Abstract

本发明属于分子生物学领域,具体涉及一种Taq酶突变体及其应用。具体技术方案为:一种Taq酶突变体,所述Taq酶突变体的氨基酸序列如SEQ ID NO.8所示。本发明对野生型Taq酶进行定向进化,提供系列适合于ARMS‑qPCR的Taq酶,相比于野生型Taq酶,其3’端错配识别能力显著高于野生型Taq酶。本发明的聚合酶可以包含在反应混合物和/或试剂盒中。另外,本发明公布的针对ARMS Taq酶优化的最适缓冲体系,可以显著提高ARMS‑qPCR的特异性和扩增效率。

Description

一种Taq酶突变体及其应用
技术领域
本发明属于分子生物学领域,具体涉及一种Taq酶突变体及其应用。
背景技术
扩增阻滞突变型PCR(Amplification Refractory Mutation System PCR,ARMS-PCR,ARMS-PCR),是目前肿瘤个体化分子检测最重要和最先进的技术之一,在临床上被广泛应用。目前ARMS-PCR结合TaqMan探针进行检测的ARMS-qPCR技术广泛应用于肿瘤靶向用药EGFR、KRAS等基因检测,市场上CFDA批准的EGFR的肿瘤靶向药检测试剂盒90%以上都是ARMS-qPCR检测方法。ARMS-PCR采用等位基因特异的两条上游引物,两者在3’端核苷酸不同,一个与野生型等位基因特异互补,另一个与突变型等位基因互补,由于Taq酶没有3’~5’外切酶活性,因此在Taq DNA聚合酶作用下,与模板不完全匹配的上游引物将不能生成PCR产物,而与模板匹配的引物体系可以扩增,结合qPCR就能很容易地分辨是否是有效扩增,从而确定基因型。
ARMS-qPCR检测能力可高达0.1%~1%,其扩增的严谨性受DNA序列自身识别特异性和Taq酶3’端错配识别能力等因素影响,其中Taq酶识别特异性是ARMS-qPCR试剂开发的重要因素,影响ARMS-qPCR 扩增模板选择能力和检测特异性。
发明内容
本发明的目的是提供一种Taq酶突变体及其应用。
为实现上述发明目的,本发明所采用的技术方案是:一种Taq酶突变体,所述Taq酶突变体的氨基酸序列如SEQ ID NO.8所示。
相应的,一种高效Taq酶突变体,在所述Taq酶突变体的基础上,融和CL7。
优选的,所述CL7的融和位置为:所述Taq酶突变体最左侧第一个蛋氨酸与精氨酸之间。
优选的,所述高效Taq酶突变体的氨基酸序列如SEQ ID NO.2所示。
相应的,一种Taq酶突变体,对野生型Taq酶进行如下任意一项或几项或全部突变:Val155Ala、Ser486Gln、Glu491Ser、Phe495Ile,将155号的缬氨酸、486号的丝氨酸、491号的谷氨酸、495 号的苯丙氨酸分别突变为丙氨酸、谷氨酰胺、丝氨酸、异亮氨酸。

本发明具有以下有益效果:本发明对野生型Taq酶进行定向进化,提供系列适合于ARMS-qPCR的Taq酶,相比于野生型Taq酶,其3’端错配识别能力显著高于野生型Taq酶。本发明的聚合酶可以包含在反应混合物和/或试剂盒中。另外,本发明公布的针对ARMS Taq酶优化的最适缓冲体系,可以显著提高ARMS-qPCR的特异性和扩增效率。

