Abstract
本发明属于轻工食品加工技术领域,具体涉及一种食管癌围术期患者专用的食品其制备方法。本发明的方法包括以下的步骤:(1)复合酶解制备水溶性植物功能多糖;(2)制备微胶囊芯材;(3)复合凝聚微胶囊的制备;(4)围术期必需蛋白质超微粉的制备;(5)复配蛋白质超微粉与复合凝聚微胶囊。本发明的有益效果在于:提供了免疫营养素;复合酶解制备含有抗肿瘤细胞因子的植物功能多糖;具有生物调节应答功效的益生菌冻干粉的引入;复合凝聚微胶囊;粉体高效复配。

技术领域
本发明属于轻工食品加工技术领域,具体涉及一种食管癌围术期患者专用的食品其制备方法。
背景技术
食管癌作为消化道肿瘤中常见的一种病症,临床表现为咽下困难并伴有严重的消化不良等症状。患者多因吞咽困难,存在不同程度的水电解质紊乱。
当前国内外对于食管癌的发病原因及机制尚未明确与解析,但众多报道中均已指出食管癌的发生、发展、围术期治疗等方面与肿瘤细胞所处的微环境及生物体体内免疫系统密切相关。种种迹象表明,当生物体内抗肿瘤细胞因子(如干扰素、白细胞介素、肿瘤坏死因子等)含量降低时,针对突变细胞的防御及清除能力大幅降低,最终引发肿瘤分化、增殖及转化。而抗肿瘤细胞因子可在激活肿瘤细胞免疫应答的前提下,诱导肿瘤细胞分化、凋亡,最终达到肿瘤治疗的目的。目前利用细胞因子进行肿瘤治疗虽已取得一些成果,但因严重的毒副作用影响了其在临床上的应用。
营养支持作为一种以解决营养需求为出发点间接进行抗肿瘤治疗的举措,因方法简便低廉、临床疗效佳、针对性强等优点而在围术期广泛采用。肠内营养(EN)因更符合生理、有利于胃肠功能、营养及免疫力的恢复、预防术后并发症,并可根据不同病人对营养的需求进行定制化设计,做到“一人一配方”,而成为营养支持的首选。肠内营养支持以饮食营养均衡为基本原则,以功能活性成分为辅,在蛋白质、脂肪、碳水化合物之间比例合理,无机盐、电解质及维生素满足患者营养需求的基础上,通过功能活性成分的引入达到辅助治疗的目的。本发明所制备开发的肠内营养配方专门针对食管癌围术期病人的营养需求,囊括了含有抗肿瘤细胞因子的免疫营养素、益生菌剂、植物功能多糖,高蛋白、高脂肪、低碳水化合物等营养成分,可在防治营养缺乏的基础上,改善患者机体免疫功能,调节机体应激反应,提高患者治疗疗效。
但是,肠内营养配方仅是肠内营养支持的前提,而肠内营养支持利用率是保证疗效正常发挥的关键点。通过技术手段使营养成分在一段时间内可以持续、均匀的释放,这是提高肠内营养支持利用率,增强肠内营养支持疗效最行之有效的方法。微胶囊技术作为一种可控制有效成分在不同靶点有效释放的缓释技术,是将一些具有反应活性、敏感或易挥发的物质用各种天然或合成的高分子材料包埋形成微小粒子的技术。但传统微胶囊在耐高温、载样量、pH敏感性、机械性能等方面有所欠缺,导致该类产品易破损、营养易流失、缓释效果不佳、靶向性不强。近年来所开发的复合凝聚微胶囊技术,因载样量高、pH敏感、温度响应性好、机械性能佳而引起各行业的广泛关注。该类微胶囊是利用带相反电荷的高分子材料间的静电相互作用自组装所形成,相对于传统微胶囊利用亲、疏水作用进行的简单包埋而言,该类微胶囊中富含有大量的自组装体系(亲、疏水作用、静电相互作用、氢键相互作用等),对提高载样量、缓释率、pH敏感性等方面具有积极的作用,在肠内营养支持方面具有巨大的潜在应用价值。
发明内容
