Description

Anti-wrinkle composition and preparation method and application thereof
Technical Field
The invention relates to the technical field of cosmetics, and particularly relates to an anti-wrinkle composition, and a preparation method and application thereof.
Background
With aging, skin aging is an inevitable aging phenomenon. Skin aging can be divided into endogenous aging and exogenous aging, wherein the former mainly refers to aging caused by irresistible factors existing in the body, such as heredity, gravity, endocrine, immune function and the like, along with the increase of age; the latter aggravates the natural aging process mainly due to the effects of the environment, sunlight, health, living habits, and the like. Particularly, in the age with great living pressure, people often stay up, smoke, drink, use electronic products for a long time, use excessive eyes and the like to accelerate skin aging, and when the water content of the skin cuticle is about 20 percent and is sufficient, the skin is soft, smooth, fine and elastic. When the stratum corneum contains less than 10% of water due to lack of lipids, water or moisturizing factors, the normal structure of the stratum corneum is unstable, and the barrier function of the skin is affected, so that increased percutaneous water loss is caused, and the skin becomes dry, rough, dull, less elastic, more wrinkled, itchy, desquamated, and the like, and also, skin aging is accelerated. Among them, facial skin aging is particularly remarkable, the number of fibroblasts in the human body is gradually reduced, collagen and elastin secreted therefrom are decomposed and broken, and the moisturizing component hyaluronic acid is reduced, the stratum corneum is thinned, etc., thereby making the skin lose elasticity and forming wrinkles, which are a sign of skin aging.
Aging is an inevitable but postponable biological process, and with the improvement of living standard and the pursuit of beauty, people have more and more demands for anti-wrinkle cosmetics.
For example, Chinese patent application 201811429898.9 discloses an anti-wrinkle moisturizing skin-brightening composition comprising alkali-soluble ganoderan ferulic ester. The research shows that the alkali-soluble ganoderma lucidum polysaccharide ferulate has the anti-aging effect, and the anti-wrinkle moisturizing and skin-brightening composition has the anti-aging and anti-wrinkle effects by adding the alkali-soluble ganoderma lucidum polysaccharide ferulate, but the anti-wrinkle effect is not obvious.
In recent years, retinol is widely used as an effective anti-aging component in medicine and cosmetics, is widely applied to anti-aging and anti-wrinkle products, has obvious effect, and is favored by consumers. For example, the chinese patent application 202110728500.7 discloses a multi-dimensional anti-wrinkle composition, a cosmetic composition and a preparation method thereof, which relate to the technical field of cosmetics, and the multi-dimensional anti-wrinkle composition is composed of the following raw materials in parts by weight: retinol palmitate: 0.05-0.2 part; acetyl tetrapeptide-2: 0.02-0.2 part; acetyl hexapeptide-8: 0.02-0.2 part; split yeast fermentation lysate: 0.5-5 parts; auxiliary components: and (4) the balance. However, the particular physicochemical properties of retinol limit its use: has great sensitivity to light, heat, polar media and the like, is easy to decay into non-bioactive substances under common conditions, and brings difficulty to use and storage. And retinol is converted by enzymes on the skin surface to release retinoic acid, which has certain irritation.
Therefore, there is a need for the development of an anti-wrinkle composition having good water retention, pore tightening, skin gloss enhancement, low irritation and photosensitivity, and good stability.

Effect test
irritation/Corrosion test-chick embryo chorioallantoic Membrane vascular test
(1) The experimental method comprises the following steps: the chick embryo chorioallantoic membrane (CAM) test is a commonly used method for evaluating the irritation of cosmetics in vitro, and utilizes the characteristic that the chorioallantoic membrane is similar to the conjunctival tissue structure of eyes in the middle stage (8-12 days) of hatching embryos. A certain amount of the test substance is dropped on the CAM, and the change of blood vessels (such as bleeding, angiolysis and blood coagulation) of the CAM is observed in a certain period of time, so that the eye irritation of the test substance is evaluated according to the damage degree of the CAM. The anti-wrinkle composition is a transparent test substance, and then is scored by adopting a reaction time method. After egg examination of the chicken embryo incubated for 10 days, marking the position of the air chamber, peeling off the eggshell part at the air chamber by using dental zigzag forceps, wetting the exposed white egg membrane by using a certain amount of 0.9% NaCl solution, and then sucking out the solution. The intima is removed with forceps and care should be taken to ensure the integrity of the vascular membrane. 0.3mL of the anti-wrinkle composition solution of the above example was directly dropped on the CAM, timed for 5min, and the change of the CAM within 5min was observed, and the initial time(s) for bleeding, clotting, and angiolysis to occur was recorded. A0.1 mol/L NaOH solution is set as a positive control, and a 0.9% NaCl solution is set as a negative control. The irritation score was calculated according to the formula (1), and the ocular irritation of the sample was graded according to the irritation score. IS < 1: no eye irritation; 1 IS < 5: mild ocular irritation; IS IS not less than 5 and less than 9: moderate ocular irritation; IS IS not less than 10: strong ocular irritation/erosion. 6 chick embryos were tested and averaged.

Effect test
1. Fibroblast protein expression assay
(1) Test samples: the experimental group is the anti-wrinkle skin care product of application examples 1-6
(2) The test method comprises the following steps: the human skin fibroblasts after recovery were subcultured in DMEM medium containing 10% Fetal Bovine Serum (FBS), with 1 change of the culture medium every 2d, to logarithmic phase. Then taking the fibroblasts in the logarithmic phase for suspension culture, adjusting the cell density (the cell density is 111111/mL and 90 muL of the cells are added into a 96-well plate in each well in the cell viability detection process, the cell density is 30000/mL and 2mL of the cells are added into a 12-well plate in each well in the I type collagen synthesis detection process), the temperature is 37 ℃ and the content of 5% CO is 5% 2 Culturing under the condition. After 24h, the cells were attached and the test sample and positive control (EGF, 10ng/mL) were added. The supernatants were harvested and tested for type I collagen content using an ELISA kit, with the cell number and the amount of collagen produced by untreated cells as references (═ 100%).
(3) And (3) test results: the results are shown in detail in FIG. 1.
According to the detection data in fig. 1, it can be seen that the anti-wrinkle skin care products prepared in examples 1 to 6 can better promote the synthesis of type I collagen, and especially the skin care products prepared in examples 5 to 6 have higher efficiency in promoting the synthesis of type I collagen.
2. Skin elasticity test
2.1 test sample: the experimental groups were the products of examples 1-6 and comparative examples 1-11.
