天山雪莲是我国稀有的传统名贵药材,入药记载可以追溯到公元8世纪的藏药典《月王药珍》,并在其后的多部著名医药典籍中出现,具有活血通经、散寒除湿、强筋助阳等功效,民间俗有“东北人参,新疆雪莲”之说,千余年来广为人们所采用,汉族人民多视为治疗风湿关节炎之珍品,维吾尔、哈萨克族则当作治疗痛经、月经不调等的妇科良药。长期的掠夺性采挖已使天山雪莲资源严重匮乏,自然资源难以满足日益增长的需要,而人工大面积栽培的关键技术仍未获得突破,雪莲细胞培养物是选取天山雪莲离体组织通过脱分化形成的愈伤组织作为继代种子,给予一定条件进行继代培养而获得的较大愈伤组织团块。天山雪莲含有丰富的蛋白质和氨基酸,可有效地调节人体酸碱度,增强人体免疫力及具有抗疲劳,抗衰老作用。雪莲能强筋壮阳、通经活血、暖经祛瘀、散寒除湿,主治风湿性关节炎,男性阳痿诸症、女性宫寒腹痛、月经不调等疾病。据现有技术表明,雪莲还有一定的抗肿瘤的功效。目前市场上有少量以雪莲培养物给主要成分的保健功能食品,但是种类少,口感欠佳,还没有利用雪莲培养物制备具有抗肿瘤功效的饮料的技术,且在人体中抗肿瘤作用的实现靠多种物质的相互配合才能实现,仅使用单种雪莲培养物难以达到有效的抗肿瘤效果。
发明内容
为弥补现有技术的不足,本发明提供一种含雪莲培养物和茶氨酸的饮料,由多种天然物质成分混合而成,具有良好的抗肿瘤效果,并赋予泡腾饮料的形式,满足消费者对口感的要求。
本发明的技术方案如下:一种含雪莲培养物和茶氨酸的固体饮料,包括以下质量份数的物质:雪莲培养物60份、茶氨酸78份、茶树花粉90份、银耳多糖70份、发酵果粉300份、壳聚糖150份、木糖醇100份、牛磺酸4份、酸源200份、碳酸氢钠200份;所述的雪莲培养物粒径为13-100nm;
所述的发酵果粉是按照如下方法制备的:(1)发酵:将新鲜蔓越莓100份、蓝莓200份进行绞碎、向其中加入低聚果糖150份再进行匀浆处理,接入酿酒酵母2-4份,在25-26℃下密闭发酵5-6天;(2)冷冻干燥:将发酵产物进行冷冻干燥,粉碎后过100目筛即得发酵果粉;
所述固体饮料的制备方法如下:
S1、将所述的茶氨酸与壳聚糖进行如下包埋处理:(1)取壳聚糖150份用纯净水洗涤后,冷冻干燥;(2)将吐温80、冷冻干燥后的壳聚糖溶于水中,加入茶氨酸78份进行超声乳化,制得水相;(3)向60份棕榈油中加入橄榄油5份,在50℃条件下进行磁力搅拌,充分混合均匀,制得油相;(4)将步骤(2)得到的水相加入到步骤(3)制得的油相中,进行超声波乳化;超声波乳化后冷冻干燥;
S2、将雪莲培养物60份、茶树花粉90份、银耳多糖70份、发酵果粉300份充分混合过高速整粒机得预混料A;
S3、将茶氨酸与壳聚糖的包埋处理物、木糖醇100份、牛磺酸4份、碳酸氢钠200份充分混合过高速整粒机得预混料B;
S4、将酸源200份、预混料A、预混料B一起混合均匀即可。
进一步的,所述的固体饮料还包括二氧化硅30-40份。
进一步的,所述的酸源为本领域常规的呈酸性的物质,优选为柠檬酸、酒石酸或苹果酸。
进一步的,所述的酿酒酵母为3份。
茶氨酸(谷氨酸γ-乙基酰胺)是茶叶中特有的游离氨基酸,在干茶中占重量的1%-2%,其含量因茶的品种、部位而变化。在化学结构上与脑内活性物质谷酰胺、谷氨酸相似,是茶叶中生津润甜的主要成份。
本发明的有益效果如下:
