Description
本發明涉及一種用於解酒和預防宿醉的組合物,尤其涉及一種下述的用於解酒和預防宿醉的組合物以及包含該組合物的解酒飲料,該組合物作為有效成分含有枳椇果實萃取濃縮液(或萃取液)、刺梨萃取物、黃杞萃取物、蓮(蓮子)萃取物以及煙醯胺,其能夠降低在體內被吸收/分解而引起宿醉的血液中乙醇和乙醛的濃度,使得有利於解酒。
宿醉是對飲酒後出現的肉體和精神上的不適感的總稱,其客觀的症狀有認知能力低下、噁心、嘔吐、乏困、頭痛、運動能力低下、血液學變化以及激素變化等。
在日常生活中,現代人承受各種壓力,尤其是在文明高度發達的現代社會中,人們所要承受的壓力強度越來越高。
雖然很多現代人利用運動、睡眠、吸煙、旅行等各種方法來消除這種壓力,但是,可以說在現代社會中最為普遍的解壓方法還是飲酒(攝取酒精)。
這是由於對於現代人來說,所謂飲酒的行為已經不僅僅是用於解壓的手段,而是在商務接待、同事或熟人之間的應酬、家庭聚會等廣泛的範圍內形成一種文化。
特別是,隨著社會變得越複雜和組織化,飲酒的機會也越增加,與此同時,暴飲、過飲等現象也增加。在韓國,存在其獨特的飲酒文化,很多人因頻繁的飲酒和過飲而發生宿醉,並影響第二天的正常生活,也有很多因長期的不良飲酒習慣而損害健康的情況。
被吸收到體內的酒精(乙醇)主要在肝臟中進行代謝,首先生成乙醛後被氧化成乙酸鹽排出體外。在從乙醇氧化成乙醛的過程中,起到催化作用的是乙醇脫氫酶(ADH,alcohol dehydrogenase),而在由乙醇氧化而生成的乙醛的分解/氧化過程中,起到催化作用的是乙醛脫氫酶(ALDH,acetaldehyde dehydrogenase)。
正常的乙醇代謝過程如下所述:攝取的乙醇在胃或小腸內被吸收,進入血管後移送至肝臟,在肝細胞中,乙醇經乙醇脫氫酶(ADH,alcohol dehydrogenase)的作用氧化成乙醛,乙醛再經乙醛脫氫酶(ALDH,aldehyde dehydrogenase)的作用分解成乙酸鹽,最後分解成二氧化碳和水排出體外。
對於引起宿醉的原因,雖然還未完全研究清楚,但是,有很多學術觀點認為其原因是:脫水、乙醇以及乙醇的代謝物(乙醛、甲醛、丙酮)的毒性作用、營養素的吸收障礙引起的營養缺乏等。例如,最近有觀點指出乙醇中少量含有的甲醇的代謝以及甲醛是引起宿醉的原因(Jones AW. Pharmaco Toxicology. 60(3) 217~220),但也有一些報告稱,乙醇的代謝產物即乙醛、丙酮等是實際上引起宿醉的原因(Tomita Y,Haseba T,Kurosu M,Watanabe T(1990). Arukoru Kenkyut Yakubutsu Ison. 25(2). 116-128)(Tsukamoto S,Muto T. Nagoya T,Shimamura M,Saito T,Tainaka H.(1989). Alcohol Alcohol. 24(2).101~108)。
人可以分為即使在體內的乙醛濃度低的情況下也開始氧化的2型、以及在高濃度的情況下開始氧化的1型,東方人一般屬於2型,與大多屬於1型的西方人相比,乙醛的氧化慢,因此相對地更容易發生宿醉。
在韓國獨特的飲酒文化的社會背景下,一直存在消費者對解酒產品的需求,從而在多個領域中正在進行為了減輕飲酒引起的宿醉的研究和實驗,並且,實際上,在市場上已有多種產品,並不斷地有新產品上市。
其中,大部分產品雖然利用的是中藥材等藥效好的材料,但是,其使用量不足,並且,對於實際製造出的最終產品的效果也缺乏客觀的驗證,因此,在現實中,實際需要解酒飲料的消費者對產品的信賴度並不高。
