技术领域
本发明涉及一种饮料,具体涉及一种以茶皂素为主体的具有解酒功效的饮料。
背景技术
酒是以粮食为原料经发酵酿造而成的,其化学成分是乙醇,这是造成酒醉的主要因素。醉酒即指大量饮酒,体内酒精代谢速度慢于乙醇吸收速度,大量酒精通过血液循环进入脑部,致使大脑亢奋,当达到一定浓度后,大脑皮层开始产生睡眠意识,最后产生麻木性昏迷,造成的急性酒精中毒现象。适量饮酒的确会对人体健康有益,例如促进血液循环。但是饮酒过度则会对人体造成很大伤害,不仅容易对肝脏等重要脏器造成损伤,而且对中枢神经系统也会产生严重损害,造成眩晕、恶心呕吐,甚至是昏迷不醒等醉酒症状。醉酒对人体的危害极大,但也是人们日常生活和交往中不可避免的。开发有效的解酒饮料越来越受到人们的普遍重视。
乙醇在体内的代谢途径:乙醇在人体内不需要经过消化作用,直接经口腔、胃、肠等吸收进入血液,然后随血液流到各个器官,其中有90%在肝脏内进行代谢,主要是在乙醇脱氢酶的作用下先把乙醇转化为乙醛,再在乙醛脱氢酶的作用下把乙醛转化为二氧化碳和水。
解酒的关键在于如何使人体重要器官以及中枢神经、血液尽量少接触由酒精(乙醇)进入人体后产生的乙醛,即可通过增强乙醇脱氢酶的活性,加快乙醇在肝脏内的代谢,进而降低乙醇对肝脏、脑等器官的危害性,或提高肝组织中超氧化物歧化酶和谷胱甘肽过氧化物酶的活性,消除肝组织和血浆中的自由基和脂质过氧化物对机体的损伤等。
目前报道的解酒药物或者保健品很多,其中以葛根与其他成分配伍的解酒产品最多,它们确有一定的解酒作用,但仍然存在各种问题。比如公布号为CN101874857A的中国专利申请,公开了一种含有葛根、枸杞、山楂和木瓜的护肝白酒,这种护肝白酒存在原料较多较成分复杂且效果欠佳的问题。又比如公布号为CN201510100822.1的中国专利申请,公开了一种以甘蔗汁作为发酵原料,接入牛樟芝发酵再经均质等得到的解酒饮料,其步骤复杂、成本高,且解酒效果不明显。
茶皂素是油茶中全部皂素的总称,油茶饼粕中含有约10-18%的皂素,因此,它有很大的利用空间。有研究表明,茶皂素具有保护肝脏的功能,目前关于茶皂素的相关解酒产品较为罕见。而葛根是中国传统医学中最具代表性的解酒药物之一,其富含黄酮类物质,可有效从乙醇的吸收、代谢、排泄三个环节发挥解酒作用。然而目前市场上常见的解酒饮料,成分复杂,解酒效果不尽如人意,且口感大多较为苦涩,不适于为广大人群所喜爱。本发明以茶皂素为主,复合葛根、陈皮等成分,并通过动物实验找到它们的最佳配合比,开发出一种高利用、成本低廉、口感好、可以保护肝胃的效果较佳的解酒饮料。

本发明提供的茶皂素解酒饮料中主要功效成分为:
茶皂素:是齐墩果烷型五环三萜类皂苷的混合物,其基本结构含有皂苷元(即配基)、糖体和有机酸三部分组成。三萜皂苷类化合物具有较好的抗氧化作用,研究表明,茶皂素在酒精吸收代谢中可降低血液中的乙醇含量,且同时可降低肝脏中的乙醛含量,既是酒精吸收的抑制剂,又是乙醇消化的促进剂,具有保护肝脏的功能。
葛根:葛中的有效成分主要包括黄酮类化合物和皂苷两大类,研究表明葛中含有的异黄酮类化合物和皂苷成分均具有一定的护肝作用。葛中的异黄酮和三萜皂苷类化合物具有较好的抗氧化作用。葛根具有解肌退热、生津止渴,解酒毒的功效。能够改善心脑血管疾病,提高学习记忆能力,降血糖、血脂及血压,保护肝脏,抗肿瘤和诱导癌细胞凋亡等。