(1)本发明制备一种新型的含雪莲培养物的固体泡腾饮料,将雪莲培养物(粒径为13-100nm)这一营养保健成分与现代新型饮料形式结合,制备的饮料不仅口感好,还具有良好的保健功能效果,且该饮料冲泡效果好,泡沫丰富,颜色鲜艳,酸甜可口;且纳米级的雪莲培养物,更易于人体吸收和利用;
(2)本发明将雪莲培养物、茶氨酸等天然活性成分按照上述质量份数比制备成泡腾颗粒固体饮料,具有携带方便、易于口服、生物利用度高的优点,另外通过将酸剂和碱剂混合的方法,在水冲泡下发生酸碱中和反应,产生二氧化碳气体的泡腾现象使得该产品具有良好的视觉欣赏效果;
(3)本发明配方中的发酵果粉经酿酒酵母发酵,发酵产物口味独特,且将果粉中的物质经微生物代谢为多种小分子物质,解决了天然果粉果味不佳、营养成分单一的问题;
(4)本发明配方中的壳聚糖具有活化细胞和保湿功能,能抑制肿瘤血管内皮细胞的生长,且将壳聚糖与茶氨酸进行包埋处理,还解决了茶氨酸容易被空气氧化以及在消化道中被酸性环境影响失活的问题,被包埋的茶氨酸还能缓慢释放,逐渐被肠道吸收;本发明将茶氨酸和雪莲培养物进行复配,二者相互促进起到了更好的抑制肿瘤的效果,对肿瘤的抑制率较二者单独使用时抑制率之和高30%左右;
(5)本发明的固体泡腾饮料将抑、清、防三维功能集成于一体,抑制:雪莲培养物可以抑制肿瘤细胞的生长,而茶氨酸与雪莲培养物并用时,茶氨酸能阻止抗肿瘤成分从肿瘤细胞中流出,可以延长抗肿瘤药物的作用时间,从而增强整体抗癌效果;清理:雪莲培养物、茶树花粉、蔓越莓中富含黄酮、茶多酚、花青素等超强抗氧化成分,联合使用可以有效清除有害自由基,保护正常细胞不被肿瘤细胞侵蚀感染;防御:低聚果糖、银耳多糖等,富含膳食纤维和多糖物质,可以促进肠道蠕动,调节肠道代谢水平,排除毒素,进而达到增强机体免疫力的作用,总体上提高了人体自身对肿瘤细胞的抵御能力。

下面结合具体实施例对本发明的技术方案作进一步的说明,但本发明不以任何形式受限于实施例内容。实施例中所述试验方法如无特殊说明,均为常规方法;如无特殊说明,所述试剂和生物材料,均可从商业途径获得。
以下实施例中各物料成分的配比如下表1:

实施例1
S1发酵果粉的制备
将新鲜蔓越莓100份、蓝莓200份进行绞碎、向其中加入低聚果糖150份再进行匀浆处理,接入酿酒酵母2份,在25-26℃下密闭发酵5天;将发酵产物进行冷冻干燥,粉碎后过100目筛即得发酵果粉;S2茶氨酸与壳聚糖进行如下包埋处理
(1)取壳聚糖150份用纯净水洗涤后,冷冻干燥;
(2)将吐温80、冷冻干燥后的壳聚糖溶于水中,加入茶氨酸78份进行超声乳化,制得水相;
(3)向60份棕榈油中加入橄榄油5份,在50℃条件下进行磁力搅拌,充分混合均匀,制得油相;
(4)将步骤(2)得到的水相加入到步骤(3)制得的油相中,进行超声波乳化,超声波乳化后冷冻干燥;
S3整粒混合
(1)将雪莲培养物(粒径30nm)60份、茶树花粉90份、银耳多糖70份、发酵果粉300份充分混合过高速整粒机得预混料A;
(2)将茶氨酸与壳聚糖的包埋处理物、木糖醇100份、牛磺酸4份、碳酸氢钠200份充分混合过高速整粒机得预混料B;
(3)将酸源200份、预混料A、预混料B一起混合均匀即可。
实施例2
S1发酵果粉的制备
将新鲜蔓越莓100份、蓝莓200份进行绞碎、向其中加入低聚果糖150份再进行匀浆处理,接入酿酒酵母4份,在25-26℃下密闭发酵6天;将发酵产物进行冷冻干燥,粉碎后过100目筛即得发酵果粉;S2茶氨酸与壳聚糖进行如下包埋处理