因此,迫切需要開發出如下所述的產品:其利用具有解酒效果的原料或組合物製造,並進行客觀的解酒效果驗證,使得消費者在飲用產品後確實能夠獲得效果,由此得到消費者的信賴,減少因過多的飲酒引起的社會損失,並且有利於個人的健康生活。
枳椇(Hovenia dulcis Thunb.)是屬於鼠李科的植物,其果實帶有肉質果柄,將其果實稱為枳椇子。枳椇子從古代開始就被用作藥材,其味甘、性平,具有止渴除煩、去濕熱、解酒毒的功效。在本草綱目等中醫古醫書中記載了枳椇子是用於解酒毒的最佳藥材。
近年來,在各個領域中對枳椇的生理活性進行了廣泛的研究,並且,事實上,在很多研究文獻中客觀地驗證了其對酒精的分解以及肝毒性的解毒等方面所起到的效果。
下面,舉例說明實際的研究事例。枳椇成分的酒精分解(等,韓國生物工學會誌Vol 21. No.1,11~15(2006),等,韓國食品營養科學會誌,35(7),828~834(2006),等,藥作誌7(4),263~268(1999)),肝解毒(等,藥作誌8(3),225~233(2000),等,獸醫學誌24(3),384~391(2007)),乙醇中毒對學習能力的影響(,藥作誌11(3),232~235(2003))。
本發明的發明人在韓國授權專利10-0620093中已經公開了利用刺梨萃取物、黃杞萃取物、蓮萃取物製備的解酒組合物具有客觀的解酒效果,但是,考慮到期待開發出具有更好的效果的解酒飲料以及組合物的消費者的需求以及社會需求,完成了本發明。
刺梨(Rosa Roxburghii)是屬於薔薇科的植物,也被稱為甜栗薔薇(sweet chestnut rose),分佈在東亞(中國喜馬拉雅)地區,其果實和種子可以食用。原產地為中國的刺梨在中國貴州省一直被當作保健以及美容食品,其包括豐富的維生素等,並且SOD活性強,從而在日本被開發成用於防止老化、祛斑、防止皺紋等的美容飲料。
在韓國專利第003413號中,公開了利用薔薇果實萃取物(rose fruit extract: RFE)製造抗壓薔薇優酪乳的方法。此外,在韓國專利第0040338號中,公開了利用薔薇萃取物製造抗壓飲料的方法,該抗壓飲料可以有效地抑制活性氧的生成,並除去活性氧,從而有效地消除壓力。
黃杞(Engelgardtia chrysolepis HANCE)是屬於鼠李科的植物,一般被稱為臺灣黃杞(Taiwan engelhardtia)。黃杞分佈在中國的南部地區以及西南地區,其葉子可以食用,由於味道微甜,在中國一直被當作甘茶(sweet tea)飲用,並作為保健茶使用,在日本等地將其稱為Kohki tea,作為飲料以及茶廣泛飲用。
蓮(Nelumbo nucifera)是屬於水蓮科的植物,也被稱為蓮花,在韓國的中南部各個地區作為觀賞用植物栽培在蓮花池中。在地理上分佈在印度、中國、日本、西伯利亞,其幼葉、果實、根莖、以及種子可以食用。其種子(蓮子)具有益腎固精、鎮靜、健脾止瀉、養心安心的作用,可以用於治療長期腹瀉、遺精帶下、心悸失眠等,其葉子具有解熱、解毒作用,其種子和果肉具有益腎固精、止血的作用,可以用於治療夜尿症、婦科病,其根莖具有解熱毒、消瘀血、止血的作用,可以用於治療一切血症、尿血、腸出血。
在韓國專利0063588號中,公開了用於保護肝細胞、預防或治療肝損傷的蓮(Nelumbo nucifera Gaertn.的成熟果實)萃取物,以及作為有效成分包含該萃取物的用於保護肝細胞、預防或治療肝損傷、抑制因肝癌誘發物質引起的突變的組合物。
在韓國正在開發和銷售很多用於解酒的產品,例如,在韓國授權專利第0181168中公開了利用赤楊、花楸、日本女貞的萃取物以及葛根萃取物製造解酒天然茶的方法,在韓國授權專利第10-0839586號中公開了利用河豚萃取物、枳椇果實、紅參、黑蒜、秘魯人參、薑黃、梅、覆盆子、五味子、葛根製造用於解酒的飲料的方法,以及在韓國專利第0040515號中公開了利用具有酒精分解酶活性的海藻萃取物、白眼狹鱈萃取物、洋蔥萃取物、豆芽萃取物、水芹萃取物、蘿蔔萃取物製造解酒飲料的方法。
