Abstract
The invention relates to the technical field of cosmetics, and discloses a traditional Chinese medicine extract composition with an acne removing effect and a preparation method thereof, wherein the composition comprises extracts of salvia miltiorrhiza, sophora flavescens, coptis chinensis, centella asiatica and andrographis paniculata, and the content of each medicinal material is as follows according to the dry weight percentage of the medicinal materials: 1-30% of salvia miltiorrhiza, 1-30% of radix sophorae flavescentis, 1-30% of coptis chinensis, 1-30% of centella asiatica and 1-30% of andrographis paniculata; the composition of the traditional Chinese medicine extract can realize multi-path antibiosis and antiphlogosis and relieve acne symptoms, is green and efficient, and does not contain hormones, so that irritation and sensitization of chemical preparations to skin are avoided.

Technical Field
The invention relates to the technical field of cosmetics, in particular to a traditional Chinese medicine extract composition with an acne removing effect and a preparation method thereof.
Background
Acne is commonly known as acne, and is treated academically. Acne is a chronic inflammatory skin disease of hair follicles and sebaceous glands, and is also one of the most common diseases in the cosmetology dermatology department. After acne occurs, pigmentation, persistent erythema, depressed or hypertrophic scars are easily caused, the appearance is influenced, and the acne is a beautiful natural enemy.
The current acne research shows that the psychological pressure and the working pressure are increased; high oil and high sugar eating habits; stay up at night, and play bad daily work and rest such as mobile phones excessively; improper use of skin care products and cosmetics, etc., can lead to acne. Acne is not limited by age, and people of all ages, except adolescents in adolescence, may suffer from it. Therefore, alleviating and treating the acne problem is a hot spot of current cosmetic research.
The pathogenesis of acne is closely related to factors such as excessive sebum secretion, follicular sebaceous gland duct keratosis, blockage, propionibacterium acnes infection and inflammatory reaction. At present, a plurality of chemical acne removing products are available on the market, and most products are added with antibiotics, retinoic acid and androgens to achieve the treatment effect. However, such products are irritating and tend to cause skin discomfort during treatment. While concurrent hormonal therapy can alleviate and treat the acne problem and symptoms within a certain period of time, if improperly used, the condition is likely to be exacerbated. Therefore, the problem of skin acne is very worthy of research by adopting a new way and a new method.
Disclosure of Invention
The invention aims to provide a traditional Chinese medicine extract composition with an acne removing effect and a preparation method thereof, and aims to solve the problem that an acne removing product in the prior art is irritant.
In a first aspect, the application provides a preparation method of a traditional Chinese medicine extract composition with acne removing efficacy, which comprises the following steps:
s1, mixing the raw materials according to a molar ratio of 3: 4: 1: 2, respectively weighing betaine, glycerol, 1, 3-butanediol and hexanediol, stirring at 50 ℃ until the mixture is colorless and transparent to obtain a deep eutectic solvent, and adding a certain amount of water to prepare a deep eutectic solvent aqueous solution;
s2, weighing the salvia miltiorrhiza, the sophora flavescens, the coptis chinensis, the centella asiatica and the andrographis paniculata, wherein the contents of the medicinal materials are as follows according to the dry weight percentage of the medicinal materials: 1-30% of salvia miltiorrhiza, 1-30% of radix sophorae flavescentis, 1-30% of coptis chinensis, 1-30% of centella asiatica and 1-30% of andrographis paniculata; soaking the dry powder of the traditional Chinese medicine composition in the deep eutectic solvent aqueous solution obtained in the step S1 to obtain a mixed solution;
s3, purifying the mixed solution by using macroporous adsorption resin, and collecting eluent after elution;
s4, concentrating the eluent, adding 1, 3-butanediol, stirring uniformly, standing for 24 hours, and collecting the supernatant;
s5, centrifuging the supernatant to obtain a centrifugal supernatant, wherein the centrifugal supernatant contains the traditional Chinese medicine extract composition with the acne removing effect.
As an embodiment, step S2 specifically includes the following steps:
s21, weighing 1-30 parts of salvia miltiorrhiza, 1-30 parts of radix sophorae flavescentis, 1-30 parts of coptis chinensis, 1-30 parts of centella asiatica and 1-30 parts of common andrographis herb by taking grams as a unit, and weighing the total weight of all the weighed medicinal materials as a numerical value A;
s22, recording the volume of the deep eutectic solvent aqueous solution as a numerical value B in milliliters, wherein the volume ratio of the deep eutectic solvent aqueous solution is 10-95%;
s23, measuring a certain amount of deep eutectic solvent water solution to enable the value B to be 5-20 times of the value A;
s24, soaking and extracting the measured medicinal materials in a deep eutectic aqueous solvent for 2-48 hours each time for 1-3 times, and combining the mixed extracting solutions.