(1)取壳聚糖150份用纯净水洗涤后,冷冻干燥;
(2)将吐温80、冷冻干燥后的壳聚糖溶于水中,加入茶氨酸78份进行超声乳化,制得水相;
(3)向60份棕榈油中加入橄榄油5份,在50℃条件下进行磁力搅拌,充分混合均匀,制得油相;
(4)将步骤(2)得到的水相加入到步骤(3)制得的油相中,进行超声波乳化,超声波乳化后冷冻干燥;
S3整粒混合
(1)将雪莲培养物(粒径100nm)60份、茶树花粉90份、银耳多糖70份、发酵果粉300份充分混合过高速整粒机得预混料A;
(2)将茶氨酸与壳聚糖的包埋处理物、木糖醇100份、牛磺酸4份、碳酸氢钠200份、二氧化硅40份充分混合过高速整粒机得预混料B;
(3)将酸源200份、预混料A、预混料B一起混合均匀即可。
实施例3
S1发酵果粉的制备
将新鲜蔓越莓100份、蓝莓200份进行绞碎、向其中加入低聚果糖150份再进行匀浆处理,接入酿酒酵母3份,在25-26℃下密闭发酵6天;将发酵产物进行冷冻干燥,粉碎后过100目筛即得发酵果粉;S2茶氨酸与壳聚糖进行如下包埋处理
(1)取壳聚糖150份用纯净水洗涤后,冷冻干燥;
(2)将吐温80、冷冻干燥后的壳聚糖溶于水中,加入茶氨酸78份进行超声乳化,制得水相;
(3)向60份棕榈油中加入橄榄油5份,在50℃条件下进行磁力搅拌,充分混合均匀,制得油相;
(4)将步骤(2)得到的水相加入到步骤(3)制得的油相中,进行超声波乳化,超声波乳化后冷冻干燥;
S3整粒混合
(1)将雪莲培养物(粒径13nm)60份、茶树花粉90份、银耳多糖70份、发酵果粉300份充分混合过高速整粒机得预混料A;
(2)将茶氨酸与壳聚糖的包埋处理物、木糖醇100份、牛磺酸4份、碳酸氢钠200份、二氧化硅35份充分混合过高速整粒机得预混料B;(3)将酸源200份、预混料A、预混料B一起混合均匀即可。
实施例4
S1发酵果粉的制备
将新鲜蔓越莓100份、蓝莓200份进行绞碎、向其中加入低聚果糖150份再进行匀浆处理,接入酿酒酵母2份,在25-26℃下密闭发酵6天;将发酵产物进行冷冻干燥,粉碎后过100目筛即得发酵果粉;S2茶氨酸与壳聚糖进行如下包埋处理
(1)取壳聚糖150份用纯净水洗涤后,冷冻干燥;
(2)将吐温80、冷冻干燥后的壳聚糖溶于水中,加入茶氨酸78份进行超声乳化,制得水相;
(3)向60份棕榈油中加入橄榄油5份,在50℃条件下进行磁力搅拌,充分混合均匀,制得油相;
(4)将步骤(2)得到的水相加入到步骤(3)制得的油相中,进行超声波乳化,超声波乳化后冷冻干燥;
S3整粒混合
(1)将雪莲培养物(粒径60nm)60份、茶树花粉90份、银耳多糖70份、发酵果粉300份充分混合过高速整粒机得预混料A;
(2)将茶氨酸与壳聚糖的包埋处理物、木糖醇100份、牛磺酸4份、碳酸氢钠200份、二氧化硅30份充分混合过高速整粒机得预混料B;
(3)将酸源200份、预混料A、预混料B一起混合均匀即可。
对本发明的四个实施例制备的固体饮料冲泡后进行感官评定,结果如下表2:

实施例5
将本发明实施例2制备的固体饮料以2倍质量的水冲泡后,进行抗肿瘤作用研究:对雌性裸鼠的腹腔内移植1×106个/1mL的人大肠癌细胞株,移植后1天2次以30g/kg连续饮用本发明的固体饮料50天,作为对照组,以30g/kg连续饮用纯净水50天,在试验结束的第二天摘出肿瘤,测定肿瘤重量,基于试验组和对照组的肿瘤重量测定抑制率,试验组对肿瘤的抑制率达35%。
