The efficacy test index I of the application: elastic R2 value
Test instrument: german CK skin elasticity tester MPA 580
The test principle is as follows:
the skin elasticity tester is a special instrument for measuring the elasticity of the surface layer of the skin, and mechanically deforms the skin by negative pressure. The measurement principle is based on inhalation. The instrument generates negative pressure to suck the skin into the aperture of the probe and release the skin after a period of time. The depth of skin penetration into the probe is measured by a non-contact optical system. The optical measuring system consists of a light source, a light receiver and two prisms facing each other, and the light receiver receives light from the emitter. The light intensity varies due to the penetration depth of the skin. The ability of the skin to resist negative pressure (compaction) and return to its original state (elasticity) during the measurement is shown graphically (length of stretch/time). This test principle allows us to understand the elastic and mechanical properties of the skin surface and to quantitatively and objectively analyze the skin age. The test probe of this experiment gave a curve, so a parameter was recorded as a reference for skin elasticity, r2= (e (a) -e (a+b))/e (a) =ua/uf=total elastoplasticity of the rebound portion (blue distance/black distance-total elasticity). The closer the value is to 1 (100%) the more elastic the curve is, a very important parameter, often used by the literature, so this experimental reference data records the R2 value, which indicates skin elasticity.
The testing method comprises the following steps:
the experiment selects 60 subjects with healthy skin and ages of 20-35 years, and the subjects are divided into 20 groups, and one group of three persons is used for parallel control. The skin elasticity of the test examples and control samples was measured, R2 values were recorded, and the mean change after 1 month and the mean change after 2 months for each sample was calculated for each of the 3 subjects and the subjects were required to avoid exposure to sunlight for as long as possible during the course of the project.
Test condition preparation: the test environment in which the experiment was carried out was kept constant at a temperature of 20℃and a relative humidity of 50%. Since thermal radiation can lead to inaccurate measurements, direct measurement of sunlight is kept avoided and the subject is required to avoid direct irradiation of sunlight during the test time so as not to bring about deviation of experimental results.
Subject preparation: the subjects were asked to make a simple clean and rest in the test laboratory for 30 minutes before the time of the test with informed consent of the subjects, and during the adaptation environment, the measured area was not blocked by clothing, emotional stress could also lead to increased evaporation and increased blood circulation, ensuring that the adaptation room was calm.
The formal start test time is 9 days earlier, and the initial value before the test subjects do not use the sample is measured on the first day, and one pump of the same product is used in the morning and evening for every three subjects. After 1 month, skin elasticity was measured for the same test area of the cheek, three times, and the average value of the three tests was recorded, and after 2 months, the operation was repeated, and the test was completed.
Test results:

The efficacy test index II of the application: wrinkle area ratio.
Test instrument: american face image analyzer VISIA-CR
The test principle is as follows:
the United states company combines years of clinical experience with the most advanced digital camera technology, and provides a professional facial image photographing and analyzing system VISIA-CR for skin researchers. Whether quantitative or qualitative, or both, the VISIA-CR can provide you with a very professional standard photographing method, allowing the following analysis of the skin: wrinkles and fine lines skin photodamage, skin texture, vascular characteristics, skin color and uniformity, porphyrin (acne).
The VISIA-CR has a variety of light source modes: standard light, UV light, cross polarized light, parallel polarized light. Due to the use of multiple light sources, the visual-CR greatly enhances the visibility of the skin to be analyzed.
The proprietary RBX technology gathers a great deal of research experience of colorimetry, provides melanin and vascular conditions of the lower layer of the skin for a great number of researchers, and is a brand new method for detecting, displaying and analyzing the lower layer of the skin. One photographing process comprises a plurality of light source modes, and at most seven pictures can be obtained.
The VISIA-CR is a face image photographing system which is simple and flexible to operate and accurate to control, and has the following characteristics:
1. the photographing of the high freshness image is completed within a few seconds.
2. The shooting time is short, the efficiency is high, and the subject can more easily keep a correct shooting position in the shooting process.
3. For different skin conditions, a preset light source pattern recommended by a clinical expert may be selected from the instrument.