陈皮:陈皮主要含有黄酮类、挥发油、生物碱和微量元素等物质,它对肝脏疾病具有积极的预防、保护作用,可缓解肝损伤、脂肪肝等疾病。其中黄酮类化合物主要有橙皮苷、川陈皮素、橘皮素等。研究表明陈皮对胃排空有一定抑制作用,橙皮甙具有一定的促胃肠动力作用。有报道称橙皮苷对刀豆蛋白A致小鼠免疫性肝损伤具有一定的保护作用,还可抑制顺铂引起的小鼠血清AST、ALT水平升高、减少MDA形成及恢复SOD活性等。
红枣:红枣富含糖类、纤维素、维生素、氨基酸、蛋白质、矿物质等营养成分,特别是含有多糖、芦丁、环磷酸腺苷等活性成分。研究表明红枣多糖具有清除自由基的作用,其粗品能提高小鼠血液、肝脏及脑组织中超氧化物歧化酶(SOD)活性和小鼠血液、肝脏中过氧化氢酶(CAT)活性;降低小鼠血液、肝脏及脑组织丙二醛(MDA)含量,表明红枣多糖粗品体内具有抗氧化作用。另外芦丁具有显著的体外抗氧化活性,能有效保护肝线粒体。
枸杞:枸杞果实中的功能成分主要是枸杞多糖、黄酮、类胡萝卜素、甜菜碱、维生素、牛磺酸及萜类化合物,这些功能成分与枸杞果实的抗氧化活性密切相关,也是枸杞果实调节机体免疫力、抑制肿瘤生长、抗疲劳、清除自由基、保肝、延缓衰老的主要成分,研究表明枸杞多糖-1具有清除·OH和抑制·OH所致小鼠肝线粒体氧化损伤的作用。
山楂:山楂含黄酮类、三萜皂甙类(熊果酸、齐墩果酸、山楂酸等),脂肪酸、维生素C、无机盐、红色素等,具有消食、降压,降脂,抗氧化等功能,可增强免疫力、清除胃肠道有害细菌等,还可预防肝癌。山楂黄酮对酒精性肝损伤具有一定的保护作用,生山楂可降低高脂饲料所致SD大鼠肝组织丙二醛(MDA)、TC、TG的量,清除肝内堆积的三酰甘油,减少脂肪酸(FFA)对肝细胞毒性作用,使谷丙转氨酶(ALT)、谷草转氨酶(AST)指标降低,达到降血脂、保肝作用。山楂的新鲜果汁可以更好保留有效成分,同时使饮料口感清新。
蜂蜜:主要的成分为糖、无机盐、蛋白质、氨基酸、有机酸、微量元素等,具有解毒、补中、止痛、润燥的作用,同时蜂蜜在食品加工中也是很好的甜味剂、抗氧化剂。具有保护肝脏、心血管系统的作用,对肠胃也有保护作用,同时还具有提高免疫力、改善睡眠等作用。
维生素:临床研究发现,对急性酒精中毒患者补充维生素VB1、VB6、VC可加速体内酒精代谢,因此产品配方中加人维生素类,不仅可缓解酒精中毒的不良症状,补充饮用酒精造成体内维生素缺乏,而且添加的VC还可作抗氧化剂抑制褐变。
有益效果
本发明提供了一种可以保护肝胃的效果较佳的解酒饮料,本饮料以以茶皂素为主,配合其他物质并找到其最佳复合比,达到原料利用率高,成本低廉,口感好,且保质期长,不含任何化学防腐剂的解酒饮料。实验检测本发明的解酒饮料能够延长了小鼠的醉酒耐受时间和减少了睡眠时间,且能显著降低血液中乙醇的浓度,增加酒后体内谷胱甘肽过氧化物酶、超氧化物歧化酶活性,加速肝脏中乙醇的代谢和自由基的清除,减少过氧化物的产生,抑制酒精引起的丙二醛含量增加,还在一定程度上增加乙醇脱氢酶活性,加快乙醇在肝脏内的代谢,减少乙醇对身体的伤害,且具有良好的口感。

制备方法:
(1)原料处理
