背景技术
(一)我国饮酒现状
在中国人的生活里,酒早已超出了一般意义。传统的酒文化让我们把酒当成了一种交际工具,喝酒则变成忠诚度和友情深浅的一种考量。在这种“酒风”盛行的趋势下,喝酒给我们带来的健康危害以及一系列社会问题也不断上涨。早在2006年,世界卫生组织就将中国列为世界酒精“重灾区”,由酒精引起的死亡率和各种疾病的发病率均高于吸烟。世卫的数据统计,有60种疾病是由于饮酒不健康造成的。而我国每年死于酒精中毒的人数超过11万人,因酒致残人数超过273万人。近年来中国饮酒人数一直呈上升趋势,目前已超过5亿人,其中又有近四成人每天饮酒1次以上,全国每年能喝掉300亿公斤左右的酒。卫生部、科技部和国家统计局曾发布的一项调查显示,我国女性的饮酒率比上世纪90年代初期上升了73%左右。按照这样计算,全国居民中真正做到滴酒不沾的可能不足2亿人,而沾染过白酒、啤酒及其他酒类饮料的人数不会少于8亿人,这是个令人震惊的数据。全国有157万酒瘾者。
(二)饮酒的危害:
过量饮酒会导致乙醇中毒,诱发各种疾病,特别是肝及神经系统的损伤。乙醇能促进体内氧自由基的生成和脂质过氧化反应,导致血液黏度增高,诱发血管神经系统疾病。体现在以下方面:(1)损害肝脏,一个健康肝脏1小时仅能分解13.6克酒精(约335毫升啤酒)。过量饮酒严重损害肝脏,约90%的酒精在肝脏内代谢,一次醉酒相当于得了一次肝炎。即便没有喝醉酒,只要是长期过量饮酒,酒精也会像“沉默杀手”一样损害肝脏:当摄入大量酒精或长期饮酒者,乙醇通过微粒体氧化酶系代谢,生成大量乙醛。由于乙醛干扰三羧酸循环的正常进行,脂肪酸氧化受阻,使磷酸二羟丙酮转变为磷酸甘油,后者与脂肪酸一起合成甘油三酯,沉着于肝细胞内,而形成肝细胞脂肪变即成酒精性脂肪肝,接着是酒精性肝炎,随后就是炎症坏死、肝纤维化,最后导致肝硬化,肝功能衰竭。重度饮酒者中80%以上有一定程度的脂肪肝,10%~35%可发展成酒精性肝炎,10%~20%将发展为肝硬化。每天摄入80克酒精,喝上十几年,50%都会出现肝硬化;(2)灼伤消化道黏膜:酒精进入消化道后,会对食管黏膜、胃黏膜和肠黏膜产生化学性灼伤,导致胃出血、胃炎、肠功能紊乱等问题。还会增加肝癌、胰腺癌、食道癌等的患病风险;(3)急性酒精中毒来势凶猛,有两个严重的并发症,一是喝多了呕吐,会因神志不清导致窒息,轻则肺炎,重则死亡;二是引发低血糖昏迷,还可以因神经麻痹导致尿意减退,憋尿憋到膀胱破裂;(4)饮酒是II型糖尿病的重要致病因素,糖尿病患者饮酒会导致血糖波动大,增加并发症的风险;(5)酒精对心脑血管的伤害也很大,会导致酒精性心肌病、高血压等多种疾病;酗酒还是导致心梗、脑梗、脑溢血等突发事故的一大因素;(6)长期饮酒,乙醛积蓄,损害中枢神经系统,表现有体重减轻、贫血、谵妄、视听幻觉、行烦懒散、不守鞭幅,智力丧失和精神障碍;(7)破坏生殖系统:酒精会导致男性精子数量减少,致使其不育。孕妇饮酒生出来的孩子易患“胎儿酒精综合征”,甚至能造成流产、死产、早产或胎儿畸形;综上所述,要纠正中国人的酒文化习俗实在太难,但饮酒危害健康事关民族兴忘,因此,研究开发解酒护肝饮料或药品就成了非常必要。
