Gegenstand der Erfindung sind l-(i\) -Aroylalkyl)-4-aryl-piperidincar"bonsäure-(4).-amide
der allgemeinen Foimel

in der Ar und Ar1 Phenylreste, die durch niedere Alkyl gruppen,
Methoxygruppen oder durch Halogenatome substituiert
sein können oder den Thienyl-(2)-Rest darstellen, Alk
eine niedere Alkylengruppe mit 3 "bis 6 Kohlenstoffatomen,
R ein Amino-, niedr. Alkylamino-, Di-(niedr. alkyl)-amino-, Phenylamino-, (niedr. Alkyl) (phenyl)-amino-, niedr.
Phenylalkylamino- oder ein über das Stickstoffatom an die

Carbonylgruppe gebundener Pyrrolidino—, Piperidino-, Morpholino-, Methylmorpholino-, Dimethylmoipholino-,
Piperazino- oder Phenylpiperazinorest und X Wasserstoff
oder .die Methylgruppe ist, sowie ein Verfahren zur Her stellung
dieser Verbindungen.
Dieses Verfahren besteht darin, daß man eine Verbindung
der allgemeinen Formel
Ar-CO- Alk - Halogen
mit mindestens der äquimolaren Menge einer Verbindung der
allgemeinen Formel
CO-R
Ar1
umsetzt, wobei Ar, Ar', Alk, R und X die oben angegebene
Bedeutung haben.
Die erfindungsgemäße Umsetzung kann in einem inerten
Lösungsmittel, z.B. einem aromatischen Kohlenwasserstoff,; wie
Benzol, Toluol, Xylol, einem niederen Alkahol, wie Äthanol,
Propanol oder Butanol oder einem niederen Alkanon, wie Aceton,
Butanon, Pentanon oder Hexanon durchgeführt werden. Ein besonders
ge eignetes Lö sungsmittel i st 4-Methyl-2-pentanon.
Durch Anwendung erhöhter Temperatur kann der Reaktiöiisablauf
beschleunigt werden. '

Beispiele für Alkylenreste in den Verfahrensprodukten
sind der Propylen-, Butylen-, Methylpropylen-, letramethyl en-
und Pentamethylenrest. Bevorzugt wird der, Propylenrest.
Die neuen Verbindungen gemäß der Erfindung stellen ausgeprägte Apomorphin-Antagonisten und Barbiturat-Potengierungsmittel
dar. Dabei zeigen sie nur geringe oder gar
keine Nebenwirkungen, wie Parkönson-artige Symptome, Hypnose und Hemmung der Nebennierenrindenhormone. Sie eignen sich daher
ausgezeichnet als selektive Antibrechmittel und sind in dieser Hinsicht dem bekannten, in gleicher Richtung wirkenden
4-(2'-Dimethylamino-äthoxy)-N-(3",4",5"-trimethoxy benzoylj-benzylamino-hydrochlorid
bei weitem überlegen.
Die erfindungsgemäß erhältlichen Verbindungen bilden in Form der freien Basen mit anorganischen und starken organischen
Säuren, wie Schwefel-, Phosphor-, Salz-, Brom wasserstoff-,
Jodwasserstoff-, SuIfamin-,. Zitronen-, Milch-,
Malein-, Äpfel-, Bernstein-, Wein-, Zimt-, Essig-, Benzoe-,
G-lucon- und Ascorbinsäure Salze. Sie können ferner in
quaternäre Ammoniumsalze übergeführt werden, a.B. mit
Methyl chlorid und -bromid, Äthyl chlorid, Propylchlorid,
Butylchlorid, I so butyl chi ο rid, Benzylchlorid und -bromid,
Phenyläthylbromid, Naphthylmethylchlorid, Dimethylsulfat,
Diathylsulfat, M«thylbenzolsulfonat, Äthyltoluolsulfonät,
Äthyl enchlorhydrin* Propylenchlorhydrin, Allyl bromid, Methallylbromid und Crotylbromid. -
Die alg Ausgangsstoffe für das erfindungsgemäße
Verfahren verwendeten Aroyl alkyl halogenide erhält man durch
Friedel-Orafts-ITmsetzung nach W.J. Close, Journ. Am.Chem,
Sog. 79, SΓ1455-58 (1957) bzw. C. Van de Westeringh et. al.,
Ind. Chim. BeIg. Uo.9, S. 1073-76 (I960) oder durch Um- '
setzung eines C^-Halogenalkannitrils mit dem entsprechenden
Aiylma^nesiumbromid nach J„B. Conant et. al, ,Journ. Am.Chem,
Soc. 46, S. 1882-85 (1924) und C. Van de Westeringh et. al.,