为了解决上述的技术问题,本发明提供了一种适于食管癌围术期患者肠道的专用食品的制备方法;
本发明所提供的适于食管癌围术期患者专用食品的制备方法,包括下述的步骤:
(1)复合酶酶解制备水溶性植物功能多糖
清洗香菇、银耳、冬虫、夏草;控干水分,并打浆,得浆液;
将所得浆液灭菌处理,在灭菌后的浆液中加入复合酶酶解4-8h,离心,取上清液,获得植物功能多糖溶液;
(2)制备微胶囊芯材
将复合菌剂冻干粉、功能性氨基酸、水溶性复合维生素、复合矿物质溶于(1)中所得的植物功能多糖溶液中,得混合物Ⅰ,在高速匀浆过程中滴入乳化剂和植物源营养素组成的混合溶液,得到复合凝聚体系;
在滴入乳化剂时将HLB值控制在10-13.5之间;
(3)复合凝聚微胶囊的制备
将(2)中所得到的复合凝聚体系在高速搅拌的条件下,加入到复合壁材溶液中,得到复合凝聚微胶囊预备液,高压均质进行二次混合处理,获得微胶囊分散液;
将该微胶囊分散液静置20-24h后,弃去上清液,得到微胶囊湿囊,离心喷雾干燥,得复合凝聚微胶囊;
(4)围术期必需蛋白质超微粉的制备
对食管癌围术期肠内营养必需的蛋白质进行超微气流粉碎预处理,过200-600目筛,得到蛋白质超微粉;
(5)复配蛋白质超微粉与复合凝聚微胶囊
将(4)中所得复合凝聚微胶囊、(5)中所得蛋白质超微粉进行粉体混合处理,得到食管癌围术期患者专用食品。
(1)中,香菇、银耳、冬虫、夏草的质量比例为2-3∶3-5∶1-3∶1.5-3;
打浆时固液比为1∶5-1∶10;
复合酶解由纤维素酶、淀粉酶、果胶酶组成,纤维素酶、淀粉酶与果胶酶的质量比分别为2-4∶1-3∶3-5,酶解时间为4-8h,酶解温度为30-50℃;
离心转速控制在4000-6000r/min,离心20—30min。
(2)中,复合菌剂冻干粉为双歧杆菌、酪酸梭菌、鼠李糖乳杆菌、乳酸杆菌中的至少两种;
功能性氨基酸为L-谷氨酰胺、组氨酸、精氨酸、赖氨酸、天冬氨酸、谷氨酸中的至少一种;
水溶性复合维生素为维生素B12、维生素B6、维生素C、维生素D、维生素A的至少两种;
复合矿物质为葡萄糖酸钙、葡萄糖酸锌、葡萄糖酸镁、葡萄糖酸亚铁、氯化钠中的至少两种;
乳化剂为Span80、大豆卵磷脂、单甘脂、聚氧乙烯蓖麻油、聚氧乙烯失水山梨醇单油酸酯中的至少一种;
植物源营养素为亚麻籽油、维生素E、番茄红素、β-胡萝卜素、大豆异黄酮中的至少一种。
复合菌剂冻干粉、功能性氨基酸、水溶性复合维生素、复合矿物质的质量比为3-5∶1-3∶0.5-2.5∶0.3-0.7;
上述复合菌剂冻干粉、功能性氨基酸、水溶性复合维生素、复合矿物质与复合酶解上清液的质量体积比为:3-6∶20;
乳化剂和植物源营养素所组成的混合溶液的体积浓度为20-35%;
混合物Ⅰ与植物源营养素和乳化剂所组成的混合溶液的质量比为1-3∶4-8;
乳化剂HLB值控制在10-13.5之间,其占复合凝聚体系质量的0.01-0.05%;
高速匀浆步骤中转速为10000-15000r/min,匀浆时间为10-20min。
复合凝聚微胶囊体系中复合壁材为β-环糊精,辛烯基琥珀酸淀粉酯,壳聚糖,阿拉伯胶,明胶,羟乙基纤维素中的至少一种;
芯材复合凝聚体系占壁材总质量的30-60%;
高压均质压力为20—45MPa,均质时间为3—7min;
离心喷雾干燥进风口温度为180—220℃,出风口温度为70—90℃,进料流量为20-35mL/min。
