式Ⅰ化合物能于存在或者没有酸接受体或适当缩合剂的情况下,通过式Ⅱ的活化羰基或硫代羰基化合物与取代的苯胺反应而得到。实现这个转变的方法在本技术领域内是众所周知的:见Zabicky“The    Chemistry    of    the    Amides”Interscience,1970。

式Ⅱ的酯(X1=C1至C6烷氧基)能用几个方法直接转变成式Ⅰ化合物。在路易氏酸如AlMe3存在下,苯胺容易地与式Ⅱ的酯反应,反应最好在室温至120℃进行,适当的溶剂包括二氯甲烷、1,2-二氯乙烷和甲苯。Weinreb等在“Organic Synthesis”59,49,(1982)中揭示了这个方法,最好与低级醇如甲醇或乙醇的酯进行。
式Ⅱ酸(X1=OH)能用肽技术领域已知的偶合剂,连同苯胺一起直接转变成式Ⅰ化合物。偶合剂包括二环己基碳二亚胺(DCC)、N-羟基琥珀酰亚胺、2-氯-N-甲基吡啶鎓碘化物、羰基二咪唑,或其它能活化酸官能的试剂,或如脱氢剂作用的试剂。Gross等在“The Peptides”3Vols.,Academic Press,New York,1979至1981,叙述了这些和其它的方法。

式Ⅰ(Y=H)的一些化合物能通过烷基化、酰化、和磺化反应(Y=H)转变成式Ⅰ的其它化合物。在酸接受体的存在下,式Ⅰ化合物与亲电试剂(Y-离去基团)反应,在氮原子上产生取代。在该方法中,如氢化钠、叔丁氧钾、氢化钾和本技术领域已知的其它碱将酰胺去质子是较好的。适当的亲电剂包括下列但并不限于这些:烷基卤、酰卤、酸酐、碳酸盐、氯甲酸酯、二硫化物和磺酰卤。该反应通常在0-25℃的温度范围内进行,但是如果使用不活泼的亲电剂时;能在直至120℃的温度下进行。在上述反应条件下,不会去质子的溶剂如:四氢呋喃、二甲基甲酰胺、二甲氧基乙烷和二乙醚是较好的。
式Ⅰ(X=O)化合物通过硫杂化试剂的方法能转变成式Ⅰ(X=S)化合物。酰胺转变成硫代酰胺在本技术领域是众所周知的。对于这个转变有效的较好试剂是五硫化磷,它可以单独使用或者与有机的或无机的碱合并使用。当单独使用五硫化磷时,有机碱如吡啶是较好的溶剂。当五硫化磷与无机碱如碳酸氢钠联合使用时,较好的溶剂是醚如二甘醇二甲醚。采用20℃-160℃之间的温度能使转变成功,而较好的温度是90至120℃之间。Lapucha在“Synthesis”(1987)第256页中叙述了这些及其它转变酰胺至硫代酰胺的方法。
通过上面的叙述,不管合成的方法,式Ⅱ化合物能转变为本发明的化合物,这对于本技术领域的技术人员来说是可以理解的。许多官能团的转变能使式Ⅰ化合物转变成新的式Ⅰ化合物,这对于本技术领域的那些技术人员是习知的,而就典型反应机制而伦,这将克服某些基团与上面所说的试剂和条件的不相容性。
实例1
N-(4-氯代苯基)-2-〔(4-氯代苯基)氨基〕2-氧代-乙烷亚肼基氯化物
在0至5℃,化合物4-氯代苯胺(7.8克)用亚硝酸钠(4.5克)进行重氮化。得到的溶液在超过20分钟时间,维持0至5℃,通过保温滴液漏斗加到剧烈搅拌着的2,4′-二氯-N-乙酰乙酰苯胺(10克)和乙酸钠(15.1克)的乙醇(260毫升)混合物中,悬浮液搅拌2小时(20℃)并过滤。固体经溶于二氯甲烷和加入硫酸镁来干燥。过滤,蒸发溶剂,用丁基氯研制得标题化合物(15.2克),熔点:180℃-181℃。

