Abstract
本发明公开了茶皂素解酒护肝饮料及其制备方法,用来缓解饮酒过量引起的不适感以及长期饮酒对胃、肝脏等造成的损伤。主要由茶皂素、枳椇子、甘草、蜂蜜、白砂糖和碳酸氢钠为原料按一定重量配比而成,同时调pH为7.5‑8.0以形成弱碱性环境。本发明原料来源方便安全,制备工艺简单,成本低廉;能用于大规模生产,具有广阔的市场前景。弱碱性环境不仅能增强茶皂素及各组分的解酒活性,还能促进人体对营养物质的吸收,同时,本产品对机体清除自由基,胃和肝脏具有良好的预防保护作用。
Description
技术领域
本发明涉及解酒饮料技术领域,尤其涉及茶皂素解酒护肝饮料及其制备方法。
背景技术
降低饮酒过度对人体带来的危害,关键在于提高乙醇脱氢酶和乙醛脱氢酶的酶活力,加速其对乙醇和乙醛的分解,从而降低乙醇和乙醛对脑、肝脏的危害,减轻酒精对肠胃的刺激,降低乙醛对细胞的氧化能力,促进新陈代谢。但是,现有的解酒类产品存在成分复杂效果欠佳等问题。
发明内容
基于背景技术存在的技术问题,本发明提出了茶皂素解酒护肝饮料及其制备方法,能够提高乙醇脱氢酶和乙醛脱氢酶的活性,从而达到很好的解酒效果。
本发明提出的一种茶皂素解酒护肝饮料的制备方法步骤如下:
S1:将枳椇子和甘草依次洗净、晾干,烘干后一起粉碎,晾干的条件为25-35℃下晾干4-8h;
S2:将S1中粉碎后的原料加水浸泡,然后进行煮制,结束后抽滤,得药液A;
S3:将S2抽滤后的滤渣加水煮制,结束后抽滤,得药液B;
S4:重复S3步骤1-3次,并将每次得到的药液B与药液A合并,合并后的药液搅拌均匀后用硅藻土过滤备用;
S5:将所述S4过滤后的药液与茶皂素、蜂蜜、白砂糖搅拌均匀,并用pH调节剂调节pH为弱碱性,然后再过滤、杀菌、真空包装得成品。
优选地,所述S1中烘干的条件为温度75-85℃、时间1.5-2.5h。
优选地,所述S2中原料与水的质量比为1:10-20,浸泡的时间为2-3h。
优选地,所述S2中的煮制条件为温度80-100℃、时间0.5-1h。
优选地,所述S3中滤渣与水的质量比为1:4-6,煮制的条件为温度80-100℃、时间20-40min。
优选地,包含如下重量份计的原料:茶皂素0.8-1.2份、枳椇子5-10份、甘草2-6份、蜂蜜6-10份、白砂糖1-3份。
优选地,饮料的pH为7.5-8.0。
优选地,所述pH调节剂为碳酸氢钠。
本发明提出的上述方法制备的茶皂素解酒护肝饮料。
本发明的S4中过滤后的药液需要进行浓缩处理,然后再加入茶皂素、蜂蜜、白砂糖和pH调节剂,保证每100重量份饮料中含有1重量份左右的茶皂素。
本发明的原料中,茶皂素又名皂甙,皂角甙,是山茶科植物中含有的一类天然糖苷,是齐墩果烷型五环三萜皂苷的混合物,基本结构由皂苷元、糖体、有机酸三部分组成。
枳椇子:枳椇子为鼠李科植物枳椇的带有肉质果柄的果实或种子,枳椇子中的黄酮类、三萜皂苷类和生物碱类物质对保护肝脏、抗氧化损伤、降压抗肿瘤具有良好的作用效果。在医书记载中也提起到枳椇子性平味甘、有止渴除烦、利尿解酒等功效。
甘草:甘草又名甜草、乌拉尔甘草是一种补益的中草药。有解毒、祛痰、止痛、解痉以至抗癌等药理作用。甘草中有效成份甘草甜素,对毒物有吸附作用能增强肝脏的解毒能力。甘草素、甘草苷等活性成分能抑制胃液、胃酸分泌,增加胃黏膜细胞的己精胺成份,保护胃黏膜,使之不受损害,促进消化道上皮细胞再生。
蜂蜜:又名蜂糖、石蜜,具有保护心脑血管、保护肝脏,促使肝细胞再生、增强免疫力、杀菌解毒、润肠通便的功效。其中蜂蜜中的果糖、葡萄糖等可促进酒精的分解吸收,加速酒精从血液中清除的速度。