围术期必需蛋白质包括大豆蛋白低聚肽、乳清蛋白、酪蛋白中的至少一种超微气流粉碎压力为0.5—1MPa,转速为1000—2000r/min,粉碎后过200-600目筛分离、细化。
复合凝聚微胶囊与蛋白质超微粉的质量比为0.5-3∶2.5-5;高速气流冲击式粉体机冲击力为5-15Kw,处理时间为5-10min,处理温度为30—60℃。
喷雾干燥对湿囊进行处理后所得复合凝聚微胶囊粒径尺寸<100μm。
采用高速气流冲击式粉体机将复合凝聚微胶囊和蛋白质超微粉进行粉体混合。
本发明的有益效果在于:
(1)提供了免疫营养素:免疫营养素作为一类特殊营养素,可以通过特定方式刺激免疫细胞,增强机体免疫应激能力,维持正常、适度的免疫反应,调整细胞因子的生产和释放,对减轻炎症反应,保护肠屏障功能完整性具有显著功效。本发明在该产品开发过程中,引入氨基酸、不饱和脂肪酸、复合维生素、复合矿物质等疗效已获公认的免疫营养素,通过这些物质与蛋白质、植物功能多糖、益生菌等活性成分间的协同增效作用提高产品疗效;
(2)复合酶解工艺制备植物功能多糖。多糖类物质是生物体内除蛋白质、脂肪、核酸以外的一类重要的生命物质材料。生物体的一切生命活性过程,包括免疫应激、细胞生长、代谢、病毒感染等过程中均有参与,在抗肿瘤、增强免疫力、抗病毒等生理功能中具有显著疗效。本发明优选含有激活一氧化氮、IL-2(白介素)、TNF-α(肿瘤坏死因子)等多种细胞因子的灵芝、银耳、冬虫、夏草为基础原料,利用温和、高效、绿色的复合酶解工艺进行植物功能多糖的提取,在最大程度的保留植物体内可激活肿瘤细胞免疫应答,诱导肿瘤细胞凋亡的功能多糖活性物质以外,巧妙引入了其他活性物质,大幅度提高植物功能多糖的功效;
(3)益生菌冻干粉的引入。食管癌围术期内因患者机体免疫力下降,加上胃肠功能未恢复,易导致肠道菌群失调,诱发全身感染、营养不良等并发症,甚至危及生命。本发明添加的益生菌冻干粉是双歧杆菌、酪酸梭菌、鼠李糖乳杆菌、乳酸杆菌中的一种或多种,在使肠内微生物菌群重获平衡的同时,可激活机体巨噬细胞、NK细胞等多种免疫细胞,诱导TNF-β、IFN-γ、IL-12、一氧化氮等效应因子发挥作用,达到提高机体免疫能力、降低术后并发症的目的。本发明针对菌剂在高温下易失活的温敏特性,选用经冷冻干燥技术预处理后的益生菌冻干粉,通过低温休眠方式维持食管癌围术期肠内营养专用食品中益生菌的活性;
(4)复合凝聚微胶囊。传统微胶囊技术因机械性能较差,在后处理过程中易出现破损、营养素损失等问题,且传统微胶囊只能采用单一的O/W或W/O的乳化体系,包埋量及适用范围大大受限。复合凝聚微胶囊采用W/O和O/W共存的复合凝聚体系,利用自组装作用将不同溶解性的活性成分溶解在不同相中,既能稳定存在又能有效防止活性成分间的相互作用,显著提高载样量。本发明选用复合凝聚微胶囊代替传统微胶囊,在提高载样量的同时,因其壁材的特殊性可显著提升pH敏感性和机械性能,最终提高产品的靶向性和缓释性;
(5)粉体高效复配。本发明采用高速气流式粉体机进行各类粉体的复配,通过粒径较小的蛋白质超微粉与复合凝聚微胶囊间通过自组装所形成的分子包覆,制备出具有高混合均匀度的多层核壳结构粉体,使产品质量的均一性及分散性显著改善。

实施例1
适于食管癌围术期患者专用食品的制备方法,包括下述的步骤:
(1)取香菇、银耳、冬虫、夏草(质量比为2∶3∶1∶1.5)清洗干净后,控干水份,破碎打浆(打浆时固液比为1∶6,固体为控干水分的香菇、银耳、冬虫、夏草,液体为纯净水),打浆液在灭菌锅中于70℃灭菌30min;
在灭菌处理过的打浆液中加入由纤维素酶、淀粉酶、果胶酶(纤维素酶、淀粉酶、果胶酶的质量比为2∶1∶3)组成的复合酶,酶解时间为4h,酶解温度为30℃。
酶解完成后将料液倒入三足离心机中,控制转速为4000r/min,离心30min后取上清液;
(2)取复合菌剂冻干粉酪酸梭菌和鼠李糖乳杆菌冻干粉(酪酸梭菌和鼠李糖乳杆菌冻的质量比为1∶1);
取功能性氨基酸(L-谷氨酰胺、组氨酸、天冬氨酸,L-谷氨酰胺、组氨酸、天冬氨酸质量比依次为3∶1∶1);
取水溶性复合维生素(维生素B12、维生素B6、维生素C、维生素D、维生素A,维生素B12、维生素B6、维生素C、维生素D、维生素A的质量比依次为1∶0.5∶2∶1∶1,5);
取复合矿物质(葡萄糖酸钙、葡萄糖酸锌、葡萄糖酸镁、葡萄糖酸亚铁、氯化钠,葡萄糖酸钙、葡萄糖酸锌、葡萄糖酸镁、葡萄糖酸亚铁、氯化钠的质量比依次为0.5∶0.8∶0.3∶1∶2);
将以上所取的复合菌剂冻干粉、功能性氨基酸、水溶性复合维生素、复合矿物质溶于多糖溶液(即复合酶解上清液)中,得混合物Ⅰ;
其中复合菌剂冻干粉、功能性氨基酸、水溶性复合维生素、复合矿物质的质量比为3∶1∶0.5∶0.3;
复合菌剂冻干粉、功能性氨基酸、水溶性复合维生素、复合矿物质之和与多糖溶液(复合酶解上清液)的质量体积比为∶1∶5;
(3)将大豆异黄酮溶于亚麻籽油中(大豆异黄酮与亚麻籽油的质量比为1∶3),得到植物源营养素;
然后将5%的Span80滴入到聚氧乙烯失水山梨醇单油酸酯中,一并滴入到植物源营养素中相混合,获得混合溶液(质量比为1∶2,混合后HLB值=11.5);
(4)将(3)中的混合溶液缓慢滴加至混合物Ⅰ中,混合物Ⅰ与混合溶液的质量比为1∶4,然后通过高速匀浆机处理,控制高速匀浆转速为10000r/min,匀浆20min,得到复合凝聚体系C;
(5)将复合凝聚体系C加入到由β-环糊精和黄原胶(质量比为4∶1)组成的复合壁材溶液中(复合凝聚体系C∶复合壁材溶液=0.3∶1(w/w)),控制转速为12000r/min,匀浆5min后进行高压均质,高压均质压力为20MPa,均质时间为7min,最终得到分散性良好的复合凝聚微胶囊分散液D,分散液D在室温下静置24h,然后离心去除上清,得到微胶囊湿囊E;
(6)对微胶囊湿囊E离心喷雾干燥,控制进风口温度为180℃,出风口温度为70℃,进料流量为35mL/min,得到复合凝聚微胶囊F;
(7)将大豆蛋白低聚肽置入超微气流粉碎机中,控制压力为0.5MPa,转速为2000r/min,粉碎后过200目筛子得到大豆蛋白低聚肽超微粉G;
(8)将复合凝聚微胶囊F与大豆蛋白低聚肽超微粉G(质量比为0.5∶2.5)置入高速气流冲击式粉体机内进行复配,控制高速气流冲击式粉体机冲击力为5Kw,处理时间为10min,处理温度为60℃,得到高混合均匀度的食管癌围术期肠内营养专用食品。
实施例1中的产品应用于体外细胞实验,具体的实验方法及所获得的效果如下:
①实验方法:
(ⅰ)细胞培养