發明要解決的問題
鑒於上述的問題,本發明人為了開發出比現有的更優秀的解酒組合物以及飲料而進行了研究。本發明的目的在於,提供一種新的用於解酒和預防宿醉的組合物以及包含該組合物的用於解酒和預防宿醉的飲料,該組合物是在已經通過研究發現有解酒效果的刺梨、黃杞、蓮的各個萃取物中,新添加下述兩種物質,即,在本草綱目等中醫古醫書中記載為用於解酒毒的最佳藥材、且在各種研究文獻中客觀地驗證了飲酒後的肝解毒效果的枳椇果實萃取液,以及作為與體內的酒精代謝有密切的關係的輔酶的煙醯胺。
利用由本發明人設定的宿醉自我效能評價方法,對包含枳椇果實萃取物或濃縮液、刺梨萃取物、黃杞萃取物、蓮萃取物以及煙醯胺的用於解酒和預防宿醉的組合物進行宿醉程度的改善與否的評價,由此實現了本發明的上述目的。
另外,為了驗證本發明的用於解酒和預防宿醉的組合物的效能,進行了消費者是否能感覺到飲酒前後的解酒效能的自我效能實驗,其結果,全體應答者的83.7%感覺到有改善解酒的效能,即,獲得了客觀的效能結果。通過該實驗,可以確認根據本發明的解酒飲料以及組合物具有優秀的解酒效能。
發明效果
通過自我效能實驗的結果可知,根據本發明的包含枳椇果實萃取物或濃縮液、刺梨萃取物、黃杞萃取物、蓮萃取物以及煙醯胺的用於解酒和預防宿醉的組合物、以及包含該組合物的用於解酒和預防宿醉的飲料,具有顯著的解酒和預防宿醉的效能。
本發明涉及一種用於解酒和預防宿醉的組合物,更具體地說,涉及一種作為有效成分含有枳椇果實萃取液或枳椇果實濃縮液、煙醯胺、刺梨萃取物、黃杞萃取物以及蓮萃取物的用於解酒和預防宿醉的組合物。
另外,本發明還涉及一種包括上述組合物的用於解酒和預防宿醉的飲料。
下面,詳細地說明根據本發明的用於解酒和預防宿醉的組合物的製備方法。
在枳椇果實100g中加入4~10倍以上的水,在熱水(95℃)中萃取1~5小時,然後,進行1次過濾,並在95℃下進行10分鐘以上的滅菌處理,然後,再次進行過濾而製備萃取液。通過對這樣製備出的萃取液進行濃縮,可以獲得固形物濃度為30~60%的枳椇果實萃取濃縮液。
可以用未經濃縮的枳椇果實萃取液或者其他濃度的濃縮液來替代本發明的實施例提出的枳椇果實萃取濃縮液,但是,此時應當使各個成分的使用量與本發明的實施例相同,即,其固形物量應當與本發明的實施例提出的枳椇果實萃取濃縮液(固形物55%)1.0g的固形物量相同。
下面,進行具體的舉例說明。為了替代本發明提出的枳椇果實萃取濃縮液(固形物55%)1.0g(一次攝取量),固形物含量為4.5%的枳椇果實萃取液需要使用的量,可以利用下述的方法進行計算。
即,利用計算式:濃縮液固形物含量55%×含有量1.0%/萃取液固形物含量4.5%來進行計算。其結果,為了與在組合物中含有1.0%的固形物含量為55%的濃縮液所含有的固形物量相同,固形物含量為4.5%的枳椇果實萃取液在組合物中需要使用的量約為12%。
為了獲得上述解酒(包括預防)效果,可以飲用本發明提出的組合物,該組合物由枳椇果實萃取濃縮液(固形物含量55%)、刺梨萃取物、黃杞萃取物、蓮萃取物以及煙醯胺組成,以體重為60kg的成人為基準,日攝取的總量為1.353g以上,飲用次數不受限制。如果使用的是固形物含量為4.5%的枳椇果實萃取液,則如上所述,需要替代枳椇果實萃取濃縮液(固形物含量55%)而在組合物中含有12%的量,此時,攝取的組合物總量為12.353g。
此外,分別準備200g的刺梨、黃杞以及蓮,並分別加入2L的熱水(95℃),萃取2小時,從而獲得刺梨、黃杞以及蓮的萃取物。過濾該萃取物而獲得約400g的萃取液,靜置12小時之後,濃縮上清液而製備固形物濃度為50%的濃縮液,然後,對該濃縮液進行噴霧乾燥而分別獲得刺梨、黃杞以及蓮的萃取粉末。