茶皂素:参照公开号为CN201610251905.5的中国专利申请,从油茶饼粕中制备具有解酒功能茶皂素方法。首先将油茶饼粕粉碎、脱脂、干燥,然后采用去离子水提取茶皂素,去离子水提得到的茶皂素粗提液,主要含有油茶多糖、蛋白质、单宁等杂质,采用两级沉淀法(即乙醇沉淀纯化法和CaO沉淀纯化法相结合的方法)纯化茶皂素,从而得到纯度较高的具有解酒功能的茶皂素。本方法生产成本低,无环境污染,得到的茶皂素能够显著增强乙醇脱氢酶活性,安全无毒,且生产工艺稳定,利于大型工业化生产。
葛根醇提物:取葛根原料,加入8倍70%乙醇浸泡30min,在70℃下回流提取2h;纱布滤过后,留滤液,滤渣中再加入8倍乙醇,在同样温度下浸提1.5h,2次;合并3次滤液,冷却,加入适量乙醇,静置过夜。滤过去除沉淀,滤液经减压浓缩至少量液体,再经冷冻干燥,研磨得到葛根粉末,于4℃下保存,使用时加蒸馏水配制成所需生药剂量浓度溶液。
陈皮黄酮提取液:将陈皮粉碎成粗粉,置干燥箱于60℃烘12小时以上,于闭光、阴凉处存放。称取约5g陈皮粗粉置于250ml量瓶中,加入石油醚(30-60℃)80mL,60℃回流脱脂2h;挥干石油醚后,125mL60%乙醇60℃进行提取,抽滤,洗涤滤渣,合并滤液和洗液,浓缩去适量的乙醇,然后转入100mL量瓶中,定容,摇匀。
红枣多糖:称取红枣50g,加15倍水,于80℃,30min,降温至50℃,加入0.01%的纤维素酶(pH4.5),保温2h,升温到80℃,30min灭酶,过滤,滤渣采取同样的方法,再提取1次,过滤,合并滤液,浓缩,加无水乙醇沉淀,沉淀复溶于水,再用无水乙醇沉淀,沉淀用无水乙醇、丙醇反复洗涤。冷冻干燥,得多糖粗品。
枸杞提取物:取干燥的枸杞500g,3000mL水加热回流提取,达到微沸状态后保持2h,再加入2000mL水重复提取1次,混合两次提取液。滤过,用旋转蒸发仪浓缩,温度控制在60℃左右,得初膏100g。在烘箱中烘干,温度控制在80℃左右,研磨得到枸杞提取物25g。
山楂提取物:将山楂磨碎后加入4倍于山楂重量的水侵泡1h,之后将混合物加入到萃取罐中,再加入重量相当于山楂4倍的水,搅拌,加热至80-85℃,萃取3次,每次3h,萃取完毕后用三层纱布过滤,弃去滤渣,将三次得到的滤液合并,之后真空浓缩至滤液为山楂重量的2倍,得山楂提取物。
蜂蜜、柠檬酸、维生素:采用市售的蜂蜜、柠檬酸、维生素。
(2)调配
将上述所得茶皂素、葛根提取物、陈皮提取物、红枣多糖、枸杞提取物、山楂提取物、蜂蜜、柠檬酸、维生素VB1、VB6、VC、饮用水按不同的配比混匀调配成饮料。
(3)灭菌:将调配好的饮料采用巴氏低温灭菌,于100℃的水浴中保持15min。
(4)当灭菌后的饮料温度降至85℃时,灌装到玻璃瓶中,这样趁热灌装可以起到防止瓶子及环境中的杂菌再次污染产品的作用。如果是在较低的温度环境下操作时,应先将玻璃瓶预热,以防爆破。
(5)成品:产品冷却到室温后,包装入库,保存。
醒酒药效动物实验:
1、小鼠灌酒量的选择:取70只小鼠,分为7组,每组10只。禁食不禁水12h后,各组按体重灌56°红星二锅头白酒10,11,12,13,14,16,18ml/kg观察醉酒表现,将使小鼠的醉酒率最高死亡率最低的灌酒剂量作为后续实验的最适醉酒量。