在紫外线消毒处理30min后的超净台中将冻存的EC109细胞株(山东省肿瘤医院中心实验室冷冻)水浴加热溶解进行细胞复苏。复苏后,离心弃上层冻存液,取细胞进行培养皿培养。取对数生长期内的LC109细胞加入胰蛋白酶进行消化处理,后置于含10%的胎牛血清的RPMI-1640培养基中终止消化后震荡成单细胞悬液。接种至96孔板中,于37℃,5%的CO2恒温箱中培养24h,PBS洗涤两次后,悬浮在无血清的成球培养基中备用。
(ⅱ)细胞凋亡检测
将胰蛋白酶消化后的EC109细胞接种于10cm培养皿中,设立对照组(空白对照组)、化学干预组1(二甲双胍)、化学干预组2(尼妥珠单抗)、生物干预组1(壳聚糖)、生物干预组2(肠内专用营养剂),每组设立3组,加入适量培养基中于培养箱中培养24h后,进行食管癌细胞收集。收集的细胞离心、去掉培养基后,PBS冲洗2次。细胞凋亡试剂盒测定,每组设立2组,收集细胞到5mL EP管,每个EP管加入100μL binding buffer,混匀细胞悬液后避光孵育15min后,利用流式细胞仪进行检测。
②实验结果
利用SPSS对流式结果进行数据统计学分析,结果表明不同处理组中均出现细胞凋亡情况(如表1所示)。由表可知,肠内营养剂相较于目前市面上常用的化学抗癌药物二甲双胍、尼妥珠单抗而言细胞凋亡情况有所减低,但比空白组及单一的生物干预剂效果好。

本发明人对实施例1中的产品进行了动物实验,以验证产品的安全性及治疗功效,具体的步骤如下:
①实验方法:
(i)NMBzA诱导裸鼠食管癌动物模型的构建兙
本发明选用目前最常用的诱导小鼠食管癌的化学致癌物非对称性亚硝胺类物质NMBzA作为小鼠食管癌模型构建的诱导物,并优选皮下注射方式进行食管癌动物模型的构建。本发明选用60只4-5周龄的雌性小鼠(体重约为15g)为实验对象,分为三组:A组为空白组,颈背部皮下注射给与15%DMSO溶液;B(造模组)、C(肠内营养干预组)两组以0.3mg/kg的剂量进行颈背部皮下注射NMBzA溶液染毒,按2次/周频率连续注射20周,每周测量记录体重,并根据体重调节注射剂量。营养干预方法如表2所示:

(ⅱ)肠内营养剂对NMBzA诱导食管癌小鼠整体形态、体重和脏器指数影响
空白组小鼠皮毛光亮、体态匀称、饮食量正常、活动正常;造模组小鼠皮毛暗淡、体态弱小、饮食量大幅下降、活动迟缓;肠内营养干预组小鼠皮毛比较光泽、体态偏瘦、饮食量较为正常、运动较正常。20周后处死小鼠,取心、肝、脾、肾进行脏器指数检测,脏器指数=脏器重量/体重(如图1、表3所示)。结果表明,造模组B组小鼠脏器指数远低于正常组A组,说明NMBzA对小鼠脏器具有毒性效应,并最终导致小鼠脏器指数增加。而肠内营养干预小鼠脏器指数相较于造模组具有显著性差异,但仍没有达到正常组水平,表明营养干预对于NMBzA所引起的脏器毒性效应具有较好的治疗功效。

(ⅲ)肠内营养剂对NMBzA诱导食管癌小鼠食管组织及血浆中抗氧化酶活性检测
将匀浆、裂解处理过的冻存的食管组织10000r离心5min,利用抗氧化酶活性检测试剂盒对组织和血浆中抗氧化酶活性进行测定(如表4所示)。实验结果表明,造模组小鼠食管组织及血浆中的GPx活性远低于正常组小鼠和肠内营养干预组小鼠。与正常组小鼠相比,肠内营养干预组小鼠GPx活性略低,但仍具有一定的治疗效果。
实施例2
(1)取香菇、银耳、冬虫、夏草(质量比为3∶5∶3∶3)清洗干净并控干水分后,破碎打浆(打浆时固液比为1∶8,固体为控干水分的香菇、银耳、冬虫、夏草,液体为纯净水),打浆液在灭菌锅中于70℃灭菌30min;