具体实施方式
本发明提供了一种新的Taq酶突变体(以下称为:Super ARMS Taq酶),具体为对野生型Taq酶进行如下的定点突变:将155号位点的缬氨酸突变为丙氨酸、486号位点的丝氨酸突变为谷氨酰胺、491号位点的谷氨酸突变为丝氨酸、495号位点的苯丙氨酸突变为异亮氨酸,并在N端添加CL7 序列,得到Super ARMS Taq。所述野生型Taq酶的氨基酸序列如SEQ IDNO.1所示,所述Super ARMS Taq酶的氨基酸序列如SEQ ID NO.2所示,其结构示意图如图1所示(图1中的Super ARMS Taq)。为了便于进行后续的效果展示,同时制备了突变1个位点(Taq 1)、突变2个位点(Taq 2、Taq 3、Taq 4)、突变3个位点(Taq 5)、突变4个位点(Taq 6)的六种对比Taq酶突变体。Taq 1、Taq 2、Taq 3、Taq 4、Taq 5、Taq 6的氨基酸序列分别如SEQ ID NO.3、SEQ ID NO.4、SEQ ID NO.5、SEQ ID NO.6、SEQ ID NO.7、SEQ ID NO.8所示。
Super ARMS Taq酶在Taq 6基础上融合了CL7(即图1的Super ARMS Taq相较于Taq6左侧多出的矩形结构,融和位置为:Taq 6最左侧第一个蛋氨酸与精氨酸之间)。CL7序列可提高Taq酶的扩增性能和3’端错配识别能力。所述Taq酶突变体可通过对野生型Taq酶进行定点突变后获得,或先制备Taq 6突变体后融合CL7获得,也可以采用其他本领域常规技术手段例如直接人工合成如SEQ ID NO.2所示的氨基酸序列。
本发明提供的Taq酶突变体适合于ARMS-qPCR,可根据实际需要,应用本领域的现有技术,对所述Taq酶突变体单独使用,或将其制备为试剂、试纸、试剂盒等后再进行使用。
下面将结合本发明实施例中的附图,对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅是本发明一部分实施例,而不是全部的实施例。若未特别指明,实施例中所用的技术手段为本领域技术人员所熟知的常规手段。所获得的数据均为进行至少3次重复后获得的平均值,且各重复获得的均为有效数据。
实施例一:Taq酶突变体粗酶性能测试
1、Taq酶突变体原核表达菌株的制备。将人工合成的Super ARMS Taq和Taq 1、Taq2、Taq 3、Taq 4、Taq 5、Taq 6的DNA分别连接到pET-28a载体上,并转化到DH5α大肠杆菌感受态中,涂布到含卡那霉素的LB固体平板上,37℃倒置培养过夜。
挑取状态良好的单克隆菌团于含卡那霉素的LB液体培养基中,37℃、200rpm/min震荡培养。将菌液进行一代Sanger测序验证。将测序验证成功的菌株进行质粒抽提,获得各Taq酶突变体表达质粒,并转化到BL21(DE3)大肠杆菌感受态中,涂布到含卡那霉素的LB固体平板上,37℃倒置培养过夜。挑取状态良好的单克隆菌团于含卡那霉素的LB液体培养基中,37℃、200rpm/min震荡培养至OD600值达到0.6。
2、Taq突变体粗酶的获取。按照1:200(V/V)的接种密度将含Taq酶突变体表达质粒的BL21(DE3)大肠杆菌接种至新的含卡那霉素的LB培养基中,37℃震荡培养3h至OD600值达到0.4。加入终浓度为0.3mM的IPTG,18℃震荡培养18h。4000×g,4℃离心收集菌体。用PBS洗涤菌体3次,加入含蛋白酶抑制剂的预冷PBS将菌体重悬至OD600值为6~7(约107cfu/uL),在冰上进行超声破碎(工作5s,停止15s,300T)。12000×g,4℃离心取上清,即为Super ARMSTaq和Taq 1、Taq 2、Taq 3、Taq 4、Taq 5、Taq 6的粗酶。
3、对Taq酶突变体进行DNA聚合酶扩增能力和3’端错配识别能力测试,按表1配置反应体系;Taq突变体粗酶指分别使用Super ARMS Taq和Taq 1、Taq 2、Taq 3、Taq 4、Taq5、Taq 6的粗酶。本发明(含本实施例)所用引物序列如表2所示。

表1中,所述2X ARMS Taq buffer组分为:40mM的Tris-SO4、10mM的(NH42SO4、50mM的K2SO4,1%Tween-20、2.5mM MgSO4、50mM betaine、0.35mM的dNTPs,pH=9.0。

3’端错配识别能力测试结果如图2所示,3’端错配识别能力通过突变型引物扩增突变型模板和野生型模板的Ct差值即阻滞能力来指征,由图2结果可知,Super ARMS Taq、Taq 6、Taq 5、Taq 4和Taq 3、Taq 2 Taq1 与 野生型DNA 聚合酶的3’端错配识别能力依次下降,3’扩增效率结果如图3所示,为Super ARMS Taq、Taq 6、Taq 5、Taq 4、Taq 3、Taq 2和Taq 1相比于野生型Taq 酶扩增能力即荧光定量PCR 中的Ct值显著提前,其中Super ARMSTaq扩增Ct值最小。
4、DNA聚合酶热加速稳定性测试。将各所述Taq酶突变体放置于37℃恒温恒湿培养箱中放置14天后,进行热加速处理。用不热加速处理的所述Taq酶突变体和处理后的所述Taq酶突变体分别配置如表3体系;Taq突变体粗酶指分别使用Super ARMS Taq和Taq 1、Taq2、Taq 3、Taq 4、Taq 5、Taq 6的粗酶。测定其扩增性能和3’端阻滞能力(阻滞突变型引物扩增野生型引物的能力)差异。

热加速稳定性测试结果如图4所示,为热加速处理对Taq 酶扩增能力和阻滞效果无显著影响。
5、DNA聚合酶灵敏度测试。将购买的菁良基因的标准品(GW-OGTS122)L858R位点标准品(5%突变概率,总浓度为50ng/μL),先用无菌水稀释到10ng/uL,然后用基因组DNA10ng/μL分别稀释到0.1%、0.05%,按照表4体系进行灵敏度验证。

