Abstract

Description
技术领域
本发明涉及一种以藻类提取物为主要成分的产品,这些产品通常用作食品补充剂以及用在保护细胞的化妆或皮肤病学应用中。
背景技术
属于紫菜属的红藻包括多个种,其中:
-在布列塔尼地区收集的脐形紫菜,
-在布列塔尼地区有的purpurea紫菜,
-在日本培养的甘紫菜,
-在日本培养的条斑紫菜,
-在澳大利亚收集的columbina紫菜
紫菜的食用品种也被称为Nori。脐形紫菜种形成一种淡紫色的环状细披针形片,其长度可以达到60厘米。该紫菜种是一种世界上消费最多的食用海藻。在法国,允许人们食用消费脐形紫菜种。这种藻类的营养价值在于富含各种不同的化合物,例如:
-蛋白,最高到45%干物质,
-多不饱和脂肪酸,如EPA(二十五烷酸),最高到5%藻脂类,
-B族维生素,特别是维生素B12,
-维生素A和类胡罗卜素。
在现有技术中描述过紫菜藻类提取物的许多性质以及它们含有的化合物,
例如:
-甘紫菜甲醇(80%)提取物的抗氧化性质,
-条斑紫菜在各种不同的极性和非极性溶剂中的提取物,特别是甲醇提取物的抗氧化性质,
-使用甲醇(50%)和丙酮提取的提取物的抗氧化性质,
-存在高浓度的牛磺酸及其衍生物,它们溶于水醇介质,它们的chaperonnes蛋白(HSP)的élicitrices性质为人们所知,
-存在可溶于水醇介质的糖衍生物,例如甘油半乳糖苷(α-D-吡喃半乳糖基-(1-2)-丙三醇)及其异构体,它们对受到强的氢应激反应的细胞介质渗透调节起着重要的作用,
-存在非提取的紫菜聚糖、甘氨酰胺核苷酸(gar)类的多糖,因为不溶于醇含量高于70%的水醇溶液,它们具有免疫和抗肿瘤的性质。
本申请人现在证明了脐形紫菜红藻的水醇提取物具有细胞受到各种物理作用时可诱发合成应激蛋白的能力。
这种出乎意料的能力使这些提取物具有制备活性组合物的显著意义,所述的组合物可以用于防止细胞不会受到由生理学或物理作用,如热冲击或受到UV辐射照射,或由病理生理学作用,特别是感染或发炎所引起的应激反应。
事实上,在应答加速蛋白变性的处理(如热)时,合成应激蛋白是一种细胞为面对非常多样性侵害而产生的一般化防御措施。这些应激蛋白与由于侵害改变其三维结构的蛋白结合,并参与其消除和变性复原,或这时这些应激蛋白与尚未形成三维结构的蛋白(在生物合成过程中的蛋白)结合,参与其成熟。这些应激蛋白于是通过保存细胞组分的完整性,并因此保存它们的功能而保护这些细胞。
因此,在应答侵害时更早诱发合成应激蛋白,并更大量地合成这些蛋白,对于赋予其更有效保护体系的细胞是有利的。
发明内容
因此,本发明的目的是一种在细胞受到物理的、生理学的作用或病理生理学侵害时能诱发合成应激蛋白的产品,其特征在于它含有红藻水醇提取物或它由红藻水醇提取物组成。
本发明优选地涉及一种属于紫菜属红藻的提取物,非常优选脐形紫菜属红藻的提取物。
该藻类提取物含有下述化合物,其比例是以相对于提取物总重量的重量百分比表示的:
-牛磺酸及其衍生物:1-5%,
-甘油半乳糖苷及其衍生物:10-25%,
-类胡萝卜素与AGPI(未测定含量),
-水溶性维生素(未测定含量),
-氨基酸及其衍生物(未测定含量),
-各种颜料。
本发明的产品含有以产品总重量计1-30重量%的红藻水醇提取物。
这种藻类水醇提取物可以使用下述化合物进行稳定:
-为以后以粉末形式使用时为麦芽糊精;本发明的粉状产品以产品总重量计含有10-30重量%,优选15-25重量%,更优选约20重量%的红藻水醇提取物。
-为以后以液体形式使用时为甘油(化妆组合物使用除外)。本发明的液体产品以产品总重量计含有10-30重量%,优选15-25重量%,更优选约20重量%的红藻水醇提取物。