实例9的产物(0.72克)与50%氢氧化钠(1毫升)的85%甲醇水溶液(10毫升)回流2小时。混合物用6N盐酸水溶液酸化,并分配于乙酸乙酯和水之间。乙酸乙酯层用硫酸镁干燥、浓缩得黄色固体。该固体悬浮于含硫酰氯(1.5毫升)的苯(20毫升)中,加热回流1.5小时。浓缩混合物并与甲苯(10毫升)共沸得油性的酰氯,将酰氯溶于四呋喃(15毫升),并用含三乙胺(1.0毫升)和4-氟苯胺(0.3毫升)的苯溶液(10毫升)处理。反应混合物搅拌18小时,并分配于乙酸乙酯和1N盐酸之间。有机层用碳酸氢钠和食盐水洗涤,有机层干燥(硫酸镁),浓缩成为残余物,残余物经色谱层析或用甲醇重结晶纯化,标题化合物(0.46克)为黄色固体;熔点:194℃-195.5℃。

4-(4-氟代苯基)-2-氧代-3-丁烯酸钾
丙酮酸和对氟苯甲醛(24.8克)的甲醇(20毫升)溶液冷至15℃,用氢氧化钾(16.8)的甲醇(50毫升)溶液处理,当加完2/3之后,移去冷却浴,让温度升至40℃,出现黄色沉淀,放置过夜后过滤沉淀物,固体用甲醇和醚洗涤,得到的标题化合物(34.5克)不用进一步纯化即可使用。
实例17
1-(3,4-二氯苯基)-5-(4-氟代苯基)-4,5-二氢-N-〔4-(三氟甲基)苯基〕-1H-吡唑-3-羰酰胺
实例16的化合物(9.0克)用3,4-二氯苯肼盐酸盐(9.5克)的水溶液(100毫升)处理。过滤橙色固体并于空气干燥,悬浮于冰乙酸(150毫升)并回流3小时。在冷却时,吡唑啉酸结晶(3.3克)。还可收集第二次产物(1.1克)。将吡啉酸悬浮于苯(100毫升)并用亚硫酰氯(6毫升)处理,混合物回流2小时并蒸发,残余物溶于干燥四氢呋喃(50毫升)并分成5等份。取一等分加入4-氨基三氟甲苯(0.35毫升)和三乙胺(0.9毫升)的四氢呋喃(10毫升)溶液,混合物搅拌30分钟并蒸发,残余物用甲醇(10毫升)研制得标题化合物(0.9克)。熔点:241.5-243℃。

实例18
4-(4-氟苯基)-2-氧代-N-〔4-(三氟甲基)苯基〕-3-丁烯酰胺
按Stecher(J.Am.Chem.Soc.,1952,74,4392)的常规方法,将实例16的化合物转变成相应的羧酸,该酸(8.5克)用二氯甲基甲醚(10毫升)的二氯甲烷(30毫升)处理。混合物加热回流并蒸发HCl,在蒸发2小时后,得到的酰基氯溶于四氢呋喃(100毫升),用三乙胺(8毫升)和4-氨基三氟甲苯(4毫升)的四氢呋喃(25毫升)混合物处理。在1小时后,混合物分配于1N    HCl和乙酸乙酯之间。放置过夜,乙酸乙酯层沉淀出标题化合物(5克)熔点:200-201℃。

4,5-二氢-4-(4-氟苯基)-1-苯基-N-〔4-(三氟甲基)苯基〕-1H-吡唑-3-羰酰胺
将实例18(0.6克)的化合物和苯肼(0.2毫升)的二甲基甲酰胺(5毫升)加热回流1小时,残余物经硅胶(50克)色谱层析,用己烷/乙酸乙酯(3∶1)作洗脱剂进行纯化,得到标题化合物(0.4克)熔点:183.5-184.5℃。

将实例13化合物(2.1克)加到氢化钠(0.2克,在油中60%)的无水四氢呋喃(25毫升)悬浮液中。30分钟后,将甲基碘(0.9克)分三次加入悬浮液,混合物搅拌18小时,用氯化铵溶液骤冷。用乙酸乙酯萃取混合物,有机层经干燥和蒸发,油状残余物经硅胶色谱层析,用己烷/乙酸乙酯(3∶1)作洗脱液得标题化合物(1.4克):熔点:181-183℃。

按实例20的方法,以1/2规模,用乙酐(0.5毫升)代替碘代甲烷制备标题化合物(0.8克),经硅胶色谱层析纯化,用己烷/乙酸乙酯S∶1)作洗脱剂得固体,以甲醇结晶得透明的黄色针状结晶。熔点:158-160℃。