(三)国内外解酒技术发展现状与趋势
解酒药总体可以分为化学药品、中药制剂、保健品三种,其解酒功效随药性快慢有关系。化学药品和中药制剂,在解酒的同时,会对肝脏、肾脏造成代谢负担而产生不同程度的伤害;而药食同源的保健品,既不会产生任何副作用,而且可以对受损肝脏细胞起修复营养的作用,安全可靠,因此,得到了越来越多的专家和消费者的青睐。
化学药品:多是兴奋剂、维生素与氨基酸等成份,能快速的起到解酒、醒酒作用,但大部分的化学药品解酒机理是仅仅能促进酒精分解,起到暂时性的缓解作用,很难起到护肝作用;中药制剂:成份多为姜黄、枳子、葛根花、白芍、肉豆蔻,解酒护肝丸,醒酒王片等中药制剂也能解酒,但效果迟缓、不甚理想;国内生产的海王金樽片:由牡蛎精粉、维生素C、L-半胱氨酸、牛磺酸、泛酸钙、淀粉、蔗糖组成,具有对化学性肝损伤有辅助保护作用。美国研发的《RU21安体普复合解酒片》,核心成分是一种产自俄罗斯的天然物质,配以维生素C、B2、B6和谷氨酸等,在饮酒时发挥高效解酒性能和对人体起到全面保护作用。但因药源不足和价格太贵(193元人民币20片)未在中国市场推开。
发明内容
本发明涉及利用现代生物技术,用中华草本原料枳椇子、葛花、决明子、荷叶萃取物,研制出一种解酒护肝的泡腾性饮料。
1、处方组成:枳椇子、葛花、决明子、荷叶。
2、方解:
(1)葛花:葛花中含有的皂角苷和异黄酮,皂角苷有强大的保肝功效,而异黄酮则有强烈的消除活性氧的作用;徐燕琴等研究提示:鲜葛花汁通过提高ADH活性,加强乙醇在胃肠道的首过效应,降低血中的乙醇浓度,激活微粒体乙醇氧化系统(MEOS),加速乙醇及其代谢产物的消除速率,达到解酒的目的。雷红伟等研究证实:葛花水提液能够抑制脂质过氧化反应。减少MDA的含量,提高谷胱甘肽酶(GST)活性,增强谷胱甘肽(GSH)的抗氧化作用,减轻急性酒精性肝损伤。柳海艳等研究进一步揭示:乙醇模型组动物肝细胞的细胞核、线粒体、内质网受到严重的损伤,出现大量的脂肪聚集和贮脂细胞,而各给药组相应减轻,其中配比组对肝细胞超微结构保护作用,强于单味葛花组和单味枳椇子组;葛花/枳椇子为2/1组,对肝细胞超微结构的保护作用最强。
(2)枳椇子,又名木蜜、树蜜、拐枣、万寿果、鸡爪子等。枳椇子功效:止渴除烦,消湿热,解酒毒;用于酒中毒、烦渴呕逆、二便不利;枳椇子含有大量的葡萄糖、苹果酸钙,有较强的利尿作用,能促进乙醇的分解和排出,显著降低乙醇在血液中的浓度,并能消除乙醇在体内产生的自由基,阻止过氧化酯质的形成,从而减轻乙醇对肝脏等的损伤;实验还证明枳椇子提取物具有明显的抗肝纤维化作用并能促进肝纤维化时I、III型胶原的降解。从而起到阻抑酒精性肝硬化的进程。
(3)荷叶具有清暑利湿、升发清阳、清心去热、止血利水活性中除了共同的碳水化合物外,还含有生物碱、黄酮、挥发油、有机酸、皂苷甾体等有效成分,能有效抑制高胆固醇血症和动脉粥样硬化、降脂减肥、抗病毒、抗菌和抗惊厥等药理作用。
