Abstract
本发明提供了一种谷糠蛋白提取物及其制备方法和应用,所述应用即该蛋白提取物在制备改善肠道健康、缓解肥胖的功能性食品,特殊膳食用食品,保健品和/或药品中的应用。蛋白提取物的制备方法是将纯天然谷糠粉碎,加入蛋白提取液低温提取获得粗蛋白液,再通过硫酸铵沉淀法富集蛋白,随后用Tris‑HCl缓冲液溶解得到谷糠蛋白提取物(EMBP)。该蛋白制备原料方便易得,工艺简单,适合大规模工业化生产。研究结果证明,口服EMBP可以重塑高脂饮食小鼠肠道菌群结构,显著提高肠道乳杆菌丰度和定植;并且能够缓解肠道屏障渗漏进而有效改善小鼠肥胖等功能。
Description
技术领域
本发明涉及植物蛋白提取物的制备和应用,具体属于一种谷糠蛋白提取物及其制备方法,以及谷糠蛋白提取物在制备改善肠道健康和/或缓解肥胖的功能性食品、特殊膳食用食品、保健食品和/或药品中的应用。
背景技术
随着社会经济的发展和生活节奏的加快,居民饮食结构发生了显著改变—高糖高脂食物摄入增多,导致人们肠道菌群发生紊乱、肠道屏障功能受损,肠道问题人群逐年增加。据统计,我国90%的人群均存在肠道健康问题,由肠道健康引起的各种代谢疾病,如:高血压及肥胖等,严重威胁居民健康。目前临床上用于改善肠道屏障功能的方法主要有合理膳食、规律作息及科学运动等,其中膳食营养是维护肠道屏障功能的主要因素,而肠道菌群是介导膳食营养组分维护肠道屏障功能的重要媒介。因此,从膳食中深入挖掘调控肠道菌群稳态,改善肠道健康的营养组分,对于提高居民整体健康水平具有重要意义。
谷子,拉丁名:Setaria italica,主要种植于我国黄河流域,我国的年产量占全世界的五分之四,其中山西是谷子的主要产地。谷糠是谷子在加工成精米过程中得到的副产品,研究发现谷糠中蛋白质含量高达18%,营养价值较高,具有抗氧化、抗菌、降血压、降血糖及抗肿瘤等多重医药功效,但是关于其改善肠道健康方面的研究尚未见报道。本发明提供了谷糠蛋白提取物在重塑肠道菌群结构、提高肠道乳杆菌定植和丰度以及缓解肠道屏障渗漏以及肥胖等方面的应用,有利于其开发为保护肠道健康等方面的产品。
发明内容
本发明的目的在于提供一种谷糠蛋白提取物(EMBP)及其制备方法,以及该提取物在改善肠道健康和/或缓解肥胖中的应用。所述制备方法工艺简单,材料方便易得,得率高,适合大规模工业化生产。
本发明的上述目的是通过以下技术方案实现的:
一种谷糠蛋白提取物(EMBP)的制备方法,包括如下步骤:
第一步:将小米谷糠粉碎后,过60目筛,得到谷糠粉,加入植物蛋白提取液于发酵罐中低温搅拌24-64h;
第二步:板框过滤,收集滤液;
第三步:滤液加热升温至80℃,孵育20-30min,待沉淀物析出,降温至室温后进行板框过滤,收集澄清滤液;
第四步:澄清滤液加入硫酸铵进行沉淀,静置,板框过滤后收集沉淀,再用蛋白缓冲液溶解,重新板框过滤去除珍珠岩,获得含盐谷糠蛋白提取物;
第五步:将上述含盐谷糠蛋白提取物用3kD分子量的超滤装置脱盐,料液进行真空冷冻干燥,冻干成粉末即为谷糠蛋白提取物,于4℃保存。
所述第一步中的植物蛋白提取液为20mM Tris-HCl溶液,含0.85%NaCl,pH 8.0;所述的谷糠和蛋白提取液的重量体积比为1︰7-9,其中重量的单位为kg,体积的单位为L;所述的低温搅拌的温度:4-6℃,时间:48-64h。
所述第二步中的板框过滤的助滤剂为珍珠岩,用量按每升蛋白提取液加入40-60g珍珠岩计。