实施例6
将本发明实施例3制备的固体饮料以2倍质量的水冲泡后,进行抗肿瘤作用研究:对雌性裸鼠的腹腔内移植1×106个/1mL的人大肠癌细胞株,移植后1天2次以50g/kg连续饮用本发明的固体饮料50天,作为对照组,以50g/kg连续饮用纯净水50天,在试验结束的第二天摘出肿瘤,测定肿瘤重量,基于试验组和对照组的肿瘤重量测定抑制率,试验组对肿瘤的抑制率达41%。
对比例7
按照实施例2的制备方法,但原料中不含茶氨酸,即无茶氨酸与壳聚糖的包埋处理过程,制备固体饮料,将制备的固体饮料以2倍质量的水冲泡后,进行抗肿瘤作用研究:对雌性裸鼠的腹腔内移植1×106个/1mL的人大肠癌细胞株,移植后1天2次以30g/kg连续饮用本发明的固体饮料50天,作为对照组,以30g/kg连续饮用纯净水50天,在试验结束的第二天摘出肿瘤,测定肿瘤重量,基于试验组和对照组的肿瘤重量测定抑制率,试验组对肿瘤的抑制率达5%。
对比例8
按照实施例2的制备方法,但原料中不含雪莲培养物,制备固体饮料,将制备的固体饮料以2倍质量的水冲泡后,进行抗肿瘤作用研究:对雌性裸鼠的腹腔内移植1×106个/1mL的人大肠癌细胞株,移植后1天2次以30g/kg连续饮用本发明的固体饮料50天,作为对照组,以30g/kg连续饮用纯净水50天,在试验结束的第二天摘出肿瘤,测定肿瘤重量,基于试验组和对照组的肿瘤重量测定抑制率,试验组对肿瘤的抑制率达6%。
从对比例7、8可以看出,当配方中不含茶氨酸或者雪莲培养物时,其对肿瘤的抑制率仅为5%-6%,而对比例5和对比例6中二者结合的配方中对肿瘤的抑制率可达到35%以上。由此可见,本发明提供的固体饮料通过将雪莲培养物和茶氨酸复配对抗肿瘤作用起到了相互增强的效果,复配饮料对肿瘤的抑制率大于二者单独使用时的抑制率之和。
应当理解,上述实施例只为说明本发明的技术构思及特点,其目的在于让熟悉此项技术的人士能够了解本发明的内容并加以实施,并不能以此限制本发明的保护范围,凡根据本发明精神实质所作的等效变化或修饰,都应涵盖在本发明的保护范围内。
Claims (4)
1.一种含雪莲培养物和茶氨酸的固体饮料,其特征在于,包括以下质量份数的原料:雪莲培养物60份、茶氨酸78份、茶树花粉90份、银耳多糖70份、发酵果粉300份、壳聚糖150份、木糖醇100份、牛磺酸4份、酸源200份、碳酸氢钠200份;所述的雪莲培养物粒径为13-100nm;
所述的发酵果粉是按照如下方法制备的:(1)发酵:将新鲜蔓越莓100份、蓝莓200份进行绞碎、向其中加入低聚果糖150份再进行匀浆处理,接入酿酒酵母2-4份,在25-26℃下密闭发酵5-6天;(2)冷冻干燥:将发酵产物进行冷冻干燥,粉碎后过100目筛即得发酵果粉;
所述固体饮料的制备方法如下:

S3、将茶氨酸与壳聚糖的包埋处理物、木糖醇100份、牛磺酸4份、碳酸氢钠200份充分混合过高速整粒机得预混料B;
S4、将酸源200份、预混料A、预混料B一起混合均匀即可。
2.如权利要求1所述的一种含雪莲培养物和茶氨酸的固体饮料,其特征在于,还包括二氧化硅30-40份。
3.如权利要求1所述的一种含雪莲培养物和茶氨酸的固体饮料,其特征在于,所述的酸源为柠檬酸、酒石酸或苹果酸。
4.如权利要求1所述的一种含雪莲培养物和茶氨酸的固体饮料,其特征在于,所述的酿酒酵母为3份。
