肠道菌群是在健康的人和动物的胃肠道内定居着的许多种类和一定数量的微生物的统称,以双歧杆菌为优势菌种,包括双歧杆菌、乳杆菌、肠球菌、肠杆菌、链球菌、葡萄球菌、梭状芽孢杆菌等种类,其不仅承担着消化和营养功能,而且在免疫调节和神经调节中都有作用。肠道菌群失调可能导致一系列消化道疾病,甚至可能与动脉粥样硬化、肥胖、糖尿病、精神疾病、多发性硬化、阿尔茨海默症等相关。
目前调节肠道菌群的方法主要为抗生素、益生元(多糖)、益生菌、肠道菌群移植等。其中可以方便口服施用的抗生素杀菌广谱性和耐药性使得其不适合作为长期调节/非紧急治疗应用;益生菌制剂由于保存条件苛刻,以及经过胃时的损失,往往效果不佳(有报道陈市售大多数益生菌剂活菌数都达不到国家标准和产品说明书标注);而纯低聚糖的益生元制剂促进多种益生菌生长的效果一般不佳,并且没有对条件致病菌和病原菌的抑制作用。
调节肠道菌群这样的长期精细调理方面,中医药有着天然的优势。五谷虫是中医药中治疗各种消化疾病常用而的成分,五谷虫是指丽蝇科大头金蝇或类似种类的幼虫,此药名始见于明朝,《本草纲目》、《本草求真》、《本草备要》等著作中认为其性寒,味甘、咸、苦,如脾胃经,有健脾化食去热消疳的功效,可用于治疗疳积腹大、翳膜遮睛、热病谵妄、毒痢作吐。现代中药研究发现,五谷虫或其中某些提取物抑菌、抗炎、抗肿瘤、降血脂等功效,也有五谷虫用于调节肠道菌群的报道,但结果显示,炒制的五谷虫调节肠道菌群作用并不显著。

三发明内容
从中医上五谷虫治疗多种消化道疾病良好疗效出发,发明人进一步研究了五谷虫对肠道菌群的调节作用,研究中发现,经过晾晒、炒制等过程,五谷虫炮制品中几乎不含双歧杆菌等益生菌,其对肠道菌群的调节作用主要依赖于其中的多糖和蛋白成分对益生菌生长的促进作用以及某些蛋白成分对某些非益生菌的抑制作用。为增强上述作用,申请人根据现有对五谷虫的研究,选择使用蛋白酶和甲壳素酶处理五谷虫后在进行常规炮制,有效提高了五谷虫对益生菌生长的促进作用。
一方面,本发明提供了一种五谷虫炮制品,其特征在于,炮制方法中包括甲壳素酶处理。
进一步地,炮制方法中包括甲壳素酶50U/g虫体处理2h。
进一步地,炮制方法中还包括碱性蛋白酶处理。
进一步地,炮制方法中包括碱性蛋白酶20U/g虫体处理2h。
进一步地,炮制方法中还包括清洗、炒制、粉碎步骤。
进一步地,炮制方法具体为:将五谷虫称重、清洗、捣碎;加入虫体重量3倍的水,按照50U/g虫体的量加入甲壳素酶,室温下处理2h后;滤去水分,文火炒至金黄色;粉碎后过50目筛。
进一步地,炮制方法具体为:将五谷虫称重、清洗、捣碎;加入虫体重量3倍的水,按照50U/g虫体和20U/g虫体的量加入甲壳素酶和碱性蛋白酶,室温下处理2h后;滤去水分,文火炒至金黄色;粉碎后过50目筛。
另一方面,本发明提供了一种炮制五谷虫的方法,具体步骤为:将五谷虫称重、清洗、捣碎;加入虫体重量3倍的水,按照50U/g虫体和20U/g虫体的量加入甲壳素酶和碱性蛋白酶,室温下处理2h后;滤去水分,文火炒至金黄色;粉碎后过50目筛。
另一方面,本发明还提供了一种药物组合物,包含本发明上述的五谷虫炮制品。
另一方面,本发明提供了本发明上述的五谷虫炮制品和药物组合物在制备调节肠道菌群的药物中的应用。
本发明的五谷虫可以为大头金蝇(Chrysomyia megacephala)幼虫,也可以为丽蝇科类似物种的幼虫。本发明的五谷虫可以使用任何符合卫生标准的饲料,如麸皮、谷壳、各种粮食、植物叶、骨粉、肉粉等饲养。
本发明的药物组合物或以本发明炮制品制备的药物可以为各种适合的药物/保健品/食品剂型,主要为口服剂型,包括但不限于汤剂、丸剂、散剂、酒剂、膏剂、锭剂、片剂、胶囊剂、混悬剂等,也可用于灌肠液、鼻饲液、胃饲液等,本领域技术人员可以根据药剂领域相关知识做出常规选择。
在制备上述药物时,本领域技术人员可以根据药剂学常识为本申请的中药组合物选用适合的辅料,包括但不限于溶剂、促溶剂、稳定剂、抗氧化剂、增稠剂、表面活性剂、包衣剂、分散剂、填充剂等。
调节肠道菌群的药物可用于各种与肠道菌群相关的疾病的治疗,包括但不限于各种中医上的疳积、西医上的消化不良、肠炎、肠应激综合征、动脉粥样硬化、肥胖、精神疾病等现有技术已知或未来发现的与肠道菌群相关的疾病。
四具体实施方式