在灭菌处理过的打浆液中加入纤维素酶、淀粉酶、果胶酶(质量比为4∶3∶5),固液比控制在1∶10,酶解时间为8h,酶解温度为50℃;
酶解完成后将料液倒入三足离心机中,控制转速为6000r/min,离心20min后取上清液;
(2)将复合菌剂冻干粉双歧杆菌冻干粉和乳酸杆菌冻干粉(质量比为1∶1),功能性氨基酸(L-谷氨酰胺、精氨酸、天冬氨酸,质量比为3∶2∶3)、复合维生素(维生素B12、维生素B6、维生素C、维生素D、维生素A,质量比为0.8∶0.8∶2.5∶1∶2)、复合矿物质(葡萄糖酸钙、葡萄糖酸锌、葡萄糖酸镁、葡萄糖酸亚铁、氯化钠,质量比为0.3∶0.6∶0.4∶2∶2)溶于复合酶解上清液中,其中复合菌剂冻干粉、功能性氨基酸、水溶性复合维生素、复合矿物质的质量比为5∶3∶2.5∶0.7,高速搅拌后得到混合物Ⅰ;复合菌剂冻干粉、功能性氨基酸、水溶性复合维生素、复合矿物质之和与多糖溶液(复合酶解上清液)的质量体积比为1∶4;
(3)将番茄红素、β-胡萝卜素(质量比为1∶4)溶于维生素E中(浓度为30%),获得植物源营养素,然后滴入乳化剂1%的大豆卵磷脂(HLB值=13),高速搅拌至溶解,得混合溶液;
(4)在高速匀浆机中,将混合溶液缓慢滴加至混合物Ⅰ中,混合物Ⅰ与混合溶液的质量比为4∶8,控制高速匀浆转速为15000r/min,匀浆20min,得到复合凝聚体系C;
(5)将所得到的复合凝聚体系C在高速匀浆机中,加入到壳聚糖、阿拉伯胶和明胶(质量比为3∶1∶2)的复合壁材溶液中(复合凝聚体系C∶复合壁材溶液=0.5∶1(w/w)),控制转速为15000r/min,匀浆5min后进行高压均质。高压均质压力为45MPa,均质时间为5min,最终得到分散性良好的复合凝聚微胶囊分散液D。D溶液在室温下静置24h后,离心去除上清,得到微胶囊湿囊E;
(6)对微胶囊湿囊E进行离心喷雾干燥,控制进风口温度为220℃,出风口温度为90℃,进料流量为20mL/min,得到复合凝聚微胶囊F;
(7)将大豆蛋白低聚肽和酪蛋白(质量比为2∶1)置入超微气流粉碎机中,控制压力为1MPa,转速为1500r/min,粉碎后过600目筛子得到蛋白质超微粉G;
(8)将复合凝聚微胶囊F与蛋白质超微粉G(质量比为3∶5)置入高速气流冲击式粉体机内进行复配,控制高速气流冲击式粉体机冲击力为15Kw,处理时间为5min,处理温度为30℃,得到高混合均匀度的食管癌围术期肠内营养专用食品。
实施例3
(1)取香菇、银耳、冬虫、夏草(质量比为2∶3∶1∶1)清洗干净后,破碎打浆,固液比控制在1∶8,酶解时间为6h,酶解温度为40℃,打浆液在灭菌锅中于70℃灭菌30min。在灭菌处理过的打浆液中加入纤维素酶、淀粉酶、果胶酶(质量比为3∶2∶4)进行复合酶解。酶解完成后将料液倒入三足离心机中,控制转速为5000r/min,离心25min后取上清液;
(2)将复合菌剂冻干粉双歧杆菌冻干粉和鼠李糖乳杆菌冻干粉(质量比为1∶2),功能性氨基酸(L-谷氨酰胺、精氨酸、赖氨酸、天冬氨酸,质量比为2∶1∶1∶2),复合维生素(维生素B12、维生素B6、维生素C、维生素D、维生素A,质量比为0.6∶0.6∶3∶2∶1),复合矿物质(葡萄糖酸钙、葡萄糖酸锌、葡萄糖酸镁、葡萄糖酸亚铁、氯化钠,质量比为0.1∶0.3∶0.2∶1.5∶1)溶于复合酶解上清液中,质量体积比为1∶4,其中复合菌剂冻干粉、功能性氨基酸、水溶性复合维生素、复合矿物质的质量比为4∶2∶1.5.5∶0.5,高速搅拌后得到混合物I;