(4)决明子除含有糖类、蛋白质、脂肪外,还含甾体化合物、大黄酚、大黄素等,还有人体必需的微量元素铁、锌、锰、铜、镍、钴、钼等。所含大黄素、大黄酸对人体有平喘、利胆、保肝、降压功效,并有一定抗菌、消炎作用。其中大黄素葡萄糖甙、大黄素蒽酮、大黄素甲醚,具有降低血清胆固醇和强心作用。决明子素具有a-羟基,可与金属素合成络合物,对金属元素吸收有很大影响。现代临床用决明子治疗高脂血症收效满意,决明子水煎或制成片剂服用。民间常用决明子炒黄末,代茶饮,有预防和治疗疾病的保健功能。
综上所述,葛花、枳椇子解酒、护肝作用明显。决明子护肝利胆,促进酒精分解化谢;荷叶降脂减肥,消除酒精性脂肪肝,复方配伍,效力增强。本发明将药食两用中华本草萃取物做成泡腾颗粒或泡腾片,既保留了涼茶的滋味、营养和多环节解酒护肝功能,又具有体积小、重量轻、便于携带,使用方便,安全性、趣味性、愉悦性强的特点。实现了内容形式双创新。
具体实施方式
(1)原料:枳椇子、葛花、决明子、荷叶。按含量为30%、25%、25%、20%配比。
(2)中草药萃取:将配方额定的漂洗干净的葛花、枳椇子、决明子、荷叶粉碎放入萃取罐中,加入10倍量的纯净水,用蒸汽加热到85-90℃。恒温30min萃取液过滤后备用。再往萃取罐中加入8倍量的纯净水,用蒸汽加热到85-90℃,恒温20min取滤液过滤后,与第一次的滤液混合、喷雾、干燥后研粉。
配料
1)制备酸粒:将葛花、枳椇子、决明子、荷叶萃取物加柠檬酸、酒石酸、木糖醇、阿斯巴甜等粉碎、混合均匀,过80目筛,加入质量分数为1%PVP的无水乙醇作为粘合剂,进行软材制粒,过14目筛,于500C烘干,再用制粒机制粒,过10目筛;
2)制备碱粒:将葛花、枳椇子、决明子、荷叶萃取物干粉加碳酸钠、碳酸氢钠、木糖醇、阿斯巴甜等,按(1)方法同法制粒。
3)将所制得的酸粒、碱粒添加5%的低取代羟丙基纤维素(L.HPC)混合均匀,制粒;压片或制丸,于50℃烘干;及时包装;得成品。(提取物∶麦芽糊精=5∶2,酸味剂的添加量为10%,碱味剂的添加量为6.5%,甜味剂的添加量为1.0%)。工艺流程见附图,“附图说明:附图是泡腾剂的生产工艺流程”。

1.该发明是一种泡腾性饮料,包括颗粒剂,片剂,丸剂。
2.该发明物的主要原材料是枳椇子、葛花、决明子、荷叶。按含量为30%、25%、25%、20%的萃取物。
3.该发明物辅料成分有麦芽糊精、柠檬酸、酒石酸、碳酸钠、碳酸氢钠、木糖醇、阿斯巴甜和5%的低取代羟丙基纤维素(L.HPC)等。
4.该发明物物料配比,萃取物∶麦芽糊精=5∶2,酸味剂添加量为10%,碱味剂添加量为6.5%,甜味剂添加量为1.0%。
5.该泡腾剂放入洁净水中,立即弃放出大量二氧化碳,使杯中液体“泡腾”,可作为凉茶---清凉饮料,清热解渴,又具有解酒、护肝、降脂、减肥功效,最适合饮酒熬夜人士和酒精性肝炎,酒精性脂肪肝及其有酒精性肝纤维化趋向、肥胖等人士饮用。