Die Ausgangsstoffe der allgemeinen Formel

werden dadurch hergestellt, daß man eine Verbindung der
allgemeinen Formel
HO-CH2-CH2-Im-CH2-OH(X) -OH
mit einer Verbindung der allgemeinen Formel Y-Cl umsetzt,
in der Y eine die Aminogruppe blockierende Gruppe ist, z.B.
der p-Ioluolsulfonyl- oder der Benzylrest (vgl. 33er. 74 B,
S. 1453 (1941); hier ist eine Verbindung beschrieben, in der X,Wasserstoff und Y der p-Toluolsulfonylrest ist; ferner
Beilstein, Bd- 12, S. 1080; in der dort beschriebenen
Verbindung ist X Wasserstoff und Y der Benzylrest; wenn X die Methylgruppe und Y der p-Ioluolsulf onylrest ist, schmilzt
die erhaltene Verbindung bei etwa 66,2 - 68,2 C). Die erhaltene Verbindung der allgemeinen Formel

Mittel nach 0. Eisleb, Ber. 74 B, S. 1453 (1941) zu einer
Verbindung der allgemeinen Formel

umwandelt. Wenn X in dieser Verbindung die Methylgruppe
bedeutet, entsteht ein Racemat. Die Isomeren werden daraus durch fraktionierte Kristallisation, vorzugsweise
aus Aceton, abgetrennt, wobei zuerst das oc-Isomere erhalten
wird (die gewählte Bezeichnung dient jedoch nur zur Unterscheidung und sagt nichts über die tatsächliche Konfiguration
aus).

anderes Verfahren angewandt. Die nitrile der oben angegebenen Formel werden zuerst zu den entsprechenden Säuren der allgemeinen
Formel hydrolysiert,

Beispiel 2 :
4,9 Teile 4-Phenyl-piperidincarbonsäure-(4)-N-tert. butylamid-hydrochlorid werden in Wasser gelöst und alkalisch
gemacht. Die alkalische Lösung wird mit Chloroföim. extrahiert und der organische Extrakt eingedampft. Der aus der
freien Base bestehende Eiickstand, 5*3 Teile Natriumcarbonat,
0,1 Teile Kaliumiodid und 60 Teile 4-Methy1-2-pentanon
werden zusammen gerührt. Dieser Lösung werden nach und nach 4,3 Teile T~Z~ Thenoyl-(2)_7-propylchlorid
in 60 Teilen 4-Methyl-2-pentanon zugesetzt. Nach 48-stündigem
Erhitzen unter Eückfluß wird das Gemisch gekühlt und
mit Wasser gewaschen. Die organische Lösung wird getrock- r
net und eingedampft. Der Rückstand wird in Methanol gelöst. Anschließend wird dieser Lösung eine Lösung von
Oxalsäure in Methanol zugesetzt. Der abgeschiedene Fest stoff wird äbfiitriert und getrocknet; man erhält l

Ein G-emisch aus 4,2. Teil en }T-(2,5-Dimethyrbenzoyl)-propylchlorid,
6 Teilen 4-:Phenyl-piperidincarbonsaure-(4)-pyrrolidid,
:-"12 Teilen Natriumcarbonat, 0,1 Teil .,
• Ealiumjodid und 280 Teilen 4-Methyl-2-pentanon wurde 59.
Stunden unter iöicltfluß erhitzt und dann filtriert. Das "FIltrat
wurde' eingedampft' und der Hückstand in 2-Propanol ge-Iqst.
Der Lösung wurde Oxalsäure in 2-Propanol zugesetzt.
Der abgeschiedene'Niederschlag wurde abfiltriert, mit Aceton
gewaschen und anschließend aus Methanol' umkristallisiert.
Man erhielt 1-f γ-(2·,5'-Dimethyibenzoyl^-propyl^-^-plienylpiperidincarbons^äure^(4)-pyrroiiaid-oxaiat,
das bei etwa r 183,6-184° schmilzt.
Claims (1)
- Patentansprüche:/-4 CO-Rin der Ar und Ar1 Phenylreste, die durch niedere Alkylgruppen, Methoxygruppen oder durch Halogenatome substituiert sein können, oder den Thienyl-(2)-Rest darstellen, Alk eine niedere Alkenylgruppe mit 3 bis 6 Kohlenstoffatomen, R ein Amino-, niedr. Alkylamino-, Di-(niedr. alkyl)-amino-, Phenylamino-, (niedr. Alkyl) (phenyl)-amino-, niedr. Phenylalkylamino- oder ein über das Stickstoffatom an die Carbonylgruppe gebundener Pyrrolidino-, Piperidino-, Morpholino- oder Phenylpiperazinorest und X Wasserstoff oder die Methylgruppe let.2. l-£ 3-(4*-KLuorbenBoyl)-propylJ7-4-(3"-phenyl) -piperidincarbonsäure- (4 )-pyrrolidi d.3. l-£"3-(4'-Pluorbenzoyl)-propylJ7-4-(4ll-chlorphenyl)-piperidincarbonsäure-(4)~H,!i-dimethylamid.8 0 9 8 0 9/111