As an embodiment, in step S3, the eluent used for elution is 1, 3-butanediol aqueous solution, and the volume ratio of the 1, 3-butanediol aqueous solution is 5% -95%.
As an embodiment, step S4 specifically includes:
concentrating the eluate by 3-20 times, adding 1, 3-butanediol of 1-3 times of the volume of the eluate, stirring, standing for 24 hr, and collecting supernatant;
in one embodiment, in step S5, the centrifugation speed is 5000r/min-10000r/min, and the centrifugation time is 5-10 minutes.
In a second aspect, the application also provides a traditional Chinese medicine extract composition with acne removing efficacy, and the composition is prepared by the preparation method.
Compared with the prior art, the method has the following beneficial effects:
(1) the active ingredients of red sage root are mainly divided into two categories of fat-soluble and water-soluble. Wherein the fat-soluble component comprises phenanthrenequinone derivatives such as tanshinone; the water soluble components include protocatechualdehyde and danshensu. Wherein the fat-soluble tanshinone has antibacterial and antiinflammatory effects; the water-soluble salvianolic acid can resist oxidation and protect cells.
(2) The chemical components of the sophora flavescens mainly comprise two main classes of alkaloids and flavones, wherein the alkaloid compounds can obviously increase the secretion of IFN-gamma of HaCaT cells stimulated by substance P. Has effects of eliminating dampness and relieving itching; prenylation of flavonoids can enhance their antibacterial activity.
(3) The coptis contains various isoquinoline alkaloids, the content of berberine is the highest and is 5% -8%, and the coptis further contains coptisine, methyl coptisine, epiberberine, magnoflorine and the like, and the components enable the coptis to have obvious antibacterial and anti-inflammatory effects.
(4) Centella asiatica contains various triterpenes, and its main ingredients are asiaticoside and madecassoside. The skin-care product has the effects of tightening the connecting part of the epidermis and the dermis, softening the skin, promoting the formation of collagen in the dermis and tightening the skin.
(5) Andrographis paniculata Nees contain mainly diterpene lactones, wherein andrographolide is used as the substance basis of the main pharmacological activity of Andrographis paniculata Nees. The andrographolide components have effect in inhibiting Diplococcus pneumoniae, inhibiting capillary permeability increase due to inflammation, and improving exudation and edema.
(6) The traditional Chinese medicine extract composition with the acne removing effect is obtained by extracting the traditional Chinese medicines through a deep eutectic solvent, and can achieve multiple ways of antibiosis and antiphlogosis and relieve acne symptoms. The product is green and efficient, and does not contain hormones, so that irritation and sensitization of chemical preparations to skin are avoided.

The following describes the implementation of the present invention in detail with reference to specific embodiments.
Example 1
A traditional Chinese medicine extract composition with acne removing effect is weighed according to dry weight of medicinal materials and comprises 20 g of salvia miltiorrhiza, 20 g of sophora flavescens, 20 g of coptis chinensis, 20 g of centella and 20 g of common andrographis herb.
A preparation method of a traditional Chinese medicine extract composition with an acne removing effect specifically comprises the following steps:
s1, mixing the raw materials according to a molar ratio of 3: 4: 1: 2, respectively weighing betaine, glycerol, 1, 3-butanediol and hexanediol, stirring at 50 ℃ until the mixture is colorless and transparent to obtain a deep eutectic solvent, and adding a certain amount of water to prepare a deep eutectic solvent aqueous solution;
s2, weighing 20 g of salvia miltiorrhiza, 20 g of radix sophorae flavescentis, 20 g of coptis chinensis, 20 g of centella asiatica and 20 g of andrographis paniculata according to dry weight of medicinal materials, soaking and extracting the dry powder of the traditional Chinese medicine composition by 1000 ml of the deep eutectic solvent aqueous solution obtained in the step S1, wherein the volume ratio of the deep eutectic solvent aqueous solution is 50%, the extraction times are 1 time, and the mixed solution solvent is obtained after soaking for 12 hours each time;
s3, purifying with macroporous adsorption resin, eluting, collecting eluent, wherein the eluent for the eluting step is 50% 1, 3-butanediol aqueous solution.