DNA聚合酶灵敏度测试结果如图5所示,Super ARMS Taq的检测灵敏度可高达0.1%、0.05%。本实施案证明本发明提供的Super ARMS Taq可显著增加DNA聚合酶的扩增能力和3’端错配识别能力。
实施例二:Taq酶突变体的纯化
1、Taq酶突变体的诱导表达。按照1:200(V/V)的接种密度将含Taq酶突变体表达质粒的BL21(DE3)大肠杆菌接种至含卡那霉素的LB培养基中,37℃震荡培养2.5h至OD600值达到0.3后,于18℃继续震荡培养0.5h至OD600值达到0.8。加入终浓度为0.3mM的IPTG,18℃震荡培养18h。4000×g,4℃离心收集菌体。预冷PBS清洗菌体三次后,称取3g菌体,加入30mL预冷的裂解缓冲液(20mM Tris-HCl、500mM NaCl、5%(v/v%)Glycerol,0.1mM PMSF、1%(v/v%)蛋白酶抑制剂,pH=8.0)以悬浮菌体。在4℃下使用高压搅拌仪(~15,000 PSI)处理3min。4℃、40000×g离心20min后取上清过0.22μm滤膜,获得Taq酶突变体粗酶液。
2、使用CL7-Im7纯化系统纯化Taq酶突变体。将过滤后的粗酶液孵育到5mL CL7-Im7填料柱中。在4℃下,用6~8倍体积(本实施例使用的7体积)的高盐缓冲液(20mM Tris-HCl、2M NaCl、5%(v/v%) Glycerol)过柱后,再使用6~8倍体积(本实施例使用的7体积)的无盐缓冲液(20mM Tris-HCl、5%(v/v%) Glycerol)过柱,0.5mL/min流速,重复4次。加入2倍柱体积的含0.5mg SUMO蛋白酶的洗脱缓冲液(20mM Tris-HCl、50mM NaCl、5%(v/v%)Glycerol,0.2mM EDTA),4℃条件下孵育2.5h。将含Taq酶突变体的洗脱缓冲液收集起来,进行透析或超滤,即可获得高浓度和高纯度的Taq酶突变体。
3、纯化后的Taq酶突变体的扩增性能结果如图3所示,CL7-Im7纯化系统纯化Super-ARMS Taq扩增效率高于野生型Taq酶。
实施例三:Taq酶突变体与现有Taq酶性能对比
使用实施例一步骤3的方法,验证Taq酶突变体(Super ARMS Taq)和现有Taq酶:ARMS Taq(MD098)的性能。
结果如图6所示,其中纵坐标代表突变型F引物扩增对应突变位点和野生型位点的Ct差值,差值越大,代表阻滞效果越强,从图可知,对于多个位点,Super-ARMS Taq扩增的Ct差值均≥MD098,即Super-ARMS Taq比MD098具有更强的阻滞效果,与已有的Taq酶相比,本发明提供的Taq酶突变体具有更强的3’端错配识别能力,更适用于ARMS-qPCR中进行突变位点的检测。

Claims (10)

1.一种Taq酶突变体A,其特征在于:所述Taq酶突变体A的氨基酸序列如SEQ ID NO.8所示。
2.一种Taq酶突变体B,其特征在于:在权利要求1所述Taq酶突变体A的基础上,融合CL7。
3.根据权利要求2所述Taq酶突变体B,其特征在于:所述Taq酶突变体B的氨基酸序列如SEQ ID NO.2所示。
4.一种Taq酶突变体C,其特征在于:对野生型Taq酶进行如下任意一项或几项或全部突变:Val155Ala、Ser486Gln、Glu491Ser、Phe495Ile,将155号的缬氨酸、486号的丝氨酸、491号的谷氨酸、495 号的苯丙氨酸分别突变为丙氨酸、谷氨酰胺、丝氨酸、异亮氨酸,所述野生型Taq酶的氨基酸序列如SEQ ID NO.1所示。
5.根据权利要求4所述Taq酶突变体C,其特征在于:所述Taq酶突变体C的氨基酸序列如SEQ ID NO.7所示。
6.根据权利要求4所述Taq酶突变体C,其特征在于:所述Taq酶突变体C的氨基酸序列如SEQ ID NO.6所示。
7.根据权利要求4所述Taq酶突变体C,其特征在于:所述Taq酶突变体C的氨基酸序列如SEQ ID NO.5所示。
8.根据权利要求4所述Taq酶突变体C,其特征在于:所述Taq酶突变体C的氨基酸序列如SEQ ID NO.4所示。
9.权利要求1~8任意一项所述Taq酶突变体在ARMS-PCR中的非疾病治疗和诊断中的应用。
10.包括权利要求1所述Taq酶突变体A或权利要求2或3所述Taq酶突变体B或权利要求4~8任意一项所述Taq酶突变体C的试剂或试纸或试剂盒。