控制昆虫和保护农作物是通过下述方法达到的,即将一种或多种有效量的本发明式Ⅰ化合物使用于昆虫蔓延地区及要保护的农作物区域,或直接施用于昆虫上来控制。使用的较好方法是通过配置喷雾器的喷雾将本发明的化合物散布到叶、土壤中,或被昆虫蔓延或需要保护的植物部份。另一方面,这些化合物的颗粒配方能使用于土壤或叶子上或选择性地将它们混到土壤中去。空中或地面上使用都可以。
本发明的吡唑化合物能直接使用,但是常常大多数使用由包括一个或多个本发明化合物在农业上适宜的载体或稀释剂组成的配方。使用的最好方法包括喷雾该化合物的水悬浮液或精制的油溶液。通常喷雾油和喷雾油浓缩物的组合能增强式Ⅰ化合物的效力。
使用式Ⅰ化合物所需速率以作有效地控制昆虫,乃取决于下列因素,如,需要受控制的昆虫的种类、害虫的生长阶段、它的大小、它的位置、宿主农作物、使用之年的时间、环境湿度、温度条件等。一般来说,在正常情况下大规模领域的操作中,每公顷使用0.05-2公斤的活性组分就足够于提供有效的控制,但是少至每公顷0.01公斤是足够的或多至每公顷8公斤也是需要的,这取决于前面所述的因素。加入如胡椒基丁醚化合物能增强式Ⅰ化合物的杀虫活性。

与喷雾油合并一起
当试验化合物(化合物30)与喷雾油或喷雾油浓缩物合并配制成可乳化的浓缩物时,试验化合物对抗草地粘虫的短期及滞留的杀虫活性得到改进。
试验化合物用5毫升丙酮稀释,然后用蒸馏水混合至1SS和50ppm。将喷雾油或喷雾油浓缩物以油:活性组份为10∶1的比率加至上述溶液中,喷雾油是在大气压下具有中间蒸馏温度(377℃)的石腊石油基油;喷雾油浓缩物由83%异链烷烃油和17%山梨醇酯及环氧山梨醇酯混合物组成;试验单位由生长在4英寸盒中的三周令大豆植株组成。三个植株为一组进行处理,每次处理是用带有喷咀的可旋转喷雾器以流量10rpm喷雾的。
在喷雾干燥后,处理的叶片切割一半,每片叶子放进6小隔盒组织培养皿中的一个隔内,在每个隔中放入第三令幼虫。然后将全部单位用湿润的吸油纸覆盖,对于每次处理放置这样的4个单位,试验单位在27℃和50%相对湿度保持72小时计算死亡率。处理的植株保持在27℃和50%相对湿度,于7天和14天重复试验测定滞留活性,测定结果记录于表8中。

用增效剂增强活性
按实例32叙述方法制备试验单位,用5个草地粘虫幼虫(Spodopt-era    frugiperda)。用试验化合物(化合物30)处理之前,按实例32叙述的技术,用5倍于所用试验化合物浓度比率的胡椒基丁醚(PBO)喷雾于试验单位上。在2.5小时后用试验化合物以10、5、2.5、1.0、0.5和0.1ppm喷雾于其上。覆盖试验单位,于27℃和50%相对湿度保持72小时后计算死亡率,所得结果记录于表9中。

Y为H、C1-C4烷基、C2-C4烷氧基烷基、C1-C4烷硫基、C1-C4卤代烷硫基、苯硫基或带有1-3个各选自W、C2-C4烷氧羰基、C(O)H、C2-C4烷羰基或C2-C4卤代烷羰基取代基取代的苯硫基;
A为H、C1-C6烷基、苯基、被(R5)p取代的苯基、CN、CO2R3、C(O)R3、C(O)NR3R4、C(S)NR3R4、C(S)R3或C(S)SR3
B为H、C1-C6烷基、C1-C6卤代烷基、C2-C6烷氧基烷基、C2-C6氰基烷基、C3-C8烷氧羰基烷基、C2-C6链烯基、C2-C6炔基、C2-C6烷氧羰基、苯基、带有1-3个各选自W、取代基取代的苯基、苄基或带有1-3个各选自W取代基取代的苄基。

R2和R5分别为卤素、CN、SCN、NO2、R3、OR3、SR3、S(O)2R3、OC(O)R3、OS(O)2R3、CO2R3、C(O)R3、C(O)NR3R4、S(O)2NR3R4或NR3R4
R3为C1-C4烷基、C1-C2卤代烷基、C2-C4链烯基、C2-C4卤代链烯基或炔丙基;
R4为H或C1-C2烷基;