所述第三步中的孵育时间为25min。
所述第四步中的硫酸铵的饱和度为70-80%;所述的蛋白缓冲液为20mM Tris-HCl溶液,pH 8.0;静置时间为:6-8h。
通过高脂饮食诱导构建小鼠肠道屏障渗漏模型和肥胖模型,灌胃EMBP,实验结果表明:EMBP能够显著降低高脂饮食小鼠的体重,并缓解其胰岛素抵抗;显著增加了肠道紧密连接蛋白的表达,减轻其肠道屏障渗漏现象;经肠道菌群16S rRNA测序表明,EMBP可以重塑小鼠肠道微生物菌群结构,显著增加乳杆菌属的定植及丰度。该谷糠蛋白提取物有潜力开发为改善肠道健康、缓解肥胖的功能性食品、特殊膳食用食品、保健食品和/或药品等相关产品。
目前已报道的植物蛋白制备技术,工艺较为复杂,提纯成本较高,不适合用于大规模工业化生产。本发明主要对传统的植物蛋白提取方式进行了优化改进,主要包括:增加了蛋白提取液和谷糠的体积重量比,提高蛋白得率;通过80℃加热去除杂蛋白,大大提高了蛋白提取物活性的稳定性;将“离心”改进为更适合工业化生产的“板框过滤”,更适合大规模工业化生产。
与现有专利相比,本发明具有以下优势:原料方便易得,蛋白提取物活性稳定,提取成本低、得率高,工艺流程简单,适合大规模工业化生产;且该蛋白提取物EMBP在重塑肠道菌群结构、提高肠道乳杆菌的定植和丰度、改善肠道屏障渗漏以及缓解肥胖等方面具有显著功效,可开发为改善肠道健康、缓解肥胖的功能性食品、保健食品、特殊膳食用食品和/或药品等相关产品。

具体实施方式
实施例1:谷糠蛋白提取物(EMBP)的制备方法
(1)称取小米米糠5kg,粉碎后,过60目筛,取谷糠粉。同时在1号罐(50L)加入40L蛋白提取液,所述的蛋白提取液为20mM Tris-HCl溶液,含0.85%NaCl,pH 8.0。开启6℃冷盐水循环系统,于罐中搅拌48h,温度计监测料液温度。
(2)板框过滤,助滤剂珍珠岩用量为2kg,滤液收集至2号罐(50L)。
(3)2号罐加热升温,升温至80℃,并孵育25min,沉淀物析出,降温后用板框过滤,助滤剂及用量同上,收集澄清滤液于1号罐。
(4)向澄清滤液加入20kg的硫酸铵粉末进行沉淀,静置8h,板框过滤,收集硫酸铵沉淀用16L的20mM Tris-HCl,pH 8.0蛋白缓冲液溶解后,重新进行板框过滤,去除珍珠岩,得到含盐的谷糠蛋白提取液。
(5)上述澄清的谷糠蛋白提取液用3kD分子量超滤脱盐。脱盐完毕,料液进行真空冷冻干燥,冻干成粉末;粉末总重为56g,计算谷糠蛋白提取物(EMBP)得率为1.12%,4℃冷藏保存,冻干粉末和提取物(如图1所示)。
实施例2:EMBP预防高脂饮食小鼠肥胖和胰岛素抵抗
1、动物来源:雄性SPF级C57BL/6小鼠;5周龄;
2、饲养条件和饲料:温度为23±2℃,相对湿度为50-55%,室内灯光模拟昼夜,12小时明暗循环。除正常对照组外均为高脂饲料喂养,正常对照组为普通维持饲料喂养,持续喂养12周。EMBP给药组采用灌胃方法,给小鼠每日灌胃不同剂量EMBP,对照和模型组灌胃同等体积生理盐水,直到实验结束。
3、实验动物分组(见表1):

4、实验进行每周对小鼠进行称重并记录,绘制折线图(如图2所示)血糖检测前24小时断饲料并停止给药,只供应水,然后尾静脉取血。使用One-Touch血糖仪检测血糖;将血液收集于无菌干燥管,室温静置半小时,打开离心机,3500rpm室温离心10min,收集上层微黄色液体即为血清,利用ELISA试剂盒(江苏酶免实业有限公司)检测小鼠血清胰岛素;胰岛素抵抗指数用空腹血糖和空腹胰岛素的乘积除以22.5计算(如图3所示)