4. Good repeatability of the test result is ensured through the target positioning and image ghost preview functions.
5. The built-in image capturing control function greatly reduces the possibility of misoperation of operators.
The VISIA-CR instrument is provided with specialized Mirror Photo Tools image management special software, and the shot images can be directly stored in a computer, so that the search and the image comparison are facilitated. All the images shot by the VISIA-CR can be searched according to file attributes or various parameters customized by users, and a new method for managing databases of a large number of images is provided for researchers. The base database images can be browsed and different images under the same subject name. The software has the functions of importing, exporting and renaming the images, and prints out according to the content required by the clients, so that the software is a very useful tool for researchers. The Photo Tools software also has the following functions:
1. distance, angle, area and percentage can be measured. 2. Images at different times are compared on the screen, the treatment result of the subject is tracked, and the test result is displayed. 3. The partial image enlarging function can carefully study the improvement condition of the skin. 4. The image can be conveniently marked, analyzed and written.
In the experiment, the wrinkle area ratio in the same AOI selected area is selected to reflect the anti-aging effect of each sample, and the smaller the numerical value is, the smaller the representation is, and the better the anti-wrinkle effect is. The wrinkle area ratio refers to the proportion of the wrinkle area in the AOI field to the total AOI field area.
The testing method comprises the following steps:
the experimental subjects and skin elasticity test subjects are in the same batch, each subject is subjected to a wrinkle area ratio test by using VISA-CR after the skin elasticity test is finished, wrinkle area ratio values are recorded, the average value of the wrinkle area ratio change of each sample after 3 subjects use for 1 month and the average value of the wrinkle area ratio change after use for 2 months are calculated for relevant evaluation, and the subjects are required to avoid exposure to sunlight for as long as possible during the project proceeding period.
Test results:
average change in skin wrinkle area ratio after 1 month of use Δ3= [ (initial skin wrinkle area ratio before use of subject 1: initial skin wrinkle area ratio before use of subject 1)/(initial skin wrinkle area ratio before use of subject 1 + (initial skin wrinkle area ratio before use of subject 2: initial skin wrinkle area ratio after use of subject 2) initial skin wrinkle area ratio before use of subject 2 + (skin wrinkle area ratio after use of subject 3: initial skin wrinkle area ratio before use of subject 3)/(initial skin wrinkle area ratio before use of subject 3 ]. 100%/3) of example 1.
Average change in skin wrinkle area after 2 months of use Δ4= [ (initial skin wrinkle area ratio before use of subject 1 to initial skin wrinkle area ratio after use of subject 1 to 2 months)/initial skin wrinkle area ratio before use of subject 1 + (initial skin wrinkle area ratio before use of subject 2 to 2 months) initial skin wrinkle area ratio before use of subject 2 + (initial skin wrinkle area ratio after use of subject 3 to 2 months-initial skin wrinkle area ratio before use of subject 3 to initial skin wrinkle area ratio before use of subject 3)/initial skin wrinkle area ratio before use of subject 3 ]. 100%/3.

Test results: as can be obtained by comparing the test results of example 1 and examples 2 to 7, example 1 is an optimal ratio combination of the components of the anti-wrinkle essence emulsion, and wherein the ratio of the components of the anti-wrinkle composition is also the optimal ratio combination. The test results of example 1 and comparative examples 1 to 7 are compared with each other, so that good compound synergism can be achieved among panthenol, adenosine and carnosine, and the effect brought by the synergistic interaction of any one or two of panthenol, adenosine and carnosine is far less excellent than that brought by the synergistic interaction of the panthenol, the adenosine and the carnosine. As can be seen from the comparison of the test results of example 1 and comparative examples 8 to 13, the amounts of panthenol, adenosine and carnosine which are used in excess of the prescribed range, either too much or too little, significantly reduce the overall efficacy of the anti-wrinkle composition.
The present embodiment is only for explanation of the present application and is not to be construed as limiting the present application, and modifications to the present embodiment, which may not creatively contribute to the present application as required by those skilled in the art after reading the present specification, are all protected by patent laws within the scope of claims of the present application.