-用于化妆组合物时添加防腐剂以及进行灭菌过滤(0.2μ);用于化妆组合物的本发明液体产品含有1-25重量%、优选3-20重量%的红藻水醇提取物。
呈粉末形式时,该产品可以很容易加入干的食品增补剂配方中,并且可以胶囊剂、片剂或袋装等包装。
呈液体形式时,该产品可用于化妆制剂,或包装成软胶囊。
本发明涉及红藻提取物或含有红藻提取物的产品在制备组合物、特别是食品或化妆组合物中的应用,这种组合物用于在细胞受到各种不同物理作用时能诱发合成应激蛋白。本发明的组合物对于调节应激蛋白HSP60、HSP27和HSP70是非常特别有效的。
在本发明范围内进行的研究工作能够证明红藻提取物或含有红藻提取物的产品具有保护这些细胞防止热冲击、UV辐射或任何其它物理或治疗冲击的能力。
这些性质使本发明的组合物无论对人,还是对动物都是有好处。
本发明涉及红藻提取物或含有红藻提取物的产品在制备组合物、特别是食品或化妆组合物中的应用,这种组合物用于保护这些细胞防止生理学应激反应和病理生理学侵害,特别是感染和炎症。
本发明还涉及红藻提取物或含有红藻提取物的产品在制备组合物、特别是食品或化妆组合物中的应用,这种组合物用于保护这些细胞防止物理侵害,特别是热冲击或UV辐射。
本发明的应用还涉及热的热冲击和冷的热冲击。于是,遇到不利的环境,例如海洋或山区的个人,也可以使用本发明的组合物,并因此特别适合于滑雪者、登山运动员和高水平运动员。
对于受到UV辐射照射,本发明的红藻提取物或含有红藻提取物的产品不作为防晒剂,而是在受到此等照射时作为对细胞所受到的应激反应的保护剂。
本发明的组合物可用于保护细胞防止物理侵害,特别是热冲击或者UV辐射,这些组合物可以口服方式或局部用药方式用药。
本发明的组合物可以构成人与动物的食品增补剂,提供每次剂量为10-1000毫克,优选25-250毫克含红藻提取物的产品。
本发明组合物构成食品增补剂,它可以为胶囊剂、软胶囊、片剂或药袋、糖浆等形式。
本发明的组合物也可以构成化妆组合物,任选地皮肤病学组合物,这时它们局部涂敷在皮肤上。本发明的组合物能够保护皮肤防止物理侵害,更具体而言,防止热冲击或UV辐射。本发明的化妆组合物还可以用作改善皮肤质量与外观的防衰老组合物。
本发明的化妆组合物可以是化妆学和皮肤病学技术领域的技术人员已知的任何形式,除涂在脸或身体皮肤上外无其它盖伦制剂限制。有利地,本发明组合物可以是凝胶、膏、乳剂、奶液、喷雾剂等形式。
这些组合物还可以含有一种或多种在化妆和皮肤病学组合物中通常已知使用的配方剂或添加剂,作为非限制性实例可以列举软化剂、着色剂、成膜活性剂、表面活性剂、香料、防腐剂、乳化剂、油类、甘醇类、皮脂吸收剂、维生素等。借助这些化妆材料知识,本技术领域的技术人员可以向本发明的组合物中添加任何配方剂,并按照所要求的性质添加任何量的配方剂。
本发明的化妆组合物含有0.1-5%、并优选0.5-2%含有红藻提取物的产品。

具体实施方式
实施例1:脐形紫菜水醇提取物在培养物中活体外细胞对热冲击或UV辐射反应方面的活性
为了进行在这个实施例中描述的实验,使用了含有0.12%该海藻干提取物的紫菜水醇(7%醇)提取物。在这些培养基中添加这种提取物高达1%。最后的海藻提取物浓度是0.0012%。同样浓度的醇溶液(7%醇,体积/体积)用作对照,并且添加到这些培养基中高达1%(最后浓度:0.07%)。
实验1:证明紫菜提取物诱发合成HSP
将如此制备的紫菜提取物和对照溶液添加到经过遗传调制(TSPc技术)的CHO细胞(中国仓鼠卵巢细胞)培养基中。然后这些细胞在43℃进行热冲击90分钟。在这种模式中,使用Elisa试剂盒(Boehringer-Roche)测定合成的生长激素,可反映出在热侵害的作用下诱导合成HSP的情况。