主要试剂:
食品级甲壳素酶:200000U/g,武汉宏信康精细化工有限公司生产;
食品级碱性蛋白酶:250000U/g,南宁东恒华道生物科技有限责任公司生产;
五谷虫样品:申请人企业自行养殖,虫品种为经过鉴定的大头金蝇,饲料主要为为麸皮和谷壳,取三龄幼虫(指取五谷虫用于炮制时,容器中的大多数幼虫为三龄幼虫);
低聚果糖/固体益生元:固体饮料粉,表明成分为低聚果糖,昆明加加宁生物制品有限公司;
培养基:双歧杆菌用BBL培养基、乳杆菌用MRS培养基、肠杆菌用EMB培养基、肠球菌用胆汁七叶苷叠氮钠培养基均购自上海博钰生物科技有限公司。
其与试剂和仪器如未指明均为常规国产型号/种类。
实施例1五谷虫虫粉的制备
各虫粉的制备使用同一容器中同一批次繁殖的五谷虫进行。
虫粉1:
将五谷虫称重、清洗、捣碎;加入虫体重量3倍的水,按照50U/g虫体和20U/g虫体的量加入甲壳素酶和碱性蛋白酶,室温下处理2h后;滤去水分,文火炒至金黄色;粉碎后过50目筛。
虫粉2:
将五谷虫称重、清洗、捣碎;加入虫体重量3倍的水,按照50U/g虫体的量加入甲壳素酶,室温下处理2h后;滤去水分,文火炒至金黄色;粉碎后过50目筛。
虫粉3:
将五谷虫称重、清洗、晒干;文火炒至金黄色;粉碎后过50目筛。
实施例2五谷虫粉对小鼠肠道菌群的影响
C57BL/6小鼠购自上海凯学生物科技有限公司。
选取饮食和活动状态良好的健康小鼠80只,随机分为7组。7组分别给予0.5mg/只虫粉1、0.25mg/只虫粉1、0.5mg/只虫粉2、0.25mg/只虫粉2、0.5mg/只虫粉3、0.25mg/只虫粉3、0.5mg/只低聚果糖、0.5mg/只蒸馏水(每日一次,虫粉加入1mL蒸馏水中灌喂)。
无菌收集给药前1日的粪便和最后一次给药后一日的粪便,生理盐水稀释后选取适合的稀释液接种于各培养基上,厌氧培养48h后分析菌种类,计算菌落数和每个粪便中的菌数的对数(Lg CFU/g)。
*统计学上显著(P<0.05)
**统计学上极显著(p<0.01)
对五谷虫粉和市售低聚糖产品的结果与现有文献基本一致,直接晒干后炒制的五谷虫粉似有增加益生菌的效果,但几无统计学意义。低聚糖产品对双歧杆菌效果显著,但对乳杆菌效果不明显,且无抑制条件致病菌的作用。经过甲壳素酶处理后炮制的五谷虫粉在低剂量和高剂量下的效果均优于直接晒干后炒制的五谷虫粉。经过甲壳素酶和碱性蛋白酶联合处理后炮制的五谷虫粉则实现了对乳杆菌和双歧杆菌的显著促进,并能显著抑制两种条件致病菌,这种作用在高剂量时(基本等同于医书中1到2钱的人用剂量)时达到了极显著的水品。
上述数据证明了甲壳素酶处理或甲壳素酶和碱性蛋白酶联合处理后能有效提高五谷虫调节肠道菌群的能力。
实施例3小鼠肠推进实验
小鼠种类为C57BL/6,购自上海凯学生物科技有限公司。
取20g±2g饮食和活动状态良好的健康雄性小鼠30只,平均分为3组;第1-7日,分别按照每次0.5mg/只(加入1ml水中灌胃给药)每日两次灌喂蒸馏水、虫粉1或虫粉2;
第2-7日,按照0.004mg/只皮下注射利血平制造疳积模型(参考国内现有技术中小鼠脾虚疳积模型的制备方法);
第8日,灌喂1ml含10%活性炭末的水,30min后处死动物,取幽门至回盲肠部的组织,将肠管铺成直线,量取小肠总长度和幽门至炭末前沿的距离,计算小肠炭末推进百分率。
肠推进率=幽门至炭末前沿距离/小肠总长度。
结果如下表
上述数据证明了五谷虫粉确有如中医著作中所记载的改善消化的功能,而且经过甲壳素酶和碱性蛋白酶联合处理,这种功能得以进一步强化,这可能与酶解产生的某些小肽或寡糖相关,有进一步研究的价值。
Claims (3)
1.一种五谷虫炮制品在制备调节肠道菌群的药物中的应用,其特征在于,所述五谷虫炮制品的炮制方法中包括甲壳素酶50U/g虫体处理2h,以及碱性蛋白酶20U/g虫体处理2h。
2.根据权利要求1的应用,其特征在于,所述五谷虫炮制品的炮制方法中还包括清洗、炒制、粉碎步骤。
3.根据权利要求1或2的应用,其特征在于,所述五谷虫炮制品的炮制方法为:将五谷虫称重、清洗、捣碎;加入虫体重量3倍的水,按照50U/g虫体和20U/g虫体的量加入甲壳素酶和碱性蛋白酶,室温下处理2h后;滤去水分,文火炒至金黄色;粉碎后过50目筛。
