式I化合物优选以α-氨基-β-烷氧羰基杂环化合物或α-氨基-β-碳环酸杂环化合物为原料进行制备,这些杂环化合物中的一些(其中Het=噻吩基)可从市场上购得(存在两种异构体)。例如,噻吩-2-氨基-3-羧酸甲酯可按照S.Gronowitz等在Suecica药学学报(Acta Pharm.Suecica)1968,第5卷,第563页甲所还万法制备。其它杂环化合物可按照文献中教导的方法制备。例如,Golankiewicz等在《四面体》1985,41,5989中描述了5-氨基噻唑-4-羧酸乙酯和5-氨基-2-甲基噻唑-4-羧酸乙酯的合成。α-氨基-β-烷氧羰基杂环化合物或α-氨基-β-碳环酸杂环化合物与酰胺(R4CONHR3)的反应(反应流程1中的步骤1)可以很方便地在RT至回流温度下在POCl3、SOCl2或SO2Cl2存在下于溶剂中进行,所述溶剂例如是ClCH2CH2Cl,CHCl3,CH2Cl2,苯,甲苯,己烷,环己烷等。所得脒(III)或者自发地环合成嘧啶-4-酮,或者通过在20℃-回流温度下在溶剂中用碱(如叔丁醇钾,NaH,KH,n-BuLi,NaOH,Na2CO3等)处理转化成环化产物,其中所述溶剂如THF,二氧六环,己烷,甲苯,DMSO,DMF,二甲基乙酰胺等。用硫将4-酮基置换为4-硫酮基的反应(反应流程1中的步骤3)通过在四氢呋喃、二氧六环或甲苯溶剂中在RT至回流温度下与P2S5或Lawesson试剂反应而进行。
上述合成路线第一次公开了如何制备包括在本发明所述式I结构范围内的3H-噻吩并[2,3-d]嘧啶-4-酮衍生物。
下列文献中描述了其中R1=R2=H的通式I化合物的制备方法:Chemical Scripta 1981,18,135、合成(Synthesis)1977,180、药物化学学报(Chem.Pharm.Bull.)1989,37,2122和DE-OS-2411273。
本发明还涉及式III、IV和V的中间体,尤其是其中A代表噻吩基[2,3-d]的那些。
在噻吩并嘧啶-4-酮化合物的5-元环上引入其它取代基也可以很方便地应用金属有机方法学进行。例如,可以将噻吩[3,2-d]嘧啶-4-酮和噻吩并[2,3-d]嘧啶-4-酮化合物的6-位选择性地脱质子。为此,特别适宜的碱为二异丙基氨基锂(LDA),环己基异丙基氨基锂(LICA)或仲丁基锂/TMEDA。上文所述的基团R1或R2中的许多可以通过使所产生的阴离子与亲电试剂反应而引入(反应流程2中的步骤1),典型的亲电试剂为Br2,NBS,F2,ICI,Cl2,F+试剂,三甲基甲氯硅烷。反应流程2:特定杂环化合物的合成E+ 1.2=NBS(N-溴代琥珀酰亚胺),NCS(N-氯代琥珀酰亚胺),I2,Cl2,Br2,FCl,F+试剂,TMS以及类似的Si试剂。