煙醯胺可利用色氨酸合成,是在動植物中廣泛存在的維生素,也被稱為B12。其在體內以NAD+和NADP+的形態存在,作為脫氫酶的輔酶,參與很多氧化/還原反應。如下所示,對參與吸收到體內的酒精的分解作用的乙醇脫氫酶以及乙醛脫氫酶起到輔助的作用,從而促進體內的代謝。煙醯胺是作為食品原料使用於多個領域的物質,只要是純度為95%以上的食品級原料,就不分廠家都能夠使用。

根據本發明的用於解酒和預防宿醉的組合物作為有效成分包括:100重量份的上述枳椇果實萃取濃縮液;0.27-5.3重量份的煙醯胺;4-20重量份的刺梨萃取物;0.4-5重量份的黃杞萃取物;以及0.4-5重量份的蓮萃取物。
在本發明的組合物中,可以用枳椇果實萃取液替代枳椇果實萃取濃縮液,但是此時,如上所述,應該使用與濃縮液的固形物量相同的固形物量,其中,該濃縮液的固形物量是根據固形物含量和組合物中的含有量來計算的。
本發明的組合物可以用作藥用組合物。此時,能夠製備成除了上述有效成分之外還選擇性地包括可藥用的載體或添加劑的劑型。上述載體或添加劑可以是稀釋劑、甜味劑、穩定劑、香料、維生素類以及抗氧化劑等。
只要是可藥用的物質,就都能夠作為上述載體或添加劑使用。例如,稀釋劑可以是乳糖、穀物澱粉等,甜味劑可以是糖、液體果糖、結晶果糖、三氯蔗糖、山梨糖醇或阿斯巴甜,穩定劑可以是羧甲基纖維素鈉、β-環糊精或黃原膠等。並且,為了改善組合物的口感,還可以包括:果香、草香等天然香料;天然果汁、類黃酮等天然色素;果糖、蜂蜜、糖等甜味成分;或檸檬酸、檸檬酸鈉等酸味劑。此外,本發明的組合物除了有效成分之外還可以包括下述的輔助成分:維生素B、C、E或β-胡蘿蔔素;Ca、Mg、Zn等無機成分;或卵磷脂等磷脂成分;或低聚糖、胺基酸、牛磺酸等。
根據本發明的上述組合物,除了可以製備飲料之外,還可以製備用於口服的解酒用液體製劑(膠體、汁)、丸劑、粉末、片劑、膠囊,但並不限定於此。
下面,說明具體的實施例。
實施例1
枳椇果實萃取液(濃縮液)的製備
在枳椇果實100g中加入4~10倍以上的水,在熱水(95℃)中萃取1~5小時。過濾該萃取液之後,在95℃下進行10分鐘以上的滅菌處理,並再次進行過濾而製備萃取液。濃縮該萃取液,最終可以獲得固形物濃度為30~60%的枳椇果實萃取濃縮液。
刺梨、黃杞、蓮萃取物的製備
分別準備200g的刺梨果實、黃杞葉以及蓮子,並分別加入2L的熱水(95℃),萃取2小時,過濾該萃取液而獲得約400g的萃取液,靜置12小時之後,濃縮上清液而製備固形物濃度為50%的濃縮液,然後,對該濃縮液進行噴霧乾燥而分別獲得刺梨、黃杞以及蓮的萃取粉末。
實施例2
飲酒前後用於解酒的飲料組合物的製備
在實施例1的枳椇果實萃取濃縮液(固形物55%)10g(或枳椇果實萃取液(固形物4.5%)120g)、刺梨萃取粉末2g、黃杞萃取粉末0.5g、蓮萃取粉末0.5g中,添加煙醯胺0.53g、大米胚芽大豆發酵萃取物100g、麩胺酸酵母抽提物3.3g、牛磺酸20g、丙氨酸1g、白糖25g、液體果糖115.7g、dL-蘋果酸1.6g、檸檬酸2.97g、檸檬酸鈉0.4g、香料1.7g(1號混合果香0.5g、2號混合果香0.8g、蜂蜜香0.5g)、維生素BD 0.4g,並加入適合飲用的淨化水而調至1L。然後,進行攪拌,直至所有組分完全溶解,並且,進行高溫瞬間滅菌(HTST,98℃下進行30秒左右)處理,以殺滅殘留的微生物,然後注入到容器中。注入到容器中之後,在使材料溫度達到80℃以上的情況下進行5~10分鐘的後滅菌,然後,冷卻並貼標、包裝,從而製備出用於解酒的飲料。