实施例3:EMBP缓解高脂饮食引起的小鼠肠道屏障渗漏
在体内评估肠道屏障通透性水平利用口服荧光素FITC-Dextran(MW:4000Da)来测定。实验取血:口服FITC后4小时,麻醉并固定小鼠,用镊子快速摘取眼球,并使血液从眼眶内流入无菌EP管中,于室温(37℃)静置半小时以上,打开离心机,3500rpm室温离心10min,收集上层微黄色液体得到血清;检测肠道通透性实验使用多功能酶标仪,在激发波长为493nm,最佳检测波长为518nm检测血清荧光强度;炎症因子检测使用ELISA试剂盒(上海西唐生物科技有限公司)检测各组小鼠血清中炎症因子TNFα和IL-1β的水平。
结果表明:与对照组相比,高脂饮食小鼠肠道通透性显著增加,血清中炎症因子TNFα和IL-1β表达的水平明显上升,即肠道屏障被破坏且有炎症发生。EMBP处理高脂饮食小鼠后,肠道通透性显著降低,血清中炎症因子表达的水平明显减少。(如图4、5所示)。
实施例4:qPCR检测EMBP处理后高脂饮食小鼠肠道紧密连接蛋白表达
小鼠肠组织总RNA的提取:麻醉处死小鼠后,切取适量结肠组织1cm左右,将切好的组织用PBS清洗三遍,浸泡入1mL的Trizol中,匀浆后分装于EP管中;加入200μL氯仿,剧烈振荡15s,静置5min;在13000rpm,4℃下离心15min,至分层(无色水相、酚-氯仿相、浅红色相)吸取上清存放在灭菌的EP管中;加入等体积的异丙醇,上下颠倒离心管,充分混匀,在室温环境下,静置10min;在13000rpm,4℃下离心10min,见白色沉淀;小心倒掉上清,沿管壁缓慢加入1mL 75%的乙醇(可轻轻上下颠倒);在13000rpm,4℃下离心5min,丢弃上清,并用枪头吸干净;吹干沉淀,直至变为透明;加入适量DEPC水,溶解沉淀,在57℃金属浴助溶10min;
RNA浓度检测:使用Nanodrop 2000测定RNA浓度。
RNA模板反转录:将上述方法提取的总RNA,用HiScriptⅡReverse Transcriptase反转录试剂盒,进行反转录,反应条件为:37℃ 15min,85℃ 5s,反应结束后,cDNA存放于-20℃长期保存。
实时定量PCR:设计并合成小鼠源的紧密连接蛋白的引物,使用ChamQ SYBR qPCRMaster Mix试剂盒进行实时定量PCR。
结果表明:与对照组相比,高脂饮食小鼠紧密连接蛋白Claudin-1、Occludin以及ZO-1表达均显著降低,EMBP处理可以明显增加高脂饮食小鼠肠道中紧密连接蛋白的表达水平(如图6所示)。
实施例5:基于llumina平台对各组小鼠肠道菌群基因组进行测序
实验流程主要包括微生物组总DNA提取、目标片段PCR扩增、扩增产物磁珠纯化回收、扩增产物荧光定量、测序文库制备以及上机进行高通量测序。主要采用Illumina MiSeq平台对群落DNA片段进行双端(Paired-end)测序。测序原始数据以FASTQ格式保存。对于细菌的16S rRNA基因,默认Greengenes数据库进行物种分类学注释。