(3)将β-胡萝卜素和大豆异黄酮(质量比为1∶1)溶于维生素E中(β-胡萝卜素和大豆异黄酮质量之和维生素E体积之比为30%),滴入3%的单甘脂和聚氧乙烯蓖麻油混合溶液中(质量比为1∶1,HLB值=12),高速搅拌至溶解后获得混合溶液;
(4)在高速匀浆机中,将混合溶液缓慢滴加至混合物I中,混合溶液与混合物I之间的质量比为2∶6,控制高速匀浆转速为13000r/min,匀浆15min,得到复合凝聚体系C;
(5)将所得到的复合凝聚体系C在高速匀浆机中,加入到壳聚糖、辛烯基琥珀酸淀粉酯和羟乙基纤维素(质量比为2.5∶0.5∶2)的复合壁材溶液中(复合凝聚体系C∶复合壁材溶液=0.6∶1(w/w)),控制转速为15000r/min,匀浆5min后进行高压均质。高压均质压力为30MPa,均质时间为7min,最终得到分散性良好的复合凝聚微胶囊分散液D。D溶液在室温下静置24h后,离心去除上清,得到微胶囊湿囊E;
(6)对微胶囊湿囊E进行离心喷雾干燥,控制进风口温度为200℃,出风口温度为80℃,进料流量为25mL/min,得到复合凝聚微胶囊F;
(7)将大豆蛋白低聚肽和乳清蛋白(质量比为2∶1.5)置入超微气流粉碎机中,控制压力为0.7MPa,转速为1500r/min,粉碎后过400目筛子得到蛋白质超微粉G;
(8)将复合凝聚微胶囊F与蛋白质超微粉G(质量比为2∶4)置入高速气流冲击式粉体机内进行复配,控制高速气流冲击式粉体机冲击力为10Kw,处理时间为7min,处理温度为40℃,得到高混合均匀度的食管癌围术期肠内营养专用食品。
表5实施例1-3与市售产品的指标对比表
从以上表格中的数据可以看出,本发明的产品与市售产品相比,其包埋率高出约10%左右,粒径远远小于市售产品,缓释率(即48小时之后,微胶囊中剩余有效成分/微胶囊中总有效成分)还能达到58%,而市售产品仅仅只有26%,且pH=3时的破损率较低,仅仅24-27%,相比于市售产品的73%的破损率,本发明的产品从以上4个方面性能占绝对的优势。
Claims (7)
1.适于食管癌围术期患者肠道的专用食品的制备方法,包括下述的步骤:
(1)复合酶酶解制备水溶性植物功能多糖
清洗香菇、银耳、冬虫、夏草;控干水分,并打浆,得浆液;香菇、银耳、冬虫、夏草的质量比例为2-3:3-5:1-3:1.5-3;打浆时固液比为1:5-1:10;
将所得浆液灭菌处理,在灭菌后的浆液中加入复合酶酶解4-8h,离心,取上清液,获得植物功能多糖溶液;
复合酶解由纤维素酶、淀粉酶、果胶酶组成,纤维素酶、淀粉酶与果胶酶的质量比分别为2-4:1-3:3-5,酶解时间为4-8h,酶解温度为30-50℃;
离心转速控制在4000-6000 r/min,离心20—30min;
(2)制备微胶囊芯材
将复合菌剂冻干粉、功能性氨基酸、水溶性复合维生素、复合矿物质溶于(1)中所得的植物功能多糖溶液中,得混合物Ⅰ,在高速匀浆过程中滴入乳化剂和植物源营养素组成的混合溶液,得到复合凝聚体系;
在滴入乳化剂时将HLB值控制在10-13.5之间;
(3)复合凝聚微胶囊的制备
