren stammende Reaktionsgemisch ist daher bedeutend
wirksamer als das aus dem Grignard-Verfahren stammende Reaktionsgemisch. Wird das Grignard-Reagenz
verwendet, so überwiegt das Isomer mit geringerer Wirksamkeit; wird das erfindungsgemäße
Reagenz verwendet, so überwiegt das wirksamere Isomer. Aus dem Grignard-Gemisch erhält
man das wirksamere Isomer durch das klassische und schwierige Verfahren der fraktionierten Kristallisation;
beim erfindungsgemäßen Verfahren ist eine fraktionierte Kristallisation nicht erforderlich.
Das erfindungsgemäße Verfahren wird wie folgt durchgeführt:
Es wird zunächst eine Lösung des trisubstituierten Piperazinopropylidenphosphorans hergestellt, indem
man das entsprechende trisubstituierte 3 - Piperazinopropylphosphoniumhalogenidhydrohalogenid
in einem wasserfreien, offenkettigen oder cyclischen Äther, z. B. Diäthyläther, Dimethoxyäthan oder Tetrahydrofuran,
suspendiert. Eine andere Möglichkeit besteht darin, Lösungsmittel wie Dimethylsulfoxyd zu
verwenden. Die Lösung wird dann je Mol des Phosphoniumhalogenid-hydrohalogenid mit 2 Mol eines
stark basischen Reagenz behandelt. Als stark basisches Reagenz lassen sich Natriummethylat, Kalium-t-Butylat,
Natriumhydroxyd, Natriumhydrid, oder Butyllithium verwenden.
In der nächsten Stufe versetzt man die Lösung mit etwa 80% einer stöchiometrisch äquivalenten Menge
des Ketons und läßt das Gemisch mehrere Stunden reagieren. Die Reaktionsgeschwindigkeit wird dadurch
gesteuert, daß man das Gemisch bei einer gewünschten Temperatur zwischen etwa —10 und etwa 100°C
hält. Dann werden etwa 10 Volumprozent Wasser zugesetzt, und das organische Lösungsmittel wird in
einem Vakuum entfernt. Der Rückstand wird mit einer Mineralsäure behandelt, bis er stark sauer ist
(etwa pH 2). Dann wird ein mit Wasser nicht mischbares organisches Lösungsmittel, vorzugsweise Benzol,
zugesetzt. Die Suspension wird gerührt, das Benzol wird abgetrennt und das übrige Gemisch, welches
das Produkt als unlösliches Hydrochlorid und Wasser enthält, wird mit Natriumhydroxydlösung alkalisch
gemacht, und die frei gewordene Base wird mit Benzol extrahiert. Der Benzolextrakt wird getrocknet, und
man erhält das Produkt durch Verdampfen des Benzols. Gegebenenfalls kann man die freie Base in das
Säureanlagerungssalz umwandeln.
Die erfindungsgemäß herstellbaren Salze sind im allgemeinen Säureanlagerungssalze. Zum Beispiel bildet
die neue Verbindung Salze mit Halogenwasserstoffsäuren, z. B. Hydrobromide und Hydrochloride,
mit Schwefelsäure, mit Phosphorsäure u. dgl. Außerdem lassen sich wertvolle Salze mit organischen
Säuren, z. B. mit Zitronensäure, Essigsäure, Milchsäure, Maleinsäure, Ameisensäure u. dgl. herstellen.
Das erfindungsgemäße Oxepin kann, wenn es nicht nach dem erfindungsgemäßen Verfahren hergestellt
wird, auch durch die folgenden Verfahren gebildet werden:
(a) Reaktion des entsprechenden Oxepin-11-ons mit einem Allylmagnesiumhalogenid nach Grignard,
anschließende Dehydratisierung der dadurch entstandenen 11-Hydroxyverbindung und
Aminierung der /S-olefinischen Bindung der Verbindung
mit einem entsprechend substituierten Piperazin oder
(b) Behandlung des Oxepin-11-ons mit einem entsprechenden
3-Piperazinopropin-(l) in Gegenwart eines Kondensationsmittels, wie Lithiumamid,
und anschließende Hydrierung und Dehydratisierung der Zwischenprodukte.
