Description

Composition with moisturizing and antioxidant effects and application thereof
Technical Field
The invention belongs to the technical field of cosmetics, relates to a composition with moisturizing and antioxidant effects and application thereof, and particularly relates to a composition with moisturizing and antioxidant effects and application thereof in preparation of cosmetics.
Background
The skin is gradually aged due to the continuous contact with the outside, including external pollution, ultraviolet irradiation and other factors, which continuously generate free radicals. Free radicals are products of human cell metabolism, and excess free radicals have destructive effects on unsaturated fatty acids, protein molecules, nucleic acid molecules, extracellular soluble components, membrane lipids, etc., resulting in wrinkles, sagging, and decreased subcutaneous fat. The free radicals destroy collagen and elastic fiber protein, so as to make skin moisturize and maintain elasticity, and accelerate the precipitation of melanin. The increase in age and certain in vitro factors can cause the body and skin tissues to produce free radicals that exceed the body's normal ability to scavenge free radicals, causing damage to the skin tissues and resulting in aging.
Meanwhile, when the environment changes, the stratum corneum of the skin cannot timely regulate enough moisturizing factors, the activity of the sebaceous glands is reduced, the oil and water on the face are reduced, the skin is tightened, and fine wrinkles are even formed under the eyes and beside the canthus. Therefore, when skin is daily protected, the moisture of the skin needs to be kept, the loss of the moisture of the skin needs to be reduced, and a certain water-oil balance state of the skin needs to be maintained.
Moisture retention and oxidation resistance are important links for maintaining young skin and delaying aging, and development of a cosmetic product with effective moisture retention and oxidation resistance is very significant.

Solution preparation: firstly, 1mg of DPPH is dissolved in about 20mL of methanol, ultrasonic treatment is carried out for 5min, and the mixture is fully shaken to ensure that the upper part and the lower part are uniform. Taking 1mL of the DPPH solution, and measuring the A value at 519nm to ensure that A = 1.2-1.3; preparing each sample solution: the compositions of examples 1 to 11 and comparative examples 1 to 4 were diluted with methanol to prepare a composition having a mass fraction of 1%.
The specific method comprises the following steps: each set of sample adding system is shown in the following table, the reaction solution is shaken up and placed in a water bath at 37 ℃ for heating for 15min, and then taken out, and an ultraviolet-visible spectrophotometer is used for measuring the absorbance value of the solution.
Reagent C1/mL C2/mL C3/mL
DPPH solution 2 2 0
Sample solution 0 2 2
Anhydrous methanol 2 0 2
The formula for the DPPH radical clearance is shown as follows:
DPPH radical clearance =1- (A2-A3)/A1 × 100%
In the formula, A1, A2 and A3 are absorbance values of C1, C2 and C3 respectively.
The test results are shown in table 1:
TABLE 1
Group of DPPH radical scavenging ratio (%)
Example 1 76.2
Example 2 74.7
Example 3 76.6
Example 4 87.4
Example 5 86.9
Example 6 87.6
Example 7 86.3
Example 8 82.9
Example 9 82.5
Example 10 80.8
Comparative example 1 74.1
Comparative example 2 65.3
Comparative example 3 73.3
Comparative example 4 70.8
As can be seen from the data in Table 1: the overall DPPH radical scavenging rate of examples 1-3 is higher than that of comparative examples 1-4, indicating that the DPPH radical scavenging effect is reduced in the absence of either component of the composition; the DPPH free radical scavenging rate of example 4 is higher than that of examples 8-10, which shows that yeast amino acids, serum albumin and decarboxylated carnosine hydrochloride also have a certain synergistic effect on DPPH free radical scavenging effect.
Test example 2
And (3) comprehensive effect experiment of moisturizing and antioxidation:
(1) 80 Chinese healthy female subjects with dark, dark yellow and uneven facial skin are selected, the age range is 25-60 years old, and the average age is 46.3 +/-5.6 years old. Stratum corneum water content, skin elasticity measurements (curometer) facial image capture (VISIA-CR, VC20 Plus), facial image analysis (VC 20 Plus) analysis of skin wrinkles were tested on the skin before (D0) and after 4 weeks with the test product (D28).
(2) The test products are emulsion products prepared in application examples 1-7 and comparative application example 1; the test environment is at a temperature of 20 +/-2 ℃ and a humidity of 50 +/-5%, and meets the requirements of scheme design; promotion/improvement after use of the product (%) = (data after use-data before use)/data before use × 100%.
(3) The comparison statistics of the moisture content of the face stratum corneum (C.U), the comparison results before and after the use of the elasticity R2 value of the cheek skin, and the comparison results before and after the use of the statutory print SEw are shown in Table 2:
TABLE 2
Figure BDA0003902798020000131
As can be seen from the data in Table 2: the overall moisturizing effect and anti-aging effect of application examples 1-7 are higher than that of comparative application example 1, which shows that the composition of the present invention has significant moisturizing and anti-oxidation effects, wherein the comprehensive effect of application example 1 is better.
Test example 3
Skin irritation test:
skin irritation measurements were performed using the emulsions of application examples 1-7. 35 women were selected as the test subjects, the spot test tapes containing 0.03mL of the samples were applied to the normal skin of the left forearm of the test subjects, and the test was conducted 6 times within a half hour after the application of the patch according to the CTFA guidelines (1981), and on the 6 th day after the application of the patch, additional tests were conducted to observe delayed reactions, and the identification criteria and results are shown in Table 3 and Table 4, respectively.
TABLE 3
Grade (symbol) Identification criteria
0 - Negative reaction: has no irritation and erythema
1 ± And (3) suspicious reaction: mild erythema
2 + Weak positive reaction: erythema
3 ++ Strong positive reaction: erythema, papules, blisters
4 +++ Very positive reaction: severe edema and bulla
TABLE 4
Figure BDA0003902798020000141
From the above data results, it can be seen that: the composition and the emulsion have no stimulation to skin, have no positive reaction and have mild and non-stimulation effects.