将(2)中所得到的复合凝聚体系在高速搅拌的条件下,加入到复合壁材溶液中,得到复合凝聚微胶囊预备液,高压均质进行二次混合处理,获得微胶囊分散液;
将该微胶囊分散液静置20-24h后,弃去上清液,得到微胶囊湿囊,离心喷雾干燥,得复合凝聚微胶囊;
(4)围术期必需蛋白质超微粉的制备
对食管癌围术期肠内营养必需的蛋白质进行超微气流粉碎预处理,过200-600目筛,得到蛋白质超微粉;
(5)复配蛋白质超微粉与复合凝聚微胶囊
将(3)中所得复合凝聚微胶囊、(4)中所得蛋白质超微粉进行粉体混合处理,得到食管癌围术期患者专用食品;采用高速气流冲击式粉体机将复合凝聚微胶囊和蛋白质超微粉进行粉体混合。
2.如权利要求1所述适于食管癌围术期患者肠道的专用食品的制备方法,其特征在于:(2)中,复合菌剂冻干粉为双歧杆菌、酪酸梭菌、鼠李糖乳杆菌、乳酸杆菌中的至少两种;功能性氨基酸为L-谷氨酰胺 、组氨酸、精氨酸、赖氨酸、天冬氨酸、谷氨酸中的至少一种;水溶性复合维生素为维生素B12、维生素B6、维生素C、维生素D、维生素A中的至少两种;
复合矿物质为葡萄糖酸钙、葡萄糖酸锌、葡萄糖酸镁、葡萄糖酸亚铁、氯化钠中的至少两种;
乳化剂为Span80、大豆卵磷脂、单甘脂、聚氧乙烯蓖麻油、聚氧乙烯失水山梨醇单油酸酯中的至少一种;植物源营养素为亚麻籽油、维生素E、番茄红素、β-胡萝卜素、大豆异黄酮中的至少一种。
3.如权利要求1所述适于食管癌围术期患者肠道的专用食品的制备方法,其特征在于:复合菌剂冻干粉、功能性氨基酸、水溶性复合维生素、复合矿物质的质量比为3-5:1-3:0.5-2.5:0.3-0.7;上述复合菌剂冻干粉、功能性氨基酸、水溶性复合维生素、复合矿物质与复合酶解上清液的质量体积比为:3-6:20;乳化剂和植物源营养素所组成的混合溶液的体积浓度为20-35%;混合物Ⅰ与植物源营养素和乳化剂所组成的混合溶液的质量比为1-3:4-8;乳化剂HLB值控制在10-13.5之间,其占复合凝聚体系质量的0.01-0.05%;高速匀浆步骤中转速为10000-15000 r/min,匀浆时间为10-20min。
4.如权利要求1所述适于食管癌围术期患者肠道的专用食品的制备方法,其特征在于:复合凝聚微胶囊体系中复合壁材为β-环糊精,辛烯基琥珀酸淀粉酯,壳聚糖,阿拉伯胶,明胶,羟乙基纤维素中的至少一种;芯材复合凝聚体系占壁材总质量的30-60%;高压均质压力为20—45 MPa,均质时间为3—7 min;离心喷雾干燥进风口温度为180—220℃,出风口温度为70—90℃,进料流量为20-35mL/min。
5.权利要求1所述适于食管癌围术期患者肠道的专用食品的制备方法,其特征在于:围术期必需蛋白质包括大豆蛋白低聚肽、乳清蛋白、酪蛋白中的至少一种;超微气流粉碎压力为0.5—1 MPa,转速为1000—2000r/min,粉碎后过200-600目筛分离、细化。
6.权利要求1所述适于食管癌围术期患者肠道的专用食品的制备方法,其特征在于:复合凝聚微胶囊与蛋白质超微粉的质量比为0.5-3:2.5-5;高速气流冲击式粉体机冲击力为5-15Kw/hr,处理时间为5-10min,处理温度为30-60℃。
7.权利要求1所述适于食管癌围术期患者肠道的专用食品的制备方法,其特征在于,喷雾干燥对湿囊进行处理后所得复合凝聚微胶囊粒径尺寸<100 μm。