作用机理:本申请的制备的饮料以茶皂素为主体,茶皂素一方面能够抑制血液对酒精的吸收,另一方面进入肝脏后由于其呈弱碱性,能够加快肝脏中乙醇脱氢酶和乙醛脱氢酶的激活,人体中的乙醇脱氢酶以多种二聚体的形式存在,由至少7种不同的基因编码。乙醇脱氢酶一共具有五个类别(Class I-V),但在人的胃肝脏中存在的主要是Class I。它催化乙醇乙醛氧化作用为:CH3CH2OH+NAD+→CH3CHO+NADH+H+,乙醛脱氢酶是随机组合的四聚体,一个突变型的亚基影响了四聚体的稳定性,进而影响酶的正常表达。研究发现无论携带ALDH2*2的是纯合子(AA)还是杂合子(GA),四聚的ALDH2均无活性,即ALDH2*2是显性遗传。杂合子GA的ALDH2四个亚基都稳定的概率是(0.5)^4=6%,因而即使杂合子的野生型与突变等位基因等量表达,其正常的ALDH2的表达量也仅有6%。有ALDH2*2突变表达出的亚基的酶无法正常代谢乙醇的氧化产物乙醛,血液乙醛浓度增高,造成一系列饮酒后的不良反应。
乙醇脱氢酶主要负责将乙醇氧化为乙醛,如果单纯提高乙醇脱氢酶的活性的话则会出现肝脏内乙醛的大量积聚,反而会对人体健康产生更大的影响,而本申请制备的饮料不仅能够提高乙醇脱氢酶的激活率,还能够提高乙醛脱氢酶的激活率,从而提高了乙醛氧化为对人体无害的乙酸的速率,使进入肝脏内的乙醇能够快速分解,本申请制备的饮料中加入的枳椇子和甘草一方面起到护肝的作用,另一方面其还可与茶皂素发生协同作用,提高茶皂素对乙醇脱氢酶和乙醛脱氢酶的激活率,从而进一步提高饮料的解酒性能。
与现有技术相比,本发明的有益技术效果:本发明通过pH调节剂使饮料为弱碱性,提高了饮料对乙醇脱氢酶和乙醛脱氢酶的激活率,乙醇脱氢酶的激活率从弱酸环境下的50%提高到65%左右,乙醛脱氢酶的激活率从弱酸环境下的20%提高到35%左右,而乙醇脱氢酶和乙醛脱氢酶活性的提高能够加快对乙醇及乙醇分解的产物乙醛的分解,从而达到解酒护肝的效果。
具体实施方式
下面结合具体实施例对本发明作进一步解说。
实施例1
本发明的茶皂素解酒饮料的制备方法包括以下步骤:
(1)将枳椇子和甘草洗净、晾干、80℃烘干2h,粉碎,加入10倍的水浸泡3小时,在80℃温度下,煮制0.5h,抽滤,药渣加5倍的水,煮制0.5h,抽滤,合并滤液搅拌均匀后硅藻土过滤备用。
(2)将(1)中的备用液加入原料重量份的茶皂素、蜂蜜和白砂糖合并后搅拌均匀,并用碳酸氢钠调节PH为8.0,过滤膜、巴氏杀菌、真空包装即为成品。
其中:茶皂素0.8份、枳椇子8份、甘草6份、蜂蜜10份、白砂糖1份。
实施例2
本发明的茶皂素解酒饮料的制备方法包括以下步骤:
(1)将枳椇子和甘草洗净、晾干、80℃烘干2h,粉碎,加入15倍的水浸泡2h,在90℃温度下,煮制1h,抽滤,药渣加5倍的水,煮制0.5h,抽滤,合并后搅拌均匀后硅藻土过滤。
(2)将(1)中的备用液加入原料重量份的茶皂素、蜂蜜和白砂糖合并后搅拌均匀,并用碳酸氢钠调节PH为7.5,过滤膜、巴氏杀菌、真空包装即为成品。
其中各原料按重量份计:茶皂素1.2份、枳椇子6份、甘草2份、蜂蜜8份、白砂糖3份。
实施例3
本发明的茶皂素解酒饮料的制备方法包括以下步骤:
(1)将枳椇子和甘草洗净、晾干、80℃烘干2h、粉碎,加入20倍的水浸泡2小时,在100℃温度下,煮制0.5h,抽滤,药渣加5倍的水,煮制0.5h,抽滤,合并后搅拌均匀后硅藻土过滤。
(2)将(1)中的备用液加入原料重量份的茶皂素、蜂蜜和白砂糖合并后搅拌均匀,并用碳酸氢钠调节PH为7.5,过滤膜、巴氏杀菌、真空包装即为成品。
其中各原料按重量份计:茶皂素1份、枳椇子10份、甘草5份、蜂蜜8份、白砂糖2份。
对比例1
该方案与实施例1的区别在于未加入pH调节剂,制备的饮料的pH在6.0左右。
对比例2
该方案与实施例1的区别在于通过碳酸氢钠调节pH为7.0。
分别对实施例1-3和对比例1-2中制备的饮料的解酒性能进行测定,主要测定饮料对乙醇脱氢酶和乙醛脱氢酶的活性促进效果,其中:
乙醇脱氢酶的活力测定采用瓦勒-霍赫法。在试管中分别加入焦磷酸钠缓冲液1.5mL(pH=8.8)、0.5mL 11.5%(V:V)的乙醇溶液1.0mL 27mmol/L的氧化型辅酶I(NAD+)和一定浓度的茶皂素口服液0.1mL,混合后放入25℃水浴中保温10min。结束后,试管立即加入0.1mL 0.25U/mL的ADH,摇匀后立即在分光光度计340nm波长处测定吸光度值(A340nm),然后每隔10s记数一次,连续测定5min,实验结果作为测定组。以0.5mL蒸馏水作空白调零,以0.1mL蒸馏水代替0.1mL茶皂素饮料作为空白组,测定空白组乙醇脱氢酶的酶活力。
乙醇脱氢酶活性计算法:
以A对时间作图,取反应最初级性部分,计算A340/10s的增加值,根据NADH在340nm处的摩尔消光系数为6.2,计算酶活力。ADH的活性以每分钟NADH生成的纳摩尔数表示。

如表1可知,实施例与对比例相比,其乙醇脱氢酶的激活率有显著地提高,从而说明弱碱条件下有利于解酒饮料对乙醇脱氢酶的激活,从而其解酒效果更好。
乙醛脱氢酶活性测定方法:
乙醛脱氢酶活性测定采用Blair&Bodley法。取干燥试管;加入100mmol/L PH为9.5的焦磷酸钠缓冲液1.6mL;加入3.6mmol/L氧化型辅酶I(NAD+)1.0mL;加入100mmol/L乙醛0.1mL;一定浓度的茶皂素饮料0.1mL(空白组加入同体积蒸馏水)0.1mL;混匀,在30℃条件下温育5min;取出立即加入18mmol/L ALDH 0.2mL,摇匀;于340nm波长处测定吸光度,每1min读数1次,直至每分钟吸光度的值达到稳定为止。

同样的,从表2可以看出实施例相对于对比例来说,乙醛脱氢酶的激活率的得到显著地提高,从而说明弱碱条件下有利于解酒饮料对乙醛脱氢酶的激活,从而其解酒效果更好。
综上所述,本申请制备的解酒饮料具有很好的激活乙醇脱氢酶和乙醛脱氢酶的效果,具有很好的解酒和护肝的功效。
此外,还对实施例1-3制得的饮料的理化性质进行了测定,其细菌总数(cfu/ml)<70,大肠杆菌(MPN/ml)<1,没有检出致病菌,符合国家标准。
以上所述,仅为本发明较佳的具体实施方式,但本发明的保护范围并不局限于此,任何熟悉本技术领域的技术人员在本发明揭露的技术范围内,根据本发明的技术方案及其发明构思加以等同替换或改变,都应涵盖在本发明的保护范围之内。
Claims (9)
1.一种茶皂素解酒护肝饮料的制备方法,其特征在于,方法步骤如下:
S1:将枳椇子和甘草依次洗净、晾干,烘干后一起粉碎;
S2:将S1中粉碎后的原料加水浸泡,然后进行煮制,结束后抽滤,得药液A;
S3:将S2抽滤后的滤渣加水煮制,结束后抽滤,得药液B;
S4:重复S3步骤1-3次,并将每次得到的药液B与药液A合并,合并后的药液搅拌均匀后过滤备用;
S5:将所述S4过滤后的药液与茶皂素、蜂蜜、白砂糖搅拌均匀,并用pH调节剂调节pH为弱碱性,然后再过滤、杀菌、真空包装得成品。
2.根据权利要求1所述的茶皂素解酒护肝饮料的制备方法,其特征在于,所述S1中烘干的条件为温度75-85℃、时间1.5-2.5h。
3.根据权利要求1所述的茶皂素解酒护肝饮料的制备方法,其特征在于,所述S2中原料与水的质量比为1:10-20,浸泡的时间为2-3h。
4.根据权利要求1所述的茶皂素解酒护肝饮料的制备方法,其特征在于,所述S2中的煮制条件为温度80-100℃、时间0.5-1h。
5.根据权利要求1所述的茶皂素解酒护肝饮料的制备方法,其特征在于,所述S3中滤渣与水的质量比为1:4-6,煮制的条件为温度80-100℃、时间20-40min。
6.根据权利要求1所述的茶皂素解酒护肝饮料的制备方法,其特征在于,包含如下重量份计的原料:
茶皂素0.8-1.2份、枳椇子5-10份、甘草2-6份、蜂蜜6-10份、白砂糖1-3份。
7.根据权利要求1所述的茶皂素解酒护肝饮料的制备方法,其特征在于,饮料的pH为7.5-8.0。
8.根据权利要求1所述的茶皂素解酒护肝饮料的制备方法,其特征在于,所述pH调节剂为碳酸氢钠。
9.一种如权利要求1-8任一项所述方法制备的茶皂素解酒护肝饮料。