S4, concentrating the eluent by 3 times, adding 1, 3-butanediol by 2 times, stirring uniformly, standing for 24 hours, and collecting the supernatant;
s5, centrifuging the supernatant at a speed of 5000r/min for 5min to obtain a centrifuged supernatant, wherein the centrifuged supernatant contains the traditional Chinese medicine extract composition with the acne removing effect.
Example 2
A preparation method of a traditional Chinese medicine extract composition with an acne removing effect specifically comprises the following steps:
s1, mixing the raw materials according to a molar ratio of 3: 4: 1: 2, respectively weighing betaine, glycerol, 1, 3-butanediol and hexanediol, stirring at 50 ℃ until the mixture is colorless and transparent to obtain a deep eutectic solvent, and adding a certain amount of water to prepare a deep eutectic solvent aqueous solution;
s2, weighing 25 g of salvia miltiorrhiza, 25 g of radix sophorae flavescentis, 20 g of coptis chinensis, 15 g of centella asiatica and 15 g of andrographis paniculata according to dry weight of medicinal materials, soaking and extracting the Chinese medicinal composition dry powder with 1500 ml of the deep eutectic solvent aqueous solution obtained in the step S1, wherein the volume ratio of the deep eutectic solvent aqueous solution is 60%, the extraction times are 2 times, and 24 hours of soaking each time is carried out to obtain a mixed solution solvent;
s3, purifying with macroporous adsorption resin, eluting, collecting eluent, wherein the eluent used in the elution step is 30% 1, 3-butanediol aqueous solution.
S4, concentrating the 6 times of eluent, adding 1.5 times of 1, 3-butanediol, stirring uniformly, standing for 24 hours, and collecting the supernatant;
s5, centrifuging the supernatant at 6000r/min for 8min to obtain a centrifuged supernatant, wherein the centrifuged supernatant contains the traditional Chinese medicine extract composition with the acne removing effect.
Example 3
A preparation method of a traditional Chinese medicine extract composition with an acne removing effect specifically comprises the following steps:
s1, mixing the raw materials according to a molar ratio of 3: 4: 1: 2, respectively weighing betaine, glycerol, 1, 3-butanediol and hexanediol, stirring at 50 ℃ until the mixture is colorless and transparent to obtain a deep eutectic solvent, and adding a certain amount of water to prepare a deep eutectic solvent aqueous solution;
s2, weighing 25 g of salvia miltiorrhiza, 25 g of radix sophorae flavescentis, 20 g of coptis chinensis, 15 g of centella asiatica and 15 g of andrographis paniculata according to dry weight of medicinal materials, soaking and extracting the dry powder of the traditional Chinese medicine composition by 2000 ml of the deep eutectic solvent aqueous solution obtained in the step S1, wherein the volume ratio of the deep eutectic solvent aqueous solution is 70%, the extraction times are 3 times, and 15 hours of soaking are carried out each time to obtain a mixed solution solvent;
s3, purifying with macroporous adsorption resin, eluting, collecting eluent, wherein the eluent for the eluting step is 60% 1, 3-butanediol aqueous solution.
S4, concentrating 10 times of eluent, adding 3 times of 1, 3-butanediol, uniformly stirring, standing for 24 hours, and collecting supernatant;
s5, centrifuging the supernatant at a speed of 10000r/min for 10min to obtain a centrifuged supernatant, wherein the centrifuged supernatant contains the traditional Chinese medicine extract composition with the acne removing effect.
Example 4
A preparation method of a traditional Chinese medicine extract composition with an acne removing effect specifically comprises the following steps:
s1, mixing the raw materials according to a molar ratio of 3: 4: 1: 2, respectively weighing betaine, glycerol, 1, 3-butanediol and hexanediol, stirring at 50 ℃ until the mixture is colorless and transparent to obtain a deep eutectic solvent, and adding a certain amount of water to prepare a deep eutectic solvent aqueous solution;
s2, weighing 15 g of salvia miltiorrhiza, 30 g of radix sophorae flavescentis, 10 g of coptis chinensis, 30 g of centella asiatica and 15 g of andrographis paniculata according to dry weight of medicinal materials, soaking and extracting the dry powder of the traditional Chinese medicine composition by 1200 ml of deep eutectic solvent aqueous solution obtained in the step S1, wherein the volume ratio of the deep eutectic solvent aqueous solution is 65%, the extraction times are 1 time, and 48 hours of soaking are carried out each time to obtain a mixed solution solvent;
s3, purifying with macroporous adsorption resin, eluting, collecting eluent, wherein the eluent used in the elution step is 10% 1, 3-butanediol aqueous solution.