Bei diesen Verfahren erhält man jedoch nicht zum überwiegenden Teil das cis-Isomer wie nach dem
erfindungsgemäßen Verfahren. Da das cis-Isomer in dem erfindungsgemäß erhaltenen Gemisch vorwiegt,
läßt sich das reine cis-Isomere der neuen Verbindung nahezu frei von dem trans-Isomeren nach bekannten
Verfahren, z. B. durch fraktionierte Kristallisation erhalten.
Das folgende Beispiel dient zur Erläuterung des erfindungsgemäßen Verfahrens:
A. 1 l-(3-Piperazinopropyliden)-2-chlor-6,1 l-dihydro-dibenz[b,e]oxepin
Eine Suspension von 71,5 g 3-(l-Piperazinopropyl) - triphenylphosphoniumbromid - hydrobromid in
220 ecm trockenem Tetrahydrofuran wird während einer Stunde mit 120 ecm 2,62molaren Butyllithium
in Heptan und nach weiteren 30 Minuten mit 25,0 g 2-Chlor-6,l 1 -dihydro-dibenz[b,e]oxepin-11 -on versetzt.
Das Gemisch wird 15,5 Stunden am Rückfluß erhitzt. 500 ecm Wasser werden bei Raumtemperatur
zugesetzt, und das Lösungsmittel wird im Vakuum entfernt. Der rohe Rückstand wird mit 10%iger Salzsäure
behandelt, bis er sauer ist (pH 2), und dann werden 1,51 Benzol zugesetzt. Nach dem Rühren
trennt sich das Gemisch in drei Phasen (eine Produktphase, in der sich das unlösliche Hydrochloridsalz
befindet, eine wäßrige Phase und eine organische Phase).
Die Benzolschicht wird durch Dekantieren entfernt, und das übrigbleibende Gemisch wird mit 10%iger
Natriumhydroxylösung basisch gemacht und dreimal mit je 1500 ecm Benzol extrahiert. Die Benzolextrakte
werden gewaschen, dann mit wasserfreiem Natriumsulfat getrocknet und in einem Vakuum konzentriert.
Man erhält 33,8 g eines zähflüssigen Öls. Ein kristallines Dihydrochlorid dieser Verbindung, 2-Chlor-11
-(3-piperazino-propyliden)-6H-dibenz[b,e]oxepin, welches in Methanol-Äther hergestellt wird, schmilzt
bei 189 bis 193° C und besteht aus einem Gemisch der cis,trans-Isomeren.
B. 1 l-[3-(4-j8-Hydroxyäthyl-l-piperazinyl)-propyliden]-2-chlor-6,ll-dihydro-dibenz[b,e]oxepin
5,1 g der unter A erhaltenen Piperazinbase werden in 100 ecm Methanol gelöst. 1 ecm Äthylenoxyd
wird zugesetzt, und das Reaktionsgemisch wird 2 Stunden am Rückfluß erhitzt. Das Lösungsmittel wird
verdampft, und der Rückstand wird in Äthanol gelöst. Die Lösung wird mit einem Überschuß an Chlorwasserstoff
in Äthanol behandelt, und die Lösungsmittel werden verdampft, so daß das Produkt in Form
seines Dihydrochloridsalzes zurückbleibt. Es werden 6 g eines Produkts mit einem Schmelzpunkt von 253
bis 255°C erzielt; die Umkristallisation aus Äthanol-Wasser steigert die Reinheit; der Schmelzpunkt des
cis-Isomeren beträgt 258 bis 261° C und der des transisomeren
271 bis 272° C.
Claims (3)
1. 11 -[3-(4-/S- Hydroxyäthyl - piperazino) - propyliden]-2-chlor-6,ll-dihydro-dibenz[b,e]oxepin
der Formel I

und dessen Salze.
2. Verfahren zur Herstellung der Verbindung nach Anspruch 1, dadurch gekennzeichnet, daß
man ein Phosphoran der allgemeinen Formel II (R)3P=CH-CH2CH2-N N-R1 (II)
in der R eine Alkylgruppe mit 1 bis 6 Kohlenstoffatomen, eine Phenyl-, Aminophenyl- oder Benzylgruppe
darstellt und R1 ein Wasserstoffatom oder eine 2-Hydroxyäthylgruppe bedeutet, mit einem
Keton der Formel III

umsetzt und gegebenenfalls das nicht substituierte Stickstoffatom des Piperazinringes durch Umsetzung
mit Äthylenoxid substituiert.
3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, daß man ein Phosphoran der Formel
II verwendet, in der R eine Phenylgruppe bedeutet.