具有杀菌性能的喹唑啉酮化合物是已知的(见WO-94/26722或EP-A-276825),而且根据WO-97/02262也知噻吩并嘧啶酮化合物是已知的。
出人意料的是,现已发现,式I新化合物在实践中具有非常有利的活性谱,能够保护植物免受真菌以及细菌和病毒的侵害。
式I化合物可以作为活性成分在农业上以及相关领域中用于防治植物病虫害。本发明新化合物的突出之处在于其在低施用量下具有优越的活性,且能被植物很好地耐受和对环境安全。它们具有非常有用的治愈、预防和内吸性能,可用于保护多种栽培植物。式I化合物可用于抑制或消灭出现在不同有用作物的植物或植物部分(果实、花、叶、茎、块茎、根)上的有害生物,同时亦能保护植物后生长出来的部分使其免受(例如)植物病原微生物的侵害。
也可以使用式I化合物作为拌种剂来处理植物繁殖材料,特别是种子(果实、块茎、谷粒)和植物插条(例如秧苗),以抵御真菌侵染和土壤中存在的植物病原真菌。
例如,化合物I对下列种类的植物病原真菌有效:半知菌类(如葡萄孢属、梨孢属(Pyricularia)、长孺孢属、镰孢属、壳针孢属、尾孢属和链格孢属)和担子菌纲(如丝核菌属、驼孢锈菌属、柄锈菌属)。此外,它们还对子囊菌纲(例如黑星菌属和白粉菌属、柄球菌属、链核盘菌属、钩丝菌属)以及卵菌纲(如疫霉属、腐霉属、单轴霉属)有效。另外,式I新化合物对植物病原细菌和病毒(例如黄单胞杆菌属的各个种、假单胞杆菌属、解淀粉欧文氏杆菌以及烟草花叶病毒)也有效。
在本发明范围内,欲保护的靶标作物一般包括下列种类的植物:禾谷类(小麦、大麦、黑麦、燕麦、稻、玉米、高粱及有关种类);甜菜(糖用甜菜和饲用甜菜);梨果、核果和浆果(苹果、梨、李、桃、杏、樱桃、草莓、树莓和黑莓);豆科植物(菜豆、小扁豆、豌豆、大豆);油料植物(油菜、芥菜、罂粟、油橄榄、向日葵、椰子、蓖麻油植物、可可豆、落花生);黄瓜属植物(南瓜、黄瓜、甜瓜);纤维植物(棉花、亚麻、大麻、黄麻);柑橘属植物(橙、柠檬、葡萄柚、柑橘);蔬菜(菠菜、莴苣、芦笋、圆白菜、胡萝卜、洋葱、番茄、马铃薯、红辣椒);樟科(鳄梨、肉桂、樟脑)或诸如烟草、坚果、咖啡、茄、甘蔗、茶树、胡椒、葡萄、啤酒花、香蕉和天然橡胶植物之类植物,以及观赏植物。
式I化合物通常以组合物形式使用,而且可以与其它化合物同时或依次施用于作物区或欲处理的植物。这些其它化合物可以是例如肥料或微量营养元素供体或能影响植物生长的其它制剂。它们也可以是选择性的除草剂以及杀虫剂、杀真菌剂、杀细菌剂、杀线虫剂、杀软体动物剂或数种这些制剂的混合物,如果需要,还含有其它载体、表面活性剂或加工领域惯用的施用促进助剂。
式I化合物可以与其它杀真菌剂一起混合,结果在一些情形下能产生意外的增效活性。

适宜的载体和助剂可以是固体或液体,而且是剂型加工技术中有用的物质,如天然或再生矿物质、溶剂、分散剂、湿润剂、粘着剂、增稠剂、粘结剂或肥料。
施用式I化合物或包含至少一种所述化合物的农药组合物的优选方法是叶面施用。施用次数和施用量取决于相应病原的侵染危险程度。然而,式I化合物也可以通过用液体制剂浇湿植物所在地,经土壤通过根部进入植物体内(内吸),或者通过将固体形式(如颗粒形式)的化合物施用于土壤(土壤施用)。在水稻作物中,这种颗粒剂可以施用到灌水的稻田中。式I化合物也可以通过用该杀菌剂的液体制剂浸渍种子或块茎、或用固体制剂包覆它们而施用于种子上(包衣)。
式I化合物可以以其原始形式、或者(优选)与剂型加工领域常用的辅助剂一起使用。为此,它们可以用已知方法很方便地加工成乳油、可涂覆性糊剂、可直接喷雾或可稀释的溶液剂、稀乳剂、可湿性粉剂、可溶性粉剂、粉剂、颗粒剂以及胶囊剂(如用聚合物胶囊化的制剂)。正如组合物的种类一样,根据预定目标和主要情形选择施用方法,如喷雾、雾化、喷粉、撒粉、涂覆或浇泼。
适宜的施用量通常为每公顷(ha)5g-2kg活性成分(a.i.),优选10g-1kg a.i./ha,最优选为20g-600g a.i./ha。当用作浸种剂时,适宜的剂量为每千克种子10mg-1g活性物质。
制剂,即包含式I化合物和如果需要还含有固体或液体助剂的组合物,是用已知的方法制备,典型的是将化合物与填料如溶剂、固体载体以及任选的表面活性化合物(表面活性剂)紧密混合和/或研磨。