S4, concentrating the eluent by 3 times, adding 1, 3-butanediol by 2 times, stirring uniformly, standing for 24 hours, and collecting the supernatant;
s5, centrifuging the supernatant at 8000r/min for 6min to obtain a centrifuged supernatant containing the Chinese medicinal extract composition with acne removing effect.
In order to verify the acne removing efficacy of the patent, the following experiments are further carried out:
the composition obtained in the above examples was subjected to an acute skin irritation test and a plurality of skin irritation tests, according to the skin irritation procedure of "2007 edition of cosmetic hygiene standards". The compositions obtained in the examples were then used for therapeutic tests, and the results of the tests were counted. The extracts obtained in example 1, example 2, example 3 and example 4 were respectively the herb extracts A, B, C, D, and the herb extract compositions obtained in the respective experimental examples were subjected to the following verification tests:

test subjects, 70, were enrolled and all subjects were asked to have a distinct inflamed pox in the same location on the cheek (determined in advance by the experimenter). The subjects cleaned the face with facial cleanser of the same brand before the experiment, and then applied the herbal extract a obtained in experimental example 1 to the cleaned pox uniformly, once a day, in the morning and at the evening, for a week. The tested part is observed before the sample is tried, on the first day of the trial, on the third day of the trial and after one week of the trial, and the number of people with different degrees of inflamed pox is recorded.

The deep eutectic solvent is a low-temperature eutectic mixture formed by combining a hydrogen bond acceptor (such as quaternary ammonium salt) and a hydrogen bond donor (such as carboxylic acid, polyalcohol and phenolic compound) in a certain stoichiometric ratio. The preparation process of the deep eutectic solvent is simple to operate, and the raw materials are mixed according to a certain molar ratio at normal pressure and low temperature (30-80 ℃) generally and stirred to obtain transparent and uniform liquid. The deep eutectic solvent also has the advantages of easily available materials, low price, no toxicity and easy degradation. Compared with the traditional extraction solvent, the deep eutectic solvent has higher extraction rate, and can simultaneously extract water-soluble and oil-soluble components. Therefore, the deep eutectic solvent is used for extracting the traditional Chinese medicine with complex components, which is more beneficial to obtaining the active components of the traditional Chinese medicine.
The application has the following beneficial effects:

s3, purifying the mixed solution by using macroporous adsorption resin, and collecting eluent after elution;
s4, concentrating the eluent, adding 1, 3-butanediol, stirring uniformly, standing for 24 hours, and collecting the supernatant;
s5, centrifuging the supernatant to obtain a centrifugal supernatant, wherein the centrifugal supernatant contains the traditional Chinese medicine extract composition with the acne removing effect.
2. The preparation method of the traditional Chinese medicine extract composition with the acne removing effect according to claim 1, wherein the step S2 specifically comprises the following steps:
s21, weighing 1-30 parts of salvia miltiorrhiza, 1-30 parts of radix sophorae flavescentis, 1-30 parts of coptis chinensis, 1-30 parts of centella asiatica and 1-30 parts of common andrographis herb by taking grams as a unit, and weighing the total weight of all the weighed medicinal materials as a numerical value A;
s22, recording the volume of the deep eutectic solvent aqueous solution as a numerical value B in milliliters, wherein the volume ratio of the deep eutectic solvent aqueous solution is 10-95%;
s23, measuring a certain amount of deep eutectic solvent water solution to enable the value B to be 5-20 times of the value A;
s24, soaking and extracting the measured medicinal materials in a deep eutectic aqueous solvent for 2-48 hours each time for 1-3 times, and combining the mixed extracting solutions.

concentrating the eluate by 3-20 times, adding 1, 3-butanediol of 1-3 times of the volume of the eluate, stirring, standing for 24 hr, and collecting supernatant.
5. The method for preparing the traditional Chinese medicine extract composition with the acne removing effect according to claim 1, wherein in step S5, the centrifugation speed is 5000r/min to 10000r/min, and the centrifugation time is 5 to 10 minutes.