为了能较为全面的评估微生物群落的多样性,本流程通过主坐标分析(Principalcoordinate analysis,PCoA)表征菌群结构(见图7);最后对所有分类水平同时进行差异富集,寻找分组之间的差异标志物种(Biomarker)。最终我们筛选出的标志物种为乳杆菌属(其丰度变化见图8)。
结果表明:与对照组相比,高脂饮食组小鼠改变了菌群结构,EMBP处理高脂饮食小鼠后重塑了小鼠肠道菌群结构;接下来,EMBP处理后,小鼠肠道中乳杆菌属显著上调,EMBP对肠道屏障的维护或与其相关。
实施例6:荧光探针检测EMBP增加肠道乳杆菌的定植
标本处理:麻醉处死小鼠后,立即取整段结肠组织,投入4%多聚甲醛中固定24h,石蜡包埋并切片;切片在50℃下烘干2-3h;浸入70%甲酰胺/2×SSC(柠檬酸钠)变性溶液,在78℃孵育5min;再依次移入80%、90%和100%的预冷酒精内脱水,每次5min;然后空气干燥。
探针杂交:根据乳杆菌的16S rDNA设计合成细菌探针(FAM荧光素双端标记);探针75℃水浴变性后,滴10μL探针在切片上,封片;37℃黑暗湿盒中孵育12h-16h;43℃预热水洗溶液,水洗切片15min;2×SSC(37℃)洗两次,每次10min;切片置于染色缸的1×PBS内待检测,勿干燥;放大杂交信号后,切片加10-20μL DAPI荧光染料,在室温下孵育2-5min并封片;在共聚焦显微镜下观察并拍照。
结果表明:与高脂饮食模型组相比,EMBP处理高脂饮食小鼠后,FAM荧光素探针标记的肠道乳杆菌定植明显增加(如图9所示,标尺为50μm)。
Claims (8)
1.一种谷糠蛋白提取物(EMBP)的制备方法,其特征在于,包括如下步骤:
第一步:将小米谷糠粉碎后,过60目筛,得到谷糠粉,加入植物蛋白提取液于发酵罐中低温搅拌24-64h;
第二步:板框过滤,收集滤液;
第三步:滤液加热升温至80℃,孵育20-30min,待沉淀物析出,降温至室温后进行板框过滤,收集澄清滤液;
第四步:澄清滤液加入硫酸铵进行沉淀,静置,板框过滤后收集沉淀,再用蛋白缓冲液溶解,重新板框过滤去除珍珠岩,获得含盐谷糠蛋白提取物;
第五步:将上述含盐谷糠蛋白提取物用3kD分子量的超滤装置脱盐,料液进行真空冷冻干燥,冻干成粉末即为谷糠蛋白提取物,于4℃保存。
2.如权利要求1所述的谷糠蛋白提取物的制备方法,其特征在于,第一步中所述的植物蛋白提取液为20mM Tris-HCl溶液,含0.85%NaCl,pH 8.0;所述的谷糠和蛋白提取液的重量体积比为1︰7-9,其中重量的单位为kg,体积的单位为L;所述的低温搅拌的温度:4-6℃,时间:48-64h。
3.如权利要求1所述的谷糠蛋白提取物的制备方法,其特征在于,第二步中所述的板框过滤的助滤剂为珍珠岩,用量按每升蛋白提取液40-60g珍珠岩计。
4.如权利要求1所述的谷糠蛋白提取物的制备方法,其特征在于,第三步中所述的孵育时间为25min。
5.如权利要求1所述的谷糠蛋白提取物的制备方法,其特征在于,第四步中所述的硫酸铵的饱和度为70-80%;所述的蛋白缓冲液为20mM Tris-HCl溶液,pH 8.0;静置时间为:6-8h。
6.如权利要求1-5任一方法制备的谷糠蛋白提取物。
7.如权利要求6所述的谷糠蛋白提取物在制备改善肠道健康和/或缓解肥胖的功能性食品或特殊膳食用食品中的应用。
8.如权利要求6所述的谷糠蛋白提取物在制备改善肠道健康和/或缓解肥胖的保健食品或药品中的应用。