虽然优选将市售产品配制成浓缩物形式,但最终使用者通常使用稀释过的制剂。
所述组合物中也可以包含有另外的助剂如稳定剂、消泡剂、粘度调节剂、粘结剂或增稠剂以及肥料、微量营养元素供体或其它能获得特定效果的制剂。

在磺化烧瓶内,向60ml 1,2-二氯乙烷中加入11.0g(70mmol)2-氨基-3-甲氧羰基噻吩和10.9g(76mmol)戊酸丙酰胺。在室温及搅拌下,缓慢滴加入7ml磷酰氯。在回流温度下保持3小时后,将混合物倒入冰水中,用碳酸氢钠调节至弱碱性。然后用二氯甲烷萃取所得混合物三次,并将分离出的有机相用硫酸钠干燥。然后利用水喷射真空泵除去溶剂。
在磺化烧瓶内,将所得粗产物加到100ml无水四氢呋喃中,并在搅拌下,小心加入4.36g(0.1mol)NaH/50ml无水THF。在回流温度下搅拌2小时后,利用水喷射真空泵除去溶剂,并将残留物溶于乙酸乙酯/水中。水相另外再用乙酸乙酯萃取。有机相用硫酸钠干燥,然后利用水喷射真空泵除去溶剂。粗产物通过硅胶柱色谱纯化(洗脱剂:TBME/己烷=1∶2),得到12.0g 2-正丁基-3-正丙基-3H-噻吩并[2,3-d]嘧啶-4-酮,为黄色粉末,m.p.70-72℃。实施例P-4a:2-正丁基-6-氯-3-正丙基-3H-噻吩并-[2,3-d]嘧啶-4-酮

5.根据权利要求4的式I化合物,其中:A为噻吩基[2,3-d],X为氧,R1为氢,氯或溴;R2为氢,氯或溴;且R1和R2中至少一个不为氢;R3为C3-C5烷基或O-C1-C4烷基;R4为C2-C5烷基或苯基,所述苯基为未取代的或被氟、氯、溴、C1-C4烷基或苯氧基单至三取代,而且其中的苯氧基则又为未取代的或被氟、氯或溴单至三取代。
6.根据权利要求4的式I化合物,其中:A为噻吩基[2,3-d],X为硫,R1为氢,氯或溴;R2为氢,氯或溴;且R1和R2中至少一个不为氢;R3为C3-C5烷基或O-C1-C4烷基;R4为C2-C5烷基或苯基,所述苯基为未取代的或被氟、氯、溴、C1-C4烷基或苯氧基单至三取代,而且其中的苯氧基则又为未取代的或被氟、氯或溴单至三取代。
7.根据权利要求4的式I化合物,其中:A为噻吩基[3,2-d],X为氧,R1为氢,氯或溴;R2为氢,氯或溴;且R1和R2中至少一个不为氢;R3为C3-C5烷基或O-C1-C4烷基;R4为C2-C5烷基或苯基,所述苯基为未取代的或被氟、氯、溴、C1-C4烷基或苯氧基单至三取代,而且其中的苯氧基则又为未取代的或被氟、氯或溴单至三取代。
8.根据权利要求1的式I化合物,其中A为吡啶基。
9.根据权利要求1的式I化合物,其中A为苯基。
10.根据权利要求1的式I化合物,其中A为噻唑基。
11.根据权利要求1的式I化合物,其中A为哒嗪基。
12.一种防治或预防有害生物的组合物,其中活性成分是与合适的载体混在一起的权利要求1中所述的化合物。
13.权利要求1的式I化合物在保护植物免受植物病原微生物侵染方面的应用。
14.一种防治或预防由植物病原微生物引起的栽培植物感染的方法,它包括对植物、植物部分或其生长场所施用权利要求1所述的式I化合物。