實驗例1
宿醉自我效能驗證實驗
基於記載在國外論文(Pittler,M. H. et al. CMAJ*2003;169:1269-1273,canadian medical association or its licensors)中的宿醉評價問卷項目以及記載在韓國食品醫藥品安全廳的「關於開發解酒效能協定的研究報告書」中的宿醉程度的評價方法和資料,進行了對於宿醉的自我效能驗證實驗。
表1中示出了本實驗的調查對象的年齡、性別構成。
實驗調查對象的平均酒量,男性中的89%為3瓶以下,女性中的86%為2瓶以下。
將43名(男性28名,女性15名)受試者作為實驗對象,讓他們飲用本發明開發的實施例2的用於解酒和預防宿醉的飲料組合物100mL(1瓶),然後,根據各自的酒量進行充分的飲酒。在飲酒的第二天,讓實驗對象應答關於宿醉現象的解酒自我效能問卷,進行對於解酒的自我效能的問卷評價。在評價中使用的用於解酒和預防宿醉的飲料組合物的成分如表2所示(參照實施例2)。為了評價解酒自我效能而在本發明中使用的評價問卷如下述的表3所示。
對於各個分數的評價基準
1 無宿醉:完全不影響生活。
2 幾乎無宿醉:幾乎不影響生活。
3 一般水準:不影響生活,比平時的宿醉強度低。
4 基本宿醉:影響生活。
5 完全宿醉:宿醉強度很強,對生活的影響很大。
對自我效能進行評價的結果,在飲用了本發明的用於解酒和預防宿醉的組合物的情況下,在大部分的項目中顯示出對於宿醉症狀的改善(減少宿醉症狀)效果,尤其是,對於在宿醉評價項目中應答率高(75%以上)的主要宿醉項目、即疲勞、頭痛、口渴、胃灼熱/胃痛、乏困、嘔吐項目,70%至85%的實驗調查對象應答了有效果,由此可知本發明的用於解酒和預防宿醉的組合物在減輕宿醉方面具有明顯的改善效果。本發明的組合物對於手顫症、急躁、敏感、無力、肌肉痛等其他項目也顯示出改善效果,但是,考慮到大部分受試者對這些症狀的應答率比其他項目的應答率低,所以這些項目似乎不是主要宿醉症狀。詳細的實驗結果如表4所示。
另外,研究了根據本發明的用於解酒和預防宿醉的組合物對於全部宿醉症狀的消除以及改善效果,如圖1所示,在參加本實驗的43名實驗調查對象中,83.7%(36名)的實驗調查對象認知到了具有解酒效能,由此可知根據本發明的組合物具有良好的解酒效果。
圖1表示的是利用根據本發明的用於解酒和預防宿醉的組合物進行解酒自我效能實驗的結果,具體示出了在飲酒前攝取根據本發明的用於解酒和預防宿醉的組合物之後對全部宿醉症狀的改善與否進行了評價的結果。
圖2表示的是利用根據本發明的用於解酒和預防宿醉的組合物進行自我效能實驗的結果,具體示出了在飲酒前攝取根據本發明的組合物之後對各個宿醉項目的改善與否進行了評價的結果。
Claims (5)
- 一種用於解酒和預防宿醉的組合物,其含有枳椇果實萃取濃縮液、煙醯胺、刺梨萃取物、黃杞(Egelgardtia chrysolepis HANCE)萃取物以及蓮萃取物作為有效成分,其中該組合物相對於100重量份的枳椇果實萃取濃縮液含有以下者作為有效成分:0.27-5.3重量份的煙醯胺、4-20重量份的刺梨萃取物、0.4-5重量份的黃杞萃取物以及0.4-5重量份的蓮萃取物。
- 如申請專利範圍第1項的組合物,其中該枳椇果實萃取濃縮液是固形成分含量為30-60重量%的枳椇果實濃縮液。
- 如專利申請範圍第1項的組合物,其中該枳椇果實萃取濃縮液為以能提供與該枳椇果實萃取濃縮液相同含量的固形成分之量被含有的枳椇果實萃取液。
- 如申請專利範圍第1項至第3項中任一項的組合物,其中該組合物為液體形式。
- 一種用於解酒和預防宿醉的飲料組合物,其包含:如申請專利範圍第4項的液體組合物;以及進一步添加於其中之可藥用的載體或添加劑。