图1表明,热应激反应时,用紫菜提取物预培育细胞可诱导刺激合成hGH,因此合成HSP,这种刺激作用显著地高于在没有接受紫菜提取物的细胞中所观察到的这种刺激作用。
实验2:鉴定由紫菜提取物诱导合成(热冲击与UV)的HSP
采用了特定的抗-HSP27、抗-HSP60、抗-HSP70抗体,和免疫印记法技术。热冲击使用的细胞是HeLa细胞,UV应激(波长为254nm的UV辐射,0.250J/cm2)使用的细胞是HaCat细胞。
图2表明紫菜提取物在没有任何侵害时都不改变HSP的产生,这意味着紫菜提取物不改变HSP合成的生理学方式。另一方面,热冲击时,紫菜提取物刺激产生HSP60、HSP70和HSP27蛋白。t=30分钟时,看到在用紫菜提取物预培育的细胞中合成的HSP27和60与对照相比显著地增加。与此相反,HSP70的合成与对照细胞的合成相当。在热冲击后的时间t=4小时30分钟和t=19小时,在用紫菜提取物预培育的细胞中HSP27、60和70的合成显著高于对照细胞。
图3表明紫菜提取物在没有任何侵害时不改变HSP的产生,但采用UV辐照时可刺激这些HaCat细胞合成HSP70。
实施例2:紫菜水醇提取物的配方
1)人的食品增补剂。
每盒30个胶囊,每个胶囊700毫克。净重:21克(折整值)。
配料:在甘油中的本发明的紫菜提取物,琉璃苣原油。胶囊(鱼胶以及土豆淀粉)。包衣剂:黄蜂蜡。可以添加维生素、矿物质、植物提取物和乳酵素。
2)动物食用增补剂
每盒30个胶囊,每个胶囊700毫克。净重:21克(折整值)。
配料:在甘油中的本发明的紫菜提取物,琉璃苣原油。胶囊(鱼胶以及土豆淀粉)。包衣剂:黄蜂蜡。
可以添加维生素、矿物质、植物提取物和乳酵素。
3)防晒口服组合物
每盒45个胶囊,每个胶囊600毫克(6号)。净重:27克。配料:胶囊(鱼胶和土豆淀粉),琉璃苣原油。在甘油中的本发明紫菜提取物。包衣剂:蜂蜡,西红柿提取物,用富含硒介质培养的酵母,维生素E。
4)抗皮肤衰老的口服组合物
每盒60个胶囊,每个胶囊673毫克。净重:40.4克(折整值)。配料:琉璃苣原油,胶囊(鱼胶和土豆淀粉)。干绿茶提取物。包衣剂:蜂蜡,在甘油中的本发明紫菜提取物,小麦蛋白水解产物,含硒酵母,硫酸锌、硫酸锰。

Claims (14)
1、包含红藻水醇提取物或者由红藻水醇提取物组成的产品在制备食品和化妆组合物中的应用,所述组合物在细胞受到热冲击或暴露于UV辐射时能诱发应激蛋白的合成,且其中所述红藻是紫菜属红藻。
2、根据权利要求1所述的应用,其特征在于所述紫菜属红藻属于脐形紫菜种。
3、根据权利要求1所述的应用,其特征在于所述应激蛋白是HSP60、HSP27或HSP70。
4、根据权利要求1或2所述的应用,其特征在于所述红藻提取物含有下述化合物:
-牛磺酸:1-5%,
-甘油半乳糖苷:10-25%,

8、根据权利要求6或7所述的应用,其特征在于所述产品含有麦芽糊精。
9、根据权利要求6或7所述的应用,其特征在于所述产品含有甘油。
10、根据权利要求9所述的应用,其特征在于所述产品含有化妆品中可使用的防腐剂,并且经受0.2μm灭菌过滤处理。
11、根据权利要求1或2所述的应用,其特征在于食品组合物提供每次剂量为10-1000毫克的所述产品。
12、根据权利要求11所述的应用,其特征在于食品组合物提供每次剂量为25-250毫克的所述产品。
13、根据权利要求1或2所述的应用,其特征在于化妆组合物含有0.1-5重量%所述产品。
14、根据权利要求13所述的应用,其特征在于化妆组合物含有0.5-2重量%的所述产品。