2、醉酒实验:取64只小鼠,分为8组,对照组(灌服10ml/kg生理盐水),茶皂素组(灌服茶皂素5mg/10g.bw),葛根组(灌服葛根提取物4mg/10g.bw),对比组(公开号为CN201410425494.8的中国专利申请的实施例部分的实施例1所制得的解酒剂),复合液1组(实例1组),复合液2组(实例2组),复合液3组(实例3组),海王金樽阳性对照组(灌服海王金樽生药片10mg/10g.bw),禁食12h后,每只小白鼠按体重灌胃各组药物,30min后按最适灌酒量灌胃白酒,开始计时,通过“翻正反射实验”判断、记录醉酒、醒酒时间。
3、取昆明种雄鼠75只,随机分为5组,模型对照组(灌服10ml/kg生理盐水),茶皂素组(灌服茶皂素5mg/10g.bw),海王金樽阳性对照组(灌服海王金樽生药片10mg/10g.bw),复合液2组(灌服茶皂素葛根复合液,复合比为4:4),对比组,同前灌胃各组药物,30min后各组分别灌胃给予白酒,灌服白酒后30、60、90、120、150min共5个时间点对小鼠进行眼眶取血。取各组小白鼠摘取眼球取血0.5mL,放入肝素抗凝,于EP管中备用。通过乙醇脱氢酶法测定血液中乙醇的浓度。
4、动物取血后脱颈椎处死,取肝脏于-80℃保存,制备10%肝组织匀浆,以3 000r/min离心,取上清液,按照试剂盒使用说明书测定GSH、MDA、ADH和SOD活性。
实验结果:

以上结果表明使小鼠的醉酒率最高死亡率最低的灌酒剂量是13mL/kg.bw,所以后续实验的最适醉酒量为13mL/kg.bw小鼠。

以上各处理对小鼠醉酒耐受时间、醒酒时间的影响结果表明复合液2组(实例2组)对延长醉酒耐受时间,缩短睡眠时间方面效果最显著,所以选取复合液2组作为后续实验的复合液最佳配比组。

以上结果表明各处理组均在一定程度上延长了小鼠的醉酒耐受时间和减少了睡眠时间,且降低了血液中乙醇的浓度,茶皂素、海王金樽、茶皂素复合物能增加酒后体内谷胱甘肽过氧化物酶、超氧化物歧化酶活性,抑制酒精引起的丙二醛含量增加,表明茶皂素在一定程度上能加速肝脏中乙醇的代谢和自由基的清除,减少过氧化物的产生;增加乙醇脱氢酶活性,加快乙醇在肝脏内的代谢,减少乙醇对身体的伤害。其中实施案例2组中的茶皂素复合物的解酒效果最显著。
Claims (3)
1.一种以茶皂素为主体的解酒饮料,其特征在于:所述茶皂素解酒饮料包括以下组分:茶皂素、葛根提取物、陈皮提取物、红枣多糖、枸杞提取物、山楂果提取物、蜂蜜、柠檬酸、维生素B1、维生素B6和维生素C。
2.如权利要求1所述的以茶皂素为主体的解酒饮料,其特征在于:按1000重量份数计,包括以下组分:
茶皂素 40-60; 葛根提取物 15-30;
陈皮提取物 10-20; 红枣多糖 5-20;
枸杞提取物 5-15; 山楂果提取物 3-9;
蜂蜜 10-25; 柠檬酸 2-5;
维生素C 0.2-0.5; 维生素B1 0.06-0.12;
维生素B6 0.05-0.1; 水 余量。
3.如权利要求1所述的以茶皂素为主体的解酒饮料,其特征在于:按1000重量份数计,包括以下组分:
茶皂素60份; 葛根提取物15份;
陈皮提取物 15份; 红枣多糖 10份;
枸杞提取物 10份; 山楂果提取物9份;
蜂蜜15份; 柠檬酸3份;
维生素C 0.3份; 维生素B1 0.08份;
维生素B6 0.06份; 水 余量。
